Description
本発明は、抗ノロウイルス効果に優れた組成物、その調製のためのテアフラビン類化合物の使用、及びテアフラビン類化合物を用いたノロウイルス感染防御方法に関する。
カリシウイルス科ノロウイルス属に分類されているノロウイルスは、手指、食品、調理器具などを介した経口感染によって人体内に入り、腸管細胞で増殖して、腹痛、下痢、嘔吐、発熱などの症状を引き起こす。また、患者自身の吐物・排泄物による二次感染力が非常に強いため、近年大規模な集団感染が相次いでいる。ノロウイルスに対するワクチンや治療薬はなく、次亜塩素酸ナトリウムや加熱による処理が推奨されてきたが、これらの薬剤を人体や調理器具類、ましてや食品に直接作用させることは適切とはいえない。
このような課題に対して、下記特許文献1には、タンニンを含有するカキノキ属(Diospyros)の植物の抽出物を有効成分とする抗ノロウイルス剤が開示されている。
しかしながら、テアフラビン類化合物に抗ノロウイルス効果があることは報告されていない。
本発明の目的は、上記従来技術に鑑み、抗ノロウイルス効果に優れた組成物、その調製のためのテアフラビン類化合物の使用、及びテアフラビン類化合物を用いたノロウイルス感染防御方法を提供することにある。
本発明者らは、テアフラビン類化合物が、ノロウイルスを不活化する効果に優れていることを見出し、本発明を完成するに至った。
本発明は、第1に、テアフラビン類化合物を、ノロウイルスの感染を防御するための有効成分として含有することを特徴とする抗ノロウイルス組成物を提供するものである。
本発明の抗ノロウイルス組成物においては、更にアルコールを含有することが好ましい。
本発明の抗ノロウイルス組成物においては、前記テアフラビン類化合物は、茶成分に由来するテアフラビン、テアフラビン-3-O-ガレート、テアフラビン-3’-O-ガレート、及びテアフラビン-3,3’-O-ジガレートからなる群から選ばれた1種又は2種以上であることが好ましい。
本発明は、第2に、テアフラビン類化合物をノロウイルス感染防御のための有効成分として含有する抗ノロウイルス組成物の調製のための該テアフラビン類化合物の使用を提供するものである。
本発明のテアフラビン類化合物の使用においては、前記抗ノロウイルス組成物は、更にアルコールを含有することが好ましい。
本発明のテアフラビン類化合物の使用においては、前記テアフラビン類化合物は、茶成分に由来するテアフラビン、テアフラビン-3-O-ガレート、テアフラビン-3’-O-ガレート、及びテアフラビン-3,3’-O-ジガレートからなる群から選ばれた1種又は2種以上であることが好ましい。

本発明のノロウイルス感染防御方法においては、前記テアフラビン類化合物を、更にアルコールとともに前記対象物に施与又は投与することが好ましい。
本発明のノロウイルス感染防御方法においては、前記テアフラビン類化合物は、茶成分に由来するテアフラビン、テアフラビン-3-O-ガレート、テアフラビン-3’-O-ガレート、及びテアフラビン-3,3’-O-ジガレートからなる群から選ばれた1種又は2種以上であることが好ましい。
本発明によれば、抗ノロウイルス効果に優れた組成物、その調製のためのテアフラビン類化合物の使用、及びテアフラビン類化合物を用いたノロウイルス感染防御方法を提供することができる。

