本発明は、標識ペプチドおよびその使用に関する。

近年、細胞膜透過性タンパク質を応用したタンパク質療法の開発が顕著である。しかし、タンパク質を用いた療法では、細胞や個体に投与後、タンパク質がプロテオソームやファゴソーム、オートファジーやタンパク質分解酵素等によって分解され、その結果、投与タンパク質があまり効力を発揮しない場合や、極めて頻繁に投与する必要が生じる場合が多い。そのため、タンパク質の分解を抑制し、安定化する方法が求められている。
これまでに、タンパク質の分解を抑制する活性を有するアミノ酸配列として、ネイスセリア・メンニギチジスのP64K抗原のN末端領域の最初の47アミノ酸から誘導されたモチーフ等が開示されている(例えば、特許文献1参照)。
特許文献1に開示されているタンパク質の分解を抑制可能なモチーフは、タンパク質の安定化に寄与するが、相同性を有するアミノ酸配列が天然に複数存在するため、タンパク質の検出又は精製用のタグとしての用途には適さない。
本発明は、上記事情に鑑みてなされたものであって、タンパク質を安定化させ、かつ、容易に検出することができる技術を提供することを目的とする。また、本発明は、標識ペプチド、核酸、ベクター、融合タンパク質、対象タンパク質を安定化する方法、特異的結合物質、融合タンパク質の検出方法および融合タンパク質の精製方法を提供することを目的とする。

本発明によれば、本来プロテアソーム、オートファジーやその他タンパク質分解酵素などによって速やかに分解されるタンパク質を安定化させ、かつ、容易に検出することができる。さらに、上記タンパク質の精製、機能解析やその他分子生物学的な解析のためのツール等として活用することができる。
以下、必要に応じて図面を参照しながら、本発明の実施形態について詳細に説明する。なお、図面中、同一又は相当部分には同一符号を付し、重複する説明は省略する。
<標識ペプチド>

本実施形態の標識ペプチドによれば、後述の融合タンパク質を分解から防ぎ安定化させることができる。さらに、天然に存在しないアミノ酸配列であることから、後述の標識ペプチドに対する特異的結合物質を用いることで、標識ペプチドと融合したタンパク質を高感度で検出することができ、また高純度で精製することができる。
本明細書において、「タンパク質が安定化した」とは、本実施形態の標準ペプチドを付加することにより、タンパク質がタンパク質分解酵素等によって分解されにくくなること、又は、タンパク質の合成速度が早まることを意味する。例えば、細胞内にタンパク質を導入した場合に、当該タンパク質の半減期が通常よりも長いことを意味する。さらに、タンパク質が安定化したか否かについては、標識ペプチドを融合したタンパク質および融合していないタンパク質を作製し、通常のウエスタンブロッティング法を用いて、タンパク質を検出することにより評価することができる。
本明細書において、「天然に存在しないアミノ酸配列」とは、天然に存在するタンパク質内に存在しないアミノ酸配列を意味する。例えば、標識ペプチド配列をクエリーとして、2015年2月25日付けのnrデータベース(ftp://ftp.ncbi.nlm.nih.gov/blast/db/)を、blastp(ver. 2.2.30)(http://blast.ncbi.nlm.nih.gov/Blast.cgi?PAGE=Proteins)プログラムを用いて検索した結果、アライメントスコアが80以下、より好ましくは60以下、さらに好ましくは50以下のタンパク質しかヒットしてこないことを意味する。
タンパク質を安定化させる活性が高い観点から、式[1]中のXで表される、グルタミン酸残基の数はアスパラギン酸残基の数より多いことが好ましい。また、mは、0~2であることが好ましい。nは、1であることが好ましい。pは、8であることが好ましい。qは、4であることが好ましい。具体的な標識ペプチドとしては、例えば、配列番号10、18および20で表されるアミノ酸配列からなる標識ペプチドが挙げられる。
(第二実施形態)
また、一実施形態において、本発明は、下記式[2]で表される天然に存在しないアミノ酸配列からなる標識ペプチドを提供する。下記式[2]で表されるように、本実施形態の標識ペプチドは、上記式[1]で表される標識ペプチドのN末端側に、N末端からC末端側に向かって、メチオニン残基およびバリン残基がこの順に付加したものである。本実施形態の標識ペプチドをN末端に有する融合タンパク質は、発現量が上昇し、安定性が向上している。

