Description
Die vorliegende Erfindung betrifft neue Mercaptoalkanolphospholipide
und deren Derivate, Verfahren zu ihrer Herstellung
und ihre Verwendung als Wirkstoffe in Arzneimitteln,
insbesondere zur Behandlung des Asthmas und von
Erkrankungen des Immunsystems. Die erfindungsgemäßen
Mercaptoalkanolphospholipide und deren Derivate entsprechen
der allgemeinen Formel I
worin:
R¹einen Alkyl- oder Alkyloxy-Rest mit 10-20 Kohlenstoffatomen,
R²ein Wasserstoffatom, einen Alkylrest mit 1-4
Kohlenstoffatomen,
R³, R⁴, R⁵gleich oder verschieden und unabhängig voneinander
ein Wasserstoffatom, einen Alkylrest
mit 1-4 Kohlenstoffatomen oder zusammen einen
heterocyclischen Rest mit 5 oder 6 Ringgliedern
mit Stickstoff, Sauerstoff, Schwefel als Heteroatome,
R⁶ein Wasserstoffatom, eine geradkettige oder
verzweigte Alkylgruppe mit 1-4 Kohlenstoffatomen
oder einen geradkettigen oder verzweigten
Acylrest mit 1-10 Kohlenstoffatomen,
meine ganze Zahl von 0-2,
neine ganze Zahl von 2-6
bedeuten.
Bevorzugt sind Mercaptoalkanolphospholipide und deren
Derivate der allgemeinen Formel I, worin
R¹einen Alkyl- oder Alkyloxy-Rest mit 14-20 Kohlenstoffatomen,
R²ein Wasserstoffatom,
R³, R⁴, R⁵einen Methylrest oder zusammen einen heterocyclischen
Rest mit 5 oder 6 Ringgliedern mit
Stickstoff, Schwefel als Heteroatomen,
R⁶ein Wasserstoffatom, eine geradkettige oder
verzweigte Alkylgruppe mit 1-4 Kohlenstoffatomen
oder einen geradkettigen oder verzweigten
Acylrest mit 1-4 Kohlenstoffatomen,
meine ganze Zahl von 0-2,
neine ganze Zahl von 2-6
bedeuten.
Besonders bevorzugt sind Mercaptoalkanolphospholipid-
Derivate der allgemeinen Formel I, worin
R¹einen Alkyl- oder Alkyloxy-Rest mit 14-18
Kohlenstoffatomen,
R²ein Wasserstoffatom,
R⁶Methyl oder Acetyl,
meine ganze Zahl von 0-2,

bedeuten.
Die neuen Mercaptoalkanolphospholipide und deren Derivate
der allgemeinen Formel I sind verwandt mit dem natürlich
vorkommenden Platelet-Activating-Factor (PAF), der ebenfalls
Phospholipidcharakter hat und folgende Struktur
besitzt:

