Abstract

Description
Verwendung von Lecithin-Analoga als antivirale Arzneimittel, sowie neue Verbindungen.
Gegenstand der vorliegenden Erfindung ist die Verwendung von
Lecithin-Analoga zur Herstellung von antiviralen Arzneimitteln.
Die vorliegende Erfindung betrifft die Verwendung von Verbindungen der Formel I +
R1-X-R2-Y-R3-Z- -R4-N(R5)3 (I),
X einen Valenzstrich, ein Sauerstoff- oder Schwefelatom, eine
Sulfinyl-, Sulfonyl-, Carbonyl-, Aminocarbonyl-, Carbonylamino- oder die Ureidogruppe -NH-CO-NH- oder einen C3-C8- Cycloalkylen- oder Phenylenrest,
Y ein Sauerstoffatom oder die Gruppen -O-CO-O-, -O-CO-NH-,
-O-CS-NH-
R1 ein Wasserstoffatom, einen geradkettigen oder verzweigten, gesättigten oder ungesättigten Alkylrest mit 1-18 bzw. 2-18 Kohlenstoffatomen, der gegebenenfalls ein- oder mehrfach durch Phenyl-, Halogen-, C1-C4-Alkoxy-, C1-C4-Alkylmercapto-, C1-C4- Alkoxycarbonyl-, C1-C4-Alkansulfinyl- oder C1-C4-Alkansulfonylgruppen substituiert ist,

Methylengruppe durch einen C5-C7-Cycloalkanring ersetzt sein kann, und die gegebenenfalls ein- oder mehrfach durch Hydroxy- Halogen-, Nitril-, eine C5-C7-Cycloalkyl-, Phenyl-, C1-C20- Alkoxycarbonyl-, C1-C20-Alkylaminocarbonyl-, C1-C20- Alkylmercapto-, C1-C20-Alkansulfinyl-, C1-C20-Alkansulfonyl-, C1-C20-Alkylcarbonylaminogruppe oder durch C1-C20-Alkoxy substituiert ist, das wiederum durch Phenyl, C1-C20- Alkylmercapto, C1-C20-Alkansulfinyl, C1-C20-Alkansulfonyl, C1- C20-Alkylcarbonylamino, C1-C20-Alkoxycarbonyl, Nitril,
Hydroxy, C1-C20-Αlkoxy oder C1-C20-Alkylaminocarbonyl
substituiert sein kann,
R4 eine geradkettige oder verzweigte Alkylenkette mit 2-5 Kohlenstoffatomen,
R5 Wasserstoff oder eine C1-C6-Alkylgruppe und
Z Sauerstoff oder Schwefel bedeuten, deren pharmakologisch unbedenkliche Salze sowie optische Isomere zur Herstellung von Arzneimitteln mit antiviraler Wirkung.
In der Europäischen Patentschrift EP-B-69,968 werden Phospholipide mit der oben angegebenen Struktur beschrieben, sowie deren Verwendung zur Herstellung von Arzneimitteln. In dieser Patentschrift ist jedoch ausschließlich die cancerostatische Wirkung dieser Phospholipide beschrieben, die sich insbesondere zur Herstellung von Antitumormitteln eignen.
Im E-A-20 09 341 wird der 3-Octadecyloxy-propyl-1-phosphorsäure- monocholinester als immunologisches Adjuvans, in DE-A- 20 09 342 dessen 2-Hydroxyderivat (3-Octadecyloxy-2-hydroxypropyl 1-phosphorsäure-monocholinester) als Mittel zur Steigerung der natürlichen Resistenz des Organismus beschrieben.
Ferner sind in der US-Patentschrift US 4.372.949
Dialkoxyphospholipide mit immunstimulierenden Eigenschaften zur Behandlung von Krebserkrankungen beschrieben. Aus der japanischen Offenlegungsschrift JP-A-5002-636 sind allgemein einige Glycerophosphorylcholine mit Urethan- bzw. Thiourethan-Gruppierungen bekannt.
