技术领域
本发明属于细胞分离技术领域,涉及一种循环肿瘤细胞分离的微流控芯片,还涉及一种循环肿瘤细胞分离方法以及计数方法。
背景技术
循环肿瘤细胞是从肿瘤原发病灶处脱落,进入人体外周循环血的各类肿瘤细胞的统称,是肿瘤开始转移的标志;因此对循环肿瘤细胞的捕获与检测对于癌症早期诊断、预后分析、个性化医疗、肿瘤单细胞测序等研究方向具有重要的意义。相关研究工作表明,患者体内循环肿瘤细胞水平与无进展生存期和总生存数呈现正相关。目前循环肿瘤细胞液体活检技术已在结肠癌、乳腺癌、前列腺癌诊断方面开展临床应用,具有极大的应用价值和市场前景。循环肿瘤细胞在血液中的含量极为稀少,血液中血细胞与循环肿瘤细胞的浓度之比大于106, 如何有效的分离与富集循环肿瘤细胞是当前研究的重点和挑战。根据原理不同,循环肿瘤细胞的分离方法主要分为生化方法与物理方法。其中使用生化方法的代表产品为获得美国FDA认证、由强生公司生产的CellSearch系统;物理方法主要有过滤、惯性力、确定性定向位移等方法。
CellSearch系统分离循环肿瘤细胞的原理为,循环肿瘤细胞壁特有的上皮粘附分子会与抗体发生特异性结合;将修饰有磁珠的抗体与人体外周循环血充分混合后,循环肿瘤细胞与抗体进行结合,细胞表面进而被修饰上磁珠;在磁场力的作用下,被磁珠修饰的循环肿瘤细胞在血样样品中发生位移,进而与血细胞分离。但是,该技术具有以下几个方面的缺陷。第一,上皮粘性分子存在低表达或不表达的情况,导致循环肿瘤细胞的漏检,从根本上影响了生化方法检测循环肿瘤细胞细胞的准确性;第二,上皮粘附分子与抗体特异性结合时间较长,血液样品处理速度慢;第三,血液样品处理量大,所需抗体较多,成本高。使用物理方法对循环肿瘤细胞进行分离,主要缺点为肿瘤细胞分离纯度不高,白细胞污染较为严重,最终导致染色过程中的假阳性,即误检情况的发生。以上缺陷限制了这些技术在临床上的推广和应用。
发明内容
针对现有技术中存在的技术问题,本发明提供一种用于循环肿瘤细胞分离的微流控芯片,一种使用所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,以及一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞计数的方法。所述分离和计数方法,通量高,效率高,方法简单,便于推广。
为达到上述目的,本发明采用以下技术方案:

所述第一壳层上设置有m个输入接口和n个输出接口,其中m≥1,n≥1;
所述第二壳层上设置有x个输入接口和y个输出接口,其中x≥1,y≥1。

作为本发明优选的技术方案,所述滤膜的材质包括二甲基硅氧烷、聚甲基丙烯酸甲酯或聚碳酸酯中任意一种或至少两种的组合,所述组合典型但非限制性实例有:二甲基硅氧烷和聚甲基丙烯酸甲酯的组合、聚甲基丙烯酸甲酯和聚碳酸酯的组合、聚碳酸酯和二甲基硅氧烷的组合或二甲基硅氧烷、聚甲基丙烯酸甲酯和聚碳酸酯的组合等。
作为本发明优选的技术方案,所述滤膜的滤孔直径为7~15μm,如7.5μm、 8μm、8.5μm、9μm、9.5μm、10μm、10.5μm、11μm、11.5μm、12μm、12.5μm、 13μm、13.5μm、14μm或14.5μm等,但并不仅限于所列举的数值,该数值范围内其他类列举的数值同样适用,优选为8~10μm。
该技术的基本原理为,血液中的循环肿瘤细胞通常比血细胞(红细胞、白细胞等)体积更大,利用横向过滤的微流控器件,可以将血细胞从血液中过滤,从而将循环肿瘤细胞分离,并通过染色与拍照对循环肿瘤细胞进行计数。
基于上述原理,本发明提供的一种用于循环肿瘤细胞分离的微流控芯片的设计原理为:循环肿瘤细胞的血液样品在一侧通道流动,并通过滤膜被过滤;其中体积较小的血细胞会通过滤膜进入另一侧通道,沿另一侧通道流动并被排出;体积较大的循环肿瘤细胞无法通过滤膜,会随血液样品在原通道流动并被收集。
本发明目的之二在于提供一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,所述方法包括以下步骤:
(1)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,血液样品从第一壳层的输入接口输入,过滤后由第二壳层的输出接口排出;
(2)打开第二壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第一壳层的输入接口,缓冲液从第二壳层的输入接口输入,由第二壳层的输出接口排出;
(3)打开第二壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第一壳层的输入接口,缓冲液从第二壳层的输入接口输入,由第一壳层的输出接口排出;
(4)打开第一壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第二壳层的输入接口,缓冲液从第一壳层的输入接口输入,由第一壳层的输出接口排出;
(5)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,缓冲液从第一壳层的输入接口输入,由第二壳层的输出接口排出。
其中,所述缓冲液包括水、磷酸盐缓冲液、加入牛血清白蛋白的磷酸盐缓冲液、培养基或血清等其中任意一种或至少两种的组合。
作为本发明优选的技术方案,所述步骤(2)到步骤(5)依次重复进行1~20 次,如1次、2次、3次、4次、5次、6次、7次、8次、9次、10次、12次、15次、18次或20次等,但并不仅限于所列举的数值,该数值范围内其他类列举的数值同样适用。
