Abstract

 本発明の解決課題は、キサントフィロマイセス属酵母の培養物から、特殊な抽出設備、煩雑な精製工程を要することなく、また必ずしも人体に有害な有機溶媒を使用することなく、アスタキサンチンなどを高含量に含んだ天然物由来のカロテノイド組成物を効率的に工業生産できる製造法を提供することにある。 本発明に係るカロテノイド組成物の製造方法は、(1)カロテノイドを含有するキサントフィロマイセス属酵母を、含水率が3~50重量%の洗浄溶媒を用いて少なくとも1回洗浄する洗浄工程、(2)上記洗浄後のキサントフィロマイセス属酵母を、熱処理溶媒中にて65℃以上の高温条件で熱処理を施す熱処理工程、(3)上記熱処理後のキサントフィロマイセス属酵母を抽出溶媒と接触させ、10~70℃の温度で酵母中のカロテノイドを抽出する抽出工程、という3つの工程を含有する点に特徴を有する。

Description

カロテノイド組成物の製造方法
 本発明はカロテノイド組成物の製造方法に関する。さらに詳しくは、キサントフィロマイセス属酵母から、アスタキサンチンなどのカロテノイドを抽出することを特徴とする、カロテノイド組成物の製造方法に関する。
 カロテノイドは、自然界に広く分布する、黄色、橙色から赤色、または紫色を呈する天然色素として知られており、中でもその一種であるアスタキサンチンは、魚類、鶏卵の色揚げ剤として飼料用途に広く使われている。またアスタキサンチンは食品添加物としても認められており、油脂加工食品、蛋白質性食品、水性液状食品、健康食品などに幅広く使用されている。さらに近年、アスタキサンチンは、そのフリーラジカルによって誘起される脂質の過酸化に対する抗酸化活性、α-トコフェロールの数百倍に達する一重項酸素消去作用などの強力な抗酸化作用を利用し、体内の過剰な活性酸素を抑え、シミやしわの改善、白内障、動脈硬化や心臓病等の予防、免疫力強化やがんの予防等、化粧品類や機能性食品、医薬品としての用途が期待されている。
 アスタキサンチンは、サケ、マス、マダイ等の魚類、カニ、エビ、オキアミ等の甲殻類など広く自然界に分布すると共に、パラコッカス(Paracoccus)属、ブレビバクテリウム(Brevibacterium)属等に属する細菌類、ヘマトコッカス・プルビアリス(Hematococcus pluvialis)等の藻類、キサントフィロマイセス・デンドロアス(Xanthophyllomyces dendrorhous)(別名:ファフィア・ロードジーマ(Phaffia rhodozyma))等の酵母類といった微生物によっても生産される。アスタキサンチンやゼアキサンチン等のカロテノイドは、供給の安定性やコストの問題から現在では化学合成品が広く用いられている。しかしながら化学合成品には製法由来の不純物、とくに合成反応に用いられる人体に有害な劇薬類の混入の不安があり、安全性の面から天然由来の原料からなる製品の供給が望まれている。
 このような背景から特に、工業規模での大量生産に適していると考えられる天然物の藻類や微生物を利用した、アスタキサンチン等のカロテノイドの製造方法が数多く報告されている。例えば、ヘマトコッカス藻類の場合、培養した藻類のシスト細胞を熱アセトン処理し、夾雑物であるクロロフィルを溶出させた後、エタノールでアスタキサンチンを抽出する方法が報告されている(特許文献1)。しかしながら、同法で得られるカロテノイド組成物には多くの生体由来の夾雑物が含まれるため、産業上、十分なアスタキサンチン含量を含んだカロテノイド組成物を製造することは難しいことに加えて、製造に使用するアセトンを例えば食品添加物使用の規定値以下のレベルまで除去することは困難であるという問題がある。
 また、細胞壁を破壊したヘマットコッカス藻類を、水、グリセリン等の補助溶剤と混練して形成した成形体を抽出槽内に充填し、超臨界流体を供給してアスタキサンチンを抽出し、人体に有害な有機溶媒を使用することなく0.5~60%含量のアスタキサンチンを得る方法が報告されている(特許文献2)。しかしながら、同法では大量生産の際に特殊な設備が必要となるほか、十分な収率を達成するため長時間の抽出操作を要する上、高含量アスタキサンチンの取得には別途濃縮操作が必要になるなど、高含量のカロテノイド組成物を工業生産するには簡便性、経済性を満足できるものではない。
 キサントフィロマイセス属(ファフィア属)酵母を用いる方法として、該酵母の菌体をn-ヘキサン-エタノールの混合溶媒中、圧縮粉砕機で粉砕しつつ抽出し、抽出物を濃縮して得られた油状物質を-50℃の冷却状態で懸濁、濾過することにより遊離脂肪酸とトリグリセリドを除き、さらに色素油中の残存遊離脂肪酸を金属セッケン化して除くことでアスタキサンチンを高濃度化する方法が開示されている(特許文献3)。また、キサントフィロマイセス属(ファフィア属)酵母の破砕菌体を有機溶媒で抽出し、抽出液を濃縮して得られた油状の粗抽出エキスを、イオン交換クロマトグラフィー、吸着クロマトグラフィー等の精製手段でアスタキサンチンまたはそのエステルを得る方法が開示されている(特許文献4)。しかしこれらの方法はいずれも工程が複雑であり、特に後者では低濃度のアスタキサンチンの粗液を複数のカラムクロマトグラフィーにて精製を行うという煩雑な工程をとっているため、工業化が困難である。
 一方、アスタキサンチンを生産する新規な細菌(E-396株)を用い、低級アルコールまたは低級アルコールと水の混合物を用いて80℃以上の温度で抽出し、その後濃縮して得られた沈殿物を低級アルコール類で洗浄することにより、アスタキサンチン含量40%以上の乾燥物を得た例が報告されている(特許文献5、6)。同法ではエタノール等の安全性の高い溶媒と水のみで高含量のアスタキサンチン組成物の製造が可能であるものの、密閉式の圧力容器中で処理する必要があり、また、80℃を越える高温操作では、天然由来のフリー体トランス型アスタキサンチンの一部がシス型化する可能性があり、目的の構造を有するアスタキサンチンの精製収率が低下する問題がある。更に同法は、実際にその製造が確認された新規な細菌以外で、適用可能かどうか不明である。
 上記以外に、培養後のキサントフィロマイセス属(ファフィア属)酵母菌体をアセトンで抽出し、得られた抽出液を濃縮して得られる粗抽出物に炭化水素系溶剤を加え、晶析させる製造方法が報告されている(特許文献7)。同法は簡便性が高く、組成物中のアスタキサンチン含量が40%以上の高含量のものが得られるなどの利点があるが、製造過程で炭化水素系溶剤を使用するため、最終的に得られる組成物中にこれらの溶剤が残留する懸念がある。
 特に、キサントフィロマイセス属酵母は、乾燥菌体重量当たり25重量%前後もの脂質を含むことが知られており、該酵母から得られる抽出液の組成は、該脂質が主成分となるため、このような抽出液を濃縮して得られる濃縮物はべとついた粘性液となってしまう。従って、キサントフィロマイセス属酵母を使用して、アスタキサンチンが高濃度に含まれる組成物を得るためには、脂質を始めとする多くの夾雑物を製造中に除去する必要があり、その結果、上述のような複雑かつ非工業的な手段を伴うか、人体に有害で最終製品中の残存量に規制を伴うヘキサン等の有機溶媒を使用する必要があった。
 こうした状況から、取扱いが容易で、なおかつ残留溶剤の少ない、もしくは残留溶剤が問題とならない方法によりアスタキサンチンを高濃度で含有する天然由来のカロテノイド組成物を製造する方法が求められていた。

 上述したように、従来の天然由来のカロテノイド組成物の製造方法では、アスタキサンチン等を高濃度で含有する組成物を得るためには、抽出操作に特殊な設備が必要であったり、菌体由来の脂質等の夾雑物を除去するために複雑かつ非工業的な手段を要したりするなど、操作の煩雑化、コストの増大を招くという問題があった。さらには製造過程で炭化水素系溶剤など、食用用途に適さない有機溶媒を使用する場合は、残留溶剤の除去が問題となっていた。特に、脂質含量の多いキサントフィロマイセス属酵母では、細菌や藻類など他の原料を使用する場合に適切とされる製造方法をそのまま応用しても、高濃度のアスタキサンチンを得るための精製操作が困難であるか、可能でも非常に煩雑なものとなっており、規制の関係もあって、アスタキサンチンを高濃度に含有し、流動性に優れた油状組成物や粉体特性に優れた粉末組成物の製造は工業的レベルでは事実上不可能と考えられていた。例えば、上記特許文献5や6の方法を、本発明者がキサントフィロマイセス属酵母に実際に適用したところ、沈殿物中に多量の脂質が残存し、洗浄工程で濾過等の操作が行えず、満足な効率にて製造することが難しいことがわかった。
 本発明は、キサントフィロマイセス属酵母の培養物から、特殊な抽出設備や煩雑な精製工程を要することなく、また必ずしも人体に有害な有機溶媒を使用することなく、アスタキサンチンなどを高濃度に含んだ天然由来のカロテノイド組成物を効率的に工業生産できる製造法を提供することを目的とする。
 上記課題を解決するために鋭意検討を行った結果、キサントフィロマイセス属酵母からカロテノイドを抽出する前に、特定の条件で該酵母を洗浄し、かつ、洗浄後のキサントフィロマイセス属酵母に熱処理を施すことによって、当該酵母からアスタキサンチン等のカロテノイドを効率的に抽出できること、また該方法は、工業生産にも適していることを見出し、本発明を完成させた。
 すなわち本発明は、下記(1)~(3)の工程を有する、カロテノイド組成物の製造方法に関する。
(1)カロテノイドを含有するキサントフィロマイセス属酵母を、含水率が3~50重量%の洗浄溶媒を用いて少なくとも1回洗浄する洗浄工程
(2)上記洗浄後のキサントフィロマイセス属酵母を、熱処理溶媒中にて65℃以上の高温条件で熱処理を施す熱処理工程
(3)上記熱処理後のキサントフィロマイセス属酵母を抽出溶媒と接触させ、10~70℃の温度で酵母中のカロテノイドを抽出する抽出工程
 さらに本発明では、抽出工程(3)が、上記熱処理後のキサントフィロマイセス属酵母にさらに抽出溶媒を加えて行われるものであることが好ましく、カロテノイドがアスタキサンチンであることが好ましく、また、上記洗浄工程(1)において、キサントフィロマイセス属酵母を、含水率3重量%未満の抽出溶媒を用いてさらに洗浄することが好ましく、抽出工程(3)での抽出温度が10~45℃であるのが好ましい。
 抽出溶媒が、アルコール類、炭化水素類及びエーテル類からなる群より選択される少なくとも1種の有機溶媒であることが好ましい。
 また本発明は、熱処理溶媒、洗浄溶媒及び抽出溶媒が、いずれもエタノールであるのが好ましい。
