Description
従来、α-フルオロアクリル酸エステルの、収率が良好である製法としては、例えば、α-フルオロホスホノアセテートとパラフォルムアルデヒドとの縮合によってα-フルオロアクリル酸エステルが得られる方法であって、当該縮合が弱無機塩基の存在下で水性媒質中で実施されることを特徴とする方法(特許文献1)が提案されている。
また、特に、医薬製造用の合成中間体等の場合、医薬の安全性の観点から、副生成物の含量が極めて低いことが望ましい。このため、α-フルオロアクリル酸エステルの選択率が非常に高いことが求められる。しかし、一般に、2-フルオロアクリル酸類の製造方法では、誘導体等の発生により、反応が複雑になり、2-フルオロアクリル酸類の収率は低く、その分離も困難である。このため、副生成物を除去して2-フルオロアクリル酸類の純度を高くするためには、大量の廃液や廃棄物が発生し、工業化生産には不利である。
本発明者らは、高い原料転化率、高い選択率、及び高い収率を有する、α-フルオロアクリル酸エステルの製造方法として、

製造方法を開発し、これを出願した(PCT/JP2013/073446、未公開)。
しかし、本発明者らは、更に、少ない触媒使用量で、高い原料転化率、及び高い収率を有する、α-フルオロアクリル酸エステル類の製造方法を提供することを目指した。

本明細書中、「フルオロアルキル基」は、少なくとも1個の水素原子がフッ素原子で置換されたアルキル基である。「フルオロアルキル基」は、パーフルオロアルキル基を包含する。「パーフルオロアルキル基」は、全ての水素原子がフッ素原子で置換されたアルキル基である。
本明細書中、「アルコキシ基」は、アルキル-O-基である。
本明細書中、「アシル基」としては、例えば、アルカノイル基(すなわち、アルキル-CO-基)等が挙げられる。

工程Aの反応原料としての前記式(3)で表されるアルコールの量は、前記式(2)で表される化合物1モルに対して、通常1~500モル、好ましくは約1.1~50モルである。
前記式(3)で表されるアルコールを工程Aの反応の溶媒としても用いる場合、当該アルコールは、通常、前記式(2)で表される化合物に対して大過剰に用いられる。具体的には、当該アルコール以外の溶媒を用いない場合、前記式(2)で表される化合物1モル当たり、当該アルコールの量は、通常0.1~20L、好ましくは約0.2~5Lであり、又は0.5~10L、若しくは約1~5Lであることもできる。
すなわち、工程Aで用いられる遷移金属錯体触媒としては、例えば、ニッケル錯体触媒、パラジウム錯体触媒、白金錯体触媒、ロジウム錯体触媒、ルテニウム錯体触媒、イリジウム錯体触媒及びコバルト錯体触媒が挙げられる。当該パラジウム錯体触媒は、好ましくは0価のパラジウム錯体触媒、又はII価パラジウム錯体触媒である。
当該遷移金属は、好ましくは、ニッケル、コバルト及びパラジウムからなる群より選択され、特に好ましくは、パラジウムである。
当該「酸素原子を有する連結基」としては、例えば、-O-、及び-(CH2)n1-O-(CH2)n2-[式中、n1、及びn2は、同一又は異なって0~6の整数を表す。]等の2価の基が挙げられる。
当該芳香環としては、例えば、ベンゼン環が挙げられる。
当該2個の芳香環は、前記「酸素原子を有する連結基」に加えて別の連結基で更に互いに連結されていてもよい。
当該2個の芳香環は、好ましくは、それぞれ、その1個の環構成原子上に1個のリン原子を有する。

このような不均一系触媒は、触媒の回収等のプロセス上の利点を有する。具体的な触媒構造としては、例えば、次の化学式:
に示すような、架橋したポリスチレン(PS)鎖にホスフィンを導入したポリマーホスフィンなどで前記遷移金属原子を固定したもの等が挙げられる。
この例における、「二座ホスフィン配位子」は、以下の化学式に示す、トリフェニルホスフィンの1つのフェニル基をポリマー鎖に結合させたトリアリールホスフィンである。
当該担体の例としては、例えば、炭素、アルミナ、シリカ-アルミナ、シリカ、炭酸バリウム、硫酸バリウム、炭酸カルシウム、酸化チタン、酸化ジルコニウム、及びゼオライト等が挙げられる。
遷移金属触媒の量の下限は、前記式(2)で表される化合物1モルに対して、通常0.000001モル、0.00001モル、より好ましくは0.00002モル、又は0.00004モルである。
工程Aで用いられる塩基としては、例えば、アミン、無機塩基、及び有機金属塩基が挙げられる。
アミンとしては、例えば、トリエチルアミン、トリ(n-プロピル)アミン、トリ(n-ブチル)アミン、ジイソプロピルエチルアミン、シクロヘキシルジメチルアミン、ピリジン、ルチジン、γ-コリジン、N,N-ジメチルアニリン、N-メチルピペリジン、N-メチルピロリジン、N-メチルモルホリン、1,8-ジアザビシクロ[5,4,0]-7-ウンデセン、1,5-ジアザビシクロ[4,3,0]-5-ノネン、及び1,4-ジアザビシクロ[2,2,2]オクタン等が挙げられる。
