リグノセルロース系バイオマスからエタノールを製造する方法
 本発明は、木質系バイオマスやソフトバイオマス等のリグノセルロース系バイオマス中のヘミセルロース又はセルロースを原料として、安価かつ効率よく糖化し、さらに酵母を用いてエタノール発酵させることによるエタノール製造方法に関するものである。
 木質系バイオマスをはじめとするリグノセルロース系バイオマスは、ヘミセルロース約20%、セルロース約50%、リグニン約30%から構成される。ヘミセルロース及びセルロースは、糖化処理によって糖類へと分解し、さらに酵母等の発酵微生物を用いて発酵させることにより、エタノールを製造することが可能である。ヘミセルロースの糖化によってC5系糖類及びC6系糖類が得られ、セルロースの糖化によってC6系糖類が得られる。
 ここで、C5系糖類とは、キシロース、アラビノース等の5炭糖とそのオリゴ糖をいう。また、C6系糖類とは、グルコース、ガラクトース等の6炭糖とそのオリゴ糖をいう。
 リグノセルロース系バイオマスの代表的な糖化方法としては、濃硫酸法と希硫酸法が挙げられる。濃硫酸法は、糖化効率が高いものの、70~80%の高濃度の硫酸を50~100℃付近で用いるために、耐酸性に優れた高価な設備が必要となり、硫酸回収コストもかかる。一方、希硫酸法の場合、ヘミセルロースの糖化では高い糖収率(65~90%)が得られるが、セルロースの糖化では非常に糖化率(25~40%)が低いという欠点があった(非特許文献2、非特許文献3)。
 リグノセルロース系バイオマスを、希硫酸を用いて150~180℃×数分間加水分解処理すると、まずヘミセルロースが加水分解されてC5系糖類であるキシロース、アラビノース等の5炭糖及びそれらのオリゴ糖や、C6系糖類であるグルコース、ガラクトース、マンノース等の6炭糖及びそれらのオリゴ糖が得られる(第一糖化工程)。
 ヘミセルロースの糖化後、固形分としてリグニンとセルロースからなる残渣が得られる。この残渣を、希硫酸を用いて230~250℃×1~3分間加水分解すると、セルロースから6炭糖であるグルコース及びそのオリゴ糖が得られる(第二糖化工程)。
 第一糖化工程では、ヘミセルロースの糖化率は80%以上であるが、第二糖化工程では、セルロースの糖化率は30~40%程度に留まり、糖化率が低い。
 上述したような二段階糖化方法では、第一糖化工程及び第二糖化工程において、糖質原料をそれぞれスラリー化するために、エタノール発酵後のエタノール濃度が低くなる。このため、エタノール蒸留工程でのエネルギー消費量が多くなり、高いエネルギー効率の確保が難しい。
 また、第一糖化工程で糖化されなかったヘミセルロース成分の固形残渣は、第二糖化工程では処理温度が高いために発酵阻害物質である酢酸、蟻酸等の有機酸まで分解されたり、レブリン酸、フルフラール、5-HMF等が生成され、後段のアルコール発酵に悪影響を及ぼすことが知られている。
 濃硫酸法及び希硫酸法による糖化技術の問題点を解決するために、特許文献1~5に開示されているようなセルロース成分を酵素で糖化する取り組みも進められている。これらの方法では、酵素糖化の前処理としてリグノセルロース系バイオマスを酸加水分解し、固液分離し、固形残渣をさらに粉砕、微粉砕等したり、あるいはアルカリ薬剤による処理等を行っている。
 また、ヘミセルロースを酸加水分解し、糖化液を中和して発酵させると共に、残渣は粉砕処理して酵素加水分解した後で発酵させるエタノール製造方法が、特許文献5に開示されている。

 しかし、希硫酸によってリグノセルロース系バイオマスを糖類に加水分解する従来技術では、(1)生成された糖類がさらに分解するために糖化率が低くなる、(2)エタノール発酵後のエタノール濃度が低く、エタノール蒸留工程でのエネルギー消費量が多いために経済性の確保が難しい、(3)ヘミセルロース、セルロースの糖化と共にリグニンが溶解し、溶解したリグニンが後段のエタノール発酵を阻害する、(4)C5系、C6系糖化液の同時発酵は難しい(C6系糖化液のみエタノール化する)、という問題があった。