紅茶発酵の過程を経た茶葉(強発酵茶)を、水、含水アルコール等の溶媒により、加熱もしくは常温条件で、数分から数時間撹拌することにより抽出し、紅茶抽出物を得ることができる。この紅茶抽出物は、精製、濃縮、粉末化などの加工度が進んだものであってもよい。このような紅茶抽出物において、テアフラビン類化合物の含量を高めるためには、例えば、紅茶発酵の過程で発酵時間を延長する方法の他、エピカテキン、エピガロカテキン、エピカテキン-3-ガレート、エピガロカテキン-3-ガレートなどのカテキン類を紅茶抽出物に追加で投入したり、ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素を添加したり、あるいはその両方を行なったりして、加熱もしくは常温条件で、数分から数時間反応させる方法が挙げられる。ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素としては、リンゴ、バナナなど果実に由来するものや、該酵素を含む茶葉抽出物、該酵素を含む茶葉粉砕物、該酵素を含む植物細胞培養液などを利用することができる。また、上記カテキン類としては、茶由来カテキン製剤やカテキンを高濃度に含む茶抽出物など茶由来のものの他、カカオ等のその他の植物由来のカテキン製剤やカテキンを高濃度に含む抽出物を利用してもよい。

水等の溶媒に溶解し又は溶解された茶抽出成分に、ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素を添加して、加熱もしくは常温条件で、数分から数時間反応させる。これにより、茶抽出成分に含まれるカテキン類からテアフラビン類化合物が生成する。ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素としては、リンゴ、バナナなど果実に由来するものや、該酵素を含む茶葉抽出物、該酵素を含む茶葉粉砕物、該酵素を含む植物細胞培養液などを利用することができる。このようにして得られた茶抽出成分の酵素処理物は、精製、濃縮、粉末化などの加工度が進んだものであってもよい。

(茶葉発酵抽出物)
茶葉を粉砕してスラリー状に調製し、必要に応じて水等の溶媒を添加して、加熱もしくは常温条件で、数分から数時間発酵させる。これにより、茶葉に含まれるカテキン類から、茶葉に含まれるポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素の作用により、テアフラビン類化合物が生成する。発酵後は、必要に応じて、固液分離したり、更に水、含水アルコール等の溶媒により、加熱もしくは常温条件で、数分から数時間撹拌することにより抽出したりしてもよい。このようにして得られた茶葉発酵抽出物は、精製、濃縮、粉末化などの加工度が進んだものであってもよい。

