Abstract
医薬品、食品素材としても利用可能な安全な免疫賦活剤及びその製造方法を提供することを目的とする。また、コーヒー抽出残渣の新規な利用方法をも提供する。本発明は、コーヒー抽出物を有効成分とする免疫賦活剤を提供する。好ましくは、コーヒー抽出物がアラビノガラクタンを含有する抽出物である免疫賦活剤である。また、この免疫賦活活性が、マクロファージなどの免疫担当細胞の増殖促進に由来することを特徴とする。ここで、免疫担当細胞が、マクロファージ様細胞株RAW264、J774.1、マウス脾細胞、マウス腹腔マクロファージ、マウス樹状細胞のいずれかであることが好ましい。また、これらの免疫賦活剤を含有する組成物は、医薬組成物、食品組成物、化粧品組成物の組成物として利用できる。

免疫賦活剤とその製造方法
技術分野
[0001] 本発明は、コーヒー抽出物を有効成分とする免疫賦活剤に関する。さらに詳しくは 、本発明は、コーヒー抽出物に含有されるァラビノガラタタン(Arabinogalactan ;AG )を有効成分として含有する、免疫賦活剤及びその利用に関する。
背景技術
[0002] 従来、ァラビノガラタタンはカラマツから抽出されたものが主として使用されてきた。力 ラマツ由来のァラビノガラタタン(Larch wood AG ; L— AG)を食品添加物として使 用する場合には、不純物を除くために高度に精製する必要があった。そこで、食経験 のある食品由来の原料から容易に精製できるァラビノガラタタン抽出物の製造方法 およびァラビノガラタタン抽出物が求められていた。
また、カラマツ由来のァラビノガラタタンは分子量が 15000〜: 18000程度であること から、水溶性に優れ、粘度が低いという特徴があった。この特徴を活かし、テクスチャ に変化を与えず食品に水溶性食物繊維として添加することができ、健康を意識した 食品が開発できる。さらに粘度を上げずに固形分濃度を上げることができるため、ィ ンクにおいては色の転写性改善、顔料安定性の向上などの特性が付与される。光沢 と透明性を必要とする食品包材、ラベル、ラップ材等の精密印刷に用いるハイエンド インクには、インクの転写性を高め、新聞、カタログ、段ボール箱等の印刷に用いる口 一エンドインクでは、顔料の安定性を増すことができる。このように、カラマツ由来のァ ラビノガラタタンは様々な用途に使用できる多糖類である。
一方、コーヒー豆中にはァラビノガラタタンが多く含まれている。コーヒー豆由来の ァラビノガラタタンは、カラマツ由来のァラビノガラタタンと比較して、その分子量が大 きいことに特徴がある。分子量が大きいことから、カラマツ由来のァラビノガラタタンの ように粘度を上げず固形分濃度を上げることができない。従ってカラマツ由来のァラ ビノガラタタンのような利用方法は期待できなかった。
従って、コーヒー生豆、コーヒー焙煎豆、コーヒー抽出後の残渣にもァラビノガラクタ
ンが含有されているにもかかわらず、ァラビノガラタタンの資源としての利用はなされ てこなかった。そこで、コーヒー抽出物、特にァラビノガラタタン含有画分の新規な用 途の開発が求められていた。
一方、今後、少子高齢化が一層進み、老人が増加することが予想され、免疫力を 高める新規な医薬、食品素材が求められている。

発明が解決しょうとする課題
[0004] 本発明は、医薬品、食品素材としても利用可能な安全な免疫賦活剤及びその製造 方法を提供することを目的とする。また、コーヒー抽出残渣の新規な利用方法をも提 供する。
課題を解決するための手段
[0005] 上記目的を達成するため、本発明者らは、コーヒー抽出物、より詳しくはァラビノガ ラタタンを含有するコーヒー抽出物の用途に関して鋭意研究を行った結果、免疫賦 活作用があることを見出し、本発明を完成した。
[0006] 本発明は、コーヒー抽出物を有効成分とする免疫賦活剤を提供する。好ましくは、 コーヒー抽出物がァラビノガラタタンを含有する抽出物である免疫賦活剤である。また 、この免疫賦活活性が、免疫担当細胞の増殖促進に由来することを特徴とする。ここ で、免疫担当細胞が、マクロファージ様細胞株 RAW264、 J774. 1、マウス脾細胞( Splenocyte) ,マウス腹腔マクロファージ(Macrophage)、マウス榭状細胞(Dendri tic cell; DC)のレ、ずれかであることが好ましレ、。
