皮膚には、皮膚表面に平行な3層の血管床からなる血管網が形成されている。この血管網により皮膚循環が作り出されている。皮膚循環の第1の機能は体温調節であり、寒冷刺激や温熱刺激が与えられたとき皮膚血管は収縮又は拡張・弛緩して皮膚循環の血流を変化させ、体熱放散を抑制又は促進することで体温を維持する。皮膚循環はまた、全身の血流配分の調節という面においても重要な役割をもっており、中枢や末梢の温熱受容器からの情報以外に、圧、容量又は化学受容器からの情報や、運動によって引き起こされた血管運動反射によっても影響を受ける。
皮膚循環は加齢による変化を受ける。加齢に伴い、皮膚血管網の構造や血流量が変化すること、及び寒冷刺激や温熱刺激に対する血流変化の幅が低減することが知られている。結果として、加齢とともに皮膚の血行が悪化し、それによってさらに皮膚の代謝が低下するという問題が起こり得る。
皮膚循環は、交感神経系を介した全身性メカニズムと、局所的調節因子による局所性メカニズムとの二元的調節を受けている。局所性メカニズムの1つは軸索反射である。これは、刺激により皮膚の感覚神経内で発生したインパルスが、中枢に向けて伝導される途中で他の分枝に逆行性に伝わり、その神経終末から神経ペプチドを放出させ、皮膚血管を拡張させる現象である。このとき放出される神経ペプチドは、substance P(SP)やcalcitonin gene-related peptide(CGRP)である。
CGRPは37個のアミノ酸からなる神経ペプチドであり、カルシトニン遺伝子の組織特異的な選択的スプライシングにより生合成される。CGRPは、中枢神経系及び末梢感覚神経系に広く分布し、特異的な受容体を介してその作用を発揮する。血管内皮細胞及び血管平滑筋細胞にCGRP受容体が発現していることはよく知られている。いずれの細胞も、CGRPがその受容体に結合することにより血管が拡張し、血流が増加する。血管内皮細胞においては、CGRPが細胞のCGRP受容体に結合すると、Gタンパク質を介してアデニレートシクラーゼの活性化を経て細胞内cAMPの増加が生じる。cAMPの増加によりプロテインキナーゼAが活性化し、次いでeNOSが活性化されるので、NOの産生が促進される。産生されたNOは血管平滑筋細胞に作用し、NOを介した細胞内cGMP活性化を経て、K+チャネルが開口する。これによって血管平滑筋は弛緩し、血管の拡張が起こり、その結果として血流は増加する。血管平滑筋細胞にCGRPが直接結合する場合には、Gタンパク質を介したアデニレートシクラーゼの活性化によってプロテインキナーゼAが活性化し、K+チャネルが開口する。この結果やはり血管平滑筋は弛緩して血管の拡張が起こり、血流は増加する。
CGRPと血流との関係はよく知られている。例えば、ラットにおいてCGRPの投与で皮膚血流が増加すること(非特許文献1)、CGRPノックアウトマウスの血圧が高くなること(非特許文献2)、CGRP中和抗体を投与したラットは皮膚血流が減少すること(非特許文献3)、末梢血管の過剰収縮に起因すると考えられているレイノー病の患者では、手指のCGRP含有神経が欠如していること(非特許文献4)、レイノー病患者にCGRPを静注することにより腕の皮膚血流量が上昇すること(非特許文献5)等が知られている。
CGRPの生理作用は、血管拡張作用の他に、炎症におけるサブスタンスPの調節、血管透過性亢進、神経筋接合部のニコチン様受容体の調節、糖新生の抑制と糖分解の促進、膵臓酵素の分泌促進、胃酸分泌抑制、心拍促進、神経活動の調節、カルシウム代謝調節、骨形成促進、インスリン分泌、体温上昇、摂食量低下など多岐にわたる。また、プロスタグランジンの合成を介した抗炎症作用、虚血再環流障害に対するプレコンディショニング、ならびに子宮筋層や子宮、胎盤に受容体が発現していることから妊娠・分娩への関与が知られている。
上記の生理作用から、CGRP又はCGRP応答性促進剤の用途としては、血行促進、血流促進、創傷治癒の促進、及び高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、妊娠中毒症、早期分娩等の治療が挙げられる。また、CGRP受容体アンタゴニストの用途として、偏頭痛、知覚過敏等の治療が挙げられる。実際、多くのCGRP受容体アンタゴニスト化合物が抗偏頭痛薬として開発されている。