Es wurde nun überraschenderweise gefunden, daß Phospholipide der Formel I eine ausgeprägte antivirale Wirkung aufweisen, und sich daher besonders gut zur Behandlung von viralen bzw. retro-viralen Infektionen eignen. Virale Infektionen von Säugern, insbesondere des Menschen, sind weit verbrt tet. Trotz intensiver Bemühungen ist es bisher nicht gelungen, Chemotherapeutika bereitzustellen, die ursächlich oder symptomatisch mit dem viral oder retroviral bedingten Krankheitsgeschehen mit erkennbar substantiellem Erfolg interferieren. Es ist heutzutage nicht möglich, bestimmte Viruserkrankungen, wie zum Beispiel das Aquired Immune Deficiency Syndrom (AIDS), den AIDS-related-complex (ARC) und deren Vorstadien, Herpes-, Cytomegalie-Virus (CMV)-, Influenza- und andere Virusinfektionen zu heilen oder chemotherapeutisch deren Symptome günstig zu beeinflussen. Derzeit steht beispielsweise für die Behandlung von AIDS fast ausschließlich das 3'-Azido-3'-deoxy-thymidin (AZT), bekannt als Zidovudine oder RetrovirR, zur Verfügung. AZT ist jedoch durch eine sehr enge therapeutische Breite bzw. durch bereits im therapeutischen Bereich auftretende, sehr schwere Toxizitäten charakterisiert (Hirsch, M.S. (1988) J.Infec.Dis. 157, 427-431).
Es besteht daher ein sehr großes Bedürfnis an Chemotherapeutika, die möglichst spezifisch mit viral oder retroviral bedingten Erkrankungen oder deren Symptomen interferieren, ohne jedoch die anderen, normal ablaufenden natürlichen Körperfunktionen zu beeinflussen.
Der vorliegenden Erfindung lag daher die Aufgabe zugrunde,
antiviral wirksame Mittel zur Verfügung zu stellen, die bei der Behandlung oder Prophylaxe von viral- oder retroviral-bedingten Erkrankungen verwendet werden können, und die die aus dem Stand der Technik bekannten Nachteile nicht besitzen.
Es wurde nun überraschenderweise gefunden, daß die Phospholipide der Formel I sehr spezifisch die Virusvermehrung hemmen.
Insbesondere sind hierbei folgende Viren zu nennen:
HTLV-I und II, HIV, Herpes, Sendai, Cytomegalie (CMV), Influenza, Parainfluenza, Epstein-Barr (EBV), Vesikular Stomatitis Virus (VSV), Hepatitis, Meningitis, Enzephalitis, etc.. Die genannten Verbindungen können vorteilhaft prophylaktisch oder therapeutisch bei der Behandlung von allen Krankheiten eingesetzt werden, bei denen eine virale bzw. retrovirale Infektion von pathophysiologischer, symptomatischer oder klinischer Relevanz ist.
Bei den Phospholipiden der Formel I bedeutet Alkyl in dem Substituenten R5 einen Kohlenwasserstoffrest mit 1-6 Kohlenstoffatomen, insbesondere den Methyl- oder Ethylrest.
Die Alkylkomponenten der in der Definition von R1-R3 genannten Substituenten, wie z. B. Alkoxy, Alkoxycarbonyl, Alkylmercapto, Alkansulfinyl oder Alkansulfonyl, bedeuten in allen Fällen in der Regel Reste mit 1-20 Kohlenstoffatomen. Insbesondere kommen solche Reste mit einer Kettenlänge von mindestens 5, bevorzugt 10 C- Atomen, und höchstens bis zu 18, bevorzugt 16 oder 15 C-Atomen in Frage, wie z. B. Alkylteile mit 5-18, insbesondere 8-16, 8-15, 10- 16 oder 10-15 C-Atomen. Beispielsweise seien der Octadecyl- mercapto-, Tetradecyloxy- oder Octylmercaptorest genannt.
Die Cycloalkylenreste der Gruppe X sind Reste mit 3-8 Kohlenstoffatomen, insbesondere der Cyclopropylen-, Cyclopentylen- und Cyclohexylenrest.
C3-C7-Cycloalkanringe, die auch Bestandteil der Alkylenkette der Gruppe R3 sein können, sind insbesondere der Cyclopentan-,
Cyclohexan- und der Cycloheptanring, wobei die Ringe noch durch eine C1-C6-Alkylgruppe substituiert sein können. In diesem Sinne kommen für R3 beispielsweise eine C1-C4-Alkyl-C3-C7-cycloalkylen oder C3-C7-Cycloalkyl-C1-C4-alkylengruppe in Frage.
Halogen bedeutet Fluor, Chlor, Brom und Jod, vorzugsweise jedoch Fluor.
Die Gruppe R1-X-R2 bedeutet für den Fall, daß X einen Valenzstrich darstellt, eine Alkylgruppe mit bis zu 20 Kohlenstoffatomen, die geradkettig oder verzweigt, gesättigt oder ungesättigt sein kann. Als geradkettige Gruppen kommen vorzugsweise der Eicosyl-, Octadecyl-, Heptadecyl-, Hexadecyl-, Tetradecyl-, Dodecyl- und der Octylrest in Frage.
