Abstract
Description
本発明はさらに、細胞の電気生理学的機能を評価するためのデバイス、並びに細胞の電気生理学的機能を評価するための方法を提供する。本発明はまた、細胞培養用シートを提供する。
本発明の第二の態様においては、培養細胞の電気生理学的機能を評価するためのデバイスを提供することを目的とする。
本発明の第三の態様においては、細胞培養の足場材として用いられる細胞培養用シートを提供することを目的とする。
(1) 配向性ファイバーシートと、該配向性ファイバー上の培養心筋細胞とを含む、心筋組織片。
(2) 配向性ファイバーシートが培養心筋細胞層の間に介在している、1の心筋組織片。
(3) 該心筋細胞が多能性幹細胞から誘導された心筋細胞である請求項1または2に記載の心筋組織片。
(4) 配向性ファイバーシートにおけるファイバーの直径が0.1μm~5μm、シートの幅1mm当たりのファイバーの本数30~15000本、シートの厚みが0.1μm~20μmである、1~3いずれかに記載の心筋組織片。
(5) 心筋細胞層の数が10層以上である、1~4いずれかに記載の心筋組織片。

播種:1~100×105/cm2
培地:20%FBS含有IMDM(非特許文献5および以下の実施例2に記載)あるいは0.04~0.4%アルブミン含有IMDM(非特許文献5および以下の実施例2に記載)等
培養条件:37℃、5%CO2インキュベーター内で培養。適宜培地交換を行う。
例えば移植に用いる心筋組織片を調製する場合、かかる条件にて約4~10日培養すればよい。
1-1.PLGA配向性ファイバーシートの製造
A.製造手順
20~25%PLGA溶液(PLGA(PLA75%:PGA25%):Sigma, P-1941, 分子量:66,000~107,000)のTHF溶液を23Gのプラント針(ニプロ)を付けたシリンジにいれ、気泡を抜き、マイクロシリンジポンプに流速10mL/分でセットした。高速回転ドラムの表面にアルミ粘着テープ(三商)を貼り付け、シリンジの針先端から10cmの距離に設置した。プラント針にプラス電極、高速回転ドラムにマイナス電極を設置し、高速回転ドラムを3000回転/分で回転させた。マイクロシリンジポンプのスイッチを入れ、8kVの電圧下で、回転ドラム上のアルミ粘着テープの上にファイバーを一方向に配向させて噴出した(噴出時間:40秒)。ファイバー噴出後、配向性ファイバーシートが収集されたアルミテープを回収し、配向性ファイバーシートを得た。また、同様の方法でランダムにファイバーを噴出させ、ランダムファイバーシート(Random Fiber,RF)を作製した(噴出時間:20秒)(図2A)。
PLGAファイバーシートの直径
上記Aで作製したPLGA配向性ファイバーシートおよびランダムファイバーシートを電子顕微鏡(JEOL JEM1400、日本)で撮影し、イメージソフトウェア(Image J)を用いてファイバーの直径サイズを測定した。直径サイズの分布図(図2B)に示すように、PLGAランダムファイバーシートおよびPLGA配向性ファイバーシートの平均直径サイズは1~1.5μmであった。
上記Aに準じて作製したPLGA配向性ファイバーシート(噴出時間:40秒、厚み:2μm、密度:300本/mm)とPLGAランダムファイバーシート(噴出時間:20秒、厚み:2μm)の弾力性を比較するために、島津精密万能試験機(AGS-X)を用いて、引っ張り実験を行った。その結果、配向性ファイバーシートと配向方向に平行する方向の弾力性はランダムファイバーシートの弾力性よりも約3倍高い(220MPa)ことが示された(図2C)。
ファイバーの直径は、装置の条件を一定とすればファイバーの素材濃度を変えることによって調節することができる。上記Aに記載のエレクトロスピニング条件下における、PLGA配向性ファイバーシート製造の最適素材濃度を検討するため、それぞれ20%、23%および25%のPLGA溶液を用いて、上記Aの方法に準じて配向性ファイバーシートを作製した。作製したファイバーの直径の分布を測定した結果、材料の濃度が増加するにつれて、ファイバーの直径も大きくなることが示された(図2D)。直径1~1.5μmのサイズのファイバーを安定して作製できる条件として、20~23%PLGA溶液を用いるのが適当であることが明らかになった。
上記Aの方法に準じて、噴出時間40秒の条件下で作製したPLGA配向性ファイバーシートを電子顕微鏡(JEOL JEM1400、日本)で撮影し、厚みを測定した。さらに、それぞれのファイバーシートについて、配向方向と垂直に切断した断面において、単位長さに対するPLGAファイバー数をカウントすることにより、シートの幅1mm当たりのファイバーの本数(密度)を調べた。該PLGA配向性ファイバーシートの厚みは2μm、密度は300本/mmであった。