また、CGRPと歯周病との関連性も知られている。非特許文献6には、炎症時にCGRPを発現している感覚神経が多数進入すること、CGRPはマクロファージにおけるH2O2産生抑制や抗原提示抑制、リンパ球の分裂抑制を介して抗炎症を示すことが報告されている。また、ヒト歯髄細胞の増殖促進活性があること、BMP-2の発現促進を介した象牙質形成が期待されることから、歯髄形成に関与していることが報告されている(非特許文献7、8)。さらに、骨吸収抑制作用があること(非特許文献9)から、CGRP又はCGRP応答性促進剤の治療用途として、歯周病も挙げられる。
CGRPに対する皮膚循環の応答性を促進又は抑制することができる物質は、上記加齢による皮膚循環の変化の改善や、上記疾患や症状の予防又は治療のために有用である。これまで、CGRP応答性を抑制することができる物質としては、CGRP受容体拮抗剤、抗CGRP抗体、アヤメ属水抽出物(特許文献1)が知られている。しかし、CGRP応答性を促進又は抑制することができ、皮膚循環の調節又は各種疾患の改善に有用なさらなる物質の開発が望まれる。
ある種のリグナン類については、これまでに、細胞接着抑制、癌転移抑制、動脈硬化の予防・治療作用(特許文献2)、NF-κB活性化抑制、遺伝子発現調節、抗ヒト免疫不全ウイルス、炎症予防又は改善、成人病予防又は改善、及び癌転移抑制作用(特許文献3)、歯周病予防又は治療作用(特許文献4)が開示されている。また、リグナン類の1種であるポドフィロトキシン類は、これまでに、細胞毒性があること、ならびに肝障害抑制活性、5-lipoxygenase阻害活性、刺激による細胞へのCa流入抑制作用、COX-2と5-lipoxygenaseをダブル阻害する作用、抗血管新生作用、抗腫瘍活性、抗酸化作用、抗ウイルス活性等の活性を有することが報告されている(非特許文献10~18)。
ポドフィロトキシン類は、Linum flavum根の培養物、Anthriscus sylvestris(シャク、ヤマニンジン)、Pulsatilla koreana(チョウセンオキナグサ)、Hernandiaceae(ハスノハギリ科)のヘルナンディア属Hernandia sp.やHernandia nymphaeifoliaに含まれていることが知られている(非特許文献10、19、20)。非特許文献21~22は、ヘルナンディア属植物やその含有成分の血管拡張作用や抗高血圧作用に関して報告しているが、当該植物の有効性は必ずしも明らかにされていない。さらにヘルナンディア属植物は、ポドフィロトキシン類以外の他の含有成分も含んでいるため、ポドフィロトキシン類のヘルナンディア属植物の薬効に対する寄与は明らかではなかった。

本発明は、細胞のCGRP応答性促進剤、皮膚循環改善剤、血行若しくは血流促進剤、皮膚代謝改善剤、又は創傷治癒促進剤の製造のための、下記式(I)

さらに本発明は、ヒト又は非ヒト哺乳動物に上記式(I)で表されるリグナン類を投与することを含む、当該ヒト又は非ヒト哺乳動物の細胞におけるCGRP応答性促進、皮膚循環改善、血行若しくは血流促進、皮膚代謝改善、又は創傷治癒促進のための方法を提供する。
本明細書において、「非治療的」とは、医療行為、すなわち治療による人体への処置行為を含まない概念である。概念である。
本明細書において、「改善」とは、疾患又は症状の好転、疾患又は症状の悪化の防止又は遅延、あるいは疾患又は症状の進行の逆転、防止又は遅延をいう。
本明細書において、「予防」とは、個体における疾患若しくは症状の発症の防止又は遅延、あるいは個体の疾患若しくは症状の発症の危険性を低下させることをいう。
本明細書において、「細胞のCGRP応答性促進」とは、CGRPにより惹起される細胞の活動を促進することをいう。CGRPが細胞のCGRP受容体に結合すると、Gタンパク質を介してアデニレートシクラーゼの活性化を経て細胞内cAMPの増加が生じる。cAMPの増加により、例えば血管平滑筋細胞では、プロテインキナーゼA活性化を介してK+チャネルが開口する。その結果、血管平滑筋は弛緩して血管の拡張が起こり、血流は増加する。あるいは血管内皮細胞においては、プロテインキナーゼA活性化を介してeNOSが活性化され、NOの産生が促進される。さらに、産生されたNOは血管平滑筋細胞に作用し、NOを介した細胞内cGMP活性化を経て、K+チャネルが開口する。この結果、血管平滑筋は弛緩して血管の拡張が起こり、血流は増加する。本発明による細胞のCGRP応答性促進剤は、CGRPにより惹起される細胞におけるこれらの一連のプロセスを促進し得る。例えば、本発明のCGRP応答性促進剤の作用としては、CGRPとその受容体との結合の促進、Gタンパク質活性の増強、アデニレートシクラーゼ活性の増強、細胞内cAMPの増加、プロテインキナーゼA活性の増強、NO産生又はcGMP活性の増強、及びK+チャネル開口促進等が挙げられる。好ましくは、「細胞のCGRP応答性」は、CGRPにより引き起こされる細胞内cAMPの増加の程度を指標として決定され得る。従って、本発明のCGRP応答性促進剤は、好ましくは、CGRPにより惹起される細胞内cAMPの増加を促進する。