 本発明の製造方法では、キサントフィロマイセス属酵母に含まれる脂質等の夾雑物の大半を事前に除去するだけでなく、さらに洗浄後の該酵母を熱処理することで、その後のカロテノイドの抽出、濃縮操作を効率良く実施でき、簡便で、工業生産にも適した、天然物由来のカロテノイド組成物の製造方法を提供できる。また本発明の製造方法では、食用用途に適用可能な安全な溶媒のみで操作することも可能であり、取扱いが容易で、なおかつ残留溶剤の少ない、もしくは残留溶剤が問題とならない方法にて製造することが可能である。
 以下に本発明の実施形態を詳細に説明する。
 本発明の製造方法は、(1)カロテノイドを含有するキサントフィロマイセス属酵母を、含水率が3~50重量%の洗浄溶媒を用いて少なくとも1回洗浄する洗浄工程、(2)上記洗浄後のキサントフィロマイセス属酵母を、熱処理溶媒中にて65℃以上の高温条件で熱処理を施す熱処理工程、(3)上記熱処理後のキサントフィロマイセス酵母を抽出溶媒と接触させ、10~70℃の温度で酵母中のカロテノイドを抽出する抽出工程、という3つの工程を実施することを特徴とする、カロテノイド組成物の製造方法である。
 本発明におけるカロテノイドとしては特に限定されないが、具体的には、αカロテン、βカロテン、γカロテン、δカロテン、リコペン等のカロテン類、ルテイン、ゼアキサンチン、カンタキサンチン、フコキサンチン、アスタキサンチン、アンテラキサンチン等のキサントフィル類等が挙げられ、特にその中でも、本発明の製造方法は、カロテノイドとして、特にアスタキサンチンを得るのに適している。この場合、得られるアスタキサンチンとしては、アスタキサンチン以外のカロテノイドとの混合物として得られる態様も含まれる。
 本発明の製造方法の原料として用いるキサントフィロマイセス属酵母は、カロテノイドを生産するものであれば特に限定されないが、特にカロテノイドとしてアスタキサンチンを製造するキサントフィロマイセス属酵母の好ましい具体例として、キサントフィロマイセス・デンドロアス(Xanthophyllomyces dendrorhous)(別名:ファフィア・ロードジーマ(Phaffia rhodozyma))等が挙げられる。
 なお、本発明においては、上記キサントフィロマイセス属酵母の野生株のみならず、例えば、上記キサントフィロマイセス属酵母の目的とするカロテノイド類の生合成に関与する遺伝子の転写及び翻訳活性、或いは発現蛋白質の酵素活性を、改変或いは改良した変異体や組換え体も好ましく使用することができる。
 本発明では、上記キサントフィロマイセス属酵母を培養することで、カロテノイドを産生させる。培養方法は特に限定されず、対象となるキサントフィロマイセス属酵母に適した、あるいは目的とするカロテノイドの生産に適した培養方法を適宜選択すればよい。培養に用いる炭素源としては、例えば、スクロース、グルコース、キシロース、マルトース、コーンシロップ、糖蜜、デンプン、グリセリン、エタノール、酢酸、乳酸、クエン酸、コハク酸等が挙げられるが、これらに限定されるものではない。これらの炭素源は単独で用いてもよく、菌体中のカロテノイド含有量の向上を目的として、2種以上組み合わせて用いてもよい。2種以上の炭素源を組み合わせて用いる場合、それらの炭素源を同時に培地中に添加してもよく、培養の途中で添加する炭素源を変更してもよい。後者の例として、培養の前半はグルコースを、培養の後半はキシロースを炭素源として培養する方法、同様に、前半はグルコース、後半はマルトースで培養する方法、前半はグルコース、後半はエタノールで培養する方法、前半はグルコース、後半はグリセリンで培養する方法、および、前半はグルコース、後半はクエン酸で培養する方法等が挙げられるが、これらの組み合わせに限定されず、如何なる炭素源の組み合わせでも、同様に実施しうる。培養に用いる窒素源としては、例えば、アンモニア、アンモニウム塩、尿素、アミノ酸類、酵母エキス、ペプトン、コーンスティープリカー等が挙げられるが、これらに限定されるものではない。これらの窒素源は単独で用いてもよく、炭素源の場合と同様に、2種以上組み合わせて用いてもよい。炭素源、窒素源以外の栄養素としては、例えば、リン酸等のリン源、マグネシウム、カリウム、ナトリウム、銅、鉄、亜鉛、マンガン、モリブデン等のミネラル類、ビオチン等のビタミン類等が挙げられるが、これらに限定されるものではない。炭素源、窒素源、および、その他の栄養素は、一括添加、分割添加、或いは連続添加等の方法で供給することができる。培養温度は特に限定されないが、15~26℃の範囲が好ましい。培養液のpHは特に限定されないが、3~7の範囲が好ましい。
 本発明の製造方法においては、上述したような培養によって得られるキサントフィロマイセス属酵母から、酵母細胞中のカロテノイドを抽出するに際して、培養後のキサントフィロマイセス属酵母をそのまま最初の洗浄工程に供することもできるが、所望により前記キサントフィロマイセス属酵母を破砕して細胞破砕物とし、該破砕物を洗浄工程に供し、引き続き熱処理工程を経た後に抽出を実施することもできる。従って、本発明の製造方法で用いられる「キサントフィロマイセス属酵母」としては、酵母細胞そのもの、凍結乾燥、噴霧乾燥等により乾燥したもの、以下に記載する細胞破砕処理を施したもののいずれもその対象である。特に酵母細胞の破砕は、キサントフィロマイセス属酵母中に生産・蓄積されたカロテノイドの効率的な抽出に寄与する。細胞破砕処理は、必ずしも必要ではないが、細胞が破砕されていないと、細胞中に生産・蓄積されたカロテノイドの回収効率は低下する。なお、本発明における酵母細胞の破砕は、目的とするカロテノイドの抽出が可能である程度に細胞壁等の表面構造が損傷を受ければよく、必ずしも細胞壁が完全に破れる、あるいは酵母細胞が断片化される必要はない。
 上記細胞破砕及び/又は後述の洗浄工程の対象となるキサントフィロマイセス属酵母の形態は、該酵母を含む培養液、その培養液を濃縮したもの、それら培養液から加圧濾過、減圧濾過、自然濾過、遠心分離、デカンテーション等の一般的な方法によりキサントフィロマイセス属酵母を湿菌体として採取したもの、該湿菌体を洗浄したもの、湿菌体を溶媒(例えば、水、生理食塩水、緩衝液等も含む)に懸濁したもの、前記湿菌体を乾燥させた乾燥菌体、これら乾燥菌体を溶媒(例えば、水、生理食塩水、緩衝液等も含む)に懸濁したもの等が挙げられ、そのいずれでもよい。また、上記濾過操作を行う場合、操作の効率化のために濾過助剤を使用してもよく、その場合、使用した濾過助剤を除去することなく湿菌体又は乾燥菌体として以降の操作を実施することもできる。上記細胞破砕の対象となるキサントフィロマイセス属酵母の形態は、好ましくはキサントフィロマイセス属酵母が水性の溶媒に懸濁している水性懸濁液であり、操作性等の面から、より好ましくは、キサントフィロマイセス属酵母を含む培養液そのもの、該培養液を濃縮したものや、キサントフィロマイセス属酵母の湿菌体を溶媒に懸濁させたものがキサントフィロマイセス属酵母の水性懸濁液として使用できる。
 上記キサントフィロマイセス属酵母の破砕は、以下の1つ、または幾つかの破砕方法を任意の順序で行うことにより行われる。破砕方法としては、例えば、物理的処理、化学的処理、酵素的処理の他、加熱処理、自己消化、浸透圧溶解、原形質溶解等を挙げることができる。
 上記物理的処理としては、例えば、高圧ホモジナイザー、回転刃式ホモジナイザー、超音波ホモジナイザー、フレンチプレス、ボールミル等の使用、あるいは、これらの組み合わせを挙げることができる。
 上記化学的処理としては、例えば、塩酸、硫酸等の酸(好ましくは強酸)を用いる処理、水酸化ナトリウムや水酸化カリウム等の塩基(好ましくは強塩基)を用いる処理等や、これらの組み合わせを挙げることができる。
 上記酵素的処理としては、例えば、リゾチーム、ザイモリアーゼ、グルカナーゼ、ノボザイム、プロテアーゼ、セルラーゼ等を用いる方法を挙げることができ、適宜これらを組み合わせて用いても良い。
 上記加熱処理としては、例えば、60~140℃で30分~3時間程度の処理を挙げることができる。
 上記自己消化としては、例えば、酢酸エチル等の溶媒による処理のほか、30~60℃で1~48時間程度、加温するといった処理等を挙げることができる。
 また、細胞内の塩濃度と異なる溶液で処理することにより、細胞の浸透圧溶解や原形質溶解を引き起こすこともできる。ただし、この方法のみでは細胞破砕効果が不十分な場合が多いため、上記のような物理的処理、化学的処理、酵素的処理、加熱処理、自己消化等と合わせて用いるのが好ましい。上記破砕方法の中でも、物理的処理、化学的処理(特に酸処理、好ましくは強酸(例えば、水溶液中におけるpKaが2.5以下の酸)や加熱処理が好ましく、破砕効率の点から物理的処理、化学的処理がより好ましい。
 本発明の製造方法における洗浄工程について説明する。本発明の製造方法においては、上記のようにして調製されたカロテノイドを含有するキサントフィロマイセス属酵母を、まず、含水率が3~50重量%の洗浄溶媒を用いて少なくとも1回洗浄を行う。ここで、洗浄は、カロテノイドを含有するキサントフィロマイセス属酵母を、湿菌体、乾燥菌体、または水性懸濁液とした状態で行うのが好ましい。また、その場合の湿菌体、乾燥菌体の含水率や、水性懸濁液中の菌体濃度は特に制限されないが、湿菌体であれば、含水率が通常20~75重量%の範囲、乾燥菌体であれば、含水率が通常0.01~20重量%の範囲が好ましく、また、水性懸濁液であれば、乾燥重量換算の菌体濃度として、通常1~25重量%の範囲のものが使用できる。なお、1回目の洗浄時のキサントフィロマイセス属酵母と洗浄溶媒の混合液の水分含量としては、10~40重量%の範囲が好ましく、15~35重量%の範囲がより好ましく、20~30重量%の範囲が特に好ましい。従って、そのような水分含量となるように、洗浄対象となるキサントフィロマイセス属酵母の形態とその水分量に応じて、洗浄溶媒の含水率や、その使用量などを適宜調整することもできる。
 本発明の製造方法において、洗浄工程で用いる洗浄溶媒としては、特に限定されないが、目的とするカロテノイドに対する洗浄条件での溶解度があまり高くないものが好ましい。そのような洗浄溶媒としては、例えば、ケトン類、アルコール類、炭化水素類、エーテル類、脂肪酸エステル類などの有機溶媒を、必要に応じて含水状態として、好ましく使用できる。好ましい有機溶媒は、ケトン類、アルコール類、エーテル類であり、特に好ましくはアルコール類である。
 上記ケトン類としては、特に制限されず、炭素数3~6のものが好適に用いられる。具体例としては、例えば、アセトン、メチルエチルケトン、メチルブチルケトン、メチルイソブチルケトン等を挙げることができる。好ましくは、アセトン、メチルエチルケトンであり、最も好ましくはアセトンである。