PAF ist biologisch sehr stark wirksam, u. a. ein sehr starker Auslöser der Thrombocytenaggregation und wird als Mediator bei Entzündungsprozessen, insbesondere allergischen Entzündungen, und beim Asthma angesehen; vgl. C.P. Page, C.B. Archer, W. Paul and J. Morley, TIPS 1984, 239 (Elsevier).
PAF ist biologisch sehr stark wirksam, u. a. ein sehr starker Auslöser der Thrombocytenaggregation und wird als Mediator bei Entzündungsprozessen, insbesondere allergischen Entzündungen, und beim Asthma angesehen; vgl. C.P. Page, C.B. Archer, W. Paul and J. Morley, TIPS 1984, 239 (Elsevier).
Überraschenderweise zeigte sich, daß die erfindungsgemäßen
Mercaptoalkanolphospholipide und deren Derivate
diesbezüglich keine oder antagonistische Wirkungen aufweisen.
So hat beispielsweise (1-Acetylthiomethyl-heptadecyl)-
2-trimethylammonioethyl-phosphat aus Beispiel 1
bei einer Dosierung von 1×10-5 Mol/l im Versuch der
Substanz-induzierten Thrombocytenaggregation nach Born,
Nature 194, 927-929 (1962) praktisch keine Wirkung (5%
Induktion), während PAF bereits bei einer Dosierung von
1×10-6 Mol/l eine 100%ige Induktion der Thrombocytenaggregation
bewirkt. Andererseits aber hemmt (1-Acetylthiomethyl-
heptadecyl)-2-trimethylammonioethyl-phosphat
in einer Dosierung von 1 mg/kg Körpergewicht die PAF-
induzierte Bronchokonstriktion am Meerschweinchen um
77%.
Der Versuch wurde am narkotisierten, künstlich beatmeten
Meerschweinchen so ausgeführt, daß zuerst die zu prüfende
Verbindung mit einer Dosierung von 1 mg/kg Körpergewicht
und 5 min später rac-C₁₆-PAF in einer Dosierung von
0,316 mg/kg Körpergewicht intravenös injiziert wurde und
nachfolgend die Kontraktion der Bronchialmuskulatur durch
Messung des Beatmungsdruckes erfaßt wurde. Die Effekte
wurden 3 min nach Injektion registriert. Der angegebene
Hemmwert in % basiert auf einem Vergleich der PAF-Effekte
in der Testgruppe mit Wirkungen, ausgelöst in einer separaten
Kontrollgruppe, in der eine isotonische Kochsalzlösung
statt der zu prüfenden Verbindung verabreicht
wurde.
Es kann daher davon ausgegangen werden, daß mit den
erfindungsgemäßen Mercaptoalkanolphospholipiden und deren
Derivaten eine wirksame Therapie eines PAF-ausgelösten
Asthmas durchgeführt werden kann und folgerichtig auch
andere Immunantworten im Sinne von entzündlichen Prozessen
günstig beeinflußt werden können, wobei schädliche
Nebenwirkungen wie eine erhöhte Thrombocytenaggregation
nicht befürchtet werden müssen.
Verbindungen der allgemeinen Formel I werden hergestellt,
indem man Epoxide der allgemeinen Formel II
in der R¹ und R² die in der allgemeinen Formel I angegebenen
Bedeutungen besitzen, in organischen oder organisch-
wäßrigen Lösungsmittelsystemen, wie z. B. Toluol,
Tetrahydrofuran, Ethanol, Methanol oder deren Mischungen
mit Wasser, oder in Substanz mit Schwefelverbindungen der
allgemeinen Formel III
HS-R⁶ (III)
in der R⁶ die in der allgemeinen Formel I angegebene
Bedeutung besitzt, umsetzt und die entstandenen Verbindungen
der allgemeinen Formel IV
in der R¹, R², R⁶ die in der allgemeinen Formel I angegebenen
Bedeutungen haben, mit einem Dichlorphosphorsäure-
ω-halogenalkylester der allgemeinen Formel V
worin n die in der allgemeinen Formel I angegebene Bedeutung
hat und Hal ein Chlor- oder Bromatom ist, in einem
indifferenten organischen Lösungsmittel, wie z. B. Chloroform,
Dichlormethan, gegebenenfalls unter Anwendung einer
Hilfsbase, wie z. B. Pyridin, Triethylamin, phosphoryliert
und nachfolgend mit einem Amin der Formel VI
worin R³, R⁴, R⁵ die in der allgemeinen Formel I angegebenen
Bedeutungen besitzen, in einem indifferenten
Lösungsmittel, wie z. B. Toluol, Dichlormethan, Acetonitril,
gegebenenfalls unter Druck, umsetzt.

Die Sulfoxide und Sulfone der allgemeinen Formel I mit m
= 1 bzw. m = 2 und R⁶ = Alkyl werden aus den Thioethern
der Formel I mit m = 0 durch Reaktion mit sauerstoffabgebenden
Reagenzien hergestellt, so z. B. mit Wasserstoffperoxid
in Eisessig oder Aceton, mit Natriumperjodat/Eisessig,
Kaliumpermanganat/Mineralsäure oder organischen
Persäuren, wie z. B. m-Chlorperbenzoesäure, in Chloroform,
Dichlormethan oder ähnlichen inerten Lösungsmitteln.
Die vorliegende Erfindung betrifft ebenfalls pharmazeutische
Präparate, welche die Mercaptoalkanolphospholipide
und deren Derivate der allgemeinen Formel I enthalten.
Bei den erfindungsgemäßen pharmazeutischen Präparaten
handelt es sich um solche zur enteralen wie oralen oder
rectalen sowie parenteralen Verabreichung, welche die
pharmazeutischen Wirkstoffe allein oder zusammen mit
einem üblichen, pharmazeutisch anwendbaren Trägermaterial
enthalten. Vorteilhafterweise liegt die pharmazeutische
Zubereitung des Wirkstoffes in Form von Einzeldosen vor,
die auf die gewünschte Verabreichung abgestimmt sind, wie
z. B. Tabletten, Drag´es, Kapseln, Suppositorien, Granulate,
Lösungen, Emulsionen oder Suspensionen. Die Dosierung
der Verbindungen liegt üblicherweise zwischen 1-1000 mg
pro Dosis, vorzugsweise zwischen 10-500 mg je Dosis, und
kann ein- oder mehrmals, bevorzugt zwei- bis dreimal täglich,
verabreicht werden.
Die Herstellung der erfindungsgemäßen Verbindungen wird
durch die folgenden Beispiele näher erläutert. Die angegebenen
Schmelzpunkte wurden mit einem Büchi-Schmelzpunktbestimmungsapparat
gemessen und sind nicht korrigiert.
Die IR-Spektren wurden mit dem Gerät Nicolet NIC-
3600 aufgenommen.
134 g 1,2-Epoxyoctadecan und 190 g Thioessigsäure
werden gemischt und die Mischung 8 Stunden auf 60°C
erwärmt. Die überschüssige Thioessigsäure wird im
Vakuum abgedampft und der Rückstand aus n-Hexan in
der Kälte kristallisiert.
Ausbeute: 63,3 g mit Schmp. 61-62°