無機塩基としては、例えば、水酸化リチウム、水酸化カリウム、水酸化ナトリウム、水酸化カルシウム、炭酸リチウム、炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、及び炭酸水素カリウム等が挙げられる。
有機金属塩基としては、例えば、
ブチルリチウム、t-ブチルリチウム、フェニルリチウム、トリフェニルメチルナトリウム、エチルナトリウム等の有機アルカリ金属化合物;
メチルマグネシウムブロミド、ジメチルマグネシウム、フェニルマグネシウムクロリド、フェニルカルシウムブロミド、ビス(ジシクロペンタジエン)カルシウム等の有機アルカリ土類金属化合物;及び
ナトリウムメトキシド、t-ブチルメトキシド等のアルコキサイド
等が挙げられる。
塩基の好ましい例としては、水酸化リチウム、トリエチルアミン、炭酸カリウム、及び炭酸リチウムが挙げられる。塩基のより好ましい例としては、トリエチルアミン、炭酸カリウム、及び炭酸リチウムが挙げられる。塩基の特に好ましい例としては、トリエチルアミンが挙げられる。
塩基は、単独で、または2種以上を組み合わせて用いることができる。
当該温度が低すぎる場合、原料転化率、及び収率が低くなる傾向がある。
一方、当該温度が高すぎる場合、後記の分析方法による分析において、工程Aの反応後の混合物中に、原料である前記式(1)で表される化合物、及び副生成物又は分解物が観測される場合がある。
[分析方法]
反応終了後、内部標準物質としてヘキサフルオロベンゼンを加えて撹拌し、しばらくの間静置して塩を沈殿させる。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施する。
当該溶媒としては、例えば、ペンタン、ヘキサン、ヘプタン、オクタン、シクロヘキサン、デカヒドロナフタレン、n-デカン、イソドデカン、トリデカン等の非芳香族炭化水素溶媒;ベンゼン、トルエン、キシレン、テトラリン、ベラトロール、ジエチルベンゼン、メチルナフタレン、ニトロベンゼン、o-ニトロトルエン、メシチレン、インデン、ジフェニルスルフィド等の芳香族炭化水素溶媒;アセトン、メチルエチルケトン、メチルイソブチルケトン、アセトフェノン、プロピオフェノン、ジイソブチルケトン、イソホロン等のケトン;ジクロロメタン、クロロホルム、クロロベンゼン等のハロゲン化炭化水素溶媒;ジエチルエーテル、テトラヒドロフラン、ジイソプロピルエーテル、メチル t-ブチルエーテル、ジオキサン、ジメトキシエタン、ジグライム、フェネトール、1,1-ジメトキシシクロヘキサン、ジイソアミルエーテル等のエーテル溶媒;酢酸エチル、酢酸イソプロピル、マロン酸ジエチル、3-メトキシ-3-メチルブチルアセテート、γ-ブチロラクトン、エチレンカーボネート、プロピレンカーボネート、炭酸ジメチル、α-アセチル-γ-ブチロラクトン等のエステル溶媒;アセトニトリル、ベンゾニトリル等のニトリル溶媒;ジメチルスルホキシド、スルホラン等のスルホキシド系溶媒;及びN,N-ジメチルホルムアミド、N,N-ジメチルアセトアミド、N-メチルピロリドン、1,3-ジメチル-2-イミダゾリジノン、N,N-ジメチルアクリルアミド、N,N-ジメチルアセトアセトアミド、N,N-ジエチルホルムアミド、N,N-ジエチルアセトアミド等のアミド溶媒等が挙げられる。
当該溶媒は、単独で、または2種以上を組み合わせて用いることができる。
一方、前記式(1)で表される化合物の沸点が高い場合、低沸点の溶媒を用いて、好適に、前記式(1)で表される化合物を精製することができる。
脱水処理を行わない化合物若しくは試薬、及び/又は溶媒を用いた場合、α-フルオロアクリル酸類が副生することにより、目的物であるα-フルオロアクリル酸エステル類の収率及び選択率が低下する虞がある。
脂肪族第一級アミン、脂肪族第二級アミン、脂肪族第三級アミン、脂環式第二級アミン、脂環式第三級アミン、芳香族アミン、複素環式アミン、及びポリマー担持アミン化合物(ポリマー型アミン化合物)等のアミン化合物;
アンモニア;
テルペン化合物;並びに
酸素原子及び硫黄原子からなる群より選択される1個以上の原子を有する化合物
等が挙げられる。
脂肪族第一級アミンとしては、例えば、メチルアミン、エチルアミン、プロピルアミン、ブチルアミン、ペンチルアミン、ヘキシルアミン、シクロヘキシルアミン、及びエチレンジアミンが挙げられる。
脂肪族第二級アミンとしては、例えば、ジメチルアミン、ジエチルアミン、ジプロピルアミン、ジブチルアミン、ジペンチルアミン、ジヘキシルアミン、及びジシクロヘキシルアミンが挙げられる。
脂肪族第三級アミンとしては、例えば、トリメチルアミン、トリエチルアミン、ジイソプロピルエチルアミン、トリ(n-ブチル)アミン、及びN,N,N’,N’-テトラメチルエチレンジアミンが挙げられる。
脂環式第二級アミンとしては、例えば、ピペリジン、ピペラジン、ピロリジン、モルホリンが挙げられる。
脂環式第三級アミンとしては、例えば、N-メチルピペラジン、N-メチルピロリジン、1,5-ジアザビシクロ[4,3,0]-5-ノネン、及び1,4-ジアザビシクロ[2,2,2]オクタンが挙げられる。