(カテキン類の酵素処理物)
エピカテキン、エピガロカテキン、エピカテキン-3-ガレート、エピガロカテキン-3-ガレートなどのカテキン類を原料にして、水等の溶媒中で、ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素を添加して、加熱もしくは常温条件で、数分から数時間反応させる。これにより、エピカテキン、エピガロカテキン、エピカテキン-3-ガレート、エピガロカテキン-3-ガレートなどのカテキン類からテアフラビン類化合物が生成する。ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素としては、リンゴ、バナナなど果実に由来するものや、該酵素を含む茶葉抽出物、該酵素を含む茶葉粉砕物、該酵素を含む植物細胞培養液などを利用することができる。また、上記カテキン類としては、茶由来カテキン製剤やカテキンを高濃度に含む茶抽出物など茶由来のものの他、カカオ等のその他の植物由来のカテキン製剤やカテキンを高濃度に含む抽出物を利用してもよい。このようにして得られたカテキン類の酵素処理物は、精製、濃縮、粉末化などの加工度が進んだものであってもよい。
エピカテキン、エピガロカテキン、エピカテキン-3-ガレート、エピガロカテキン-3-ガレートなどのカテキン類を原料にして、水等の溶媒中で、ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素を添加して、加熱もしくは常温条件で、数分から数時間反応させる。これにより、エピカテキン、エピガロカテキン、エピカテキン-3-ガレート、エピガロカテキン-3-ガレートなどのカテキン類からテアフラビン類化合物が生成する。ポリフェノールオキシダーゼ活性及び/又はペルオキシダーゼ活性を有する酵素としては、リンゴ、バナナなど果実に由来するものや、該酵素を含む茶葉抽出物、該酵素を含む茶葉粉砕物、該酵素を含む植物細胞培養液などを利用することができる。また、上記カテキン類としては、茶由来カテキン製剤やカテキンを高濃度に含む茶抽出物など茶由来のものの他、カカオ等のその他の植物由来のカテキン製剤やカテキンを高濃度に含む抽出物を利用してもよい。このようにして得られたカテキン類の酵素処理物は、精製、濃縮、粉末化などの加工度が進んだものであってもよい。
なお、本明細書において「茶抽出物」とは、上記に例示の調製法以外でも、茶を原料にしてその成分として、あるいはその成分であるカテキン類から生成させた、茶由来のテアフラビン類化合物を含むものであればよく、その全般を包含する意味である。
上記茶抽出物の原料となる茶としては、ツバキ科の多年性植物である茶の樹から得られるものであればよく、特に制限はない。一般に栽培されている茶品種としては、例えば、あさつゆ、おおいわせ、おくひかり、おくみどり、かなやみどり、こまかげ、さみどり、はつもみじ、やまとみどり、まきのはらわせ、みねかおり、めいりょく、やぶきた、やまなみ等の緑茶品種、烏龍、色種、水仙、鉄観音等のウーロン茶品種、からべに、ひめみどり、べにひかり、べにふうき、べにふじ、べにほまれ等の紅茶品種などが挙げられる。茶葉の採取時期は、1番茶、2番茶、3番茶などのいずれでもよく、また、その栽培国・地域に特に制限はない。
本発明の抗ノロウイルス組成物は、その有効成分であるテアフラビン類化合物をノロウイルスに接触させるようにして用いて、ノロウイルスの感染能力を破壊、減退、もしくは抑制する等して不活化し、ノロウイルスの感染を防ぐことができる、というものである。
本発明の抗ノロウイルス組成物は、テアフラビン類化合物を、固形分中10~100質量%含有することが好ましく、25~100質量%含有することがより好ましく、40~100質量%含有することが最も好ましい。テアフラビン類化合物の含有量が、固形分中に10質量%未満では、ノロウイルスに接触する際のテアフラビン類化合物の濃度が低くなり、ノロウイルスの感染を防ぐ効果が低減する傾向となる場合がある。なお、テアフラビン類化合物として茶抽出物を用いる場合、該茶抽出物は、その固形分中に、テアフラビン類化合物を、10~100質量%含有するものであることが好ましく、25~100質量%含有するものであることがより好ましく、40~100質量%含有するものであることが最も好ましい。