69 g 2-Bromethylphosphorsäuredichlorid werden in
300 ml absol. Chloroform gelöst und 60 g Pyridin hinzugefügt.
In die Mischung werden unter Eiskühlung
67,1 g S-(2-Hydroxyoctadecyl)methanthioat, gelöst in
200 ml absol. Chloroform, eingetropft und die

46,2 g (1-Acetylthiomethyl-heptadecyl)-2-bromethyl-
phosphat werden in 100 ml absol. Toluol gelöst,
100 ml 20%ige Trimethylaminlösung in Toluol hinzugefügt
und die Mischung 4 Stunden auf 60°C im Druckgefäß
erwärmt. Die Reaktionsmischung wird im Vakuum eingeengt
und der Rückstand unter Säulenchromatographie
(Kieselgel//CHCl₃/Methanol/Wasser) gereinigt.
Ausbeute: 29 g mit Schmp. 215-218°C

85 g Hexadecyl-2,3-epoxypropylether und 108 g Thioessigsäure
werden gemischt und die Mischung 8 Stunden
auf 60°C erwärmt. Die überschüssige Thioessigsäure
wird im Vakuum abgedampft und der Rückstand durch
Säulenchromatographie (Kieselgel//Hexan/Essigsäureethylester)
gereinigt.

60,2 g 2-Bromethylphosphorsäuredichlorid werden in
300 ml absol. Chloroform gelöst und 52 g Pyridin hinzugefügt.
In die Mischung werden unter Eiskühlung
59,7 g S-(3-Hexadecyloxy-2-hydroxy-propyl)methanthioat,
gelöst in 200 ml absol. Chloroform, eingetropft
und die Mischung 4 Stunden unter DC-Kontrolle
bei Raumtemperatur gerührt. Nach Zugabe von Eiswasser
wird noch 1 Stunde nachgerührt, die Chloroformphase
abgetrennt und mit viel Wasser gewaschen. Die Chloroformphase
wird mit Natriumsulfat getrocknet, eingeengt
und der Rückstand durch Säulenchromatographie
(Kieselgel//Chloroform/Methanol) gereinigt.

28,8 g (1-Acetylthiomethyl-2-hexadecyloxy-ethyl)-2-
bromethyl-phosphat werden in 50 ml absol. Toluol
gelöst, 50 ml 20%ige Trimethylaminlösung in Toluol
hinzugefügt und die Mischung 4 Stunden mit 60°C im
Druckgefäß erwärmt. Die Reaktionsmischung wird im
Vakuum eingeengt und der Rückstand durch Säulenchromatographie
(Kieselgel//CHCl₃/Methanol/Wasser)
gereinigt.

40,3 g 1,2-Epoxyoctadecan und 200 ml 30%ige Methylmercaptanlösung
in Methanol werden gemischt und die
Mischung im Druckgefäß 8 Stunden auf 120°C erwärmt.
Die Mischung wird im Vakuum eingeengt und der Rückstand
durch Säulenchromatographie (Kieselgel//Hexan/
Essigsäureethylester) gereinigt.

25 g 2-Bromethylphosphorsäuredichlorid werden in
150 ml absol. Chloroform gelöst und 21,8 g Pyridin
hinzugefügt. In die Mischung werden unter Eiskühlung
22 g 2-Hydroxyoctadecyl-methylthioether, gelöst in
50 ml absol. Chloroform, eingetropft und die Mischung
3 Stunden unter DC-Kontrolle bei Raumtemperatur
gerührt. Nach Zugabe von Eiswasser wird noch 1 Stunde
nachgerührt, die Chloroformphase abgetrennt und mit
viel Wasser gewaschen. Die Chloroformphase wird mit
Natriumsulfat getrocknet und der Rückstand durch Säulenchromatographie
(Kieselgel//Chloroform/Methanol)
gereinigt.