[0007] また、本発明は、これらの免疫賦活剤を含有する組成物を提供する。これら組成物 は、医薬組成物、食品組成物、化粧品組成物等の組成物として利用できる。
また、本発明の免疫賦活剤の製造工程としては、コーヒー生豆、コーヒー焙煎豆ま たはコーヒー抽出残渣に水を添加し、加熱する工程と、加熱抽出液を回収し、減圧 濃縮する工程と、減圧濃縮した液体にエタノールを加えて沈殿させる工程とを有する ことを特徴とする。
更に、減圧濃縮した液体にエタノールをカ卩えて沈殿させる工程の後に、沈殿を水酸
化ナトリウム溶液に溶解する工程と、室温で:!〜 48時間、ついで、 50°C〜70°Cで 1 〜48時間攪拌する工程と、 pHを 7· 0〜8. 0に調製する工程と、有機溶媒で抽出す る工程と、タンパク分解酵素によりタンパク質を分解する工程と、水で透析する工程と を設けても良い。
ここで、使用する水としては、例えば、脱イオン水、蒸留水、ミリ Q水等が好ましく用 いられるが、より好ましくは蒸留水である。 pHはより好ましくは、 7. 2〜7. 8、さらに好 ましく ίま、 7. 4〜7. 6、特 (こ好ましく ίま 7. 45〜7. 55である。
[0008] また、本発明の免疫賦活剤は、ァラビノガラタタンの平均分子量が 1万〜 300万で あることを特徴とする。
また、本発明の免疫賦活剤は、ァラビノガラタタンのァラビノース/ガラクトースの比 力 SO. 02-1. 0であることを特徴とする。
また、本発明は、コーヒー抽出物を添加することによりマウス脾細胞または樹状細胞 のインターロイキン 12 (IL— 12)産生量を、コーヒー抽出物無添加の場合に比べ 増加させる方法を提供する。
また、本発明は、コーヒー抽出物投与により、マウス血中インターロイキン一 12 (IL 12)量を、コーヒー抽出物非投与の場合に比べ増加させる方法を提供する。 また、本発明は、コーヒー抽出物を摂取させることによりマウス脾細胞の、マイトジェ ン PMA/Ionomycinによる増殖促進活性を非摂取の場合に比べ高める方法を提 供する。
発明の効果
[0009] 本発明の免疫賦活剤は、 1し_ 12の産生または 1^_ の産生を増強するため、 細胞性免疫賦活作用を有する。そのため、癌の免疫療法や癌の予防への利用が期 待される。また、本発明の免疫賦活剤の有効成分は、従来から食品として用いられて レ、る糖および Zまたは乳酸菌であり、安全であることが知られているため、医薬品とし てだけでなぐ健康食品としても有用である。
発明を実施するための最良の形態
[0010] 以下、本発明の実施例について、図面を参照しながら詳細に説明していく。
本発明の免疫賦活剤は、コーヒー抽出物を有効成分として含有する。コーヒー抽出
物を得るための材料としては、例えば、コーヒーの生豆、コーヒー抽出後の残渣、コー ヒー焙煎豆等を用いることができる力 これらに限られない。
[0011] 本発明において、コーヒーとは、コーヒー属植物をいう。ァカネ科植物に属するコー ヒー属の栽培種は、ァラビ力種、ロブスタ種、リベリカ種の三原種とそれをもとにした数 十品種がある。力ネフオラ種、等が挙げられるがこれらに限られない。
[0012] また、粗精製物(Crude AG)、準精製物(Quasi— crude AG)、高度に精製され たもの等、精製度はどの段階でもよい。要するに免疫賦活作用を有する成分を含有 すればよい。
[0013] コーヒー抽出物は、コーヒー植物体の一部あるいは、コーヒー抽出後の残渣を抽出 処理することにより得られる。抽出対象となる植物部位として、例えば、前記コーヒー の豆の部分が好ましく用いられるがこれに限られなレ、。
[0014] また、抽出に使用される溶媒としては、特に制限されず、極性及び非極性溶媒のい ずれであってもよレ、。当該抽出溶媒として、具体的には、水、酢酸ェチル等の極性溶 媒が例示される。これらの溶媒は単独で用いてもよぐ二種以上を組み合わせて使用 してもよい。前記抽出溶媒として好ましくは極性溶媒であり、より好ましくは水である