 上記アルコール類としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、飽和のものが好ましく用いられる。通常、炭素数1~20、好ましくは炭素数1~12、より好ましくは炭素数1~6である。なかでも、炭素数1~5の1価アルコール、炭素数2~5の2価アルコール、炭素数3の3価アルコールが好ましい。
 これらアルコール類の具体例としては、例えば、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール、1-ヘキサノール、2-メチル-1-ペンタノール、4-メチル-2-ペンタノール、2-エチル-1-ブタノール、1-ヘプタノール、2-ヘプタノール、3-ヘプタノール、1-オクタノール、2-オクタノール、2-エチル-1-ヘキサノール、1-ノナノール、1-デカノール、1-ウンデカノール、1-ドデカノール、アリルアルコール、プロパルギルアルコール、ベンジルアルコール、シクロヘキサノール、1-メチルシクロヘキサノール、2-メチルシクロヘキサノール、3-メチルシクロヘキサノール、4-メチルシクロヘキサノール等の1価アルコール;1,2-エタンジオール、1,2-プロパンジオール、1,3-プロパンジオール、1,2-ブタンジオール、1,3-ブタンジオール、1,4-ブタンジオール、2,3-ブタンジオール、1,5-ペンタンジオール等の2価アルコール;グリセリン等の3価アルコールを挙げることができる。
 1価アルコールとしては、好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール、1-ヘキサノール、2-メチル-1-ペンタノール、4-メチル-2-ペンタノール、2-エチル-1-ブタノール、1-ヘプタノール、2-ヘプタノール、3-ヘプタノール、1-オクタノール、2-オクタノール、2-エチル-1-ヘキサノール、1-ノナノール、1-デカノール、1-ウンデカノール、1-ドデカノール、ベンジルアルコール、シクロヘキサノール、1-メチルシクロヘキサノール、2-メチルシクロヘキサノール、3-メチルシクロヘキサノール、4-メチルシクロヘキサノール等である。より好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール、1-ヘキサノール、2-メチル-1-ペンタノール、4-メチル-2-ペンタノール、2-エチル-1-ブタノール、シクロヘキサノール等である。さらに好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール等である。特に好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、2-メチル-1-ブタノール、イソペンチルアルコール等であり、最も好ましくは、エタノールである。
 2価アルコールとしては、1,2-エタンジオール、1,2-プロパンジオール、1,3-プロパンジオール等が好ましく、1,2-エタンジオールが最も好ましい。3価アルコールとしては、グリセリンが好ましい。
 上記炭化水素類としては、特に制限されないが、例えば、脂肪族炭化水素、芳香族炭化水素、ハロゲン化炭化水素等を挙げることができる。このなかでも脂肪族炭化水素、芳香族炭化水素が好ましく、脂肪族炭化水素がより好ましい。
 脂肪族炭化水素としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、飽和のものが好ましく用いられる。通常、炭素数3~20、好ましくは炭素数5~12、より好ましくは炭素数5~8のものが用いられる。具体例としては、例えば、プロパン、ブタン、イソブタン、ペンタン、2-メチルブタン、ヘキサン、2-メチルペンタン、2,2-ジメチルブタン、2,3-ジメチルブタン、ヘプタン、ヘプタン異性体(例えば、2-メチルヘキサン、3-メチルヘキサン、2,3-ジメチルペンタン、2,4-ジメチルペンタン)、オクタン、2,2,3-トリメチルペンタン、イソオクタン、ノナン、2,2,5-トリメチルヘキサン、デカン、ドデカン、2-ペンテン、1-ヘキセン、1-ヘプテン、1-オクテン、1-ノネン、1-デセン、シクロペンタン、メチルシクロペンタン、シクロヘキサン、メチルシクロヘキサン、エチルシクロヘキサン、p-メンタン、シクロヘキセン等を挙げることができる。好ましくは、ペンタン、2-メチルブタン、ヘキサン、2-メチルペンタン、2,2-ジメチルブタン、2,3-ジメチルブタン、ヘプタン、2-メチルヘキサン、3-メチルヘキサン、2,3-ジメチルペンタン、2,4-ジメチルペンタン、オクタン、2,2,3-トリメチルペンタン、イソオクタン、ノナン、2,2,5-トリメチルヘキサン、デカン、ドデカン、シクロペンタン、メチルシクロペンタン、シクロヘキサン、メチルシクロヘキサン、エチルシクロヘキサン、p-メンタン等である。より好ましくは、ペンタン、2-メチルブタン、ヘキサン、2-メチルペンタン、2,2-ジメチルブタン、2,3-ジメチルブタン、ヘプタン、2-メチルヘキサン、3-メチルヘキサン、2,3-ジメチルペンタン、2,4-ジメチルペンタン、オクタン、2,2,3-トリメチルペンタン、イソオクタン、シクロペンタン、メチルシクロペンタン、シクロヘキサン、メチルシクロヘキサン、エチルシクロヘキサン等であり、さらに好ましくは、ペンタン、ヘキサン、シクロヘキサン、メチルシクロヘキサン等であり、特に好ましくは、酸化からの防護効果が特に高いという点から、ヘプタン、メチルシクロヘキサンであり、最も好ましくはヘプタンである。
 芳香族炭化水素としては、特に制限されないが、通常、炭素数6~20、好ましくは炭素数6~12、より好ましくは炭素数7~10のものが用いられる。具体例としては、例えば、ベンゼン、トルエン、キシレン、o-キシレン、m-キシレン、p-キシレン、エチルベンゼン、クメン、メシチレン、テトラリン、ブチルベンゼン、p-シメン、シクロヘキシルベンゼン、ジエチルベンゼン、ペンチルベンゼン、ジペンチルベンゼン、ドデシルベンゼン、スチレン等を挙げることができる。好ましくは、トルエン、キシレン、o-キシレン、m-キシレン、p-キシレン、エチルベンゼン、クメン、メシチレン、テトラリン、ブチルベンゼン、p-シメン、シクロヘキシルベンゼン、ジエチルベンゼン、ペンチルベンゼン等である。より好ましくは、トルエン、キシレン、o-キシレン、m-キシレン、p-キシレン、クメン、テトラリン等である。最も好ましくは、クメンである。
 ハロゲン化炭化水素としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、非環状のものが好ましく用いられる。より好ましくは塩素化炭化水素、フッ素化炭化水素であり、さらに好ましくは塩素化炭化水素である。また、炭素数1~6、好ましくは炭素数1~4、より好ましくは炭素数1~2のものが好適に用いられる。具体例としては、例えば、ジクロロメタン、クロロホルム、四塩化炭素、1,1-ジクロロエタン、1,2-ジクロロエタン、1,1,1-トリクロロエタン、1,1,2-トリクロロエタン、1,1,1,2-テトラクロロエタン、1,1,2,2-テトラクロロエタン、ペンタクロロエタン、ヘキサクロロエタン、1,1-ジクロロエチレン、1,2-ジクロロエチレン、トリクロロエチレン、テトラクロロエチレン、1,2-ジクロロプロパン、1,2,3-トリクロロプロパン、クロロベンゼン,1,1,1,2-テトラフルオロエタン等を挙げることができる。好ましくは、ジクロロメタン、クロロホルム、四塩化炭素、1,1-ジクロロエタン、1,2-ジクロロエタン、1,1,1-トリクロロエタン、1,1,2-トリクロロエタン、1,1-ジクロロエチレン、1,2-ジクロロエチレン、トリクロロエチレン、クロロベンゼン、1,1,1,2-テトラフルオロエタン等である。より好ましくは、ジクロロメタン、クロロホルム、1,2-ジクロロエチレン、トリクロロエチレン、クロロベンゼン、1,1,1,2-テトラフルオロエタン等である。
 上記エーテル類としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、飽和のものが好ましく用いられる。通常、炭素数3~20、好ましくは炭素数4~12、より好ましくは炭素数4~8のものが用いられる。具体例としては、例えば、ジエチルエーテル、メチルtert-ブチルエーテル、ジプロピルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジヘキシルエーテル、エチルビニルエーテル、ブチルビニルエーテル、アニソール、フェネトール、ブチルフェニルエーテル、メトキシトルエン、ジオキサン、フラン、2-メチルフラン、テトラヒドロフラン、テトラヒドロピラン、エチレングリコールジメチルエーテル、エチレングリコールジエチルエーテル、エチレングリコールジブチルエーテル、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル、エチレングリコールモノブチルエーテル等を挙げることができる。好ましくは、ジエチルエーテル、メチルtert-ブチルエーテル、ジプロピルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジヘキシルエーテル、アニソール、フェネトール、ブチルフェニルエーテル、メトキシトルエン、ジオキサン、2-メチルフラン、テトラヒドロフラン、テトラヒドロピラン、エチレングリコールジメチルエーテル、エチレングリコールジエチルエーテル、エチレングリコールジブチルエーテル、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル等である。より好ましくは、ジエチルエーテル、メチルtert-ブチルエーテル、アニソール、ジオキサン、テトラヒドロフラン、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル等である。さらに好ましくは、ジエチルエーテル、メチルtert-ブチルエーテル、アニソール等であり、最も好ましくは、メチルtert-ブチルエーテルである。
 上記脂肪酸エステル類としては、特に制限されないが、例えば、プロピオン酸エステル、酢酸エステル、ギ酸エステル等を挙げることができる。好ましくは、酢酸エステル、ギ酸エステルであり、より好ましくは酢酸エステルである。エステル基としては、特に制限されないが、通常、炭素数1~8のアルキルエステル、炭素数7~12のアラルキルエステルが、好ましくは炭素数1~6のアルキルエステルが、より好ましくは炭素数1~4のアルキルエステルが用いられる。
 プロピオン酸エステルの具体例としては、例えば、プロピオン酸メチル、プロピオン酸エチル、プロピオン酸ブチル、プロピオン酸イソペンチル等を挙げることができる。好ましくはプロピオン酸エチル等である。