0,95 g (1-Methylthiomethyl-heptadecyl)-2-bromethyl-phosphat
werden in 10 ml Pyridin gelöst und die
Lösung 20 Stunden bei 80-90°C gerührt. Das überschüssige
Pyridin wird im Vakuum abgezogen und der Rückstand
durch Säulenchromatographie (Kieselgel//
Dichlormethan/Methanol/Wasser) gereinigt.

0,6 g (1-Methylthiomethyl-heptadecyl)-2-pyridinioethyl-
phosphat werden in 20 ml Chloroform gelöst, 0,2 g 3-
Chlorperbenzoesäure hinzugefügt und die Mischung 2 Stunden
bei Raumtemperatur gerührt. Die Mischung wird im
Vakuum eingeengt und der Rückstand durch Säulenchromatographie
(Kieselgel//Dichlormethan/Methanol/Wasser)
gereinigt.
Ausbeute: 0,3 g mit Schmp. 203-204°C

47 g Hexadecyl-2,3-epoxypropylether und 250 ml
30%ige Methylmercaptanlösung in Methanol werden
gemischt und die Mischung 8 Stunden im Druckgefäß auf
120°C erwärmt. Die Mischung wird im Vakuum eingeengt
und der Rückstand durch Säulenchromatographie (Kieselgel//
Hexan/Essigsäureethylester) gereinigt.
Ausbeute: 24 g mit Schmp. 58-59°C

16,7 g 2-Bromethylphosphorsäuredichlorid werden in
100 ml absol. Chloroform gelöst und 14,6 g Pyridin
hinzugefügt. In die Mischung werden unter Eiskühlung
16 g 3-Hexadecyloxy-2-hydroxypropyl)methylthioether,
gelöst in wenig Chloroform, eingetropft und die
Mischung 3 Stunden unter DC-Kontrolle bei Raumtemperatur
gerührt. Nach Zugabe von Eiswasser wird noch
1 Stunde nachgerührt, die Chloroformphase abgetrennt
und mit viel Wasser gewaschen. Die Chloroformphase
wird mit Natriumsulfat getrocknet, eingeengt und der
Rückstand durch Säulenchromatographie (Kieselgel//
Chloroform/Methanol) gereinigt.
Ausbeute: 5,6 g Öl

2,6 g (1-Methylthiomethyl-2-hexadecyloxy-ethyl)-2-
bromethyl-phosphat werden in 10 ml Pyridin gelöst und
die Lösung 20 Stunden bei 80-90°C gerührt. Das überschüssige
Pyridin wird im Vakuum abgezogen und der
Rückstand durch Säulenchromatographie (Kieselgel//
Dichlormethan/Methanol/Wasser) gereinigt.
Ausbeute: 1,9 g Wachs

2,6 g (1-Methylthiomethyl-2-hexadecyloxy-ethyl)-2-bromethyl-
phosphat werden mit 15 ml 25%iger Trimethylaminlösung
in Toluol gemischt und die Mischung 4 Stunden auf
60°C im Druckgefäß erwärmt. Die Reaktionsmischung wird im
Vakuum eingeengt und der Rückstand durch Säulenchromatographie
(Kieselgel//CHCl₃/Methanol/Wasser) gereinigt.
Ausbeute: 2,6 g Wachs

1,8 g (1-Methylthiomethyl-2-hexadecyloxy-ethyl)-2-pyridinioethyl-
phosphat werden in 20 ml Chloroform gelöst,
0,58 g 3-Chlorperbenzoesäure hinzugefügt und die Mischung
2 Stunden bei Raumtemperatur gerührt. Die Mischung wird
im Vakuum eingeengt und der Rückstand durch Säulenchromatographie
(Kieselgel//Dichlormethan/Methanol/Wasser)
gereinigt.
Ausbeute: 0,5 g mit Schmp. 208-109°C

2,5 g (1-Methylthiomethyl-2-hexadecyloxy-ethyl)-2-bromethyl-
phosphat werden in 20 ml Chloroform gelöst, 0,86 g
3-Chlorperbenzoesäure hinzugefügt und die Mischung
2 Stunden bei Raumtemperatur gerührt. Die Mischung wird
im Vakuum eingeengt und der Rückstand durch Säulenchromatographie
(Kieselgel//Dichlormethan/Methanol/Wasser)
gereinigt.
Ausbeute: 0,8 g mit Schmp. 227-229°C