さらに、PLGA配向性ファイバーシート(噴出時間:40秒)中のファイバーの配向性を調べるために、PLGA配向性ファイバーシートの電子顕微鏡写真から、イメージソフトウェア(Image J) を用いてファイバーの角度を測定した。配向性ファイバーシートを構成するファイバーのうち、90.8%以上のファイバーが配向方向(0°)の±20°以内にあることが確認できた(図2E)。また、上記Aに記載の条件で作製したPLGAランダムファイバーシートの厚みは、2μmであった。
A.製造手順
ファイバーの材料として、PLGA溶液の代わりに、16%PMGI溶液(Michro chem)を用いて、上記1-1.Aの方法に準じて、PMGI配向性ファイバーシートを作製した。エレクトロスピニング法では、ファイバー噴出時間を増やすことによって、より高密度のファイバーを得ることが可能となる。噴出時間をそれぞれ、90秒および300秒として、低密度PMGI配向性ファイバーシート(AF(Low))および高密度PMGI配向性ファイバーシート(AF(High))を作製した(図2F;スケールバーは50μmを示す)。
B.PMGI配向性ファイバーシートの性質
それぞれの条件下で得られたPMGI配向性ファイバーの厚みおよび密度を上記1-1に記載の方法に準じて測定した。低密度PMGI配向性ファイバーシート(噴出時間:90秒)の厚みは2μmであり、高密度PMGI配向性ファイバーシート(噴出時間:300秒)の厚みは4μmであることがわかった(図2G)。さらに、各配向性ファイバーシートの密度は、低密度PMGI配向性ファイバーシートの場合は、300本/mm、高密度PMGI配向性ファイバーシートの場合は、400本/mmであることがわかった(図2H)。さらに、PMGI低密度配向性PMGIファイバーシートを用いて、上記1-1に記載の方法に準じて該PMGI配向性ファイバーシート中のファイバーの配向性を計測した結果、97%以上のファイバーが配向方向(0°)の±20°以内にあることがわかった(図2I)。
A.製造手順
30%ポリスチレン溶液(PS:Sigma、182435、分子量:130,000~290,000)のDMF溶液を27Gのプラント針(ニプロ)を付けたシリンジにいれ、気泡を抜き、マイクロシリンジポンプに流速1mL/時間でセットした。高速回転ドラムの表面にアルミ粘着テープ(三商)を貼り付け、シリンジの針先端から15cmの距離に設置した。プラント針にプラス電極、高速回転ドラムにマイナス電極を設置し、高速回転ドラムを1000回転/分で回転させた。マイクロシリンジポンプのスイッチを入れ、20kVの電圧下で、回転ドラム上のアルミ粘着テープの上にファイバーを一方向に配向させて噴出した(噴出時間:10分および50分)。ファイバー噴出後、配向性ファイバーシートが収集されたアルミテープを回収し、PS配向性ファイバーシートを得た。ナノファイバー作成装置:MECC、NF500、日本。
上記Aで得られたPS配向性ファイバーシートの厚みおよび密度を、上記1-1に記載の方法に準じて測定した。低密度PSナノファイバー(噴出時間:10分、厚み:2μm、密度:50本/mm);高密度PSナノファイバー(噴出時間:50分、厚み:10μm、密度:300本/mm)。さらに、上記1-1に記載の方法に準じた測定により、該PS配向性ファイバーシート中のPSファイバーの平均直径サイズは2μmであり、該ファイバーシート中のファイバーは、0±5°以内にあることが明らかになった。
2-1.PLGA配向性ファイバーシート上の心筋細胞
A.ヒトiPS細胞(IMR90-1)由来心筋細胞の培養
特許文献1および非特許文献5に記載の方法にしたがってヒトiPS(IMR90-1)細胞から分化誘導した心筋細胞を用いた。心筋細胞のコロニーを50ml遠心管に移し、50g×2分間遠心した。上清を捨て、PBS(GIBCO)で一回洗浄した。プロテアーゼ溶液(0.1%コラゲナーゼタイプI(Life technologies)、0.25%トリプシン、1U/mL DNaseI(Applied Biosystems)、116mM NaCl、20mM HEPES、12.5mM NaH2PO4、5.6mM グルコース、5.4mM KCl、0.8mM MgSO4)を10ml加えて、ガスバーナーで滅菌したスターラーバーを遠心管中に2個入れ、20~30分間、37℃で撹拌した。細胞がある程度溶解している該プロテアーゼ溶液(pH7.35)を、別の回収用50ml遠心管にセットした40μmフィルター(MilliQ)に通した。