(上記式中、3つのR2は、各々独立して、水素原子、又は、炭素数1~5の直鎖もしくは分岐鎖状のアルキル基のいずれかであることを表す。)
反応点が嵩高い3級アルコール3aの場合、反応の完結には過剰の反応剤(3当量のNAPOMCl及び6当量のDIPEA)と反応時間の延長(32.5 h)を要したが別段大きな問題を生じることなく、室温下で望む生成物が得られた。また、チオール6aに関しても同様にNAPOM保護体6bに導くことができた(88%, エントリー6)。
一方NAPOMスルフィド6bに対してDDQを作用させたところ、NAPOM基の除去により生じたチオール6a同士が酸化的条件下ジスルフィドを形成した6cが94%の収率で得られることが分かった(エントリー6。なおジスルフィド(6c)からチオール(6a)への変換は文献既知(非特許文献6参照)である)。これらの結果はNAPOM基が、酸化的条件で除去できる新規BOM系保護基となり得ることを示している。
そこで種々反応条件を検討したところ(例えば塩基としてDMAPを用いたところ、アシル転位が進行した)、DIPEAの代わりに2,6-ルチジンとテトラブチルアンモニウムヨージド(TBAI)を用いることで(エントリー2)、アシル転位反応を3%にまで抑制し、望む8aを89%の収率で得ることに成功した。本法は反応性の高いクロロ基を有するために、Ag2Oを用いるアルキル化法が適用できない、アルコール9のNAPOM化にも適用できた(エントリー3、75%)。
アルコール 1a (220 mg, 1.61 mmol) のジクロロメタン(19 mL)溶液に、細かく粉砕した塩化カルシウム (220 mg, 1.98 mmol)及びエチルジイソプロピルアミン(1.13 mL, 6.48 mmol)を加え、室温下30分撹拌した。NAPOM クロリド(92%, 667 mg, 2.97 mmol)を加え、さらに室温下6.5時間撹拌した。0 ℃に冷却し、エーテルで希釈後、水を加えて反応を停止させた。得られた混合物をエーテルで3 回抽出し、有機相を合わせて飽和硫酸水素カリウム水溶液と飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別後、ろ液を濃縮して得られた粗生成物をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル = 50/1 → 30/1)にて精製することで、NAPOMエーテル 1b (450 mg, 1.47 mmol, 91%)を無色油状物として得た。NAPOMエーテル 1b: 1H-NMR (400 MHz, CDCl3): δ 7.84-7.80 (m, 4H) , 7.52-7.44 (m, 3H) , 7.29-7.26 (m, 2H) , 7.20-7.17 (m, 3H), 4.82 (s, 2H) , 4.78 (s, 2H) , 3.65 (t, J = 6.4 Hz, 2H) , 2.72 (t, J = 7.8 Hz, 2H) , 1.95 (tt, J = 7.8, 6.4 Hz, 2H)。

ベンジルアルコール 9 (235 mg, 1.50 mmol)のジクロロメタン(15 mL)溶液に、細かく粉砕した塩化カルシウム(235 mg, 2.12 mmol)と2,6-ルチジン(1.39 mL, 18.0 mmol)を加えて30分撹拌した。0 ℃に冷却し、NAPOMクロリド(92%, 2.02 g, 9.00 mmol)を加え、さらに室温下16時間撹拌した。0 ℃に冷却し、エーテルで希釈後、飽和硫酸水素カリウム水溶液を加えて反応を停止させた。得られた混合物をエーテルで3回抽出し、有機相を合わせて飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別後、ろ液を濃縮して得られた粗生成物をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル = 50/1 → 30/1 → 20/1)にて精製することで、NAPOMエーテル 10a (369 mg, 1.13 mmol, 75%)を無色の粉末状固体として得た。NAPOMエーテル 10a: 1H-NMR (400 MHz, CDCl3) : δ 7.85-7.80 (m, 4H), 7.51-7.45 (m, 3H), 7.39-7.34 (m, 4H), 4.89 (s, 2H), 4.81 (s, 2H), 4.69 (s, 2H), 4.59 (s, 2H)。