本发明目的之三在于提供一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞计数的方法,所述方法包括以下步骤:
(1′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,血液样品从第一壳层的输入接口输入,过滤后由第二壳层的输出口排出;
(2′)打开第二壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第一壳层的输入接口,缓冲液从第二壳层的输入接口输入,由第二壳层的输出接口排出;
(3′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,缓冲液从第一壳层的输入接口输入,由第二壳层的输出接口排出;
(4′)打开第一壳层的输入接口,关闭第一壳层的输出接口、第二壳层的输入接口和、第二壳层的输出接口,染液从第一壳层的输入接口输入,直至染液充满芯片内部,静置;
(5′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,清洗液从第一壳层的输入接口输入,由第二壳层的输出接口排出;
(6′)打开第二壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第一壳层的输入接口,清洗液从第二壳层的输入接口输入,由第一壳层的输出接口排出;
(7′)在荧光显微镜下观察芯片内部被染色细胞的荧光信号并拍照,即可对循环肿瘤细胞进行计数。
其中,所述缓冲液包括水、磷酸盐缓冲液、加入牛血清白蛋白的磷酸盐缓冲液、培养基或血清等其中任意一种或至少两种的组合。
其中,所述染液包括异硫氰酸荧光素和/或藻红蛋白等。
其中,所述清洗液包括加入1%胎牛血清的磷酸盐缓冲液和/或磷酸盐缓冲液等。
作为本发明优选的技术方案,进行所述步骤(4′)前,步骤(2′)到步骤(3′) 依次重复进行1-20次,如1次、2次、3次、4次、5次、6次、7次、8次、9 次、10次、12次、15次、18次或20次等,但并不仅限于所列举的数值,该数值范围内其他类列举的数值同样适用。
作为本发明优选的技术方案,进行所述步骤(7′)前,步骤(5′)到步骤(6′) 依次重复进行1~20次,如1次、2次、3次、4次、5次、6次、7次、8次、9 次、10次、12次、15次、18次或20次等,但并不仅限于所列举的数值,该数值范围内其他类列举的数值同样适用。
本发明所述第一、第二等仅是为了叙述清楚和方便而采取的命名方式,并不是对命名的结构的顺序以及使用顺序的限制。
与现有技术方案相比,本发明至少具有以下有益效果:
(1)本发明提供的一种用于循环肿瘤细胞分离的微流控芯片,所述芯片通量高,效率高,血细胞的过滤效率可达99.99%以上,对于循环肿瘤细胞的分离率达86%以上;
(2)本发明提供的一种使用所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,所述方法采用循环过滤的方法,大大提高了物理过滤的效率,提高了循环肿瘤细胞的分离纯度;
(3)本发明提供的一种使用所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,所述方法无需使用昂贵的抗体,成本低廉。

具体实施方式
下面结合附图并通过具体实施方式来进一步说明本发明的技术方案。
为更好地说明本发明,便于理解本发明的技术方案,本发明的典型但非限制性的实施例如下:
实施例1
一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,所述方法包括以下步骤:
(1)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,将人体外周循环血用磷酸盐缓冲液稀释5倍后,取1毫升样品,以40mbar的压力从第一壳层的输入接口输入,过滤后由第二壳层的输出接口排出;
(2)打开第二壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第一壳层的输入接口,800μL磷酸盐缓冲液从第二壳层的输入接口输入,由第二壳层的输出接口排出;
(3)打开第二壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第一壳层的输入接口,800μL磷酸盐缓冲液从第二壳层的输入接口输入,由第一壳层的输出接口排出;
(4)打开第一壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第二壳层的输入接口,800μL磷酸盐缓冲液从第一壳层的输入接口输入,由第一壳层的输出接口排出;
(5)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,800μL磷酸盐缓冲液从第一壳层的输入接口输入,由第二壳层的输出接口排出。
步骤(5)后,依次重复上述步骤(2)到步骤(5)9次。
其中,步骤(1)的过滤流速为51mL/h,步骤(2)的过滤流速为48mL/h,步骤(3)的过滤流速为48mL/h,步骤(4)的过滤流速为48mL/h,步骤(5) 的过滤流速为48mL/h。