FBS20%含有IMDM培地(SIgma-Aldrich)(組成:20%FBS(GIBCO)、1%MEM非必須アミノ酸溶液(Sigma)、1%ペニシリン-ストレプトマイシン(Gibco)、2mM L-グルタミン(Sigma)、0.5mM L-カルニチン(Sigma)、0.001%2-メルカプトエタノール(Gibco)、0.005N NaOHおよび10ng/mlBMP4含有、以下FBS20%培地)をフィルターの上からゆっくり20ml加えた。フィルター上に残ったコロニーを回収するために、6cmペトリディッシュ内でフィルターを裏返して、プロテアーゼ溶液5mlで裏側から洗い落とし、最初に用いたスターラーバー入り遠心管に戻し、20~30分間、37℃にて撹拌した。該プロテアーゼ溶液を、該回収用遠心管にセットした40μmフィルターに通し、FBS20%培地をフィルターの上から10ml加えた(プロテアーゼ溶液15mlとFBS培地30mlで計45ml)。該プロテアーゼ溶液45ml中の全細胞数を計測した。細胞を1000rpm×3分間遠心して、上清を捨てた。細胞に必要な濃度に合わせてFBS20%培地を加え(FBS20%培地100μlにつき細胞数が5x105個となるように調整し、次に2x105個に調整する)、ピペッティングにより懸濁した。
各基板に対する細胞接着性を検討するために、細胞を播種した5時間後に、接着していない細胞を除き、接着している細胞数を測定した(図3A)。その結果、PLGAファイバーを接着した基板はファイバー化していないフラットな基板よりも明らかに細胞接着性が高いことが確認された。
細胞接着性に対する、配向性ファイバーシートの密度の影響を検討した。上述したとおり、エレクトロスピニング法では、ファイバー製造時の噴出時間を増やすことによって、より高密度のファイバーを得ることができる。噴出時間10秒、40秒、10分および15分の条件下で、実施例1-1.Aの方法に準じてPLGA配向性ファイバーを作製し、電子顕微鏡で撮影した(図3B)。さらに、それぞれの条件下で得られたファイバーシートの厚みを、実施例1-1.C記載の方法に準じて測定した。ファイバーの密度が高くなることにより、ファイバーシートの厚みも1μmから16μmまで上昇することが確認できた(図3C)。
PMGI配向性ファイバーシートを用いて、同様に細胞接着性に対するファイバーシート密度の影響を検討した。実施例1-2.Aと同様の方法で製造した低密度PMGI配向性ファイバーシート(噴出時間:90秒、厚み:2μm、密度:300本/mm)および高密度PMGI配向性ファイバーシート(噴出時間:300秒、厚み:4μm、密度:400本/mm)をそれぞれ接着した細胞培養用基板およびゼラチンで表面をコーティングした基板(Flat)上に、2-1の方法に準じて心筋細胞を播種し、5時間後における各基板に対する細胞接着性を比較した。その結果、PMGI配向性ファイバーシートにおいても、ファイバー密度が高い場合に、細胞接着率が上昇することが示された(図3F)。一方、低密度PMGI配向性ファイバーシート上の細胞とゼラチンコーティング基板上の細胞は、同程度の接着率を示した。
実施例1に準じてエレクトロスピニング法により製造し、アルミ粘着テープの上に収集されたPMGI配向性ファイバーシート(AF、噴出時間:90秒、厚み:2μm、密度:300本/mm)を、圧力プレス機(MNP-001、アズワン)を用いて、64チャネル多電極アレイ(nitride-coated gold electrodes、multi-channel systems、ドイツ)(電極サイズ:30μm,spaced 200μm,8×8grid array)上に転写し、細胞機能評価用デバイスを得た(図4A、左)。さらに、細胞培養用PDMS製チャンバー(2cmx2cm)を作製し、電極上のPMGI配向性ファイバーシート上に接着させた(図4A、右)。一晩乾燥させて溶媒を蒸発させ、30分間UV照射により滅菌を行った後に、心筋細胞の機能評価に用いた。
PMGI配向性ファイバーシートの代わりにPSファイバーシート(噴出時間:10分、厚み:2μm、密度:50本/mm)およびPLGA配向性ファイバーシート(噴出時間:40秒、厚み:2μm、密度300本/mm)を多電極アレイ上に転写することにより、同様に細胞機能評価用デバイスが得られた(図4B)。
4-1.PMGI配向性ファイバーシート上の心筋細胞の活動電位
A.培養心筋細胞の活動電位測定
