[0001] 本発明は、 tRNA合成方法、核酸、アミノアシル tRNA合成方法及び非天然型アミ ノ酸組み込み蛋白質の製造方法に関する。
本願は、 2005年 8月 2日に出願された特願 2005— 224638号に基づレヽて優先権 を主張し、その内容をここに援用する。
背景技術
[0002] 蛋白質中の所望の位置のアミノ酸残基を、通常の蛋白質合成に関わる 20種類以 外のアミノ酸 (非天然型アミノ酸)で置換した、非天然型アミノ酸組み込み蛋白質 (以 下、「ァロ蛋白質」という)は、蛋白質の機能 ·構造解析のための有効な手段となり得る 。リジン誘導体の中には、ァセチルリジン、メチルリジン等のように、翻訳後修飾により 合成されるアミノ酸がある。それらは、特にヒストンにおける遺伝子発現調節に関わる ことで有名であるが、その他多くの蛋白質の転写活性化調節、蛋白質 蛋白質相互 作用調節、ュビキチン化の抑制/促進に関わることが知られている。このようなリジン 誘導体が、真核生物に部位特異的に導入できれば、リジンのァセチル化、メチルイ匕 等について多くの知見が得られると期待される。
[0003] ピロリジル tRNA合成酵素(PylRS)は、メタン生成古細菌(Methanosarcina属)から 発見された新しいアミノアシル tRNA合成酵素(aaRS)である。その対応する tRNA ( ピロリジン(pyrrolysine) tRNA)は、サプレッサー tRNAであり、異常に小さい Dルー プを有する等特異な二次構造を有する。最近、 PylRSとピロリジン tRNAが、大腸菌 内で、内在性の aaRS及び tRNAとは相互作用をせず(直交性)、ピロリジンをアンバ 一 'コドン特異的に蛋白質に導入し得ることが見出された (非特許文献 1)。また、野 生型 PylRS力 大腸菌内で N ε _Boc_L—リジン等の非天然型アミノ酸をピロリジ ン tRNAに結合させ得ることを見出した(非特許文献 1)。

発明の開示
発明が解決しょうとする課題
[0004] しかしながら、 PylRSとピロリジン(pyrrolysine) tRNA力 真核生物、特に動物細胞 において直交性を有し、これらを遺伝暗号の拡張に用いることができるか否か明らか になっていなかった。
[0005] したがって、本発明は、アミノアシル tRNA合成酵素、アミノアシル tRN Aを用い、 真核細胞中で、非天然型アミノ酸組み込み蛋白質を製造する方法を提供することを 目的とする。
また、本発明は、力かるアミノアシル tRNAを真核細胞中で合成する方法を提供す ることを目的とする。
また、本発明は、 tRNAを真核細胞中で合成する方法を提供することを目的とする また、本発明は、力かる tRNAを真核細胞中で合成するための核酸を提供すること を目的とする。
課題を解決するための手段
[0006] 本発明者らは、上記目的を達成するため鋭意研究した結果、プロモーターに着目 した。一般に、真核細胞内での tRNAの発現は、 tRNAコーディング配列内の 2つの 内部プロモーターを必要とし、そのコンセンサス配列は、ボックス A、ボックス Bとして 知られている。例えば、バチルス ステアロサーモフィラス(Bacillus stearothermophilu s)のサプレツサーチ口シン tRNAは、原核生物由来であるが、そのサプレツサーチ口 シン tRNA配列内には、ボックス Aとボックス Bが存在しているため(M. Sprinzlら、 Nuc leic Acids Research 17, 1-172 (1989) )、なんら改変を加えなくても動物細胞内で発 現させることができる。このため、ボックス Aとボックス Bを有しない原核生物を用いる 場合、これにボックス Aとボックス Bを導入することが必要であると考えられる。
[0007] しかしながら、上記したように、メタン生成古細菌由来のピロリジン tRNAの Dループ は、数塩基が欠損した異常に小さいものであり、これにボックス Aとボックス Bを導入す ると、機能が失われてしまうため、ボックス Aとボックス Bを導入したピロリジン tRNAを 用いても、リジン誘導体を組み込んだァロ蛋白質を合成することはできなかった。そこ