[0015] 抽出方法は、粗抽出の場合は、コーヒー生豆、コーヒー焙煎豆またはコーヒー抽出 残渣に蒸留水を添加し、加熱して抽出する。その後、抽出液を減圧濃縮した後、 3〜 4倍量 (V/V)のエタノールを添加し、沈殿を回収し、粗抽出画分とする。
高度に精製する場合には、粗抽出画分を水酸化ナトリウム溶液に溶解し、室温で 数時間、ついで 55°C〜60°Cで数時間攪拌し、硫酸、塩酸等の酸により、 pH7. 0〜 8. 0に調整後、クロロフオルム、酢酸ェチル、ジェチルエーテルで順次抽出し、水層 にトリプシンをカ卩え、 40°C、 48時間反応させ、タンパク質を分解する。このものを蒸留 水に透析し、精製ァラビノガラタタンを得る。
[0016] コーヒー抽出物がァラビノガラタタン(Coffee AG ; Cof—AG)を含有する抽出物で あるとは、前記抽出物がコーヒー由来のァラビノガラタタンを含有することを意味する 。コーヒー由来のァラビノガラタタンの分子量は、好ましくは、 5000以上 300万以下、 より好ましくは、 1万以上 300万以下、さらに好ましくは、 2万以上 300万以下である。 なお、ここでレヽぅ分子量は HPLC (High Performance Liquid Chromatograph)を用レヽ
てゲルろ過クロマトグラフィーにより測定した値である。
[0017] コーヒー抽出物のァラビノース/ガラクトースの比率は、 0· 02-1. 0、より好ましく は 0· 3〜0· 5である。なお、この数値は、 0. 02〜: 1. 0の範囲内であればどの部分で 区切ってもそれなりの効果を発揮するため、この範囲内であれば任意の数値で好ま しい範囲を区切ることができる。
[0018] 前記免疫賦活剤を含有する組成物としては、例えば、医薬組成物、食品組成物等 が挙げられる。
[0019] (医薬組成物)
医薬の分野では、免疫賦活作用を有効に発揮できる量のコーヒー抽出物とともに、 薬学的に許容される担体や添加剤を配合することにより、免疫賦活作用を有する医 薬組成物が提供される。当該医薬組成物は、医薬品であっても、医薬部外品であつ てもよい。
当該医薬組成物は、内用的に適用されても、また、外用的に適用されてもよい。し たがって、当該医薬組成物は、内服剤、静脈注射、皮下注射、皮内注射、筋肉注射 及び/または腹腔内注射等の注射剤、経粘膜適用剤、経皮適用剤等の製剤形態で 使用すること力できる。
当該医薬組成物の剤型としては、適用の形態により、適当に設定できるが、例えば 、錠剤、顆粒剤、カプセル剤、粉末剤、散剤、等の固形製剤;液剤、懸濁剤等の液状 製剤、軟膏剤、ゲル剤等の半固形剤があげられる。
当該医薬組成物の用途としては、抗ウィルス剤、抗癌剤、肝炎の予防'治療剤、アト ピー性皮膚炎の予防'治療剤、花粉症予防'治療剤、整腸剤等があげられる。
[0020] (食品組成物)
食品の分野では、免疫賦活作用を生体内で発揮できる有効な量のコーヒー抽出物 を食品素材として、各種食品に配合することにより、免疫賦活作用を有する食品組成 物を提供することができる。すなわち、本発明は、食品の分野において、免疫賦活用 と表示された食品組成物を提供することができる。当該食品組成物としては、一般の 食品の他、特定保健用食品、栄養補助食品、機能性食品、病院患者用食品等をあ げ'ること力 Sできる。
当該食品組成物としては、例えば、調味料、畜肉加工品、農産加工品、飲料 (清涼 飲料、アルコール飲料、炭酸飲料、乳飲料、果汁飲料、茶、コーヒー、栄養ドリンク等 )、粉末飲料 (粉末ジュース、粉末スープ等)、濃縮飲料、菓子類 (キャンディ、クッキ 一、ビスケット、ガム、グミ、チョコレート等)、パン、シリアル等をあげることができる。ま た、特定保健用食品、栄養補助食品、機能性食品等の場合、カプセル、トローチ、シ 口ップ、顆粒、粉末等の形状であってもよい。
