따라서 본 발명의 목적은 알도오즈 환원효소 억제제 화합물을 포함하는 근원 세포의 증식 및 분화 촉진용 조성물을 제공하는 것이다.

이하, 본 발명을 상세하게 설명한다.
본 발명의 일 양태로서, 본 발명은 알도오즈 환원효소 억제제 화합물을 포함하는 근원 세포의 증식 및 분화 촉진용 조성물에 대한 것이다. 본 발명의 상기 알도오즈 환원효소 억제제는 그 종류에 제한되는 것은 피다레스타트(fidarestat), 톨레스타트(tolrestat), 에팔레스타트(epalrestat), 라니레스타트(ranirestat), 포놀레스타트(ponolrestat), 조폴레스타트(zopolrestat), 제나레스타트(zenarestat), 소르비닐(sorbinil), 메토소르비닐(methosorbinil), 이미레스타트(imirestat) 및 미날레스타트(minalrestat)로 이루어진 군으로부터 선택된 하나 이상의 물질일 수 있으며, 보다 바람직하게는 피다레스타트, 톨레스타트 에팔레스타트 또는 소르비닐 중에서 선택될 수 있다. 상기 물질은 하기의 화학식으로 표시될 수 있다.
본 발명의 일 양태로서, 본 발명의 알도오즈 환원효소 억제제는 근육 줄기세포 또는 근원세포의 증식 및 분화를 촉진하는 효과를 가지며, 이를 통하여 근육 재생 효과 증대, 근육량 증진 효과를 가질 수 있으며, 나아가 근 기능 저하, 근육 감소, 근육 위축, 근육 소모 또는 근육 퇴화로 인한 근육 질환의 예방 또는 치료 효과를 가질 수 있다.
상기 “근원세포의 증식 촉진”이란, 근원 세포의 줄기 세포인 위성 세포의 모집, 활성 또는 증식을 촉진하여, 근원 세포의 개수가 증가하고, 이로 인해 근원 세포의 증식이 증가되는 것을 의미한다. 이와 같은 근원 세포의 증식 촉진을 통하여 근감소증과 같은 다양한 근육 질환에서의 치료 효과를 가져올 수 있다.
또한 본 발명의 알도오즈 환원효소 억제제는 “근원세포 분화 촉진” 효과를 가지며, “근원 세포 분화” 란, 단핵인 근원 세포(myoblast)가 융합을 통해 다핵의 근관(myotube)를 형성하는 과정을 의미하며, 이러한 과정을 유도하는 것을 말한다. 근관을 형성하는 분화단계의 세포는 Pax7-, MyoD+, MyoG+ 등의 마커의 발현 여부를 통해 확인된다. 근원세포의 분화 촉진을 통하여 근육 세포가 증가되며, 개체 측면에서는 궁극적으로 근육량이 증가되는 효과를 가져올 수 있다.
또한 본 발명의 알도오즈 환원효소 억제제는 “근육 재생” 효과를 가지며, 상기 "근육 재생"이란, 손상된 근육이 정상으로 회복될 수 있는 능력을 의미한다. 상기 근육 재생은 근육 세포의 분화가 촉진되어 절대적인 근육 세포의 양이 증가된 것이거나 또는 개개의 근관의 직경이 증가된 것에 의한 효과일 수 있다.
또한 본 발명은 알도오즈 환원효소 억제제는 근육량 증진 또는 근력 강화 효과를 가진다. "근력 강화"란, 신체 수행의 강화, 최대 지구력의 강화, 근육량의 증가, 근육 회복의 강화, 근육피로의 감소, 에너지 수지의 개선 또는 이들의 조합 효과를 말한다. 앞서 설명한 바와 같이, 본 발명의 조성물은 근원세포를 근육 세포로 분화 시키는 능력을 통하여 근육량을 증가시켜 전체 근육량을 증가시킬 수 있고, 근육 재생을 촉진함으로써, 최대 지구력이 강화되고, 이에 따라 신체 수행이 강화되고 근육 피로도 감소할 수 있다. 또한, 근육 세포가 빠르게 대체될 수 있기 때문에 근육의 손상에 대하여 빠르게 치유될 수 있다.
