Description
本発明は、細胞分散液中に含まれる癌細胞を捕捉する細胞捕捉デバイスに関する。
癌は世界各国で死因の上位を占め、わが国においては年間30万人以上が癌によって死亡しており、その早期発見および治療が望まれている。癌による人の死亡は、癌の転移再発によるものがほとんどである。癌の転移再発は、癌細胞が原発巣から血管またはリンパ管を経由して、別臓器組織の血管壁に定着、浸潤して微小転移巣を形成することで起こる。このような血管やリンパ管を通じての人の体内を循環する癌細胞は、血中循環癌細胞(Circulating Tumor Cell、以下、場合により「CTC」という。)と呼ばれている。
血液には赤血球や白血球および血小板等の血球成分が多く含まれ、その個数は血液1mL中に3.5~9×109個ともいわれている。その中でCTCは僅か数個程度しか存在しない。血球成分の中からCTCを効率的に検出するためには血球成分を分離する必要があり、観察および測定が非常に困難であった。
CTCの検出により癌の早期発見等が期待されることから、様々な検討がなされている。例えば、特許文献1には、微細な貫通孔を有するパリレン製のフィルターで癌細胞を含む細胞分散液をろ過し、癌細胞(CTC)を捕捉して効率的に検出することが記載されている。
CTC等の癌細胞は、血液中の血球細胞、例えば赤血球や白血球、あるいは血小板等に比べてサイズが一回り大きい。したがって、理論的には、機械的濾過法を適用してこれらの血球成分を除去し、癌細胞を濃縮することが可能である。
しかしながら、文献1に記載のフィルターを組み込んだデバイスは、上下に細胞分散液が流入する流入管及び流出する流出管が突出している。このため、流入管や流出管が障害となり、顕微鏡の対物レンズをフィルターの観察位置に移動することができない場合があった。また、デバイスを顕微鏡の台座に直接固定することが困難な場合があった。
また、上記の問題を別にしても、特許文献1のデバイスは、パリレン製の樹脂フィルターを剛性の低いポリジメチルシロキサン(PDMS)で挟み込んで固定する方式であるため、フィルターのデバイスへの取り付け時にフィルターの平坦度が得られず、顕微鏡等による直接観察を行う場合は、観察時に焦点深度がずれてしまい、対象視野ごとに焦点を調整する必要があり、作業効率が低下する場合があった。
以上のことから、特許文献1のデバイスでは、CTCを捕捉後のフィルターを観察するために、デバイスを分解してフィルターを取り出す必要があった。このため、取り出したフィルターに作業雰囲気環境からゴミなどが汚染し、誤判定してしまう場合があった。また、デバイスの分解において、各種病原菌やウイルスで汚染されている可能性のある、ヒト由来のサンプルが付着したデバイス内部が露出するため、作業の安全性を確保することが煩雑な場合があった。また、フィルターが極薄膜であるため、その取り扱いが煩雑な場合があった。
本発明は、以上のような問題点に鑑みてなされたものであり、デバイスを分解することなく、フィルター上に捕捉した細胞を観察可能な、細胞捕捉デバイスを提供することを目的とする。
本発明は、細胞分散液が流入する流入管が接続された流入口と、細胞分散液が流出する流出管が接続された流出口とを有する筐体と、筐体内に配置され、細胞分散液に含まれる癌細胞を捕捉する捕捉領域を有するフィルターとを備え、フィルターは筐体に接合されており、捕捉領域の少なくとも一部は外部から前記捕捉領域を観察するための観察領域となっており、流入管及び流入口は、フィルターの法線方向から見て観察領域よりも外側の位置に配置され、流入管は、フィルターの面内方向に沿って延びている、細胞捕捉デバイスを提供する。
上記本発明の細胞捕捉デバイスによれば、デバイスを分解することなく、直接顕微鏡の台座に固定して、フィルター上に捕捉した細胞を観察することができる。また、フィルターが、接合により筐体に強固に固定されていることから、より高い圧力で細胞分散液をろ過することが可能となる。これにより、CTCの回収効率を高めることができる。また、フィルターをデバイスの筐体に接合することにより、デバイス内のフィルターを平坦にすることができる。これにより、顕微鏡観察時に焦点深度を一定にすることができ、対象視野ごとに焦点を調整する煩雑さを解消することができる。