当該食品組成物におけるコーヒー抽出物の配合率としては、適宜実験により決定 できるが、例えば、 0. 01mg/L〜5mgZL、より好ましくは、 0. 05mg/L~lmg/ Lが望ましい。
[0021] 当該食品組成物の用途としては、例えば整腸剤、花粉症予防 ·治療剤、食品添カロ 物、アトピー性皮膚炎用食品等が揚げられる。
[0022] 本発明の免疫賦活剤は、さらに乳酸菌を有効成分として含有することが好ましい。
乳酸菌としては、食品の加工などに通常用いられる乳酸菌類が用いられ、特に、ヒト の腸内に棲んでいる腸内乳酸菌類が好適である。代表的には、ラクトバチルス ·ガセ リ、ラクトバチルス 'カゼィ、ラクトバチルス'ァシドフィルス、ビフイドバタテリゥム'ロンガ ム、ビフイドバタテリゥム 'インファンテイス、ビフイドバタテリゥム'ビフィダム、ビフィドノく クテリゥム'ブレーべ、ビフイドバタテリゥム'アドレツセンテス、ストレプトコッカス'フエ力 リスなどが挙げられる。これらは、単独で用いてもよぐあるいは 2種以上を組み合わ せて用いてもよい。
[0023] 本発明の免疫賦活剤は、上記抽出物を単独でまたは組み合わせて、あるいは上記 乳酸菌との混合物として用いることにより、マクロファージの IL— 12の産生能または 腸管上皮細胞間リンパ球の IFN_ γ産生能を増強することができる。
[0024] (調製例)
乳酸菌培養培地である MRS培地(商品名 Lactobacilli MRS Broth」、 Dif co 社製) 5mlにラクトバチルス Zガセリ JCM1131を接種し、 32°Cで 24時間静置培養し た。この培養液を 100mlの MRS培地に 1 %になるように接種し、 32°Cで 24時間静置 培養した。得られた培養液を 10, 000 X gで 20分間遠心分離し、菌体を回収した。こ の菌体を PBSに懸濁し、 10, 000 X gで 20分間遠心分離し、菌体を回収した。この
操作を 3回繰り返した後、菌体を蒸留水に懸濁した。この懸濁液を 70°Cに 10分間置 レ、て殺菌した後、ドライアイス一エタノール中で急速凍結した。これを凍結乾燥し、ラ タトバチルス'ガセリ乾燥死菌体 0. 73gを得た。
実施例 1

マウス由来のマクロファージ様細胞株である RAW264細胞株(理研より入手可能 R CB00535)を、細胞数が 20 X 105/mlとなるように、 10%FBS (ゥシ胎児血清)を含 む DMEM培地(以下、単に培地という)で希釈した。これを 96穴組織培養プレートに 1穴当たり 50 μ 1を播種し、 37°Cの 5%炭酸ガス培養器内で 2時間培養した。これに 上記調製例で得たコーヒー抽出物を 0· 0625 / g/ml〜0. 5 μ §/πι1の濃度になる ように培地に加え、 1穴あたりの容量を 100 μ ΐとした。比較としてカラマツ由来のァラ ビノガラタタン画分を同じ濃度で培地に添加した。さらに、免疫細胞を刺激する物質、 すなわち免疫応答に優れた物質として一般に知られている LPS (リポポリサッカライド )や conA (コンカナパリン A)を 20 μ g/ml添加した。これらを 5%炭酸ガス培養器内 で 37°Cにて 1〜4時間培養後、細胞の増殖量をモニターした。増殖試験を行う試薬と しては、タカラバイオ株式会社の Premix WST- 1 Cell Proliferation Assay Systemを用レ、、計測装置としては BI〇一 RAD社製の MICROPLATE READE R Model 550を用レヽた。
[0026] マクロファージ様細胞株 RAW264を用いた増殖試験の結果を図 1に示す。マウス マクロファージ様細胞株において、コーヒー由来のァラビノガラタタンを添加すると、コ ントロールに対して有意な増殖促進活性が認められた。また、マウスマクロファージ様 細胞株において、コーヒー抽出残渣抽出物(Crude AG from residue ; CrudeR- AG)、準精 物 (Quasi— crude AG from green coffee beans ; Q. CmdeB— AG)、生豆抽出物(Crude AG from green coffee beans ; CrudeB_AG)の 3 種の粗抽出物を添加することによって、コントロールに対する有意差、または有意傾 向を示す増殖促進活性が認められた。