NAPOMエーテル 8a (298 mg, 0.850 mmol)のメタノール(5.6 mL)溶液を0 ℃に冷却し、ナトリウムメトキシド(91.8 mg, 1.70 mmol) を加えた後、室温下で2.5時間撹拌した。0 ℃に冷却し、エーテルで希釈後、水を加えて反応を停止させた。得られた混合物をエーテルで3回抽出し、有機相を合わせて飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別後、ろ液を濃縮することでモノNAPOMエーテル11 (257.9 mg, 0.84 mmol)を無色の粉末状固体として得た。モノNAPOMエーテル 11: 1H-NMR (400 MHz, CDCl3): δ 7.85-7.83 (m, 3H), 7.51-7.44 (m, 3H), 7.79 (s, 1H), 7.39-7.27 (m, 5H), 4.92-4.86 (m, 3H), 4.79 (d, J = 12.1 Hz, 1H), 4.76 (d, J = 12.1 Hz, 1H), 3.85 (dd, J = 10.5, 3.2 Hz, 1H), 3.67 (dd, J = 10.5, 8.7 Hz, 1H), 2.97 (d, J = 2.3 Hz, 1H)。

モノNAPOMエーテル11 (30.4 mg, 0.0986 mmol)をジクロロメタン-水 2 : 1 混合溶媒(1.5 mL)に溶かし、0 ℃下2,3-ジクロロ-5,6-ジシアノ-p-ベンゾキノン(DDQ, 35 mg, 0.15 mmol)を加え室温下で2.5時間激しく撹拌した。その後メタノール(0.5 mL)を加え更に4時間撹拌した。0 ℃に冷却し、酢酸エチルで希釈後、飽和炭酸水素ナトリウム水溶液と飽和チオ硫酸ナトリウム水溶液を加えて反応を停止させた。得られた混合物を酢酸エチルで3回抽出し、有機相を合わせて飽和塩化ナトリウム水溶液で洗浄後、 無水硫酸ナトリウムで乾燥した。乾燥剤をろ別後、ろ液を濃縮して得られた粗生成物をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル = 4/1 → 2/1 → 1/1)にて精製することで、ジオール 12a (12.2 mg, 0.0883 mmol, 90%)を無色の粉末状固体として得た。
NAPOMエーテル 1b (153 mg, 0.50 mmol)及びPMBエーテル14 (135 mg, 0.50 mmol) をアセトン-水 9 : 1 混合溶媒(5 mL)に溶かし0 ℃下、硝酸セリウムアンモニウム(CAN, 822 mg, 1.50 mmol)を加え、 室温下で6時間撹拌した。0 ℃に冷却し、エーテルで希釈後、飽和炭酸水素ナトリウム水溶液と飽和チオ硫酸ナトリウム水溶液を加えて反応を停止させた。
得られた混合物をエーテルで3回抽出し、有機相を合わせて飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別後、ろ液を濃縮して得られた粗生成物をシリカゲルカラムクロマトグラフィー (ペンタン/エーテル = 20/1 → 10/1 → 3/1)にて精製することで、アルコール 1a (76.9 mg, 0.565 mmol, 定量的)を淡黄色油状物として得た。同時に未反応のNAPOMエーテル 1b (165 mg, 0.538 mmol, 定量的)が回収された。

2-ナフタレンメタノール 17 (24.0 g, 0.152 mol)のペンタン(250 mL)とジクロロメタン(50 mL)の混合溶液中にパラホルムアルデヒド(5.39 g, 0.152 mol)を加え、 -20 ℃に冷却した。別のフラスコに量り取った無水硫酸マグネシウム (300 g)に、臭化水素酸 (48%水溶液, 140 mL, 1.23 mol)を滴下漏斗を用いてゆっくり加えることで臭化水素ガスを発生させた。発生した臭化水素ガスをガラス管を通して反応溶液に吹き込みながら、-20℃下5時間撹拌した。その後アルゴンガスを1時間吹き込み、反応溶液をアルゴン雰囲気下でろ過した。ろ液を濃縮し乾燥することで、2-NAPOMブロミド 18 (12.1 g, 0.0481 mol, 32%)を白色固体として得た。2-NAPOMブロミド 18: 1H-NMR (400 MHz, CDCl3): δ7.88-7.80 (m, 4H), 7.53-7.45 (m, 3H), 5.75 (s, 2H), 4.89 (s, 2H).