本発明の抗ノロウイルス組成物は、テアフラビン類化合物と共に更にアルコールを含有することが好ましい。これによれば、テアフラビン類化合物による抗ノロウイルス効果を、アルコールにより、更に高めることができる。アルコールとしては、エタノールやイソプロピルアルコール等が挙げられる。アルコールは、合成アルコールを使用してもよく、発酵アルコールを使用してもよい。アルコールの含有量としては、抗ノロウイルス組成物中に10~95v/v%であることが好ましく、50~95v/v%であることがより好ましい。
本発明の抗ノロウイルス組成物は、テアフラビン類化合物やアルコールの他に、他の原料や素材を含んでいてもよい。例えば、必要に応じて、製剤的に有利な素材として、界面活性剤、増粘剤、乳化剤、着色剤、着香剤、防腐剤などを含むことができる。また、必要に応じて、栄養的に有利な原料として、砂糖、果糖、糖アルコールなどの糖類や、タンパク質、脂質、ミネラル分などを含むことができる。更にまた、必要に応じて、他の抗ウイルス剤や抗菌剤、制菌剤など、独自の機能性を発揮し得る素材を含むことができる。
本発明の抗ノロウイルス組成物の有効成分であるテアフラビン類化合物をノロウイルスに接触させるには、例えば、本発明の抗ノロウイルス組成物もしくはそれに含有されるテアフラビン類化合物を、感染防御の対象物に施与又は投与すること等により行うことができる。ここで「施与」とは、生体もしくは非生体等である対象物に噴霧したり、散布したり、塗布したり、添加したり、混合したり、固着したりすること等を意味する。また、「投与」とは、ヒト又は動物等である対象物に、経口剤等の形態で経口摂取せしめたり、注射剤等の形態で注射したり、クリームやローション等の形態で表皮に局所的に塗布したり、入浴剤等の形態で体全体に作用させたり、マウスウオッシュ、ハンドソープ、シャンプー等の形態で体の一部、例えば、口腔内や手足、髪の毛等に作用させたりすること等を意味する。
本発明が適用される感染防御の対象物に特に制限はない。例えば、既にノロウイルスが存在する対象物であってもよく、ノロウイルスが存在するであろうと目される対象物であってもよく、感染の危険性がある対象物であってもよく、感染を予防したい対象物であってもよい。施与又は投与する対象物に既にノロウイルスが存在している場合、それに施与又は投与することによってテアフラビン類化合物をノロウイルスに接触させることができることは勿論、未だノロウイルスが存在していない場合であっても、施与又は投与した対象物にはテアフラビン類化合物が有効に残存するので、その後に対象物に付着したり、付着しようとしたりするノロウイルスに対して、その感染能力を破壊、減退、もしくは抑制する等して不活化し、ノロウイルスの感染を防ぐことができる。具体的には、例えば、調理の場面において、包丁、まな板、鍋、レンジ、オーブンなどの調理器具、調理台、流し台、食器、ふきん、タオル、調理作業者の着衣、調理作業者の手指、顔、髪の毛などが挙げられる。この場合、調理前、調理中、又は調理後の食品材料や加工された食品、生鮮食品等に施与したりすることなども挙げられる。更には、例えば、医療の場面において、病院のトイレ、床、ベッド、医療作業者の着衣、医療作業者の手指、顔、髪の毛などが挙げられる。あるいは更には、例えば、公共交通、公共広場、学校、病院を訪れた者の吐瀉物、排泄物などが挙げられる。更にはあるいは、例えば、ノロウイルスに対する感染予防の目的でヒト又は動物に経口的に投与したり、ヒトが着用するマスクに固着させたりすることなども挙げられる。更にはあるいは、例えば、ガム、キャンディー、予防用飲料、ゼリー飲料等の飲食品に含有せしめることなどが挙げられる。ただし、これらに限られるものではない。