実施例 2

マウスから脾細胞を調製し、実施例 1と同様に増殖促進活性を調べた。細胞数が 1 00 X 105/mlとなるように、 10%FBS (ゥシ胎児血清)を含む RPMI1640培地(以 下、単に培地という)で希釈した。これを 96穴組織培養プレートに 1穴当たり 50 μ ΐを 播種し、 37°Cの 5%炭酸ガス培養器内で 2時間培養した。これに上記調製例で得た コーヒー抽出物を 0. 125 x gZml〜0. 5 μ g/mlの濃度になるように培地に加え、 1 穴あたりの全量を 100 a 1とした。比較としてカラマツ由来のァラビノガラタタン画分を 同じ濃度で培地に添加した。これらを 5 %炭酸ガス培養器内で 37°Cにて 1〜4時間 培養後、増殖量をモニターした。増殖試験を行う試薬としては、タカラバイオ株式会 社の Premix WST—l Cell Proliferation Assay Systemを用レヽ、計測装置 としては BIO— RAD社製の MICROPLATE READER Model 550を用レヽた。
[0028] マウス脾細胞を用いた増殖試験の結果を図 2 _ 1〜図 2 _ 3に示す。図 2_ 1〜図 2 _ 3は、それぞれァラビノガラタタン含有画分 (コーヒー抽出残渣、生豆抽出物、準精 製物のそれぞれの各粗抽出物)の脾細胞増殖活性を示す。近交系の脾細胞におい てコーヒー由来のァラビノガラタタンを添加すると、コントロールと比較して有意に増殖 活性が亢進していることが観察されている。また、近交系 balb/cマウスではカラマツ 由来のァラビノガラタタンとコーヒー由来のァラビノガラタタンとの間で増殖活性の有 意差が確認されている。また、粗抽出物についてはコーヒー抽出物を高度に精製し た純精製物と同程度力それ以上の増殖活性が認められる。
以上まとめると、カラマツのァラビノガラタタンに比べ、コーヒー抽出物の方が、増殖 促進活性は高かった。また、コーヒー抽出残渣抽出物、準精製物、生豆抽出物のい ずれに関しても有意な増殖促進活性が見られた。
実施例 3

マウス由来の榭状細胞を調製し、サイト力インの発現を調べた。マウス下肢大腿骨 から注射器で無菌的に骨髄細胞を採取し、細胞数が 4 X 106/mlとなるように、 4ng /mlの IL - 4 (和光純薬)と 1 Ong/mlの GM-CSF (和光純薬)、 10 %FBS (ゥシ胎 児血清)を含む RPMI1640培地(以下、単に培地という)で希釈した。これを 24穴組 織培養プレートに 1穴当たり 250 μ 1を播種し、 37°Cの 5 %炭酸ガス培養器内で 1週 間培養した。
尚、 1週間の培養期間のうち 2、 4日目に培地で浮遊細胞を洗浄除去した。これに 上記調製例で得たコーヒー抽出物を 0. 25 μ gZmlの濃度になるように培地に加え、 1穴あたりの全量を 500 a 1とした。比較としてカラマツ由来のァラビノガラタタン画分を 同じ濃度で培地に添加した。
これらを 5%炭酸ガス培養器内で 37°Cにて 20時間培養後、上清を回収した。 IL- 12量の(発現)確認は Immunoassay Kit Mouse IL- 12p40 (BIOSOURCE) を、 IFN— γ量の(発現)確認は Immunoassay Kit Mouse IFN— γ (BIOSO URCE)を用いて行った。計測装置としては BIO— RAD社製の MICROPLATE R EADER Model 550を用レヽた。
