Abstract
Description
柑橘系の原料(脱汁液)からエタノールを製造する方法については、脱汁液を濃縮して得られる柑橘糖蜜を原料とする第一の方法と、柑橘糖蜜を生成することなく脱汁液を直接の原料とする第二の方法がある。
第一の方法については、すでに検討されている(例えば、特開2002-153231号公報)。この方法の場合、濃縮工程においてリモネンを除去できる利点がある反面、濃縮工程に多大なエネルギーが必要となると共に、発酵プロセスが高濃度の糖液に対応できないため、糖濃度20%以下になるように水で希釈する必要がある。そのため、経済的なエタノール製造プロセスの確立という観点からは問題があった。
一方、第二の方法については、脱汁液中には糖分が10%前後含まれており、発酵には最適な濃度であり、希釈や濃縮することなくそのまま発酵することができれば、経済的なエタノール製造プロセスの確立が可能となる。しかしながら、発酵阻害物質であるリモネンに対応する技術を確立する必要がある。
リモネンは、大部分が皮に含有されている。しかし、搾汁工程や脱汁工程で、果皮を含む果実を機械的に絞ることにより、その一部は液中に放出される。前述した工程により皮から遊離したリモネンは、脱汁液中において主としてミセル状で存在している。発明者の行った測定結果によれば、皮含有リモネンとミセル状リモネンを合計した脱汁液中のリモネン(0.2~0.5wt%)のうち、ミセル状で存在しているリモネン濃度は0.01~0.04wt%と微量である。
ところが、YPD培地にミセル状(油状)のリモネンを添加し、一般酵母を使ってエタノール発酵させた実験の結果によれば、添加したリモネン濃度が高くなると、エタノール生成率が急激に低下した。例えば、リモネンを0.1%v/v添加した場合は、エタノール生成率は約20%まで大きく低下した。一般に、酵母は糖液中に固体微粒子として存在し、油状の物質を吸着しやすい。そのため、このような実験結果となった理由は、添加された微量(0.05%v/v)のリモネン(油状)が酵母に吸着し、発酵阻害を引き起こしたものと推定される。従って、脱汁液の中から発酵阻害物質であるリモネンを除去するためには、リモネンの存在比率で75%以上を占める固形分を除去するとともに、軽液分(油状物質)として存在するミセル状のリモネンをも除去する必要があった。ただし、三相遠心分離等の処理によって事前に原料からリモネンを除去しても、従来の一般酵母には誘導期間があるため、発酵が安定せず、発酵速度も遅かった。そのため、コンタミネーション等の影響で発酵性が不安定になりやすく、長期に亘って高い発酵性を維持することが困難であった。
本発明は、かかる事情に鑑みてなされるもので、発酵阻害物質リモネンに耐性を有するアルコール発酵性酵母及びこれを用いたエタノール製造方法を提供することを目的とする。
なお、以下「リモネン耐性」とは、原料中、濃度0.1~0.5wt%のリモネンの存在下において、86%以上の収率でエタノール発酵することが可能な酵母の性質をいう。「高温耐性」とは、発酵温度40℃以上の環境下において、86%以上の収率でエタノール発酵することが可能な酵母の性質をいう。また、リモネン耐性及び高温耐性のないサッカロマイセス・セルビジエ(Saccharomyces.Cerevisiae)を「一般酵母」ともいう。
以下、本発明を実施例により説明する。
愛媛県内で収穫された柑橘類ジュース工場の製造ラインからみかん脱汁液を採取した。このみかん脱汁液を1000倍の濃度に希釈し、YPD寒天培地(酵母抽出物1%、ペプトン2%、グルコース2%含有)に塗沫し、30℃にて約2日間培養した。
培養シャーレにコロニー形成した各種酵母を、各々ピックアップしてYPD培地(酵母抽出物2%、ペプトン2%、グルコース10%含有、pH6.5~7.0)にて純粋培養した。
純粋培養した各酵母の中から、リモネン耐性を有する酵母をスクリーニングした。
