哈特維希-布赫瓦爾德(Hartwig-Buchwald)反應隨著時間的推移已經進入工業製造過程,特別地對於製作有機發光二極體中之中間體。為此目的,EP-A1-2407502描述了樹枝狀聚合物的製備,藉由哈特維希-布赫瓦爾德反應獲得了二級胺中間體。EP-A2-2421064同樣使用哈特維希-布赫瓦爾德反應作為二級胺的一合成路徑,該等二級胺用作中間體。
中等產率係一缺點,但是代表更大問題的是缺乏選擇性,不僅產生二級胺而且還產生三級胺副產物。該等副產物難以藉由昇華去除。此外,鑒於不希望的雜質,甚至結構相關之雜質,並且甚至處於小量值,可能導致不希望的發射波長變化或量子產率下降,並且因此還導致不希望的發熱,在此一特別缺點係,加於中間體(用於製備在製作

本專利申請之目的係提供一種方法,該方法允許在哈特維希-布赫瓦爾德偶合的條件下以相對高產率和選擇性製備二聯苯胺,從而允許與純化該等化合物(通常藉由昇華完成)相關聯之成本以及複雜程度之降低。根據以上說明,那麼,有必要主要藉由增加選擇性來實現增加的產率。

該聯苯基單元可以是未取代的或取代的,包括被一個或多個苯基取代,因此形成,例如,三聯苯、四聯苯或苯並菲單元,它們可以是取代的或未取代的。該聯苯基單元還可以是橋聯的,如像在茀以及它的衍生物情況下。

相對於該一級芳香胺A-NH2和該鹵代芳香族化合物X-B這兩種反應物之總莫耳量,該鹼通常可以40%至80%、更具體地說50%至70%或55%至65%的量使用。這意味著如果,例如,該一級芳香胺A-NH2和該鹵代芳香族化合物X-B各使用20mmol,因此總計40mmol,則該鹼可以16mmol至32mmol,更具體地說20mmol至28mmol或從22mmol至26mmol之量使用。
替代於這個程序,也可以將該一級芳香胺A-NH2、該鹵代芳香族化合物X-B以及該鹼引入,同時添加一溶劑以及該催化劑溶液(以適當量計量的)。

確保在哈特維希-布赫瓦爾德偶合的條件下它們係惰性的,因為胺基或鹵素基團可能破壞所希望之反應。
對於沒有作為一次級反應(形成三級胺)發生的雙芳基化之轉化,所需要的催化劑量係在0.01mol%至1.5mol%之間、或從0.1mol%至1mol%、或從0.3mol%至0.8mol%。以mol%計的數字係基於以莫耳計的該一級芳香胺A-NH2和該鹵代芳香族化合物X-B兩種反應物的總量。
該等反應物之轉化率係至少90%,更具體地說至少95%或至少97%。經常作為一次級反應(形成三級胺)發生的雙芳基化在此幾乎完全被抑制。
反應溫度通常是5℃至150℃、或20℃至140℃、或30℃至130℃,更具體地說60℃至120℃或70℃至111℃。反應時間通常是1小時至36小時、或4小時至24小時、或6小時至16小時或8小時至12小時。

因此,本專利申請還涉及化合物1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-馬來醯亞胺[Pd(dippf)(馬來醯亞胺)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-1,3-二乙烯基-1,1,3,3-四甲基二矽氧烷[Pd(dippf)(VTS)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-雙(二苯亞甲基丙酮)[Pd(dippf)(dba)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-萘醌[Pd(dippf)(萘醌)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-馬來酸酐[Pd(dippf)(馬來酸酐)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-馬來酸二乙酯[Pd(dippf)(馬來酸二乙酯)]、以及1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-降冰片烯[Pd(dippf)(降冰片烯)]。
該鈀原子(處於氧化態0的鈀)常常被三配位並且該等錯合物通常是三角平面的,意味著包含多於一個雙鍵的化合物如1,3-二乙烯基-1,1,3,3-四甲基二矽氧烷(VTS或VS)或雙(二苯亞甲基丙酮)僅僅用它們的雙鍵之一來錯合該鈀原子。第二個雙鍵或者什麼也不錯合或著錯合一個另外的鈀原子。