また、一実施形態において、本発明は、下記式[2]で表される天然に存在しないアミノ酸配列からなる標識ペプチドを提供する。下記式[2]で表されるように、本実施形態の標識ペプチドは、上記式[1]で表される標識ペプチドのN末端側に、N末端からC末端側に向かって、メチオニン残基およびバリン残基がこの順に付加したものである。本実施形態の標識ペプチドをN末端に有する融合タンパク質は、発現量が上昇し、安定性が向上している。

<標識ペプチドをコードする核酸>
一実施形態において、本発明は、上述した標識ペプチドをコードする核酸を提供する。前記標識ペプチドをコードする核酸としては、例えば、配列番号9、17、19に記載の塩基配列からなる核酸、又は、配列番号9、17、19に記載の塩基配列と80%以上、例えば85%以上、例えば90%以上、例えば95%以上の同一性を有し、タンパク質を安定化する活性を有するペプチドをコードする塩基配列からなる核酸等が挙げられる。なお、配列番号9、17、19に示す塩基配列は、後述の実施例で用いられた配列番号10、18、20のアミノ酸配列からなる標識ペプチドをコードする核酸の塩基配列である。
一実施形態において、本発明は、上述した標識ペプチドをコードする核酸を提供する。前記標識ペプチドをコードする核酸としては、例えば、配列番号9、17、19に記載の塩基配列からなる核酸、又は、配列番号9、17、19に記載の塩基配列と80%以上、例えば85%以上、例えば90%以上、例えば95%以上の同一性を有し、タンパク質を安定化する活性を有するペプチドをコードする塩基配列からなる核酸等が挙げられる。なお、配列番号9、17、19に示す塩基配列は、後述の実施例で用いられた配列番号10、18、20のアミノ酸配列からなる標識ペプチドをコードする核酸の塩基配列である。
<標識ペプチドをコードする核酸を含むベクター>
一実施形態において、本発明は、上述した核酸を含むベクターを提供する。本実施形態のベクターは、発現ベクターであることが好ましい。発現ベクターとしては特に制限されず、例えば、pBR322、pBR325、pUC12、pUC13等の大腸菌由来のプラスミド;pUB110、pTP5、pC194等の枯草菌由来のプラスミド;pSH19、pSH15等の酵母由来プラスミド;λファージ等のバクテリオファージ;アデノウィルス、アデノ随伴ウィルス、レンチウィルス、ワクシニアウィルス、バキュロウィルス等のウィルス;及びこれらを改変したベクター等を用いることができる。これらは融合タンパク質発現ベクター作製用であってもよい。
一実施形態において、本発明は、上述した核酸を含むベクターを提供する。本実施形態のベクターは、発現ベクターであることが好ましい。発現ベクターとしては特に制限されず、例えば、pBR322、pBR325、pUC12、pUC13等の大腸菌由来のプラスミド;pUB110、pTP5、pC194等の枯草菌由来のプラスミド;pSH19、pSH15等の酵母由来プラスミド;λファージ等のバクテリオファージ;アデノウィルス、アデノ随伴ウィルス、レンチウィルス、ワクシニアウィルス、バキュロウィルス等のウィルス;及びこれらを改変したベクター等を用いることができる。これらは融合タンパク質発現ベクター作製用であってもよい。
上述の発現ベクターにおいて、標識ペプチド発現用プロモーターとしては特に限定されず、例えば、EF1αプロモーター、SRαプロモーター、SV40プロモーター、LTRプロモーター、CMV(サイトメガロウィルス)プロモーター、HSV-tkプロモーター等の動物細胞を宿主とした発現用のプロモーター、カリフラワーモザイクウイルス(CaMV)の35Sプロモーター、REF(rubber elongation factor)プロモーター等の植物細胞を宿主とした発現用のプロモーター、ポリヘドリンプロモーター、p10プロモーター等の昆虫細胞を宿主とした発現用のプロモーター等を使用することができる。これらプロモーターは、融合タンパク質を発現する宿主に応じて、適宜選択することができる。
上述の発現ベクターは、さらに、マルチクローニングサイト、エンハンサー、スプライシングシグナル、ポリA付加シグナル、選択マーカー、複製起点等を有していてもよい。
<融合タンパク質>
(第一実施形態)
一実施形態において、本発明は、対象タンパク質と上述した標識ペプチドとの融合タンパク質を提供する。