本発明においてCGRP応答性を促進する「細胞」は、CGRP受容体を発現する細胞又はCGRP受容体を有する細胞であれば特に限定されない。好ましくは、細胞としては、血管平滑筋細胞、血管内皮細胞、繊維芽細胞、骨芽細胞、エナメル芽細胞、象牙質芽細胞が挙げられ、より好ましくは血管平滑筋細胞及び血管内皮細胞が挙げられる。あるいは、当該「細胞」は、上記で挙げた細胞の細胞片又は細胞分画物であってもよく、上記で挙げた細胞を含む組織又は上記で挙げた細胞に由来する培養物であってもよい。
本発明は、細胞のCGRP応答性促進剤の提供に関する。
本発明者らは、細胞のCGRP応答性を促進することができる素材について検討し、特定の構造のリグナン類にCGRP応答性促進作用があることを見出した。
本発明によれば、細胞のCGRP応答性を促進することができるCGRP応答性促進剤が提供される。当該CGRP応答性促進剤は、加齢等による皮膚循環の変化の改善や、CGRPに関連する各種疾患や症状の予防又は治療のために有用である。
本発明に係る細胞のCGRP応答性促進剤は、下記式(I)で示されるリグナン類を有効成分とする。
上記式(I)中、R1は、水素原子又はアルコキシ基を示し、R2、R3及びR4は、同一又は異なって、アルコキシ基を示す。上記R1~R4におけるアルコキシ基としては、炭素数1~10の直鎖又は分岐鎖のものが好ましく、例えばメトキシ基、エトキシ基、n-プロピルオキシ基、イソプロピルオキシ基、n-ブチルオキシ基、sec-ブチルオキシ基、tert-ブチルオキシ基、ペンチルオキシ基、ヘキシルオキシ基、ヘプチルオキシ基、オクチルオキシ基、ノニルオキシ基、デシルオキシ基等を挙げることができ、好ましくは、炭素数1~3のアルコキシ基であり、より好ましくはメトキシ基である。
上記式(I)で表されるリグナン類(本明細書において以下、式(I)リグナン類と称する)は、例えばアスナロ(主に葉部、小枝部)から抽出することができる。抽出は、アスナロ又はその乾燥末を水、有機溶媒(石油エーテル、n-ヘキサン、シクロヘキサン、四塩化炭素、トルエン、ベンゼン、ジクロロメタン、クロロホルム、エーテル、酢酸エチル、ブタノール、アセトン、プロパノール、エタノール、メタノール、ピリジン、ポリエチレングリコール、プロピレングリコール、ブチレングリコール等)、それらの混合物(例えば、水性アルコール等)を用い、通常3~70℃で抽出処理することにより行う。
アスナロ原体からの式(I)リグナン類の好ましい抽出例としては、アスナロの乾燥粉砕物100gをエタノール1リットルに浸漬し、室温で時々攪拌しながら7日間抽出を行い、得られた抽出液をろ過し、ろ液を5℃で3日間静置した後、再度ろ過して上澄みを得る方法が挙げられる。次いで得られた抽出液から溶媒を留去して得られた残渣を、適宜メタノール、エタノール、酢酸エチル等の溶媒に溶解し、更に水、メタノール、エタノール、酢酸エチル、クロロホルム、ジクロロメタン、ヘキサン、アセトン、ベンゼン等を溶出溶媒として、アンバーライトXAD-2、ダイアイオンHP-20、TSKゲルHW-40等の親水性ポリマーやセファデックスLH-20等のセファデックス、逆相系シリカゲルやシリカゲル、セルロース等を担体に用いたカラムクロマトグラフィーに付し、薄層クロマトグラフィーなどで目的成分を確認しながら分画することにより目的物を得ることができる。また、場合によりベンゼン、エーテル、ヘキサン、アセトン、メタノール、エタノール、水等の適当な溶媒を用いて再結晶することにより精製してもよい。
また式(I)リグナン類は、文献(特開平10-175861号公報、Natural Product Report,183-205,1995等)記載の方法により合成することができ、その由来は特に限定されるものではない。あるいは、Quality Phytochemicals、2Daybiochem、BOC Sciences、Jamson PharmaChem Technology等から購入することができる。
