背景技術
ァミノ酸は細胞の生存に必須の分子であり書、 特に必須ァミノ酸とその関連物質 は哺乳類の生存に極めて重要な役割を果たしている。 メチォニン (methionine) はそのような必須アミノ酸のひとつであり、 これまで脂肪肝や肝硬変などでその 量が減少し、 外因性に補給すると細胞障害の抑制が認められることが報告されて きた。
Methionineは、 S'adenosyl methionine (SAM)に代謝され、 メチル基を DNA、 蛋白質などに転移させた後 S-adenosyl homocysteineとなり、 さらにホモシステ, イン (homocysteine) に変換される。 またこの homocysteineは、 システィン ( cysteine) の原料として transsulfuration pathwayで代謝され、 抗酸化物質であ' るダルタチオン (glutathione) の原料として使われる。 したがって、
methionineや homocysteineの補給は、 上記の肝疾患にとどまらず、 各種の酸化 ス トレス病態の改善薬としての利用可能性を考えることができる。
しかしながら、 homocysteineは強力な還元力を有し、 試験管内では酸素を還元 してスーパーオキサイド (superoxide) を生成することが知られており、 また臨 床的にも血中濃度が高くなると心血管ィベントが增加するいわゆる coronary risk factorのひとつとして認知されている。 このため、 methionineや homocysteineの 補給により酸化ス トレス病態を制御する試みはされていなかった。
発明の開示
本発明が解決しょうとする課題は、 生体内におけるアミノ酸.(methionine及び homocysteine) の細胞保護剤としての新規用途、 並びに生体内においてこれらの ァミノ酸濃度を増加誘導させる方法を提供することにある。
本発明者は、 上記課題を解決するべく鋭章検討を行った。 血液中や細胞内には、 methionineや homocysteineは本来豊富に存在しており、 homocysteineは皿液中 ではジスルフイ ド (disulfide)を形成してホモシスチン (homocystine) として存在 し、 homocystineは細胞内に取り込まれて再び homocysteineとして利用される。 このため、 本発明者は、 その強力な還元作用が細胞保護作用をもたらす可能性を 改めて検証すべきであるという着想に至った。 その結果、 本発明者は、
methionine及び homocysteineの投与が、 肝臓等の組織において酸化ストレス病 態を軽減し、 細胞保護作用を有することを見出した。
さちには、 生体内で heme oxygenaseを介したヘムの分解により生じるガス分 子である一酸化炭素 (CO) に着目し、 この CO処理により、 細胞保護性アミノ酸 の細胞内含量を生理的濃度の範囲で人為的かつ有効に増加させることができるこ とを見出し、 本発明を完成した。
すなわち、 本発明は以下の通りである。
(1) —酸化炭素、 protoheme IX、 protoporphyrin IX, ドーパミン及びドブタミ ンからなる群から選ばれる少なくとも 1つを含有することを特徴とする、 生体内 の細胞保護性ァミノ酸の増加誘導剤。
(2) 生体内に、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及 ぴドブタミンからなる群から選ばれる少なくとも 1つを投与することを特徴とす る、 細胞保護性ァミノ酸の増加誘導方法。
細胞保護性アミノ酸としては、 例えばメチォニン及ぴ Z又はホモシスティンが 挙げられる。
本発明において、 一酸化炭素の投与は、 10 ppm以上 250ppm以下の濃度で吸入 により行うことができ、 一酸化炭素含有錯体ゃ cytochrome P450で代謝されて CO
を体内で生成する arylhydrocarbonなどの化合物の形態で投与することもできる。 また、 一酸化炭素を、 赤血球、 修飾ヘモグロビン及びリボソーム内包へモグロビ ンからなる群から選ばれる少なくとも 1つに結合させて投与することが可能であ る。
protoheme IX又は protoporphyrin IXの濃度は、 例えば 10〜100 μ mol/Lである。 本発明の方法において、 ドーパミン及び/又はドブタミンを投与すると、 細胞 の cyclic AMPを増加させて heme oxygenase- 1を誘導し、 生体内の一酸化炭素を 増加させることができる。