 また、非特許文献1に開示されているような、ソフトバイオマス等のリグノセルロース系バイオマスの酵素糖化においては、生成糖類の分解が起こらない利点があるが、リグニンが充分除去されずセルロースを覆っているため、効率的な酵素糖化反応が行われにくいという欠点がある。このために、多量の酵素が必要となり、経済性確保が難しいという問題があった。
 本発明は、リグノセルロース系バイオマスを原料とした、安価かつ効率的なエタノール製造方法の提供を目的とする。
 本発明者らは、リグノセルロース系バイオマスを爆砕処理又は亜臨界処理した後の固形残渣について、エタノールに浸漬することによってセルロースを覆っているリグニンを除去可能であることを見出し、本発明を完成させるに至った。
 具体的に、本発明は、
 リグノセルロース系バイオマスからエタノールを製造する方法であって、
 リグノセルロース系バイオマスを爆砕処理するか、亜臨界状態で加水分解した後フラッシュ処理するリグノセルロース分解工程と、
 分解工程後の固形残渣をエタノール浸漬してリグニンを除去するリグニン除去工程と、
 リグニン除去工程後の固形残渣を、酵素によって糖化し、さらにエタノールへと発酵させるC6系糖化発酵工程と、
を有することを特徴とする方法に関する。
 本発明では、リグノセルロース系バイオマスを爆砕処理等して糖化処理し、固液分離にて糖化処理液(液相)と固形残渣とに分離する。そして、固形残渣を水洗浄することにより、C5系糖類を充分に回収する。このC5系糖類を含む洗浄水と固液分離した糖化処理液(爆砕処理液又は亜臨界処理液)を混合し、C5系糖化液とする。
 水洗浄した固形残渣には、爆砕処理等によって糖化されないセルロースが残存しているが、そのままではメイセラーゼ等の酵素を添加しても、セルロースがリグニンによって被覆されているため、セルロースを糖化してC6系糖類へと高い効率で分解することは困難である。しかし、本発明では、リグニン分解工程として、固形残渣をエタノールに浸漬させることによってリグニンを溶解及び除去することにより、酵素によるセルロース糖化率が向上する。
 分解工程後の液相を濃縮した後、C5系酵母によってC5系糖化液のエタノール発酵を行うC5系発酵工程を有することが好ましい。
 糖化処理液(液相)及び固形残渣の洗浄水には、C5系糖類(キシロース、アラビノース、キシロオリゴ糖等)が含まれているが、そのままではC5系糖類濃度が低いため、逆浸透膜装置等を用いて濃縮し、C5系糖類濃度を3%~6%程度とする。この濃縮操作によって、C5系発酵微生物によるエタノール発酵速度が向上する。
 前記C5系発酵工程後のC5系発酵液から発酵微生物(C5系発酵微生物)を分離した後、C5系発酵液を前記C6系糖化・同時発酵工程へと供給することが好ましい。
 エタノール濃度の上限濃度は、C5系発酵では3%程度であり、C6系発酵では15%程度である。一貫プラントでバイオマスからエタノールを製造する場合、エタノールの最終濃度はなるべく高くすることが好ましい。エタノール発酵が終了してエタノール濃度3%程度となったC5系発酵液から遠心分離器等の手段によってC5系発酵微生物を分離した後、C6系糖化液に混合することにより、C5系糖化液及びC6系糖化液を個別に発酵させるよりも、蒸留前のエタノール濃度を高くすることができ、エタノール蒸留工程でのエネルギー消費を削減できる。
 前記分解工程前に、リグノセルロース系バイオマスをエタノール又はアンモニア水に浸漬する浸漬工程をさらに有することが好ましい。
