窒素雰囲気下、室温で化合物(4)3.59g(18.8ミリモル)、NMP5.11g及びトリエチルアミン1.99gを混合した。得られた混合物全量及びオキシ塩化リン2.74g(17.9ミリモル)の各々を、化合物(3)5.12gとNMP10.25gとの混合物に、25℃で7時間かけて同時且つ別々に滴下して、同時に滴下を終了した。その後、25℃で2時間撹拌した。滴下速度は、一方が不適切に過剰に添加されないように、化合物(4)及びオキシ塩化リンを同時に滴下し始め、概ね一定の滴下速度を保ち、且つ同時に滴下が終了するように調整した。得られた混合物を、安息香酸イソプロピルを内部標準物質に用いて高速液体クロマトグラフィーにより分析(高速液体クロマトグラフィー分析条件1)したところ、化合物(5)の収率は93.2%であった。
実施例2
窒素雰囲気下、室温で化合物(4)3.59g(18.8ミリモル)、NMP5.11g及びトリエチルアミン1.99gを混合した。得られた混合物全量及びオキシ塩化リン2.98g(19.4ミリモル)の各々を、化合物(3)5.12gとNMP10.22gとの混合物に、25℃で20時間かけて同時且つ別々に滴下して、同時に滴下を終了した。その後、25℃で2時間撹拌した。滴下速度は、一方が不適切に過剰に添加されないように、化合物(4)及びオキシ塩化リンを同時に滴下し始め、概ね一定の滴下速度を保ち、且つ同時に滴下が終了するように調整した。得られた混合物を、安息香酸イソプロピルを内部標準物質に用いて高速液体クロマトグラフィーを用いて分析(高速液体クロマトグラフィー分析条件1)したところ、化合物(5)の収率は96.2%であった。
実施例3
窒素雰囲気下、室温で化合物(4)29.9g(156ミリモル)、NMP46.3g及びトリエチルアミン16.8gを混合した。得られた混合物全量及びオキシ塩化リン24.7g(161ミリモル)の各々を、化合物(3)46.3g(164ミリモル)とNMP92.6gとの混合物に、5℃で5時間かけて同時且つ別々に滴下して、同時に滴下を終了した。滴下速度は、一方が不適切に過剰に添加されないように、化合物(4)及びオキシ塩化リンを同時に滴下し始め、概ね一定の滴下速度を保ち、且つ同時に滴下が終了するように調整した。その後、5℃で2時間撹拌した後、0℃に冷却し13時間攪拌した。得られた混合物を、安息香酸イソプロピルを内部標準物質に用いて高速液体クロマトグラフィーを用いて分析(高速液体クロマトグラフィー分析条件1)したところ、化合物(5)の収率は94.7%であった。
実施例4
窒素雰囲気下、室温で化合物(5)0.20g、濃硫酸45mg、水0.20g及びDMF0.40gを混合し、120℃に昇温して9時間撹拌した。得られた混合物に水を加え、析出した固体を濾過し、水で洗浄した後、減圧下で乾燥した。ビフェニルを内部標準物質に用いて高速液体クロマトグラフィーを用いて分析(高速液体クロマトグラフィー分析条件2)したところ、化合物(6)の白色固体が収率86.4%で得られたことが確認された。
1H-NMR (CDCl3) δ: 9.16 (1H, s), 8.81 (1H, br s), 8.72 (1H, d), 8.36 (1H, br s), 8.31 (1H, d), 8.21 (1H, s), 3.93 (3H, s), 3.82 (2H, q), 1.39 (3H, t)
実施例5
窒素雰囲気下、室温で化合物(4)14.63g、NMP35.01g及びトリエチルアミン8.53gを混合した。得られた混合物全量及びオキシ塩化リン12.92gの各々を、化合物(3)20.01gとNMP45.02gとの混合物に、25℃で7時間かけて同時且つ別々に滴下して、同時に滴下を終了した。その後、25℃で2時間撹拌した。滴下速度は、一方が不適切に過剰に添加されないように、化合物(4)及びオキシ塩化リンを同時に滴下し始め、概ね一定の滴下速度を保ち、且つ同時に滴下が終了するように調整した。
実施例6
化合物(5)1.83g、90%乳酸水溶液1.00g及びプロピレングリコール4.00gの混合物を120℃に昇温し、120℃で12時間撹拌した。得られた反応混合物を85℃まで冷却し、水5.00gを加えて30分間撹拌した。室温まで冷却して析出した固体を濾過し、水5.00gで洗浄、減圧乾燥することにより化合物(6)を1.78g得た(含量92.6%、収率93.8%:ビフェニルを内部標準物質に用いて高速液体クロマトグラフィー分析条件2により測定した。)。
実施例7
化合物(5)と水との混合物(含水率61.6%)76.12g、90%乳酸水溶液15.58g及びプロピレングリコール85.58gの混合物をバス温135℃で加熱することにより、内温が120℃になるまで脱水を行い、11時間撹拌した。得られた反応混合物を75℃まで冷却し、水142.46gを加えて30分間撹拌した。40℃まで冷却して得た固体を濾過し、水143gで2回洗浄し、減圧乾燥することにより化合物(6)を24.54g得た(含量98.53%、収率86.9%:安息香酸イソプロピルを内部標準物質に用いて高速液体クロマトグラフィー分析条件1により測定した。)。
X線回折の測定条件は以下の通りである。