(3) 一酸化炭素、 protoheme IX、 protoporphyrin IX, ドーパミン及びドブタミ ンからなる群から選ばれる少なくとも 1つを含有することを特徴とする、 細胞保 護用又は細胞障害治療用医薬組成物。
(4) メチォニン及び Z又はホ ΐシスティンを含有することを特徵とする、 細胞 保護用又は細胞障害治療用医薬組成物。
(5) 生体内に、 (3)又は (4)に記載の医薬組成物を投与することを特徴.とする細胞 障害の治療方法。
(6) 生体内の細胞保護性アミノ酸の增加誘導剤の製造における、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタミンからなる群から 選ばれる少なくとも 1つの使用。
(7) 細胞保護用又は細胞障害治療用医薬組成物の製造のためのメチォニン及ぴ 又はホモシスティンの使用。
(8) 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタ ミンからなる群から選ばれる少なくとも 1つを含むことを特徴とする、 生体内の 細胞保護性アミノ酸の増加誘導用キット。
(9) 一酸化炭素、 protoheme IX、 protoporphyrin IX, ドーパミン、 ドブタミン、 メチォニン及ぴホモシスティンからなる群から選ばれる少なくとも 1つを含むこ とを特徴とする、 細胞保護用又は細胞障害治療用キット。
本発明の細胞保護性ァミノ酸の増加誘導剤、 及び細胞保護性ァミノ酸の増加 誘導方法を用いることにより、 例えば methionine、 homocysteineなどの細胞保 護性アミノ酸の細胞内含有量を生理的濃度範囲で人為的かつ有効に増加させるこ とができる。 そして、 methionineや homocysteineなどのアミノ酸は細胞保護作 用を有するため、 これらのアミノ酸は、 細胞保護用又は細胞障害治療用医薬組成 物として有用である。 図面の簡単な説明
図 1は、 acetaminophen (AAP)による急性肝障害モデルにおける COの生成と HO阻害剤 ZnPPの効果を示す図である。 CTZ (chlotrimazole)は cytochrome P450 の阻害剤であり、 AAPの代謝を抑制することが示されている。

図 3は、 AAP肝障害における heme oxygenase阻害による細胞障害の増悪と homocysteine (Hcy)の投与による抑制効果を示す図である。 抑制効果は最小用量 である 60 nmol/25 g body weightで最も強く、 用量を増加させてもさらなる抑制 効果は認められなかった (*P<0.05 versus the values for AAP+ZnPP, 実験数は 各 6匹) 。
図 4は、 虚血再灌流障害 (I/R3H) における GLDH の増加と L-methionine による抑制を示す図である。 Methionineに細胞保護作用があることが分かる。 発明を実施するための最良の形態
以下、 本発明について詳しく説明するが、 本発明の範囲はこれらの説明に拘束 されることはなく、 以下の例示以外についても、 本発明の趣旨を損なわない範囲 で適宜変更し実施し得る。
なお、 本明細書において引用された全ての刊行物、 ^1えば先行技術文献、 及び
公開公報、 特許公報その他の特許文献は、 その全体が参照として本明細書に組み 入れられる。 また、 本明細書は、 本願優先権主張の基礎となる米国仮出願第 60/753, 387号の開示内容を包含する。 本発明は、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及び ドブタミンからなる群から選ばれる少なくとも 1つを用いて、 生体内において細 胞保護性アミノ酸を増加させることを特徴とするものである。 これらの物質を生 体に投与すると生体内の CO量が増加する。 生体内で増加した COは、 血液中や細 胞内の methionine, homocysteineなどの細胞保護性ァミノ酸の細胞内含有量を 増加させる。 このようにして増加した細胞保護性アミノ酸の細胞保護作用により、 各種の酸化ス トレス病態を軽減することができる。 したがって、 生体内で増加し た細胞保護性アミノ酸を細胞保護剤として使用することにより、 細胞障害又は細 胞障害に起因する各種疾患を治療することが可能となる。 1 . 細胞保護性ァミノ酸の増加誘導剤及び増加誘導方法
本発明の '「生体内の細胞保護性アミノ酸の増加誘導剤」 は、 一酸化炭素、 protoheme IX, protoporphyrin IX, ドーパミン及びドブタミンからなる群から 選ばれる少なくとも 1つを含有する。