本実施形態の融合タンパク質によれば、対象タンパク質は、上述した標識ペプチドと結合した融合タンパク質として存在することで、細胞内に導入した場合でも、タンパク質分解酵素等による分解が抑制され、安定化させることができる。
本実施形態の対象タンパク質は、特に限定されないが、例えば、細胞の核内に局在するタンパク質、細胞質又は細胞膜に局在するタンパク質等が挙げられる。核内に局在するタンパク質としては、細胞の初期化因子であるSox-2、Oct3/4、Klf4、c-Myc、Glis1等の転写因子、細胞周期に係る因子であるDP-1、p53等の転写因子、転写を活性化させる因子であるGAL4、Foxp3等の転写因子、核内受容体であるRARα等の受容体等が挙げられる。細胞質又は細胞膜に局在するタンパク質としては、シグナル伝達に関連する因子であるSocs3、Dkk1、Wnt3等の因子等が挙げられる。
本実施形態の融合タンパク質において、対象タンパク質が分解されず、かつ、対象タンパク質の有する活性等に影響がない限り、対象タンパク質と標的ペプチドとの融合の位置は特に限定されない。後述する実施例で示すように、標的ペプチドは融合タンパク質のどこに位置していてもよく、対象タンパク質のN末端側、C末端側又は対象タンパク質の内部のいずれに位置していても、対象タンパク質を安定化する効果を発揮することができる。
(第二実施形態)
本実施形態の融合タンパク質は、細胞透過性のモチーフをさらに有していてもよい。細胞透過性のモチーフとしては、例えば、MTM(Membrane translocating motif)等が挙げられる。MTMは、繊維芽細胞増殖因子であるヒトのFGF4(Fibroblast growth factor4)の疎水性シグナル配列領域に由来するアミノ酸配列からなペプチドであり、例えば、配列番号21で表されるアミノ酸配列からなるペプチド(Hawiger et al., 2005 Nature Medicine vol.11 No.(8):892‐8)等が挙げられるがこれに限定されない。融合タンパク質はMTMを有することで、細胞膜透過性を有し、容易に細胞膜内に導入され、活性を失うことなく機能することができる。本実施形態の融合タンパク質において、MTMの融合の位置は特に限定されず、融合タンパク質のN末端側又はC末端側のいずれに連結してもよい。

融合タンパク質は、例えば次のような方法により作成することができる。まず、融合タンパク質をコードする核酸を含む発現ベクターを用いて、宿主を形質転換する。続いて、当該宿主を培養して融合タンパク質を発現させる。培地の組成、培養の温度、時間、誘導物質の添加等の条件は、形質転換体が生育し、融合タンパク質が効率よく産生されるよう、公知の方法に従って当業者が決定できる。また、例えば、選択マーカーとして抗生物質抵抗性遺伝子を発現ベクターに組み込んだ場合、培地に抗生物質を加えることにより、形質転換体を選択することができる。続いて、宿主が発現した融合タンパク質を適宜の方法により精製することにより、融合タンパク質が得られる。
<融合タンパク質をコードする核酸>
一実施形態において、本発明は、上述した融合タンパク質をコードする核酸を提供する。本実施形態の核酸を導入した発現ベクターを作製することで、安定性が向上したタンパク質を発現させることができる。

