本発明は抗腫瘍作用を有する ビスージォキソピペラジン誘導体又 はその薬学的に許容される塩に関する。

ビス—ジォキソ ピペラジン誘導体は現在まで種々報告されており、 抗腫瘍作用を有する化合物として 1 , 2-ビス(4- モルホリ ノメチル - 3 , 5-ジォキソ ピペラジン -卜ィル) ェタ ン [第 8回国際薬理学会議 予稿集、 441頁、 1981年] 、 -1 , 2-ビス(4- モルホリ ノ メ チル - 3 , 5-ジォキソ ピペラジン -1-ィル) プロパン [特開昭 59-190976号 公報] 及び 1 , 2-ビス(4-i so- ブ トキシカルボニルォキシメ チル -3 , 5-ジォキソピペラジン -1-ィル) ェタ ン [第 14回国際化学療法会議 予稿集、 324頁、 1985年及び特開昭 60-97963号公報] が知られてい o
しかし、 これらの化合物より も更に強い抗腫瘍作用を有する ビス —ジォキソピペラジン誘導体が望まれていた。
発 明 の 開 示
本発明者らはかかる状況において新規なビス一ジォキソ ピペラジ ン誘導体の研究を行なつた結果、 後記式①で示される化合物が著し く優れた抗腫瘍作用を有することを知見して本発明を完成した。

「低級」 とは特に限定がなければ炭素数 1 〜 6個を有する基を意 味する。
「低級アルキル基」 と しては、 例えばメ チル、 ェチル、 n-プロ ピ ル、 iso-プロ ピノレ、 n-ブチノレ、 iso-ブチル、 tert— プチル、 n-ペン チル、 n-へキシル等の直鑌又は分枝鎖状のアルキル基が挙げられ、 R 1 と しては特にメ チルが好ま しい。
「ァ ミ ノ基の保護基」 と しては、 例えばァセチル、 tert- ブ トキ シ力ルボニル、 ベンジルォキシカルボニル等が挙げられる。
又、 フヱニル基、 スチリ ル基、 ベンジル基又はフヱノキシ基等の ベンゼン環、 あるいはフ リ ル基が置換基を有する場合、 この置換基 と しては例えばフ ッ素、 塩素、 臭素等のハロゲン原子、 メ チル、 .ェ チル等の低級アルキル基、 メ トキシ、 エ トキン等の低級アルコキシ 基、 ニ ト 口基、 ァ ミ ノ基又はシァノ基が挙げられ、 1 ないし 2置換 してもよい。

反応温度は 0 〜50 Cの範囲であり、 反応時間は反応温度により変 化するが 4 〜 24時間の範囲である。
(B) 化合物 (1の反応性誘導体を用いる場合,、 反応性誘導体としては 酸ハロゲン化物、 酸無水物又はハロゲン化ギ酸エステルが好ま しい, 反応温度は - 20 °C〜室温の範囲であり、 反応時間は反応温度によ り変化するが 1 〜 8時間の範囲である。
上記 (A)法及び ( 法のいずれの場合も、 化合物 0D 1 モルに封して化 合物!)又はその反応性誘導体をそれぞれ 0 . 8〜 5 モル使用し、 反応 溶媒としては DMF 、 ピリ ジン、 ジクロロメ タン、 クロ口ホルム、 ァ セ 卜ニ 卜 リル等の非プロ ト ン性極性溶媒を単独又は混合して使用で きる。
なお、 上記製法においては、 化合物 に対する化合物 D又はその 反応性誘導体の使用量によつて生成比が変化するが、 いずれの場合

本発明の化合物①を製造するに際し、 必要に応じて化合物!)の官 能基を常法により保護する。
又、 本発明の化合物 0)の酸付加塩、 例えば塩酸塩、 シユウ酸塩、 P-卜ルエルスルホン酸塩、 酢酸塩、 ト リ フルォロ酢酸塩等は常法に
より製造することができる。
なお、 前記製造方法の出発原料である化合物 は英国特許第
1234935号公報記載の公知化合物である。