で、本発明者らは、さらに検討した結果、真核生物の tRNAをプロモーターとしてピロ リジン tRNAの 5 '末端側に結合させれば、メタン生成古細菌由来のピロリジン tRNA を、動物細胞中で効果的に発現させることができることを見出し、本発明を完成した。
[0008] すなわち、本発明は、合成対象となる tRNAの核酸配列の 5 '末端に真核生物由来 tRNA核酸配列を結合させた配列を含む核酸を、真核細胞内にぉレ、て転写すること を特徴とする tRNA合成方法を提供するものである。
また、本発明は、力かる tRNAの合成方法に用いる核酸であって、 tRNAの核酸配 列の 5 '末端に、真核生物由来 tRNA核酸配列を結合させた配列からなる核酸を提 供するものである。
また、本発明は、 tRNAに対応するアミノアシル tRNA合成酵素の存在下、 tRNA の核酸配列の 5 '末端に真核生物由来 tRNA核酸配列を結合させた配列を含む核 酸を、真核細胞内において転写することを特徴とするアミノアシノレ tRNA合成方法を 提供するものである。
また、本発明は、アミノアシル tRNA合成酵素と、アミノアシル tRNA合成酵素の存 在下で非天然型アミノ酸と結合可能な tRNAの核酸配列の 5 '末端に、真核生物由 来 tRNA核酸配列を結合させた配列を含む核酸と、非天然型アミノ酸と、所望の位 置にナンセンス変異を受けた所望の蛋白質遺伝子と、を真核細胞中で発現させ、前 記蛋白質のナンセンス変異の位置に前記非天然型アミノ酸を取り込ませて非天然型 アミノ酸組み込み蛋白質を発現させることを特徴とする非天然型アミノ酸組み込み蛋 白質の製造方法を提供するものである。
発明の効果
[0009] 本発明の製造方法を用いれば、 tRNA,アミノアシル tRNAを、真核細胞中で効果 的に発現させることができ、ボックス A、ボックス Bを有さない tRNAを真核細胞中で 発現させることが可能である。
また、本発明の製造方法を用いれば、野生型のアミノアシル tRNA合成酵素を用 いて、特に真核生物に存在する N ε—ァセチルリジン、 Ν ε—トリメチルリジン、蛍光 基を有する Ν ε —2—メチルァミノ一ベンゾィルリジン等のリジン誘導体が導入された ァロ蛋白質を合成することができる。

本発明に用いられる非天然型アミノ酸としては、特にリジン誘導体であることが好ま しい。リジン誘導体は、非天然型のアミノ酸であり、 ε位の窒素原子に結合した水素 原子が他の原子又は原子団に置換されたものが好ましい。例えば、ピロリジン(pyrrol ysine)、 N ε —t—ブトキシカルボ二ルリジン(Ν ε —Boc—リジン)、 Ν ε—ァセチルリ ジン、 Ν ε—トリメチルリジン、 Ν ε —2—メチルァミノ一ベンゾィルリジン(Nma—リジ ン)力 S挙げられる。特に、真核生物に存在する修飾リジンであるメチルリジン、ァセチ ルリジンを部位特異的に蛋白質に導入することにより、リジンのァセチル化、メチル化 について多くの知見が得られる可能性がある。また、これらのリジン誘導体が導入さ れたァ口蛋白質は、蛋白質機能 ·構造解析の材料として有用であり、創薬のターゲッ トともなる可能性がある。
[0012] (アミノアシル tRNA合成酵素)
本発明で用いられるアミノアシル tRNA合成酵素は、非天然型アミノ酸を認識し、か つアミノアシル tRNAを特異的に認識して、該非天然型アミノ酸が結合したサプレツ サー tRNAを生成させることができる tRNA合成酵素である。
このうち、アミノ酸としてリジン誘導体を認識し、かつ tRNAとして併用するピロリジン
tRNAを特異的に認識して、該リジン誘導体が結合したサプレッサー tRNAを生成さ せることができるメタン生成古細菌由来の PylRSが好ましい。メタン生成古細菌として は、メタノサルシーナ 'マゼィ(M. mazei)が好ましい。