対象タンパク質に標識ペプチドを結合する工程は、具体的には、対象タンパク質をコードする核酸断片と、標識ペプチドをコードする核酸断片とをLigation等により連結させることにより、実施することができる。上記の核酸を発現させて得られる融合タンパク質は、標識ペプチドを有していない対象タンパク質と比較して、安定性が向上している。
<標識ペプチドに対する特異的結合物質>
一実施形態において、本発明は、上述した標識ペプチドに対する特異的結合物質を提供する。本実施形態の特異的結合物質としては、上述した標識ペプチドに対する抗体、抗体断片、アプタマー、受容体等が挙げられる。
一実施形態において、本発明は、上述した標識ペプチドに対する特異的結合物質を提供する。本実施形態の特異的結合物質としては、上述した標識ペプチドに対する抗体、抗体断片、アプタマー、受容体等が挙げられる。
抗体は、例えば、マウス等のげっ歯類の動物に標識ペプチドを抗原として免疫することによって作製することができる。また、例えば、ファージライブラリーのスクリーニングにより作製することができる。抗体断片としては、Fv、Fab、scFv等が挙げられる。
アプタマーとは、標識物質に対する特異的結合能を有する物質である。アプタマーとしては、核酸アプタマー、ペプチドアプタマー等が挙げられる。標識ペプチドに特異的結合能を有する核酸アプタマーは、例えば、systematic evolution of ligand by exponential enrichment(SELEX)法等により選別することができる。また、標識ペプチドに特異的結合能を有するペプチドアプタマーは、例えば酵母を用いたTwo-hybrid法等により選別することができる。
詳細については後述するが、本実施形態の特異的結合物質によれば、上述した融合タンパク質を高感度に検出することができる。また、融合タンパク質を高純度に精製をすることができる。
<融合タンパク質の検出方法>
一実施形態において、本発明は、上述した融合タンパク質に、上述した標識ペプチドに対する特異的結合物質を接触させる工程を備える前記融合タンパク質の検出方法を提供する。

本実施形態の検出方法によれば、上述した標識ペプチドが天然に存在しないアミノ酸配列からなることから、上述した融合タンパク質を容易に検出することができる。
融合タンパク質に特異的結合物質を接触させる工程は、例えば、次のように実施することができる。まず、融合タンパク質を発現させた宿主を緩衝液に懸濁して超音波破砕等の方法で破壊する。続いて、例えば、破砕液をSDS(Sodium dodecyl sulfate)-ポリアクリルアミドゲル電気泳動(SDS-PAGE)法により、分子量の違いにより分離する。さらに、電気泳動で分離したタンパク質をゲルからメンブレンに移し、ブロッキング剤でメンブレン上をコーティングする。続いて、メンブレン上の融合タンパク質に、上述した標識ペプチドに対する特異的結合物質を接触させ、検出する。
図1は、融合タンパク質の検出方法の一実施形態を説明する模式図である。例えば、マウス由来の上述した標識ペプチドに対する一次抗体4を用いた場合、前記一次抗体4を融合タンパク質3に直接結合させる。さらに、マウス由来のタンパク質に結合する抗体に標識をつけた標識二次抗体5を前記一次抗体4に反応させて、最終的に融合タンパク質3を検出することができる(図1参照)。
図1は、融合タンパク質の検出方法の一実施形態を説明する模式図である。例えば、マウス由来の上述した標識ペプチドに対する一次抗体4を用いた場合、前記一次抗体4を融合タンパク質3に直接結合させる。さらに、マウス由来のタンパク質に結合する抗体に標識をつけた標識二次抗体5を前記一次抗体4に反応させて、最終的に融合タンパク質3を検出することができる(図1参照)。
標識には酵素を用いる系、蛍光を用いる系など、公知の方法を用いることができる。例えば、酵素を用いる系では、HRP(Horseradish Peroxidase)や AP(Alkaline Phosphatase)などで標識した二次抗体を一次抗体に反応させて、酵素活性による発色や化学発光により検出する。化学発光による検出には、X線フィルムへの露光検出、あるいは化学発光を検出可能なスキャナーを必要とするが、発色法に比べ10~50倍以上も感度が高いので、微量タンパク質の検出が容易になる。蛍光法は、Cy3やCy5で標識した二次抗体を蛍光検出するので、定量性に優れている。
<融合タンパク質の精製方法>
一実施形態において、本発明は、上述した融合タンパク質に、上述した標識ペプチドに対する特異的結合物質を接触させる工程を備える前記融合タンパク質の精製方法を提供する。