[0033] マウス脾細胞培養上清を用いた ELISA試験とマウス榭状細胞培養上清を用いた E LISA試験の結果を図 4—1〜図 4— 3、図 9— 1、図 9— 2に示す。近交系 balb/cマ ウスより単離した脾細胞、及び樹状細胞に 0. 25 / g/mlのコーヒー由来のァラピノ ガラクタンを添加し、 20時間培養したところ、脾細胞上清の IL— 12の濃度がコント口 ールに対して有意傾向で、樹状細胞上清の IL 12、 IFN- γの濃度が共にコント口 ールに対して有意に亢進していることが観察されている(図 4—1〜図 4— 3を参照)。 また、同マウスより単離した脾細胞に 0. 25 x gZmlのコーヒー由来のァラビノガラタ タンを添加し、 37時間培養したところ、上清の IL_ 12、 IFN- yの濃度が共に亢進 する傾向にあり、特にコーヒー抽出残渣粗抽出物において IFN_ 産生に有意差が 認められた(図 9— 1,図 9— 2を参照)。

マウス由来の脾細胞を調製し、マイトジェンである PMA/Ionomycinに対する増 殖促進活性を調べた。細胞数が 20 X 105/mlとなるように、 10%FBS (ゥシ胎児血 清)を含む RPMI1640培地(以下、単に培地という)で希釈した。これを 96穴組織培 養プレートに 1穴当たり 50 μ ΐを播種した。これに PMA (SIGMA)を 50ng/ml、 Io nomycin (SIGMA)を lng/mlの濃度になるように培地に加えて添加し、 1穴あたり の全量を 100 β 1とした。これらを 5%炭酸ガス培養器内で 37°Cにて 24時間培養後、 増殖量をモニターした。増殖試験を行う試薬としては、タカラバイオ株式会社の Prem ix WST- 1 Cell Proliferation Assay Systemを用レヽ、計測装置としては BI O— RAD社製の MICROPLATE READER Model 550を用いた。

上記のァラビノガラタタン投与試験終了後、マウスの血清を回収し、サイト力インの 発現を調べた。 IL— 12量の確認は Immunoassay Kit IL- 12 + p40 (BIOSOU RCE)を用いて行った。計測装置としては BIO— RAD社製の MICROPLATE RE ADER Model 550を用いた。結果を図 5— 2に示す。近交系 balbZcマウスに対
し、 1日あたり 2. 5mgのコーヒー由来ァラビノガラタタン精製物、および抽出残渣由 来粗抽出物を一週間投与したところ、血中のサイト力イン濃度がコントロールに対して 有意に亢進してレ、ることが観察されてレ、る。
[0041] (ァラビノガラタタンの調製)
コーヒー生豆または ーヒー由出残澄 100gに蒸留水 1500〜2000mlをカロ免、 12 1°Cで 2時間抽出した。得られた抽出液を、 lOOOOrpm, 20分間遠心した上清を回 収し、ロータリーエバポレータで減圧濃縮し、 3〜4倍量のエタノールを加え沈殿を回 収した。この沈殿を粗精製物として用いた。図 6 (A)にコーヒー生豆、(B)にコーヒー 抽出残渣からのァラビノガラタタンの調製フローを示す。
さらに精製する場合は、粗精製物 2. 5gを 0. 2M水酸化ナトリウム溶液 100mlに溶 解し、室温(25°C)で 3時間、 55〜60°Cで 3時間攪拌した後、硫酸で pH7. 5に調整 し、クロロフオルム、酢酸ェチル、ジェチルエーテルで順次抽出した後、水層にトリプ シンを加え、 40°C、 48時間反応させ、タンパク質を分解除去した。このものを蒸留水 に透析することにより、精製ァラビノガラタタン 1 · 4gを得た。図 7に調製フローに示す

HPLCによるゲルろ過クロマトグラフィー (カラム: TSK— GEL G6000PW φ 7. 5 mm X 300mm,ガードカラム: TSK— GUARD COLUMN PWH 7. 5 X 75mm, 移動相: 0. 1M NaCl in 0. 1Mリン酸緩衝液 (pH6. 6)、検出器: RI、検出温度: 45 °C、流速: 0. 2ml/min)により、 pullulan (昭和電工製)を標準物質として平均分子 量を測定した。その結果、コーヒー由来のァラビノガラタタンは平均分子量 27000で カラマツ由来のァラビノガラタタンと同程度であった力 分子量分布はカラマツ由来の ァラビノガラタタンよりも幅広レ、ことが分かった。 (図 8参照)
実施例 5