In der Definition von R1-R3 versteht man unter der ungesättigten Alkylgruppe insbesondere soche, die bis zu vier Doppelbindungen, vorzugsweise jedoch ein oder zwei Doppelbindungen enthalten.
Die Gruppe R3 besteht bevorzugt aus einer -CH2-CH2-CH2-Gruppe, deren mittlere Methylengruppe ein oder zweifach durch Alkyl-, Alkoxy-, Alkylmercapto-, Alkansulfinyl-, Alkansulfonyl-, Alkoxy- alkylen-, Benzyloxy-, Hydroxy- oder Halogen substituiert sein kann und die gegebenenfalls an den Kohlenstoffpositionen C1-C3 durch Alkylgruppen substituiert sein kann, wobei diese auch einen Ring bilden können.
Bevorzugte Verbindungen der Formel I sind insbesondere Derivate des Propanol-(1)-, Propanol-(2) - bzw. Propanthiol-(1)-phosphor- säuremonocholinesters, wobei die 3-Stellung des Propanols durch einen Alkoxyrest und gegebenenfalls auch in 1-Stellung oder 2- Stellung substituiert ist.
Bevorzugt kommen solche Verbingungen der Formel I zur Anwendung, in denen X einen Valenzstrich, ein Sauerstoff- oder Schwefelatom, Y ein Sauerstoffatom, Z Sauerstoff- oder Schwefelatom, R1 ein Wasserstoffatom, einen geradkettigen oder verzweigten, gesättigten
oder ungesättigten Alkylrest mit 1-18 Kohlenstoffatomen, R2 eine geradkettige oder verzweigte, gesättigte oder ungesättigte
Alkylenkette mit 1-18 Kohlenstoffatomen, R3 eine geradkettige oder verzweigte, gesättigte oder ungesättigte Alkylenkette mit 2-8 Kohlenstoffatomen, in der eine Methylengruppe durch einen C5-C7- Cycloalkanring ersetzt sein kann, und die gegebenenfalls ein- oder mehrfach durch Hydroxy, C1-C20-Alkylmercapto-, C1-C20- Alkansulfinyl-, C1-C20-Alkansulfonyl- oder durch C1-C20-Alkoxy substituiert ist, R4 eine geradkettige oder verzweigte
Alkylenkette mit 2-4 Kohlenstoffatomen und R5 Wasserstoff oder eine C1-C6-Alkylgruppe bedeuten.
Insbesondere sind Verbindungen der Formel I bevorzugt, in denen X einen Valenzstrich, ein Sauerstoff- oder Schwefelatom, Y ein
Sauerstoffatom, Z ein Sauerstoff- oder Schwefelatom, R1 ein
Wasserstoffatom, ein geradkettiger oder verzweigter, gesättigter oder ungesättigter Alkylrest mit 1-18 Kohlenstoffatomen ist, R2 eine geradkettige oder verzweigte, gesättigte oder ungesättigte Alkylenkette mit 1-18 Kohlenstoffatomen bedeutet, R3 eine
Alkylenkette mit 3 Kohlenstoffatomen ist, die ein- oder mehrfach durch C9-C15-Alkylmercapto-, C9-C15-Alkansulfinyl-, C9-C15- Alkansulfonyl oder durch C9-C15-Alkoxy substituiert ist, R4 eine Alkylenkette mit 2 Kohlenstoffatomen und R5 Wasserstoff, Methyl oder Ethyl bedeuten.
Gegenstand der vorliegenden Erfindung sind jedoch auch neue Verbindungen, die eine Auswahl aus den in der EP-B-69,968 beschriebe nen Phospholipiden darstellen. Es hat sich gezeigt, daß
insbesondere Phospholipide der Formel II
A die Gruppe
bedeutet, und
R1 eine C1-C20-Alkoxycarbonyl-, C1-C20-Alkylaminocarbonyl-, C1- C20-Alkylmercapto-, C1-C20-Alkansulfinyl-, C1-C20-Alkansulfonyl-, C1-C20-Alkylcarbonylamino- oder C1-C20-Alkoxygruppe darstellt, und die übrigen Reste R1, X, R2, Y, Z, R4 und R5 im wesentlichen die oben angegebene Bedeutung haben.