生体内に投与され取り込まれた一酸化炭素 (CO)は、 ヘモグロ ビン (hemoglobin)に結合して末梢組織に運ばれる。 また、 生体内に投与され取り込ま れ 7こ protoheme IX又 ίま protoporpnyrin丄 Xf¾、 heme oxygenase- 1 (Ηυ-l)を介し た分解により、 一酸化炭素を発生する。 さらに、 ドーパミン (dopamine) 及び ドブタミン (dobutamine) は、 カテコールアミンの一種であるが、 これらの力 テコールアミンは、 生体に投与することにより細胞の cyclic AMPを増加させて HO-1を誘導する。 そして、 生体内で HO-1を介したヘムの分解により、.一酸化炭 素が発生する。
したがって、 本発明の増加誘導剤を生体に投与することにより、 一酸化炭素の 濃度が上昇して細胞保護性アミノ酸、 例えば、 メチォニン及び 又はホモシステ
ィンの細胞内含有量を生理的濃度の範囲で人為的かつ有効に増加誘導させること ができる。
また本発明では、 細胞保護性ァミノ酸の増加誘導剤を製造するために、 一酸化 炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタミンからなる 群から選ばれる少なくとも 1つを使用することができる。
本発明の増加誘導剤を生体内に投与するためには、 例えば以下の通り実施する ことができる。
(i) 1 O ppm以上 2 5 O ppm以下の濃度の一酸化炭素を含む雰囲気の吸入によ り行う。
(ii) 一酸化炭素含有錯体ゃ cytochrome P450で代謝されて COを体内で生成する arylhydrocarbonなどの化合物を生体内に投与する。
(iii)赤血球、 修飾へモグロビン及びリボソーム内包へモグロビンからなる群か ら選ばれる少なぐとも 1つに一酸化炭素を結合させて投与する。
(IVノ protoheme IX又は protoporphyrin IXを 与する。
(V) ドーパミン及び/又はドブタミンを投与する。 一酸化炭素の投与を、 一酸化炭素を含む雰囲気の吸入により行う場合、 その濃 度は一酸化炭素中毒を引き起こさない濃度、 例えば 2 5 O ppm以下、 好ましくは 1 0 O ppm以下と低濃度にすることが好ましい。
「一酸化炭素含有錯体」 としては、 CORM-l、 CORM-2 ( [Ru(CO)3Cl2]2)
(Ozawa N, Go da , Makino N, Yamaguchi T, Yoshimura Y, Suematsu M (2002) Leydig cell-derived heme oxygenase -1 regulates apoptosis of premeiotic germ cells in response to stress. J Clin Invest 109: 457-467.)) 、 CORM.3、 CORM-Al、 CORM-F3などの CO releasing moleculeを挙げることができる。 そ れぞれ中心金属は、 マンガン (CORM-1) 、 ルテニウム (CORM-2、 ·3) 、 ホウ 素 (CORM-A1) 及ぴ鉄 (CORM-F3) である。
錯体の形態では、 使用し得る一酸化炭素濃度 (又は錯体の量) は、 ΙμΜ— lmM、 好ましくは 10μΜ— ΙΟΟμΜである。
また、 一酸化炭素を、 赤血球、 修飾ヘモグロビン、 及びリボソーム内包へモグ ロビンからなる群から選ばれる少なくとも 1つに結合させて投与する場合、 生体 内八の投与は、 例えば、 一酸化炭素を結合させた赤血球等の 「修飾型一酸化炭素 」 を血管内に投与すればよい。 赤血球は、 自己血、 輸血用血液、 非ヒ ト動物の血 液を問わず、 あらゆる赤血球源からのものを使用することができる。 リボソーム 内包ヘモグロビンの作製方法は公知である (H.Sakai et al., J. Biochem., 131, 611-617 (2002); J.G.Riess, Chem.Rev., 101, 2797-2919 (2001)) 。
さらに、 protoheme IX又は protoporphyrin IXを生体内に投与する場合、 投与 ルートは、 例えば経口投与、 又は腹腔内若しくは筋肉内注射することにより行う ことができる。 投与量は特に限定されるものではないが、 例えば 1〜: L000 pmol/kg、 好ましくは 40〜100pmol/kgである。
2 . 医薬組成物
上記の通り、 本発明の増加誘導剤を生体に投与すると、 メチォニン、 ホモシス ティンなどのアミノ酸の細胞内含有量が増加する。 これらのアミノ酸は細胞保護 作用を有するため、 細胞保護用又は細胞障害治療用医薬組成物として有用である。 また、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタ ミンのいずれも、 細胞保護性アミノ酸の増加を誘導させることができるため、 こ れらの物質も、 細胞保護用又は細胞障害治療用医薬組成物として有用である。