本実施形態の精製方法によれば、上述した標識ペプチドが天然に存在しないアミノ酸配列からなることから、上述した融合タンパク質を容易に精製することができる。
本明細書において、「精製」は、「分離」、「回収」、「吸着」等といいかえることができる。
本実施形態の精製方法は、例えば、次のように実施することができる。まず、融合タンパク質を発現させた宿主を緩衝液に懸濁して超音波破砕等の方法で破壊する。続いて、標識ペプチドに対する特異的結合物質が固定された固相と、前記融合タンパク質を含む破砕液を接触させる。すると、融合タンパク質中の標識タンパク質が標識ペプチドに対する特異的結合物質に吸着することにより、融合タンパク質を固相に吸着させることができる。
続いて、固相に吸着した融合タンパク質を緩衝液等で洗浄する。その後、固相に吸着した融合タンパク質に、例えば、pHを変化させること、又は、塩を添加すること等により、融合タンパク質を固相から解離させ回収すればよい。
上述の固相としては、従来の精製方法において用いられるものであれば特に限定はなく、例えば、ビーズ、膜、中空糸膜、プラスティック基板、ウェルプレート、スライドガラス、表面プラズモン共鳴測定装置等の測定機器のセンサーチップ等が挙げられる。ビーズ、膜等の形態である固相は、カラムに充填されていてもよい。
以下、実施例により本発明を説明するが、本発明は以下の実施例に限定されるものではない。
<実験例1>
1-1.発現プラスミドの作製
標識ペプチドおよびヒトSox2タンパク質(以下、「hSox2」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、下記表1に示す試験例1~27の融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグ(以下、「Flag」という場合がある。)も導入した。
1-1.発現プラスミドの作製
標識ペプチドおよびヒトSox2タンパク質(以下、「hSox2」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、下記表1に示す試験例1~27の融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグ(以下、「Flag」という場合がある。)も導入した。
表1中、「kozak-Met」は、コザック配列を示している。コザック配列(Kozak sequence)は、真核生物のmRNAに出現する共通配列であり、主に翻訳の開始に関与している。脊椎動物では「(gcc)gccRccAUGG」と表され、なかでも開始コドン(AUG)の3塩基上流のR(プリン塩基・アデニンまたはグアニン)と開始コドンの次のGが重要な役割を果たすと考えられている。「AUG」の部分は、タンパク質のN末端のメチオニン残基をコードしている。今回は、「ccgcc(kozak)-AUG(Met)」で表される塩基配列を用いた。
今回使用した標識ペプチドの塩基配列およびアミノ酸配列については、配列表の配列番号1~20に記載した。また、ヒトSox2の塩基配列およびアミノ酸配列については、配列表の配列番号23および24に記載した。