施与又は投与の方法・用量等の態様は、特に限定されないが、典型的な例を挙げれば、感染防御の対象物に表面積を観念できる場合、その表面積1cm2当たりにテアフラビン類化合物を0.0001~1.0mg程度の量で施与又は投与することが好ましく、0.001~1.0mg施与又は投与することがより好ましく、0.01~1.0mg程度の量で施与又は投与することが最も好ましい。対象物の表面積1cm2当たりにテアフラビン類化合物を上記範囲未満の量で施与又は投与したのでは、ノロウイルスに接触する際のテアフラビン類化合物の濃度が低くなり、ノロウイルスの感染を防ぐ効果が低減する傾向となる場合がある。
また、テアフラビン類化合物は、更にアルコールとともに対象物に施与又は投与してもよい。これによれば、上述したように、アルコールにより、テアフラビン類化合物による抗ノロウイルス効果を更に高めることができる。用いるアルコールの濃度としては、10~90v/v%であることが好ましく、25~90v/v%であることがより好ましい。
本発明の抗ノロウイルス組成物は、適当な基材や担体等を使用して、公知の方法によって、種々の形態をとり得る。例えば、錠剤、顆粒剤、粉剤、カプセル、ゼリー、ドリンク等の形状のサプリメント、予防用飲料、消毒液、うがい液、マウスウオッシュ、歯磨き剤、口腔用スプレー、トローチ錠、口腔内溶解錠剤、ガム、キャンディー、ゼリー飲料、グミなどが挙げられる。あるいは、食品添加物や医薬品などの利用形態や、疾病に罹患していない者を対象とした、いわゆる機能性表示食品(機能性関与成分によって健康の維持及び増進に資する特定の保健の目的が期待できる旨を科学的根拠に基づいて容器包装に表示した食品)やその成分としての利用形態も排除されるものではない。また、本発明の抗ノロウイルス組成物は、それ自体が有効成分であってもよく、あるいは他の有効成分と併用して、ノロウイルスの感染防御の効果を得るために使用されてもよい。
より具体的には、以下のような製品が挙げられる。
・消毒液:抗菌・抗ウイルス目的で、アルコールもしくは殺菌剤を含み、手または全身の皮膚に噴霧、もしくはジェル状又は液状で手の表面に塗布するもの。主に屋外から屋内に入った際に利用する製品。
・うがい液:抗菌・抗ウイルス目的で、少量のアルコールもしくは殺菌剤を含み、うがいもしくは口腔の濯ぎをすることで、口腔内の菌やウイルスの増殖を防ぐ為に用いられる製品。主に屋外から屋内に入った際に利用する。
・マウスウオッシュ:上記のうがい液と同様、口濯ぎをすることで、口腔内の菌・ウイルスの増殖を防ぐ為に用いられる製品。
・口腔用スプレー:上記のうがい液と同様、口腔内の奥側に噴霧することで、口腔内の菌・ウイルスの増殖を防ぐ為に用いられる製品。
・ハンドソープ:菌・抗ウイルス目的で殺菌剤や抗菌剤を含み、手の表面の洗浄と同時に、手に付着した菌及びウイルスの増殖を防ぐ為に用いられる製品。主に屋外から屋内に入った際に利用する。
・感染予防食品(ガム、キャンディー):口腔内に比較的長時間滞在させることで、食品に含まれる抗菌成分により、菌・ウイルスの感染を防ぐ製品。
・感染予防飲料・ゼリー食品:口腔内に比較的長時間滞在させ、食品に含まれる抗菌成分により、菌・ウイルスの感染を防ぐ製品。
・不織布・マスク:抗菌・抗ウイルス目的で、機能を付与したマスク。抗菌素材を不織布に添着もしくは結合させ、それを用いてマスクに加工した製品。マスクを着用することで、菌・ウイルスの感染を防ぐ。
以下に実施例を挙げて本発明について更に具体的に説明するが、これらの実施例は本発明の範囲を限定するものではない。
<製造例1> テアフラビン含有粗抽出物 その1
緑茶由来カテキン製剤(カテキン含量90%以上、商品名「サンフェノン90」、太陽化学株式会社製)0.3gを水400mLに溶解した。別途、茶葉3gに水100mLを加えて、粉砕し、茶粉砕物を得た。この茶粉砕物に上記のカテキン溶液を合わせ、30~35℃で3時間反応させた。反応後の反応液を合成吸着剤「セパビーズSP-700」(商品名、三菱化学株式会社製)に通して精製を行い、フリーズドライにより粉末化して、テアフラビン含有粗抽出物0.1gを得た。HPLCで分析したところ、このテアフラビン含有粗抽出物中にはテアフラビンが6質量%、テアフラビン-3-O-ガレートが14質量%、テアフラビン-3'-O-ガレートが4質量%、テアフラビン-3,3'-O-ジガレートが18質量%含まれており、テアフラビン類化合物の合計含量は42質量%であった。
