発酵生成物および食中毒防止方法
技術分野
本発明は、 サルモネラ菌の生体内での増殖および腸管粘膜内への侵入を阻止し、 サルモネラ菌による食中毒の予防を目的とした発酵生成物および食中毒防止方法 に関する。 背景技術
メチニコフの長寿説以来、 乳酸菌あるいは乳酸菌を用いた発酵生成物の生理効 果に関して多くの研究がなされている。
ヨーグルトなどの発酵乳に含まれる乳酸菌の機能としては、 腸内菌叢改善効果、 整腸作用などがよく知られている。 このように、 乳酸菌には種々の保健効果が期 待できることから、 特にヒトの腸内からも検出されるラクトバチラス · ァシドフ ィラス、 ラクトバチラス · 力ゼィ、 ビフイ ドバクテリウム属を中心にこれらの菌 種を用いた発酵乳や乳酸菌飲料等の種々の発酵生成物が市販されている。
ところで、 グラム陰性の通性嫌気性桿菌であるサルモネラ菌は、 例えば食物と ともに人間の体内に入って繁殖し、 中毒症状を引き起こす病原菌である。 サルモ ネラ菌による中毒症状は、 悪寒、 高熱、 嘔吐、 下痢、 腹痛等を伴うものであって、 感染後数時間から 2 4時間後に発症し、 その症状は一般に重く、 回復が遅いとい う特徴を有する。
通常、 サルモネラ菌に感染し、 発病した場合には、 クロラムフエ二コール系等 の抗生物質を投与する治療が行われる。 しかし、 かかる抗生物質を投与すること により、 過敏症、 ショック症状、 肝障害、 ビタミン欠乏症等の副作用のおそれが あるため、 多量に投与するのは好ましくない。
従って、 サルモネラ菌の感染を予防する実用的な対策が要望されている。 発明の開示
本発明の目的は、 副作用のおそれがなく、 サルモネラ菌の感染予防に有効な発
酵生成物びよびサルモネラ菌に起因する食中毒の防止方法を提供することにある。 本発明者らは、 上記課題を解決するために鋭意研究を重ねた結果、 ラクトバチ ラス · プランタラムで発酵させた発酵生成物が、 サルモネラ菌に対する感染予防 に有効であり、 しかも副作用のおそれがないという新たな事実を見出し、 さらに 研究して本発明を完成するに至った。
すなわち、 本発明は、
( 1 ) ラクトバチラス · プランタラムで食品素材を発酵させたことを特徴とする、 サルモネラ菌に対する感染予防用の発酵生成物、
(2) 茶抽出物を加えて発酵させた上記 ( 1 ) 項記載の発酵生成物、
(3) 発酵乳または乳酸飲料の形態である上記 ( 1 ) 項または (2 ) 項記載の発 酵生成物、
(4) 食品素材が動物乳またはその脱脂乳である上記 (1 ) 項記載の発酵生成物、
( 5) ラクトバチルス · プランタラムを生体内に摂取しサルモネラ菌による感染 を予防することを特徴とするサルモネラ菌による食中毒の防止方法、
(6) ラクトバチラス · プランタラムで食品素材を発酵させ、 該発酵生成物を生 体内に摂取しサルネラ菌による感染を予防することを特徴とするサルモネラ菌に よる食中毒の防止方法、 および
( 7) 食品素材が動物乳またはその脱脂乳である上記 (6) 項記載の食中毒の防止 方法、

発明を実施するための最良の形態
本発明に用いられるラクトバチラス · プランタラム種は乳酸桿菌の 1種であ つて、 具体的には、 Lac tobaci l lus plan tarum [ATCC14431株] 等があげられる。 このラクトバチラス . プランタラムは、 生育環境内のマンガンを菌体内に取り 込むことで良好に生育することが知られている。 従って、 マンガンを多く含有す る茶抽出物を加えて発酵させたときは、 発酵を促進でき、 単位容量当たりの菌数 を増加させることができる。
サルモネラ菌内毒素による毒作用の発現を防止するには、 サルモネラ菌の増殖 を阻害すると同時にサルモネラ菌の腸管粘膜内への侵入を阻止することが重要で ある。 よって、 茶抽出物を加えて、 ラクトバチラス ' プランタラムで発酵させて 得られた発酵生成物におけるサルモネラ菌に対する感染予防作用と、 乳酸菌によ る腸内菌叢改善効果、 整腸作用などとの連関作用は、 サルモネラ菌に対する感染 予防という観点から非常に重要であると考えられる。