[0013] PylRSは、真核細胞、好ましくは動物細胞、特に好ましくは哺乳動物細胞中で発現 させる。 PylRSを哺乳動物細胞中で発現させるためには、例えばメタノサルシーナ · マゼィ由来の野生型遺伝子に、 N末端領域に FLAGタグ等が付加するようにした D NA配列を PCR法を用いて増幅し、これを市販の pcDNA3. 1 (インビトロジェン社製 ) , pAGE107 (Cytotechnology, 3, 133 (1990) )、 pAGE103 (J. Biochem. 101, 1307 (1987) )などの Nhel— BamHIサイトに組み込んで構築したプラスミドを、哺乳動物 細胞に導入すればよい。
細胞へのベクターの導入方法としては、例えば、電気穿孔法(Nucleic, Acids Res. 15, 1311-1326 (1987) )、リン酸カルシウム法(Mol. Cell Biol. 7, 2745-2752 (1987) )、 リポフエクシヨン法(Cell 7, 1025-1037 (1994) ;Lamb, Nature Genetics 5, 22-30 (1993 ) )などが挙げられる。
[0014] (tRNA)
上記アミノアシル tRNA合成酵素と組み合わせて使用される tRNAは、サプレッサ 一 tRNAとして機能するためのナンセンスコドンに相補的なアンチコドン及び立体構 造を保持しており、かつ真核細胞、好ましくは動物細胞、特に好ましくは哺乳動物細 胞中で発現する。アミノアシル tRNA合成酵素が PylRSである場合、対応するピロリ ジン tRNAは、サプレッサー tRNAとして機能するためのナンセンスコドンに相補的 なアンチコドン及び立体構造を保持しており、かつ真核細胞中で発現する、非真核 細胞由来の tRNAである。すなわち、通常の 20種類のアミノ酸に割り当てられたコド ンではないナンセンスコドンに割り当てられ、かつ、上記リジン誘導体に特異的な Pyl RSにのみ認識され、宿主の通常の aaRSには認識されなレ、(直交性)、という要件を 備え、かつ動物細胞中で発現するサプレッサー tRNAである。
ここで、ナンセンスコドンとしては、 UAG (アンバー)、 UAA (オーカ一)、 UGA (ォ パール)が挙げられる力 UAG (アンバー)コドンを用いることが好ましい。
[0015] 上記したように、真核細胞での tRNAの発現は、 tRNAコーディング配列内の 2つ
の内部プロモーターを必要とし、そのコンセンサス配列は、ボックス A、ボックス Bとし て知られている。
ピロリジン tRNAのクローバー型構造を図 1に示す。図 1において、左側のループ( Dループ)中、〇は塩基の欠損を示す。このように、ピロリジン tRNAは、 Dループ中 3 塩基が欠損しており、他の tRNAの Dループに比べて異常に小さい。動物細胞中で ピロリジン tRNAを発現させるために、ピロリジン tRNAにボックス A、ボックス B配列を 導入しても、ピロリジン tRNAの Dループが異常に小さいため、構造が大きく変化して しまい、このためサプレッサー活性を保持することができなかった。
[0016] (tRNA,アミノアシル tRNAの合成)
本発明のアミノアシル tRNAの合成方法は、野生型の tRNAの 5'末端に真核生物 由来の tRNAを結合させ、アミノアシノレ tRNA合成酵素の存在する真核細胞内、好 ましくは動物細胞内、特に好ましくは哺乳動物細胞内で転写させる。このとき、 tRNA の 3'末端に転写終結配列を結合させることが好ましい。より具体的には、まず、メタノ サルシーナ ·マゼィ由来の野生型ピロリジン tRNAの 5,末端に真核生物由来の tRN Aを結合させ、また 3 '末端に転写終結配列をそれぞれ結合させた配列を DNAブラ イマ一力 合成し、例えば pCR4Blunt— TOPO (インビトロジェン社製)等に組み込 んで構築したプラスミドを、動物細胞に導入して発現させた後、動物細胞内で転写し てプロセッシングすることにより得られる。このとき、野生型ピロリジン tRNAの 5 '末端 と真核生物由来の tRNAとは、リンカ一を介して結合させることが好ましい。リンカ一と しては、特に制限はなぐ例えば、 BglII、 Xbal、 Xhol等によって切断されるものが挙 げられる。 5 '末端に結合する tRNAは、真核生物由来である力 真核生物としては、 特に制限はなぐ例えば動物、植物、昆虫等が挙げられる。このうち、ヒト由来の tRN Aが好ましい。 tRNAが結合するアミノ酸も、通常の 20種類の天然型アミノ酸であれ ば特に制限はなレ、。このうち、ノ リンが好ましい。