 酢酸エステルの具体例としては、例えば、酢酸メチル、酢酸エチル、酢酸プロピル、酢酸イソプロピル、酢酸ブチル、酢酸イソブチル、酢酸sec-ブチル、酢酸ペンチル、酢酸イソペンチル、酢酸sec-ヘキシル、酢酸シクロヘキシル、酢酸ベンジル等を挙げることができる。好ましくは、酢酸メチル、酢酸エチル、酢酸プロピル、酢酸イソプロピル、酢酸ブチル、酢酸イソブチル、酢酸sec-ブチル、酢酸ペンチル、酢酸イソペンチル、酢酸sec-ヘキシル、酢酸シクロヘキシル等である。より好ましくは、酢酸メチル、酢酸エチル、酢酸プロピル、酢酸イソプロピル、酢酸ブチル、酢酸イソブチル等であり、最も好ましくは、酢酸エチルである。
 ギ酸エステルの具体例としては、例えば、ギ酸メチル、ギ酸エチル、ギ酸プロピル、ギ酸イソプロピル、ギ酸ブチル、ギ酸イソブチル、ギ酸sec-ブチル、ギ酸ペンチル等を挙げることができる。好ましくは、ギ酸メチル、ギ酸エチル、ギ酸プロピル、ギ酸ブチル、ギ酸イソブチル、ギ酸ペンチル等である。最も好ましくは、ギ酸エチルである。
 本発明における洗浄溶媒は、言うまでもなく上記有機溶媒を単一で(必要に応じて水と混合して)使用することも可能であるし、2種類以上を組み合わせて使用することもできる。
 前記洗浄溶媒としては、融点が例えば-250~10℃程度、好ましくは-150~-30℃程度、より好ましくは-100~-50℃程度のものが好ましい。また沸点が例えば30~300℃、好ましくは40~250℃、より好ましくは50~150℃程度のものも好ましい。或いは1体積倍以上、好ましくは10体積倍以上、より好ましくは100体積倍以上の水と温度25℃で分離することなく混和し得るものも好ましい。これら融点、沸点、及び水との混和性を満足する溶媒を、全洗浄溶媒中、例えば50重量%以上、好ましくは70重量%以上、より好ましくは90重量%以上、特に100重量%にするのが推奨される。
 洗浄溶媒の使用量は、洗浄後にキサントフィロマイセス属酵母菌体との分離が可能な量である限り特に限定されないが、該菌体の乾燥重量1重量部に対して、例えば、0.1重量部以上、好ましくは1重量部以上、さらに好ましくは5重量部以上である。また洗浄溶媒の使用量の上限は特に限定されないが、前記菌体の乾燥重量1重量部に対して、例えば、200重量部以下、好ましくは50重量部以下、より好ましくは20重量部以下である。
 本発明の製造方法においては、特に、カロテノイドとしてアスタキサンチンを得ることを目的とする場合、洗浄溶媒としては、ケトン類、アルコール類を使用するのが好ましく、その中でもアセトン又はエタノールを使用するのがさらに好ましく、エタノールを使用するのがより好ましい。例えば、洗浄溶媒としてアセトン又はエタノールを使用する場合の使用量としては、特に限定されないが、例えば、菌体の乾燥重量1重量部に対して、好ましくは0.1重量部以上、より好ましくは1重量部以上、さらに好ましくは5重量部以上であり、また、好ましくは200重量部以下、より好ましくは50重量部以下、さらに好ましくは20重量部以下である。
 本発明の洗浄工程において、少なくとも1回目の洗浄に使用される洗浄溶媒の含水率は、3~50重量%の範囲であれば特に限定されないが、5~30重量%の範囲がより好ましく、10~20重量%の範囲が特に好ましい。洗浄溶媒中の含水率が3重量%未満では、キサントフィロマイセス属酵母由来のカロテノイド以外の夾雑物の洗浄効率は高くなるものの、目的物質であるカロテノイドの洗浄溶媒に対する溶解度が大きくなるため、カロテノイドの回収率が悪化するという問題がある。一方、洗浄溶媒中の含水率が50重量%を超える条件では、カロテノイドの回収率は良好となるものの、カロテノイド以外の夾雑物の除去率が著しく低下するため、洗浄工程の観点からは好ましくない。
 本発明の製造方法においては、上記「含水率が3~50重量%の洗浄溶媒による洗浄」は、1回でも良く、2回以上実施しても良い。特に後述する「含水率が3重量%未満の洗浄溶媒による洗浄」をさらに実施する場合は、「含水率が3~50重量%の洗浄溶媒による洗浄」は1回でも十分に洗浄効果を得ることが出来る。
 さらに本発明の製造方法においては、上記「含水率が3~50重量%の洗浄溶媒による洗浄」を少なくとも1回実施すれば、キサントフィロマイセス属酵母由来のカロテノイド以外の夾雑物を除去できるが、さらにその除去効果を高めるため、上記「含水率が3~50重量%の洗浄溶媒による洗浄」を少なくとも1回実施した後に、さらに含水率が3重量%未満の洗浄溶媒を用いて少なくとも1回洗浄することが好ましい。この場合、「含水率が3重量%未満の洗浄溶媒」としては含水率が3%未満であれば特に限定されないが、その含水率は1.5重量%以下であるのが好ましく、当然ながら水を全く含まない溶媒を用いることもできる。工業的には、一般に入手できる規格の有機溶媒を、特に含水させることなく用いるのが好ましい。なお、「含水率が3~50重量%の洗浄溶媒による洗浄」と「含水率が3重量%未満の洗浄溶媒による洗浄」は、異なる種類の有機溶媒を使用しても良いが、含水率が異なる以外は同じ有機溶媒を使用するのが好ましい。本発明の製造方法においては、洗浄工程として、含水率が3~50重量%のエタノールによる洗浄を少なくとも1回行った後に、含水率が3重量%未満のエタノールによる洗浄を少なくとも1回実施するのが特に好ましい。含水率が3~50重量%の洗浄溶媒による洗浄と含水率が3重量%未満の洗浄溶媒による洗浄は、それぞれ1回づつ実施するのが最も好ましい。
 本発明の製造方法において、洗浄工程における温度条件は特に限定されないが、洗浄溶媒とキサントフィロマイセス属酵母の混合液の温度が30℃以下となる条件が好ましく、20℃以下の条件がより好ましく、10℃以下がさらに好ましい。洗浄時の混合液の温度が30℃を超えると、洗浄液中のカロテノイド含有率が高くなり、結果、酵母中から抽出し得るカロテノイド量が低下し、カロテノイドの回収率が悪化するという問題がある。洗浄温度の下限は適宜設定でき、例えば、-50℃程度、好ましくは-20℃程度、より好ましくは0℃程度である。
 洗浄の圧力は、前記洗浄溶媒が液体となる範囲で適宜設定でき、例えば、0.5気圧~2気圧程度、好ましくは0.8気圧~1.2気圧程度であり、より好ましくは大気圧(約1気圧)である。
 本発明の製造方法において、洗浄溶媒を用いてカロテノイドを含有するキサントフィロマイセス属酵母を洗浄する際の洗浄方式は、該酵母菌体と洗浄溶媒とを接触させ、次いで不純物を含む洗浄溶媒から酵母菌体を分離し得る限り特に制限はなく、例えば、攪拌翼や静止型ミキサー、流通式カラム、ポンプ等による混合や、菌体の濾過操作時に濾板上のキサントフィロマイセス属酵母に洗浄溶媒を添加し、濾過洗浄しても良いが、洗浄効率の観点から攪拌翼を用いて洗浄溶媒とカロテノイドを含有するキサントフィロマイセス属酵母を一定時間接触・混合させ、その後、該キサントフィロマイセス属酵母と洗浄溶媒を分離することで洗浄操作を行うことが好ましい。その場合の洗浄時間(すなわち洗浄溶媒とカロテノイドを含有するキサントフィロマイセス属酵母の接触時間)は、特に制限されないが、24時間以内が好ましく、より好ましくは1分~6時間の範囲であり、さらに好ましくは、10分~3時間の範囲である。なおここで、洗浄時間を適宜調整することで、洗浄時のカロテノイドの損失をある程度抑えることもできる。また、洗浄終了後のキサントフィロマイセス属酵母と洗浄溶媒との分離操作の種類には特に制限は無く、加圧濾過、減圧濾過、自然濾過、遠心分離、デカンテーション等の一般的な方法により実施することができ、言うまでもなく、これらの操作を組み合わせて行うこともできる。濾過操作を行う場合、操作の効率化のために濾過助剤を使用しても良い。
 本発明の製造方法においては、上記のようにして抽出操作の前に洗浄溶媒を用いてキサントフィロマイセス属酵母を予め洗浄することで、菌体内に多量の脂質を含むキサントフィロマイセス属酵母から、脂質をはじめとする菌体由来の夾雑物の大半が除去され、最終的に製造し得る目的のカロテノイド組成物の高含有量化、油状組成物とした場合の粘性低下や、粉末組成物として得た場合の粉体流動性の向上が図れるほか、製造過程においても残存脂質に起因して生じるべとつきが抑えられ、装置内の付着等の問題が大幅に改善されるなど、生産効率や生産性を向上させることができる。
 次に、本発明の製造方法における熱処理工程について説明する。本発明の製造方法では、上記洗浄工程を実施した後に、洗浄後のキサントフィロマイセス属酵母を、熱処理溶媒中にて65℃以上の高温とすることで熱処理を実施する。抽出工程の前に、洗浄後のキサントフィロマイセス属酵母を、熱処理溶媒と接触させた状態で熱処理を行うことで、その後の抽出工程での抽出効率を向上させ、次の抽出工程で使用する溶媒量を大幅に低減させることができる。なお、本熱処理工程において、キサントフィロマイセス属酵母からカロテノイドが一部抽出されても良い。
 本発明の製造方法において、熱処理工程に用いる熱処理溶媒としては、キサントフィロマイセス属酵母からカロテノイドが抽出されやすくなるよう作用するものであれば特に限定されないが、例えば、アルコール類、炭化水素類及びエーテル類が好ましく適用でき、アルコール類及びエーテル類がより好ましく適用でき、アルコール類が最も好ましく適用できる。
 上記アルコール類としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、飽和のものが好ましく用いられる。通常、炭素数1~20、好ましくは炭素数1~12、より好ましくは炭素数1~6である。なかでも、炭素数1~5の1価アルコール、炭素数2~5の2価アルコール、炭素数3の3価アルコールが好ましい。
 これらアルコール類の具体例としては、例えば、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール、1-ヘキサノール、2-メチル-1-ペンタノール、4-メチル-2-ペンタノール、2-エチル-1-ブタノール、1-ヘプタノール、2-ヘプタノール、3-ヘプタノール、1-オクタノール、2-オクタノール、2-エチル-1-ヘキサノール、1-ノナノール、1-デカノール、1-ウンデカノール、1-ドデカノール、アリルアルコール、プロパルギルアルコール、ベンジルアルコール、シクロヘキサノール、1-メチルシクロヘキサノール、2-メチルシクロヘキサノール、3-メチルシクロヘキサノール、4-メチルシクロヘキサノール等の1価アルコール;1,2-エタンジオール、1,2-プロパンジオール、1,3-プロパンジオール、1,2-ブタンジオール、1,3-ブタンジオール、1,4-ブタンジオール、2,3-ブタンジオール、1,5-ペンタンジオール等の2価アルコール;グリセリン等の3価アルコールを挙げることができる。