後記実施例に示すように、式(I)リグナン類は、細胞のCGRP応答性を有意に促進する作用を有する。従って、リグナン類は、細胞のCGRP応答性促進剤として有用である。また、リグナン類は、当該CGRP応答性促進作用を介して、皮膚循環の改善、歯髄形成の促進等の効果を発揮することができ、結果として、皮膚代謝改善、血行促進、血流促進、創傷治癒の促進、あるいは高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善のために有用である。
すなわち、式(I)リグナン類は、細胞のCGRP応答性促進のために使用することができる。あるいは、式(I)リグナン類は、皮膚循環の改善若しくは皮膚代謝改善のため、血行若しくは血流促進のため、又は創傷治癒の促進のために使用することができる。またあるいは、式(I)リグナン類は、高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善のために使用することができる。当該使用は、ヒト若しくは非ヒト動物における使用、又はそれらに由来する組織、器官、細胞等におけるインビトロでの使用であり得る。
また、本発明における式(I)リグナン類の使用は、治療的使用であっても非治療的使用であってもよい。本明細書において、式(I)リグナン類の非治療的使用とは、例えば、健康の維持又は促進を目的とした皮膚循環改善、血行若しくは血流促進、又は皮膚代謝改善のために、あるいは美容又は審美的目的での皮膚代謝改善や創傷治癒促進のために、医療行為としてではなく、式(I)リグナン類を使用することであり得る。また例えば、式(I)リグナン類の非治療的使用とは、他者に、医療行為としてではなく、健康の維持又は促進あるいは美容又は審美的目的で皮膚循環改善、血行若しくは血流促進、皮膚代謝改善、創傷治癒促進等の効果を得るために式(I)リグナン類を使用させることを目的として、当該効果を謳って式(I)リグナン類を提供することであり得る。
従って、一態様として、本発明は、式(I)リグナン類を有効成分として含有する細胞のCGRP応答性促進剤を提供する。当該CGRP応答性促進剤は、細胞のCGRP応答性促進のために使用することができ、また当該CGRP応答性促進作用に基づいて、皮膚循環の改善又は皮膚代謝改善のため、血行又は血流促進のため、創傷治癒の促進のため、あるいは高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善のために使用することができる。一実施形態として、本発明のCGRP応答性促進剤は、本質的に式(I)リグナン類から構成される。好ましくは、当該CGRP応答性促進剤は、皮膚循環の改善又は皮膚代謝改善のため、血行又は血流促進、又は創傷治癒のために使用される。
別の態様として、本発明は、式(I)リグナン類を有効成分として含有する皮膚循環改善剤を提供する。また別の態様として、本発明は、式(I)リグナン類を有効成分として含有する皮膚代謝改善剤を提供する。好ましくは、当該皮膚代謝改善剤は、加齢等による皮膚代謝低下の改善剤である。一実施形態として、これらの剤は、本質的に式(I)リグナン類から構成される。
また別の態様として、本発明は、式(I)リグナン類を有効成分として含有する血行又は血流促進剤を提供する。また別の態様として、本発明は、式(I)リグナン類を有効成分として含有する創傷治癒促進剤を提供する。さらに別の態様として、本発明は、式(I)リグナン類を有効成分として含有する高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善剤を提供する。一実施形態として、これらの剤は、本質的に式(I)リグナン類から構成される。
また別の態様として、本発明は、細胞のCGRP応答性促進剤の製造のための式(I)リグナン類の使用を提供する。なお別の態様として、本発明は、皮膚循環改善剤、血行促進剤、血流促進剤、創傷治癒促進剤、あるいは高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善剤の製造のための式(I)リグナン類の使用を提供する。好ましくは、当該皮膚代謝改善剤は加齢等による皮膚代謝低下の改善剤である。一実施形態において、上記の剤の皮膚循環改善作用、血行若しくは血流促進作用、創傷治癒促進作用、又は上記疾患の予防及び/又は改善作用は、細胞のCGRP応答性促進作用によってもたらされる。