將1,1’-雙(二異丙基膦基)二茂鐵(dippf)(213mg、0.50mmol)溶解在二乙醚(5mL)中,並且逐滴添加在2,4,6,8-四甲基環四矽氧烷中的1,3-二乙烯基-1,1,3,3-四甲基二矽氧烷鈀(0)(Pd-VTS)(0.50mL、0.50mmol Pd)。將該橙色混合物在室溫下攪拌1小時。
該程序最初與實例1中的一樣。然後,添加在二乙醚(5mL)中的馬來醯亞胺(99.1mg、1.00mmol),並且在一超聲浴中處理該混合物十分鐘,此時形成一黃色沈澱。在該固體已經沈降之後,去除上清液並且使用二乙醚(3×5

在圖1中示出了該反應方程序。沒有在任何實例中觀察到該一級胺131被該芳基溴132多重芳基化。
在20ml的用於氣體層析法的頂空小瓶(headspace vial)中進行所有的試驗,該等小瓶用卷邊的、特徵為聚四氟乙烯塗覆的丁基橡膠隔膜的鋁蓋(兩者都可從,例如,VWR公司獲得)密封。為了加熱該等容器,使用8cm高的圓柱形的鋁塊,在它們的直徑上精確地與實驗室磁力攪拌器(例如Heidolph Mr 2002)之加熱板相配。該等鋁塊配備有十個7cm深並且與該等反應器相同直徑的孔,以及一用來容納溫度感測器之孔。
製成用於連接到施倫克(Schlenk)線之真空分配器用於10個容器每次的同時抽真空和再充填。為此目的,將十個真空相容的3mm聚四氟乙烯管各自在一端用適配器連接到調節鎖緊套口注射針(accommodate Luer-Lock syringe needle)上,並且在另一端連接到一鋼管上,該鋼管可以經由一真空軟管連接到該施倫克線上。

隨後依次同時將該等反應器抽空和裝入氮氣三次。當已經以此方式給予該等反應器一惰性氣氛時,藉由一超壓閥在真空線上產生與外部氣氛之壓力補償。使用一注射器,通過該等隔膜蓋注入在甲苯(2mL)中的[Pd(dippf)(馬來醯亞胺)](1.24mg、0.002mmol)和dippf(0.85mg、0.002mmol)的一種儲備溶液。然後,使該鋁塊達到70℃並且去除該真空分配器的針。
在20小時的反應時間之後,冷卻該等容器並且小心地打開,並且使用二氯甲烷(30ml)和水(30ml)稀釋該反應介質。使用1N的鹽酸將該水相調節至7的pH,從該有機相中分離出來並且使用二氯甲烷(2×20mL)萃取。結合的有機相經過硫酸鎂乾燥、過濾並且最終藉由薄層層析法進行分析。

