Abstract
Die vorliegende Erfindung betrifft ein Verfahren zur Extraktion von Pflanzen, insbesondere Wurzeln, der Pflanzengattung Pelargonium, dadurch gekennzeichnet, daß die Pflanzen mit einem ethanolfreien Lösungsmittel oder einer ethanolfreien Lösungsmittelmischung extrahiert werden, das bzw. die ausgewählt wird bzw. werden aus Wasser und zumindest einem einwertigen Alkohol mit mindestens drei Kohlenstoffatomen, Wasser und zumindest einem mehrwertigen Alkohol, Wasser und zumindest einer anorganischen, organischen, ein- oder mehrwertigen Säure oder einem Derivat derselben, zumindest einem pflanzlichen Öl und Kohlendioxid.

Verfahren zur Extraktion von Pflanzen der Pflanzengattung Velargonium, danach hergestellter
Extrakt und Verwendung desselben
Die vorliegende Erfindung betrifft ein Verfahren zur Extraktion von Pflanzen, bevorzugt Wurzeln, der Pflanzengattung Pelargonium, sowie einen insbesondere nach dem Verfahren hergestellten Extrakt sowie dessen Verwendung.
Pelargonium reniforme/sidoides ist eine Pflanze, die im südlichen Afrika in der Volksmedizin tradtionell bei gastrointestinalen Beschwerden und Atemwegserkrankungen gebraucht wird. Die Pflanze wurde im Jahre 1897 nach England gebracht, und in den zwanziger Jahren wurde die Pflanze und Extrakte durch Sechehaye in die Schweiz gebracht. Extrakte wurden vor allem in homöopathischen Zubereitungen in der Tuberkulosetherapie eingesetzt (Kolodziej, H., Kayser, O., Z. Phytotherap. 19, 141-151, 1998).
In wissenschaftlichen Untersuchungen konnte gezeigt werden, daß ethanolische Extrakte eine antimikiObielle Wirkung gegen einige grampositive und gramnegative Bakterien haben (Kayser, O., Kolodziej, H., Planta Med. 63, 508-510, 1997) und über immunmodulatorische Wirkungen verfügen (Kayser, O., Kolodziej, H., Kiderlen, A.F., Radtke, O., Phytomedicine 2003; 10 Suppl. 4:18-24.
Hauptinhaltsstoffe von Pelargonium sidoides-Wurzoln sind Proanthocyanidine, oligomere hy- drolysierbare Gerbstoffe, Cumarine, Flavonoide und Kaffeesäurederivate (Kayser, O., Dissertation, Freie Universität Berlin, 1997). Charakteristisch ist das Vorkommen der zum Teil hocho- xygenierten Cumarine, die für die biologische Wirkung zum Teil mit verantwortlich gemacht werden (Kayser, O., Kolodziej, H., Phytochemistry, 39, 1181-1185, 1995).
Extrakte des Wurzelmaterials von Pelargonium sidoides und auch Pelargonium reniforme sowie anderer Pflanzen der Gattung Pelargonium wurden in der Vergangenheit durch Mazeration und/oder Perkolation unter Verwendung von reinem Wasser, Gemischen aus Wasser und einem organischen Lösungsmittel wie Aceton (Kayser, O., Kolodziej, H., Phytochemistry, 39, 1181- 1185, 1995), Methanol (Bladt, S., Wagner, H., Dtsch. Apoth.-Ztg., 128, 292-296, 1988) oder Ethanol (WO 03/028746 Al) hergestellt. Zwischenzeitlich sind aus der Pflanzengattung Pelargonium auch Pelargonium lobatum und Pelargonium triste bekannt. In der therapeutischen An-
wendung und im Handel befindet sich ein mit Glycerol stabilisierter ethanolischer Auszug aus Pelargonium reniforme/sidoides (Rote Liste 2006). Gegenüber Extraktionen mit Wasser, Methanol oder Aceton zeigt sich bei der ethanolischen Extraktion die verbesserte Gewinnung von Extraktivstoffen und die Extraktion bei Raumtemperatur. Allerdings wird der Gebrauch ethanolischer Extrakte aus P. reniforme/sidoides in Fachkreisen als problematisch angesehen, da insbesondere Kinder als bevorzugte Patientengruppe arzneilich genutzte Extrakte einnehmen. Diese Problematik gilt auch für Patienten, die Ethanol aus gesundheitlichen Gründen nicht einnehmen sollen oder dürfen, zum Beispiel alkoholkranke Personen, Epileptiker oder Parkinson-Patienten.