緑茶由来カテキン製剤(カテキン含量90%以上、商品名「サンフェノン90」、太陽化学株式会社製)0.3gを水400mLに溶解した。別途、茶葉3gに水100mLを加えて、粉砕し、茶粉砕物を得た。この茶粉砕物に上記のカテキン溶液を合わせ、30~35℃で3時間反応させた。反応後の反応液を合成吸着剤「セパビーズSP-700」(商品名、三菱化学株式会社製)に通して精製を行い、フリーズドライにより粉末化して、テアフラビン含有粗抽出物0.1gを得た。HPLCで分析したところ、このテアフラビン含有粗抽出物中にはテアフラビンが6質量%、テアフラビン-3-O-ガレートが14質量%、テアフラビン-3'-O-ガレートが4質量%、テアフラビン-3,3'-O-ジガレートが18質量%含まれており、テアフラビン類化合物の合計含量は42質量%であった。
<製造例2> テアフラビン含有精製抽出物
製造例1と同様の調製により得たテアフラビン含有粗抽出物の4gを、20%アセトンに溶解し、同じく20%アセトンで膨潤したゲル濾過カラム「SephadexLH-20」(商品名、GEヘルスケア・ジャパン株式会社製)に供して分画し、テアフラビン含有抽出液を得た。このテアフラビン含有抽出液をフリーズドライにより粉末化して、テアフラビン含有精製抽出物1.5gを得た。HPLCで分析したところ、このテアフラビン含有精製抽出物中にはテアフラビンが16質量%、テアフラビン-3-O-ガレートが27質量%、テアフラビン-3'-O-ガレートが7質量%、テアフラビン-3,3'-O-ジガレートが40質量%含まれており、テアフラビン類化合物の合計含量は90質量%であった。
製造例1と同様の調製により得たテアフラビン含有粗抽出物の4gを、20%アセトンに溶解し、同じく20%アセトンで膨潤したゲル濾過カラム「SephadexLH-20」(商品名、GEヘルスケア・ジャパン株式会社製)に供して分画し、テアフラビン含有抽出液を得た。このテアフラビン含有抽出液をフリーズドライにより粉末化して、テアフラビン含有精製抽出物1.5gを得た。HPLCで分析したところ、このテアフラビン含有精製抽出物中にはテアフラビンが16質量%、テアフラビン-3-O-ガレートが27質量%、テアフラビン-3'-O-ガレートが7質量%、テアフラビン-3,3'-O-ジガレートが40質量%含まれており、テアフラビン類化合物の合計含量は90質量%であった。
<製造例3> テアフラビン含有粗抽出物 その2
緑茶用茶葉50gに熱水400mLを添加し、撹拌した後、固液分離して緑茶抽出液を得た。別途、緑茶用茶葉3gに水100mLを加えて、粉砕し、緑茶粉砕物を得た。この緑茶粉砕物に上記の緑茶抽出液を合わせ、30~35℃で3時間反応させた。反応後の反応液を合成吸着剤「セパビーズSP-700」(商品名、三菱化学株式会社製)に通して精製を行い、フリーズドライにより粉末化して、テアフラビン含有粗抽出物0.1gを得た。HPLCで分析したところ、このテアフラビン含有粗抽出物中にはテアフラビンが4質量%、テアフラビン-3-O-ガレートが10質量%、テアフラビン-3'-O-ガレートが3質量%、テアフラビン-3,3'-O-ジガレートが14質量%含まれており、テアフラビン類化合物の合計含量は31質量%であった。
緑茶用茶葉50gに熱水400mLを添加し、撹拌した後、固液分離して緑茶抽出液を得た。別途、緑茶用茶葉3gに水100mLを加えて、粉砕し、緑茶粉砕物を得た。この緑茶粉砕物に上記の緑茶抽出液を合わせ、30~35℃で3時間反応させた。反応後の反応液を合成吸着剤「セパビーズSP-700」(商品名、三菱化学株式会社製)に通して精製を行い、フリーズドライにより粉末化して、テアフラビン含有粗抽出物0.1gを得た。HPLCで分析したところ、このテアフラビン含有粗抽出物中にはテアフラビンが4質量%、テアフラビン-3-O-ガレートが10質量%、テアフラビン-3'-O-ガレートが3質量%、テアフラビン-3,3'-O-ジガレートが14質量%含まれており、テアフラビン類化合物の合計含量は31質量%であった。
<試験例1> (マウスノロウイルスに対する抗ウイルス性)