Claims (6)
1. Mercaptoalkanolphospholipide und deren Derivate der
allgemeinen Formel I
worin:R¹einen Alkyl- oder Alkyloxy-Rest mit 10-20 Kohlenstoffatomen,
R²ein Wasserstoffatom, einen Alkylrest mit 1-4
Kohlenstoffatomen,
R³, R⁴, R⁵gleich oder verschieden und unabhängig voneinander
ein Wasserstoffatom, einen Alkylrest
mit 1-4 Kohlenstoffatomen oder zusammen einen
heterocyclischen Rest mit 5 oder 6 Ringgliedern
mit Stickstoff, Sauerstoff, Schwefel als Heteroatomen,
R⁶ein Wasserstoffatom, eine geradkettige oder
verzweigte Alkylgruppe mit 1-4 Kohlenstoffatomen
oder einen geradkettigen oder verzweigten
Acylrest mit 1-10 Kohlenstoffatomen,
meine ganze Zahl von 0-2,
neine ganze Zahl von 2-6bedeuten.
2. Mercaptoalkanolphospholipid-Derivate der allgemeinen
Formel I,
worin R¹einen Alkyl- oder Alkyloxy-Rest mit 14-20 Kohlenstoffatomen, R²ein Wasserstoffatom, R³, R⁴, R⁵einen Methylrest oder zusammen einen heterocyclischen Rest mit 5 oder 6 Ringgliedern mit Stickstoff, Schwefel als Heteroatomen, R⁶ein Wasserstoffatom, eine geradkettige oder verzweigte Alkylgruppe mit 1-4 Kohlenstoffatomen oder einen geradkettigen oder verzweigten Acylrest mit 1-4 Kohlenstoffatomen, meine ganze Zahl von 0-2, neine ganze Zahl von 2-6bedeuten.

3. Mercaptoalkanolphospholipid-Derivate der allgemeinen
Formel I, worin
R¹einen Alkyl- oder Alkyloxy-Rest mit 14-18
Kohlenstoffatomen,
R²ein Wasserstoffatom,
R⁶Methyl oder Acetyl,
meine ganze Zahl von 0-2,
n2bedeuten.
4. Verfahren zur Herstellung von Verbindungen der allgemeinen
Formel I gemäß den Ansprüchen 1, 2 und 3,
dadurch gekennzeichnet, daß man Epoxide der allgemeinen
Formel II
in der R¹ und R² die in der allgemeinen Formel I
angegebenen Bedeutungen besitzen, in organischen oder
organisch-wäßrigen Lösungsmittelsystemen oder in
Substanz mit Schwefelverbindungen der allgemeinen
Formel IIIHS-R⁶ (III)in der R⁶ die in Formel I angegebene Bedeutung
besitzt, umsetzt und die entstandenen Verbindungen
der Formel IV
in der R¹, R², R⁶ die in Formel I angegebenen Bedeutungen
haben, mit einem Dichlorphosphorsäure-ω-halogenalkylester
der Formel V
worin n die in der allgemeinen Formel I angegebene
Bedeutung hat und Hal ein Chlor- oder Bromatom ist,
in einem indifferenten organischen Lösungsmittel,
gegebenenfalls unter Anwendung einer Hilfsbase
phosphoryliert und nachfolgend mit einem Amin der
Formel VI
worin R³, R⁴, R⁵ die in der allgemeinen Formel I
angegebenen Bedeutungen besitzen, in einem indifferenten
Lösungsmittel, gegebenenfalls unter Druck,
umsetzt.
5. Verfahren zur Herstellung von Verbindungen der allgemeinen
Formel I mit R⁶ = Alkyl und m = 1 oder 2 gemäß
den Ansprüchen 1, 2 und 3, dadurch gekennzeichnet,
daß man Verbindungen der allgemeinen Formel I mit R⁶ =
Alkyl und m = 0 mit sauerstoffabgebenden Reagenzien,
bevorzugt Wasserstoffperoxid und organischen
Persäuren, in inerten Lösungsmitteln umsetzt.
6. Pharmazeutische Präparate, dadurch gekennzeichnet,
daß sie eine Verbindung der allgemeinen Formel I
gemäß den Ansprüchen 1, 2 und 3 als Wirkstoff im
Gemisch mit üblichen pharmazeutischen Hilfs- und
Trägerstoffen enthalten.