Flagタグは、配列番号22で表されるアミノ酸配列からなるペプチドであり、この配列をペプチドすることで、ウエスタンブロッティングによる検出が極めて容易になること(ウエスタンブロッティングに用いるHRP融合Flag抗体(SIGMA社)は極めて高感度でFlagタグ融合タンパク質を検出可能である。)から使用した。
各プラスミドを含む単一クローン化した大腸菌を各々アンピシリン入りLB培地を用いて培養した。遠心分離により集菌後、アルカリSDS法およびPEG沈殿法を用い、発現プラスミドを精製した。
1-2.細胞培養
HEK293T細胞(human embryonic kidny(HEK)の細胞を無血清培地に馴化し浮遊細胞化した細胞)を10%FBS入りダルベッコ改変イーグル培地(Dulbecco’s Modi-ed Eagle Medium、DMEM)(抗生物質としてペニシリンとストレプトマイシンを含む)により培養した。トランスフェクション用には、トランスフェクション時に50%コンフルエントとなるように、前日に12ウェルディッシュにHEK293T細胞を撒いた。
HEK293T細胞(human embryonic kidny(HEK)の細胞を無血清培地に馴化し浮遊細胞化した細胞)を10%FBS入りダルベッコ改変イーグル培地(Dulbecco’s Modi-ed Eagle Medium、DMEM)(抗生物質としてペニシリンとストレプトマイシンを含む)により培養した。トランスフェクション用には、トランスフェクション時に50%コンフルエントとなるように、前日に12ウェルディッシュにHEK293T細胞を撒いた。
1-3.トランスフェクション
HEK293T細胞に試験例1~27の発現プラスミドをトランスフェクションした。発現プラスミドは、上述のものを各200ng用い、補正用にpcDNA発現プラスミドを各々計3.0μgとなるように加え、さらにTEバッファー20μL(10mM Tris-HCl(pH8.0)、1mM エチレンジアミン四酢酸(ethylenediaminetetraacetic acid、EDTA))、2M塩化カルシウム溶液3μLを加えた。トランスフェクション試薬には、HBSバッファー(20mM HEPES(pH7.1)、280mM NaCl、1.5mM Na2HPO4)を用いた。発現プラスミド溶液に、HBSバッファー20μLを2回に分けてゆっくりと加え、撹拌懸濁した。発現プラスミド溶液中のDNAと、リン酸カルシウムとの複合体を形成するため、30分間静置した。その間、細胞の培地を交換した。発現プラスミド溶液中のDNAと、リン酸カルシウムとの複合体を培地に添加し、ゆっくりと混ぜ合わせた。CO2インキュベーターに置き、5%CO2濃度、37℃の条件下で培養を続けた。
HEK293T細胞に試験例1~27の発現プラスミドをトランスフェクションした。発現プラスミドは、上述のものを各200ng用い、補正用にpcDNA発現プラスミドを各々計3.0μgとなるように加え、さらにTEバッファー20μL(10mM Tris-HCl(pH8.0)、1mM エチレンジアミン四酢酸(ethylenediaminetetraacetic acid、EDTA))、2M塩化カルシウム溶液3μLを加えた。トランスフェクション試薬には、HBSバッファー(20mM HEPES(pH7.1)、280mM NaCl、1.5mM Na2HPO4)を用いた。発現プラスミド溶液に、HBSバッファー20μLを2回に分けてゆっくりと加え、撹拌懸濁した。発現プラスミド溶液中のDNAと、リン酸カルシウムとの複合体を形成するため、30分間静置した。その間、細胞の培地を交換した。発現プラスミド溶液中のDNAと、リン酸カルシウムとの複合体を培地に添加し、ゆっくりと混ぜ合わせた。CO2インキュベーターに置き、5%CO2濃度、37℃の条件下で培養を続けた。
1-4.ウエスタンブロッティング法による融合タンパク質の検出
(1)細胞抽出液の調製
トランスフェクション後、24時間培養を続けた細胞から培地を捨て、1mLリン酸緩衝生理食塩水(Phosphate buffered saline、PBS)を加え、軽いピペッティングで細胞を剥がし、1.5mLのチューブに移した。1.5krpmの遠心分離で細胞を回収し、上清を捨てた。沈殿した細胞に80μLのNET-N+バッファー(20mM Tris-HCl(pH7.9)、1mM EDTA(pH7.9)、150mM NaCl、1% NP-40およびプロテアーゼインヒビターカクテルを使用直前に添加したもの)を加え、数回ピペッティングにより抽出し、13.5krpm、4℃で遠心分離を行った。上清を別の新しい1.5mLのチューブに移し、80μLの2×SDS-PAGEサンプルバッファーを加え、撹拌した。98℃で1分間煮沸し、電気泳動用のサンプルを調製した。

(2)SDS-PAGE法による融合タンパク質の分離
12%SDS-PAGEゲルにより、各レーンの細胞抽出液サンプルを5μLずつロードし電気泳動した。電気泳動後、ゲル板からゲルを外した。

(3)ゲルからメンブレンへの転写
電気泳動により得られたゲルをトランスファーバッファーに浸した。ゲルをトランスファーブロット(BioRAD社)にセットし、ゲル上に分離したサンプルタンパク質をPVDF(ポリビニリデンジフロライド)メンブレン(ミリポア社)に転写した。