具体的には、リモネン濃度が0.02~0.2%v/vの範囲でそれぞれ異なる数種類の糖液を原料として、それぞれエタノール発酵を行った。その結果、各リモネン濃度の糖液に対し、エタノール発酵性の優れた株をリモネン耐性株とした。そして、これらリモネン耐性株の中でも特にリモネン耐性が高い株を寄託した(受託番号 NITE BP-890)。以下、本明細書中において、この寄託した株を「BP-890株」と呼ぶ。BP-890株は、本出願人の申請により以下の寄託機関に寄託されている。

BP-890株からDNAを抽出し、D2LSUrDNA領域をPCRにより増幅した。得られたPCR産物について、ダイレクトシーケンスにより塩基配列を決定した(図1参照)。この塩基配列に基づいて、データベース上で既知の微生物のD2LSUrDNA配列とのブラスト(BLAST)検索を行い、近縁種と推定される微生物を特定した。その結果、表1に示すように、BP-890株はサッカロマイセス(Saccharomyces)属セルビジエ(Cerevisiae)種の酵母であることが明らかとなった。
BP-890株は、以下の菌学的性質を有する。
1.細胞形状 :球状(図2参照)
2.コロニー形状 :白色(光沢無)、皺状
3.増殖形式 :多極出芽
4.最適生育温度 :30℃
5.最適生育pH :4.0
6.凝集性 :なし
7.資化糖 :スクロース、グルコース、フルクトース
BP-890株について、任意の濃度のリモネンを添加してバッチ発酵(振とう培養)を行い、エタノール発酵性を調べた。実験条件は以下の通りである。
<実験条件>
1.発酵温度:30℃
2.振とう数:120rpm(振幅10mm)
3.発酵時間:24時間
4.原料:YPD培地
図から明らかなように、一般酵母については、リモネン濃度が高くなるに従って、エタノール生成率が低下する。一方、BP-890株については、リモネン添加濃度が0.1%v/v程度になるまで、エタノール生成率は90%を超え、エタノール生成率が低下しなかった。
YPD培地を用いて、発酵温度30℃、37℃、40℃、42℃、45℃にて、24時間バッチ発酵実験を実施した。実験条件は以下の通りである。

1.振とう数:120rpm(振幅10mm)
2.発酵時間:24時間
3.原料:YPD培地
リモネン存在下でバッチ培養を行い、エタノール生成及び酵母の生育について調べた。実験条件は以下の通りである。

1.発酵温度:30℃
2.攪拌速度:120rpm
3.発酵時間:24時間
4.原料:みかん(温州みかん)脱汁液を三相遠心分離して得られた重液分によりpH3.5に調整)
BP-890株についての結果を図5(A)に示す。図中、横軸は時間を表し、縦軸はスクロース(ショ糖)濃度(g/L)、グルコース(ブドウ糖)濃度(g/L)、フルクトース(果糖)濃度(g/L)、エタノール濃度(g/L)、及び酵母濃度(cells/ml)を示している。
図5(B)に示す一般酵母については、8時間程度の誘導期間が存在した。しかし、BP-890株については誘導期間が存在せず、培養開始後より増殖を開始した。また、BP-890株は、一般酵母に比べて、酵母増殖速度及びエタノール生成速度ともに速かった。このような特性により、BP-890株をエタノール発酵に用いた場合、一般酵母に比べてより短期間で安定した培養が可能であり、かつ平均滞留時間の短い効率的な発酵が可能である。
1)硝酸添加によりpH3.5に調整したみかん脱汁液(三相遠心分離なし)
2)みかん脱汁液を三相遠心分離して得られた重液分(pH調整なし)
3)みかん脱汁液(三相遠心分離なし、pH調整なし)
なお、三相遠心分離の効果はリモネンの除去である。ここで、BP-890株は、リモネン耐性を有することからリモネンの除去は不要とも思われる。しかし、1)BP-890株がリモネン耐性を有するとはいえ、リモネン濃度が低ければエタノール生成率の更なる向上が期待できること、及び2)三相遠心分離によりリモネンを完全に除去することはできないことから、みかん脱汁液に対する三相遠心分離の有無を条件として考慮している。また、硝酸添加の効果は、雑菌繁殖の抑制である。三相遠心分離と硝酸添加の効果の詳細については後述する。