 1価アルコールとしては、好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール、1-ヘキサノール、2-メチル-1-ペンタノール、4-メチル-2-ペンタノール、2-エチル-1-ブタノール、1-ヘプタノール、2-ヘプタノール、3-ヘプタノール、1-オクタノール、2-オクタノール、2-エチル-1-ヘキサノール、1-ノナノール、1-デカノール、1-ウンデカノール、1-ドデカノール、ベンジルアルコール、シクロヘキサノール、1-メチルシクロヘキサノール、2-メチルシクロヘキサノール、3-メチルシクロヘキサノール、4-メチルシクロヘキサノール等である。より好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール、1-ヘキサノール、2-メチル-1-ペンタノール、4-メチル-2-ペンタノール、2-エチル-1-ブタノール、シクロヘキサノール等である。さらに好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、tert-ブチルアルコール、1-ペンタノール、2-ペンタノール、3-ペンタノール、2-メチル-1-ブタノール、イソペンチルアルコール、tert-ペンチルアルコール、3-メチル-2-ブタノール、ネオペンチルアルコール等である。特に好ましくは、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール、2-ブタノール、イソブチルアルコール、2-メチル-1-ブタノール、イソペンチルアルコール等であり、最も好ましくは、エタノールである。
 2価アルコールとしては、1,2-エタンジオール、1,2-プロパンジオール、1,3-プロパンジオール等が好ましく、1,2-エタンジオールが最も好ましい。3価アルコールとしては、グリセリンが好ましい。
 上記炭化水素類としては、特に制限されないが、例えば、脂肪族炭化水素、芳香族炭化水素、ハロゲン化炭化水素等を挙げることができる。このなかでも脂肪族炭化水素、芳香族炭化水素が好ましく、脂肪族炭化水素がより好ましい。
 脂肪族炭化水素としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、飽和のものが好ましく用いられる。通常、炭素数3~20、好ましくは炭素数5~12、より好ましくは炭素数5~8のものが用いられる。具体例としては、例えば、プロパン、ブタン、イソブタン、ペンタン、2-メチルブタン、ヘキサン、2-メチルペンタン、2,2-ジメチルブタン、2,3-ジメチルブタン、ヘプタン、ヘプタン異性体(例えば、2-メチルヘキサン、3-メチルヘキサン、2,3-ジメチルペンタン、2,4-ジメチルペンタン)、オクタン、2,2,3-トリメチルペンタン、イソオクタン、ノナン、2,2,5-トリメチルヘキサン、デカン、ドデカン、2-ペンテン、1-ヘキセン、1-ヘプテン、1-オクテン、1-ノネン、1-デセン、シクロペンタン、メチルシクロペンタン、シクロヘキサン、メチルシクロヘキサン、エチルシクロヘキサン、p-メンタン、シクロヘキセン等を挙げることができる。好ましくは、ペンタン、2-メチルブタン、ヘキサン、2-メチルペンタン、2,2-ジメチルブタン、2,3-ジメチルブタン、ヘプタン、2-メチルヘキサン、3-メチルヘキサン、2,3-ジメチルペンタン、2,4-ジメチルペンタン、オクタン、2,2,3-トリメチルペンタン、イソオクタン、ノナン、2,2,5-トリメチルヘキサン、デカン、ドデカン、シクロペンタン、メチルシクロペンタン、シクロヘキサン、メチルシクロヘキサン、エチルシクロヘキサン、p-メンタン等である。より好ましくは、ペンタン、2-メチルブタン、ヘキサン、2-メチルペンタン、2,2-ジメチルブタン、2,3-ジメチルブタン、ヘプタン、2-メチルヘキサン、3-メチルヘキサン、2,3-ジメチルペンタン、2,4-ジメチルペンタン、オクタン、2,2,3-トリメチルペンタン、イソオクタン、シクロペンタン、メチルシクロペンタン、シクロヘキサン、メチルシクロヘキサン、エチルシクロヘキサン等であり、さらに好ましくは、ペンタン、ヘキサン、シクロヘキサン、メチルシクロヘキサン等であり、特に好ましくは、酸化からの防護効果が特に高いという点から、ヘプタン、メチルシクロヘキサンであり、最も好ましくはヘプタンである。
 芳香族炭化水素としては、特に制限されないが、通常、炭素数6~20、好ましくは炭素数6~12、より好ましくは炭素数7~10のものが用いられる。具体例としては、例えば、ベンゼン、トルエン、キシレン、o-キシレン、m-キシレン、p-キシレン、エチルベンゼン、クメン、メシチレン、テトラリン、ブチルベンゼン、p-シメン、シクロヘキシルベンゼン、ジエチルベンゼン、ペンチルベンゼン、ジペンチルベンゼン、ドデシルベンゼン、スチレン等を挙げることができる。好ましくは、トルエン、キシレン、o-キシレン、m-キシレン、p-キシレン、エチルベンゼン、クメン、メシチレン、テトラリン、ブチルベンゼン、p-シメン、シクロヘキシルベンゼン、ジエチルベンゼン、ペンチルベンゼン等である。より好ましくは、トルエン、キシレン、o-キシレン、m-キシレン、p-キシレン、クメン、テトラリン等である。最も好ましくは、クメンである。
 ハロゲン化炭化水素としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、非環状のものが好ましく用いられる。より好ましくは塩素化炭化水素、フッ素化炭化水素であり、さらに好ましくは塩素化炭化水素である。また、炭素数1~6、好ましくは炭素数1~4、より好ましくは炭素数1~2のものが好適に用いられる。具体例としては、例えば、ジクロロメタン、クロロホルム、四塩化炭素、1,1-ジクロロエタン、1,2-ジクロロエタン、1,1,1-トリクロロエタン、1,1,2-トリクロロエタン、1,1,1,2-テトラクロロエタン、1,1,2,2-テトラクロロエタン、ペンタクロロエタン、ヘキサクロロエタン、1,1-ジクロロエチレン、1,2-ジクロロエチレン、トリクロロエチレン、テトラクロロエチレン、1,2-ジクロロプロパン、1,2,3-トリクロロプロパン、クロロベンゼン,1,1,1,2-テトラフルオロエタン等を挙げることができる。好ましくは、ジクロロメタン、クロロホルム、四塩化炭素、1,1-ジクロロエタン、1,2-ジクロロエタン、1,1,1-トリクロロエタン、1,1,2-トリクロロエタン、1,1-ジクロロエチレン、1,2-ジクロロエチレン、トリクロロエチレン、クロロベンゼン、1,1,1,2-テトラフルオロエタン等である。より好ましくは、ジクロロメタン、クロロホルム、1,2-ジクロロエチレン、トリクロロエチレン、クロロベンゼン、1,1,1,2-テトラフルオロエタン等である。
 上記エーテル類としては、環状、非環状を問わず、又、飽和、不飽和を問わず、特に制限されないが、一般に、飽和のものが好ましく用いられる。通常、炭素数3~20、好ましくは炭素数4~12、より好ましくは炭素数4~8のものが用いられる。具体例としては、例えば、ジエチルエーテル、メチルtert-ブチルエーテル、ジプロピルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジヘキシルエーテル、エチルビニルエーテル、ブチルビニルエーテル、アニソール、フェネトール、ブチルフェニルエーテル、メトキシトルエン、ジオキサン、フラン、2-メチルフラン、テトラヒドロフラン、テトラヒドロピラン、エチレングリコールジメチルエーテル、エチレングリコールジエチルエーテル、エチレングリコールジブチルエーテル、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル、エチレングリコールモノブチルエーテル等を挙げることができる。好ましくは、ジエチルエーテル、メチルtert-ブチルエーテル、ジプロピルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジヘキシルエーテル、アニソール、フェネトール、ブチルフェニルエーテル、メトキシトルエン、ジオキサン、2-メチルフラン、テトラヒドロフラン、テトラヒドロピラン、エチレングリコールジメチルエーテル、エチレングリコールジエチルエーテル、エチレングリコールジブチルエーテル、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル等である。より好ましくは、ジエチルエーテル、メチルtert-ブチルエーテル、アニソール、ジオキサン、テトラヒドロフラン、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル等である。さらに好ましくは、ジエチルエーテル、メチルtert-ブチルエーテル、アニソール等であり、最も好ましくは、メチルtert-ブチルエーテルである。
 本発明における熱処理溶媒は、言うまでもなく上記溶媒を単一で使用することも可能であるし、2種類以上を組み合わせて使用することもできる。
 前記熱処理溶媒としては、融点が例えば-250~10℃程度、好ましくは-150~-30℃程度、より好ましくは-100~-50℃程度のものが好ましい。また沸点が例えば30~300℃、好ましくは40~250℃、より好ましくは50~150℃程度のものも好ましい。これら融点及び沸点を満足する溶媒を、全熱処理溶媒中、例えば50重量%以上、好ましくは70重量%以上、より好ましくは90重量%以上、特に100重量%にするのが推奨される。
 熱処理工程における熱処理溶媒の使用量は、キサントフィロマイセス属酵母が十分に加熱され、なおかつ熱処理中の混合液の流動性が確保できれば特に制限はなく、菌体の乾燥重量1重量部に対し、好ましくは1~100重量部、より好ましくは5~50重量部程度の熱処理溶媒を使用することができる。
 熱処理工程における温度は後工程の抽出工程におけるカロテノイドの回収率の観点から、65℃以上の条件であれば特に限定されないが、熱処理温度の上限は、溶媒の沸点、操作圧力、カロテノイドの熱安定性などの観点から適宜設定でき、例えば、熱処理溶媒の沸点以下(エタノールの場合は約78℃)にしてもよい。熱処理工程の温度は、好ましくは70~80℃の範囲、より好ましくは70~76℃の範囲で実施される。