式(I)リグナン類は、細胞のCGRP応答性促進のための、皮膚循環の改善又は皮膚代謝改善のための、血行又は血流促進のための、創傷治癒の促進のための、あるいは高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善のための組成物、医薬、医薬部外品、化粧料、飲食品等の製造のために使用することができる。当該組成物、医薬、医薬部外品、化粧料、飲食品等もまた、本発明の範囲内である。
上記組成物、医薬、医薬部外品、化粧料、飲食品等は、ヒト又は非ヒト動物用として製造され、又は使用され得る。式(I)リグナン類は、当該組成物、医薬、医薬部外品、化粧料、飲食品等に配合され、細胞のCGRP応答性促進のため、皮膚循環の改善又は皮膚代謝改善のため、血行又は血流促進のため、創傷治癒の促進のため、あるいは高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善のための有効成分であり得る。
上記医薬又は医薬部外品は、式(I)リグナン類を有効成分として含有する。当該医薬又は医薬部外品は、任意の投与形態で投与され得る。投与は経口でも非経口でもよい。経口投与のための剤型としては、例えば、錠剤、被覆錠剤、顆粒剤、散剤、カプセル剤のような固形投薬形態、ならびにエリキシル、シロップ及び懸濁液のような液体投薬形態が挙げられ、非経口投与のための剤型としては、注射、輸液、経皮、経粘膜、経鼻、経腸、吸入、坐剤、ボーラス、貼布剤等が挙げられる。
上記医薬又は医薬部外品は、式(I)リグナン類を単独で含有していてもよく、又は薬学的に許容される担体と組み合わせて含有していてもよい。斯かる担体としては、例えば、賦形剤、被膜剤、結合剤、増量剤、崩壊剤、滑沢剤、希釈剤、浸透圧調整剤、pH調整剤、分散剤、乳化剤、防腐剤、安定剤、酸化防止剤、着色剤、紫外線吸収剤、保湿剤、増粘剤、活性増強剤、抗炎症剤、殺菌剤、香料、矯味剤、矯臭剤等が挙げられる。また、当該医薬や医薬部外品は、式(I)リグナン類のCGRP応答性促進作用が失われない限り、他の有効成分や薬理成分を含有していてもよい。
上記医薬又は医薬部外品は、式(I)リグナン類から、あるいは必要に応じて上記担体及び/又は他の有効成分や薬理成分を組みあわせて、常法により製造することができる。当該医薬又は医薬部外品における式(I)リグナン類の含有量は、0.001~100質量%とするのが好ましく、0.002~100質量%とするのがより好ましく、0.005~100質量%とするのがさらに好ましく、0.01~100質量%とするのがさらにより好ましく、0.05~100質量%とするのがなお好ましい。
上記飲食品は、式(I)リグナン類を有効成分として含有する。当該飲食品は、CGRP応答性促進や、皮膚循環改善、血行又は血流促進、皮膚代謝改善、創傷治癒等をコンセプトとし、必要に応じてその旨を表示した飲食品、機能性飲食品、保健食品、病者用飲食品、特定保健用飲食品とすることができる。これら飲食品は表示によって一般の飲食品と区別することが可能である。
上記食品の形態としては、パン類、ケーキ類、麺類、菓子類、ゼリー類、冷凍食品、アイスクリーム類、乳製品、スープ類、食用油、調味料等の各種食品の他、上述した経口投与製剤と同様の形態(錠剤、カプセル剤、顆粒剤、散剤、シロップ等)、ならびに家畜用飼料、ペットフード等が挙げられ、飲料の形態としては、例えば、果汁飲料、炭酸飲料、茶系飲料、ニアウオーター、スポーツ飲料、乳飲料、アルコール飲料、清涼飲料等が挙げられる。また、飲料は、容器に充填した容器詰飲料とすることができる。
上記種々の形態の飲食品を調製するには、式(I)リグナン類を単独で、又は他の飲食品材料や、溶剤、軟化剤、油、乳化剤、防腐剤、香科、安定剤、着色剤、酸化防止剤、保湿剤、増粘剤等を適宜組み合わせて用いることが可能である。上記飲食品等における式(I)リグナン類の含有量は、0.00001~100質量%とするのが好ましく0.00002~75質量%とするのがより好ましく、0.00005~50質量%とするのがさらに好ましく、0.0001~30質量%とするのがさらにより好ましく、0.0002~10質量%とするのがなお好ましい。