 分解工程前に、リグノセルロース系バイオマスをエタノール又はアンモニア水に浸漬することにより、爆砕処理又は亜臨界処理状態で加水分解処理において生成するフルフラール、5-ヒドロキシメチルフルフラール(5-HMF)、有機酸等のエタノール発酵に有害な物質の濃度を低下させることが可能となる。
 エタノールは、無水エタノールを使用してもよく、濃度20%以上の水溶液を使用してもよい。また、アンモニア水は、濃度20%以下のものを使用することがよい。なお、浸漬時間は、1時間以上24時間以下とすることが好ましい。
 C5系発酵液から分離されたC5系発酵微生物を、C5系糖化液のエタノール発酵に再利用することが好ましい。同様に、C6系糖化・同時発酵工程後のC6系発酵液からC6系発酵微生物を分離して、C6系糖化液のエタノール発酵に再利用することが好ましい。
 酵母などの発酵微生物を再利用することにより、エタノール製造のコストを抑制できる。
 本発明の上記目的、他の目的、特徴及び利点は、添付図面参照の下、以下の好適な実施態様の詳細な説明から明らかにされる。
 本発明のリグノセルロース系バイオマスからエタノールを製造する方法では、爆砕処理等とエタノール浸漬とを組み合わせることにより、従来は不十分であったC6系酵素によるセルロースの糖化率が向上する。また、C5系発酵液を爆砕処理等による固形残渣と混合し、糖化・同時発酵させることにより、一貫プラントとして得られるエタノール濃度を最大8%程度にまで高めることが可能となる。さらに、爆砕処理等の前処理としてリグノセルロース系バイオマスをエタノール又はアンモニア水に浸漬することにより、爆砕処理等におけるエタノール発酵の阻害物質の生成量を抑制し、エタノール収率を向上させることも可能である。
図1は、本発明の実施の形態1の工程フロー図である。 図2は、爆砕装置の概念図である。 図3は、本発明の実施の形態2の工程フロー図である。
 以下、本発明の実施の形態について、適宜図面を参酌しながら説明する。なお、本発明は以下の記載に限定されない。
 [実施の形態1]
 本発明の実施の形態1の工程フロー図を、図1に示す。
 (分解工程)
 まず、サトウキビバガス等のリグノセルロース系バイオマス(以下、「バイオマス」と称する)を破砕機又は粉砕機等により平均径30~50mm以下(好ましくは、10mm以下)の小片とする。この小片を爆砕装置にて水蒸気による爆砕処理(200~240℃、1.5~4MPa、1~15min;好ましくは225~230℃、2.5~3Mpa、1~5min)を行い、バイオマス中のヘミセルロース成分を糖化する。
 爆砕装置で処理されたバイオマス(爆砕処理物)には、ヘミセルロースに由来するC5系糖化液、糖の分解物及びリグニン溶解物等の発酵阻害物、固形残渣が含まれる。 この爆砕処理物をフィルタープレス等によって固液分離し、爆砕処理液と固形残渣に分ける。固形残渣は、さらに水で適宜洗浄し、固形残渣に含まれる糖類が回収される。水洗浄によって糖回収率が向上する。
 ここで、爆砕装置の概念図を、図2に示す。所定の爆砕圧力になるように、飽和水蒸気曲線よりボイラ加熱温度を決定し、ボイラ1を立ち上げる。次に、バルブ2及びバルブ3を「閉」とする。所定量のバガスを投入口4から反応器に投入し、密閉する。そして、バルブ2を「開」とし、ボイラ1から反応器5へと水蒸気を供給し、加熱する。
  所定時間加熱後、バルブ2を「閉」とし、直ちにバルブ3を「開」として爆砕処理を行う。固形物(固形残渣)及び爆砕処理液は、圧力開放時にセパレータ6に搬送され、そこで水蒸気と分離され、受器7に爆砕処理液及び固形物が捕集される。そして、受器7を取り外し、爆砕処理液及び固形物を回収する。
 (C5系発酵工程)