実施例3で得られた、機能評価用デバイスのPDMSチャンバー内に、iPS細胞(IMR90-1)由来の心筋細胞を、実施例2の方法に準じて播種した。5時間後、細胞がしっかり接着していることを確認し、チャンバー全体に培地を添加した(FBS20%培地)。37℃インキュベーター内で細胞を培養し、2日目以降、培地をFBS(-)培地に交換し、4日間に一回培地交換を行った。培地を交換する前に、電気信号の測定を行った。測定は、多電極システムを37℃ヒートプレートの上に設置し、温度が安定するまで5分間待った後に行った。測定後、培地交換を行い、インキュベーターで培養を続けた。活動電位測定用のソフトウェアとしては、Multi-channel systems MC-RACK、データ解析用ソフトウェアとしてはOriginPro9.0を使用した。
上記Aの方法にしたがって、PMGI配向性ファイバーシートを転写した多電極アレイ(AF)および対照として、ゼラチンコーティングした多電極アレイ(Flat)上に心筋細胞を播種し、培養を行った。開始2日後には、顕微鏡観察により、PMGI配向性ファイバーシート上の培養細胞が配向性を示すことを確認した。一方、フラットな多電極アレイ上では、細胞が塊状に凝集しやすく、接着性が弱い様子が観察された(図5A)。14日間培養した後、ピンセットで該心筋組織片をファイバーシートごと電極から取り外した。その結果、細胞はファイバーシート側にしっかりと接着しており、多電極アレイ側には細胞およびファイバーは残っていない様子が観察された(図5B)。
実施例3および上記Aの方法に準じて、多電極アレイ上に転写されているPMGI配向性ファイバーシート(AF)上で培養した心筋細胞から、培養開始後2日目、6日目、10日目、14日目において、電気信号を測定し、ゼラチンコーティングした多電極アレイ(Flat)上の細胞と比較した(図5C)。その結果、配向性ファイバーシート上で培養した細胞は、シグナル検出率が時間と共に増加し、6日目以降は全てのチャネルから電気信号を検出することができた(100%)。一方、フラットな多電極アレイ上で培養した心筋細胞は、6日目におけるシグナル検出率が配向性ファイバーシート上の細胞と比較して低かった(約60~80%)。また、培養開始から14日経過すると、フラットな多電極アレイ上の細胞は基板から剥離する傾向が見られたのに対し、PMGI配向性ファイバーシート上の細胞は培養開始から14日以上経過しても細胞の剥離は観察されず(図5D)、電気信号の検出率も低下しなかった。
同様に、PLGA配向性ファイバーシート(AF、噴出時間:40秒、厚み:2μm、密度:300本/mm)およびPLGAランダムファイバーシート(RF、噴出時間:20秒、厚み2μm)をそれぞれ転写した多電極アレイおよびゼラチンでコーティングした多電極アレイ(Flat)上でヒトiPS(IMR90-1)由来心筋細胞を培養し、細胞の電気信号を測定した。ファイバーの構造によらず、ファイバーシートを転写した多電極アレイ上の心筋細胞からのシグナル検出率は時間と共に増加し、6日目からは全てのチャネルから電気信号を検出することができ(100%)、14日経過してもシグナルの検出率は低下しなかった。一方、フラットな多電極アレイを用いた場合、チャネルからのシグナル検出率が低かった(約80~90%)(図5E)。また、フラットな多電極アレイ上では、培養10日目の時点で細胞が剥離する傾向が見られたのに対し、PLGA配向性ファイバーシート上で培養した心筋細胞は、培養開始から32日後においても細胞の剥離は観察されなかった(図5F)。このことから、ファイバーシート上では、細胞接着性が高いために電気信号図の検出率が高く、ゼラチンコーティングした基板上では細胞の接着性が弱く、細胞密度が不均一なためにシグナル検出率が低くなると考えられる。
5-1.PMGI配向性ファイバーシート上の心筋細胞の活動電位パターン
不整脈の検出
実施例3の方法に準じて、PMGI配向性ファイバーシート(AF、噴出時間:90秒、厚み:2μm、密度:300本/mm)を転写した多電極アレイおよびゼラチンコーティングした多電極アレイ(Flat)上で、実施例4の方法に準じて、ヒトiPS(IMR90-1)細胞由来心筋細胞を培養し、該心筋細胞から電気信号を測定した(n=6)。PMGI配向性ファイバーシート上の心筋細胞からは、培養開始から6日間、不整脈(arrhythmia)に対応する不規則な電気活動が全く検出されなかった(図6A下段並びに図6B)。これに対し、フラットな多電極アレイ上で培養した心筋細胞からは、実験サンプル数に対して40%の確率で不整脈が検出された(図6A上段並びに図6B)。
また、PMGI配向性ファイバーシート(AF)上で培養した心筋細胞は、フラット上の細胞よりも電位振幅の値が高いことが確認された(図6Aおよび図6C)。さらに、電位振幅を、14日間経時的に測定した結果、フラットな電極上で培養した心筋細胞では、電位振幅値が変化しなかったのに対し、配向性ファイバーシート上の心筋細胞は、電位振幅値が経時的に上昇することがわかった(図6D)。