또한, 본 발명의 알도오즈 환원효소 억제제 화합물은 근력 강화를 통하여 운동수행능력을 향상시킬 수 있으며, 이 때, "운동 수행능력"이라 함은, 근력을 이용하여 운동을 수행하는 능력을 말한다. 상기 근력은 근육의 양, 근 지구력, 산화성 근육량의 증가, 근육 회복력 및 근육 내 에너지 수지의 개선에 의해 향상될 수 있으며, 또한 근육 내 피로물질 감소 등에 의해서도 증진될 수 있으나, 본 발명의 알도오즈 환원효소 억제제 화합물은 특히 근원세포 증식 촉진 또는 근원세포 분화, 근육량 증가, 근육 재생 등의 효과를 통해 근력을 증진시키는 효과를 가진다.
본 발명의 알도오즈 환원효소 억제제는 근 기능 개선 효과를 가지는데, “근 기능 개선”이란 근육의 수축에 의해 힘을 발휘하는 능력으로서, 근육이 저항을 이겨 내기 위하여 최대한으로 수축력을 발휘할 수 있는 능력인 근력, 근육이 주어진 중량에 얼마나 오랫동안 또는 얼마나 여러 번 수축과 이완을 반복할 수 있는지를 나타내는 능력인 근지구력, 단시간 내에 강한 힘을 발휘하는 능력인 순발력을 포함한다. 이러한 근 기능은 근육량에 비례하고, "근 기능 개선"은 근 기능을 더 좋게 향상시키는 것을 의미한다. 또한, 본 발명의 알도오즈 환원효소 억제제는 근육 내 미토콘드리아 활성을 증진시키기 때문에 결과적으로 근육에서의 에너지 수지를 개선하므로, 근 기능을 개선하는 효과를 가질 수 있다.
본 발명의 알도오즈 환원효소 억제제를 포함하는 근원세포 증식 촉진 또는 근원세포의 분화능 촉진용 조성물, 근육 재생용 조성물, 근육량 증가용 조성물, 근력 강화용 조성물, 운동수행능력 향상용 조성물 및 근 기능 개선용 조성물은 식품 조성물 또는 식품첨가제 또는 건강식품 조성물의 형태로 제조될 수 있다. 본 발명의 근력 강화용 조성물은 노화 또는 질병에 의한 근육 질환 등의 예방 또는 치료 뿐만 아니라, 근육 기능이 정상인 일반인에 있어서도 근육 생성, 근육량 증가, 근력 강화 효과를 가진다. 따라서 기능성 식품 또는 이에 대한 보조제 등의 형태로 이용될 수 있다.
다른 양태로서 본 발명은 알도오즈 환원효소 억제제 또는 이의 약학적으로 허용 가능한 염을 포함하는 근육 질환에 대한 예방, 개선 또는 치료용 조성물에 대한 것이다. 즉, 본 발명의 알도오즈 환원효소 억제제는 근육 질환에 대한 예방, 개선 또는 치료 효과를 가진다. 상기 '근육 질환'은 근육이 노화 혹은 질병에 의해 손상되거나 손실되어 근력이 약화된 상태에서 발생한 근육 질환인 것을 의미하며, 이는 유전적 소인, 고혈압, 내당능장애, 당뇨, 비만, 이상지질혈증, 아테롬성경화증 또는 심혈관질환 등 노화와 관련된 질환; 암, 자가면역질환, 감염성 질환, AIDS, 만성 염증성 질환, 관절염, 영양 실조, 신장 질환, 만성 폐쇄성 폐질환, 폐기종, 구루병, 만성 하부 척추통증, 말초 신경 손상, 중추 신경 손상 및 화학적 손상과 같은 질환 등의 만성 질환; 골절, 외상 등과 같은 원인 혹은 장기간의 침상 요양에 의한 운동 손실; 노화 등 다양한 원인에 의해 기인할 수 있다. 즉, 상기 근육 질환은 노화, 근 기능 저하, 근육 소모, 근육 퇴화, 불용(disused) 또는 근육 손상으로 인하여 유발되는 것.