Der Wunsch nach ethanolfreien pflanzlichen Liquida auf anderen pharmazeutischen Gebieten hat in der Vergangenheit zur Entwicklung von Zubereitungsformen ohne Ethanol geführt, bei denen im Gegensatz zu Frischpflanzensäften, Teezubereitungen und wäßrigen Tonika nunmehr auch lipophile Pflanzeninhaltsstoffe enthalten sind. Als Lösungs- und Extraktionsmedium dienten beispielsweise Polyethylenglykol, Glycerol, Sorbitol oder Propylenglykol, einzeln oder in Kombination mit Wasser. In einer Dissertation von Vogel, K., Disseratation, Universität Tübingen, 1992, konnte gezeigt werden, daß die lipophilen Inhaltsstoffe von Kamille, nämlich Chama- zulen und Bisabolol, nicht nur gut löslich sind in Propylenglykol, sondern auch chemisch und physikalisch stabilisiert werden können. Kamille gehört einer anderen Pflanzengattung als Pe- largonium an. Bei der Kamillenextraktion zeigte sich jedoch, daß mit dem mehrwertigen Alkohol Propylenglycol im überwiegenden Maße lediglich lipophile Bestandteile extrahiert wurden, hydrophile Bestandteile jedoch kaum erhalten wurden (Vogel, K., Dissertation, Universität Tübingen, 1992). Ethanol- Wasser-Gemische besitzen gegenüber ethanolfreien Extrakten gewisse technologische und mikrobiozide Vorteile, insbesondere kann eine höhere Extraktausbeute erwartet werden. Gegenwärtig wird ein Propylenglykol-Glycerol- Wasser-Gemisch genutzt, um in einigen Fertigarzneimitteln den ethanolischen Spissum oder acetonischen Extrakt zu lösen (bspw. Tebonin forte Lösung). Sorbitol-Wasser-Gemische werden eingesetzt, um das in der Pädiatrie gebrauchte Arzneimittel Prospan anzubieten (Rote Liste 2006).
Der vorliegenden Erfindung liegt die Aufgabe zugrunde, das gattungsgemäße Verfahren derart weiter zu entwickeln, daß die Nachteile des Stands der Technik überwunden werden. Insbesondere soll ein Verfahren bereit gestellt werden, das eine ethanolfreie Extraktion von Pflanzen, insbesondere Wurzeln, der Pflanzengattung Pelargonium ermöglicht, um eine Verabreichung an Patienten zu ermöglichen, die Ethanol aus gesundheitlichen Gründen nicht einnehmen sollen
oder dürfen (beispielsweise Kinder, alkoholkranke Personen, Epileptiker, Parkinson-Patienten). Das erfindungsgemäße Verfahren soll ferner zu einem Extrakt führen, der dem aus einer Wasser- Ethanol-Extraktion gewonnenen Extrakt im wesentlichen äquivalent ist. Die eingesetzten Extraktionsmittel sollen toxikologisch unbedenklich sein. Insbesondere soll das Verfahren einen Extrakt bereitstellen, der qualitativ und quantitativ den wäßrig-ethanolischen Extrakten gleicht.
Desweiteren ist es eine Aufgabe der vorliegenden Erfindung, einen insbesondere nach dem Verfahren hergestellten Extrakt bereitzustellen und dessen Verwendung zu ermöglichen.
Die erste Aufgabe wird dadurch gelöst, daß die Pflanzen mit einem ethanolfreien Lösungsmittel oder einer ethanolfreien Lösungsmittelmischung extrahiert werden, das bzw. die ausgewählt wird bzw. werden aus Wasser und zumindest einem einwertigen Alkohol mit mindestens drei Kohlenstoffatomen, Wasser und zumindest einem mehrwertigen Alkohol, Wasser und zumindest einer anorganischen, organischen, ein- oder mehrwertigen Säure oder einem Derivat derselben, zumindest einem pflanzlichen Öl und Kohlendioxid. Als mehrwertige Alkohole sollen bevorzugt auch Zucker und Zuckeralkohole verstanden werden.
Bevorzugt ist dabei, daß die Extraktion eine Perkolation und/oder eine einstufige oder mehrstufige, vorzugsweise zweistufige Mazeration umfaßt.
Besonders bevorzugt ist, daß der mehrwertige Alkohol ausgewählt wird aus der Gruppe bestehend aus Polyethylenglycol, Glycerol, Propylenglycol, Sorbitol, Xylitol oder Mischungen derselben.
Dabei wird vorgeschlagen, daß das Polyethylenglycol ein Molekulargewicht von etwa 40 bis etwa 40000, bevorzugt von etwa 200 bis etwa 2000, besonders bevorzugt von etwa 200 bis etwa 800 aufweist.