5-1と同様に、高密度PLGA配向性ファイバーシート(AF-H、噴出時間:10分、厚み:10μm、密度:10000本/mm)、低密度PLGA配向性ファイバーシート(AF、噴出時間:40秒、厚み:2μm、密度:300本/mm)、およびPLGAランダムファイバーシート(RF、噴出時間:20秒、厚み:2μm)をそれぞれ転写した多電極アレイ、および表面をゼラチンコーティングした多電極アレイ(Flat)を用いて、6日間培養したヒトiPS(IMR90-1)由来心筋細胞の電気信号を測定し、さらに14日間の電位振幅値を経時的に測定した。その結果、ファイバー構造の違いによらず、PLGAファイバー上で培養した心筋細胞は、電位振幅の値がフラットな多電極アレイ上で培養した細胞よりも高いことが確認でき、さらに、PLGAファイバー上で培養した心筋細胞からは、不整脈に相当する不規則な電気活動が全く見られなかった(図6E)。これに対し、従来細胞培養に使われているゼラチンコーティングした基板(Flat)を用いた場合には、頻繁に不整脈が検出された(図6EおよびF)。ゼラチンコーティング基板上の心筋細胞における不整脈の検出率は実験サンプル数に対して34.5%であった(図6F下段)。また、14日間の培養の間、フラットな多電極アレイ上の細胞の電位振幅値は変化しなかったのに対し、ファイバーシート上の心筋細胞の電位振幅値は、時間と共に上昇することが分かった(図6G)。特に培養開始から10日目以降において、配向性ファイバーシート培養条件で培養した心筋細胞が、ランダム培養条件下で培養した心筋細胞よりも振幅値が高い傾向が見られた。
6-1.PMGI配向性ファイバーシート上の心筋組織の興奮伝播
心機能に重要である、心筋組織の興奮伝播方向および伝播速度について測定・評価を行った。実施例3および4の方法に準じて、PMGI配向性ファイバーシート(AF、噴出時間:90秒、厚み:2μm、密度:300本/mm)を転写した多電極アレイおよびゼラチンコーティングした多電極アレイ(Flat)上でヒトiPS(IMR90-1)由来心筋細胞を培養した。培養開始から14日後において、電極を介して、視野の中央部分に電気刺激(±1500 mV、40μs持続)をかけ、心筋細胞の電気信号を測定し、刺激によって引き起こされる電気的興奮の到達分布図を作製した(図7A)。等高線の形の変化は到達方向を表し、色の変化は伝播速度を表す。また、培養開始14日後における、興奮の伝播速度を定量化した(図7B)。
上記6-1と同様に、PLGA配向性ファイバーシート(AF、噴出時間:40秒、厚み:2μm、密度:300本/mm)およびPLGAランダムファイバーシート(RF、噴出時間:20秒、厚み:2μm)をそれぞれ転写した多電極アレイならびにゼラチンコーティングした多電極アレイ(Flat)上で培養した心筋細胞に、6日目、14日目において、視野の中央部分に電気刺激(±1500 mV, 40 μs持続)をかけ、心筋細胞の電気信号を測定し、電気的興奮の到達分布図を作製した(図7D)。また、6日目および14日目の興奮伝播速度をそれぞれ定量化した(図7E)。PLGA配向性ファイバーシートにおいても、興奮伝播方向は配向性ファイバーシートの方向(視野の左右方向)と一致していることがわかった(図7D、左欄)。また、ファイバーがない場合は、6-1の結果と同様に、電気刺激に対して、刺激点から応答反応が起こらず、離れている場所から興奮が誘発される様子が観察された(図7D、右欄)。
7-1.PMGI配向性ファイバー上の心筋細胞の心電図
心筋細胞の電気生理学的成熟度と心毒性試験において、心電図QT間隔(電位依存性Na+電流由来のQ波と電位依存性K+電流由来のT波の間隔)が一つの代表的指標となる。実施例3および4に準じて、電極に転写したPMGI配向性ファイバーシート(AF、噴出時間:90秒、厚み:2μm、密度:300本/mm)およびゼラチンコーティングした多電極アレイ(Flat)上でヒトiPS(IMR90-1)由来心筋細胞を培養した。培養開始後10日目において、それぞれの細胞におけるQT間隔を測定した結果、PMGI配向性ファイバーシート上の心筋細胞では、QT間隔が、フラットな多電極アレイ上の心筋細胞より短いことが確認された(図8A)。さらに、それぞれの条件下で培養した心筋細胞からQT間隔を14日間、経時的に測定した結果、PMGI配向性ファイバーシート上の心筋細胞のQT間隔は、フラットな多電極上の細胞と比較して明らかに短くなっていることが分かった(図8B)。この結果は、配向性ファイバーシート上の培養心筋細胞は電気生理学的成熟度が高いことを示すものである。
A.T波の測定