化合物(5)1.83g及び90%乳酸水溶液4.00gの混合物を120℃に昇温、120℃で10時間撹拌した。得られた反応混合物を85℃まで冷却し、水5.00gを加えて30分間撹拌した。反応混合物を室温まで冷却し、析出した固体を濾過し、水5.00gで洗浄、減圧乾燥することにより化合物(6)を1.80g得た(含量87.8%、収率89.5%:ビフェニルを内部標準物質に用いて、高速液体クロマトグラフィー分析条件2により測定した。)。
実施例9
化合物(5)1.83g、90%乳酸水溶液1.00g及びキシレン4.00gの混合物を150℃に昇温し、150℃で10時間撹拌した。得られた反応混合物を濃縮し、残渣に水5.00gを加えて30分間撹拌した。析出した固体を濾過し、水5.00gで洗浄、減圧乾燥することにより化合物(6)を1.68g得た(含量89.3%、収率85.2%:ビフェニルを内部標準物質に用いて高速液体クロマトグラフィー分析条件2により測定した。)。
実施例10
化合物(5)1.83g、90%乳酸水溶液1.00g及びメチルイソブチルケトン4.00gの混合物を120℃に昇温し、120℃で43時間撹拌した。得られた反応混合物を濃縮し、残渣に水5.00gを加えて30分間撹拌した。析出した固体を濾過し、水5.00gで洗浄、減圧乾燥することにより化合物(6)を1.65g得た(含量84.6%、収率79.5%:ビフェニルを内部標準物質に用いて、高速液体クロマトグラフィー分析条件2により測定した。)。

実施例12
窒素雰囲気下、室温で、6-クロロ-3-(エタンスルホニル)-2-ピリジンカルボン酸カリウムの水溶液(含量:38.9%)150.10gと1H-1,2,4-トリアゾール28.27gとを混合し、85℃まで昇温した。得られた混合物に85℃で48%水酸化カリウム水溶液33.26gを3.5時間かけて滴下した後、85℃で5時間撹拌し、その後、55℃まで冷却した。得られた混合物を、55℃に維持した水145.8gと濃硫酸44.61gとの混合物に4時間かけて滴下した。得られた混合物を20℃まで冷却して得た固体を濾過し、水87.5gで洗浄した。その後、減圧下で乾燥して、収率96.9%で化合物(3)の白色固体を得た。収率は、絶対検量線法により、高速液体クロマトグラフィー分析条件3を用いて測定した。
参考例1
窒素雰囲気下、室温で、化合物(3)5.12g、化合物(4)3.59g、NMP15.41g及びトリエチルアミン2.01gを混合した。混合物に25℃でオキシ塩化リン3.02gを20時間かけて滴下し、その後、25℃で2時間撹拌した。得られた混合物を、安息香酸イソプロピルを内部標準物質に用いて高速液体クロマトグラフィーを用いて分析(高速液体クロマトグラフィー分析条件1)したところ、化合物(5)の収率は78.3%であった。
実施例13
(1)
メチル=3,6-ジクロロピリジン-2-カルボキシラート3.0g、エタンチオール2.27mL及びTHF29mLの混合物に氷冷下、水素化ナトリウム1.28gを加えた。室温で1時間撹拌した後、この混合物に飽和塩化アンモニウム水溶液を加えた後、酢酸エチルで抽出した。有機層を硫酸ナトリウムで乾燥した後、濃縮した。残渣をシリカゲルカラムクロマトグラフィーに付し、メチル=3,6-ビス(エチルスルファニル)ピリジン-2-カルボキシラート2.80gを得た。

メチル=3,6-ビス(エチルスルファニル)ピリジン-2-カルボキシラート2.80g及びクロロホルム36mLの混合物に氷冷下、メタクロロ過安息香酸10.28gを加えた。この混合物を室温で8時間撹拌した後、飽和チオ硫酸ナトリウム水溶液及び飽和重曹水を順次加え、酢酸エチルで抽出した。有機層を硫酸ナトリウムで乾燥した後、濃縮してメチル=3,6-ビス(エタンスルホニル)ピリジン-2-カルボキシラート3.43gを得た。

メチル=3,6-ビス-エタンスルホニル-2-ピリジンカルボキシレート1.0g及びDMF10mLの混合物に室温下、1H-1,2,4-トリアゾール237mgと炭酸カリウム473mgとを加えた。室温で5時間撹拌した後、この混合物に飽和塩化アンモニウム水溶液を加えて酢酸エチルで抽出した。有機層を硫酸ナトリウムで乾燥した後、濃縮してメチル=3-(エタンスルホニル)-6-(1H-1,2,4-トリアゾール-1-イル)ピリジン-2-カルボキシラート790mgを得た。