したがって、 本発明の医薬組成物は、 メチォニン及びノ又はホモシスティンを 含有する。 あるいは、 本発明の医薬組成物は、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタミンからなる群から選ばれる少なくと も 1つを含有するものとすることもできる。 メチォニン、 ホモシスティンを生体 内に投与した場合はもちろん、 前述のように一酸化炭素、 protoheme IX、 protoporphyrin IX, ドーパミン又はドブタミンを投与した場合でも、 メチォニン、 ホモシスティンなどの細胞内含有量を増加させることができる。 メチォニン及ぴ ホモシスティンは、 例えば、 肝臓のス トレス病態を軽減するなど、 細胞保護作用 を有する。 このため、 これらのアミノ酸は、 細胞保護用又は細胞障害治療用医薬
組成物として有用であり、 生体内に、 この組成物を投与することにより、 細胞障 害を治療することができる。
本発明の医薬組成物を投与する対象は、 ヒ 卜に限定されるものではなく、 非ヒ ト哺乳動物を挙げることができる。 非ヒ ト哺乳動物としては、 例えばマウス、 ラ ット、 モルモッ ト、 ゥシ、 ネコ、 ゥサギ、 ゥマ、 ヒッジ、 サル、 ィヌなどが挙げ られる。
本発明において保護の対象となる細胞は特に限定されず、 細胞保護性アミノ酸 により保護することのできる全ての細胞を挙げることができる。 例えば、 肝細胞、 神経細胞、 血管内皮細胞、 血球細胞、 肥満細胞などが挙げることができる。
ここで、 「細胞障害」 には、 例えば、 虚血再灌流により生じたもの、 薬剤誘起 性のもの、 感染症により生じたものなどがある。 また、 細胞障害に起因する疾患 としては、 限定されるものではないが、 例えば虚血性臓器障害、 出血性ショック、 敗血症などが挙げられ、 これらの疾患を医薬組成物の使用の目的とすることがで きる。
本発明の医薬組成物の有効成分として使用される一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタミン、 並びにメチォニン及びホモシ スティンは、 前記 1項において記載した投与方法により投与することができる。 これらの有効成分は、 例えば、 一酸化炭素についてはガスボンベとして発注 ·購 入することにより入手するこ ができる。 - protoheme IX又は protoporphyrin IX、 一酸化炭素は、 例えば、 ガスボンベ として発注 ·購入することにより入手することができるが、 市販品 CORM (CO- releasing molecules) (シグマ社) を購入すること等もできる。 上記有効成分は、 そのまま使用しても良いし、 薬理学的に許容された担体と組 み合わせて製剤化したものを使用することができる。
薬理学的に許容される担体としては、 例えば通常医薬に使用される、 賦形剤、 結合剤、 崩壊剤、 滑沢剤、 潤滑剤、 乳化剤、 着色剤、 矯味矯臭剤、 界面活性剤、 溶解補助剤、 懸濁化剤、 等張剤、 安定化剤、 緩衝剤、 抗酸化剤などを挙げること
ができる。
製剤としては、 錠剤、 散剤、 顆粒剤、 カプセル剤、 シロップ剤などの経口剤、 坐剤、 軟膏剤、 点眼剤、 パップ剤などの外用剤、 又は注射剤を挙げることができ る。
上記注射剤は、 点滴、 筋注、 皮下注、 静注などの方法で使用することができる。 注射剤は、 上記の薬理学的に許容される担体を適宜組み合わせて、 リボソーム製 剤として製剤化してもよレ、。
生体内への一酸化炭素 (気体の一酸化炭素、 一酸化炭素含有錯体、 修飾型一酸 化炭素) 、 protoheme IX、 protoporphyrin IX、 ドーパミン、 ドブタミンの投与 量は、 年齢、 性別、 症状、 投与経路、 投与回数、 剤形によって異なる。
気体の一酸化炭素の場合は、 成人 (60 kg) 、 好ましくは 10〜250ppm、 より 好ましくは lOOppmの濃度で 30分〜 3時間、 好ましくは 1〜 2時間、 吸入投与 する。 吸入は、 一酸化炭素元 (例えば、 一酸化炭素ボンべ又は濃縮装置) に接続 したマスク、 鼻カテーテル、 鼻力ニューレを用いて行うことができる。 一酸化炭 素の飽和溶液 (生理食塩水など) を血管内に投与することもできる。