QT間隔に加えて、心電図T波の検出率もまた、心筋細胞の電気生理学的な成熟度や心毒性を測定する指標の一つとして用いられる。実施例3および4に準じて、多電極アレイ上に接着させた低密度PLGA配向性ファイバーシート(AF、噴出時間:40秒、厚み:2μm、密度:300本/mm)および高密度PLGA配向性ファイバーシート(AF-H、噴出時間:10分、厚み:10μm、密度:10000本/mm)、PLGAランダムファイバーシート(RF、噴出時間:20秒、厚み:2μm)を転写した多電極アレイおよびゼラチンコーティングした多電極アレイ(Flat)上でそれぞれ培養したヒトiPS細胞(IMR90-1)由来心筋細胞から、心電図のT波にあたる電位依存性K+電流由来のT波を多電極システムを用いて14日間経時的に検出し、検出効率を比較した(図8C)。
さらに、上記7-1と同様に、高密度PLGA配向性ファイバーシート(AF-H、噴出時間:10分、厚み:10μm、密度:10,000本/mm)、低密度PLGA配向性ファイバーシート(噴出時間:40秒、厚み:2μm、密度:300本/mm)およびランダムファイバーシート(噴出時間:20秒、厚み:2μm)をそれぞれ転写した多電極アレイ、ならびにゼラチンコーティングした多電極アレイ(Flat)を用いて、培養10日目における心筋細胞のQT間隔を測定した。その結果、配向性ファイバーシート上の心筋細胞のQT間隔が、ランダムファイバーシート上およびフラットな電極上の心筋細胞よりも短いこと、ならびに、高密度配向性ファイバーシート(AF-H)上で培養することで、QT間隔がさらに短くなることがわかった(図8D)。このことより、配向性ファイバーシート上で培養された心筋細胞は電気生理学的成熟度が高く、ファイバーの密度が高くなると成熟度がさらに高くなることが示唆される。
実施例1-3の方法で作製したPS配向性ファイバーシート(噴出時間:50分、厚み:10μm、密度:300本/mm)を転写した多電極アレイを用いて、上記7-1と同様に培養開始6日目時点における細胞のQT間隔を測定した結果、該PS配向性ファイバーシート上の心筋細胞は0.16秒という非常に短いQT間隔を示した(図8F)。この値は、成人のQT間隔(約0.2秒)と近い値であり、このことは、該配向性ファイバーシート上の細胞は成人と同程度の電気生理学的成熟度が高いことを示すものである。
8-1.PLGA配向性ファイバーシート上の心筋組織の免疫染色
さらに、配向性ファイバーシート上の心筋組織片の細胞成熟度を調べるために、心筋細胞マーカーであるα-MHC、β-MHC、myosin light chain(MLC)-2V、cardiac troponin T(cTnT)2およびα-Actininについての免疫染色を行った。PLGA配向性ファイバーシート(噴出時間:40秒、厚み:2μm)、PLGAランダムファイバーシート(噴出時間:20秒、厚み:2μm)を接着させたガラス板およびゼラチンコーティングしたガラス板上で、iPS細胞由来培養組織片を14日間培養した。該細胞を4%ホルムアルデヒドで15分間固定し、PBSで5分間洗浄した(3回)。PBS-0.5%Triton X-100を用いて30分間透過処理を行い、PBSで5分間洗浄した(3回)。ブロッキング液(PBS中、5%正常ヤギ血清、5%正常ロバ血清、3%BSA、0.1%Tween20)を用いて1時間ブロッキング処理を行い、PBSで5分間洗浄した(3回)。一次抗体(α-actinin(マウスモノクローナル抗体 EA-53,Sigma)、cTnT2; マウスモノクローナル,Santa Cruz)、MLC-2v(ウサギポリクローナル、Proteintech Group,IL、米国)、α-MHC(ウサギモノクローナル抗体、Sigma)、β-MHC(マウスモノクローナル抗体、Santa Cruz)を用いて、4℃で一晩反応させた後、PBSで5分間洗浄した(3回)。二次抗体(DyLight 488-結合型抗マウスIgG、DyLight 488-結合型抗-ウサギIgG、DyLight 594-結合型抗マウスIgG、DyLight 594-結合型抗-ウサギIgG)を用いて、室温で1時間反応させ、PBSで5分間洗浄した(3回)。300nM 4,6-ジアミジノ-2-フェニルインドール(DAPI、invitrogen)を用いて、室温にて30分間核を染色し、PBSで5分間洗浄した(3回)。観察は、共焦点レーザー走査型顕微鏡(FV10i,Olympus)を用いて行った。