芳香族アミンとしては、例えば、アニリン、メチルアニリン、ジメチルアニリン、N,N-ジメチルアニリン、ハロアニリン、及びニトロアニリンが挙げられる。
複素環式アミンとしては、例えば、ピリジン、メラミン、ピリミジン、ピペラジン、キノリン、及びイミダゾールが挙げられる。
ポリマー担持アミン化合物(ポリマー型アミン化合物)としては、例えば、ポリエチレンイミン、ポリアリルアミン、及びポリビニルピリジンが挙げられる。
テルペン化合物としては、例えば、α-ピネン、カンフェン、α-テルピネン、D-リモネン、γ-テルピネン、p-シメン、及びテルピノーレン等が挙げられる。
酸素原子及び硫黄原子からなる群より選択される1個以上の原子を有する化合物としては、1個以上の芳香性6員炭素環、並びに当該1個以上の芳香性6員炭素環の1個以上の環構成炭素原子に直接結合している酸素原子又は硫黄原子を有する炭素数6~20の化合物が好ましい。その例としては、具体的には、例えば、ヒドロキノン、4-メトキシフェノール、2,5-ジ-tert-ブチルヒドロキノン、メチルヒドロキノン、tert-ブチルヒドロキノン(TBH)、p-ベンゾキノン、メチル-p-ベンゾキノン、tert-ブチル-p-ベンゾキノン、2,5-ジフェニル-p-ベンゾキノン、2,6-ジ-tert-ブチル-4-メチルフェノール(BHT)、及びフェノチアジン等が挙げられる。
当該反応時間は、より高い反応温度を採用することにより、より短くすることができる。
特に、本発明の製造方法では、副生成物及び分解物が極めて微量であるので、蒸留等の簡便な方法により、極めて純度の高い式(1)で表される化合物を得ることができる。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 9.45g(75.64mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロビス(トリフェニルホスフィン)パラジウム(II) 50.5mg(0.072mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で14時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが53.02mmol(収率70.1%)及び未反応の1-ブロモ-1-フルオロエテンが20.88mmol(回収率27.6%)であった。
転化率は72.5%、選択率は96.7%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.76g(70.11mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[ビス(ジフェニルホスフィノフェニル)エーテル]パラジウム(II) 51.5mg(0.072mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で13時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが60.08mmol(収率85.7%)及び未反応の1-ブロモ-1-フルオロエテンが9.11mmol(回収率13.0%)であった。
転化率は86.8%、選択率は98.7%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.88g(71.07mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 54.4mg(0.072mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で8時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが64.03mmol(収率90.1%)及び未反応の1-ブロモ-1-フルオロエテンが5.83mmol(回収率8.2%)であった。
転化率は91.0%、選択率は99.0%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 9.11g(72.93mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[1,1’-ビス( ジシクロヘキシルホスフィノ)フェロセン]パラジウム(II) 54.4mg(0.072mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で12時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが65.27mmol(収率89.5%)及び未反応の1-ブロモ-1-フルオロエテンが7.07mmol(回収率9.7%)であった。