[0043] 実施例 5は、コーヒー由来ァラビノガラタタン投与による血中総 IgE抗体産生抑制効 果を調べた結果にっレ、て示す。コーヒー由来ァラビノガラタタンとカラマツ由来ァラビ ノガラタタンを摂取させたマウスに卵白アルブミン(以下 OVA、 SIGMA)を投与し、 I gE抗体産生を誘導した時のァラビノガラタタンによる血中総 IgE抗体産生の抑制効
果を確認したものである。
具体的には、 OVAによる IgE抗体産生誘導はメスの近交系 balb/cマウスを用レ、、 以下の (A)〜(C)の試験区及び匹数 (n)で実施した。
(A)水(コントロール); n= 6
(B)コーヒー由来ァラビノガラタタン純精製物 2. 5mgZ日; n= 5
(C)カラマツ由来ァラビノガラタタン純精製物 2. 5mg/日; n= 5
[0044] 感作は次のように実施した。初回感作としてマウス 1匹につき OVA10 μ g及びアジ ュバントとして水酸化アルミニウムゲル(SIGMA) 2mgをリン酸緩衝生理食塩水(PB S) 0. 3mlに懸濁した〇VA溶液を準備し、 6週齢のそれぞれのマウスに感作開始初 日及び 4日目に腹腔内投与した。二次感作として OVAを 25mgZmlとなるよう PBS に溶解し、この抗原溶液にマウスの鼻部を約 3秒間浸漬した。この処置を 1回につき 3 度繰り返した。二次感作の操作は初回感作開始日より 10日後から連日 10日間、 1日 朝夕 2回実施した。
ァラビノガラタタンの投与は初回感作の 1週間前より開始した。なお、それぞれのサ ンプルは水に溶解し、自由摂取させた。
[0045] 初回感作開始から 20日後に採血を行い、血清を回収して総 IgE濃度を調べた。血 中総 IgE抗体濃度はモリナガマウス IgEキット(森永生科学研究所)により測定した。 図 10に示されるように、コーヒー由来ァラビノガラタタン純精製物 2. 5mg/日試験 区でコントロール試験区及びカラマツ由来ァラビノガラタタン純精製物試験区に対し て、総 IgE量の減少傾向が確認できた。
実施例 6
[0046] (腸内細菌の増殖試験)
コーヒー由来ァラビノガラタタン (Cof_ AG)の腸内菌叢を構成する菌類に対する 資化性を、他の糖類と比較した結果を含めて以下に示す。
前段階培養には、 GAMブイヨン液体培地を用い、試験の培地には Pepton— Yea st-Fildes solution (PYF)液体培地に供試糖類を添加した後、オートクレーブ滅 菌したものを用いた。上記 PYF培地は、下表 1の組成からなるものである。また、 Fild es溶液は、下表 2のように調製されるものである。

[0049] 上記の成分を混合し、 55°C温浴槽水中で 1夜保持し、消化させた。これに 20%Na OH溶液 12mlを加えた後、 NaOHにより ρΗ7· 6になるように調整し、フィルタ一によ る滅菌を行った。

GAMブイヨンで培養した新鮮な菌を、供試糖類を添加した PYF培地に、各菌株が 各々 107〜108CFU/チューブとなるように接種し、 37°Cで 72時間嫌気培養した。 菌数の増殖は、接種後 72時間後に、また接種後 72時間後に培地の pHの低下によ る菌増殖の判定を行った。判定基準は、 (サンプノレの pH)— (糖無添加培地の pH) = [pH]とし、 [pH] < 0. 5を(―)、 0. 5≤[ρΗ] < 1. 0を(土)、 1. 0≤[pH] < 1. 5を(+ )、 1. 5≤[pH]を(++)とした。
なお、試験法および判定法に関しては、文献「Suzuki et al, Utilization by Intestin al Bacteria and Digestibility of Arabino— oligosaccharides In Vitro(J. Japan. Soc.Hort. Sci.73(6) : 574-579,2005)」を参照レ、ただきたレ、。
炭素源に関しては、表 1に示すように、グルコース(対照)、コーヒー由来ァラビノガ ラタタン、カラマツ由来ァラビノガラタタンを用いた。また、供試菌株については、下表 3に示す。試験結果を、下表 4および表 5に示す。