(4)ブロッキング
サンプルタンパク質を移したPVDFメンブレンをブロッキングバッファー(ナカライ社)に浸した。

(5)抗体反応
TBSTバッファー(20mM Tris-HCl(pH7.5)、150mM NaCl、0.1% Tween20)で4回洗浄後、一次抗体としてHRP融合抗Flag抗体、またサンプルの抽出液の均一性を補正するためにハウスキーピング遺伝子であるβ-アクチンに特異的な抗体(抗β-アクチン, モノクローナル抗体, ペルオキシダーゼ結合)を用いた。
TBSTバッファー(20mM Tris-HCl(pH7.5)、150mM NaCl、0.1% Tween20)で4回洗浄後、一次抗体としてHRP融合抗Flag抗体、またサンプルの抽出液の均一性を補正するためにハウスキーピング遺伝子であるβ-アクチンに特異的な抗体(抗β-アクチン, モノクローナル抗体, ペルオキシダーゼ結合)を用いた。
(6)検出
検出はケミルミワン(Chemi-Lumi One)(ナカライ社)をHRPの基質として用い、発色をハイパーフィルム(GEヘルスケア社)によって検出した。結果は、図2に示した。

1-5.結果
対照である試験例1よりも発現量が多いものは、分解抑制活性を有すると判断できる。図2から、N末端領域に標識ペプチドが配置した融合タンパク質では、配列番号4、10、12の標識ペプチドを融合したヒトSox2の発現が特に高かった。
また、C末端領域に標識ペプチドを配置した融合タンパク質では、配列番号2、10、18、20の標識ペプチドを融合したヒトSox2の発現が特に高かった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号2、10、18、20の標識ペプチドを融合したヒトSox2の発現が特に高かった。
従って、本発明の標識ペプチドは、ヒトSox2と融合したタンパク質において、分解耐性活性を十分に有することが明らかになった。
対照である試験例1よりも発現量が多いものは、分解抑制活性を有すると判断できる。図2から、N末端領域に標識ペプチドが配置した融合タンパク質では、配列番号4、10、12の標識ペプチドを融合したヒトSox2の発現が特に高かった。
また、C末端領域に標識ペプチドを配置した融合タンパク質では、配列番号2、10、18、20の標識ペプチドを融合したヒトSox2の発現が特に高かった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号2、10、18、20の標識ペプチドを融合したヒトSox2の発現が特に高かった。
従って、本発明の標識ペプチドは、ヒトSox2と融合したタンパク質において、分解耐性活性を十分に有することが明らかになった。
<実験例2>
2-1.発現プラスミドの作製
標識ペプチドおよびヒトRARαタンパク質(以下、「hRARα」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、表2に示す融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグも導入した。また、ヒトRARαの塩基配列およびアミノ酸配列を配列番号25および26に記載した。
2-1.発現プラスミドの作製
標識ペプチドおよびヒトRARαタンパク質(以下、「hRARα」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、表2に示す融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグも導入した。また、ヒトRARαの塩基配列およびアミノ酸配列を配列番号25および26に記載した。
2-2.細胞培養
実験例1の1-2.と同様に行った。
2-3.トランスフェクション
実験例1の1-3.と同様の手順により、HEK293T細胞に試験例28~54の発現プラスミドをトランスフェクションした。
2-4.ウエスタンブロッティング法による融合タンパク質の検出
実験例1の1-4.と同様に行った。結果を図3に示した。