転化率は90.1%、選択率は99.3%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.80g(70.43mmol)、トリエチルアミン 8.01g(79.2mmol)、塩化パラジウム(II) 12.8mg(0.072mmol)、2,2’-ビス(ジフェニルホスフィノ)-1,1’-ビナフチル 4.5mg(0.072mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で14時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが54.65mmol(収率77.6%)及び未反応の1-ブロモ-1-フルオロエテンが14.23mmol(回収率20.2%)であった。
転化率は78.9%、選択率は98.3%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 9.14g(73.16mmol)、トリエチルアミン 8.01g(79.2mmol)、塩化パラジウム(II) 12.8mg(0.072mmol)、1,4-ビス(ジシクロヘキシルホスフィノ)ブタン 32.4mg(0.072mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で14時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが58.09mmol(収率79.4%)及び未反応の1-ブロモ-1-フルオロエテンが13.46mmol(回収率18.4%)であった。
転化率は81.0%、選択率は98.0%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.95g(71.63mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 2.7mg(0.0036mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で8時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが59.81mmol(収率83.5%)及び未反応の1-ブロモ-1-フルオロエテンが10.60mmol(回収率14.8%)であった。
転化率は84.9%、選択率は98.3%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 9.02g(72.19mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 54.4mg(0.072mmol)、及び予め脱水処理を施したエタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で8時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸エチルエステルが63.46mmol(収率87.9%)及び未反応の1-ブロモ-1-フルオロエテンが7.29mmol(回収率10.1%)であった。
転化率は88.6%、選択率は99.2%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.86g(70.91mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 54.4mg(0.072mmol)、メタノール 4.61g(0.144moL)、及び予め脱水処理を施したテトラヒドロフラン 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で7時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが60.42mmol(収率85.2%)及び未反応の1-ブロモ-1-フルオロエテンが9.43mmol(回収率13.3%)であった。
転化率は86.0%、選択率は99.1%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.94g(71.55mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 54.4mg(0.072mmol)、メタノール 4.61g(0.144moL)、及び予め脱水処理を施したN-メチルピロリドン 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で7時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが62.75mmol(収率87.7%)及び未反応の1-ブロモ-1-フルオロエテンが7.01mmol(回収率9.8%)であった。
転化率は89.1%、選択率は98.4%であった。