In einer Ausführungsform werden die Pflanzen zerkleinert, gemahlen und/oder getrocknet und/oder frisch eingesetzt.
Besonders bevorzugt ist, daß das Gewichtsverhältnis Wasseπmehrwertiger Alkohol in einem Bereich von 95:5 bis etwa 5:95 liegt.

Auch wird vorgeschlagen, daß das Gewichtsverhältnis Wasser:organische ein- oder mehrwertige Säure in einem Bereich von 99,9:0,1 bis etwa 80:20 liegt.
Alternativ wird vorgeschlagen, daß zumindest ein pflanzliches Öl oder Kohlendioxid als Extraktionsmittel eingesetzt wird.
Ebenfalls wird alternativ vorgeschlagen, daß das Gewichtsverhältnis Wasseπanorganische Säure in einem Bereich von 99,9:0,1 bis etwa 80:20 liegt. Bevorzugt ist die anorganische Säure Phosphorsäure oder ihre Alkali- oder Erdalkalisalze.
Bevorzugt ist, daß das Gewichtsverhältnis von Wasseπmehrwertigem Alkohol bei einer Mazeration von etwa 80:20 bis etwa 60:40, bevorzugt etwa 70:30 reicht, und daß das Gewichtsverhältnis von Wasser:mehrwertigem Alkohol bei einer Perkolation von etwa 90:10 bis etwa 70:30, bevorzugt etwa 80:20 reicht.
Auch kann vorgesehen sein, daß vor der Perkolation ein Anmaischen der Pflanzen erfolgt.
Die zweite Aufgabe wird gelöst durch einen Extrakt der Pflanzengattung Pelargonium, gelöst in einem ethanolfreiem Lösungsmittel oder einer ethanolfreien Lösungsmittelmischung, ausgewählt aus der Gruppe bestehend aus Wasser und zumindest einem einwertigen Alkohol mit mindestens drei Kohlenstoffatomen, Wasser und zumindest einem mehrwertigen Alkohol, Wasser und zumindest einer anorganischen, organischen, ein- oder mehrwertigen Säure oder einem Derivat derselben, zumindest einem pflanzlichen Öl und Kohlendioxid.
Am bevorzugtesten ist, daß der Extrakt von Wurzeln der Pflanze Pelargonium sidoides, Pelargonium reniforme, Pelargonium lobatum und/oder Pelargonium triste ist.
Auch wird in einer Ausführungsform vorgeschlagen, daß der Extrakt zusätzlich ein Eindik- kungsmittel enthält und in fester, erstarrter Form vorliegt.
Dabei kann der Extrakt in Form eines Lutschbonbons, einer Lutschtablette, einer Bukal- oder Lingualarzneiform oder eines Lutschers vorliegen.
Schließlich wird die Verwendung des erfindungsgemäßen Extrakts zur Behandlung von akuten und/oder chronischen Entzündungserkrankungen und/oder Infektionen bei Säugetieren vorgeschlagen.
Bevorzugt ist die Verwendung beim Menschen, Haus- und Nutztieren, bevorzugt Pferd, Hund, Katze, Schwein oder Huhn.
Auch wird vorgeschlagen, daß der Wurzelextrakt oral verabreicht wird.
Der Extrakt kann auch bevorzugt zur Behandlung oder Prophylaxe von HIV-Infektionen oder HlV-assoziierten Infektionen eingesetzt werden. Entsprechende HIV-assoziierte Infektionen können Bakterien-, andere Virus-, Pilz- und/oder Parasiteninfektionen sein. Konkretere Beispiele werden in der DE 10 2004 032 439 Al gegeben.
Überraschenderweise wurde festgestellt, daß durch das erfindungsgemäße Verfahren ein Extrakt, vorzugsweise Wurzelextrakt, der Pflanzengattung Pelargonium in einer gleichen Ausbeute und Qualität wie für eine Extraktion auf Basis Wasser-Ethanol erhalten werden kann. Gleichzeitig wurde gefunden, daß die bekannten antimikrobiellen und immunstimulierenden Wirkungen bei Einsatz des erfindungsgemäßen Verfahrens erhalten wurden. Mit dem erfindungsgemäßen Verfahren können hohe Anteile an lipophilen und hydrophilen Bestandteilen extrahiert werden. Da das erfmdungsgemäße Verfahren ethanolfrei betrieben wird, kann der gewonnene Extrakt unbedenklich für die Behandlung von Kindern, alkoholkranken Personen, Epileptikern und Parkinson-Patienten eingesetzt werden.