测试前,取1mL血液样品,经过流式细胞仪测试,得到样品的总颗粒数为 1.11×109,而经10次过滤后,收集到的样品的颗粒数为1.09×109,10次过滤操作对样品中血细胞的过滤效率为99.99%。
实施例2
一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,所述方法除了步骤(5)后,不重复上述步骤(2)到步骤(5)外,其他条件均与实施例1相同。
经1次过滤后,收集到的样品的颗粒数为9.63×108,1次过滤操作对样品中血细胞的过滤效率为86.79%。
实施例3
一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞计数的方法,所述方法包括以下步骤:
(1′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,将浓度约为1×106每毫升SK-BR-3细胞用磷酸盐缓冲液稀释一万倍后,取600μL样品从第一壳层的输入接口输入,过滤后由第二壳层的输出口排出;
(2′)打开第二壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第一壳层的输入接口,100μL磷酸盐缓冲液从第二壳层的输入接口输入,由第二壳层的输出接口排出;
(3′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,100μL磷酸盐缓冲液从第一壳层的输入接口输入,由第二壳层的输出接口排出;
(4′)打开第一壳层的输入接口,关闭第一壳层的输出接口、第二壳层的输入接口和、第二壳层的输出接口,染液从第一壳层的输入接口输入,直至染液充满芯片内部,静置;
(5′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,100μL清洗液从第一壳层的输入接口输入,由第二壳层的输出接口排出;
(6′)打开第二壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第一壳层的输入接口,100μL清洗液从第二壳层的输入接口输入,由第一壳层的输出接口排出;
(7′)在荧光显微镜下观察芯片内部被染色细胞的荧光信号并拍照,即可对循环肿瘤细胞进行计数。
所述计数结果为分离得到的SK-BR-3细胞的细胞数为56个。
取1mL过滤前的样品,使用细胞计数仪计数,得到SK-BR-3细胞的细胞数 7.561×105,经过1次过滤与清洗操作后,芯片对肿瘤细胞的获取效率为74.2%。
实施例4
一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞计数的方法,所述方法除了进行步骤(4′)之前依次重复步骤(2′)到步骤(3′)20次,以及进行步骤(7′)前,依次重复步骤(5′)到步骤(6′)20次外,其他条件均与实施例3相同。
所述计数结果为分离得到的SK-BR-3细胞的细胞数为45。
经过20次过滤与清洗操作后,芯片对肿瘤细胞的获取效率为59.6%。
实施例5
将平均浓度为1×105到1×106每毫升的SK-BR-3细胞溶液,使用磷酸盐缓冲液稀释10倍后取1mL,将此1mL细胞溶液加入1mL血液样品中,并按照实施例2的方法进行操作。
使用细胞计数仪对分离得到的样品以及过滤前的肿瘤细胞样品进行计数,得到芯片对SK-BR-3细胞的获取效率为85.72%,处理后肿瘤细胞数量与血细胞数量的比例提高60倍
实施例6
将平均浓度为1×105到1×106每毫升的SK-BR-3细胞溶液,使用磷酸盐缓冲液稀释10倍后取1mL,将此1mL细胞溶液加入1mL血液样品中,并按照实施例4的方法进行操作。
使用细胞计数仪对过滤前的肿瘤细胞样品进行计数,根据计数结果得到芯片对SK-BR-3细胞的获取效率为59.6%,处理后肿瘤细胞数量与血细胞数量的比例提高到455倍。
对比例1
一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,所述方法除了只进行步骤(1)外,其他条件均与实施例1相同。
使用细胞计数仪对分离得到的样品以及过滤前的样品进行计数,得到芯片对血细胞的获取效率为43%。
对比例2
一种所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞计数的方法,所述方法除了只进行步骤(1′)外,其他条件均与实施例3相同。
使用细胞计数仪对分离得到的样品以及过滤前的肿瘤细胞样品进行计数,得到芯片对SK-BR-3细胞的获取效率为65%。
对比例3
根据文献(Lin,H.K.,Zheng,S.,Williams,A.J.,Balic,M.,Groshen,S.,Scher,H.I.,Cote,R.J.(2010).Portable filter-based microdevice for detection andcharacterization of circulating tumor cells.Clinical Cancer Research,16(20),5011–5018)中使用强生公司的CellSearch系统对混有乳腺癌细胞的血液样品进行测试,测试结果为,对肿瘤细胞的平均获取效率为13%。
根据实施例1-6可以看出,利用本发明提供的用于循环肿瘤细胞分离的微流控芯片以及提供的使用方法,对血液中的血细胞的获取效率可达99.99%,而对癌细胞的分离效率页要高于50%,对于将癌细胞与血液混合后的样品,分离得到的癌细胞与残留的血细胞的比例为初始状态的455倍。对比例1和对比例2 均未按照本发明提供的使用方法进行分离,癌细胞和血细胞的分离效率均有所下降。