[0017] 本発明の tRNAの合成方法は、合成対象となる tRNAの 5'末端に真核生物由来 の tRNAを結合させ、真核細胞内で転写させる。合成対象となる tRNAは、特に限定 されず、すべての tRNAを指す力 例えば、真核生物由来 tRNA (真核生物ミトコンド リア tRNAを含む)、原核生物由来 tRNA等が挙げられる。好ましくは真正細菌、古
細菌等の原核生物由来 tRNAであり、さらに好ましくはピロリジン(pyrrolysine) tRNA である。また 3 '末端に転写終結配列を結合させることが好ましい。真核細胞は、動物 細胞、特に哺乳動物細胞であることが好ましい。真核生物由来の tRNAは、例えば 動物、植物、昆虫等由来のものであり、このうち、ヒト由来の tRNAが好ましレ、。 tRNA が結合するアミノ酸としては、通常の 20種類の天然型アミノ酸であれば特に制限はな レヽ。このうち、ノ リンが好ましい。
[0018] (非天然型アミノ酸を組み込ませるための蛋白質)
本発明で非天然型アミノ酸を組み込ませる蛋白質の種類は、限定されるものではな ぐ発現可能ないかなる蛋白質でもよぐ異種の組換え蛋白質でもよい。例えば、蛋 白質の種類として、いわゆるシグナル伝達関連蛋白質、受容体、増殖因子、細胞周 期関連因子、転写因子、翻訳因子、輸送関連蛋白質、分泌蛋白質、細胞骨格関連 蛋白質、酵素、シャペロン又は癌、糖尿病若しくは遺伝病等を含む疾患関連蛋白質 などが挙げられる。
[0019] 本発明において、非天然型アミノ酸、特にリジン誘導体を組み込ませる位置にナン センスコドン(サプレッサー tRNAがアンバーサプレッサーのときはアンバーコドン)を 導入することが必要であり、これによりこのナンセンスコドン(アンバーコドン)部位に、 特異的に非天然型アミノ酸、特にリジン誘導体を組み込むことができる。
[0020] 蛋白質に部位特異的に変異を導入する方法としては、周知の方法を用いることが でさ、特に限定されないが、 Gene 152, 271-275 (1995)、 Methods Enzymol. 100, 468 —500 (1983)、 Nucleic Acids Res. 12, 9441—9456 (1984)、 Pro Natl. Acad. Sci. USA 82, 488-492 (1985)、「細胞工学別冊「新細胞工学実験プロトコール」、秀潤社、 241 — 248頁(1993)」に記載の方法、または「Quick Change Site-Directed Mutagenesis Kit] (ストラタジーン社製)を利用する方法などに準じて、適宜実施することができる。 本発明は動物細胞内で発現させることができるので、これまで、大腸菌や無細胞蛋 白質系では、発現しない、あるいは発現量が低レ、、または活性型となるための翻訳後 の修飾を受けることができないような蛋白質へ、非天然型アミノ酸を取りこませることが できる。このような蛋白質としては、当業者には種々のものが知られている力 例えば 、ヒト EGFR等のチロシンキナーゼ型レセプターの細胞外ドメイン(Cell, 110, 775-787
(2002) )、ヒト Groucho/TLEl蛋白質(Structure 10, 751- 761 (2002) )、ラット筋肉特異 的キナーゼ(Structure 10. 1187-1196 (2002) )などについて、ァロ蛋白質を合成する ことができる力 これらに限定されるものではない。
また、本発明の方法においては、動物細胞内でァロ蛋白質を発現させるので、糖 鎖と結合した糖蛋白質に非天然型アミノ酸、特にリジン誘導体を組みこませることもで きる。特に、無細胞蛋白質系における糖鎖付加のパターンが、本来のパターンと異な るようなタイプの糖蛋白質の場合には、本発明の動物細胞内での系は、 目的の(本 来の)パターンの糖鎖が付加されたァロ蛋白質を得るための有効な手段と考えられる