Die Alkylkomponenten der in der Definition von Formel II genannten Substituenten R1, R2 und R3, wie z. B. Alkoxy, Alkoxycarbonyl, Alkylmercapto, Alkansulfinyl oder Alkansulfonyl, bedeuten in allen Fällen in der Regel Reste mit 1-20 Kohlenstoffatomen, die
geradkettig oder verzweigt sein können. Insbesondere kommen solche Reste mit einer Kettenlänge von mindestens 5, bevorzugt 10 C- Atomen, und höchstens bis zu 18, bevorzugt 16 oder 15 C-Atomen in Frage, wie z. B. Alkylteile mit 5-18, insbesondere 8-16, 8-15, 10- 16 oder 10-15 C-Atomen. Beispielsweise seien der Octadecylmercapto-, Tetradecyloxy- oder Octylmercaptorest genannt.
In der allgemeinen Formel I und II zeigen insbesondere solche Phospholipide eine gute antivirale Wirkung, wenn sie durch die folgenden Merkmale gekennzeichnet sind:
R1 bedeutet vorzugsweise eine geradkettige C5-C15-Alkylgruppe, die durch eine C1-C6-Alkoxygruppe substituiert sein kann.
Bevorzugt ist R1 eine Pentyl-, Hexyl-, Heptyl-, Decyl-,
Dodecyl-, Tridecyl-, Tetradecyl- oder Pentadecylgruppe. Als C1-C6-Alkoxy-Substituent kommt vorzugsweise die n- Butyloxygruppe in Frage. Für den Fall, daß X einen Valenz
strich darstellt, kommt insbesondere für R1 ein Wasserstoffatom in Frage.
X bedeutet vorzugsweise einen Valenzstrich oder ein Sauerstoffoder Schwefelatom oder die Gruppe -SO2- vorzugsweise jedoch einen Valenzstrich.
R2 ist vorzugsweise eine geradkettige C2-C18-Alkylen-, insbesondere C9-C15-Alkylengruppe, wie beispielsweise die Ethylen-, Propylen-, Pentylen-, Decylen-, Undecylen-, Dodecylen-,
Tridecylen-, Tetradecylen-, Pentadecylen-, Hexadecylen,
Heptadecylen- oder Octadecylengruppe, wobei die Decylen-, Undecylen-, Dodecylen-, Tridecylen- und Tetradecylengruppe besonders bevorzugt sind.
In der Formel I kommt für R3 insbesondere eine geradkettige C2-C6- Alkylengruppe, beispielsweise die Ethylen-, Propylen- oder
Butylengruppe, vorzugsweise die. Propylengruppe in Frage, die durch eine Hydroxy-, C1-C20-Alkylmercapto-, C1-C20-Alkansulfinyl-, C1- C20-Alkansulfonyl-, C1-C20-Alkoxy-, C1-C20-Alkoxycarbonyl- oder C1-C20-Alkyl-aminocarbonylgruppe substituiert sein kann, wobei die zuvor genannten C1-C20-Alkylteile vorzugsweise 9-15 Kohlenstoffatome enthalten. Besonders bevorzugte Substituenten der Gruppe R3 sind die C1-C20-Alkylmercapto- oder C1-C20-Alkoxygruppe, wie z. B. die Methylmercapto-, Methoxy- und Ethoxygruppe, wobei die C1-C20- Alkoxygruppe durch eine C1-C4-Alkoxygruppe, wie z. B.
Methoxygruppe, substituiert sein kann. Für den Fall, daß R3 eine Propylengruppe darstellt, können die zuvor genannten Substituenten in 1- oder 2-Stellung der Propylengruppe stehen, wobei die Gruppe R1-X-R2-Y- als 3-ständig bezeichnet werden soll, und die Gruppe - Z-P(O2)O-R4-N(R5) 3 dann in Abhängigkeit der oben genannten
Substituenten in 2-bzw. 1-Stellung stehen muß.
Als Substituenten der Gruppe R3 kommen in diesem Sinne vorzugsweise die folgenden Gruppen in Frage: C9-C15-Alkoxy-, wie z.B. die Decyloxy-, Undecyloxy-, Dodecyloxy- oder Tetradecyloxygruppe; eine C9-C15-Alkylmercapto-, wie z.B. Nonylmercapto, Decylmercapto- oder Undecylmercaptogruppe, sowie eine C9-C15-Alkoxycarbonyl-, wie z.B.
die Decyloxycarbonylgruppe; eine C9-C15-Alkylaminocarbonyl-, wie z.B. die Decylaminocarbonyl-; eine C9-C15-Alkylcarbonylaminogruppe, wie z.B. Decylcarbonylaminogruppe.