Bevorzugt wird im erfindungsgemäßen Verfahren die Extraktion in Form einer Perkolation und/oder einer zweistufigen Mazeration durchgeführt.
Die Perkolation wird bevorzugt mit unterschiedlichen Gemischen aus Wasser und mehrwertigem Alkohol durchgeführt. Ein bevorzugter mehrwertiger Alkohol ist Sorbitol, beispielsweise in einer Mischung aus Wasser-Glycerol-Sorbitol 70:15:15 (Gew.-%), oder Polyethylenglykol, beispielsweise in einer Mischung aus Wasser-Polyethylenglykol-Glycerol 70:20:10 (Gew.-%). Die Perkolation kann aber auch durch Anmaischen erfolgen, wobei das Anmaischen und anschließende Perkolieren mit gleich oder unterschiedlich konzentrierten Wasser-Alkohol-Gemischen
erfolgen kann. Es zeigte sich als Vorteil, daß ein Anmaischen mit einem Wasser-Alkohol-Anteil von 70:30 (Gew.-%) und die Perkolation mit einem Wasser-Alkohol-Anteil von 80:20 (Gew.-%) zu besten Ausbeuten führt. Bei einem Verhältnis von Wurzelmaische zu Extraktionsmedium von 20:80 (Gew.-%) ergibt sich eine Endkonzentration des bzw. der mehrwertige Alkohol(e) bei circa 22 %. Die erfindungsgemäßen Extrakte enthalten einen Cumarinanteil, bezogen auf Scopole- tin, der im gleichen Größenbereich wie derjenige der ethanolischen Extrakte liegt.
Bei Extraktionsversuchen mit der gemahlenen und getrockneten Wurzel von Pelargonium sidoi- des mit Gemischen aus Wasser und mehrwertigem Alkohol wurde überraschenderweise festgestellt, daß in Abhängigkeit von der Zusammensetzung des Extraktionsmediums lipophile Stoffe wie Cumarine auf der einen Seite, aber auch wasserlösliche Phenole auf der anderen Seite sich im gleichen Extraktionsschritt extrahieren lassen. Vor dem Hintergrund des bestehenden Kenntnisstandes zu Pelargonium sidoides und Pelargonium reniforme ist dieses nicht zu erwarten gewesen. Sowohl bei der Perkolation als auch bei der zweistufigen Mazeration, die unten in den Beispielen näher beschrieben werden, konnten Extraktivmengen gewonnen werden, die qualitativ und quantitativ denen ethanolischer Extrakte gleichen.
Eine besonders bevorzugte Ausführungsform des erfindungsgemäß erhaltenen Wurzelextrakts liegt darin, dem Extrakt ein Eindickungsmittel, beispielsweise Glukosesirup, in einer solchen Menge zuzugeben, daß sich der Extrakt verfestigt bzw. erstarrt. Dann ist es möglich, den erstarrten Extrakt beispielsweise in Form eines Lutschbonbons, Lutschtablette, Bukal- oder Lingual- arzneiform oder eines Lutschers zu verabreichen, was insbesondere die Behandlung von Kindern stark erleichtern kann. Daneben kann der erfindungsgemäße Extrakt auch in einer üblichen flüssigen Zubereitung, wie in Form von Tropfen, Saft, Sirup, usw. formuliert und verabreicht werden.
Bei Prüfung der erfindungsgemäßen Extrakte auf ihre antimikrobielle und immunstimulierende Wirkung konnte eine deutliche Wirkung gegen grampositive Keime wie Staphylococcus aureus, Streptococcus pneumoniae und gramnegative Keime wie E. coli, Proteus mirabilis und Hämophilus influenzae gezeigt werden. Bei Prüfung auf Immunstimulierung zeigte sich eine Induktion von IFN-Gamma und TNF-alpha in Leishmania donovani infizierten murinen Makrophagen.
AIs Extraktionsmittel können bestimmte ein- oder mehrwertige Alkohole und Derivate, wie aliphatische und aromatische ein- oder mehrwertige Alkohole, anorganische oder organische, ein- oder mehrwertige Säuren und Derivate, pflanzliches Öl und Kohlendioxid eingesetzt werden. Bevorzugte Mischungsverhältnisse sind oben angegeben.