上記化粧料は、式(I)リグナン類を有効成分として含有する。当該化粧料は、式(I)リグナン類を単独で含有していてもよく、又は化粧料として許容される担体と組み合わせて含有していてもよい。斯かる担体としては、例えば、賦形剤、被膜剤、結合剤、増量剤、崩壊剤、滑沢剤、希釈剤、浸透圧調整剤、pH調整剤、分散剤、乳化剤、防腐剤、安定剤、酸化防止剤、着色剤、紫外線吸収剤、増粘剤、活性増強剤、抗炎症剤、殺菌剤、香料、矯味剤、矯臭剤等が挙げられる。また、当該化粧料は、式(I)リグナン類のCGRP応答性促進作用が失われない限り、他の有効成分や化粧成分、例えば、保湿剤、美白剤、紫外線保護剤、細胞賦活剤、洗浄剤、角質溶解剤、メークアップ成分(例えば、化粧下地、ファンデーション、おしろい、パウダー、チーク、口紅、アイメーク、アイブロウ、マスカラ、その他)等を含有していてもよい。化粧料とする場合の形態としては、クリーム、乳液、ローション、懸濁液、ジェル、パウダー、パック、シート、パッチ、スティック、ケーキ等、化粧料に使用され得る任意の形態が挙げられる。
上記化粧料は、式(I)リグナン類から、あるいは必要に応じて上記担体及び/又は他の有効成分や化粧成分を組みあわせて、常法により製造することができる。当該化粧料における式(I)リグナン類の含有量は、0.00001~100質量%とするのが好ましく0.00002~75質量%とするのがより好ましく、0.00005~50質量%とするのがさらに好ましく、0.0001~30質量%とするのがさらにより好ましく、0.0002~10質量%とするのがなお好ましい。
あるいは、式(I)リグナン類は、CGRP応答性促進のため、皮膚循環の改善又は皮膚代謝改善のため、血行又は血流促進のため、創傷治癒の促進のため、あるいは高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患の予防及び/又は改善のため、それらを必要とするか又はそれらが望まれる動物を対象に有効量で投与され得る。一態様において、当該投与は、健康の維持若しくは促進又は美容若しくは審美的目的での皮膚循環の改善、皮膚代謝改善、血行若しくは血流促進、又は創傷治癒促進等を企図して、非治療的に行われる。
投与の対象としては、皮膚循環の改善若しくは皮膚代謝の改善が必要であるか又は望まれる動物、好ましくは加齢等による皮膚代謝の低下の改善が必要であるか又は望まれる動物が挙げられる。あるいは、投与の対象としては、血行促進又は創傷治癒の促進が必要であるか又は望まれる動物、歯髄形成促進が必要であるか又は望まれる動物、ならびに高血圧、心不全、虚血再環流による肝障害、腎不全、消化管潰瘍、敗血症、骨粗鬆症、不整脈、くも膜下出血、肺性高血圧、遅延型過敏症、歯周病、妊娠中毒症、早期分娩等の疾患に罹患している動物、その疑いのある動物、又はその危険性の高い動物が挙げられる。好ましくは、皮膚循環改善若しくは皮膚代謝改善、血行若しくは血流促進、又は創傷治癒を必要とするか又はそれらが望まれる動物が投与の対象として挙げられる。動物は、好ましくはヒト又は非ヒト哺乳動物であり、より好ましくはヒトである。
好ましい投与量は、対象の種、体重、性別、年齢、状態又はその他の要因に従って変動し得る。投与の用量、経路、間隔、及び摂取の量や間隔は、当業者によって適宜決定され得る。例えば、ヒトに経口投与する場合、投与量は、式(I)リグナン類として、成人1人当たり、1μg~6000mg/日とすることが好ましく、10μg~5000mg/日がより好ましく、100μg~4000mg/日がさらに好ましく、500μg~3000mg/日がなお好ましい。あるいは、ヒトに経皮投与する場合、投与量は、式(I)リグナン類として、成人1人当たり、0.001μg~6000mg/日とすることが好ましく、0.002μg~5000mg/日がより好ましく、0.005μg~4000mg/日がさらに好ましく、0.01μg~3000mg/日がなお好ましい。
あるいは、投与対象は、動物由来の組織、器官、細胞、又はそれらの培養物若しくは分画物であり得る。当該組織、器官、細胞、又はそれらの分画物は、CGRP受容体を発現するか又はCGRP受容体を有する、天然由来又は生物学的若しくは生物工学的に改変された組織、器官、細胞、又はそれらの分画物である。したがって本発明はまた、当該組織、器官、細胞、又はそれらの培養物若しくは分画物においてCGRP応答性を促進する方法を提供する。本方法は、例えば、CGRP受容体を有し且つCGRP応答性を促進させたい細胞を式(I)リグナン類の存在下で培養する工程を含む。細胞が細胞培養物の場合、添加される式(I)リグナン類の濃度は、0.00001~1000μMであり、好ましくは0.00002~500μMであり、より好ましくは0.00005~200μMである。式(I)リグナン類を添加するタイミングは、CGRP刺激以前及び/又はCGRP刺激時であればよい。