製造例2で得たテアフラビン含有精製抽出物(以下、「テアフラビン精製品」という。)を、被検試料として使用した。また、比較のための被検試料として、非特異的な蛋白結合能を有し、ノロウイルスに対する有効性も報告されているタンニン酸や、既存のウイルス対策用製品である製品A及び製品Bを使用した。

テアフラビン精製品は、ジメチルスルホキシドに71.7mg/mLとなるように溶解し、以下の試験に使用した。

(5)抗ウイルス試験用細胞の調製
RAW264.7細胞を96穴プレートのウェルに単層培養し、PBS (Phosphate Buffered Saline)で2回洗浄し、200μLの細胞維持培地(2%ウシ胎児血清含DMEM(Dulbecco's Modified Eagle Medium)培地)を添加して、以下の試験に使用した。
RAW264.7細胞を96穴プレートのウェルに単層培養し、PBS (Phosphate Buffered Saline)で2回洗浄し、200μLの細胞維持培地(2%ウシ胎児血清含DMEM(Dulbecco's Modified Eagle Medium)培地)を添加して、以下の試験に使用した。

各被検試料について、濃度を以下のように調整し、試験液とした。

テアフラビン精製品については、上記(4)で調製した抗ウイルス試験用試料を、テアフラビン精製品濃度が1.5mg/mLとなるように25%エタノール添加血清不含DMEM培地で希釈し、試験液とした。
タンニン酸については、上記(4)で調製した抗ウイルス試験用試料を用い、タンニン酸濃度が1.5mg/mLとなるように25%エタノール添加血清不含DMEM培地で希釈し、試験液とした。
製品Aについては、原液をそのまま試験液とした。
製品Bについては、原液をそのまま試験液とした。
マウスノロウイルス液5μLと試験液45μLを混和し、60分間反応させた。
反応液に血清不含DMEM培地を450μL添加し、反応を停止した。
遠心式フィルター(分画分子量:100K)で遠心処理し(室温,20,000×g,1min→培地450μL添加→室温,20,000×g,1min)、ウイルスを回収した。
回収したウイルスを300μLの血清不含DMEM培地で希釈して、ウイルス試験液とし、そのウイルス試験液について血清不含DMEM培地を用いて5倍段階希釈列を調製した。
希釈したウイルス試験液を、上記(5)で準備した抗ウイルス試験用細胞の上清に50μLずつ添加し(各濃度につき3ウェル)、37℃、5%CO2存在下で3日間培養した。
細胞変性効果(Cytopathic effect: CPE)を観察し、ウイルス感染価の指標である50%組織培養感染量(50% Tissue culture infectious dose: TCID50)をBehrens-Karber法にて算出した。また、同様の手順で、被検試料で処理しないウイルス試験液を調製し、それを接種した細胞についても、細胞変性効果を観察して、50%組織培養感染量(logTCID50値)を求めた。下記式(1)により、被検試料によるウイルス感染価の減少値を算出した。

結果を表1及び図1に示す。

表1及び図1に示すように、テアフラビン精製品は、マウスノロウイルスの感染能力を不活化する効果が、タンニン酸や既存のウイルス対策用製品である製品A及び製品Bなどと比較して、格段に優れていることが明らかとなった。
<試験例2> (ネコカリシウイルスに対する抗ウイルス性 その1)
[試験方法]
(1)ウイルス
ノロウイルスの代替ウイルスとして、ネコカリシウイルスF9株を入手し、使用した。

表2及び図2に示すように、テアフラビン精製品により、ネコカリシウイルス(ノロウイルス代替ウイルス)の感染能力が不活化した。その効果は、テアフラビン精製品による処理時間に依存的であった。
<試験例3> (ネコカリシウイルスに対する抗ウイルス性 その2)
[試験方法]
試験例2において、その「(4)抗ウイルス試験用試料の調製」及び「(6)抗ウイルス試験」を下記のようにして行った以外は、試験例2と同様にして、被検試料のネコカリシウイルスに対する抗ウイルス性を調べた。

テアフラビン精製品は、ジメチルスルホキシドに0.717mg/mL(低濃度)、7.17mg/mL(中濃度)、及び71.7mg/mL(高濃度)となるように溶解し、以下の試験に使用した。