これら条件による結果をそれぞれ表2~表4に示す。
なお、発酵後については、発酵後(1)及び発酵後(2)として、データを2回計測している。平均エタノール生成率(%)は、この2回計測したエタノール生成率(%)の平均である。
みかん脱汁液を三相遠心分離して得られた重液分を原料として流加培養を行い、エタノール生成及び酵母の生育について調べた。実験条件は以下の通りである。
<実験条件>
1.温度 :30℃
2.攪拌速度 :150rpm
3.原料供給 :3.3ml/min
4.リアクタ容量:5L
5.空気吹込み量:0.05vvm
6.添加酵母量 :3.0%v/v
7.原料 :みかん(温州みかん)脱汁液
結果を図6に示す。図中、横軸は時間を表し、縦軸はエタノール濃度(g/L)及び酵母濃度(cells/槽)を示している。
図から明らかなように、三相遠心分離処理したリモネン濃度の低い液において、一般酵母については約4時間の誘導期間が存在する。これに対して、BP-890株については誘導期間が存在せず、培養開始直後より増殖を開始している。また、BP-890株は、培養開始直後よりエタノール生成を開始し、15時間目にはエタノール生成がピークに達した。つまり、BP-890株は、一般酵母よりもエタノール生成速度が速いことが分かった。即ち、三相遠心分離したリモネン濃度の低い原料に対してもBP-890株は、誘導期間がなく、安定して発酵することができる。
BP-890株を使用し、伊予かんを原料の一例としてエタノール製造を行った。まず、その製造方法について説明する。
エタノール製造方法は、図7に示すように前工程と後工程で構成されている。前工程は、搾汁工程P1、混合工程P2、及び脱汁工程P3を有する。後工程は、三相遠心分離工程P4、酸混合工程P5、発酵工程P6、及び蒸留工程P7を有する。以下、各工程について説明する。
三相遠心分離工程P4にて、脱汁液13に含まれる発酵阻害物質であるリモネンを、軽液分(油状物質)16、及び固形分17として共に除去する。これにより、重液分15の液中リモネン濃度を0.01wt%以下とすると共に、固形分を含めた全体のリモネン濃度を0.1wt%以下に低減することが可能である。
なお、本工程で使用する三相遠心分離機は、比重差の少ない軽液、重液を分離することから、沈降面積を大きくすることが可能なディスク型の遠心分離機を用いることが好ましい。遠心力としては、工業的には5000G以上有すれば、分離可能である。
硝酸は三相遠心分離工程P4にて生成された重液分15よりも比重が重いため、硝酸添加後は常時又は定期的に攪拌する。
<連続発酵条件>
1.発酵温度 :30℃
2.攪拌速度 :150rpm
3.発酵時間 :24時間(滞留時間)
4.添加酵母量 :3.0%v/v
5.リアクタ容量 :5L
6.希釈率 :0.04(1/h)
7.滅菌空気吹込量:0.05vvm
8.その他 :連続発酵中に希釈率Dを0.014~0.04の範囲で変更
本実施例では、希釈率Dを0.014~0.04の範囲で変更したが、希釈率Dの変更にかかわらず、安定してエタノールが生成された。通常、一般酵母を用いた連続発酵において希釈率Dを変更させた場合、発酵性は不安定であり、経過時間に伴いエタノール生成量および酵母数は減少し、発酵不良となる。平均エタノール生成率は、105.4%であった。なお、平均エタノール生成率が100%を超えているが、その理由は以下の2つが考えられる。第1に、平均エタノール生成率を計算する際に考慮した糖は、スクロース、グルコース、及びフルクトースであり、酵母がこれら以外の糖を資化したためであると考えられる。第2に、測定誤差により平均エタノール生成率が大きめに算出されたものと考えられる。