I ) 試験管内における P388白血病腫瘍細胞の増殖阻害効果
CDFi 雌マウスに P388白血病腫瘍細胞 1X 106 個を移植し、 移植 後 5 日目のマウス腹水細胞をキヤ ビラリ一で無菌的に採取して実験 腫瘍細胞と した。 上記実験腫瘍細胞は RPMI1640培地に牛胎児血清 (10%) 、 カナマイ シン(0. Img/ 及び 2-ヒ ドロキシェチルジス ルフィ ド (O.OlmM) を添加した培養液に懸濁して、 細胞数を Q.5 H 当り 5 X 104 個に調整した。 又試料は上記培養液に溶解又は懸蜀し て試料濃度を 1 X 10—1 〜 1 X 10 πιΜに調整した。
試験管にこの細胞懸濁液 と試料溶液 Q.5 を入れ、 モル 卜 ン栓をゆるめにしめて 5 %炭酸ガスィ ンキュベ一夕一中、 37てで 48 時間培養した。 培養後、 試験管に 4 SJ?の 0.25% ト リ プシン溶液を加
え、 37 Cで 5分間捩盪したのちコール夕一カウンターを用いて細胞 数を測定し、 増殖阻害度を下記式により求めた。

そして、 種々の試料濃度での増殖阻害度から 50%増殖阻害濃度 (IC50 ) を算出した。
その結果を下記第 1表に示す。 第 1 表
すなわち、 本発明の化合物は比較化合物 Aより明らかに活性が強 く 、 大幅に少ない投与量で有効性を発揮し得ることから、 投与時の 患者の負担を軽減し得ると共に全身的な副作用の防止につながる高 濃度の局所用剤 (マイ クロカプセル剤、 注射剤等) への応用が可能 になった。
Π ) 試験管内における V 79細胞のコロニ一形成阻止効果
プラスティ ッ クディ ッ シュ Φ 60mm) 中で継代増殖させたチヤィ ニーズハムスター肺線維芽細胞腫由来の V79細胞に ト リ プシンー
EDTA溶液(GIBC0社製) 1 a?を加え、 5 %炭酸ガスイ ンキュベータ一 中 37てで 5分間処理し、 均一な浮遊細胞液とした。 そ.の後 RPMI1640 培地に牛胎児血清 (10%) 及びカナマイ シ ン ( O.imgZK?) を添加 した培養液を加えて稀釈し、 生細胞数が 1 当り 100個、 200個、 400個の浮遊細胞液を調製した。 別のプラスチッ クディ ッ シュ ( ø 60min) に上記培養液各し 85¾?を加え、 更に先に調製した浮遊細胞液 各 1 を加えて生細胞数が 1 ディ ッ シュ当り 100個、 200個、 400 個の試験用ディ ッ シュと した。 また、 試料は 20mMとなるよう DMS0に 溶解し、 上記培養液で稀釈して 2 Χ 10·3 〜2 X 10_1D oiMの濃度に調整 した。
試験用ディ ッ シュを 5 %炭酸ガスィ ンキュベータ一中 37 Cで 24時 間培養し、 更に調製済みの試料液 150 J? を加えて 96時間培養した。 その後ディ ッ シュより培養液を静かに除去し、 マグネシウム及び力
ルシゥムを含まないリ ン酸緩衝生理食塩水 [P B S (—) ] でディ ッ シュを洗浄した。 次いで 3.5%ホルマリ ン P B S (-) 溶液 1 を加え、 2時間以上静置してコロニ一を固定した後ク リスタルバイ ォレツ トのエタノール溶液で染色した。
肉眼的に判定可能なコロニー数を数え、 対照群のコ π二一数との 比を求めてコロニー形成を 90%阻止する試料濃度 ICaを算出した。
その結果を下記第 2表に示す。 第 2 表
Π) P388白血病腫瘍移植マウスに対する延命効果
対照群一群^)匹、 投与群一群 7匹の CDFi 雄マウス ( 6週齢、 体 重 25± 2 g)の腹腔内に 1.0 X 106 個の P388白血病腫瘍細胞をそれぞ れ移植し、 翌日と 5 日目の 2回試料を所定量腹腔内投与することに より行なった。 試料の効果は延命率(Rate of Increase in Life
Span) と して下記式より算出した