アルコール 3a (82.1 mg, 0.500 mmol)のトルエン(1 mL)溶液に、エチルジイソプロピルアミン(0.513 mL, 3.00 mmol)及び2-NAPOM ブロミド(377 mg, 1.50 mmol)を加え、室温下15.5時間撹拌した。0℃に冷却し、エーテルで希釈後、水を加えて反応を停止させた。得られた混合物をエーテルで3回抽出し、有機相を合わせて飽和硫酸水素カリウム水溶液と飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別後、ろ液を濃縮して得られた粗生成物をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル = 50/1 → 30/1)にて精製することで、NAPOMエーテル 3b (125 mg, 0.374 mmol, 75%)を無色油状物として、また、原料3a (8.6 mg, 0.0524 mmol, 10%)を無色油状物として得た。
CBr4 (6.0 mg, 0.018 mmol)を2―ナフチルメチル3―フェニルプロピルエーテル(27.6 mg, 0.100 mmol)及び2―ナフチルメトキシメチル 3―フェニルプロピル エーテル(30.6 mg, 0.100 mmol)を、メタノール(MeOH) 溶液(500μL)とTHF溶液(50μL)の混合溶液に0℃で加えた。還流条件下で10時間撹拌した後、反応液をエーテルで希釈し、飽和炭酸水素ナトリウム水溶液を0℃下で加えて反応を停止させた。得られた混合物をエーテルで抽出し、有機相を合わせて飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別し、ろ液を減圧下で濃縮して得られた粗生成物を、シリカゲルカラムクロマトグラフィー (ペンタン/エーテル = 50/1 → 30/1 → 10/1 → 5/1 →1/1)にて精製することで、NAPエーテル(26.6 mg, 0.0962 mmol, 96%)及び3―フェニル―1―プロパノール(12.8 mg, 0.0940 mmol, 94%)を得た。
2―ナフチルメトキシメチル 3―フェニルプロピル エーテル(153 mg, 0.500 mmol)及びp―メトキシベンジル3―フェニルプロピルエーテル(135 mg, 0.500 mmol)を、アセトン (4.5 mL) -水(0.5 mL)混合溶媒に溶かし0℃下、硝酸セリウムアンモニウム(CAN, 548 mg, 1.00 mmol) を加えた。混合液を0℃から常温まで1時間激しく撹拌した後、さらにCAN(137 mg, 0.25 mmol) を0℃で加えた。さらに混合液を0℃から常温まで1.5時間激しく撹拌した後、反応液をエーテルで希釈し、飽和炭酸水素ナトリウム水溶液と飽和チオ硫酸ナトリウム水溶液を加えて反応を停止させた。得られた混合物をエーテルで抽出し、有機相を合わせて飽和塩化ナトリウム水溶液で洗浄後、無水硫酸ナトリウムで乾燥した。乾燥剤をろ別し、ろ液を減圧下で濃縮して得られた粗生成物を、シリカゲルカラムクロマトグラフィー (ペンタン/エーテル = 20/1 → 10/1 → 3/1)にて精製することで、NAPOMエーテル(165 mg、定量的(quantitative))及び3―フェニル―1―プロパノール(76.9 mg、定量的(quantitative))を得た。
NAPOM(2-ナフチルメトキシメチル 3-フェニルプロピル) エーテル(153 mg, 0.500 mmol)及びPMB(p-メトキシベンジル 3-フェニルプロピル) エーテル(135 mg, 0.500 mmol)のメタノール(10 mL)溶液を10% Pd/C (26.6 mg)に加え、混合液を水素雰囲気下 (風船)、室温で2.25時間激しく撹拌した。反応液は、セライト(celite)パッドでろ過し、減圧下で濃縮した。ろ液を濃縮して得られた粗生成物をシリカゲルカラムクロマトグラフィー (ペンタン/エーテル = 50/1 → 30/1 → 3/1 → 1/1)にて精製することで、PMBエーテル (127 mg, 0.470 mmol, 94%)及び3-フェニル-1-プロパノール (80.6 mg, 定量的(quantitative))を得た。
Claims (6)
- ヒドロキシ基及び/又はメルカプト基を有する基質化合物の当該ヒドロキシ基及び/又はメルカプト基を保護する保護基導入剤であって、下記の式(I)で表されることを特徴とするヒドロキシ基及び/又はメルカプト基用保護基導入剤。
(上記式中、Aは、隣接するベンゼン環の2つの炭素原子を含んで環数1~5の環構造が形成されていることを表し、当該環構造は、置換もしくは無置換の5員環又は6員環からなり、複素環を含んでいてもよく、R1、R2、R3及びR4は、各々独立して、水素原子、又は、炭素数1~5の直鎖もしくは分岐鎖状のアルキル基であり、Xは、ハロゲン原子、あるいはOSO2R5(R5 =アリール基またはアルキル基)である。) - 請求項1又は請求項2に記載のヒドロキシ基及び/又はメルカプト基用保護基導入剤において、
前記基質化合物に、上記の式(I)で表される化合物を、塩基の共存下反応させることにより、前記基質化合物に含まれるヒドロキシ基及び/又はメルカプト基を保護することを特徴とするヒドロキシ基及び/又はメルカプト基用保護基導入剤。