また、 一酸化炭素含有錯体、 又は赤血球、 修飾ヘモグロビン及びリボソーム内 包ヘモグロビン等に結合した一酸化炭素 (修飾型一酸化炭素) の場合は、 成人 1 日当たりの投与量は、 臨床医によって適宜選択される。 protoheme IX又は protoporphyrin IXの成人 1日当たりの投与量は、 1〜4 0 micromole/kg 、 好ま しくは 5〜20 micromole/kgである。 本発明では、 細胞保護用又は細胞障害治療用医薬組成物を製造するためにメチ ォニン及びノ又はホモシスティンを使用する。 3 . キット
本発明の細胞保護性アミノ酸の増加誘導用キットは、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及びドブタミンからなる群から選ばれる少 なくとも 1つを含むものである。 また、 本発明の細胞保護用又は細胞障害治療用
キットには、 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及び ドブタミン並びにからなる群から選ばれる少なくとも 1つを含むことを特徴とす る。
キットに含まれる一酸化炭素は、 例えば気体の一酸化炭素、 修飾型一酸化炭素、 又は製剤化した一酸化炭素である。 また、 protoheme IX及び protoporphyrin IX、 ドーパミン及びドブタミン、 メチォニン及びホモシスティンも適宜製剤化できる。 本発明のキットは、 一酸化炭素等の他に、 吸入器、 注射器、 希釈用緩衝液、 生 理食塩水等を含めることができる。 さらに、 本発明のキットには、 操作説明書な どを含める.こともできる。 以下に、 実施例を挙げて本発明をより具体的に説明するが、 本発明はこれらに 限定されるものではない。
[実施例]
急性肝障害モデルにおける methionine, homocvsteineの細胞保護作用の解析
<方法〉
急性臓器障害時に外因性に methionine, homocystein を投与した際の細胞保 護作用を、 マウスの薬剤惹起性肝臓障害モデルおよびラットの虚血再灌流モデル を用いて検証した。
マウス薬剤惹起性肝臓障害は acetaminophen (AAP)を 200 mg/kg腹腔内注射 を行って 2 時間後に犠牲死させた。 一方、 肝臓の虚血再灌流障害では尾状葉を 残して、 他の門脈血流を微小クランプで停止させ、 1 時間後にクランプを解除し て 6時間後に犠牲死させた。
いずれの群においても犠牲死に際して心臓採血を行い肝臓細胞障害の指標とし て Glutamate dehydrogenase (GLDH)を計測するとともに、 肝臓組織を短時間 で切り落として液体窒素内に瞬時に落とし、 既報の方法により液体クロマトダラ フィー · タンデム型質量分析法 ( LC-MS/MS ) によ り methionine 及び
homocysteine などの低分子代謝物の定量的分析を行った (Sugiura, Y., Kashiba, M" Hosnikawa, K" Sasaki, R., Saito, K" Kimura, ri" Maruyama; K. Goda, N., Suematsu, M. Cadmium exposure alters metabolomics of sulfur- containing amino acids in rat testes. Antioxid. Redox Signaling 7(5-6), 781- 787, 2005.; Tian, J., Bryk, R., Itoh, M., Suematsu, M., Nathan, C. Variant tricarboxylic acid cycle in Mycobacterium tuberculosis'- Identification of 0i - ketoglutarate decarboxylase. Proc. Natl. Acad. Sci. USA 102, 10670-10675, 2005.) 。
液体窒素で凍結した組織の一部は蛋白量の定量に用い、 代謝物量は蛋白質量で 標準化して比較検討した。 またそれぞれのモデルにおいて、 肝臓内 CO量をガス クロマトグラフィーを用いて既報の方法により定量した (McLaughlin BE, Lash GE, Smith GN, Marks GS, Nakatsu K, Graham CH, Brien JF. Heme oxygenase expression in selected regions of term human placenta. Exp Biol Med (Maywood). 2003 May; 228(5): 564-7.) 。 同様の実験を、 肝臓の門脈本幹 を 1 時間クランプして解除した後、 再灌流を 3 時間行って肝臓障害を惹起する モデルにおいても検討した。
<結果 > .