이에 제한되는 것은 아니나, 상기 근육 질환은 긴장감퇴증(atony), 근위축증(muscular atrophy), 근이영양증(muscular dystrophy), 근육 퇴화, 근경직증, 근위축성 축삭경화증, 근무력증, 악액질 (cachexia) 및 근육 감소증(sarcopenia)으로 이루어진 군에서 선택되는 하나 이상의 근육질환일 수 있고, 보다 구체적으로는, 노화 또는 암과 같은 질환에 의한 근위축증(muscular atrophy), 근이영양증(muscular dystrophy), 근육 퇴화, 근경직증, 근위축성 축삭경화증, 근무력증, 악액질(cachexia), 근육감소증(sarcopenia) 및 근육소실증을 포함할 수 있다.
본 명세서에서 용어 "예방"은 본 발명에 따른 상기 약학적 조성물의 투여에 의해 근육 질환을 억제시키거나 발병을 지연시킬 수 있는 모든 행위를 의미한다.
본 명세서에서 용어 "치료"는 본 발명에 따른 상기 약학적 조성물의 투여에 의해 증세가 호전되거나 이롭게 되는 모든 행위를 말한다.
본 발명의 상기 약학적 조성물은 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있으며, 상기 제형화에 필요한 담체 혹은 부형제 등을 추가로 포함할 수 있다. 상기 유효 성분에 추가로 포함될 수 있는 약학적으로 허용 가능한 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 마그네슘 스테아레이트 및 광물유 등이 포함된다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
예를 들어, 경구 투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 추출물 또는 화합물에 적어도 하나 이상의 부형제 적어도 면, 전분, 칼슘카보네이트 (calcium carbonate), 수크로스 (sucrose) 또는 락토오스 (lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 경구 투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜 (propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔 (witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 또는 비경구 투여(정맥주사, 피하, 복강 내 또는 국소에 적용)될 수 있으며, 투여량은 환자의 상태 및 체중, 질병의 정도 및 약물 형태, 투여 경로 및 시간에 따라 다르며, 당업자에 의해 적절한 형태로 선택될 수 있다.
본 발명의 상기 약학적 조성물은, 약학적으로 유효한 양으로 투여한다. 본 발명에서 "약학적으로 유효한 양"이란 의학적 치료에 적용 가능한 합리적인 양으로, 질병을 치료하기에 충분한 양을 의미하며, 그 기준은 환자의 질환, 중증도, 약물 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 병용되는 성분 및 기타 사항에 따라 결정될 수 있다. 본 발명의 상기 약학적 조성물은, 개별 치료제 혹은 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 상기 요소들을 모두 고려하여 부작용을 최소화할 수 있는 수준으로 투여량을 결정할 수 있고, 이는 당업자에 의해 용이한 수준으로 결정될 수 있다. 구체적으로 상기 약학적 조성물의 투여량은 환자의 연령, 체중, 중증도, 성별 등에 따라 달라질 수 있으며, 일반적으로 체중 1kg 당 0.001 내지 150 mg, 보다 바람직하게는 0.01 내지 100mg의 양을 매일 또는 격일, 1일 1 내지 3회 투여할 수 있다. 다만, 이는 예시적인 것으로, 상기 투여량은 필요에 달리 설정할 수 있다.