In der allgemeinen Formel II kommen für R' vorzugsweise
Hydroxy-, C1-C20-Alkylmercapto-, C1-C20-Alkansulfinyl-, C1-C20- Alkansulfonyl-, C1-C20-Alkoxy-, C1-C20-Alkoxycarbonyl-, C2-C20- Alkylcarbonylamino- oder C1-C20-Alkylaminocarbonylgruppe in Frage, wobei die Alkylteile insbesondere 9-15, vorzugsweise 10-12
Kohlenstoffatome enthalten. Bevorzugte Alkylteile sind die
Decylen-, Undecylen- und Dodecylengruppe. R4 bedeutet insbesondere eine geradkettige oder verzweigte C2-C5-Alkylengruppe, wobei die Ethylengruppe bevorzugt ist. Für R5 kommt insbesondere ein
Wasserstoffatom oder eine C1-C4-Alkylgruppe, vorzugsweise die Methylgruppe in Frage.
Unter der Definition der N+(R5)3-Gruppe sollen sowohl die identisch substituierten Stickstoffderivate, wie z.B. die Trimethylammoniumgruppe, als auch die gemischt substituierten Derivate, wie z.B. Dimethylammonium,- Diethylammonium-, n-Butyl-dimethylammonium oder Methyl-di-ethylammonium-gruppe verstanden werden bzw. alle durch die Kombination der in der Definition von R5 genannten
Gruppen möglichen Varianten.
Besonders bevorzugte Verbindungen der allgemeinen Formel II sind solche, in denen die Gruppe R1-X-R2- eine C9-C16-Alkylgruppe darstellt, wie beispielsweise die Decyl-, Undecyl-, Dodecyl-, Tridecyl-, Pentadecyl- und Hexadecylgruppe, und R' eine C9-C15-Alkyl- mercapto-, C9-C15-Alkoxy-, C9-C15-Alkoxycarbonyl-, C9-C15-Alkyl- aminocarbonyl- oder C9-C15-Alkylcarbonylaminogruppe, wie beispielsweise die Decylkercapto-, Undecylmercapto-, Decyloxy-, Undecyloxy-, Dodecyloxy-, Decyloxycarbonyl-, Decylaminocarbonyl- oder Decylcarbonylaminogruppe. Bevorzugt enthalten die Alkylteile in den zuvor genannten Gruppen 10-13 Kohlenstoffatome.
Die Verbindungen der Formel I können nach den in EP-B-69,968 beschriebenen Verfahren hergestellt werden.
Zur Herstellung der Verbindungen der Formel I und II eignet sich jedoch besonders vorteilhaft das folgende Verfahren:
Eine Verbindung der allgemeinen Formel III
R1-X-R2-Y-R3-ZH (III) in der
R1, R2, R3, X, Y und Z die oben angegebene Bedeutung haben, wird mit einer Verbindung der allgemeinen Formel IV
in der
R4 die oben genannte Bedeutung hat, in Gegenwart eines säure-bindenden Mittels umgesetzt und das Reaktionsprodukt direkt mit gegebenenfalls alkyliertem Ammoniak behandelt.
Verbindungen der Formel III können nach an sich bekannten Verfahren wie z.B. in Lipids 22, 947 (1987) beschrieben hergestellt werden. Verbindungen der Formel IV sind bekannte Verbindungen und im Handel erhältlich.
Das Verfahren wird in der Regel so durchgeführt, daß ein Alkanol bzw. Alkanthiol der allgemeinen Formel III mit einer Verbindung der allgemeinen Formel IV in Gegenwart eines säurebindenden
Mittels, wie z.B. Triethylamin in einem absoluten, inerten
organischen Lösungsmittel, wie z.B. chlorierter Kohlenwasserstoff oder Toluol, bei Temperaturen um den Gefrierpunkt bis Raumtemperatur reagiert, und das Reaktionsprodukt direkt mit gegebenenfalls alkyliertem Ammoniak behandelt wird. Dazu löst man den Ammoniak oder das Alkylamin in einem Medium, das sowohl den Phosphorsäurediester als auch Ammoniak oder das Amin genügend gut löst, wozu
sich besonders Mischungen aus Acetonitril oder niederen Alkoholen mit chlorierten Kohlenwasserstoffen eignen und vervollständigt die Reaktion bei einer Temperatur von 20 bis 70°C.
Man kann auch stufenweise vorgehen, also zuerst eine Alkyl-ammoniumgruppe einführen und mit Alkylhalogenid anschließend zum Dioder Trialkylammoniumalkylester umsetzen.
