본 출원은 2015년 9월 23일에 출원된 대한민국 특허출원 제 10-2015-0134540 호를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다. 모발 (hair)은 피부 진피 (skin dermis)의 모낭 (hair follicle)에서 자라는 단 백질 필라멘트이다. 모낭은 크게 모근 부분에 위치한 진피유두 (dermal papilla), 진피유두를 둘러싼 기질 (matrix), 그리고 근초 (root sheath)의 구조로 되어 있다. 진피유두는 주로 결합조직 (connective tissue)와 모세혈관으로 되어 있으며 모근을 둘러싸 영양분과 산소를 공급한다. 기질은 상피세포 (epithelial cell)와 멜라닌형 성세포 (melanocyte)로 되어 있으며, 세포분열이 활발하게 일어난다. 여기서 분열된 세포들이 각화 (keratinization)되면서 모발이 만들어지고 피부 밖으로 자라게 된 다. 근초는 내모근초 (inner root sheath)와 외모근초 (outer root sheath)가 있으 며, 성장하고 있는 모발이 완전히 각화될 때까지 보호하고 피부표피까지 도달하게 하는 역할을 한다. 모발은 성장기 /생장기 (anagen)-퇴화기 (catagen)-휴지기 (telogen)-탈락기 (자연탈모)를 거치면서 주기적으로 성장하고 빠지고 다시 자라나 는 과정을 반복하게 된다. 모낭에서 모발 성장기가 시작되면서 모발이 자라는데, 외모근초 팽윤부 (bulge)에 존재하는 즐기세포들이 활성화되어 초기 성장기 동안 새 로운 기질을 형성하고, 상대적으로 미분화된 기저부 세포로부터 모발줄기 (hair shaft)와 내모근초가 생성된다. 모발줄기의 크기와 길이는 각각 모낭의 크기와 모 발 성장기의 지속시간에 비례하게 된다. 탈모증은 정상적으로 털이 있어야 하는 부분의 털이 밀도가 비정상적으로 낮 거나 소실된 상태를 말하며, 크게 반흔성과 비반흔성으로 나뉜다. 반흔성 탈모증은 모낭이 파괴되고 섬유조직으로 회복되어 영구적 탈모 상태가 되는 것으로 외상, 화 상, 종양, 수술 등으로 인한 흉터와 감염이 주요 원인이다. 비반흔성 탈모증은 조 직이 섬유화되지 않고 모낭도 보존된 상태로, 원형 탈모증, 휴지기 /생장기 탈모증, 여성형 탈모증, 남성형 유전성 안드로겐 탈모증 등이 있다. 탈모의 원인은 정확히 밝혀지지는 않았으나, 대기오염, 식이습관 등 환경적 요인과 유전적 요인, 스트레 스, 호르몬 불균형, 자율신경계 및 혈액순환 장애, 과도한 피지 생성, 모근의 영양 결핍, 알레르기, 세균 감염, 노화 등 생리적 요인들이 복합적으로 작용하는 것으로 추정되고 있다. 탈모증은 과거 중장년층에서만 발생되었으나, 최근에는 20~30대의 젊은 층에서도 빈번하게 발생되고 있다. 이에 탈모 현상을 개선하고 발모를 촉진하기 위한 화장품 또는 제약 연구개 발이 활발히 진행되어 왔으며, 현재 미국 식품의약국 (FDA)의 의약품 허가를 받고 시판 중인 탈모 치료제로는 도포제인 미녹시딜 (minoxidi l )과 경구용 피나스테라이 드 (f inasteride)가 있다. 미녹시딜은 혈관을 확장시켜 혈류량을 증가시키고 모근에 영양분을 공급함으로써 모발 성장을 유도하며, 피나스테라이드는 Π형 5 α -환원효 소의 특정억제제로 테스토스테론이 디하이드로테스토스테론 (dihydrotestosterone) 으로 전환되는 것을 방지함으로써 탈모를 억제하는 것으로 알려져 있다. 이들 발모 치료제는 모근이 활성화 되어 있는 부위에서만 효과가 있기 때문에 탈모를 예방하 는데는 효과가 있지만 장기간 휴지기 상태에 있는 모발의 성장 효과는 미미하거나, 투약하는 동안에만 효과가 지속되는 등의 단점이 있다. 특히 남성호르몬을 조절하 는 약물은 성욕감퇴, 발기부전 등의 부작용이 나타날 수 있다. 이에 탈모를 예방하고 모발 성장을 촉진하며, 그 효과가 안정적으로 유지될 수 있고, 부작용은 적은 안전하고 경제적인 발모 촉진 기술 개발이 필요하다.