本発明はまた、例示的実施形態として以下の用途又は方法を包含する。但し、本発明はこれらの実施形態に限定されない。
(1)上記式(I)〔式中、R1は水素原子又はアルコキシ基を示し、R2、R3及びR4は、同一又は異なって、アルコキシ基を示す〕で表されるリグナン類を有効成分とする細胞のCGRP応答性促進剤。
(2)好ましくはR1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、(1)記載の剤。

以下、実施例を示し、本発明をより具体的に説明する。
実施例1
リグナン類による細胞のCGRP応答性促進効果を調べた。
1.方法
(1)細胞培養
正常ヒト冠状動脈平滑筋細胞(HCASMC;クラボウ社、細胞lot#01272)を増殖用培地(基礎培地HuMedia-SB2 500mlにFBS 25ml、hEGF 0.5ml、hFGF-B 0.5ml、インスリン0.5ml、抗菌剤0.5mlを添加したもの;クラボウ社)で培養した。継代には0.025%トリプシン/0.01%EDTAを用いた。
リグナン類による細胞のCGRP応答性促進効果を調べた。
1.方法
(1)細胞培養
正常ヒト冠状動脈平滑筋細胞(HCASMC;クラボウ社、細胞lot#01272)を増殖用培地(基礎培地HuMedia-SB2 500mlにFBS 25ml、hEGF 0.5ml、hFGF-B 0.5ml、インスリン0.5ml、抗菌剤0.5mlを添加したもの;クラボウ社)で培養した。継代には0.025%トリプシン/0.01%EDTAを用いた。
(2)細胞の調製
正常ヒト冠状動脈平滑筋細胞(HCASMC)を4000個/cm2の密度で増殖用培地を用いて48ウェルプレートに播種し、翌日分化用培地(基礎培地HuMedia-SB2 500mlにFBS 5ml、ヘパリン0.5ml、抗菌剤0.5mlを添加したもの;クラボウ社)に交換した。培地を1日おきに交換して7日間培養し、CGRP応答性の測定に供した。
正常ヒト冠状動脈平滑筋細胞(HCASMC)を4000個/cm2の密度で増殖用培地を用いて48ウェルプレートに播種し、翌日分化用培地(基礎培地HuMedia-SB2 500mlにFBS 5ml、ヘパリン0.5ml、抗菌剤0.5mlを添加したもの;クラボウ社)に交換した。培地を1日おきに交換して7日間培養し、CGRP応答性の測定に供した。
(3)CGRP応答性の測定
細胞を、ホスホジエステラーゼ阻害剤(PDI)であるIBMX(3-Isobutyl-1-methylxanthine、500μM;シグマ社)、Ro-20-1724(100μM;和光純薬工業株式会社)、及び所定の濃度の下記に示すリグナン類化合物を含む基礎培地(Humedia-SB2;クラボウ社)で2回洗浄し、さらに同培地を200μl添加して37℃にて60分間培養し、細胞内に阻害剤を浸透させた。培地を除去して、同じ濃度の下記に示すリグナン類化合物、0.5nM CGRP、PDIを含む基礎培地を添加し、15分間37℃にて培養した。反応の停止は、cAMP測定EIAキット(cAMP EIA(non-acetylation);GEヘルスケアバイオサイエンス株式会社)に付属の細胞溶解液を添加することにより行い、取扱説明書に記載の方法に従って細胞内cAMP濃度([cAMP]i)を測定した。化合物のCGRP応答性促進能は、0.5nM CGRPのみで刺激したときの[cAMP]iを基準(コントロール)とし、これに対する百分率をImprove Indexとして表した。
(4)試験化合物
試験には、以下のリグナン類化合物TD-1及びTD-2を用いた。これらの化合物は、特開平10-175861号公報に記載の方法に基づいて、以下の手順で調製した。アスナロ乾燥粉砕物(180g)をエタノール1Lに浸漬し、室温において時々攪拌しながら7日間抽出を行い、得られた抽出液を濾過し、濾液を5℃において3日間静置した後、再度濾過し、上澄みを得た。次に溶媒を留去して得られた残渣(2.4g)をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル、CHCl3:CH3OH、H2O:CH3OH)、高速液体クロマトグラフィー(YMC-ODS S-10、YMC社製)に供することによって調製した(収量:TD-1 21mg、TD-2 25mg)。