Beispiele konkreter besonders bevorzugter Extraktionsmittel sind:
Mehrwertige Alkohole, einschließlich Zucker und Zuckeralkohole, Polyethylenglycol (PEG) 100-4000, Propylenglycol, Polypropylenglycol 100-4000, Butandiol in möglichen Substituti¬
onsformen und seine Derivate, Butylalkohol, 1,3-Butylenglycol, Carboxymethylcellulose und ihre Derivate, beispielsweise Carboxymethylcellulose, Carboxyethylcellulose, Dextran, Dextrin, Dextrose, Ethylcellulose, Ethylenglycolderivate, beispielsweise Ethylenglycoldistearat, Ethylen- glycolmonobutylether, Ethylenglycolmonoethylether, Glucose, Glycerin und seine veretherten und veresterten Derivate, insbesondere Glycerin verestert mit langkettigen Fettsäuren oder ve- rethert mit langkettigen Fettalkoholen, Lactose, Mono- und Diglyceride mit ihren Estern und Alkali- und Erdalkalisalzen, Octanol, Polysorbat 20-80, Polyvinylacetat, Polyvinylalkohol, Pro- pylenglycolalginat, Sorbitan und seine ein- und mehrfach veresterten Derivate, Sorbitol, Xylose, Xylit, organische ein- und mehrwertige Säuren und ihre Alkali- und Erdalkalisalze, beispielsweise Ascorbinsäure, Weinsäure, Äpfelsäure, Heteropolysaccharide, Monosaccharide, Disaccharide, Trisaccharide, Tetrasaccharide, Kohlendioxid, pflanzliche Öle, Lecithin und seine Derivate, Phosphorsäure und ihre Alkali- und Erdalkalisalze und Polyoxyethylen, vorzugsweise verestert mit gesättigten oder ungesättigten Fettsäuren, zum Beispiel Polysorbat 20-80.
Bevorzugt sind ebenfalls aliphatische Phenylalkohole, beispielsweise Isobutyl-, Propyl- oder Ethylphenylalkohol, Methylbenzylacetat, -butyrat und weitere Säurederivate, flüssige, gesättigte und ungesättigte Fettsäuren und Fettalkohole mit Kettenlängen von C6 bis C16, Benzylalkohol und seine Derivate, Isoamylalkohol und seine Derivate, Isobutylalkohol und seine Derivate.
Die Erfindung wird anhand folgender, nicht einschränkender Beispiele erläutert. Um vergleichbare analytische und pharmakologische Daten zu erhalten, wurde der Scopoletingehalt aller Extrakte bestimmt.
Beispiel 1
100 g getrocknete und gemahlene Wurzel von Pelargonium sidoides wurden 24 h mit 200 g einer Wasser-Sorbitol-Mischung (70g Sorbitol in 100 ml Wasser) vorgequellt und anschließend mit 800 ml des gleichen Wasser-Sorbitol-Gemisches über 8 Stunden perkoliert. Der Rohextrakt wurde über Seitz Supra 1500 filtriert und abgefüllt.

Beispiel 2
100 g getrocknete und gemahlene Wurzel von Pelargonium sidoides wurden 24 h mit 200 g einer Wasser-PEG400-Mischung (Wasser:PEG400 = 10:90 (Gew.-%)) vorgequellt und anschließend mit 800 ml des gleichen Wasser-PEG400-Gemisches über 8 Stunden perkoliert. Der Rohextrakt wurde über Seitz Supra 1500 filtriert und abgefüllt.

Beispiel 3
Je 20 g getrocknete und gemahlene Wurzeldroge von P, sidoides wurde in unterschiedlichen Ansatzmedien (A1-A15) über 16 h vorgequellt (siehe Tabelle 3) und danach mit je 160 g zweier unterschiedlicher Extraktionsmedien (El, E2) perkoliert. Das Extraktionsmedium E 1 bestand aus Wasser-Glycerol-Sorbitol 80:10:10 (Gew.-%). und Extraktionsmedium E2 aus Wasser- Glycerol-PEG400 85:12,5:2,5 (Gew.-%). Die Perkolation erfolgte über 8 h und der Rohextrakt wurde über Glasfritten Größe G4 filtriert. Neben den ethanolfreien Extrakten wurde zu Vergleichszwecken ein ethanolhaltiger Extrakt (Al 6) erstellt, der nach den Vorgaben in Beispiel 1 der WO 03/028746 Al erstellt wurde.

Im Vergleich zur Perkolation wurde eine modifizierte Mazeration entsprechend der DAB 10 Vorschrift (Devrtsches Arzneibuch) durchgeführt. 20 g der gemahlenen und getrockneten Wurzel von P. sidoides wurden mit 140 ml einer Wasser-Sorbitol-Mischung (70g Sorbitol in 100 ml Aqua dest.) suspendiert und über 5 Tage mit gelegentlichem Umschütteln stehen gelassen. Anschließend wurde filtriert und der Rohextrakt aufgefangen. Das feuchte Drogenmaterial wurde nun ein zweites Mal in der gleichen Art und Weise mazeriert. Die Extraktlösung wurde ebenfalls aufgefangen und mit dem ersten Rohextrakt vereinigt.