Claims (16)
- 一種使用具有式X-B之芳香族化合物選擇性芳基化具有式A-NH2的一級芳香胺以產生二級芳香胺A-NH-B之方法,基團A和B彼此獨立地是相同或不同的、取代或未取代的芳族基團並且基團X係一鹵素或三氟甲基磺酸基團,該二級芳香胺中的芳香碳原子直接與氮原子結合,並且該等基團中的至少一個,A或B,包括一聯苯單元,並且該反應在一鹼和一鈀錯合物存在下進行,該鈀原子被至少一個雙(二烷基膦基)二茂鐵配位基錯合。
- 如申請專利範圍第1項所述之方法,該等雙(二烷基膦基)二茂鐵配位基之烷基取代基具有二至五個碳原子。
- 如申請專利範圍第1或2項所述之方法,該等烷基取代基選自由以下各項組成之群組:異丙基、異丁基、三級丁基以及其組合。
- 如申請專利範圍第1或2項所述之方法,芳香族取代基,A和B,兩者都包括一聯苯基單元,該等單元係彼此相同或不同的。
- 如申請專利範圍第1或2項所述之方法,該聯苯基單元直接與該胺的二級氮原子結合。
- 如申請專利範圍第1或2項所述之方法,該聯苯基單元係具有式2或3的一橋聯聯苯基單元 其中,D是氧、硫、氮或碳並且可以在氮情況下被甲基、乙基、聯苯基、萘基或苯基單取代或在碳情況下被甲基、乙基、聯苯基、萘基或苯基雙取代;R31是氫、苯基、聯苯基或吡啶基;且R32是一離去基團、鹵素、NH2或三氟甲基磺酸基團,取決於該基團是否用作A或B;亦或R32係一間隔基,分別被安排在A或B與X或NH2之間。
- 如申請專利範圍第1或2項所述之方法,該聯苯基單元係2-茀、3-茀、2-(9,9-二苯基茀)、2-(9,9-二甲基茀)、3-(9,9-二苯基茀)、3-(9,9-二甲基茀)、3-(4-苯基)-9-苯基-9H-咔唑、3-(4-苯基)-9-甲基-9H-咔唑、3-(4-苯基)-9-聯苯基-9H-咔唑、2-(4-苯基)-9-苯基-9H-咔唑、2-(4-苯基)-9-甲基-9H-咔唑、2-(4-苯基)-9-聯苯基-9H-咔唑、3-(4-聯苯基)-9-苯基-9H-咔唑、3-(4-聯苯基)-9-甲基-9H-咔唑、3-(4-聯苯基)-9-聯苯基-9H-咔唑、2-(4-聯苯基)-9-苯基-9H-咔唑、2-(4-聯苯基)-9-甲基-9H-咔唑、2-(4-聯苯基)-9-聯苯基-9H-咔唑、3-(9-苯基-9H-咔唑)、3-(9-甲基-9H-咔唑)、3-(9-聯苯基-9H-咔唑)、2-(9-苯基-9H-咔唑)、2-(9-甲基-9H-咔唑)、2-(9-聯苯基-9H-咔唑)或苯並菲。
- 如申請專利範圍第1或2項所述之方法,基於該一級芳香胺A-NH2和該鹵代芳香族化合物X-B兩種反應物之總莫耳量以0.01mol%至1.5mol%的量使用該鈀錯合物。
- 如申請專利範圍第1或2項所述之方法,該鈀錯合物以一固體、溶液或與固體雙(二烷基膦基)二茂鐵的粉末混合物之形式使用。
- 如申請專利範圍第1或2項所述之方法,包括以下步驟:- 在一反應器中提供該一級芳香胺A-NH2、該鹵代芳香族化合物X-B、一適合的溶劑以及任選地雙(二烷基膦基)二茂鐵;- 添加一呈固體或溶液形式之鈀錯合物,其中該鈀原子被至少一個雙(二烷基膦基)二茂鐵配位基錯合;- 加熱該反應器中的所得的反應混合物;- 分離該反應產物,二級芳香胺A-NH-B;並且- 任選地純化該二級芳香胺A-NH-B。
- 如申請專利範圍第1或2項所述之方法,其中該一級芳香胺A-NH2係且該鹵代芳香族化合物X-B係
- 如申請專利範圍第1或2項所述之方法,在該鈀錯合物中的鈀原子附加地被2,4,6,8-四甲基環四矽氧烷、雙(二苯亞甲基丙酮)或馬來醯亞胺錯合。
- 如申請專利範圍第1或2項所述之方法,在該鈀錯合物中之鈀原子被1,1’-雙(二異丙基膦基)二茂鐵錯合。
- 一種化合物,其係1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-馬來醯亞胺[Pd(dippf)(馬來醯亞胺)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-1,3-二乙烯基-1,1,3,3-四甲基二矽氧烷[Pd(dippf)(VTS)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-雙(二苯亞甲基丙酮)[Pd(dippf)(dba)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-萘醌[Pd(dippf)(萘醌)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-馬來酸酐[Pd(dippf)(馬來酸酐)]、1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-馬來酸二乙酯[Pd(dippf)(馬來酸二乙酯)]、或1,1’-雙(二異丙基膦基)二茂鐵鈀(0)-降冰片烯[Pd(dippf)(降冰片烯)]。
- 一種粉末混合物,包括一種雙(二烷基膦基)二茂鐵和一鈀錯合物,其中該鈀原子被至少一個雙(二烷基膦基)二茂鐵配位基錯合,惟當該雙(二烷基膦基)二茂鐵係[Fe(η5-C5H4PEt2)2]時,該鈀錯合物不為Fe(η5-C5H4PEt2)2PdCl2。
- 一種如申請專利範圍第14項所述之化合物或如申請專利範圍第15項所述之粉末混合物作為催化劑用於哈特維希-布赫瓦爾德偶合之用途。