또한 본 발명의 상기 조성물은 식품 또는 건강기능식품일 수 있으며, 특히 상기 "건강기능식품"은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
본 발명의 상기 식품 또는 건강기능식품은, 근육 질환의 예방 및 개선을 위한 목적으로 산제, 과립제, 정제, 캡슐제, 환제, 현탁액, 에멀젼, 시럽 등 약학적 투여 형태 또는 티백, 침출자, 음료, 캔디, 젤리, 껌 등의 건강 기능식품으로 제조 및 가공이 가능하다.
본 발명의 상기 식품 또는 건강기능식품 조성물은, 식품 첨가물로 사용될 수 있으며, 단독으로 혹은 다른 성분과 조합하여 제품화 될 수 있다. 또한, 영양제, 비타민, 전해질, 풍미제, 착색제 및 증진제, 펙트산 및 이의 염, 알긴산 및 이의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등이 포함될 수 있다. 상기 성분은 단독 혹은 조합하여 사용될 수 있으며, 적절한 함량으로 조합되어 사용할 수 있다.
본 발명의 다른 실시 양태로서, 본 발명은 알도오즈 환원효소 억제제 화합물을 포함하는 사료 또는 사료 첨가용 조성물에 대한 것이다.
본 발명에서, '사료'란, 동물의 생명을 유지하는데 필요한 유기 또는 무기 영양소를 공급하는 물질을 의미한다. 상기 사료는 가축 등의 동물이 필요로 하는 에너지, 단백질, 지질, 비타민, 광물질 등의 영양소를 포함하며, 곡물류, 근과류, 식품가공부산물류, 조류, 섬유질류, 유지류, 전분류, 박류, 곡물부산물류 등의 식물성 사료 또는 단백질류, 무기물류, 유지류, 광물성류, 유지류, 단세포 단백질 등의 동물성 사료가 될 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서, '사료 첨가제'란, 동물의 생산성 향상이나 건강을 증진시키기 위해 사료에 첨가되는 물질을 의미하며, 이에 제한되는 것은 아니나, 성장 촉진, 질병 예방 등을 위한 아미노산제, 비타민제, 효소제, 향미제, 규산염제, 완충제, 추출제, 올리고당 등이 더욱 포함될 수 있다. 본 발명의 상기 사료 또는 사료 첨가제 조성물은 동물의 근원 세포 분화 촉진, 근육 재생 또는 근력 강화 효과를 가질 수 있으며, 나아가 동물의 근육 관련 질환에 대하여 예방, 치료 또는 개선 효과를 가질 수 있다.
본 발명은 알도오즈 환원효소 억제제 화합물을 포함하는 근원세포 증식 및 분화 촉진용 조성물로서, 상기 조성물은 근원세포의 분화 촉진을 통하여 근육 재생을 증가시키며, 근육량 증가, 근력 강화, 근 기능 개선 및 운동 수행능력 증가로 인하여 다양한 근육 질환에 대한 예방 또는 치료 효과를 가질 수 있고, 일반인의 근육 기능 개선에도 효과적이므로, 약학적 조성물, 기능성 식품 및 첨가제 등 다양한 용도로 사용될 수 있다.
도 1은 알도오즈 환원효소 억제제의 농도 별 세포 독성을 분석하기 위하여 생쥐 근원세포인 C2C12 세포주를 대상으로 알도오즈 환원효소 억제제 중 4종(에팔레스타트, 톨레스타트, 피다레스타트, 소르비닐)을 각각 0, 1, 10, 100, 1,000, 10,000nM 농도로 처리한 다음 MTT 분석을 실행한 결과이다. 알도오즈 환원효소 억제제 종류별로, 각 왼편은 알도오즈 환원효소 억제제의 MTT 반응 결과물의 사진이며, 각 오른편은 농도별 세포 생존율을 나타낸 그래프이다.

도 13은 알도오즈 환원효소 억제제에 의한 근육 활성 조절을 분석하기 위하여 미토콘드리아 관련 유전자(ATP5a, Sdhb, MTCO1, MTCO2, Uqcc1)의 발현을 실시간 중합 효소 연쇄반응으로 분석한 결과이다.