Sämtliche Zwischenstufen sowie Endprodukte lassen sich bequem säulenchromatographisch mit üblichen Elutionsmitteln, wie z.B. Ether, Ligroin, chlorierten Kohlenwasserstoffen, niederen Alkoholen oder Mischungen derselben, an Kieselgel reinigen, wobei im Falle des betainartigen Endprodukts zweckmässig etwas Wasser zugesetzt wird.
Die pharmakologisch verträglichen Salze erhält man in üblicher Weise, z.B. durch Neutralisation der Verbindungen der Formel I mit nichttoxischen anorganischen oder organischen Säuren, wie z.B.
Salz-, Schwefel-, Phosαhor-, Bromwasserstoff-, Essig-, Milch-, Zitronen-, Äpfel-, Salicyl-, Malon-, Malein- oder Bernsteinsäure.
Die Arzneimittel enthaltend Verbindungen der Formel I oder II zur Behandlung von viralen Infektionen können in flüssiger oder fester Form enteral oder parenteral appliziert werden. Hierbei kommen die üblichen Applikationsformen in Frage, wie beispielsweise Tabletten, Kapseln, Dragees . Sirupe, Lösungen oder Suspensionen. Als Injektionsmedium kommt vorzugsweise Wasser zur Anwendung, das die bei Injektionslösungen üblichen Zusätze wie Stabilisierungsmittel, Lösungsvermittler und Puffer enthält. Derartige Zusätze sind z.B. Tartrat- und Zitratpuffer, Ethanol, Komplexbildner, wie Ethylendiamintetraessigsäure und deren nichttoxischen Salze, hochmolekulare Polymere, wie flüssiges Polyethylenoxid zur Viskositätsregulierung. Flüssige Trägerstoffe für Injektionslösungen müssen steril sein und werden vorzugsweise in Ampullen abgefüllt. Feste Trägerstoffe sind beispielsweise Stärke, Lactose, Mannit, Methylcellulose, Talkum, hochdisperse Kieselsäuren, höher molekulare Fettsäuren, wie Stearinsäure, Gelatine, Agar-Agar, Calziumphosphat, Magnesiumstearat, tierische und pflanzliche Fette, feste
hochmolekulare Polymere, wie Polyethylenglykole, etc.. Für orale Applikationen geeignete Zubereitungen können gewünschtenfalls Geschmacks- oder Süßstoffe enthalten.
Die Dosierung kann von verschiedenen Faktoren, wie Applikationsweise, Spezies, Alter oder individuellem Zustand abhängen. Die erfindungsgemäßen Verbindungen werden üblicherweise in Mengen von 0,1 - 100 mg, vorzugsweise 0,2 - 80 mg pro Tag und pro kg Körpergewicht appliziert. Bevorzugt ist es, die Tagesdosis auf 2-5
Applikationen zu verteilen, wobei bei jeder Applikation 1-2 Tabletten mit einem Wirkstoffgehalt von 0,5 - 500 mg verabreicht werden. Die Tabletten können auch retardiert sein, wodurch sich die Anzahl der Applikationen pro Tag auf 1-3 vermindert. Der Wirkstoffgehalt der retardierten Tabletten kann 2 - 1000 mg betragen. Der Wirkstoff kann auch durch Dauerinfusion gegeben werden, wobei die Mengen von 5 - 1000 mg pro Tag normalerweise ausreichen.
Im Sinne der vorliegenden Erfindung kommen außer den in den Beispielen genannten Verbindungen die folgenden Verbindungen der Formel I in Frage:

2.2 g 3-Dodecyloxy-2-decyloxypropanol-1 in 15 ml Dichlormethan sowie 2.1 ml Triethylamin werden bei -25°C mit einer Lösung aus
1.3 g 2-Chlor-2-oxo-1,3,2-dioxaphospholan (Fluka) in 10 ml Dichlormethan versetzt und 30 Minuten gerührt. Dann läßt man auf Raumtemperatur erwärmen, entfernt das Lösungsmittel im
Rotationsverdampfer und suspendiert den Rückstand in Ether. Nach Abtrennen das Triethylammoniumsalzes wird das Filtrat erneut eingedampft und der ölige Rückstand in 30 ml Acetonitril
aufgenommen. Die klare Lösung wird durch Einleiten bei 20°C mit Trimethylamin gesättigt und 96 h bei RT im geschlossenen Gefäß gerührt. Danach wird der Niederschlag abgesaugt, mit Acetonitril gewaschen und aus Dichlormethan/Aceton umkristallisert.