上記8-1と同様に、多電極アレイに転写したPMGI配向性ファイバーシート(AF、噴出時間:90秒、厚み:2μm)とゼラチンコーティングした電極(Flat)の上でそれぞれ14日間培養したヒトiPS(IMR90-1)由来心筋細胞に対して、心筋細胞特異的なマーカーとしてのβ-MHCおよびMLC2V、ならびに細胞骨格系マーカーとしてのActin(一次抗体:ヤギポリクローナル抗体(Santa Cruz, SC 1616)、二次抗体:Alexa Fluor 488結合型抗ヤギIgG(Jackson ImmnoResearch #705-546-147))について、免疫染色を行った(図9D)。PLGA配向性ファイバーシートを用いた場合と同様に、β-MHCの発現は、PMGI配向性ファイバーシート上の細胞において、フラットな電極上の細胞と比較して非常に強いことが示された。また、MLC2VおよびActinについての染色像から、PMGI配向性ファイバーシート上の心筋細胞が配向性ファイバーシートに沿って配向していることが示された。この結果より、ファイバーシートが、心筋細胞の成熟度を亢進していることが示唆される。また、MLC2Vおよびアクチンの染色像から、心筋細胞は配向性ファイバーの方向に沿って配向していることが明らかになった。
9-1.PLGA配向性ファイバーシート上の心筋組織片の観察
ファイバーシートと心筋細胞との接着構造、配向構造および組織片の3次元構造を調べるために、透過型電子顕微鏡(TEM)による観察を行った。PLGA配向性ファイバーシートおよびPLGAランダムファイバーを接着させたプラスチック製切片および、ゼラチンでコーティングしたフラットな切片を作製した。実施例2の方法に準じて、iPS細胞由来心筋細胞を各切片の表面に播き、14日間培養を行った。NaHCa緩衝液(100mM NaCl、30mM HEPES、2mM CaCl2、pH7.4)中2%グルタルアルデヒドを用いて細胞を固定した。それぞれの試料を0.25%オスミウム、0.25%K4Fe(CN)6、1%タンニン酸、50mM 酢酸ウラニルで順番に処理し、後期固定処理を行った。エタノールシリーズで脱水処理を行い、65℃で重合処理を行った。ウルトラミクロトーム(Leica FC6,Vienna,AU)を用いて、厚みが60~100nmになるように、配向性ファイバーシートに対して垂直方向に凍結切片を作製し、断面を観察した。観察は、透過型電子顕微鏡(JEOL JEM1400、日本)を用いて行った。
PMGI配向性ファイバー(噴出時間:90秒、厚み:2μm)およびゼラチンコーティング上で心筋細胞を培養して作製した心筋組織片を用いて、上記9-1と同様に透過型電子顕微鏡(TEM)による観察を行った。その結果、PLGA配向性ファイバーシートを用いた場合と同様に、PMGI配向性ファイバー上で14日間培養した心筋細胞は、ファイバーに巻き付くように密着して結合していること、筋線維の配列構造はファイバーの方向性と一致していることが観察された(図10B、右)。さらに、PMGI配向性ファイバーシート上の心筋細胞は、厚みが約30μmであり、心筋細胞層がファイバーシート両面に形成された、約10層の多層構造を有していることも明らかになった(図10B、左)。
A.HERGチャネル阻害剤(E-4031)添加時のQT間隔延長
実施例3および4の方法に準じて、PLGA配向性ファイバーシート(AF、噴出時間:40秒、厚み:2μm、密度:300本/mm)およびランダムファイバーシート(RF、噴出時間:20秒、厚み:2μm)をそれぞれ転写した多電極アレイならびにゼラチンコーディングした多電極アレイ(Flat)上で培養した心筋細胞に、代表的なヒト遅延整流性カリウムイオンチャネル遺伝子(HERG)チャネル阻害剤であるE-4031を添加し、QT間隔の延長を5~10分間連続して調べた。hERGチャネルは、心臓内の内向き整流性電位開口型カリウムチャネルであって、遮断することによって、QT間隔延長が観察されることが知られている。使用濃度が増加、また細胞成熟度が低いすることによって、不整脈が起こされる。E-4031の添加により、ファイバーの構造およびファイバーの有無によらず、E-4031の濃度依存的にQT間隔の延長が観察された(図11A)。
E-4031投入後5~10分間、心筋細胞からの電気信号を測定している間における、不整脈の有無を調べた。その結果、薬剤投入後のフラットな電極上の心筋細胞からは、ファイバーシート上の細胞と比較して不整脈が高い頻度で観察された(図11B~Cおよび表1)。これは、フラットな多電極アレイ上では細胞と基板の接着性が低く、また細胞の成熟度が低い(図9A)ためと考えられる。
A.細胞培養部が配向性ファイバーシートである細胞培養用シートの製造
実施例1の方法に準じて、エレクトロスピニング法でPLGA配向性ファイバーシート(噴出時間:10分、厚み:10μm、密度:10000本/mm)を作製し、PDMSで作製したスペーサー(1.8 cmx1.8cm)上に転写した。さらに、配向性ファイバーシートと同じPLGA溶液にて転写部分を塗布してフレーム部分を形成し、一晩乾燥させてUV照射により滅菌した後に細胞培養に用いた(図12A)。
