Abstract
Description
NeuAcは、発酵法、酵素法、天然物からの抽出法、又は化学合成法等によって製造することができる。既知のNeuAcの結晶としては、2水和物(非特許文献1)及び無水和物(非特許文献2)の結晶が知られているが、当該結晶の溶解液は強酸性を示し、その溶解度は常温で100g/L程度と比較的小さい。さらに当該結晶およびその溶解液が次第に茶褐色に変色するなど、保存安定性についても問題があった。
このような状況において、常温及び高温条件下における保存安定性が高い、NeuAcのアンモニウム塩の結晶が望まれていた。
(1)NeuAcアンモニウム塩・無水和物の結晶。
(2)粉末X線回折において、回折角(2θ)が、12.3±0.2°、好ましくは±0.1°、13.7±0.2°、好ましくは±0.1°、14.2±0.2°、好ましくは±0.1°、22.4±0.2°、好ましくは±0.1°、および22.6±0.2°、好ましくは±0.1°にピークを有する、上記(1)に記載の結晶。
(3)粉末X線回折において、回折角(2θ)が、さらに17.1±0.2°、好ましくは±0.1°、21.2±0.2°、好ましくは±0.1°、21.6±0.2°、好ましくは±0.1°、23.5±0.2°、好ましくは±0.1°、および24.8±0.2°、好ましくは±0.1°にピークを有する、上記(2)に記載の結晶。
(4)粉末X線回折において、回折角(2θ)が、さらに27.7±0.2°、好ましくは±0.1°、28.1±0.2°、好ましくは±0.1°、28.4±0.2°、好ましくは±0.1°、28.6±0.2°、好ましくは±0.1°、および42.3±0.2°、好ましくは±0.1°にピークを有する、上記(3)に記載の結晶。
(5)pHが3.0~9.0であるアンモニウム含有化合物を含むNeuAcの水溶液に、アルコール類およびケトン類からなる群より選ばれる溶媒を添加または滴下することによりNeuAcアンモニウム塩・無水和物の結晶を析出させ、該水溶液からNeuAcアンモニウム塩・無水和物の結晶を採取することを特徴とする、NeuAcアンモニウム塩・無水和物の結晶の製造法。
(6)アルコール類およびケトン類からなる群より選ばれる溶媒が、C1~C6のアルコール、ならびにアセトン、メチルエチルケトン、およびジエチルケトンから選ばれる溶媒である、上記(5)に記載の製造法。
本発明は、NeuAcアンモニウム塩・無水和物の結晶に関する。
NeuAcの結晶が無水和物の結晶であることは、後述の分析例に記載のカールフィッシャー法を用いて測定した水分含量が、1.0重量%以下、好ましくは0.95重量%以下、最も好ましくは0.9重量%以下であることにより確認することができる。
例えば、NeuAcの無水物の結晶が1アンモニウム塩の結晶であることは、該結晶中のアンモニウム含量が、5.2±1.0重量%、好ましくは5.2±0.5重量%、最も好ましくは5.2±0.3重量%であることにより確認することができる。
また、本発明の結晶としては、後述の分析例に記載の赤外分光(IR)分析に供した場合、図4に示す赤外吸収スペクトルを示すNeuAcアンモニウム塩・無水物の結晶を挙げることができる。
2.本発明のNeuAcアンモニウム塩・無水和物の結晶の製造法
本発明のNeuAcアンモニウム塩・無水和物の結晶の製造法は、pHが3.0~9.0、好ましくは4.5~8.5、最も好ましくは5.5~8.0であるアンモニウム含有化合物を含むNeuAcの水溶液に、アルコール類およびケトン類からなる群より選ばれる溶媒を添加または滴下することによりNeuAcアンモニウム塩・無水和物の結晶を析出させ、該水溶液からNeuAcアンモニウム塩・無水和物の結晶を採取することを特徴とする方法である。
NeuAcの水溶液に、結晶化の障害となる固形物が含まれる場合には、遠心分離、濾過またはセラミックフィルタ等を用いて固形物を除去することができる。また、NeuAcの水溶液に、結晶化の障害となる水溶性の不純物や塩が含まれる場合には、イオン交換樹脂等を充填したカラムに通塔する等により、水溶性の不純物や塩を除去することができる。
アンモニウム含有化合物としては、アンモニア水溶液及びアンモニアガスのような塩基性化合物や、アンモニウムの炭酸化物、アンモニウムの硫酸化物、アンモニウムの硝酸化物、アンモニウムの塩化物のような中性塩を挙げることができる。中性塩としては、例えば、炭酸アンモニウム、硫酸アンモニウム、硝酸アンモニウム、または塩化アンモニウムを挙げることができる。