[0021] 非天然型アミノ酸、特にリジン誘導体を組み込ませるための蛋白質は、例えば所望 の蛋白質の所望のアミノ酸の位置に対応するコドンを、ナンセンスコドンに置換し、さ らに C末端に所望のタグを付加するように構築した配列を有する遺伝子を、例えば pc DNA4/TO等の BamHI— XhoIサイトに組み込んでプラスミドを構築し、これを動 物細胞に導入して発現させることができる。

本発明に用いられる、宿主の動物細胞としては、遺伝子組換え系が確立されてい る、哺乳類細胞が好ましい。有用な哺乳動物宿主細胞系の実例は、チャイニーズハ ムスター卵巣(CHO)と COS細胞を含む。より特有な例は、 SV40によって形質転換 したサル腎臓 CV1系(COS_7、 ATCC CRL 1651);ヒト胚腎臓系(293又は懸濁培 養での増殖用にサブクローンした 293細胞、 J. Gen Virol , 36: 59 (1977) );チヤィニ 一ズノヽムスター卵巣細胞/- DHFR (CHO、 Proc. Natl. Acad. Sci. USA, 77: 4216 (19 80) );マウスセルトーリ細胞(TM4、 Biol. R印 rod" 23: 243-251 (1980) );ヒト肺細胞 ( W138、 ATCC CCL 75);ヒト肝臓細胞(Hep G2, HB 8065);及びマウス乳癌(MMT 060562, ATCC CCL51)を含む。これらの宿主は、各々発現系が確立されており、適 切な宿主細胞の選択は、当業者の技術範囲内である。
これらは、 列えば、 Molecularし lomng第 J片 j¾、 Cold Spring Harbor Laboratory Press (2001)などに記載された方法に準じて行なうことができる。
[0023] 上記 3種類のプラスミドを、動物細胞に組み込んで発現させることにより、 目的とす
るリジン誘導体が組み込まれたァロ蛋白質を発現させることができる。
すなわち、(A)アミノアシル tRNA合成酵素、特に PylRSを動物細胞内で発現させ る発現ベクターと、 (B)上記アミノアシル tRNA合成酵素、特に PylRSの存在下で、 非天然型アミノ酸、特にリジン誘導体と結合可能なメタノサルシーナ ·マゼィ由来のピ 口リジン tRNAを、上記動物細胞内で発現させる発現ベクターと、 (C)所望の位置に ナンセンス変異を受けた所望の蛋白質を発現させる発現ベクターと、非天然型ァミノ 酸、特にリジン誘導体と、を有する動物細胞を、その動物細胞の増殖に適した培地( 例えば、 CHO細胞の場合、 Optト MEM I (Gibco BRL社)など)で、適当な条件でイン キュペートする。例えば、 CHO細胞の場合は、 37°C程度の温度で、 24時間程度、ィ ンキュペートする。
[0024] 次に実施例を示して本発明をさらに詳細に説明するが、本発明は以下の実施例に 限定されるものではない。
実施例 1
[0025] Grb2は、細胞内で、上皮成長因子受容体と相互作用する、細胞の癌化に関わるタ ンパク質である。本実施例では、ヒト Grb2の 111位にリジン誘導体を組み込む実験を 行った。
[0026] (PylRS発現プラスミドの構築)
メタノサルシーナ 'マゼィ由来の野生型 PylRS遺伝子に、 N末端領域に FLAGタグ が付加するようにした DNA配列(配列番号 1)を、 PCR法を用いて増幅した。これを、 pcDNA3. 1の Nhel— BamHIサイトに組み込んで、プラスミドを構築した。
(ピロリジン tRNA発現プラスミドの構築)
メタノサルシーナ 'マゼィ由来の野生型ピロリジン tRNAの 5'末端に、リンカ一(酉己 列番号 2)を介してヒトバリン tRNAを結合させ、また 3'末端に、転写終結配列をそれ ぞれ結合させた配列(配列番号 3)を DNAプライマーから合成した。これを、 pCR4Bl unt— TOPOに組み込んで、プラスミドを構築した。
(リジン誘導体を組み込む蛋白質を発現させるプラスミドの構築)
ヒトの grb2の 111位のロイシンコドン 、 Quick Change site-directed mutagenesis ki t (ストラタジーン社)を用いて、アンバーコドンに変換した(grb2 (Ami 11) )。次いで