CGRP応答性の測定結果を図1A及びBに示す。Improve Index 100%は、1189.3fmol/wellの[cAMP]iに相当する。化合物TD-1及びTD-2は、濃度依存的に細胞のCGRP応答性を有意に促進した(Dunnett検定、N=3)。

実施例2
化合物の添加条件を様々に変更して細胞のCGRP応答性を調べた。
1.方法
実施例1と同様の条件でHCASMC(クラボウ社、細胞lot#01272)を調製した。その後、表1に記載の条件で培養物にホスホジエステラーゼ阻害剤(PDI)、試験化合物(TD-1又はTD-2、100nM)、及び/又はCGRP(0.5nM)を添加して培養し、その後、実施例1と同様の方法で細胞のCGRP応答性を測定した。
化合物の添加条件を様々に変更して細胞のCGRP応答性を調べた。
1.方法
実施例1と同様の条件でHCASMC(クラボウ社、細胞lot#01272)を調製した。その後、表1に記載の条件で培養物にホスホジエステラーゼ阻害剤(PDI)、試験化合物(TD-1又はTD-2、100nM)、及び/又はCGRP(0.5nM)を添加して培養し、その後、実施例1と同様の方法で細胞のCGRP応答性を測定した。
2.結果
CGRP応答性の測定結果を図2に示す。Improve Index 100%は、1189.3fmol/wellの[cAMP]iに相当する。CGRP刺激時に試験化合物を添加することにより、Improve Indexは増加した(条件2~3)。一方、CGRP非存在下ではImprove Indexは有意に低かった(条件4)ことから、試験化合物による作用は、CGRPを代替するもの(例えば、CGRP受容体アゴニスト)ではなく、CGRPとその受容体との結合により惹起される一連の応答を促進するものであることが示された。
CGRP応答性の測定結果を図2に示す。Improve Index 100%は、1189.3fmol/wellの[cAMP]iに相当する。CGRP刺激時に試験化合物を添加することにより、Improve Indexは増加した(条件2~3)。一方、CGRP非存在下ではImprove Indexは有意に低かった(条件4)ことから、試験化合物による作用は、CGRPを代替するもの(例えば、CGRP受容体アゴニスト)ではなく、CGRPとその受容体との結合により惹起される一連の応答を促進するものであることが示された。
Claims (41)
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項1記載の使用。
- 前記細胞が血管平滑筋細胞である、請求項1~3のいずれか1項記載の使用。
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項5~8のいずれか1項記載の使用。
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項11記載の使用。
- 前記細胞が血管平滑筋細胞である、請求項11~13のいずれか1項記載の使用。
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項15~18のいずれか1項記載の使用。
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項21記載のリグナン類。
- 前記細胞が血管平滑筋細胞である、請求項21~23のいずれか1項記載のリグナン類。
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項25~28のいずれか1項記載のリグナン類。
- 好ましくはR1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項31記載の方法。
- 好ましくは上記細胞が血管平滑筋細胞である、請求項31~33のいずれか1項記載の方法。
- R1が水素原子又は炭素数1~3のアルコキシ基であり、且つR2、R3及びR4がメトキシ基である、請求項35~38のいずれか1項記載の方法。
- 非治療的方法である、請求項31~40のいずれか1項記載の方法。