이하, 본 명세서를 구체적으로 설명하기 위해 실시예를 들어 상세히 설명한다. 그러나, 본 명세서에 따른 실시예들은 여러 가지 다른 형태로 변형될 수 있으며, 본 명세서의 범위가 아래에서 상술하는 실시예들에 한정되는 것으로 해석되지는 않는다. 본 명세서의 실시예들은 당업계에서 평균적인 지식을 갖춘 자에게 본 명세서를 더욱 완전하게 설명하기 위해 제공되는 것이다.
실시예 1. 근원세포주 배양 및 분화 유도
C2Cl2는 C3H종의 생쥐에서 얻은 근원세포주로서, 근육세포 분화 연구에 널리 사용되고 있다. 17세(남성, 백인) 또는 66세(남성, 백인) 인체-유래 근원세포주는 Cook MyoSite (Cat# SK-1111, 표준 공여 근육세포)로부터 구매하여 사용하였다. 상기 세포는 세포 배양용 배지(GM, Growth media)에서 배양하고, 분화용 배지(DM, Differentiation media)에서 근육세포 분화를 유도하였다. 세포 배양용 배지는 15% 소태아혈청(FBS, Fetal bovine serum)이 첨가된 DMEM을 사용하였으며, 분화용 배지는 2% 말혈청(Horse serum)이 포함된 DMEM을 사용하였다.
근육세포 분화를 유도하기 위해, 세포배양용 배지에 세포를 분주하여 배양한 후 세포 밀도가 약 80-90%에 도달하였을 때, 배지를 분화용 배지로 교체하고 약 2-3일간 분화를 유도하였다.
실험예 1. 알도오즈 환원효소 억제제-유발 독성에 대한 세포 생존 평가
C2C12 근원세포를 대상으로 알도오즈 환원효소 억제제 중 4종(에팔레스타트, 톨레스타트, 피다레스타트, 소르비닐)을 각각 0, 1, 10, 100, 1,000, 10,000nM 농도로 18시간 동안 처리한 다음 세포 생존력(Cell viability)을 MTT 분석을 실행하여 분석 하였다. 세포 배양용 배지 상에서 근원세포(3.5X105cells/96well)를 0.2mg/ml의 MTT와 6시간 동안 반응시키고, 이후 용출된 반응물을 570nm 흡광도에 측정하였다. 그 결과, MTT 분석에 이용된 4종의 알도오즈 환원효소 억제제 모두가 ~1,000nM 농도 처리까지는 대조군과 거의 유사한 세포 생존력을 나타냈으나, 톨레스타트의 경우 10,000nM 처리 시 약간의 세포독성이 확인되었다 (도 1).
실험예 2. 알도오즈 환원효소 억제제에 의한 세포 증식 촉진 효과 분석
알도오즈 환원효소 억제제가 생쥐 근원세포의 증식에 끼치는 영향을 분석하기 위하여 BrdU 염색 실험을 실행하였다. C2C12 근원세포에 대조군인 DMSO 또는 4종의 알도오즈 환원효소 억제제(1μM)를 각각 약 20시간 동안 처리한 후, BrdU와 15분간 반응한 다음에 BrdU에 대한 항체를 이용하여 면역형광 염색을 수행하였다(도 2). 그 결과, 사용된 4종의 알도오즈 환원효소 억제제 모두, 생쥐 근원세포에 처리하였을 때 세포증식이 증가되었다. 이후, 알도오즈 환원효소 억제제가 인체-유래 근원세포의 증식에 끼치는 영향을 분석하기 위하여, 17세 청소년-유래 근원세포(도 3) 또는 66세 노인-유래 근원세포(도 4)를 대상으로 BrdU 염색 실험을 진행하였다. 17세 또는 66세 인체-유래 근원세포에 대조군인 DMSO 또는 4종의 알도오즈 환원효소 억제제(1μM)를 각각 2일 동안 처리한 후, BrdU와 30분간 반응한 다음에 BrdU에 대한 면역형광 염색을 수행하였다. 그 결과, 사용된 알도오즈 환원효소 억제제 중 소르비닐을 제외한 3종을 각각 처리하였을 때 인체-유래 근원세포의 증식이 증가되었다.