 熱処理工程における加熱時間は、特に限定されないが、後の抽出工程における抽出率向上の観点から、キサントフィロマイセス酵母と熱処理溶媒の混合液が65℃以上となる時間が、好ましくは10秒以上、より好ましくは1分以上、さらに好ましくは15分以上となるように実施するのが好ましい。加熱時間の上限も特に限定されないが、製造効率及びカロテノイドの分解抑制の観点から、10時間以下が好ましく、120分以下がより好ましく、60分以下が特に好ましい。
 熱処理工程の圧力は、前記熱処理溶媒の液体状態を維持できる範囲で適宜設定でき、例えば、0.8気圧~5気圧程度、好ましくは1気圧~1.5気圧程度であり、より好ましくは大気圧(約1気圧)である。
 熱処理方法としては特に限定されず、例えば、洗浄工程後のキサントフィロマイセス属酵母を熱処理溶媒中で加温できる限り、該酵母と熱処理溶媒の添加方向や加熱のタイミングは適宜設定できるが、好ましくは洗浄工程後のキサントフィロマイセス属酵母に、所定温度に加温した熱処理溶媒を加えてもよいし、常温の熱処理溶媒を加えた後に当該混合液を所定温度まで加温してもよい。また熱処理時において熱処理溶媒とキサントフィロマイセス属酵母の混合液は静置状態でも攪拌された状態でもかまわないが、適宜攪拌された状態であるのが好ましい。その場合の攪拌方式には特に制限はなく、後述する抽出工程において使用する混合方法及び/又は装置がそのまま使用できる。
 熱処理終了後、熱処理溶媒は、その一部または全部を菌体と分離してもよく、分離しなくてもよい。分離する場合は、分離後の熱処理溶媒からもカロテノイドを回収できる。分離しない場合は、熱処理溶媒は、後述の抽出溶媒を兼ねることができる。
 次に、本発明の製造方法における抽出工程について説明する。抽出工程とは、上記熱処理後のキサントフィロマイセス属酵母を抽出溶媒と接触させて抽出を行う工程をいう。抽出溶媒としては、菌体と分離されることなく熱処理工程で残った熱処理溶媒と、新たに用いられる溶媒のいずれも用いることができ、それら両方を用いてもよい。すなわち抽出工程では、
(i)上記熱処理後のキサントフィロマイセス属酵母(熱処理溶媒を含んでいてもよい)に、必要に応じて抽出溶媒を加えるか、
(ii)用いる抽出溶媒に、上記熱処理後のキサントフィロマイセス属酵母(熱処理溶媒を含んでいてもよい)を加えるか、或いは
(iii)抽出溶媒を用いることなく、キサントフィロマイセス属酵母と熱処理溶媒を混合させたままにしておき、
 10~70℃の温度で当該酵母中のカロテノイドを抽出する。本発明の製造方法においては、上記熱処理工程後、熱処理溶媒を一部又は全量除去してから、抽出工程を実施してもよいが、熱処理後の熱処理溶媒とキサントフィロマイセス属酵母の混合液をそのまま、引き続き本抽出工程に供するのが好ましい。熱処理溶媒が残っているキサントフィロマイセス酵母に、さらに抽出溶媒を加え、本抽出工程に供するのが最も好ましい態様である。
 本発明の製造方法において、抽出工程に用いる抽出溶媒としては、目的のカロテノイドを効率よく抽出できるものであれば特に限定されないが、例えば、アルコール類、炭化水素類及びエーテル類が好ましく適用でき、アルコール類及びエーテル類がより好ましく適用でき、アルコール類が最も好ましく適用できる。これらアルコール類、炭化水素類及びエーテル類の具体例及び好ましい例は、熱処理溶媒の説明と同じである。
 また、本発明の製造方法において、洗浄溶媒、熱処理溶媒、及び抽出溶媒は、それぞれ同一でも異なっていても良いが、使用した溶媒の回収・再利用の点からは、洗浄溶媒、熱処理溶媒及び抽出溶媒は同一の有機溶媒を使用するのが好ましい。また、残留溶剤が問題とならないという観点では、洗浄溶媒、熱処理溶媒及び抽出溶媒は、いずれもアルコール類から選択するのが好ましく、すべてエタノールを使用するのが最も好ましい。
 本発明の製造方法において、抽出溶媒の使用量は特に限定されないが、抽出溶媒の使用量が菌体の乾燥重量に対し著しく多い場合、得られる抽出液中のカロテノイド濃度が極端に希薄となってしまい、効率的な精製処理が困難となる。一方、菌体の乾燥重量に対し、抽出溶媒の使用量が著しく少ない場合は、高濃度のカロテノイド抽出液が得られるものの、抽出溶媒のカロテノイドに対する飽和溶解度の観点から、菌体中のカロテノイドを効率良く抽出することは極めて難しい。そのような観点から、本発明の製造方法における、熱処理工程後に新たに使用する抽出溶媒の好ましい使用量は、菌体の乾燥重量1重量部に対し1重量部以上であり、より好ましくは5重量部以上であり、最も好ましくは10重量部以上である。また、その上限も特に限定されないが、好ましくは1000重量部以下であり、より好ましくは500重量部以下であり、最も好ましくは300重量部以下である。或いは200重量部以下、特に100重量部以下であってもよい。
 なお、熱処理後に熱処理溶媒の一部又は全部を除去せずそのまま本抽出を行う場合、抽出工程において新たに抽出溶媒を追加しないで抽出を行うことも可能であり、それも本発明の範囲内である。本抽出工程において使用する溶媒の合計量(抽出工程において熱処理溶媒が残存する場合はその残存量と、必要に応じて新たに使用される抽出溶媒の合計量)は、菌体の乾燥重量1重量部に対し1重量部以上であり、好ましくは2重量部以上であり、より好ましくは5重量部以上であり、更に好ましくは10重量部以上であり、より好ましくは15重量部以上であり、最も好ましくは20重量部以上である。また、その上限も特に限定されないが、1100重量部以下であり、好ましくは600重量部以下であり、より好ましくは400重量部以下であり、更に好ましくは300重量部以下であり、最も好ましくは200重量部以下である。
 前述の熱処理工程後本抽出工程において使用する熱処理溶媒の量(残存量)と熱処理工程後に新たに使用する抽出溶媒の量の比率(重量基準)も特に制限されないが、運転のタイムサイクル、溶媒使用量の観点から、熱処理溶媒の残存量に対する抽出溶媒の使用量として、0.1倍量~20倍量の範囲が好ましく、0.3倍量~10倍量の範囲がより好ましく、0.8倍量~5倍量の範囲が特に好ましい。新たに使用する抽出溶媒の温度が室温程度(例えば、0~40℃)であれば、抽出溶媒の添加量が増える程、特段の冷却手段なしに熱処理後の溶媒の温度を下げることができるため好ましい。
 本発明の製造方法において、抽出工程での抽出時の温度は、10~70℃の範囲であれば特に限定されないが、抽出効率の観点から20℃以上が好ましく、25℃以上がより好ましい。10℃未満の抽出温度では、抽出溶媒に対するカロテノイドの溶解度が低いため、満足な収率を得ることが困難となる。一方、抽出時の温度は高いほど抽出効率は向上するが、抽出温度が70℃を超える場合、カロテノイド中のアスタキサンチンの一部がシス体化するなど、カロテノイドの構造が変質する問題がある。そのような観点から、抽出工程はその前の熱処理工程よりも低い温度、例えば60℃以下で実施するのが好ましく、50℃以下で実施するのがより好ましく、45℃以下で実施するのが特に好ましい。なお、熱処理工程の温度よりも抽出工程の温度が低い場合は、熱処理後のキサントフィロマイセス属酵母を所望の温度まで冷却した後、抽出工程を実施しても良いし、熱処理後のキサントフィロマイセス属酵母に適宜温度を調整した抽出溶媒を添加することで、抽出工程における抽出温度を所定の温度に設定することもできる。本発明では、抽出工程を比較的室温に近い状態で実施したとしても、熱処理工程の実施により効率良くアスタキサンチンを抽出することができるため、有用な方法と言える。
 抽出工程の圧力は、前記熱処理工程と同様であってよく、例えば、0.8気圧~5気圧程度、好ましくは1気圧~1.5気圧程度であり、より好ましくは大気圧(約1気圧)である。
 本発明の製造方法においては、抽出溶媒を用いた抽出操作を行う際の、抽出溶媒とキサントフィロマイセス属酵母の混合方式には特に制限はなく、例えば、攪拌翼や静止型ミキサー、流通式カラム、ポンプ等や、高圧ホモジナイザー、回転刃式ホモジナイザー、超音波ホモジナイザー等の破砕・乳化装置を用いることもでき、言うまでもなく、これらの装置を組み合わせて抽出することもできる。
 本発明の製造方法において、抽出手段としては、回分抽出、連続抽出のどちらの方法でも採用することができ、これらの操作を複数回繰り返すこともできるが、工業的には連続抽出が生産性の面で好ましく、連続抽出の中でも向流多段抽出が特に好ましい。回分抽出の場合の攪拌時間は、特に制限されないが、通常5分以上であり、連続抽出の場合の平均滞留時間は、特に制限されないが、通常10分以上である。また回分抽出の回数も特に限定されず、1回の抽出でも効率よく抽出できるが、2回以上抽出を繰り返すのが収率の観点からは好ましい。抽出工程において、2回以上の回分抽出を適用する場合、それぞれの回分抽出の前に熱処理工程を実施するなど、熱処理工程と抽出工程をそれぞれ2回目以上繰り返しても良く、その場合よりカロテノイドの回収率を向上させることができる。
 上記抽出工程が完了した後、キサントフィロマイセス属酵母と抽出溶媒との分離を行う。分離操作の種類には特に制限は無く、加圧濾過、減圧濾過、自然濾過、遠心分離、デカンテーション等の一般的な方法により実施することができ、言うまでもなく、これらの操作を組み合わせて行うこともできる。
 本発明の製造方法においては、キサントフィロマイセス属酵母中に含まれるアスタキサンチンなどのカロテノイドを高い効率で抽出することが可能であり、その抽出率は、通常70%以上、より好ましくは80%以上、さらに好ましくは85%以上である。
 本発明の製造方法においては、上記のようにして得られたカロテノイドの抽出液をそのまま、あるいは適宜加工することでカロテノイド組成物とすることが出来る。本発明の製造方法によって得られるカロテノイド組成物の性状は特に限定されないが、好ましくは、以下に説明するような処理を行って得られる、油状組成物、または粉末組成物である。上記油状組成物は、抽出溶媒によるカロテノイドの抽出液を濃縮することで製造できる。この場合の濃縮操作の方式には特に制限は無く、タンク内での減圧濃縮、フラッシュ濃縮、薄膜濃縮等の操作を適用することができる。
 上記濃縮操作は油状組成物中の洗浄溶媒、または抽出溶媒が十分に除去されるまで実施するのが好ましい。油状組成物中の洗浄溶媒及び抽出溶媒の残存濃度は、食品、化粧品、医薬品として人体に害を及ぼさない程度であれば特に制限はないが、油状組成物中の残存量として1重量%以下が好ましく、より好ましくは0.1重量%以下であり、さらに好ましくは0.01重量%以下である。
 上記濃縮操作における温度条件は、濃縮中にカロテノイドの分解やシス体化等の変質が著しく生じることのない条件であれば特に制限はなく、好ましくは10~80℃の範囲であり、より好ましくは20~70℃の範囲であり、さらに好ましくは25~60℃の範囲である。また、本発明における濃縮操作においては、必要に応じて、減圧下で操作してもよい。
