Description
[1] 間葉系幹細胞、組織幹細胞および前駆細胞からなる群より選択される幹細胞を培養するための細胞培養用基板であって、ドットパターンを有し、当該ドットパターンにおける各ドットの直径が50nm以上200nm以下であり、各ドットの配置間隔が50nm以上400nm以下である、前記細胞培養用基板。
[3] 基板および/またはドットパターンの材質が、石英;ガラス;チタン、シリコン、金および白金からなる群より選択される金属、もしくはこれらいずれかの金属を含む合金;コバルト-クロム合金もしくはステンレス鋼;または、ポリスチレン、ポリ乳酸、ポリカプロラクトン、ポリグリコール酸、ポリカーボネート、およびシクロオレフィンポリマーからなる群より選択されるプラスチック;である、上記[1]または[2]に記載の細胞培養用基板。
(1)当該ドットパターンにおける各ドットの直径が50nm以上200nm以下であり、各ドットの配置間隔が50nm以上400nm以下であり、そして各ドットの高さが50nm以上300nm未満である;または
(2)当該ドットパターンにおける各ドットの直径が50nm以上200nm未満であり、そして各ドットの配置間隔が50nm以上200nm未満である;
前記細胞培養用基板。
[7] 基板および/またはドットパターンの材質が、石英;ガラス;チタン、シリコン、金および白金からなる群より選択される金属、もしくはこれらいずれかの金属を含む合金;コバルト-クロム合金もしくはステンレス鋼;または、ポリスチレン、ポリ乳酸、ポリカプロラクトン、ポリグリコール酸、ポリカーボネート、およびシクロオレフィンポリマーからなる群より選択されるプラスチック;である、上記[5]または[6]に記載の方法。
[9] 培養中の幹細胞の核上部アクチンの発達が抑制される、上記[5]~[8]のいずれか一項に記載の方法。
本明細書で特段に定義されない限り、本発明に関連して用いられる科学用語及び技術用語は、当業者によって一般に理解される意味を有するものとする。
一態様において本発明は、細胞培養用基板であって、ドットパターンを有し、当該ドットパターンにおける各ドットの直径が50nm以上200nm以下であり、各ドットの配置間隔が50nm以上400nm以下である、前記細胞培養用基板に関する。
(1)当該ドットパターンにおける各ドットの直径が50nm以上200nm以下であり、各ドットの配置間隔が50nm以上400nm以下であり、そして各ドットの高さが50nm以上300nm未満である;または
(2)当該ドットパターンにおける各ドットの直径が50nm以上200nm未満であり、そして各ドットの配置間隔が50nm以上200nm未満である;
前記細胞培養用基板に関する。
一態様において、本発明は、幹細胞の分化を抑制する方法であって、ドットパターンを有する細胞培養用基板上で当該幹細胞を培養することを含み、当該ドットパターンにおける各ドットの直径が50nm以上200nm以下、各ドットの配置間隔が50nm以上400nm以下である、前記方法に関する。好ましくは、本発明の方法を適用する幹細胞は間葉系幹細胞、組織幹細胞および前駆細胞からなる群より選択される幹細胞(以下、本明細書において、「間葉系幹細胞等の幹細胞」または「間葉系幹細胞等」と表記する場合がある)であり、さらに好ましくは間葉系幹細胞である。
一態様において、本発明は、間葉系幹細胞等の幹細胞の核上部アクチンの発達を抑制する方法であって、ドットパターンを有する細胞培養用基板上で当該幹細胞を培養することを含み、当該ドットパターンにおける各ドットの直径が50nm以上200nm以下、各ドットの配置間隔が50nm以上400nm以下である、前記方法に関する。
アクチンの発達度は、当業者に公知の手法により評価することができ、例えば、細胞のアクチンを蛍光標識したファロイジンで染色し、蛍光顕微鏡により比較観察することにより評価することができる。核上部アクチンは、当業者に公知の手法により評価することができ、例えば、細胞のアクチンを染色した上で、共焦点レーザー顕微鏡等の焦点深度を核の上部に合わせることにより観察することができる。したがって、ドットパターンを有する細胞培養用基板上で間葉系幹細胞等の幹細胞を培養した場合に、ドットパターンを有しない平滑な表面を有する同材質の基板上で当該幹細胞を培養した場合と比較して、核上部アクチンの発達が抑制されている様子が観察された場合に、ドットパターンを有する細胞培養用基板の使用により核上部アクチンの発達が抑制されたと判断することができる。