申请人声明,本发明通过上述实施例来说明本发明的详细结构特征,但本发明并不局限于上述详细结构特征,即不意味着本发明必须依赖上述详细结构特征才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明所选用部件的等效替换以及辅助部件的增加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于上述实施方式中的具体细节,在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,这些简单变型均属于本发明的保护范围。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合,为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。
此外,本发明的各种不同的实施方式之间也可以进行任意组合,只要其不违背本发明的思想,其同样应当视为本发明所公开的内容。
Claims (10)
1.一种用于循环肿瘤细胞分离的微流控芯片,其特征在于,所述微流控芯片包括第一壳层和第二壳层,以及设置于所述第一壳层和第二壳层之间的滤膜,所述滤膜和第一壳层之间形成第一通道,所述滤膜和第二壳层之间形成第二通道;
所述第一壳层上设置有m个输入接口和n个输出接口,其中m≥1,n≥1;
所述第二壳层上设置有x个输入接口和y个输出接口,其中x≥1,y≥1。
2.根据权利要求1所述的微流控芯片,其特征在于,所述第一壳层的材质包括二甲基硅氧烷、聚甲基丙烯酸甲酯或聚碳酸酯中任意一种或至少两种的组合。
3.根据权利要求1或2所述的微流控芯片,其特征在于,所述第二壳层的材质包括二甲基硅氧烷、聚甲基丙烯酸甲酯或聚碳酸酯中任意一种或至少两种的组合。
4.根据权利要求1-3任一项所述的微流控芯片,其特征在于,所述滤膜的材质包括二甲基硅氧烷、聚甲基丙烯酸甲酯或聚碳酸酯中任意一种或至少两种的组合。
5.根据权利要求1-4任一项所述的微流控芯片,其特征在于,所述滤膜的滤孔直径为7~15μm,优选为8~10μm。
6.一种使用1-5任一项所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞分离的方法,其特征在于,所述方法包括以下步骤:
(1)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,血液样品从第一壳层的输入接口输入,过滤后由第二壳层的输出接口排出;
(2)打开第二壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第一壳层的输入接口,缓冲液从第二壳层的输入接口输入,由第二壳层的输出接口排出;
(3)打开第二壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第一壳层的输入接口,缓冲液从第二壳层的输入接口输入,由第一壳层的输出接口排出;
(4)打开第一壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第二壳层的输入接口,缓冲液从第一壳层的输入接口输入,由第一壳层的输出接口排出;
(5)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,缓冲液从第一壳层的输入接口输入,由第二壳层的输出接口排出。
7.根据权利要求6所述的方法,其特征在于,所述步骤(2)到步骤(5)依次重复进行1~20次。
8.一种使用1-5任一项所述循环肿瘤细胞分离的微流控芯片进行循环肿瘤细胞计数的方法,其特征在于,所述方法包括以下步骤:
(1′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,血液样品从第一壳层的输入接口输入,过滤后由第二壳层的输出口排出;
(2′)打开第二壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第一壳层的输入接口,缓冲液从第二壳层的输入接口输入,由第二壳层的输出接口排出;
(3′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,缓冲液从第一壳层的输入接口输入,由第二壳层的输出接口排出;
(4′)打开第一壳层的输入接口,关闭第一壳层的输出接口、第二壳层的输入接口和、第二壳层的输出接口,染液从第一壳层的输入接口输入,直至染液充满芯片内部,静置;
(5′)打开第一壳层的输入接口和第二壳层的输出接口,关闭第一壳层的输出接口和第二壳层的输入接口,清洗液从第一壳层的输入接口输入,由第二壳层的输出接口排出;
(6′)打开第二壳层的输入接口和第一壳层的输出接口,关闭第二壳层的输出接口和第一壳层的输入接口,清洗液从第二壳层的输入接口输入,由第一壳层的输出接口排出;
(7′)在荧光显微镜下观察芯片内部被染色细胞的荧光信号并拍照,即可对循环肿瘤细胞进行计数。
9.根据权利要求8所述的方法,其特征在于,进行所述步骤(4′)前,步骤(2′)到步骤(3′)依次重复进行1-20次。
10.根据权利要求8或9所述的方法,其特征在于,进行所述步骤(7′)前,步骤(5′)到步骤(6′)依次重复进行1~20次。