被検試料は、(A)上記(4)で調製した抗ウイルス試験用試料(低濃度)を、エタノール非添加の血清不含イーグルMEM培地又は25%エタノール添加血清不含イーグルMEM培地で、テアフラビン精製品濃度が0.018mg/mLとなるように希釈し、(B)上記(4)で調製した抗ウイルス試験用試料(中濃度)を、エタノール非添加の血清不含イーグルMEM培地又は25%エタノール添加血清不含イーグルMEM培地で、テアフラビン精製品濃度が0.18mg/mLとなるように希釈し、(C)上記(4)で調製した抗ウイルス試験用試料(高濃度)を、エタノール非添加の血清不含イーグルMEM培地又は25%エタノール添加血清不含イーグルMEM培地で、テアフラビン精製品濃度が1.8mg/mLとなるように希釈し、それぞれジメチルスルホキシド濃度が等しくなるようにして、試験液とした。
被検試料は、(A)上記(4)で調製した抗ウイルス試験用試料(低濃度)を、エタノール非添加の血清不含イーグルMEM培地又は25%エタノール添加血清不含イーグルMEM培地で、テアフラビン精製品濃度が0.018mg/mLとなるように希釈し、(B)上記(4)で調製した抗ウイルス試験用試料(中濃度)を、エタノール非添加の血清不含イーグルMEM培地又は25%エタノール添加血清不含イーグルMEM培地で、テアフラビン精製品濃度が0.18mg/mLとなるように希釈し、(C)上記(4)で調製した抗ウイルス試験用試料(高濃度)を、エタノール非添加の血清不含イーグルMEM培地又は25%エタノール添加血清不含イーグルMEM培地で、テアフラビン精製品濃度が1.8mg/mLとなるように希釈し、それぞれジメチルスルホキシド濃度が等しくなるようにして、試験液とした。

結果を表3及び図3に示す。

表3及び図3に示すように、テアフラビン精製品により、ネコカリシウイルス(ノロウイルス代替ウイルス)の感染能力が不活化した。その効果は用量依存的であった。特に、添加濃度が0.018mg/mlの場合、エタノールを併用しないときのウイルス感染価の減少値が0.2であるのに比べ、25%エタノールを併用したときのウイルス感染価の減少値が2を超えていた。一方で、25%エタノールのみで処理したときのウイルス感染価の減少値は0.5であり、テアフラビン精製品とエタノールとの併用により抗ノロウイルス効果が相乗的に向上することが明らかとなった。

下記表4の配合にて原料を撹拌混合し、抗ノロウイルス用消毒剤を得た。

単位面積当たり0.24g/m2となるように、不織布に製造例1で得たテアフラビン含有粗抽出物(テアフラビン類化合物の含有量:42質量%)を添着させ、テアフラビン添着不織布を得た。その不織布を用いてマスクを成形し、抗ノロウイルス用マスクを得た。

Claims (9)
- テアフラビン類化合物を、ノロウイルスの感染を防御するための有効成分として含有することを特徴とする抗ノロウイルス組成物。
- 更にアルコールを含有する請求項1記載の組成物。
- 前記テアフラビン類化合物は、茶成分に由来するテアフラビン、テアフラビン-3-O-ガレート、テアフラビン-3’-O-ガレート、及びテアフラビン-3,3’-O-ジガレートからなる群から選ばれた1種又は2種以上である請求項1又は2記載の組成物。
- テアフラビン類化合物をノロウイルス感染防御のための有効成分として含有する抗ノロウイルス組成物の調製のための該テアフラビン類化合物の使用。
- 前記抗ノロウイルス組成物は、更にアルコールを含有する請求項4記載の使用。
- 前記テアフラビン類化合物は、茶成分に由来するテアフラビン、テアフラビン-3-O-ガレート、テアフラビン-3’-O-ガレート、及びテアフラビン-3,3’-O-ジガレートからなる群から選ばれた1種又は2種以上である請求項4又は5記載の使用。
- テアフラビン類化合物を感染防御の対象物に施与又は投与することを特徴とするノロウイルス感染防御方法(但し医療行為を除く)。
- 前記テアフラビン類化合物を、更にアルコールとともに前記対象物に施与又は投与する請求項7記載の方法。
- 前記テアフラビン類化合物は、茶成分に由来するテアフラビン、テアフラビン-3-O-ガレート、テアフラビン-3’-O-ガレート、及びテアフラビン-3,3’-O-ジガレートからなる群から選ばれた1種又は2種以上である請求項7又は8記載の方法。