図 1に示すように、 AAP肝障害モデルでは、 組織内の CO が急速に増加する ことが明らかになった (図 1A) 。 またこの COの増加は、 AAP投与 2時間では heme oxygenase- 1 (ΗΟ·1) の誘導が増加していないこと、 6時間では増加する ことが示された (図 1A中の上側のパネル) 。 COの増加は、 HOの阻害剤であ る Zn protoporphyrin (ZnPP)により抑制されることから、 COは HO 由来の生 成であることが判明した (図 IB) 。 COが最も高くなる 2時間で肝臓及び血液 中の代謝物を測定したと ころ、 肝臓内の methionine 及び血液中の homocysteineの増加が確認された (表 1 ) 。

*pく 0.05 significantly decreased compared to control. †p<0.05 significantly increased compared, to control. 図 2に示すように、 Homocysteineの増加は COの阻害剤である ZnPPにより 完全に抑制された。 このことから、 CO には血液中の homocysteine を增加させ る作用があることが明らかになった。 同様に、 肝臓内の methionine の量も ZnPPにより低下した。
以上の結果から、 CO には homocysteine及び methionineを増加させる作用 があることが明らかになった。 次に、 これら 2つのアミノ酸 (homocysteine, methionine) に細胞保護効果 があるかどうかを検証しこ。
図 3に示すように、 AA モデルでは、 HO の阻害をかけることにより GLDH の増加による明らかな細胞障害の増悪が惹起された。 ここに外因性の
homocysteineを投与すると、 顕著に細胞障害が軽減した。
また、 図 4に示すように、 虚血再灌流障害モデル (I/R3H) では、 GLDH.が 有意に増加するが、 Methionineを 40 μπιοΐ/kgで腹腔内投与すると (I/R3H+L- met) 、 GLDHの増加が顕著に抑制されることが判明した。 く考察〉
COの増加は、 肝臓の細胞保護性アミノ酸 (methionine, homocysteine) の増 加を促すことが本発明により明らかになった。 上記の実験モデルでは、 Methionine 及び homocysteine 自身に細胞保護作用があることが明らかになつ たため、 他の肝障害モデルでの効果も期待できると考えられる。 また、 これらの 細胞保護作用は、 COの投与によっても再現できると考えられる。
CO は hemoglobin に結合して末梢組織に運ばれるものである。 そのため、 COの投与の方法に関しては、 (1) 低濃度 (10〜250ppm、 特に lOOppm程度) の吸入とすること、 (2) 赤血球、 修飾ヘモグロビン及び liposome -encapsulated hemoglobin等に結合させて血管内投与すること、 並びに (3) protoheme IX及 び protoporphyrin IX等を体内に投与すること (経口投与、 又は腹腔内若しくは 筋肉内注射 : 40〜: 100 moiyL) が好適であると考えられ、 さらには、 (4) dopamine及び dobutamine等の細胞の cyclic AMPを増加させて HO-1を誘導 する能力のあるすベての薬剤等により代替投与が可能であると考えられる。 産業上の利用可能性
本発明の細胞保護性アミノ酸の増加誘導剤、 細胞保護性アミノ酸の增加方法、 及び細胞保護用又は細胞障害治療用医薬組成物は、 各種の酸化ス トレス病態の改 善薬及び改善方法としての利用可能性がある。
Claims
1 . 一酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーノヽ。ミン及ぴドブタ ミンからなる群から選ばれる少なくとも 1つを含有することを特徴とする、 生体内の細胞保護性アミノ酸の増加誘導剤。
2 . 細胞保護性アミノ酸が、 メチォニン及び Z又はホモシスティンである、 請 求項 1に記載の増加誘導剤。
3 . 生体内に、 一酸ィ匕炭素、 protoheme IX、 protoporphyrin IX、 ドーノ ミン 及ぴドブタミンからなる群から選ばれる少なくとも 1つを投与することを特 徴とする、 細胞保護性アミノ酸の増加誘導方法。
4 . 一酸化炭素の投与は、 lOppm以上 250ppm以下の COを含む雰囲気の吸入に より行うものである、 請求項 3に記載の方法。
5 . —酸化炭素の投与は、 一酸化炭素含有錯体の形態で投与するものである、 請求項 3に記載の方法。
6 . —酸化炭素の投与は、 一酸化炭素を、 赤血球、 修飾ヘモグロビン及びリポ ソーム内包へモグロビンからなる群から選ばれる少なくとも 1つに結合させ て投与するものである、 請求項 3に記載の方法。
7 . protoheme IX又は protoporphyrin IXの濃度力 40〜: 100 ju mol/Lである、 請 求項 3に記載の方法。 -
8 . ドーパミン及びノ又はドプタミンの投与により、 細胞の cyclic AMPを増加 させて heme oxygenase- 1を誘導し、 生体内の一酸化炭素を増加させることを 特徴とする、 請求項 3に記載の方法。
9 . 細胞保護性アミノ酸が、 メチォニン及び 又はホモシスティンである、 請 求項 3 〜 8のいずれか 1項に記載の方法。
1 0 . —酸化炭素、 protoheme IX、 protoporphyrin IX、 ドーパミン及ぴドブ タミンからなる群から選ばれる少なくとも 1つを含有することを特徴とする、 細胞保護用又は細胞障害治療用医薬組成物。