추가로, 인체-유래 근원세포의 증식에 알도오즈 환원효소 억제제가 끼치는 영향을 분석하기 위하여, 세포증식 마커인 Ki67의 발현 양 차이를 분석하였다(도5). 17세 또는 66세 인체-유래 근원세포에 대조군인 DMSO 또는 4종의 알도오즈 환원효소 억제제(1μM)를 각각 2일 동안 처리한 후, Ki67의 mRNA의 양 차이를 실시간 중합 효소 연쇄반응을 수행하여 분석하였다. 그 결과, 17세, 66세 인체-유래 근원세포 모두에서 소르비닐을 제외한 3종의 알도오즈 환원효소 억제제에 의한 세포증식의 증가가 확인되었다.
실험예 3. 근원세포의 근육세포 분화 촉진 효과 확인
알도오즈 환원효소 억제제가 근원세포의 근육세포 분화에 끼치는 영향을 분석하기 위하여 MHC 면역형광 염색을 수행하였다(도 6). C2C12 세포주에 대조군 DMSO 또는 4종의 알도오즈 환원효소 억제제를 각각 10, 100, 1,000nM로 처리한 후 실시예 1과 같이 근육세포 분화를 유도하고, 분화 유도 3일째에 MHC 항체를 이용하여 면역형광 염색을 수행함으로써 근관 형성 정도를 비교 분석하였다. 그 결과, 대조군에 비하여 4종의 알도오즈 환원효소 억제제 모두가 근육세포 분화를 촉진하였으며, 촉진 정도는 처리 농도가 높을수록 증가하였다.
알도오즈 환원효소 억제제에 의한 근육세포 분화 촉진을 확인하기 위하여, MHC 단백질 발현 차이를 웨스턴 블럿팅을 수행하여 분석하였다(도 7). C2C12 근원세포에 DMSO 또는 100nM의 알도오즈 환원효소 억제제 4종을 각각 처리한 후, 근육세포 분화 유도 3일째에 단백질을 분리하고 MHC 발현 정도를 웨스턴 블럿팅으로 분석하였다. 그 결과, 소르비닐을 제외한 3종의 알도오즈 환원효소 억제제 처리를 통해서 MHC의 발현이 증가, 즉 근육세포 분화가 대조군에 비하여 촉진되었다.
근육세포 분화 유도과정에서 알도오즈 환원효소 억제제가 끼치는 영향을 분석하기 위하여, 근육세포 분화-관련 신호전달 인자인 AKT의 활성을 분석하였다(도 8). C2C12 근원세포에 DMSO 또는 4종의 알도오즈 환원효소 억제제를 각각 100nM 농도로 처리한 다음, 근육세포 분화를 유도하고 3일째에 단백질을 분리한 후 AKT와 인산화된 AKT(p-AKT)의 양 차이를 웨스턴 블럿팅을 수행하여 분석하였다. 그 결과, 에팔레스타트를 제외한 3종의 알도오즈 환원효소 억제제를 각각 처리하였을 때 인산화된 AKT 즉, 활성화된 AKT의 양이 대조군에 비하여 증가된 것을 확인하였다.
알도오즈 환원효소 억제제가 젊은 인체-유래 근원세포(도 9) 또는 노화된 인체-유래 근원세포(도 10)의 근육세포 분화를 촉진하는지의 여부를 분석하기 위하여 MHC 면역형광 염색을 실행하였다. 17세 또는 66세 인체-유래 근원세포에 DMSO 또는 1μM의 알도오즈 환원효소 억제제 4종을 각각 처리한 후 근육세포 분화를 4일간 유도하고 근유세포 분화 정도, 즉 근관 형성 정도를 MHC 면역형광 염색을 수행하여 분석하였다. 그 결과, 인체-유래 근원세포에 4종의 알도오즈 환원효소 억제제를 처리하였을 때, 대조군에 비하여 근육세포 분화가 증가하였다.