[0054] 上記表 4から明らかなように、コーヒー由来ァラビノガラタタンは、腸内有用細菌であ る Bifidobacterium属に良好に資ィ匕され、 Bifidobacterium longumにはカラマツ 由来ァラビノガラタタンと同程度に資化され、 Bifidobacterium pseudocatenulat umにはカラマツ由来ァラビノガラタタンよりもよく資化されたことがわかる。
また表 5の結果より、コーヒー由来ァラビノガラタタンは腸内有害菌とされている Clos tridium属ゃ Escherichia coliに対してほとんど資化されなかった。
ここで、 Bifidobacterium属はヒトにおける腸内有用細菌の代表的な菌種として知 られている。
以上のことから、コーヒー由来ァラビノガラタタンを摂取することにより腸内環境が改 善されるなどのプレバイオテイクス効果が期待できることが理解されるであろう。
産業上の利用可能性
[0055] 本発明の免疫賦活剤は、マクロファージの増殖を促進するため、細胞性免疫賦活 作用を有する。そのため、癌免疫療法や癌予防、ウィルス疾患の予防'治療薬として の利用が期待される。また、健康食品、コーヒー抽出残渣の有効利用法としても利用 可能である。

Claims
[1] コーヒー抽出物を有効成分とする、免疫賦活剤。
[2] コーヒー抽出物がァラビノガラタタン(Arabinogalactan ;AG)を含有する抽出物で ある請求項 1の免疫賦活剤。
[3] 免疫賦活活性が、マクロファージなどの免疫担当細胞の増殖促進に由来することを 特徴とする、請求項 1の免疫賦活剤。
[4] 免疫担当細胞力 マクロファージ様細胞株 RAW264、J774. 1、マウス脾細胞(Sp lenocyte) ,マウス腹腔マクロファージ(Macrophage)、マウス榭状細胞(Dendritic cell ; DC)のいずれかである、請求項 3の免疫賦活剤。
[5] 請求項 1乃至 4のいずれか 1項に記載の免疫賦活剤を含有する組成物。
[6] 前記組成物が医薬組成物である、請求項 5に記載の組成物。
[7] 前記組成物が食品組成物である、請求項 5に記載の組成物。
[8] コーヒー由来のァラビノガラタタン(Coffee AG ; Cof—AG)を有効成分とする免疫 賦活剤。
[9] コーヒー生豆、コーヒー焙煎豆、またはコーヒー抽出残渣に、
水を添加し、加熱する工程と、
加熱抽出液を回収し減圧濃縮する工程と、
減圧濃縮した液体にエタノールを加えて沈殿させる工程と、
を有することを特徴とするコーヒー抽出物の製造方法。
[10] コーヒー生豆またはコーヒー抽出残渣に、
水を添加し、加熱する工程と、
加熱抽出液を回収し、減圧濃縮する工程と、
減圧濃縮した液体にエタノールを加えて沈殿させる工程と、
沈殿を水酸化ナトリウム溶液に溶解する工程と、
室温で:!〜 48時間、ついで、 50°C〜70°Cで:!〜 48時間攪拌する工程と、 pHを 7· 0〜8· 0に調製する工程と、
タンパク分解酵素により、タンパク質を分解する工程と、
水で透析する工程と、
を有することを特徴とするコーヒー抽出物の製造方法。
[11] ァラビノガラタタンの平均分子量が 10, 000-3, 000, 000である請求項 1乃至 8 のレ、ずれか 1項に記載の免疫賦活剤。
[12] ァラビノガラタタンのァラビノース/ガラクトースの比が 0. 02〜: 1. 0である請求項 1 乃至 8のレ、ずれか 1項に記載の免疫賦活剤。
[13] 乳酸菌を配合した、請求項 5乃至 7のいずれか 1項に記載の組成物。
[14] コーヒー抽出物を添カ卩することにより、マウス脾細胞または樹状細胞のインターロイ キン— 12産生量を無添加の場合に比べ増加させる方法。
[15] コーヒー抽出物を投与することにより、マウス血中インターロイキン一 12量を非投与 マウスに比べ増加させる方法。
[16] コーヒー抽出物を摂取させることによりマウス脾細胞のマイトジヱン PMA/Ionomy cinによる増殖促進活性を非摂取の場合に比べ高める方法。