Anschließend wurde filtriert und der Rohextrakt aufgefangen. Das feuchte Drogenmaterial wurde nun ein zweites Mal in der gleichen Art und Weise mazeriert. Die Extraktlösung wurde ebenfalls aufgefangen und mit dem ersten Rohextrakt vereinigt.

Analytische und pharmakologische Methoden
A. Bestimmung der Gesamtphenole
Die Bestimmung des Gesamtphenolanteils erfolgt nach der im Deutschen Arzneibuch angegebenen Methode für Gerbstoffe (DAB 2002). Die Bestimmung erfolgt photometrisch nach Umsetzung mit Molybdat-Wolframat-Reagenz. Dazu wird der Rohextrakt direkt genommen, mit Natri- umcarbonat-Lösung alkalisiert und anschließend mit Molybdat-Wolframat-Reagenz versetzt. Nach Zentrifugation wird die Extinktion der überstehenden Lösung bei 720 nm gegen Wasser gemessen. Die Bestimmung gegen Extraktionsmedium El oder E2 zeigte keinen Unterschied im Vergleich zu Wasser. Die Berechnung erfolgt auf Basis des Epicatechins.

B. Bestimmung der Gesamtcumarine
Die Bestimmung der Gesamtcumarine erfolgt mit Hilfe der HPLC über eine RP- 18 Säule. Als mobile Phase wird ein Acetonitril-Wasser-Phosphorsäure-Gradient (10:990:4 zu 205:795:4) verwendet. Detektiert wird bei 330 nm. Die Berechnung der einzelnen Cumarin-Peaks erfolgt als Scopoletin.
C. Bestimmung der antimikrobiellen Wirkung
Je 50 μl der erfindungsgemässen Extrakte werden in 150 ml Muellr-Hinton-Medium zu 200 μl verdünnt, und der antimikrobielle Effekt nach DIN 58 940, 1995, und der Methodik nach Van den Berghe D. A., Vlietnick, A. J., In: Methods in Plant Biochemistry, Assays for Bioactivity, VoI 6, Dey, P.M., Harborne, J. B. (Hrsg.), Academic Press, London, San Diego, New York, Boston, Sydney, Tokyo, Toronto, 47-99 (1991) bestimmt. Die Bestimmung erfolgte durch Ausmessen des Hemmhofes auf Agarplatten, in die vor Beimpfen Vertiefungen gestanzt wurden. Die Referenzstämme wurden von der deutschen Stammsammlung für Mikrooroganismen und Zellkulturen (DSMZ) in Braunschweig bezogen: Escherichia coli ATCC 25922, Hämophilus influenzae ATCC 33379. Alle weiteren Stämme wurden als in-house Stamm aus Patientenmaterial am Universitätsmedizinischen Zentrum UMCG, Groningen, NL, gewonnen.
D. Bestimmung der immunmodulierenden Wirkung
Prüfung auf TNF-Synthese und Freisetzung aus murinen L-929 (TNF) und humanen WEHI-Fibroblasten
Zellen der Maus-Fibroblastenzellinie L-929 (TNF) #5707 und WEHI 164/E clone 13 #5676 wurden in R5-Medium vorkultiviert bei 370C, 6% CC^. Das Kulturmedium in der Nährflasche wurde verworfen, und die adhärenten Zellen in der Flasche wurden mit 10 mL HBSS (Henk ba- lanced salt Solution) (ohne FCS (Fetal calf serum)) gespült. Die HBSS-Lösung wurde ebenfalls verworfen, in die Flasche 5 mL Trypsin/EDTA-Lösung gegeben und 15 Minuten bei 37°C, 6% CO2 inkubiert. Die enzymatische Ablösung der Zellen wurde durch Zugabe von 5 mL R5-
Medium gestoppt und die Zellsuspension in Falconröhrchen gesammelt. Zur Zellgewinnung wurde die Suspension zwei Mal bei 1100 U/min 12 Minuten bei Raumtemperatur zentrifugiert

und drei Mal mit 10 mL HBSS-Lösung gewaschen. Nach Neubauer erfolgte die Zellzahlbestimmung (3,97xlO7 für L-929 (TNF) und l,3xlθ7 für WEHI 164/E), die jeweils auf 3xlO4 Zellen/Vertiefung in 100 μL (entspricht 3x1 O^ Zellen/mL) mit R5-Lösung eingestellt werden. Die normierte Zellsuspension wurde auf Mikrotiterplatten (96 Vertiefungen) ausplattiert. Als Kontollen wurden in zwei Vertiefungen Mediumkontrollen (zellfrei), in weiteren zwei Vertiefungen Negativkontrollen (Zellen mit R5-Medium) und eine Vertiefung eine Positivkontrolle (Zellen in R5-Lösung mit 5 μL einer auf 10U/mL eingestellten TNF-Lösung) pipettiert. Die ausplattierten Mikrotiterplatten wurden über Nacht bei 37°C, 6% CO2 inkubiert.
