  • the medium was changed to a drug-free medium and the cells were further cultured for 96 hr.
  • FK228 was dissolved in ethanol and diluted with a medium before use, and cisplatin was diluted with a medium before use. The antitumor activity after each treatment was evaluated by MTT assay.
  • 3- (4 , 5-dimethylthiazol-2-yl) - 2,5-diphenyltetrazonium bromide (MTT) was dissolved in PBS at a concentration of 2.5 mg/mL, 20 ⁇ l thereof was added to each well and cultured for 4 hr.
  • Formazan formed by living cells was dissolved by adding acidic isopropanol (100 ⁇ l) and the absorbance of each well was measured at a wavelength of 580 nm. With the absorbance (OD) of non-treated cells as 100%, concentration—response curves of antitumor activity of a single pharmaceutical agent and a combination of pharmaceutical agents were drawn.