本実施例は、遠心分離工程P4を省略している点と希釈率Dの変更範囲の2点が実施例3と相違する。即ち、脱汁工程P3にて脱汁液を生成した後、遠心分離工程P4を省略し、酸混合工程P5にて、脱汁液に硝酸を添加しpH3.5の酸混合液を生成した。酸混合液をバイオリアクタ22に送り込み、発酵工程P6を実施した。なお、脱汁液13のpHを調整した酸混合液のリモネン濃度(固形分に付着したリモネンと液中に存在するリモネンの合計値)は0.2%v/vであった。
本実施例では、希釈率Dを0.04~0.08の範囲で変更したが、希釈率Dの変更にかかわらず、安定してエタノールが生成された。平均エタノール生成率は、88.5%であった。
本実施例は、連続発酵装置20(図8参照)の運転条件が前述の実施例3と相違する。具体的には、一般酵母及びBP-890株を使用し、それぞれについて、ジュース工場の運転に合わせて発酵処理と発酵処理の停止とを繰り返す運転条件にて連続発酵させた。なお、脱汁液13のpHを調整した酸混合液のリモネン濃度は約0.03vol%であった。
<運転条件>
1.エタノール発酵させる発酵期間(12時間)と、原料供給を停止し発酵を停止する停止期間(12時間)を繰り返す(原料を半日で処理し、残りの半日は処理を停止する)。
2.発酵期間中の希釈率Dは、0.08(平均滞留時間12時間)とする。
3.停止期間中、バイオリアクタ22の内部温度は30℃に維持した。攪拌及び滅菌空気の吹込は継続する(原料供給のみを停止した)。
また、同図(B)中、横軸は経過時間(h)を示し、縦軸はスクロース濃度(g/L)、グルコース濃度(g/L)、フルクトース濃度(g/L)、全糖濃度(g/L)、エタノール濃度(g/L)、流入液3糖の合計濃度(g/L)及び酵母濃度(cells/ml)を示している。
本実施例では、生育阻害物質であるリモネン存在条件下で原料供給を停止した状態において、リモネン耐性をもたない一般酵母については、リモネンによる生育阻害を受けることに加え、栄養不足等による生育への悪影響をも受けることになる。そのため、酵母生育速度の著しい低下や酵母の死滅を招き、酵母数が減少して、その後、発酵性が悪化した。一方、BP-890株については、原料供給を停止した状態においても酵母の減少はみられず、安定して高い発酵性が維持された。即ち、BP-890株は、ジュース工場の稼働に合わせて夜間に供給を停止するような一時的な原料供給停止条件下において、一般酵母は安定的な操業が不可能であるが、リモネン耐性のあるBP-890株は、安定的な発酵が可能であることが判明した。
前述の実施例3及び実施例4においては、脱汁工程P3にて得られた脱汁液13からエタノール発酵を行ったが、搾汁工程P1にて得られた搾汁残さ11を原料としてエタノール発酵(固体発酵)を行うことも可能である。
Claims (9)
- リモネンに耐性を有するサッカロマイセス(Saccharomyces)属のアルコール発酵性酵母。
- 請求項1記載のアルコール発酵性酵母において、更に高温耐性を有するアルコール発酵性酵母。
- 請求項1又は2記載のアルコール発酵性酵母において、サッカロマイセス・セルビジエ(Saccharomyces.Cerevisiae)であるアルコール発酵性酵母。
- 請求項3記載のアルコール発酵性酵母において、サッカロマイセス・セルビジエ(受託番号 NITE BP-890)であるアルコール発酵性酵母。
- 請求項1~4のいずれか1項に記載のアルコール発酵性酵母を使用するエタノール製造方法。
- 請求項5記載のエタノール製造方法において、柑橘類を原料とするエタノール製造方法。
- 請求項5記載のエタノール製造方法において、柑橘類脱汁液を三相遠心分離して得られた重液分を原料とするエタノール製造方法。
- 請求項5記載のエタノール製造方法において、酸性物質が添加された柑橘類脱汁液を原料とするエタノール製造方法。
- 請求項8記載のエタノール製造方法において、前記酸性物質が硝酸であるエタノール製造方法。