至適投与量 本発明の化合物は P388白血病腫瘍細胞移植マウスに対して優れた 延命効果を示したことから、 強い抗腫瘍作用を有することが明らか であり、 動物又は人間の抗腫瘍剤と して有用であることが示された 又、 本発明の化合物は L1210白血病腫瘍、 B-16悪性黒色腫、 ルイ
ス肺癌、 MM-46 乳癌、 MH-134肝癌、 ェ一リ ッ ヒカルシノ一マ
(Ehrlich carcinoma) 等を用いた抗腫瘍試験において幅広い抗腫瘍
スペク トルを有することが示された。 ' 次に、 本発明の化合物の急性毒性は下記試験により確認した。
急性毒性試験は一群 5匹の ddY系雄マウス ( 6週齢、 体重 30± 2
g)に 1 %ヒ ドロキシプロピルセルロース(HPC) を添加した生理食塩
水にて調製した試料を腹腔内投与し、 14日間観察して LD50 を求めた。
その結果、 エリス口 -2- C4-(3- トルオイルォキシメチル)-3, 5-ジ
ォキソピペラジン -1-ィル〕 -3-(3,5-ジォキソ ピペラジン -1-ィル) ブタ ンの LD50 は 6.7〜10.0nig/kg であった。
次に、 本発明のビス一ジォキソピペラジン誘導体を人体へ適用す- る場合の投与方法、 剤型、 投与量について説明する。
本発明の化合物は経口的又は非経口的に投与可能であり、 経口投
与の剤型としては錠剤、 コ一ティ ング錠剤、 散剤、 顆粒剤、 カプセ
ル剤、 マイ ク ロカプセル剤、 シロップ剤などが、 又非経口投与の剤
型と しては注射剤 (用時溶解して用いる注射用凍結乾燥剤を含む) 、 坐剤などが使用できる。 これらの剤型の調製は薬学的に許容される
賦形剤、 結合剤、 滑沢剤、 崩壊剤、 懸濁化剤、 乳化剤、 防腐剤、 安
定化剤及び分散剤、 例えば乳糖、 白糖、 でんぶん、 デキス ト リ ン、
結晶セルロース、 カオリ ン、 炭酸カルシウム、 タルク、 ステア リ ン - 酸マグネシウム、 蒸溜水又は生理食塩水を用いて行なわれる。 . 投与量は患者の症状、 年齢、 体重などに応じて異なるが、 成人に
対する 1 日量と して 1 〜 600oig、 好ま しく は 5 〜 lOOmgを 1 〜 3回
に分けて投与することができる。

実施例 6 メ ソ -2, 3-ビス 〔4- (3- トルオイルォキシメ チル〉-3,5- ジォキソ ピペラジン -1-ィノレ〕 プタ ン及びエリ ス口- 2 - C4-(3- トルオイルォキシメ チル) -3, 5-ジォキソ ピペラ ジン -1 -ィル〕 -3- (3, 5-ジォキソ ピペラジン -1-ィル) プ夕 ン
メ ソ -2, 3-ビス(3,5- ジォキソ ピペラ ジン -1-ィル) ブタ ン 200 Dig(0.7mniol) に DMF 41?を加え 110。Cで 10分間加熱し、 次いでホル ムアルデヒ ドの 37%水溶液 0.2 ^を加え、 更に 140°Cで 1.5B 間加 熱攪拌した。 反応混合物より減圧下溶媒を留去し、 残留物に DMF 20 ^及びピリ ジン 1 を加えて一 10°Cに冷却した混合物に 3-トルオイ ルクロ リ ド 150ing (i.Ommol) を徐々に加え— 10。Cで 5時間攪拌した 後、 反応混合物より減圧下溶媒を留去し、 残留物をク ロ口ホルムで 抽出した。 抽出液を希塩酸、 飽和炭酸水素ナ ト リ ゥム水溶液、 水の 順で洗浄した後、 無水硫酸マグネシウムで脱水した。 抽出液より減 圧下溶媒を留去して得られた残留物をシリカゲルカラムクロマ トグ ラフィ 一 (酢酸ェチル : n-へキサン = 1:1 ) にて分離、 精製し標記
化合物を得た。

以上述べた如く 、 本発明の化合物①は従来知られている ビス一ジ
ォキソピペラジン誘導体とは異なる構造を有する新規化合物であり、 幅広い抗腫瘍スぺク トルを有すると共に公知の抗腫瘍性ビス一ジォ
キソ ピペラジン (比較化合物 A ) より著しく強い抗腫瘍作用を有す
ることから、 抗腫瘍剤として極めて有用である。
