技術分野
[0001] 本発明は新規エステル誘導体、その用途、及びその製造方法に関する。詳しくは、 本発明は新規エステル誘導体及びこれを有効成分として含有する化粧料組成物、 医薬組成物、食品組成物等に関する。
背景技術
[0002] トウガラシ属に属する植物体 (以下、トウガラシ類という。 )に含有される天然の辛味 成分であるカブサイシンには、抹消血管拡張作用により血行促進作用があることが知 られている。しかしながら、カブサイシンには強い刺激性を有するという問題点がある 。一方、カブサイシン等のカブサイシノイド類の類似体として、カプシエイト、ジヒドロ力 プシエイト等の力プシノイド類が報告されており、これら力プシノイド類は、カプサイシ ノイド類に比べ、辛味刺激が少ないことからダイエット食品用途等での使用に期待が もたれている (特許文献 1、非特許文献 1)。
[0003] また、力プシノイド類であるノナン酸バニリルにっ 、て、外用塗布時に血行促進作 用を示すことが知られている(特許文献 2)。しかし、バニリルアルコールと脂肪酸がェ ステル結合した力プシノイド類は、安定性の面で必ずしも十分と言えず (J.Agric.Food .Chem. 2001, 49, 4026-4030)、製剤調製等の観点から、より安定性の高い成分が望 まれていた。
[0004] バニリル構造(3—メトキシー4ーヒドロキシフヱ-ル基)を有するアルコールまたは力 ルボン酸と脂肪族カルボン酸または長鎖アルコールとを縮合して得られるエステルイ匕 合物は、上記力プシノイド以外にいくつかの化合物が知られている力 血行促進作用 などの生理活性は報告されて!ヽな ヽ。
例えば下記化合物(1)の合成報告があるが(非特許文献 2)、この化合物の生理活 性に関する報告は無い。

[0011] 本発明は、力プシノイド類の安定な誘導体および該誘導体を含有する組成物を提 供することを目的とする。
課題を解決するための手段
[0012] 本発明者らは、上記目的を達成するために鋭意研究を続けた結果、力プシノイド類 の分子構造中、バニリルアルコール由来のベンゼン環構造を保ちつつ、エステル結 合近傍の構造を変換することによって、安定化を達成した。また、これらの化合物の 一部は、天然の力プシノイド類に比して安定であるだけでなぐ血行促進作用を示す ことを見出し、本発明を完成した。
すなわち、本発明は以下の内容を含む。

これらのリパーゼは、それらを生産する微生物の培養等によって得られるが、巿販 品を好適に使用することができる。かかる市販のリパーゼとしては、 Novozyme 435 (ノ ボザィム社製)、 Lipase AK (天野製薬社製)、 Lipase PL (名糖産業社製)、 Lipase QL (名糖産業社製)等の固定化酵素が挙げられる。

滴下終了してから 90分後、 10%塩ィ匕アンモ-ゥム水溶液 (120 ml)をゆっくり滴下 してタエンチし、 n—へキサン (120 ml)で抽出した。 n—へキサン層を 10%塩化アン モニゥム水溶液(100 ml)、水(100 ml)および飽和食塩水(50 ml)にて洗浄した。 n 一へキサン層を無水硫酸マグネシウムにて乾燥後、濾過して濾液を減圧濃縮し、 8 ーメチルノナン酸ェチルエステルの粗生成物(24. 2 g)を薄黄色油状物質として得 た。 GC— MSにより純度を測定したところ、 97. 5%であった。

[0133] 得られた 8—メチルノナン酸ェチルエステルのうち、 22. 2 gを 500 mlナス型フラス コに入れ、エタノール(77 ml)に溶解させて室温にて 2M NaOH水溶液(77 ml, 1 54 mmol)を 5分かけて滴下した。滴下終了後、 60 °Cの油浴を用いて 90分加熱撹 拌し、 TLCにて原料の消失を確認した後、室温に冷却した。
エタノールを減圧濃縮した後、水(40 ml)、 t ブチルメチルエーテル (80 ml)をカロ えて分液操作を行った。水層に対してさらに t ブチルメチルエーテル (80 ml)で 2 回分層し、洗浄した。続いて水層に対して 2M HC1水溶液 (120 ml)をゆっくり加え て酸性にした後、 n—へキサン(80 ml)にて抽出した。 n—へキサン層を水(80 ml+ 40 ml)および飽和食塩水(40 ml)にて洗浄し、 n—へキサン層を無水硫酸マグネシ ゥムにて乾燥後、濾過して濾液を減圧濃縮し、 17. 3 gの 8—メチルノナン酸を薄黄 色油状物質として得た。このうち、 15. 3 gに対して減圧蒸留を行い、 12. 7 gの 8—メ チルノナン酸を薄黄色油状物質として得た。 GC— MSにより純度を測定した所、 99 . 9%以上であった。 6 ブロモー n—へキサン酸ェチルエステルからの総収率 81%

[0159] [試験方法]バニリル 8—メチルノナノエートの PTLCによる単離精製
実施例 1のホモバニリルアルコールの代わりにバニリルアルコール (1. 70 g, 11. 0 mmol)を、 8—メチルノナン酸の代わりに n—デカン酸 (1. 72 g, 10 mmol)を用い て同様の反応を行った。残渣を PTLCで精製したところ得られたバニリル デカノエ ートの収量は 1. 33 g(4. 31 mmol, 43. 1 %)であり、バニリル デカノエートがシリ 力ゲルとの接触によって分解していること (すなわち不安定であること)が示された。一 方、同様の操作を行って得たホモバニリル 8—メチルノナノエートの単離収率は前 述のとおり 93. 2 %であり、ホモバニリル 8—メチルノナノエートはシリカゲルと接触 させてもバニリル デカノエートよりもはるかに安定であることが示された。また、同様 に PTLCで単離精製を行っている実施例 2〜23においても、良好な単離収率が示さ れていることから、本発明における化合物は力プシノイド類よりも安定性に優れるもの であることが分力ゝる。

結果を表 2〜4に示す。本発明者らにより新規に合成された、化合物 、 K 1、 K 2、 K—3、 L、 L—l、 L— 2、 L— 3、 L— 4は強い血管拡張効果が確認され、特に 化合物 K 1、 K 2、 K 3でより強い血管拡張効果が確認された。また化合物 I、 I 1、 1— 2、 1— 3、 J—l、 J— 2、 J— 3、 J—4、 J— 5でも弱い血行促進効果が確認され た。

産業上の利用可能性
本発明の化合物は、外用血行促進作用を有することにより、化粧料組成物の活性 成分として有用である。また、交感神経賦活剤等の医薬、ダイエット用途の食品素材 等としても好適に使用可能である。
本出願は、日本で出願された特願 2006— 084298を基礎としており、その内容は 本明細書にすべて包含されるものである。