流入管及び流入口は、フィルターの法線方向から見て捕捉領域よりも外側の位置に配置されていてもよい。
これにより、より広い観察領域を確保することができ、CTCの検出精度を高めることができる。
フィルターは、実質的に平坦であることが好ましい。これにより、顕微鏡観察時に焦点深度を一定にすることができ、対象視野ごとに焦点を調整する煩雑さを解消することができる。
フィルターは、金属からなることが好ましい。これにより、血球成分の残存を低減し、CTCを高い捕獲率で濃縮することができる。ここで、残存とは、細胞が、フィルターを通過せずにフィルター上に残ることをいう。金属は加工性に優れているため、フィルターの加工精度を高めることができる。これにより、血球成分の残存を低減させ、CTCの高い捕獲率を実現することができる。
また、金属はプラスチックなどの他の材料と比べて剛直であるため、外部から力が加わってもそのサイズや形状が維持される。このため、フィルターの孔(貫通孔)よりも若干大きな血液成分(特に白血球)を変形させて通過させ、高精度の分離・濃縮が可能になると考えられる。白血球の中にはCTCと同じ程度のサイズを有する細胞が存在し、サイズの違いだけではCTCのみを高精度で区別できない場合がある。しかしながら、白血球は癌細胞よりも変形能が大きいため、吸引や加圧などによる外部の力により、自分より小さな孔を通過することができ、CTCと分離することが可能となる。
また、フィルターの材質を金属とすることで高い剛性が得られることから、筐体に固定する時に所定の張力を加えることができる。このため、フィルターがしわやたるみを生じることなく平滑なフィルター面を維持し、フィルターの平坦性を確保することが容易になる。
筐体の少なくとも一部は、可視光領域において実質的に透明であることが好ましい。これにより、デバイスを分解することなく、外部からフィルター上の捕捉領域を観察することができる。また、フィルターをデバイスから取り出す必要がないため、フィルターへのゴミなどの汚染による誤判定を防止することができる。また、各種病原菌やウイルスで汚染されている可能性のある、ヒト由来のサンプルが付着したデバイス内部を露出する必要がないことから、作業の安全性の確保における煩雑さを低減することができる。また、極薄膜であるフィルターを取り扱う作業における煩雑さを低減することができる。
本発明により、デバイスを分解することなく、フィルター上に捕捉した細胞を観察可能な、細胞捕捉デバイスを提供することができる。
以下、本発明の細胞捕捉デバイスの好適な実施形態について図面を参照しながら詳細に説明するが、本発明はこれらに制限されるものではない。
図1は本実施の細胞捕捉デバイスの一実施形態を示す斜視図である。図2は図1におけるII-II線断面図である。
図1および図2に示すように、細胞捕捉デバイス100は、細胞分散液が流入する流入管125が接続された流入口130と、細胞分散液が流出する流出管135が接続された流出口140とを有する筐体120と、筐体120内に配置され、細胞分散液に含まれる癌細胞を捕捉する捕捉領域を有するフィルター105とを備え、捕捉領域の少なくとも一部は外部から前記捕捉領域を観察するための観察領域145となっており、流入管125及び流入口130は、フィルター105の法線方向から見て観察領域145よりも外側の位置に配置され、流入管125は、フィルター105の面内方向に沿って延びている。
ここで、面内方向とは、フィルター105の面内のあらゆる方向をいい、フィルター105の面上の2次元の全方向である。本明細書において「面内方向に沿って」とは、面内方向に対して60°未満、好ましくは45°未満、さらに好ましくは30°未満の角度をなす方向であることを意味する。流入管125及び流入口130が、フィルター105の法線方向から見て観察領域145よりも外側の位置に配置され、流入管125が、フィルター105の面内方向に沿って延びていることにより、細胞捕捉デバイス100の外部から、面160を通してフィルター105上の観察領域145を顕微鏡観察する場合に、観察の障害となる構造物が存在しないため、細胞捕捉デバイスを分解することなく観察することができる。
一方、下部部材115は、流出管135及び流出口140を備える。流出管135は下部部材115の側面に配置されているため、観察領域145を顕微鏡観察する際に、細胞捕捉デバイス100を直接顕微鏡の台座に安定的に固定することができる。