Ausb. 1.27 g (41 % d.Th.), Schmp. 214-218°C (Zers.).
Das Ausgangsmaterial wurde analog zu LIPIDS 22, 947 (1987) und dort zitierter Literatur hergestellt. a) 5-Decyloxy-2-phenyl-1,3-dioxan
13 g (72 mmol) 5-Hydroxy-2-phenyl-1,3-dioxan in 50 ml abs. DMF wurden bei RT innerhalb 10 min zu einer Suspension aus 1.95 g (79 mmol) Natriumhydrid (97 %) in 35 ml abs. DMF getropft und nach vollständiger Zugabe 30 min bei Raumtemperatur gerührt. Dann wurden 17.5 g (79 mmol) 1-Decylbromid in 90 ml abs. Toluol auf einmal zugegeben und die Lösung 24 h bei Raumtemperatur
gerührt. Von der entstandenen Suspension wurde danach das
Lösungsmittel im Vakuum abgedampft, der Rückstand mit Wasser versetzt und mit Ether extrahiert. Die org. Phase wurde nochmals mit Wasser gewaschen, über Na2SO4 getrocknet und vom Lösungsmittel befreit. Der Rückstand wurde durch Flash-Säulenchromatographie mit Ether/Isohexan 1:5 als Eluens gereinigt. Gelbes öl, Ausbeute 9.7 g (42 %). b) 2-Decyloxy-1,3-propandiol
9.7 g (30.3 mmol) 5-Decyloxy-2-phenyl-1,3-dioxan wurden in 170 ml abs. Ethanol gelöst und nach Zugabe von 2 g Palladium auf Kohle (10 %) bei Raumtemperatur und Atmosphärendruck bis zur Aufnahme der theoretischen Wasserstoffmenge hydriert. Danach wurde der Katalysator abgetrennt und das Filtrat im Rotationsverdampfer eingeengt. Das zurückbleibende öl (Ausbeute fast quantitativ) kristallisierte beim Stehen durch. Schmp. 36-38°C. c) 3-Dodecyloxy-2-decyloxypropanol-1
5.4 g (23.3 mmol) 2-Decyloxy-1,3-propandiol in 17 ml abs. DMF wurden bei Raumtemperatur innerhalb von 15 min zu einer Suspension aus 617 mg (24 mmol) Natriumhydrid (97 %) in 15 ml abs. DMF getropft und bis zur Beendigung der Gasentwicklung nachgerührt. Dann wurden 5.98 g (24 mmol) 1-Dodecylbromid in 35 ml abs. Toluol zugegeben und die Lösung 24 h bei RT gerührt. Die entstandene Suspension wurde danach vom Lösungsmittel befreit, mit Wasser
versetzt und mehrfach mit Ether extrahiert. Die vereinigten organischen Phasen wurden über Na2SO4 getrocknet, eingedampft und der Rückstand durch Flash-Säulenchromatographie mit Ether/Isohexan als Eluens gereinigt. Ausbeute 4.6 g (49 % d.Th.) als gelbliches öl.
Beispiel 2
3-Decyloxy-2-dodecyloxypropyl-1-phosphorsäure-monocholinester wurde analog zu Bsp. 1 hergestellt.
Ausbeute 53 %.
Das als Edukt verwendete 3-Decyloxy-2-dodecyloxypropanol-1 wurde wie unter Bsp. 1 beschrieben hergestellt (farbloses öl, Ausb.
43 %).
Beispiel 3
Untersuchungen zur pharmakologischen Wirksamkeit
Die Verbindungen der vorliegenden Erfindung weisen eine antivirale Wirksamkeit auf und können somit beispielsweise zur Behandlung von HIV-bedingten Krankheiten oder Infektionen eingesetzt werden. De Nachweis der anti-HIV-Wirksamkeit erfolgt in Zeil- oder Gewebekulturen, wobei die direkte Hemmung der HIV-Replikation gemessen wird.
Zunächst erfolgt eine Behandlung von HIV-infizierten humanen T- Lymphomzellen bzw. HIV-infizierten humanen fetalen Lungenzellen mit der zu untersuchenden Verbindung in Verdünnungsreihe. Anschließund erfolgt eine Quantifizierung der Virusmenge im überstand durch Neuinfektion von humanen fetalen Lungenzellen. Die Abnahme der entstandenen HlV-positiven Zellkolonien gilt als direktes Maß für die antivirale Wirksamkeit der untersuchten
Verbindung.