 本発明の製造方法においては、上記濃縮により得られる濃縮物が油状である場合、それをそのまま油状組成物として利用することもできるが、濃縮物が乾固物として得られた場合や油状物として得られた場合でもその他製品としての取り扱い易さの観点から、該濃縮物に液体油脂を添加、混合することで、目的とする油状組成物を調製しても良い。なお、本発明における「油状組成物」とは、カロテノイドとその他の成分が均一に油状状態で混合している場合だけでなく、カロテノイドが一部析出し、スラリー状となっている形態も含まれる。
 上記添加することのできる液体油脂としては、アスタキサンチンなどのカロテノイドの濃縮物を溶解または分散させることのできる油脂であれば特に制限はなく、例えば、動植物からの天然油脂であってもよく、合成油脂や加工油脂であってもよい。より好ましくは、食品、化粧品又は医薬用に許容されるものである。例えば、植物油脂としては、例えば、ヤシ油、パーム油、パーム核油、アマニ油、つばき油、玄米胚芽油、菜種油、米油、落花生油、コーン油、小麦胚芽油、大豆油、エゴマ油、綿実油、ヒマワリ種子油、カポック油、月見草油、シア脂、サル脂、カカオ脂、ゴマ油、サフラワー油、オリーブ油等を挙げることができ、動物油脂としては、例えば、豚脂、乳脂、魚油、牛脂等を挙げることができ、さらに、これらを分別、水素添加、エステル交換等により加工した油脂(例えば、硬化油)も挙げることができる。言うまでもなく、中鎖脂肪酸トリグリセリド(MCT)も使用しうる。又、これらの混合物を使用しても良い。中鎖脂肪酸トリグリセリドとしては、例えば、脂肪酸の炭素数が各々6~12、好ましくは8~12のトリグリセリドを挙げることができる。
 上記、油脂のうち、取り扱い易さ、臭気等の面から植物油脂、合成油脂や加工油脂等が好ましく、そのより好ましい具体例として、ヤシ油、パーム油、パーム核油、菜種油、米油、大豆油、綿実油、サフラワー油、オリーブ油、MCT等を挙げることができる。
 上記液体油脂は、カロテノイドの抽出液の濃縮物を作製後に添加、混合することで目的とするカロテノイドの油状組成物を調製してもよいし、抽出操作、または濃縮操作の前やその途中で予め添加しておき、その後の処理を行っても良い。
 本発明におけるカロテノイドを含有する油状組成物中のカロテノイド含量としては特に限定されず、目的とする製品の求められる用途に応じて適宜選択しうるが、例えばカロテノイドとしてアスタキサンチンを目的とする場合、取り扱いやすい粘度・性状を有する油状組成物を調製するという観点においては、油状組成物中のアスタキサンチン含有量の上限として、例えば、30重量%以下、好ましくは、20重量%以下、より好ましくは15重量%以下である。また油状組成物中のアスタキサンチン含有量の下限としては特に限定されず、例えば0.1重量%以上、好ましくは1重量%以上、より好ましくは3重量%以上である。
 一方、本発明の製造方法では、抽出工程で得られたカロテノイドの抽出液に対し、濃縮、冷却及び/又は貧溶媒添加などの処理を行うことにより、カロテノイドを析出させ、得られる析出物を固液分離することで、カロテノイド組成物として、アスタキサンチンが高濃度に含まれた粉末組成物を製造することもできる。
 本発明の製造方法において、カロテノイドを析出させる方法には特に制限は無く、例えば抽出液を濃縮しながら蒸発晶析する方法、抽出液をそのままあるいは適宜濃縮したものにカロテノイドに対する貧溶媒を添加してカロテノイドを析出させる方法、抽出液をそのままあるいは適宜濃縮したものをカロテノイドの飽和溶解度を超える条件に冷却し晶析する方法等が挙げられる。本発明は、言うまでもなく、これらの操作を組み合わせて製造することもできるが、収率や操作性の観点から、抽出液を濃縮し、ある程度カロテノイド結晶が析出した状態で、貧溶媒を添加するか冷却することでカロテノイドを析出させることが望ましい。
 上記粉末組成物の製造において、抽出液の濃縮操作を行う場合、抽出液の濃縮倍率が低すぎると、目的のカロテノイドのほとんどが母液側に溶解したままとなり、製造し得る粉末組成物の総量は少なくなり、満足な収率、生産性は得られない傾向がある。一方、抽出液の濃縮倍率が高すぎる場合、カロテノイド以外のキサントフィロマイセス属酵母由来の夾雑物の析出が顕著となり、製造し得る粉末組成物中のアスタキサンチン含有量が低下する問題がある。従って、粉末組成物の製造にあたっては、十分な収率、生産性、含有量を満たすためには適切な濃縮倍率まで抽出液を濃縮するのがよく、例えば、抽出液の濃縮倍率が10~1000倍の範囲まで濃縮するのが好ましく、50~500倍の範囲まで濃縮するのがより好ましい。
 粉末組成物の製造においては、次に、上記カロテノイドを析出させた後、抽出に用いた有機溶媒と析出物の固液分離を行う。上記固液分離には通常の固液分離操作を適用することができ、例えば、デカンテーション、遠心分離、加圧濾過、減圧濾過、自然濾過等が挙げられ、さらに、必要に応じてケーキ洗浄を行い、さらに、真空乾燥などの乾燥処理を行って残存する有機溶媒等を除去することにより、カロテノイド組成物を乾燥粉末として取得することができる。
 従来の製造方法では、キサントフィロマイセス属酵母由来のカロテノイド(アスタキサンチンなど)を高濃度で含有し、夾雑物が少ない粉末組成物を得ることは事実上困難であったが、本発明の製造方法によって、例えば粉末組成物中に20~80重量%の範囲の高濃度のカロテノイド(アスタキサンチンなど)を含有した粉末組成物を製造することもできる。好ましくは前記カロテノイドをキサントフィロマイセス属酵母から高い収率で回収できる。さらには洗浄、抽出、熱処理などで用いた溶媒の混入を著しく抑制することも可能である。

 次に本発明を実施例により具体的に説明するが、本発明はこれら実施例のみに限定されるものではない。なお、洗浄や熱処理、抽出に使用した有機溶媒は、含水率を特に記載していない限り、純度99%以上(含水率1%未満)のものを使用した。

 (製造例1)
 5mLのYM培地(ポリペプトン0.5%、酵母エキス0.3%、モルトエキス0.3%、グルコース1.0%)を含む試験管4本に、キサントフィロマイセス・デンドロアス(Xanthophyllomyces dendrorhous)NBRC 10129株(独立行政法人製品評価技術基盤機構生物遺伝資源部門(〒292-0818千葉県木更津市かずさ鎌足2-5-8)より入手)をNTGで変異処理して得られた変異株であるKNK-11株を接種し、20℃で48時間培養した。培養液を50mLのYM培地を含む500mL容の坂口フラスコ4本に移し、20℃で48時間の培養を行なった。培養液を2500mLの培地(リン酸アンモニウム1.3%、リン酸カリウム0.7%、酵母エキス0.3%、グルコース1%を含む)を含む5000mL容のジャーファーメンターに移し、20℃で培養し、アスタキサンチンおよびHDCOを含有する菌体を含む培養液を得た。培養中は、pHを4.4~5.6の間にコントロールし、溶存酸素濃度は飽和の30~80%となるようにグルコースを流加した。
 得られた培養液のうち、3000mLに硫酸400gを添加し、70℃で1時間攪拌を行った。その後、30%水酸化ナトリウムを1300g添加して中和した培養液を減圧濾過し、湿菌体(水分含量60重量%、菌体含量20重量%(乾燥重量として))を調製した。
 (実施例1)
 製造例1にて得られた湿菌体36.7g(アスタキサンチン0.15g含有)に10℃に冷却した含水エタノール(含水率12重量%)100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-1と濾液-1を得た。濾液-1中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.005重量%であった。得られたケーキ-1に10℃に冷却したエタノール100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-2と濾液-2を得た。濾液-2中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.006重量%であった。得られたケーキ-2にエタノール375mLを加え、75℃にて30分攪拌した。その後さらにエタノール375mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-3中のアスタキサンチン量は0.13gであった。この液を1/50になるまで減圧濃縮した後、中鎖脂肪酸トリグリセリド(MCT)1.5gを加え、さらに濃縮を継続した。得られた油状物2.3g中のアスタキサンチン含有量は5.6重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (実施例2)
 製造例1にて得られた湿菌体36.7g(アスタキサンチン0.15g含有)に10℃に冷却した含水エタノール(含水率6重量%)100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-1と濾液-1を得た。濾液-1中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.007重量%であった。得られたケーキ-1に10℃に冷却したエタノール100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-2と濾液-2を得た。濾液-2中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.006重量%であった。得られたケーキ-2にエタノール375mLを加え、75℃にて30分攪拌した。その後さらにエタノール375mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-3中のアスタキサンチン量は0.13gであった。この液を1/50になるまで減圧濃縮した後、サフラワー油2.4gを加え、さらに濃縮を継続した。得られた油状物3.3g中のアスタキサンチン含有量は3.9重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (実施例3)