フィルター105は、接合領域155において、上部部材110及び下部部材115と接合されている。接合方法としては溶着が好ましい。溶着とは、高温で材料自体を溶かしてこれらを直接に結合することをいう。溶着は、融着、熱接着、ヒートシールと呼ばれる場合もある。溶着方法としては、熱溶着、超音波溶着、高周波溶着等が挙げられる。フィルター105の周囲を熱または超音波等の処理によって溶着し固定することにより、フィルター105を、しわやたるみのない張力のかかった平坦な状態で配置することができる。これにより、フィルター105を顕微鏡観察する場合の焦点深度が一定となるため、フィルター上に捕捉された細胞の顕微鏡または拡大鏡による観察において焦点の調整を頻繁に行う必要がなく、効率的にCTCを検出することができる。
本実施形態では、上部部材110、フィルター105及び下部部材115が、それぞれ溶着により接合されている。このため、筐体120の気密性を確実に得ることができ、細胞捕捉デバイス100内に細胞分散液を流入させた時の高い液シール性を確保することができる。また、溶着による接合は、短時間で接合処理が可能であるため生産性にも優れる。
また、上部部材110及び下部部材115の材料として、剛性の高いものを選定することにより、フィルター105を接合する時の加圧や熱による変形を抑制することができる。ここで、材料の剛性は、例えばヤング率で表すことができる。上部部材110及び下部部材115の材料としては、ヤング率で0.1GPa以上のものが好ましく、1GPa以上のものがより好ましい。
溶着以外の接合方法としては、接着剤の塗布による方法が挙げられる。しかしながら、接着剤による接合では、好ましい接合が困難な場合がある。例えば、接着剤の塗布量が少なすぎる場合には、接着剤が必要箇所の全体に行き渡らない。このため、完全な接合ができず、上部部材110と下部部材115の間から液漏れが生じる。反対に、接着剤の塗布量が多すぎる場合には、接着剤が必要箇所の範囲を超えて塗布され、フィルター105の貫通孔を塞いでしまう恐れがある。また、接着剤の塗布及び硬化に時間を要し、細胞捕捉デバイス100の生産性が低下する場合がある。
フィルター105の捕捉領域とは、フィルター105が筐体内に配置された状態において、細胞を捕捉可能な領域をいう。フィルター105は、その全面に貫通孔が存在してもよいし、中央部分のみに貫通孔が存在してもよい。例えば、中央部分のみに貫通孔が存在する場合には、細胞を捕捉可能な領域は、フィルター105の貫通孔が存在する中央部分である。また、フィルター105の全面に貫通孔が存在する場合には、フィルターと筐体120との接合領域155は、細胞を捕捉することができないため、捕捉領域は、フィルター105の全面から接合領域155を除いた領域である。捕捉領域の少なくとも一部は観察領域145であり、捕捉領域内に捕捉された細胞のうち、観察領域145内に捕捉された細胞を観察対象とする。
筐体120は、上部部材110及び下部部材115から構成される。筐体120の形状は直方体や円筒等であってよく、特に制約はない。上部部材110の面160及び下部部材115の面170は、平坦で、かつ互いに平行であることが好ましく、特に面160は平滑であることが好ましい。これにより、細胞捕捉デバイス100を顕微鏡の台座に直接固定して、細胞捕捉デバイス100の外部から、面160を通してフィルター105上の観察領域145を顕微鏡観察することが容易になる。
下部部材115の内壁150及びフィルター105により囲まれる空間は空洞であり、この空間内にはフィルターを支える支持体等の構造物が存在しない。このため、フィルター105を通過した細胞分散液の流路が妨げられることがなく、細胞分散液がフィルター105を通過する場合の抵抗が最小限に抑えられ、均一にフィルター105を通過することができる。これにより、細胞捕捉性能のむらや局所的な閉塞を抑制でき、安定した捕捉性能を発揮することができる。