Aus den über Nacht inkubierten Zellsuspensionen werden die Überstände von den am Boden adhärierenden Zellen abgesaugt und mit unterschiedlich verdünnten Actinomycin D-Lösungen sowie Testlösungen eingestellt. Die aktivierten Mikrotiterplatten wurden bei 37°C, 6% CO2, 18
Stunden inkubiert. Nach der Inkubation wurde die Positivkontrolle mikroskopisch auf hinreichende Zellyse kontrolliert. Das Medium der Mikrotiterplatte wurde verworfen und die Platte auf Vlies trocken geklopft. In jede Vertiefung wurden 100 μL einer 0,5%ige Kristallviolettlösung gefüllt und 3 Minuten angefärbt. Anschließend wurde die Färbelösung verworfen, jede Mikroti- terplatte wird vier Mal in destillierten Wasser gewaschen, auf Vlies trocken geklopft und 30 Minuten lang bei 50°C nachgetrocknet. In jede Vertiefung werden anschließend 100 μL 33%ige Essigsäure gefüllt, die Mikrotiterplatten 10 Minuten geschüttelt und die Absorption bei 592 nm im ELISA Gerät gemessen.
Bestimmung des zytoprotektiven Effekts durch Wertbestimmung von IFN im EMC-Virus/L929 (IFN) -Testmodell
In 100 mL Kulturschalen wurden L929 (IFN)-Zellen (Charge Nr. 5577) angezüchtet, trypsiniert, gewaschen und gezählt. Die Suspension wurde auf 2x10-" Zellen/mL eingestellt und in Mikrotiterplatten mit 2x104 Zellen/Vertiefung eingesät, die über Nacht bei 370C, 5% CO2, inkubiert wurden. Anschließend wurde der Überstand jeweils durch je 100 μL Mediumkontrolle, Zellkontrolle, Viruskontrolle mit r-mu-IFN-γ-Kontrolle, r-mu-IFN-γ-Standard (100 LVmL) und Testlösung ersetzt, die alle linear verdünnt wurden.

Die Proben wurden genau acht Stunden lang bei 370C, 5% CO2, inkubiert. Nach Ablauf der Inkubationszeit wurden in die Virus-, Interferonkontrolle und in die Testlösungen je 100 μL der Virus-Gebrauchssuspension pipettiert und über Nacht bei 370C, 5% CO2, erneut inkubiert.
Nach 22 Stunden zeigte sich vollausgeprägte Virusaktivität (Zellysis muß in der Viruskontrolle deutlich ausgeprägt sein), so daß die Lysis abgebrochen wurde. Die Überstände wurden über dem Waschbecken verworfen. In jede Vertiefung wurde 100 μL 0,5%ige Kristallviolett-Lösung pipettiert. Nach drei Minuten Färbezeit wurde die überschüssige Lösung entfernt und die Mikro- titerplatten mit destillierten Wasser nachgewaschen und bei 50°C 10 Minuten nachgetrocknet. In die vollständig trockenen Vertiefungen wurden 100 μL Essigsäure (33%) pipettiert und 10 Minuten geschüttelt. Die Bestimmung der Absorption der Kristallviolettfärbung erfolgt am ELISA- Reader bei 550 nm. Die mittlere Absorption der r-mu-IFN-γ-Kontrolle wurde gleich 100% und die der Viruskontrolle gleich 0% Zellprotektion gesetzt. Der EDgQ- Wert des r-mu-IFN-γ-
Standards lag bei 1,076 LVmL (bestimmt aus 8 Standardkurven).
Die in der vorstehenden Beschreibung sowie in den Ansprüchen offenbarten Merkmale der Erfindung können sowohl einzeln aus auch in beliebiger Kombination für die Verwirklichung der Erfindung in ihren verschiedenen Ausführungsformen wesentlich sein.