3. Verwendung von Verbindungen der Formel I gemäß Anspruch 1 oder 2, wobei
X einen Valenzstrich, ein Sauerstoff- oder Schwefelatom, Y ein Sauerstoffatom, Z ein Sauerstoff- oder Sσhwefelatom, R1 ein Wasserstoffatom, ein geradkettiger oder verzweigter, gesättigter oder ungesättigter Alkylrest mit 1-18 Kohlenstoffatomen ist, R2 eine geradkettige oder verzweigte, gesättigte oder ungesättigte Alkylenkette mit 1-18 Kohlenstoffatomen bedeutet, R3 eine Alkylenkette mit 3 Kohlenstoffatomen ist, die ein- oder mehrfach durch C9-C15-Alkylmercapto-, C9-C15-Alkan- sulfinyl-, C9-C15-Alkansulfonyl oder durch C9-C15-Alkoxy substituiert ist, R4 eine Alkylenkette mit 2 Kohlenstoffatomen und R5 Wasserstoff, Methyl oder Ethyl bedeuten.
4. Verbindungen der Formel II +
R1-X-R2-Y-CH2-A-Z O-R4-N(R5)3 (II),
R1, R2, X, Y, Z, R4 und R5 die in den Ansprüchen 1-3 angegebenen Bedeutungen haben und
A die Gruppe -CH-CH2- oder -CH- darstellt, und
R1 eine C1-C20-Alkoxycarbonyl-, C1-C20-Alkylaminocarbonyl-, C1-C20-Alkylmercapto-, C1-C20-Alkansulfinyl-, C1-C20-Alkansulfonyl-, C1-C20-Alkylcarbonylamino- oder C1-C20-Alkoxy- gruppe bedeutet, sowie deren pharmakologisch unbedenkliche Salze und optische Isomere.
5. Verbindungen der Formel II gemäß Anspruch 4, dadurch
gekennzeichnet, daß die Gruppe R1-X-R2- einen C9-C16-Alkylrest bedeutet.
6. Verbindungen der Formel II gemäß Anspruch 4 oder 5, dadurch gekennzeichnet, daß R' eine C9-C15-Alkylmercapto-, C9-C15- Alkoxy-, C9-C15-Alkoxycarbonyl-, C9-C15-Alkylaminocarbonyl- oder C9-C15-Alkylcarbonylaminogruppe, wie beispielsweise die Decylmercapto-, Undecylmercapto-, Decyloxy-, Undecyloxy-, Dodecyloxy-, Decyloxycarbonyl-, Decylaminocarbonyl- oder
Decylcarbonylaminogruppe bedeutet.
7. Verbindungen gemäß Anspruch 4 oder 5, ausgewählt aus der
Gruppe der folgenden Verbindungen:
3-Dodecyloxy-2-decyloxypropyl-1-phosphorsäure-monocholinester
3-Decyloxy-2-dodecyloxypropyl-1-phosphorsäure-monocholinester
3-Dodecyloxy-2-dodeoyloxypropyl-1-phosphorsäure-monocholinester
3-Undecyloxy-2-undecylmercaptopropyl-1-phosphorsäure-monocholinester
3-Tridecyloxy-2-deoylmercaptopropyl-1-phosphorsäure-monocholinester
3-Dodecyloxy-2-decylmercaptopropyl-1-phosphorsäure-monocholinester
3-Dodecyloxy-2-decylsulfonylpropyl-1-phosphorsäure-monocholinester
3-Decyloxy-2-dodecylsulfonylpropyl-1-phosphorsäure-monocholinester
8. Arzneimittel, enthaltend mindestens eine Verbindung gemäß
einem der Ansprüche 4-7 neben üblichen Träger- und
Hilfsstoffen.
9. Verwendung von Verbindungen gemäß einem der Ansprüche 4-6 zur Herstellung von Arzneimitteln zur Behandlung oder Prophylaxe von viral- oder retroviral-bedingten Erkrankungen.
10. Verfahren zur Herstellung von Verbindungen der Formel II gemäß einem der Ansprüche 4-7, dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen Formel III
R1-X-R2-Y-CH2-A-ZH (III) in der R1, R2, X, Y, A und Z die in den Ansprüchen 4-7
angegebenen Bedeutungen haben, mit einer Verbindung der allgemeinen Formel IV
(IV)
in der R4 die in den Ansprüchen 4-7 angegebenen Bedeutungen hat, in Gegenwart eines säurebindenden Mittels umsetzt und das Reaktionsprodukt direkt mit gegebenenfalls alkyliertem
Ammoniak behandelt.