本発明の発酵生成物の原料となる食品素材としては、 ラクトバチラス · プラン タラムを接種させることにより発酵し得る素材であれば、 特に限定されないが、 例えば牛乳、 ャギ乳等の動物乳またはそれらの脱脂乳があげられる。
本発明の発酵生成物の形態には、 例えば、 ヨーグルトなどの発酵乳、 乳酸菌飲 料等が含まれる。
また、 本発明の発酵生成物は、 ラクトバチラス ' プランタラム種と、 必要に応 じて茶抽出物とを含有する乾燥品 (好ましくは凍結乾燥品) であってもよい。 こ の乾燥品は粉末、 顆粒、 錠剤などの任意の形態で用いられる。 この乾燥品はその まま摂取してもよく、 あるいは家庭などにおいて市販牛乳に添加し、 発酵させて ヨーグルトを作製し摂取してもよい。 また、 この乾燥品は冷凍品などの形態であ つてもよい。
さらに、 本発明の発酵生成物は、 ラクトバチラス ' プランタラム種と、 必要に 応じて茶抽出物とからなる医薬形態の製剤であってもよい。 この製剤は医薬上許 容しうる種々の単体と混合して調整される乾燥品 (錠剤、 粉末、 顆粒、 カプセル
などの形態) であってもよく、 あるいは液剤の形態であってもよい。
ラクトバチラス · プランタラムは、 生育のためにマンガンを必要とするが、 マ ンガン自体は食品添加物として認可されたものが存在しないことから、 菌体にマ ンガンを供給するには、 無機化合物としてではなく、 マンガンを多く含有する茶 葉のほか、 キャベツ、 ホウレンソゥに代表される野菜類やハーブ類などのマンガ ン含有天然素材が用いられる。 これらの天然素材の添加は、 ラクトバチラス ' プ ランタラムの生育に必要なマンガンを供給するうえで重要であるが、 とくに茶葉 はそれ自体がカテキン、 ビタミン c、 各種微量元素などを多く含有し、 栄養補給 効果も期待されるので、 好都合に利用される。
茶葉等のマンガン含有天然素材は、 抽出物または粉末の状態で添加するのが、 菌によるマンガンの取込みが容易になるので好ましい。 抽出物としては、 前記天 然素材を水で抽出した水溶液をそのまま使用してもよい。 粉末化は、 天然素材を 水および Zまたは水混和性有機溶媒 (例えばエチルアルコールなどのアルコール 類) で抽出し、 次いで水に対して非混和性の有機溶媒 (例えば、 クロ口ホルム、 酢酸ェチルあるいは n—ブタノールなど) で抽出し、 有機相と水相とに分けて、 水相から溶解固形分を回収し乾燥することによって行われる。 また、 水および または水混和性有機溶媒で抽出した溶液から固形分を回収し、 粉末化したもので あってもよく、 あるいは単に前記天然素材を細かく粉碎したものでもよい。
前記茶葉としては、 例えば玉露、 玉露茶、 抹茶、 煎茶、 番茶、 粉茶、 芋茶ある いはほうじ茶などの緑茶 (不発酵茶) 、 紅茶などの発酵茶、 ウーロン茶あるいは バオチョン茶 (ジャスミン茶) などの半発酵茶などがあげられる。
マンガン含有天然素材の添加量は、 発酵製品の場合で製品 1 k gに対してマン ガン換算で 4〜 2 0 m g程度、 好ましくは 4〜 8 m g程度であるのがよい。 また、 主に菌体とマンガン含有天然素材と賦形剤とからなる乾燥品の場合には、 マンガ ン含有天然素材の添加量は、 製品 1 0 gに対してマンガン換算で 4〜 2 O m g程 度、 好ましくは 4〜 8 m g程度であるのがよい。
本発明の発酵生成物の形態としては、 前述のように、 ヨーグルトなどの発酵乳、 乳酸菌飲料および乾燥品が代表例としてあげられる。
上記発酵乳は、 牛乳または脱脂乳 1 リットルに対し、 必要に応じて茶葉等のマ ンガン含有天然素材の所定量を加え、 ラクトバチラス ' プランタラムを接種し、
3 0〜 3 5 で 1 2〜4 8時間程度発酵させることによって得られる。 乳酸菌ス ターター、 マンガン含有天然素材の添加量、 発酵時間などを調整することにより、 固形 (静置ヨーグルト) 、 半固形 (攪拌ヨーグルト) および液状 (ドリンクョ一 ダルト) のヨーグルト形態のものが製造できる。 また発酵乳に甘味料 (例えば、 ブドウ糖、 ショ糖など) や果肉 (例えば、 グレープフルーツ、 リンゴ、 オレンジ あるいはレモンなど) 、 ミネラル源としての無機電解質 (例えば、 塩化ナトリウ ム、 塩化カリウム、 塩化マグネシウムあるいは塩化カルシウムなど) 、 ビタミン 類、 香味料などを適宜加えてもよい。
