Abstract
Description
The invention relates to Microbiology and agriculture, and specifically to a method for cultivating microorganisms based on lactic acid bacteria to obtain a probiotic additive in order to increase the meat productivity of quails.
Modern poultry farming in many ways solves the problem of maximizing the bird's bioresource potential and at the same time preserving its productive health. Accelerating the dynamics of growth and increasing daily increase in live weight of poultry while reducing economic costs per 1 kg increase in live weight is one of the main tasks of the poultry industry, in particular, quailing.
Known use for the cultivation of microbial culture molasses medium composed of 1 liter of water: 45 g beet, molasses, 2 g KN 2 PO 4 , 0.02 g yeast extract (Patent RF №243864,
… с.-х. В. - 24 с.).The known method provides for the use in the composition of animal feed for quails of a probiotic feed additive, which includes three types of lactic acid bacteria: Lactobacillus delbrueckii subsp. bulgaricus B-5788, Lactobacillus acidophilus B-3235, Lactococcus lactis ssp. lactis B-3145 grown on a medium consisting of water, beet molasses, milk or whey, which improves the safety and productivity of quails (Kobylyatskaya GV Receipt and efficacy of using probiotic Trilatobact in quailing: abstract of thesis ... cand. of agricultural sciences / GV Kobylyatskaya. - Krasnodar, 2013. - 24 p.).
The disadvantage of this method is that the probiotic contains microorganisms that are not natural representatives of the microbiome of the gastrointestinal tract of quails, and the nutrient medium used allows the titer of each culture to be only 5.0 × 10 7 CFU / ml, as a result, will ensure the maximum realization of the biological potential of the bird.
- 2017. - №04 (128). - С.1102-1115).The closest is the technical solution used to obtain probiotic supplements for quails, the medium consisting of feed molasses, K 2 HPO 4 , yeast extract and water (Intensification of the cultivation process of physiologically adapted strains of lactobacilli as the basis for the creation of microbial biopreparations for poultry farming / A. G. Koshchaev, Yu.A. Lysenko, Mishchenko VA [and others] // Polytematic network electronic scientific journal of the Kuban State Agrarian University. - 2017. - №04 (128). - P.1102-1115).
Lactobacillus agilis.A disadvantage of the known technical solution is that an insufficient increase in the titer of lactic acid cultures of Lactobacillus agilis.
The technical result is to increase the titer of lactic acid cultures of Lactobacillus agilis, which are representatives of the natural microflora of the gastrointestinal tract of quails, due to the use of molasses nutrient medium.
The technical result is achieved by the fact that in a nutrient medium for the cultivation of lactobacilli consisting of feed molasses, K 2 HPO 4 , yeast extract and water, according to the invention, feed molasses consists of beet and corn molasses taken in equal parts and microorganisms of Lactobacillus agilis with no titer less than 1.0 × 10 5 CFU / ml in the following ratio of initial components, g / l:
Меласса свекловичная - 22-23;Beet molasses - 22-23;
Меласса кукурузная - 22-23;Corn molasses - 22-23;
К2НРО4 - 2,0;K 2 NRA 4 - 2.0;
Дрожжевой экстракт - 0,02;Yeast extract - 0.02;
Lactobacillus agilisMicroorganisms Lactobacillus agilis
с титром не менее 1,0×105 КОЕ/мл - 99-101;with a titer of at least 1.0 × 10 5 CFU / ml - 99-101;
Water - the rest
The determination of the microflora titer was carried out according to GOST 10444.11-89 (clause 4.2.2). They took 1.0 ml of each culture grown and placed in a flask with a volume of 100 cm 3 and poured 99.0 ml with sterile saline, left for 1 hour. A dilution of 1: 100 was obtained. After that they prepared a series of consecutive tenfold dilutions up to 10 -10 . Separate sterile tips were used for each dilution. Sowing in Petri dishes was carried out according to (on Laktobakagar from
X=N×P,X = N × P,
где: N - среднеарифметическое значение числа колоний в чашках Петри; Р - порядковый номер десятикратного разведения, в котором отмечается рост бактерий.where: N is the arithmetic mean value of the number of colonies in Petri dishes; P - the serial number of tenfold dilution, in which the growth of bacteria.
In the process of cultivation, an analysis of the dynamics of consumption of reducing substances (PB) with an initial concentration of 4% was carried out. The method of determining the reducing substances is based on the reducing ability of the monoforms of sugars - glucose and fructose.
Results on the number of microbial cultures studied, obtained under laboratory conditions during cultivation in a molasses medium, are presented in Figure 1, where the composition numbers of the molasses medium are indicated on the abscissa axis and the number of microorganisms on the ordinate axis.
As a result of the study, molasses medium No. 3 turned out to be the most effective, where 50% of beet and 50% of corn molasses, Lb. titer, were used as fodder molasses. agilis was 3.3 × 10 10 CFU / ml, while on other variants the titer of the cultures used did not exceed 1.0 × 10 9 CFU / ml. So on molasses nutrient No. 1 amount of Lb. agilis was 2.1 × 10 9 CFU / ml; on
Also in the process of cultivation, the dynamics of consumption of reducing substances (PB) on molasses medium No. 3 were recorded, the results of which are shown in Figure 2, where the time of cultivation of microorganisms is indicated on the abscissa and the number of reducing substances is shown on the ordinate axis.
Based on the results of monitoring the carbon substrate consumption, it can be concluded that it is depleted by as early as 24 hours from the start of cultivation.