 次に、C5系糖類を含む洗浄水を爆砕処理液と混合し、C5系糖化液とする。洗浄水を混合することにより糖化液中の糖濃度は低下する。そこで、C5系糖化液は、逆浸透膜等のC5系糖類を濃縮できる膜を用いて糖濃縮を行なう。糖濃度を高めることによって、C5系糖化液の発酵速度も増大する。また、糖化液量が減少することにより、C5系糖化液の発酵槽を小型化することも可能となる。
 糖濃縮後、C5系糖化液はC5系糖化液の発酵槽に送られ、C5系糖化液の発酵微生物(C5系発酵微生物、例えば、Pichia stipitis)によって27~35℃、48~72時間、エタノール発酵させる。
 C5系糖化液は、C5系発酵液となり、エタノール濃度は最大3%程度にまでエタノール発酵させることができる。C5系発酵微生物を遠心分離器によって分離した後、C5系発酵液はそのままプラント外へと取り出すことも可能であるが、本発明では、後述するように、爆砕処理等による固形残渣と混合し、酵素とC6系糖化液の発酵微生物(C6系発酵微生物)によって同時に糖化及びエタノール発酵させることが好ましい。
 なお、C5系糖化液にキシロオリゴ糖が含まれている場合には、外部からキシラナーゼ、等の酵素を投入すれば、キシロオリゴ糖が単糖に分解され、C5系糖化液のエタノール発酵率がより向上する。
 (リグニン除去工程)
 爆砕処理によって得られた固形残渣は、水洗浄されて糖類及びリグニン溶解物が除去された後、エタノール濃度30%以上、好ましくは50%以上エタノール水に常温で0.5時間以上48時間以下、好ましくは1時間以上24時間以下の時間浸漬され、セルロースを被覆しているリグニンが溶解及び除去される。エタノール浸漬後、固形残渣を機械的に脱エタノール処理するか、加熱又は減圧加熱等により脱エタノール処理を行う(高濃度エタノール水を用いる場合には、一旦水洗浄した後に脱エタノール処理してもよい)。
 (C6系発酵工程)
 脱エタノール処理後の固形残渣は、C6系糖化・同時発酵槽に送られる。ここで、上述したC5系発酵液を加え、所定の固形分濃度(10~20%)のスラリーを製造することが好ましい。酵素及びC6系発酵微生物(例えば、Saccharomyces cerevisiae)を所定量(糖質原料:酵素=5~2000:1、好ましくは10~1000:1/発酵微生物菌体量は発酵液量に対して0.1w/v%以上3w/v%以下、好ましくは0.3w/v%以上1.5w/v%以下)投入して、27~35℃、24~48時間、エタノール発酵させる。
 すなわち、本発明のC6系糖化・同時発酵工程では、水ではなくC5系発酵液を用いてスラリーを製造することが好ましい。C6系発酵微生物は、C5系発酵微生物と比較しての耐有機酸性及び耐エタノール性に優れているため、固形残渣を、C5系発酵液を用いてスラリー製造しても、C6系糖化・同時発酵が可能となる。
 爆砕処理された固形残渣では、瞬時の圧力開放により機械的に大きな力がかかり、セルロースのミクロフィブリルがほぐれる。さらにエタノール浸漬することにより、リグニンが極力除去される。その結果、セルラーゼ等の酵素がミクロフィブリル内部に容易に侵入することができ、従来技術よりも糖化反応が起こりやすく、エタノール収率も高くなる。
 C6系糖化・同時発酵槽ではセルラーゼとC6系発酵微生物が混在するために、セルロースの糖化反応が進んでも、発酵微生物によってグルコース等のC6系糖類濃度が常に低く維持できるので、安定したエタノール発酵が実現できる。
 <1.エタノール浸漬による糖化率の向上>
 1辺約30mmのさとうきびバガス小片を原料100gとして、爆砕処理とエタノール浸漬によるリグニン除去とを組み合わせたことによる、C6系糖化・同時発酵工程におけるセルロースの酵素糖化率の変化を調べた。エタノール浸漬を行う場合には、爆砕処理によって得られた固形残渣を水洗浄した後、無水エタノールに室温で1時間浸漬させた。また、セルロースの酵素糖化率は、[((C6系糖類生成量×0.9)/セルロース量)×100]という式で算出した。その結果を、表1に示す。