実施例1:間葉系幹細胞の分化抑制に関するナノドットパターンの影響
ドットパターンを有する石英基板を準備した。具体的には、ドットパターンの各ドットの直径(DF)として100nm、150nm、200nm、250nmまたは500nmの5種、及び各ドットの配置間隔(DI)として100nm、150nm、200nm、250nmまたは500nmの5種、のそれぞれを組み合わせた25種類のドットパターンを有する石英基板を準備した。各ドットの高さは150nmであった。また、対照として、ドットパターンのない平面を有する石英基板を用いた。これらの石英基板を用いて、間葉系幹細胞の培養中のドットパターンの影響を評価した。
(1)評価に用いたすべてのパターン(直径100nm~500nm、配置間隔100nm~500nm、高さ150nm、の範囲内のドットパターン)における間葉系幹細胞の培養において、ドットパターンのない平面を有する石英基板と比較して骨芽細胞への分化を抑制する傾向が観察された;
(2)直径100nm、配置間隔100nm、高さ150nm、のドットパターンにおける間葉系幹細胞の培養では、培養日数に関わらず、骨芽細胞分化の抑制効果が高いことが見出された;
(3)直径100nmのドット構造上での培養により、間葉系幹細胞が分化して生ずる骨芽細胞の割合は、より大きな配置間隔を有するドットパターン上での培養で増加傾向にあった。すなわち、直径100nmのドット構造において、ドットの配置間隔の増大に伴い、間葉系幹細胞の骨芽細胞分化を抑制効果が低下する傾向が観察された。
まず、比較のために、間葉系幹細胞をカバーガラス上で、骨分化誘導培地(Lonza;デキサメタゾン、アスコルベート、β-グリセロホスフェート含有培地)中で培養して、間葉系幹細胞を骨芽細胞へと分化させた。間葉系幹細胞は、骨髄由来のヒト間葉系幹細胞(ATCC)を用い、継代数は5以内とした。
結果を図3および4に示す。
間葉系幹細胞の分化に影響を及ぼす細胞側の因子として、伸展面積およびアスペクト比に関する報告がある。そこで、ドットパターン上、またはドットパターンなしの平面上で培養した間葉系幹細胞について、細胞形態を観察し、細胞の伸展面積-アスペクト比と骨芽細胞分化との関係を分析した。
ドットパターン上で培養した細胞と平面上で培養した細胞の伸展面積-アスペクト比の分布を比較した。ドットパターン上で培養下細胞の伸展面積-アスペクト比の分布は、平面上で培養した細胞とは傾向が異なった。同じ培養日数で比較すると、ドットパターン上で培養した細胞の伸展面積は、平面上で培養した細胞よりも小さい。ドットパターン上で培養した細胞は、培養3日目では伸展面積5,000μm2よりも小さい領域に、培養14日目では伸展面積が10,000μm2よりも小さい領域に分布する頻度が高かった。平面上で培養した細胞の伸展面積-アスペクト比はこれらの領域の外に分布する頻度が高かった。
Claims (7)
- 間葉系幹細胞の分化を抑制する方法であって、細胞外マトリックスを構成するタンパク質でコーティングされたドットパターンを有する細胞培養用基板上で当該間葉系幹細胞を培養することを含み、当該基板およびドットパターンの材質は石英であり、ドットパターンにおける各ドットの直径が50nm以上200nm以下、各ドットの配置間隔が50nm以上400nm以下である、前記方法。
- 細胞培養用基板上のドットパターンにおける各ドットの高さが、50nm以上1000nm以下である、請求項1に記載の方法。
- 細胞培養用基板のコーティングに用いるタンパク質が、フィブロネクチン、コラーゲン、ラミニン、またはビトロネクチンである、請求項1または2に記載の方法。
- 各ドットの直径が100nm、配置間隔が100nmである、請求項1~3のいずれか一項に記載の方法。
- 培養中の間葉系幹細胞の核上部アクチンの発達が抑制される、請求項1~4のいずれか一項に記載の方法。
- 間葉系幹細胞の核上部アクチンの発達を抑制する方法であって、ドットパターンを有する細胞培養用基板上で当該間葉系幹細胞を培養することを含み、
当該ドットパターンにおける各ドットの直径が50nm以上200nm以下、各ドットの配置間隔が50nm以上400nm以下、各ドットの高さが、50nm以上1000nm以下であり、
