본 발명에 따른 나노 구조를 갖는 세포 배양용 용기는 성체 줄기세포를 점착하여 줄기세포를 증식 및 분화시키기 위한 세포 배양면을 포함하는 세포 배양용 용기에 있어서, 세포 배양면은 세포 배양면 상에서 일정한 간격을 두고 배치되는 나노 구조물을 포함하고, 나노 구조물은 세포 배양면으로부터 돌출한 나노 기둥을 포함하고, 나노 기둥의 폭은 40 nm 내지 500 nm 사이의 범위이고, 나노 기둥의 높이는 10nm 내지 1 ㎛ 사이의 범위이다.

<2> 현재 사람 몸 속의 세포 (특히, 줄기세포)를 체외에서 배양한 다음 이를 다시 환자의 몸 속에 넣어 질환올 치료하는 세포 치료가 확대되고 있다. 이에 따라 저가 의 쉬운 방법에 의해 세포의 증식과 분화 효율올 향상 시 킬 수 있는 배양 방법과 배양계 (cul ture system)에 대한 관심 이 높아지고 있다. 이 러 한 배양계에는 다양한 기기들이 관계하고 있으며 , 이에 있어서 세포배양액과 세포를 담을 수 있는 세포 배양용 용기가 중요한 요소 중 하나이다.

<4> 본 발명은 상기 한 문제점을 해결하기 위 한 것으로서 , 세포 배양용 용기에 나 노구조를 도입하여 세포가 원래 안주하고 있는 환경올 모사한다. 이를 통해 성체 줄기세포를 비룻한 각종 세포의 부착, 증식 및 분화 효율을 높이는 것올 목적으로 한다.

【기술적 해결방법】

<86> 도 16 및 도 17올 참조하면, 나노 기둥을 가지는 실시여 U에서 나노 기공을 가지는 실시예 2 및 비교예에서 배양한 지방유래 줄기 세포보다 골세포로의 분화효 율이 상대적으로 높게 나타남을 확인할수 있다.
<87> 도 11 내지 도 17을 통해서 볼 때, 직경 200ntn, 깊이 500 nm인 나노 기공 형 상과 직경 200 nm, 높이 500ntn인 나노 기등 형상이 규칙적으로 형성된 세포 배양면 에서 성체 줄기세포를 배양한 경우를 평평한 배양면과 비교할 때, 부착율, 증식율 및 분화 효율이 상대적으로 향상되는 효과를 확인할수 있다.
<88> 따라서 적정한 크기의 나노 기공, 나노 기등 구조의 나노 구조물을 포함하는 세포 배양면올 이용하여 세포를 배양하는 경우, 안정적으로 세포 점착, 증식 및 분
화를 유도할 수 있다. 그리고 보다 넓은 면적으로 안정적으로 세포가 세포 배양면 표면에 점착되는 효과를 얻올 수 있다. 따라서 이 러한 구조물을 포함하는 세포 배 양용 용기에서 성체 줄기세포를 배양하는 경우, 세포 분화 효율을 증가시 키 고, 많 은 수의 세포를 얻을 수 있을 것이다.

【청구의 범위】

성체 줄기세포를 점착하여 상기 줄기 세포를 증식 및 분화시 키기 위 한 세포 배양면을 포함하는 세포 배양용 용기에 있어서,
상기 세포 배양면은 상기 세포 배양면 상에서 일정 한 간격을 두고 배치되는 나노 구조물을 포함하고,
상기 나노 구조물은 상기 세포 배양면으로부터 돌출한 나노 기둥올 포함하 고 ,
상기 나노 기등의 폭은 40 nm 내지 500 nm 사이의 범위 이고 ,
상기 나노 기등의 높이는 lOnm 내지 1 사이의 범위 인 나노 구조를 갖는 세포 배양용 용기 .
【청구항 2】
저 u항에서 ,
상기 나노 기둥은 반원 형상의 기단과, 상기 기단으로부터 일정한 폭으로 돌 출하며 상부가 반원 형상올 이루는 기등으로 이루어지는 나노 구조를 갖는 세포 배 양용 용기 .

성체 줄기세포를 점착하여 증식 및 분화시 기기 위 한 세포 배양면올 포함하는 세포 배양용 용기에 있어서,
상기 세포 배양면은 상기 세포 배양면 상에서 일정 한 간격을 두고 배치되는 나노 구조물을 포함하고,
상기 나노 구조물은 상기 세포 배양면으로부터 오목한 나노 기공올 포함하 고 ,
상기 나노 기공의 폭은 40 nm 내지 500 nm 사이 의 범위 이고,
상기 나노 기공의 깊이는 lOnm 내지 1 사이의 범위 인 나노 구조를 갖는 세 포 배양용 용기 .

제 1항 또는 제 3항에서,
상기 세포 배양면은 열가소성 수지, 열경화성 수지 및 탄성중합체 중 적어도 하나로 이루어지는 나노 구조를 갖는 세포 배양용 용기 .

제 4항에서,
상기 세포 배양용 용기는 표면에 플라즈마 처리 , 오존 처리 또는 세포 점착 증진 물질의 코팅 중 어느 하나가 처리되어 있는 나노 구조를 갖는 세포 배양용 용 기 .

2단계 양극산화알루미늄 공정을 이용하여 예비 기공올 포함하는 알루미늄 템 플릿을 형성하는 단계,
상기 알루미늄 템플릿 위에 고분자 물질층을 형성하고, 상기 고분자 물질층 상부를 상기 알루미늄 템플릿으로 가압하여 고분자 템플릿을 형성하는 단계,
상기 고분자 템플릿 및 상기 알루미늄 템플릿의 표면에 씨앗층을 형성하는 단계,
상기 씨앗층 위에 금속을 도금한 후 상기 고분자 템플릿과 상기 알루미늄 템 플릿을 제거하여 금속 몰드를 형성하는 단계, 그리고
상기 금속 몰드 위에 세포 배양용 고분자 물질층을 형성 한 후 상기 금속 몰 드를 제거하여 나노 구조물이 형성된 고분자 물질로 이루어지는 세포 배양면을 형 성하는 단계
를 포함하는 나노 구조를 갖는 세포 배양용 용기의 제조 방법 .

제 6항에서 ,
상기 금속 몰드는 니켈, 철, 구리, 은, 금 및 아연 주석 -납 합금 중 적어도 하나를 포함하여 이루어지는 나노 구조를 갖는 세포 배양 용기 의 제조 방법 .

계 6항에서,
상기 세포 배양면올 형성하는 단계는
캐비티를 포함하는 계 1 금형,
상기 캐비티 내에 상기 금속 몰드를 장착하는 단계,
상기 제 1 금형과 일정 한 간격을 두고 제 2 금형을 정 렬하는 단계,
상기 제 1 금형과 상기 제 2 금형 사이에 상기 세포 배양용 고분자 물질충을 형성하기 위한 수지를 주입하는 단계,
상기 수지를 경화시킨 후 상기 제 1 금형과 상기 제 2 금형을 제거하여 상기 세포 배양면이 바닥에 형성된 세포 배양용 용기를 완성하는 단계
를 더 포함하는 나노 구조를 갖는 세포 배양용 용기의 제조 방법 .