 セルロースの酵素糖化率は、爆砕処理を行わずエタノール浸漬も行わない場合には10%未満、エタノール浸漬だけを行う場合には約20%であった。25~35atmで5分間爆砕処理した場合でも、エタノール浸漬を行わない条件下ではセルロースの酵素糖化率は73.6~87.9%であったが、同じ爆砕処理を行った後でエタノール浸漬を行うと、30atm及び35atmの場合にはセルロースの酵素糖化率が2割以上向上した。特に、30atm×5分間の条件では、セルロースの酵素糖化率は100%であり、完全にセルロースが分解された。
 爆砕処理後の固体残渣からリグニンを除去した結果、酵素糖化が容易となり、C6系糖類の糖化・発酵率が高くなる。その結果、原料バイオマス単位重量あたりのエタノール生成量は、希硫酸等を用いる酸加水分解方法と比較して、約2倍となる200L/トン・バイオマスとすることもできる。
 <2.エタノール濃度とリグニン除去率>
 上記1の35atm×5分間の条件で爆砕処理したさとうきびバガス小片100gを、エタノール濃度及び浸漬時間を変化させた場合のリグニン除去率の変化を調べた。リグニン除去率は、重量変化から推測した。その結果を、表2に示す。なお、表2の数値の単位は、すべて%である。

 30%エタノール水では、浸漬時間を長くしてもリグニン除去率は30%未満であった。50%エタノール水及び無水エタノールでは24時間浸漬時に最もリグニン除去率が高くなった。同様の実験を繰り返したところ、エタノール濃度は、50%以上とすることが好ましく、浸漬時間は1時間以上24時間以下とすることが好ましいことが確認された。
 <3.爆砕処理前のエタノール水又はアンモニア水への浸漬と有害物質濃度>
 上記1と同じさとうきびバガス小片100gを、爆砕処理前に無水エタノール、20%エタノール水又は20%アンモニア水に室温で24時間浸漬する前処理を行った。その後、上記1と同様に爆砕処理を行い、爆砕処理液の糖濃度(w/v%)と、有害物質として5-HMF、フルフラール及び有機酸の糖分解物質3種類の合計濃度(mg/L)を測定した。その結果を、表3に示す。なお、糖濃度及び糖分解物質3種類の濃度は、高速液体クロマトグラフィを用いて測定した。

 20atm×2分間の爆砕処理した場合には、糖分解物質濃度は爆砕前処理の有無による有意な変化は認められなかった。しかし、35atm×3分間の爆砕処理した場合には、糖分解物質濃度は無水エタノール浸漬で約1/4、20%アンモニア水浸漬で約1/20となった。また、無水エタノール浸漬については、糖濃度が前処理なしの場合よりも2倍以上となった。
 <4.C6系糖化・同時発酵スラリー調製におけるエタノール添加>
 上記1と同じさとうきびバガス小片100gを爆砕処理し、固形残渣を水洗浄した後、無水エタノールに室温で3時間浸漬した。エタノールを水洗浄によって除去し、水又は2%エタノール水を用いてC6系発酵用のスラリーを調製した。セルラーゼ及びC6系発酵微生物(Saccharomyces cerevisiae)を添加し、固形分濃度10%、固形分:酵素=10:1、糖化発酵温度37℃、糖化発酵時間48時間という条件で糖化及びエタノール発酵させた。原料セルロース単位重量あたりに生成されたエタノール重量(エタノール生成原単位:kg/kg)を [爆砕さとうきびバガスから生成したエタノール重量/爆砕さとうきびバガス中のセルロース重量]で算出した。その結果を、表4に示す。

 C6系発酵スラリーを2%エタノール水によって調製したのは、C5系発酵微生物を除去した後のC5系発酵液を用いることを想定したものであるが、エタノールがC6系糖化・同時発酵スラリーに当初から存在しても、酵素糖化・同時発酵は阻害されないことが確認された。
 [実施の形態2]