移植を目的として組織片を製造する場合、安全性の高い生体内分解材料を用いて配向性ファイバーを作製することが望ましい。PLA(ポリ乳酸)とPGA(ポリグリコール酸)のコポリマーであるPLGAは、毒性のない生体分解性素材である。PLAとPGAの組み合わせ比率によって、分解スピードを調節することが可能である。上記Aに準じて作製したPLGA(PLA75%:PGA25%)配向性ファイバーの分解時間を培養条件下で調べた。実施例2の方法に準じて心筋細胞を播いてから、3ヶ月後には、PLGAファイバー(噴出時間:10分、厚み:10μm、密度:10000本/mm)が完全に分解されている様子が観察された(図12E)。
A.心筋細胞組織片のマウス心臓への接着
配向性ファイバーシート上で製造した心筋組織とマウス摘出心臓との接着性を調べた。実施例11、Aの方法に準じて作製した、PLGA製フレームを有するPLGA配向性ファイバーシート(噴出時間:10分、厚み:10μm、密度:10000本/mm)上で、実施例2の方法に準じて心筋細胞を6日間培養し、培養組織片を作製した。該培養組織片をPLGA配向性ファイバーシートとともに剥がし、生後3ヶ月のマウスの心臓の表面に該ファイバー心筋組織片を静置した。静置から3時間後には、該心臓組織片が心臓膜に接着している様子が観察された(図13A)。ピンセットで該心筋組織片を持ち上げても、マウス心臓から剥がれることはなかった。また、PDMS製のフレーム部分を有するPLGA配向性ファイバーシート上で同様に心筋組織片を作製し、該フレーム部分の三辺を切り落として、細胞培養部分のみを同様にマウスの心臓表面に静置した場合にも、心筋組織片の心臓表面への接着が観察された(図13B)。
上記Aと同様に、PLGA製のフレーム部分を有するPLGA配向性ファイバーシート(噴出時間:10分、厚み:10μm、密度:10000本/mm)上で作製した心筋組織片を、細胞培養開始6日後に、ヌードラットの心臓表面に移植した。移植2週間後、心臓を回収して、4%パラホルムアルデヒドで固定して脱水した後、パラフィン包埋して2μm切片を作製した。該切片の脱パラフィン処理を行った後、ヘマトキシリン・エオジン染色、ヒト特異的プローブによるIn situ hybridization(ISH)および心筋マーカーであるcTnTについての組織染色を行い、顕微鏡観察を行った(図13C)。その結果、移植された心筋組織片は宿主の心臓表面に生着していること、宿主心臓表面にヒトiPS細胞由来の心筋細胞が存在していることが明らかになった。
A.配向性ファイバー上の心筋組織片の二層化
配向性ファイバー上の心筋組織片を重ねることにより、さらに厚みのある心筋組織片を作製することが可能かどうかを検討した。実施例11の方法に準じて、PDMSスペーサーに接着した、PLGA製のフレーム部を有するPLGA配向性ファイバーシートを作製した(噴出時間:10分、厚み:10μm)。実施例2の方法に準じて、該PLGA配向性ファイバーシート上にiPS細胞(IMR90-1)由来心筋細胞を播種し、培養した。培養開始後14日目に、PLGA配向性ファイバーシートの両面に形成された心筋移植片を、配向方向が同一となるよう2層に重ねた(図14A、上段)。重ねた移植片は2時間後にはお互いに接着し、その接着強度はピンセットでは容易に剥がせない程度の強さであった(図14A、下段)。この結果から、配向性ファイバー上に形成した心筋組織片は容易に多層化できることが明らかになった。
上記Aで得た多層のPLGA配向性ファイバー上心筋組織片の機能を評価するために、該組織片をPDMSスペーサーから剥離して多電極アレイ(30μm spaced 200μm、8×8 grid array)上におき、電気信号を測定した。その結果、二層の心筋移植片は、重ねて1分後の時点では、それぞれの心筋移植片は単独で拍動していたが、3時間後には、二層の心筋組織片は同期して拍動することが確認された(図14B)。

- 配向性ファイバーシートと、該配向性ファイバー上の培養心筋細胞とを含む、心筋組織片。
- 配向性ファイバーシートが培養心筋細胞層の間に介在している、心筋組織片。
- 該心筋細胞が多能性幹細胞から誘導された心筋細胞である請求項1または2に記載の心筋組織片。
- 配向性ファイバーシートにおけるファイバーの直径が0.1μm~5μm、シートの幅1mm当たりのファイバーの本数30~15000本、シートの厚みが0.1μm~20μmである、請求項1~3いずれかに記載の心筋組織片。