Claims
1. Verfahren zur Extraktion von Pflanzen, insbesondere Wurzeln, der Pflanzengattung Pe- largonium, dadurch gekennzeichnet, daß die Pflanzen mit einem ethanolfreien Lösungsmittel oder einer ethanolfreien Lösungsmittelmischung extrahiert werden, das bzw. die ausgewählt wird bzw. werden aus Wasser und zumindest einem einwertigen Alkohol mit mindestens drei Kohlenstoffatomen, Wasser und zumindest einem mehrwertigen Alkohol, Wasser und zumindest einer anorganischen, organischen, ein- oder mehrwertigen Säure oder einem Derivat derselben, zumindest einem pflanzlichen Öl und Kohlendioxid.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß die Extraktion eine Perkolation und/oder eine einstufige oder mehrstufige, vorzugsweise zweistufige Mazeration umfaßt.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß der mehrwertige Alkohol ausgewählt wird aus der Gruppe bestehend aus Polyethylenglycol, Glycerol, Propy- lenglycol, Sorbitol, Xylitol oder Mischungen derselben.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß das Polyethylenglycol ein Molekulargewicht von etwa 40 bis etwa 40000, bevorzugt von etwa 200 bis etwa 2000, besonders bevorzugt von etwa 200 bis etwa 800 aufweist.
5. Verfahren nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, daß die Pflanzen zerkleinert, gemahlen und/oder getrocknet und/oder frisch eingesetzt werden.
6. Verfaliren nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, daß das Gewichtsverhältnis Wasseπmehrwertiger Alkohol in einem Bereich von 95:5 bis etwa 5:95 liegt.
7. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, daß das Gewichtsverhältnis Wasser: organische ein- oder mehrwertige Säure in einem Bereich von 99,9:0,1 bis etwa 80:20 liegt. 46
8. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, daß zumindest ein pflanzliches Öl oder Kohlendioxid als Extraktionsmittel eingesetzt wird.
9. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, daß das Gewichtsverhältnis Wasseπanorganische Säure in einem Bereich von 99,9:0,1 bis etwa 80:20 liegt.
10. Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß das Gewichtsverhältnis von Wasseπmehrwertigem Alkohol bei einer Mazeration von etwa 80:20 bis etwa 60:40, bevorzugt etwa 70:30 reicht.
11. Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß das Gewichts Verhältnis von Wasseπmehrwertigem Alkohol bei einer Perkolation von etwa 90:10 bis etwa 70:30, bevorzugt etwa 80:20 reicht.
12. Verfahren nach einem der Ansprüche 2 bis 11, dadurch gekennzeichnet, daß vor der Perkolation ein Anmaischen der Pflanzen erfolgt.
13. Extrakt der Pflanzengattung Pelargonium, gelöst in einem ethanolfreiem Lösungsmittel oder einer ethanolfreien Lösungsmittelmischung, ausgewählt aus der Gruppe bestehend aus Wasser und zumindest einem einwertigen Alkohol mit mindestens drei Kohlenstoffatomen, Wasser und zumindest einem mehrwertigen Alkohol, Wasser und zumindest einer anorganischen, organischen, ein- oder mehrwertigen Säure oder einem Derivat derselben, zumindest einem pflanzlichen Öl und Kohlendioxid, insbesondere hergestellt nach einem Verfahren der Ansprüche 1 bis 12.
14. Extrakt nach Anspruch 13, dadurch gekennzeichnet, daß der Extrakt von Wurzeln der Pflanze Pelargonium sidoides, Pelargonium reniforme, Pelargonium lobatum und/oder Pelargonium triste ist.
15. Extrakt nach Anspruch 13 oder 14, dadurch gekennzeichnet, daß der Extrakt zusätzlich ein Eindickungsmittel enthält und in fester, erstarrter Form vorliegt. 47
16. Extrakt nach Anspruch 15, dadurch gekennzeichnet, daß der Extrakt in Form eines Lutschbonbons, einer Lutschtablette, einer Bukal- oder Lingualarzneiform oder eines Lutschers vorliegt.
17. Verwendung des Extrakts nach einem der Ansprüche 13 bis 16 zur Behandlung von akuten und/oder chronischen Entzündungserkrankungen und/oder Infektionen bei Säugetieren.
18. Verwendung nach Anspruch 17 beim Menschen, Haus- und Nutztieren, bevorzugt Pferd, Hund, Katze, Schwein oder Huhn.
19. Verwendung nach Ansprach 17 oder 18, dadurch gekennzeichnet, daß der Extrakt oral verabreicht wird.
20. Verwendung nach einem der Ansprüche 17 bis 19 zur Behandlung oder Prophylaxe von HIV-Infektionen oder HIV-assoziierten Infektionen.