上部部材110はCTCを検出する時に使用する波長の光に対して実質的に透明な材料からなる。上部部材110の材料としては、ガラス、石英ガラス、アクリル樹脂、ポリジメチルシロキサン等の高分子が挙げられるがこれらに限定されない。一実施形態において、上部部材110及び下部部材115の双方が上記の材料からなる。上部部材110と下部部材115とは必ずしも同一材料である必要はないが、接合処理のしやすさの観点から同一材料であることが好ましい。上部部材110及び下部部材115の材料としては、デバイスの量産が可能であることから、低自家蛍光性のアクリル樹脂が好ましく、ポリメチルメタクリレートが特に好ましい。一般的に、癌細胞等を観察する場合には、対象となる細胞に蛍光試薬による染色処理を行った後、波長が300から800nmの紫外または可視光領域の光を照射して、蛍光観察を行う。そのため、上部部材110は前記波長域の光の照射時に材料自体が発する自家蛍光が低い材料を選定することが好ましい(低自家蛍光性)。一般的に、芳香環を有する有機高分子、例えばポリスチレン、ポリカーボネート等の樹脂は、自家蛍光性が強く、上記の目的に適さない場合が多い。
続いて、本発明の細胞捕捉デバイスの変形例について説明する。
図3は細胞捕捉デバイスの一実施形態を示す断面図である。細胞捕捉デバイス200においては、下部部材215にフィルター205を保持する段差が設けられている。フィルター205は、この段差部分にはめ込まれ、接合領域255で溶着により下部部材215と接合されている。その他の点については細胞捕捉デバイス100と同様である。
図4は細胞捕捉デバイスの一実施形態を示す断面図である。細胞捕捉デバイス300においては、下部部材315の内壁350に、流出口340に向かう傾斜が付けられている。これにより、フィルター305を通過した細胞分散液の収集性を向上させ、液だまりを防止することができる。下部部材315の内壁350の傾斜は、直線に限られず、例えば円弧状であってもよい。その他の点については細胞捕捉デバイス100と同様である。
図5(A)は細胞捕捉デバイスの一実施形態を示す断面図である。細胞捕捉デバイス400においては、フィルター405及び下部部材415の間にガスケット480が設けられている。その他の点については細胞捕捉デバイス100と同様である。
図5(B)は細胞捕捉デバイスの一実施形態を示す断面図である。細胞捕捉デバイス401においては、フィルター405は下部部材415に接合されている。そして、フィルター405及び上部部材410の間にガスケット480が設けられている。その他の点については細胞捕捉デバイス100と同様である。
ガスケット480は、上部部材410、下部部材415、及びフィルター405の間の耐流体用シールとして用いることができる。ガスケット480を設置することにより、フィルター405の外周部からの細胞分散液の漏れを、より確実に防止することができる。ガスケット480の代わりにOリングを用いても同様の効果が期待できる。
ガスケット480は弾性のある材料からなることが好ましい。ガスケット480の材料としては、例えば、熱可塑性樹脂、ゴム、エラストマ等が挙げられる。
図6は細胞捕捉デバイスの一実施形態を示す断面図である。細胞捕捉デバイス500においては、流入管525が上部部材510の側面、すなわち、フィルター505の法線方向から見て捕捉領域よりも外側の位置に配置されている。この構成においては、細胞捕捉デバイス500の外部から、面560を通してフィルター505上の観察領域545を顕微鏡観察する場合に、観察の障害となる構造物が存在しない。このため、観察領域545をより広く設定することが可能であり、最大で、フィルター505の捕捉領域と同一の領域にまで広げることが可能となる。また、流出管535は下部部材515の側面に配置されている。その他の点については細胞捕捉デバイス100と同様である。
続いてフィルターについて説明する。
図7(A)は、フィルターの一実施形態を示す概略図である。フィルター600は、複数の貫通孔610が形成された基板620からなり、面630の表面にCTCが捕獲される。貫通孔610の配置は、図7(A)のような整列配置でもよく、列毎に配置がずれた千鳥配置でもよく、任意に配置されたランダム配置であってもよい。
図7(B)は、フィルター600の貫通孔610の上面図である。貫通孔610の開口形状は、短辺がa、長辺がbである長方形の短辺に隣接して、半径がcである2つの半円形が結合した形状である。一実施形態において、a、b、cはそれぞれ8、37及び4μmである。