アルコール類としては、C1~C6のアルコール類を、より好ましくは、C1~C3のアルコール類を、さらに好ましくは、メタノール、エタノール、n-プロパノールおよびイソプロピルアルコールからなる群より選ばれるアルコール類を、最も好ましくは、メタノールおよびエタノールからなる群より選ばれるアルコール類を挙げることができる。
アルコール類およびケトン類からなる群より選ばれる溶媒を添加または滴下するときの該水溶液の温度としては、NeuAcが分解しない温度ならいずれでもよいが、溶解度を下げてNeuAcアンモニウム塩・無水和物の結晶の結晶化率を向上させるために、80℃以下、好ましくは70℃以下、より好ましくは60℃以下、最も好ましくは50℃以下を挙げることができる。
アルコール類およびケトン類からなる群より選ばれる溶媒を添加または滴下する時間としては、1~48時間、好ましくは3~30時間、最も好ましくは5~20時間を挙げることができる。
種晶を添加する時間としては、アルコール類およびケトン類からなる群より選ばれる溶媒の滴下または添加を開始してから、1~5時間以内、好ましくは1~4時間以内、最も好ましくは1~3時間以内を挙げることができる。
熟成させるとは、アルコール類およびケトン類からなる群より選ばれる溶媒を添加または滴下する工程を一旦停止して、結晶を成長させることをいう。
NeuAcアンモニウム塩・無水和物の結晶を採取する方法としては、特に限定されないが、加圧濾過、吸引濾過、遠心分離等を挙げることができる。さらに母液の付着を低減し、結晶の品質を向上させるために、適宜、結晶を洗浄することができる。結晶洗浄に用いる溶液に特に制限はないが、水、メタノール、エタノール、アセトン、n-プロパノール、イソプロピルアルコールおよびそれらから選ばれる1種類または複数種類を任意の割合で混合したものを用いることができる。
上記の製造法によって製造することができるNeuAcアンモニウム塩・無水和物の結晶としては、例えば、X線源としてCuKαを用いた粉末X線回折パターンが、図1~3ならびに表1、4、および6に示す値で規定される、NeuAcアンモニウム塩・無水和物の結晶を挙げることができる。
[分析例]
(1)粉末X線回折
粉末X線回折装置(XRD)Ultima IV(リガク社製)を用い、測定は使用説明書に従って行った。
(2)溶解度の測定
NeuAcアンモニウム塩・無水和物の結晶、または既知であるNeuAc2水和物の結晶を、各温度に調節した水に溶け残るまでそれぞれ添加し、十分な時間、撹拌保持した後、結晶を含まない上澄み液を採取し、以下のHPLC条件を用いてNeuAc濃度を測定した。
ガードカラムShodex SUGAR SH-G φ6.0×50mm
カラム: SUGAR SH1011 φ8.0×300mm×2本直列
カラム温度:60℃
緩衝液:0.005mol/Lの硫酸水溶液
流速:0.6mL/min
検出器:UV検出器(波長210nm)
(3)カールフィッシャー法による結晶の水分含量の測定
自動水分測定装置AQV-2200(平沼産業社製)を用い、使用説明書に従って行った。
(4)アンモニウム含量の測定
NeuAcアンモニウム塩・無水和物の結晶を水に溶解し、以下のHPLC条件を用いて、結晶中に含まれるアンモニウムイオンの濃度を測定した。
カラム: YMC-Pack ODS-AQ φ6.0×150mm
カラム温度:40℃
緩衝液:クエン酸三ナトリウム二水和物(14.7g)、無水硫酸ナトリウム(7.1g)、ラウリル硫酸ナトリウム(15g)、1-プロパノール(600mL)を脱イオン水に溶解して5Lとし、硫酸を用いてpHを3.8に調整する。
反応液:ホウ酸(18.5g)、水酸化ナトリウム(10.7g)、N-アセチルシステイン(4.6g)、o-フタル酸アルデヒド(0.6g)、Brij-35(4.1mL)を脱イオン水に溶解して1Lとする。
流速:1.2mL/min(移動相)、0.4mL/min(反応液)
検出器:励起波長:355nm、蛍光波長:455nm
(5)融点の測定
Melting Point M-565(BUCHI社製)を用い、使用説明書に従って行った。
(6)赤外分光(IR)分析
FTIR-8400型(島津製作所製)を用い、使用説明書に従って行った。
[参考例1]
NeuAcアンモニウム塩の非結晶性アモルファスの取得