 製造例1にて得られた湿菌体36.7g(アスタキサンチン0.15g含有)に10℃に冷却した含水エタノール(含水率12重量%)100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-1と濾液-1を得た。濾液-1中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.005重量%であった。得られたケーキ-1にエタノール375mLを加え、75℃にて30分攪拌した。その後さらにエタノール375mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-2と濾液-2を得た。濾液-2中のアスタキサンチン量は0.13gであった。この液を1/50になるまで減圧濃縮した後、サフラワー油2.8gを加え、さらに濃縮を継続した。得られた油状物3.8g中のアスタキサンチン含有量は3.4重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (実施例4)
 実施例1にて得られたケーキ-2にエタノール250mLを加え、70℃にて30分攪拌した。その後さらにエタノール750mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。得られたケーキ-3にエタノール250mLを加え、再度70℃にて30分攪拌した。その後さらにエタノール750mLを加え、40℃にて1時間攪拌した。それを減圧濾過しケーキ-4と濾液-4を得た。濾液-3と濾液-4を合わせた液中のアスタキサンチン量は0.14gであった。この液を1/100になるまで減圧濃縮した後、MCT1.6gを加え、さらに濃縮を継続した。得られた油状物2.7g中のアスタキサンチン含有量は5.2重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (実施例5)
 実施例1にて得られたケーキ-2にエタノール375mLを加え、75℃にて3時間攪拌した。その後さらにエタノール375mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-3中のアスタキサンチン量は0.13gであった。この液を1/50になるまで減圧濃縮した後、サフラワー油2.6gを加え、さらに濃縮を継続した。得られた油状物3.5g中のアスタキサンチン含有量は3.7重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (実施例6)
 実施例1にて得られたケーキ-2にエタノール375mLを加え、65℃にて30分攪拌した。その後さらにエタノール375mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-3中のアスタキサンチン量は0.12gであった。この液を1/50になるまで減圧濃縮した後、サフラワー油2.5gを加え、さらに濃縮を継続した。得られた油状物3.2g中のアスタキサンチン含有量は3.8重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (実施例7)
 製造例1にて得られた湿菌体36.7g(アスタキサンチン0.15g含有)に10℃に冷却した含水メタノール(含水率12重量%)100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-1と濾液-1を得た。濾液-1中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.002重量%であった。得られたケーキ-1に10℃に冷却したメタノール100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-2と濾液-2を得た。濾液-2中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.003重量%であった。得られたケーキ-2に2-プロパノール375mLを加え、75℃にて30分攪拌した。その後さらに2-プロパノール375mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-3中のアスタキサンチン量は0.13gであった。この液を1/50になるまで減圧濃縮した後、サフラワー油2.4gを加え、さらに濃縮を継続した。得られた油状物3.3g中のアスタキサンチン含有量は3.9重量%であった。また、得られた油状物中の残存メタノールと2-プロパノール含量をガスクロマトグラフィーにて測定したところ、それぞれ0.1重量%以下であった。
 (実施例8)
 実施例1にて得られた濾液-3を1/100になるまで減圧濃縮した後、濃縮液を減圧濾過して固液分離し、さらに得られた固形分を室温で減圧乾燥して黒紫色の粉末を得た。得られた粉末0.20g中のアスタキサンチン含有量は50.8重量%であった。
 (比較例1)
 製造例1にて得られた湿菌体32.3g(アスタキサンチン0.13g含有)にアセトン1600mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキと濾液を得た。濾液中のアスタキサンチン量は0.12gであった。この液を減圧濃縮した後、MCT1.63gを加え、更に濃縮を継続した。得られた油状物2.9g中のアスタキサンチン含有量は3.5重量%であった。また、得られた油状物中の残存アセトン含量をガスクロマトグラフィーにて測定したところ、0.2重量%であった。
 (比較例2)
 製造例1にて得られた湿菌体32.3g(アスタキサンチン0.13g含有)にエタノール800mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-1と濾液-1を得た。得られたケーキ-1に更にエタノール800mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-2と濾液-2を得た。濾液-1と濾液-2を合わせた液中のアスタキサンチン量は0.11gであった。この液を減圧濃縮した後、MCT1.63gを加え、更に濃縮を継続した。得られた油状物3.6g中のアスタキサンチン含有量は2.8重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%であった。
 (比較例3)
 製造例1にて得られた湿菌体32.3g(アスタキサンチン0.13g含有)にエタノール800mLを加え、85℃にて1時間攪拌した。それを減圧濾過し、ケーキと濾液を得た。濾液中のアスタキサンチン量は0.12gであった。この液を1/100になるまで減圧濃縮した後、得られた濃縮液から沈殿物を固液分離するため減圧濾過を試みたが、沈殿物のべとつきが著しく、濾過することができなかった。
 (比較例4)
 実施例1にて得られたケーキ-2にエタノール250mLを加え、60℃にて30分攪拌した。その後さらにエタノール500mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-3中のアスタキサンチン量は0.086gであった。この液を1/50になるまで減圧濃縮した後、サフラワー油2.0gを加え、さらに濃縮を継続した。得られた油状物2.8g中のアスタキサンチン含有量は2.8重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%以下であった。
 (比較例5)
 製造例1にて得られた湿菌体32.3g(アスタキサンチン0.13g含有)に5℃に冷却したエタノール100mLを加え、その温度で30分攪拌した。それを減圧濾過しケーキ-1と濾液-1を得た。濾液-1中のアスタキサンチン濃度を高速液体クロマトグラフィーにて測定したところ、0.005重量%であった。得られたケーキ-1にエタノール800mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-2と濾液-2を得た。得られたケーキ-2に更にエタノール800mLを加え、40℃にて1時間攪拌した。それを減圧濾過し、ケーキ-3と濾液-3を得た。濾液-2と濾液-3を合わせた液中のアスタキサンチン量は0.11gであった。この液を1/100になるまで減圧濃縮した後、サフラワー油1.63gを加え、更に濃縮を継続した。得られた油状物2.5g中のアスタキサンチン含有量は4.0重量%であった。また、得られた油状物中の残存エタノール含量をガスクロマトグラフィーにて測定したところ、0.1重量%であった。
 表1に実施例1~8、比較例1、2、4、5における仕込み条件と、得られたカロテノイド油状物中のアスタキサンチン含有量の結果を示す。

Claims (12)

  1. 下記(1)~(3)の工程を含有する、カロテノイド組成物の製造方法。
    (1)カロテノイドを含有するキサントフィロマイセス属酵母を、含水率が3~50重量%の洗浄溶媒を用いて少なくとも1回洗浄する洗浄工程
    (2)上記洗浄後のキサントフィロマイセス属酵母を、熱処理溶媒中にて65℃以上の高温条件で熱処理を施す熱処理工程
    (3)上記熱処理後のキサントフィロマイセス属酵母を抽出溶媒と接触させ、10~70℃の温度で酵母中のカロテノイドを抽出する抽出工程
  2. 抽出工程(3)が、上記熱処理後のキサントフィロマイセス属酵母にさらに抽出溶媒を加えて行われるものである請求項1に記載のカロテノイド組成物の製造方法。
  3. カロテノイドがアスタキサンチンである、請求項1又は2に記載の製造方法。
  4. 洗浄工程(1)において、キサントフィロマイセス属酵母を、含水率が3~50重量%の洗浄溶媒を用いて洗浄した後に、さらに含水率3重量%未満の洗浄溶媒を用いて洗浄する、請求項1~3いずれか1項に記載の製造方法。
  5. 洗浄溶媒が、ケトン類、アルコール類、炭化水素類、エーテル類及び脂肪酸エステル類からなる群より選択される少なくとも1種類の有機溶媒である請求項1~4いずれか1項に記載の製造方法。
  6. 熱処理溶媒が、アルコール類、炭化水素類及びエーテル類からなる群より選択される少なくとも1種の有機溶媒である請求項1~5いずれか1項に記載の製造方法。
  7. 抽出溶媒が、アルコール類、炭化水素類及びエーテル類からなる群より選択される少なくとも1種の有機溶媒である請求項1~6いずれか1項に記載の製造方法。
  8. 洗浄溶媒、熱処理溶媒及び抽出溶媒が、いずれもエタノールである請求項1~4いずれか1項に記載の製造方法。
  9. 熱処理工程(2)における熱処理時間が10秒~10時間である請求項1~8いずれか1項に記載の製造方法。
  10. 抽出工程(3)での抽出温度が10~45℃である請求項1~9いずれか1項記載の製造方法。
  11. 得られたカロテノイドの抽出液を濃縮して油状組成物とする工程をさらに含む、請求項1~10いずれか1項記載の製造方法。
  12. 得られる油状組成物中のアスタキサンチン含有量が1~20重量%である請求項11に記載の製造方法。