실험예 4. 근원세포에서 근위축증 유발 및 알도오즈 환원효소 억제제에 의한 근위축증 방지, 완화 효과 확인
알도오즈 환원효소 억제제에 의한 근위축증 억제 효과를 분석하기 위하여 덱사메타손을 처리하고 MHC 면역형광 염색을 실행하였다(도 11). C2C12 근원세포를 이용하여 실시예 1과 같이 2일간 근육세포 분화를 유도한 후, DMSO 또는 1μM의 알도오즈 환원효소 억제제 4종을 각각 4시간 동안 처리하고, 100μM의 덱사메타손을 처리하여 근위축 증상을 유발하였다. 20시간 동안 덱사메타손을 처리한 후, 알도오즈 환원효소 억제제에 의한 근위축증 방지 효과를 분석하기 위하여 MHC 면역형광 염색을 수행하였다. 그 결과, 덱사메타손을 처리하여 유도된 근위축증 정도가 에팔레스타트를 제외한 3종의 알도오즈 환원효소 억제제의 전처리를 통해서 억제되어, 덱사메타손을 처리한 대조군에 비하여 근유세포 분화가 증가되어 있음을 확인하였다.
추가로, 알도오즈 환원효소 억제제에 의한 근위축증 억제 효과를 분석하기 위하여, 덱사메타손을 처리하고 근위축 관련 마커들의 발현 차이를 실시간 중합 효소 연쇄반응으로 분석하였다(도 12). C2C12 근원세포를 이용하여 2일간 근육세포 분화를 유도한 후, DMSO 또는 1μM의 알도오즈 환원효소 억제제 4종을 각각 4시간 동안 처리하고, 100μM의 덱사메타손을 처리하여 근위축 증상을 유발하였다. 20시간 동안 덱사메타손을 처리한 후, 근위축 마커인 Atrogin-1과 MuRF-1 mRNA의 발현을 실시간 중합 효소 연쇄반응으로 분석하였다. 그 결과, 덱사메타손 처리로 증가되는 근위축 마커들의 발현이 알도오즈 환원효소 억제제의 전처리로 인해 그 발현 정도가 감소된 것을 확인하였다.
실험예 5. 근원세포의 활성 촉진 효과 확인
알도오즈 환원효소 억제제에 의한 근육 활성 조절을 분석하기 위하여 미토콘드리아 관련 유전자의 발현을 실시간 중합 효소 연쇄반응으로 분석 하였다(도 13). C2C12 근원세포에 DMSO 또는 100nM의 알도오즈 환원효소 억제제 4종을 각각 처리한 후 3일간 근육세포 분화를 유도하였다. 이후, 미토콘드리아 관련 유전자인 ATP5a, Sdhb, MTCO1, MTCO2 및 Uqcc1의 mRNA 발현 정도를 실시간 중합 효소 연쇄반응을 수행하여 분석하였다. 그 결과, 알도오즈 환원효소 억제제 처리 시 전반적으로 근육 활성 조절에 중요한 미토콘드리아의 활성이 증가되는 것을 확인하였다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
본 발명은 알도오즈 환원효소 억제제 화합물을 포함하는 근원 세포의 증식 및 분화 촉진용 조성물에 대한 것이다.
본 발명의 다른 양태로서, 본 발명은 상기 알도오즈 환원효소 억제제 화합물을 포함하는 근육 재생용 조성물에 대한 것이다.
본 발명의 다른 양태로서, 본 발명은 상기 알도오즈 환원효소 억제제 화합물을 포함하는 근육량 증진 또는 근력 강화용 조성물에 대한 것이다.