上記乳酸菌飲料は、 脱脂乳、 糖 (例えばブドウ糖、 ショ糖など) などの混合液 に、 必要に応じて茶葉等のマンガン含有天然素材の所定量を加え、 ラクトバチラ ス · プランタラムを接種し、 3 0〜 3 5 °Cで 1 2〜4 8時間程度発酵させること によって得られる。 脱脂乳と水、 果汁などの希釈液との割合で、 液状ヨーグルト タイプやジュースタイプなどが製造できる。 発酵ベースとしては、 前記脱脂乳の ほか、 乳清、 低脂肪乳なども使用可能である。 希釈液としては、 前記水のほか、 果肉、 ラクトコーヒーなどがあげられる。
また、 本発明における乾燥品は、 5 X 1 0 8〜5 X 1 0 1°相当の菌体と、 必要 に応じて 2〜4 gの前記マンガン含有天然素材とを混合し、 賦形剤を加えて乾燥 したものである。 賦形剤としては、 例えば乳糖、 ブドウ糖、 ショ糖あるいはオリ ゴ糖などがあげられる。
上記乾燥品は、 前述のように、 そのまま摂取してもよく、 あるいは一般家庭な どにおいて牛乳に添加し、 室温で 1 2〜 2 4時間発酵させてヨーグルトを作製し 摂取してもよい。
かく して得られる発酵生成物を、 ヒトを含む哺乳動物が前述のような形態で安 全に経口摂取することができる。 摂取量および摂取時期も特に制限はなく、 例え ば通常の食生活において、 適宜、 摂取すればよい。 本発酵生成物を摂取すること により、 サルモネラ菌の感染を予防し、 食中毒の発症例えば、 悪寒、 高熱、 嘔吐、
下痢あるいは腹痛などの防止に有効である。 サルモネラ菌による食中毒の原因と なる血清型は動物由来であり、 例えば■ a oTJe a typhi murium, Salmonella ha dar, Salmonella virchow, Salmonella agona などカ'あげられる力 とりわけ Salmonella typhi murium に起因する食中毒の予防に有効である。
本発明において、 サルモネラ属菌の感染予防は、 上述のようにラクトバチラ ス · プランタラムによる発酵生成物の形態で摂取することのほかに、 ラクトバチ ラス · プランタラム菌体と必要に応じてマンガン供給天然素材 (例、 茶抽出物) とを含有する調製物 (例、 凍結乾燥品) のかたちで直接に摂取してもよい。 実施例
以下に実施例および試験例を挙げて、 本発明をさらに具体的に説明する。
〔発酵生成物の製造〕
実施例 1 (ヨーグルトタイプ)
滅菌した牛乳 1 リットルに対して、 抹茶エキス [長谷川香料 (株) 製] を 7m 1添加した。 この抹茶エキス添加牛乳にラクトバチラス · プランタラムを接種し、 30〜33 °Cの温度を保ちながら 1 2〜24時間発酵させた。
常法に従って、 乳酸菌スターターおよび抹茶エキスの添加量、 発酵時間などの 製造条件を調整することにより、 以下のヨーグルト形態を作製することができた。

実施例 2 (乳酸菌飲料タイプ)
所定量の脱脂乳およびショ糖の混合液に抹茶エキスを添加したのち、 ラクトバ チラス · ブランタラムを接種し、 30〜33 °Cの温度を保ちながら 1 2〜24時 間発酵させた。 次いで、 発酵液を水で希釈し、 安定剤、 香料などを加えて、 液状 ヨーグルトタイプの乳酸菌飲料を得た。 このものの組成を表 1に示す。
実施例 3 (乳酸菌飲料タイプ)
所定量の脱脂乳およびショ糖の混合液に茶葉 (粉末化緑茶) であるシングルセ
ル化茶葉 (シングルセル食品 (株) 製) を添加したのち、 ラクトバチラス ' ブラ ンタラムを接種し、 3 7 °Cの温度を保ちながら 1 8時間発酵させた。 ついで、 発 酵液を水およびオレンジ果汁で希釈し、 安定剤、 香料などを加えて、 ジュースタ イブの乳酸菌飲料を得た。 このものの組成を表 1に示す。 表 1

5 X 101Q相当の菌体 (ラクトバチルス · プランタラム) と 4 gの茶葉 (粉末化緑 茶) とを混合し、 乳糖およびブドウ糖を賦形剤として凍結乾燥し、 錠剤タイプ の凍結乾燥品を得た。 このものはそのまま摂取するのに適していた。