一実施形態において、フィルターの基板620の材質としては、木綿、麻等の天然高分子、ナイロン、ポリエステル、ポリアクリロニトリル、ポリオレフィン、ハロゲン化ポリオレフィン、ポリウレタン、ポリアミド、ポリスルホン、ポリエーテルスルホン、ポリ(メタ)アクリレート、ハロゲン化ポリオレフィンエチレン-ポリビニルアルコール共重合体、ブタジエン-アクリロニトリル共重合体等の合成高分子あるいはこれらの混合物が挙げられる。また、金属、セラミックス及びこれらの複合材料等も例示できる。
一実施形態において、フィルターの基板620の材質は金属である。金属としては、金、銀等の貴金属、銅、アルミニウム、タングステン、ニッケル、クロム等の卑金属、及びこれらの金属の合金が例示できるがこれらに限定するものではない。金属は単体で用いてもよく、機能性を付与するために他の金属との合金又は金属の酸化物として用いてもよい。酸化や腐食の影響を受けにくいニッケルやステンレス、チタンを主成分とする金属を用いることがより好ましい。ここで、主成分とは、上記基板を形成する材料のうち50重量%以上を占める成分をいう。これらの金属にフォトリソグラフィーなどの方法を使って、貫通孔を形成することができる。
貫通孔の開口形状として、円、楕円、長方形、角丸長方形、多角形等が例示できる。効率良く癌細胞を捕獲できる観点からは、円、長方形又は角丸長方形が好ましい。また、フィルターの目詰まり防止の観点からは、長方形又は角丸長方形が特に好ましい。
一般的なCTCの大きさは直径10μm以上である。ここで、細胞の直径とは、顕微鏡で観察した場合の細胞の輪郭上の任意の2点を結ぶ直線のうち最も長い直線の長さをいう。そこで、血球成分の透過性とCTCの捕捉性能の観点から、貫通孔の平均孔径は、5~15μmであることが好ましく、6~12μmであることがより好ましく、7~10μmであることが特に好ましい。本明細書において、開口形状が楕円、長方形、多角形等の円以外の形状における孔径とは、それぞれの貫通孔を通過できる球の直径の最大値とする。貫通孔の孔径は、例えば開口形状が長方形の場合には、その長方形の短辺の長さとなり、開口形状が多角形の場合には、その多角形の内接円の直径となる。開口形状が長方形や角丸長方形の場合、CTCや白血球が貫通孔に捕捉された場合であっても、開口部において、開口形状の長辺方向に隙間ができる。この隙間を通して液体が通過可能であるため、フィルターの目詰まりを防止することが可能になる。
フィルターの貫通孔の平均開口率は0.1~50%が好ましく、0.5~40%がより好ましく、1~30%が特に好ましく、1~10%が最も好ましい。ここで、開口率とは、フィルター全体の面積に対する貫通孔が占める面積をいう。平均開口率は、目詰まり防止の観点から、大きいほど好ましいが、50%を超えると、フィルターの強度が低下したり、加工が困難になる場合がある。また0.1%より小さいと目詰まりを発生しやすくなるため、フィルターの癌細胞捕捉性能が低下する場合がある。
フィルターの基板の厚さは3~100μmであることが好ましく、5~50μmであることがより好ましく、10~30μmであることが特に好ましい。基板の厚さが3μmより薄いと、フィルターの強度が低下し、取り扱いが困難になる場合がある。また、基板の厚さが100μmを超えると、必要以上の材料を消費したり、加工に長時間かかったりするため、コスト的に不利になったり、精密加工そのものが困難になる場合がある。
フィルター表面の平坦度は、観察に使用する顕微鏡の倍率にもよるが、JIS B0601-2001で規定されるRz(最大高さ粗さ)として10μm以下であることが好ましい。