、 C末端に FLAGタグ (DYKDDDDK)が付加するように構築した遺伝子(配列番号 4)を、 pcDNA4/TOの BamHI— XhoIサイトに組み込み、サブレッシヨン検出用の プラスミドとした。
[0027] (遺伝子の細胞への導入とサプレツシヨン反応)
上記 3種類のプラスミドを 2. OmL培養スケール 6-wellプレートで培養されたチヤィ ニーズハムスター卵巣細胞(CHO細胞、継代時の培養液は DMEM/F-12 (Gibco)、 10%FBS (ICN)、 1/100 penicillin-streptomycin (Gibco)を使用))の 1ゥエルあたりに 0. ずつ、様々な組み合わせで (結果参照)、 90%コンフルェント時に形質導入 を行った。开質導入は、 Lipofectamine 2000 (Invitrogen)を用いる方法をとり、そのマ ニュアルに沿って行った。形質導入時は培養液として Opti-MEM (Gibco)を使用した 形質導入を行った細胞培養液を、 ImM Boc リジン(Bachem)が存在する力、又 は存在しない DMEM/F_12 (Gibco)で置換し、 1 μ g/mLテトラサイクリンの添加によ り発現誘導し、 20時間ほど COインキュベーターにおいて 37°Cで培養を続けた。

細胞培養液を除き、緩衝液で洗浄後、細胞を溶解させ、蛋白質を回収した。 SDS ポリアクリルアミド電気泳動を行い、分子量で蛋白質を分離した後、メンブレンにェ レクトロブロット(100V、 1時間)を行った。抗体は ΑΝΉ-FLAG M2 (Sigma)を一次抗 体、 Mouse Ig Horseradish Peroxidase-Linked whole antioody rrom sheep Amersha m)を二次抗体として用いた。検出試薬は、 ECL Western Blotting Detection Reagent (Amersham)を用いた。測定は冷却 CCDカメラ LAS 1000 plus (富士フィルム)を用レヽ て行った。

図 2に、ウェスタンブロットによる Grb2 ( 1 1 l amb)のサブレッシヨンの検出結果を示 す。左端の 1レーンは、 Grb2全長のバンドの位置を示すための対照である。野生型 Grb2の C末端には FLAGタグが付加されており、 C末端まで合成された場合に Grb 2の矢印の部分にバンドが検出される。 2〜4レーンは、 PylRS、ピロリジン tRNA、 N ε —Boc リジンのいずれかがない場合である。これらの場合、 Grb2の全長が合成
されていなレヽ。一方、レーン 5は、 PylRS、ピロリジン tRNA及び N ε _ Boc _リジン のすべてが細胞内に導入された場合であり、 Grb2の全長が合成されている。このこ と力ら、 Grb2の 1 1 1位に導入したアンバーコドンに、 PylRS及びピロリジン tRNAに よって、 N ε _ Boc _リジンが導入されたことがわかる。

実施例 1において、ピロリジン tRNA発現プラスミドの構築方法を用いず、メタノサル シーナ.マゼィ由来の野生型ピロリジン tRNAにボックス A、ボックス Bを導入したもの を用いた以外は、実施例 1と同様にプラスミドの構築、遺伝子の導入を行い、サブレ ッシヨン反応の検出を行った。ボックス A、ボックス Bを導入したピロリジン tRNAのクロ 一バー型構造を図 4に示す。ボックス A、ボックス Bは、常法に従って導入した。

図 6は、試験管内で、精製 PylRS、精製ピロリジン tRNAの存在下で、各リジン誘導 体がピロリジン tRNAに結合するか否かを検討したものである。バンドが上にシフトし
たものがピロリジン tRNAに結合したことを示す。 Bocは N ε —Boc—リジン、 Acは Ν ε—ァセチルリジン、 Argはアルギニンを示す。図 6では、 N ε —Boc—リジンは 8m Mであり、このときピロリジン tRNAに結合している力 N ε —Boc—リジンが ImM以 下でも、 PylRSが N ε —Boc—リジンをピロリジン tRNAに結合させることができること が確認されている。
産業上の利用可能性
本発明は、リジン誘導体を導入してァロ蛋白質を合成することに有効に利用するこ とがでさる。