実施例 5 (凍結乾燥タイプ)
5 X 108相当の菌体 (ラクトバチルス · プランタラム) と 4 gの茶葉 (粉末化緑 茶) とを混合し、 乳糖およびブドウ糖を賦形剤として凍結乾燥し、 錠剤タイプの 凍結乾燥品を得た。 このものを市販牛乳に添加し、 室温で 12〜24時間発酵させ たところ、 ヨーグルトを作製できた。 従って、 この凍結乾燥品は一般家庭でョー ダルトを作製するのに適している。
[サルモネラ菌の感染予防作用の評価]
試験例 1
実施例 1で得られたドリンクヨーグルト (発酵生成物) および牛乳を水で希釈 して調整した 75%水溶液 (以下、 それぞれ 「発酵生成物」 および 「牛乳」 とい う) を用いて、 以下の試験を行った。
1グループが I CR系雄性マウス ( 3週齢、 体重 8〜 g) 30匹からなる 2つの グループのうち、 一方のグループには発酵生成物を、 他方のグループには牛乳を、 それぞれ 8日間自由に摂取させた。 なお、 牛乳を摂取させたグループを対照とし た。
次いで、 発酵生成物または牛乳の摂取開始から 9日目に、 それぞれのグループ に対し、 サルモネラ菌 Sal議 el la typh i murium, ネズミチフス菌、 S I C—8 401株) 3. 0 X 101QCFUを経口投与し、 引き続き、 それぞれのグループに発酵生成 物または牛乳を 20日間自由に摂取させて、 サルモネラ菌投与後のマウスの生存 数を観察した。
両方のグループのマウスについて、 サルモネラ菌投与した日 (0日目) 以降に おける死亡数の経日変化を表 2に示す。 2 0日経過後のマウスの生存率は、 発酵 生成物を投与したマウスが 76. 7 %、 対照のマウスが 6. 7 %であった。 表 2

また、 サルモネラ菌投与後におけるマウスの生存率 (%) の経日変化を図 1に、 サルモネラ菌投与翌日における体重変化量 (g) およびサルモネラ菌投与後 2 0 日間におけるマウスの下痢発生匹数をそれぞれ図 2および図 3に示す。
表 2および図 1より明らかなように、 発酵生成物を継続して摂取させたマウス の生存率は、 牛乳を摂取させたマウス (対照) に比べて高く、 有意な差が見られ た。
図 2に示すように、 サルモネラ菌を投与した翌日の体重減少量は、 牛乳を摂取 させたマウス (対照) の平均値が 2. 4 gであるのに対し、 発酵生成物を摂取させ たマウスの平均値は 0. 7 gであって、 対照に比べて体重の減少が抑制されていた。 従って、 サルモネラ菌の感染初期の段階から感染予防作用を発現することが示唆 された。
また図 3に示すように、 牛乳を摂取させた場合は 30匹中 2 4匹ものマウスが 下痢症状を示したのに対し、 発酵生成物を摂取させた場合は 30匹中 11匹にとど まっており、 有意な差が見られた。 産業上の利用可能性
本発明の発酵生成物、 ラクトバチラス · プランタラムと、 必要に応じて茶抽出 物とを加えて発酵させたものであって、 例えば発酵乳、 乳酸飲料として提供され る。 かかる発酵生成物はサルモネラ菌に対する感染予防作用を有することから、 サルモネラ菌による食中毒を効果的に予防できる。
Claims
1 . ラクトバチラス · プランタラムで食品素材を発酵させたことを特徴とする、 サルモネラ菌に対する感染予防用の発酵生成物。
2. 茶抽出物を加えて発酵させた請求項 1記載の発酵生成物。
3. 発酵乳または乳酸飲料の形態である請求項 1 または 2記載の発酵生成物。
4. 食品素材が動物乳またはその脱脂乳である請求項 1記載の発酵生成物。
5. ラクトバチルス · プランタラムを生体内に摂取しサルモネラ菌による感染を 予防することを特徴とするサルモネラ菌による食中毒の防止方法。
6. ラクトバチラス , プランタラムで食品素材を発酵させ、 該発酵生成物を生体 内に摂取しサルネラ菌による感染を予防することを特徴とするサルモネラ菌 による食中毒の防止方法。
7. 食品素材が動物乳またはその脱脂乳である請求項 6記載の食中毒の防止方法。