150mLのステンレス製オートクレーブに2-ブロモ-2-フルオロビニルベンゼン 7.12g(35.4mmol)、トリエチルアミン 4.01g(39.6mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 27.2mg(0.036mmol)、及び予め脱水処理を施したメタノール 18mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で9時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロ-3-フェニルアクリル酸メチルエステルが32.64mmol(収率92.2%)及び未反応の2-ブロモ-2-フルオロビニルベンゼンが1.88mmol(回収率5.3%)であった。
転化率は93.6%、選択率は98.5%であった。
150mLのステンレス製オートクレーブに1-(2-ブロモ-2-フルオロビニル)-4-メトキシベンゼン 8.11g(35.1mmol)、トリエチルアミン 4.01g(39.6mmol)、ジクロロ[4,5-ビス(ジフェニルホスフィノ)-9,9’-ジメチルキサンテン]パラジウム(II) 27.2mg(0.036mmol)、及び予め脱水処理を施したエタノール 18mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で9時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロ-3-(4-メトキシフェニル)アクリル酸エチルエステルが30.54mmol(収率87.0%)及び未反応の1-(2-ブロモ-2-フルオロビニル)-4-メトキシベンゼンが3.47mmol(回収率9.9%)であった。
転化率は89.3%、選択率は97.4%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.98g(71.87mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[ビス(ジフェニルホスフィノフェニル)エーテル]パラジウム(II) 51.5mg(0.072mmol)、及び脱水処理を施していないメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で13時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈し、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが50.93mmol(収率70.9%)、2-フルオロアクリル酸が10.13mmol(収率14.1%)及び未反応の1-ブロモ-1-フルオロエテンが8.12mmol(回収率11.3%)であった。
転化率は87.5%、選択率は81.0%であった。
150mLのステンレス製オートクレーブに1-ブロモ-1-フルオロエテン 8.89g(71.15mmol)、トリエチルアミン 8.01g(79.2mmol)、ジクロロ[ビス(ジフェニルホスフィノフェニル)エーテル]パラジウム(II) 51.5mg(0.072mmol)、2,6-ジ-tert-ブチル-4-メチルフェノール 75.0mg(0.34mmol)、及び予め脱水処理を施したメタノール 36mLを仕込み、一酸化炭素 1.0MPaGを導入し100℃で13時間撹拌した。
反応終了後、オートクレーブを冷却した後、未反応のガスをパージして開栓し、内部標準物質としてヘキサフルオロベンゼン 186mg(1.0mmol)を加えて撹拌し、しばらくの間静置して塩を沈殿させた。上澄みを重クロロホルムで希釈して、19F-NMR積分値による定量を実施したところ、2-フルオロアクリル酸メチルエステルが62.85mmol(収率88.3%)及び未反応の1-ブロモ-1-フルオロエテンが6.81mmol(回収率9.6%)であった。
転化率は90.4%、選択率は97.7%であった。
Claims (7)
- 式(1):
R1、及びR2は、同一又は異なって、アルキル基、フルオロアルキル基、1個以上の置換基を有していてもよいアリール基、ハロゲン原子、又は水素原子を表し;及び
R3はアルキル基、フルオロアルキル基、又は1個以上の置換基を有していてもよいアリール基を表す。]
で表される化合物の製造方法であって、

二座ホスフィン配位子を含有する遷移金属錯体触媒、及び塩基の存在下で、
式(3):

[式中の記号は前記と同意義を表す。]
で表されるアルコール及び一酸化炭素
と反応させて前記式(1)で表される化合物を得る工程Aを含む
製造方法。 - R1は、水素原子、又はアリール基であり、且つ
R2は、水素原子、又はアリール基である
請求項1に記載の製造方法。 - 前記遷移金属がパラジウムである請求項1又は2に記載の製造方法。
- 前記二座ホスフィン配位子が、酸素原子を有する連結基で互いに連結された2個の芳香環を有する二座ホスフィン配位子である請求項1~3のいずれか1項に記載の製造方法。
- 前記遷移金属錯体触媒の量は、前記式(2)で表される化合物1モルに対して、0.001モル以下である請求項1~4のいずれか1項に記載の製造方法。
- 前記塩基がアミンである請求項1~5のいずれか1項に記載の製造方法。
- 工程Aが、60~120℃の範囲内の温度で実施される請求項1~6のいずれか1項に記載の製造方法。