NeuAc無水和物の結晶519.4gを水に溶解し、アンモニア水を用いてpHを6.80に調整し、NeuAcアンモニウム塩含有水溶液650mLとした。この水溶液の一部を凍結乾燥することで、白色の粉末を得た。当該粉末の粉末X線回折を測定したところ、X線回折ピークが確認されなかったことから、当該粉末は非結晶性アモルファスであるであることがわかった。
無水和物換算で1546.7gのNeuAcを水に溶解し、アンモニア水を用いてpHを7.67に調整し、3800mLとした。この水溶液を濃縮して1800mLとし、得られた濃縮溶液のうち500mLを次の工程に供した。
当該500mLの濃縮溶液を45℃に維持しつつ、これに1500mL(3倍量)のメタノールを3時間かけて滴下添加したのち、アセトン400mLを添加した。1時間熟成させたのち、アセトン1100mL(最終添加量:3倍量)を3時間かけて追加添加し、結晶を析出させた。結晶スラリーを5℃まで冷却して3時間熟成させた後に当該結晶を濾取し、80%メタノール水溶液で洗浄した後、25℃で減圧乾燥させることにより、430.8gの結晶を得た。
以上より、当該結晶は、NeuAcアンモニウム塩・無水和物の結晶であることがわかった。
NeuAcアンモニウム塩・無水和物の結晶の溶解度は、既知のNeuAc・2水和物結晶の溶解度に比べて大きく、溶解性に優れていることが示された。
また、NeuAcアンモニウム塩・無水和物の結晶の融点が166.2℃と、既知のNeuAcアンモニウム塩・1水和物塩の105℃付近よりも高いことから、NeuAcアンモニウム塩・無水和物の結晶は、高温条件下においても安定であることが示された。
無水和物換算で1475.8gのNeuAcを水に溶解し、アンモニア水を用いてpHを6.81に調整し、4000mLとした。この水溶液を濃縮して1810mLとし、得られた濃縮溶液のうち40mLを次の工程に供した。
当該濃縮溶液40mLに10mLの水を加えて50mLとした。当該50mLの溶液を40℃に維持しつつ、これに40mLのエタノールを1時間かけて滴下添加した。実施例1で取得した結晶を種晶として添加し、結晶を析出させた。当該結晶を9時間かけて熟成させた後、さらに110mL(最終添加量:3倍量)のエタノールを8時間かけて滴下添加した。結晶スラリーを10℃まで冷却して3時間熟成させた後に当該結晶を濾取し、80%エタノール水溶液で洗浄した後、25℃で減圧乾燥させることにより、32.9gの結晶を得た。
当該結晶の各種物性を表4に示す。pHは、NeuAcアンモニウム塩として100g/Lの水溶液を測定した。
実施例2で調整した濃縮溶液50mLを40℃に維持しつつ、これに25mL(0.5倍量)のメタノールを1時間かけて滴下添加した。種晶を添加して4時間熟成させたのち、225mL(最終添加量:5倍量)のメタノールを12時間かけて滴下添加することで結晶を析出させた。結晶スラリーを10℃まで冷却して3時間熟成させた後に当該結晶を濾取し、80%メタノール水溶液で洗浄した後、25℃で減圧乾燥させることにより、40.6gの結晶を得た。
当該結晶の各種物性を表7に示す。pHは、NeuAcアンモニウム塩として100g/Lの水溶液を測定した。

- N-アセチルノイラミン酸アンモニウム塩・無水和物の結晶。
- 粉末X線回折において、回折角(2θ)が、12.3±0.2°、13.7±0.2°、14.2±0.2°、22.4±0.2°、および22.6±0.2°にピークを有する、請求項1に記載の結晶。
- 粉末X線回折において、回折角(2θ)が、さらに17.1±0.2°、21.2±0.2°、21.6±0.2°、23.5±0.2°、および24.8±0.2°にピークを有する、請求項2に記載の結晶。
- 粉末X線回折において、回折角(2θ)が、さらに27.7±0.2°、28.1±0.2°、28.4±0.2°、28.6±0.2°、および42.3±0.2°にピークを有する、請求項3に記載の結晶。
- pHが3.0~9.0であるアンモニウム含有化合物を含むN-アセチルノイラミン酸の水溶液に、アルコール類およびケトン類からなる群より選ばれる溶媒を添加または滴下することによりN-アセチルノイラミン酸アンモニウム塩・無水和物の結晶を析出させ、該水溶液からN-アセチルノイラミン酸アンモニウム塩・無水和物の結晶を採取することを特徴とする、N-アセチルノイラミン酸アンモニウム塩・無水和物の結晶の製造法。
- アルコール類およびケトン類からなる群より選ばれる溶媒が、C1~C6のアルコール、ならびにアセトン、メチルエチルケトン、およびジエチルケトンから選ばれる溶媒である、請求項5に記載の製造法。