続いて、本実施形態のフィルターの製造方法を説明する。フィルターの製造方法は特に制限されず、例えば、フォトリソグラフィー法を利用して、エッチングや電気めっきを施すことによる製造方法が挙げられる。フォトリソグラフィー法等を利用した製造方法を以下に説明する。まず、ステンレス等からなる支持体上に感光性のレジストフィルム(感光層)を貼り合わせる。次に、フィルターの貫通孔の開口形状のパターンを有するマスクを感光層上に固定する。続いて、マスク上から光(活性光線)を照射する。光照射後、感光層上に支持体がある場合にはこれを除去し、アルカリ性水溶液、水系現像液及び有機溶剤等の現像液によるウエット現像、又はドライ現像等で未露光部を除去することによって現像し、レジストパターンを形成する。続いて、現像されたレジストパターンをマスクとして、マスクされずに露出している基板上にめっきを行う。めっきの方法としては、例えば、銅めっき、はんだめっき、ニッケルめっき、金めっきなどが挙げられる。めっき処理後、めっき層を支持体及び感光層から剥離すると、めっき層が得られる。このめっき層がフィルターである。得られたフィルターの表面を粗化処理してもよい。粗化処理の方法としては、酸性又は塩基性水溶液等による化学エッチング、サンドブラスト等の物理的処理等が挙げられる。
続いて細胞捕捉デバイスの使用方法について説明する。
細胞分散液としては、骨髄、脾臓、肝臓等にプールされている血液、リンパ、組織液、臍帯血等を利用することが可能であるが、体内を循環している末梢血を使うことが最も簡便である。末梢血中のCTCの存在を検出することは、癌の病状進行を判断する有用な手段である。
細胞分散液中のCTCの存在の有無を検出する場合、細胞捕捉デバイスの流入管に細胞分散液を導入し、フィルター上にCTCを含む細胞を濃縮し、濃縮された細胞の中にCTCが存在するか否かを確認すればよい。流入管への細胞分散液の導入には、加圧又は減圧による方法、ペリスタルティックポンプを使用する方法等が例示できる。また、フィルターの捕捉領域の面積は、例えば1mLの血液からCTCを濃縮する場合、0.25~10cm2が適している。
上記の方法でCTCを濃縮した場合、CTCだけでなく白血球等の血球細胞も同時に濃縮される。このため、回収された細胞に癌原発巣に由来する上皮細胞が含まれているか否かを確認することが必要である。例えば、上記の方法でCTCを濃縮した後、蛍光標識された上皮細胞マーカーに対する抗体で染色し、上皮細胞であることを確認することができる。上皮細胞マーカーに対する抗体としては、抗Cytokeratin抗体等が例示できる。
濃縮した細胞の染色及び観察は、例えば次のようにして行うことができる。細胞を濃縮後の細胞捕捉デバイスの流入管に抗Cytokeratin抗体溶液を導入し、所定時間静置する。続いて、細胞捕捉デバイスの流入管に、洗浄用のバッファーを導入し、未反応の抗体を洗浄除去する。続いて、細胞捕捉デバイスを直接顕微鏡の台座に固定し、蛍光顕微鏡観察を行う。細胞捕捉デバイスに抗体溶液を導入する前に、抗体の非特異的な反応を抑制するためのブロッキング用バッファーを導入してもよい。
また、上記の方法で濃縮された細胞を回収し、遺伝子を解析することにより、癌細胞であることを確認してもよい。細胞の回収は、例えば、細胞捕捉デバイスの流出管側からバッファーを導入し、流入管側から回収することにより行うことができる。例えば、p53、K-RAS、H-RAS、N-RAS、BRAF、APC等の遺伝子における変異を解析し、癌細胞であることを確認できる。また、これらの遺伝子解析の結果は、その後の患者の治療方針の決定等にも利用することができる。あるいは、上記の方法で濃縮した細胞のテロメラーゼ活性等を測定することにより、癌細胞であることを確認してもよい。
細胞分散液中のCTCの存在の有無の検出が終了した後、細胞捕捉デバイスの流出管側から洗浄バッファーを導入し、流入管側から排出することにより、フィルターに捕捉された細胞を洗浄除去し、細胞捕捉デバイスを再利用することも可能である。

- 細胞分散液が流入する流入管が接続された流入口と、前記細胞分散液が流出する流出管が接続された流出口とを有する筐体と、
前記筐体内に配置され、前記細胞分散液に含まれる癌細胞を捕捉する捕捉領域を有するフィルターとを備え、
前記フィルターは、前記筐体に接合されており、
前記捕捉領域の少なくとも一部は外部から前記捕捉領域を観察するための観察領域となっており、
前記流入管及び前記流入口は、前記フィルターの法線方向から見て前記観察領域よりも外側の位置に配置され、
前記流入管は、前記フィルターの面内方向に沿って延びている、細胞捕捉デバイス。 - 前記流入管及び前記流入口は、前記フィルターの法線方向から見て前記捕捉領域よりも外側の位置に配置されている、請求項1に記載の細胞捕捉デバイス。
- 前記フィルターは、実質的に平坦である、請求項1又は2に記載の細胞捕捉デバイス。