図3から、N末端領域に標識ペプチドが配置した融合タンパク質では、配列番号2、4、6、16の標識ペプチドを融合したヒトRARαの発現が特に高かった。
また、C末端領域に標識ペプチドを配置した融合タンパク質では、配列番号10、18、20の標識ペプチドを融合したヒトRARαの発現が特に高かった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号12の標識ペプチドを融合したヒトRARα以外はすべてにおいて発現が高かった。
従って、本発明の標識ペプチドは、ヒトRARαと融合したタンパク質においても、分解耐性活性を十分に有していることが明らかになった。
図3から、N末端領域に標識ペプチドが配置した融合タンパク質では、配列番号2、4、6、16の標識ペプチドを融合したヒトRARαの発現が特に高かった。
また、C末端領域に標識ペプチドを配置した融合タンパク質では、配列番号10、18、20の標識ペプチドを融合したヒトRARαの発現が特に高かった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号12の標識ペプチドを融合したヒトRARα以外はすべてにおいて発現が高かった。
従って、本発明の標識ペプチドは、ヒトRARαと融合したタンパク質においても、分解耐性活性を十分に有していることが明らかになった。
<実験例3>
3-1.発現プラスミドの作製
標識ペプチドおよびヒトDP-1タンパク質(以下、「hDP-1」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、表3に示す融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグも導入した。また、ヒトDP-1の塩基配列およびアミノ酸配列を配列番号27および28に記載した。
3-1.発現プラスミドの作製
標識ペプチドおよびヒトDP-1タンパク質(以下、「hDP-1」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、表3に示す融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグも導入した。また、ヒトDP-1の塩基配列およびアミノ酸配列を配列番号27および28に記載した。
なお、「hDP-1ΔC395」とは、ヒトDP-1タンパク質中のタンパク質分解に抵抗性を有するC末端領域を欠損した変異体を示す。「hDP-1ΔC395」のcDNAは、以下のプライマー(FowardおよびReverse1)を用いたPCRにより増幅した。また、表3中の試験例57および58に示す融合タンパク質をコードするcDNAは、以下のプライマー(FowardおよびReverse2)を用いたPCRにより増幅した。
PCR反応液は以下の組成であった。
以上の組成のPCR反応液を以下のサイクルで反応させた。
PCR増幅産物を制限酵素処理し、同制限酵素処理したpcDNA3-FlagベクターにLigationし挿入した。挿入はミニプレップ後、制限酵素処理し確認した。また全配列が正しいことをシークエンスにより確認した。
3-2.細胞培養
実験例1の1-2.と同様に行った。
3-3.トランスフェクション
実験例1の1-3.と同様に行った。
3-4.ウエスタンブロッティング法による融合タンパク質の検出
実験例1の1-4.と同様に行った。結果を図4に示した。

図4から、C末端領域に標識ペプチドを配置した融合タンパク質では、ヒトDP-1の発現が見られなかった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号20の標識ペプチドを融合したヒトDP-1の発現が特に高かった。
従って、本発明の標識ペプチドとヒトDP-1との融合したタンパク質では、N末端領域に配置した場合において、分解耐性活性を有することが明らかになった。
図4から、C末端領域に標識ペプチドを配置した融合タンパク質では、ヒトDP-1の発現が見られなかった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号20の標識ペプチドを融合したヒトDP-1の発現が特に高かった。
従って、本発明の標識ペプチドとヒトDP-1との融合したタンパク質では、N末端領域に配置した場合において、分解耐性活性を有することが明らかになった。
<実験例4>
4-1.発現プラスミドの作製
標識ペプチドおよびヒトFoxp3タンパク質(以下、「hFoxp3」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、表7に示す融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグも導入した。また、ヒトFoxp3の塩基配列およびアミノ酸配列を配列番号31および32に記載した。
4-1.発現プラスミドの作製
標識ペプチドおよびヒトFoxp3タンパク質(以下、「hFoxp3」という場合がある。)からなる融合タンパク質を哺乳動物培養細胞で発現させるため、pcDNA発現プラスミド(Invitorogen社)に、表7に示す融合タンパク質をコードするcDNAをLigationにより挿入した。融合タンパク質には、Flagタグも導入した。また、ヒトFoxp3の塩基配列およびアミノ酸配列を配列番号31および32に記載した。
4-2.細胞培養
実験例1の1-2.と同様に行った。
4-3.トランスフェクション
実験例1の1-3.と同様に行った。
4-4.ウエスタンブロッティング法による融合タンパク質の検出
実験例1の1-4.と同様に行った。結果を図5に示した。

さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号18および20の標識ペプチドを融合したヒトFoxp3の発現が特に高かった。
従って、本発明の標識ペプチドは、ヒトFoxp3と融合したタンパク質においても、分解耐性活性を十分に有していることが明らかになった。
図5から、C末端領域に標識ペプチドを配置した融合タンパク質では、配列番号18および20の標識ペプチドを融合したヒトFoxp3の発現が特に高かった。
さらに、Kozak-Metおよび標識ペプチドをN末端領域に配置した融合ペプチドでは、配列番号18および20の標識ペプチドを融合したヒトFoxp3の発現が特に高かった。
従って、本発明の標識ペプチドは、ヒトFoxp3と融合したタンパク質においても、分解耐性活性を十分に有していることが明らかになった。
本発明によれば、タンパク質を安定化させ、かつ、容易に検出又は精製することができる。本発明によれば、分解が速いタンパク質を安定化させ、機能解析やその他分子生物学のツールとして活用することができる。