 実施の形態1では、爆砕装置を用いてさとうきびバガス小片等のバイオマスを爆砕処理するが、爆砕装置を亜臨界(水)装置に置き換えることができる。例えば、図3に示すように、バガス小片を爆砕処理する替わりに、亜臨界状態でバガス小片を加水分解した後、フラッシュ装置によって急激に減圧することでも同様の効果がある。
 亜臨界(水)装置では、亜臨界水温度を160~240℃、処理時間を1~90分間とすることが好ましい。また、亜臨界溶媒は水に限らず、酢酸(例えば、0.1M濃度以下)等の有機酸やエタノール混合溶液であってもよい。
 なお、亜臨界溶媒として水を用いる場合でも、亜臨界処理の前処理として、エタノール又はアンモニア水浸漬を行うと、糖の分解が抑制され、フルフラール等の有機酸の生成を抑制できる。
 [実施の形態3]
 実施の形態1では、糖質原料であるバイオマスを、糖化処理する前に破砕機又は粉砕機を用いて平均径30~50mm以下(好ましくは10mm以下)に細かくしているが、バイオマスを25~35atm×5分間以上の条件で爆砕処理する場合には、爆砕によりバイオマスが100μm以下の微粉末に粉砕されることから、糖質原料が後の工程で容易にハンドリングできるサイズに破砕されるならば、爆砕処理前に平均径30~50mm以下に細かくする必要はない。
 [実施の形態4]
 C5系発酵微生物及びC6系発酵微生物を用いる代わりに、酵素を表層提示している発酵微生物を使用することも可能である。すなわち、特開2008-193935号公報に開示されているようなキシラナーゼ及びセルラーゼを表層提示した発酵微生物を用いると、C5系発酵及びC6系糖化・同時発酵に同じ表層提示発酵微生物が投入可能となる。この場合、発酵微生物培養槽が一つとなり、設備費の削減につながる。
 上記説明から、当業者にとっては、本発明の多くの改良や他の実施の形態が明らかである。従って、上記説明は例示としてのみ解釈されるべきであり、本発明を実行する最良の態様を当業者に教示する目的で提供されたものである。本発明の精神を逸脱することなく、その構造及び/又は機能の詳細を実質的に変更できる。
 本発明のリグノセルロース系バイオマスからエタノールを製造する方法は、化学プラントを使用するバイオエタノール製造分野において、特に有用である。

Claims (7)

  1.  リグノセルロース系バイオマスからエタノールを製造する方法であって、
     リグノセルロース系バイオマスを爆砕処理するか、亜臨界状態で加水分解するリグノセルロース分解工程と、
     分解工程後の固形残渣をエタノール浸漬してリグニンを除去するリグニン除去工程と
     リグニン除去工程後の固形残渣を、酵素によって糖化し、さらにC6系発酵微生物によりエタノールへと発酵させるC6系糖化・同時発酵工程と、
    を有することを特徴とする方法。
  2.  分解工程後の液相を濃縮した後、C5系発酵微生物によってC5系糖類のエタノール発酵を行うC5系発酵工程をさらに有することを特徴とする、請求項1に記載の方法。
  3.  前記C5系発酵工程後のC5系発酵液から発酵微生物を分離した後、C5系発酵液を前記C6系糖化・同時発酵工程へと供給する、請求項2に記載の方法。
  4.  C5系発酵液から分離されたC5系発酵微生物を再利用する、請求項3に記載の方法。
  5.  前記分解工程前に、リグノセルロース系バイオマスをエタノール又はアンモニア水に浸漬する浸漬工程をさらに有する、請求項1に記載の方法。
  6.  前記C6系糖化・同時発酵工程後のC6系発酵液からC6系発酵微生物を分離して再利用する、請求項1に記載の方法。
  7.  C5系発酵微生物及びC6系発酵微生物を用いる代わりに、C5系酵素及びC6系酵素を表層提示した、グルコース及びキシロースをエタノール資化できる発酵微生物を用いる、請求項2に記載の方法。