The next stage of the research was to determine the effect on growth of the used cultures of various nutrient media, which are often used for the cultivation of lactic acid bacteria. The cultivation time for all cultures was 48 h, the temperature optimum was 37 ° C.
For evidence of the effectiveness of the proposed environment for comparison were used media of the following composition:

The composition of the medium HPS, g / l: Na 2 HPO 4 12-water - 3.2; KH 2 PO 4 - 0.3; MgSO 4 - 0.5; NaCl - 0.5; peptone - 2.0; yeast extract - 0.05; glucose - 25.
4. The composition of the molasses medium: 45 g / l molasses fodder (50% beet and 50% corn molasses), K 2 HPO 4 - 2 g / l, yeast extract - 0.02 g / l.
Results on the number of microbial cultures studied, obtained under laboratory conditions during cultivation in various nutrient media, are presented in Figure 3, where different nutrient media are indicated on the abscissa axis and the number of microorganisms on the ordinate axis.
As a result of the study, the molasses medium turned out to be the most effective, since the Lb. agilis was 4.7 × 10 10 CFU / ml. The environment for lactic acid bacteria was less effective. Uglich - Lb. agilis - 8.7 × 10 9 CFU / ml. When growing lactobacilli on the environment HPS were obtained the following results: Lb. agilis - 2.7 × 10 9 CFU / ml, and on MPC: Lb. agilis - 2.0 × 10 9 CFU / ml.
Next, we determined the optimal cultivation temperature of the tested microorganisms in order to increase the cell titer in the shortest possible time. The above cultivation results were obtained at a growing temperature of 37 ° C.
A series of experiments was laid to determine the maximum thermotolerance of these crops when grown on molasses medium No. 3 for 48 hours. The results of temperature dependence of the number of lactic acid bacilli cells are presented in Figures 4 and 5, where the time of cultivation of microorganisms is indicated on the abscissa axis and - the number of microorganisms.
The result of cultivation at 39 ° C is not represented, since at this temperature no culture showed growth relative to the seed titer. However, after the cultivation temperature was reduced to 37 ° C, the growth was restored in the same volume, which indicates that the culture did not die off, but entered the phase of growth inhibition, which continued until the temperature decreased by 2-3 ° C.
Based on the results of cultivation on various media and at different temperatures, it was established that the highest cell titer was achieved by 24 hours from the start of the cultivation, regardless of the composition of the media. Longer growth and increase in temperature leads to a decrease in titer and, as a rule, an increase in the number of non-viable cells.
The results of the studies showed that the most effective nutrient medium is molasses medium of the following composition: 45 g / l molasses feed (50% beet and 50% corn molasses), K 2 HPO 4 - 2 g / l, yeast extract - 0.02 g / l, while the optimum temperature and time regimes are 37 ° C and 24 h, respectively. The developed molasses environment can be used under production conditions during the further development of biological products for animal husbandry, including poultry farming.
To prove the effectiveness of the obtained probiotic supplement on the basis of Lactobacillus agilis microorganisms, scientific studies were conducted on quails of the Japanese breed.
Применение пробиотической добавки в опытных группах осуществлялось ежедневно (таблица 1).By the method of groups of analogues, five groups of quails of 20 animals each were formed: the control group - only the main complete feed and drinking water were present in the diet; The 1st experimental group - with the basic ration and drinking water to the bird, they also drank a probiotic supplement in a dose of 0.2 ml / goal; 2nd experimental group - with the basic ration and drinking water to the bird, they also drank a probiotic supplement in a dose of 0.5 ml / goal; The 3rd experimental group - with the basic ration and drinking water to the bird, also drank a probiotic supplement in a dose of 1.0 ml / goal. The use of probiotic supplements in the experimental groups was carried out daily (Table 1).
Quails were grown in industrial multi-tier metal cages.
2.The results of economic indicators for growing quails are presented in table 2.
2.The results of economic indicators for growing quails are presented in table 2.
As can be seen from the table, a significant increase in live weight of quails in comparison with the control group was detected on the 21st day in the 2nd and 3rd experimental groups, respectively, at 7.80 and 7.16%. On the 28th day, in the 2nd experimental group, the live weight of quails was greater than in the control by 13.46%, and in the 3rd group - by 14.21%. On the 35th day of bird rearing, a significant increase in live weight was observed in the 2nd and 3rd experimental groups by 10.60 and 10.83%. A similar trend in these experimental groups for the studied indicator was observed on the 42nd day, in which the live weight was higher by 9.12% and 9.75%. It should be noted that in the 1st experimental group there was a slight increase in live weight over the entire growing period.
10,10%.The safety of quails in all groups was the same and amounted to 100.0%. The increase in live weight of quails for the entire period of cultivation in the experimental groups was also greater than in the control group by 2.91; 9.45 and 10.10%.
6,78%.As can be seen from the data of table 2, with an increase in the live weight of the experienced birds, their consumption of compound feed also increases. The cost of feed per 1 kg increase in live weight in the experimental groups remained lower than in the control by 2.01; 7.79; and 6.78%.
In general, to justify the increased dynamics of live weight of quails in the experimental groups, it is possible due to the positive effects of biological products based on Lactobacillus agilis cultures, which contribute to better absorption of energy and nutrients of the compound feed by poultry.
Thus, the test results showed that growing quails using the developed probiotic supplement based on the culture of the species Lactobacillus agilis, isolated from the gastrointestinal tract of quails, and also grown on a molasses medium, in test doses provides an increase in the productivity of quails, which was especially detected at a dose of 0.5 ml / goal.