본 발명의 또 다른 양태로서, 본 발명은 상기 알도오즈 환원효소 억제제 화합물을 포함하는 운동 수행능력 향상용 조성물에 대한 것이다.
본 발명의 또 다른 양태로서, 본 발명은 상기 알도오즈 환원효소 억제제 화합물을 포함하는 근 기능 개선용 조성물에 대한 것이다.
상기 본 발명의 알도오즈 환원효소 억제제는 피다레스타트(fidarestat), 톨레스타트(tolrestat), 에팔레스타트(epalrestat), 라니레스타트(ranirestat), 포놀레스타트(ponolrestat), 조폴레스타트(zopolrestat), 제나레스타트(zenarestat), 소르비닐(sorbinil), 메토소르비닐(methosorbinil), 이미레스타트(imirestat) 및 미날레스타트(minalrestat)로 이루어진 군으로부터 하나 이상 선택될 수 있다.
또한 본 발명의 상기 조성물은 식품, 기능성 식품, 건강기능 식품, 의약품, 동물용 사료 또는 사료 첨가제 중 하나 이상 선택될 수 있다.
본 발명의 또 다른 양태로서, 본 발명은 상기 알도오즈 환원효소 억제제 화합물 및 이의 약학적으로 허용 가능한 염을 포함하는 근육 질환의 예방 또는 치료용 약학적 조성물에 대한 것이다.
본 발명의 또 다른 양태로서, 본 발명은 상기 알도오즈 환원효소 억제제 화합물 또는 이의 식품학적으로 허용 가능한 염을 포함하는 근육 질환의 예방 또는 개선용 건강기능 식품 조성물에 대한 것이다.
본 발명의 또 다른 양태로서, 본 발명은, 상기 알도오즈 환원효소 억제제 화합물 또는 이의 염을 포함하는 근육 질환의 예방 또는 개선용 동물용 사료 조성물에 대한 것이다.
본 발명에서 상기 근육 질환은 긴장감퇴증(atony), 근위축증(muscular atrophy), 근이영양증(muscular dystrophy), 근육 퇴화, 근경직증, 근위축성 축삭경화증, 근무력증, 악액질 (cachexia), 근육소실증 및 근육 감소증(sarcopenia)을 포함하는 군으로부터 선택될 수 있다.
또한 본 발명에서 상기 근육 질환은 노화, 근 기능 저하, 근육 소모, 근육 퇴화, 불용(disused) 또는 근육 손상으로 인하여 유발되는 것일 수 있다.
본 발명의 다른 양태로서, 본 발명은 상기 알도오즈 환원 효소 억제제 화합물을 이용하여, 근원 세포의 증식 및 분화를 촉진시키는 방법에 대한 것이다.
본 발명의 또 다른 양태로서, 본 발명은 상기 알도오즈 환원 효소 억제제 화합물을 이용하여 근육 재생, 근육량 증가, 근력 강화 또는 운동능력을 증가시키는 방법에 대한 것이다.
또한 본 발명의 또 다른 양태로서 본 발명은, 상기 알도오즈 환원효소 억제제 화합물을 이용하여, 긴장감퇴증(atony), 근위축증(muscular atrophy), 근이영양증(muscular dystrophy), 근육 퇴화, 근경직증, 근위축성 축삭경화증, 근무력증, 악액질 (cachexia), 근육소실증 및 근육 감소증(sarcopenia)과 같은 근육 질환의 예방, 개선 또는 치료하는 방법에 대한 것이다.

- 알도오즈 환원효소 억제제 화합물을 포함하는 근육 재생용 조성물.
- 알도오즈 환원효소 억제제 화합물을 포함하는 근육량 증진 또는 근력 강화용 조성물.
- 알도오즈 환원효소 억제제 화합물을 포함하는 운동 수행능력 향상용 조성물.
- 알도오즈 환원효소 억제제 화합물을 포함하는 근 기능 개선용 조성물.
