Claims (42)
1. Вариантный полипептид Cry1B, содержащий аминокислотную последовательность, характеризующуюся по меньшей мере 95% идентичностью последовательности с SEQ ID NO: 62, SEQ ID NO: 63, SEQ ID NO: 64, SEQ ID NO: 65, SEQ ID NO: 66, SEQ ID NO: 67, SEQ ID NO: 68, SEQ ID NO: 69, SEQ ID NO: 70, SEQ ID NO: 71, SEQ ID NO: 72, SEQ ID NO: 73, SEQ ID NO: 74, SEQ ID NO: 75, SEQ ID NO: 76, SEQ ID NO: 77 или SEQ ID NO: 78, где вариантный полипептид Cry1B обладает повышенной инсектицидной активностью против кукурузной совки и/или травяной совки по сравнению с полипептидом Cry1B под SEQ ID NO: 1.

Вариантный полипептид Cry1B по п. 1, где полипептид Cry1B содержит аминокислотную последовательность под SEQ ID NO: 62, SEQ ID NO: 63, SEQ ID NO: 64, SEQ ID NO: 65, SEQ ID NO: 66, SEQ ID NO: 67, SEQ ID NO: 68, SEQ ID NO: 69, SEQ ID NO: 70, SEQ ID NO: 71, SEQ ID NO: 72, SEQ ID NO: 73, SEQ ID NO: 74, SEQ ID NO: 75, SEQ ID NO: 76, SEQ ID NO: 77 или SEQ ID NO: 78.

8. Вариантный полипептид Cry1B по любому из пп. 1-5, где вариантный полипептид Cry1B соединен с гетерологичной сигнальной последовательностью или транзитной последовательностью.
9. Рекомбинантный полинуклеотид, кодирующий вариантный полипептид Cry1B по любому из пп. 1-8.
10. Рекомбинантный полинуклеотид по п. 9, где последовательность нуклеиновой кислоты была оптимизирована для экспрессии в растении.
11. Рекомбинантный полинуклеотид по п. 10, где последовательность нуклеиновой кислоты представляет собой синтетическую нуклеотидную последовательность, имеющую предпочтительные для растения кодоны, которые были сконструированы для экспрессии в растении.
12. Рекомбинантный полинуклеотид по п. 11, где последовательность нуклеиновой кислоты была оптимизирована для экспрессии в маисе или сое.
13. ДНК-конструкция, содержащая полинуклеотид по любому из пп. 9-12, функционально связанный с гетерологичным регуляторным элементом.
14. Клетка-хозяин, содержащая ДНК-конструкцию по п. 13.

20. Семя, содержащее ДНК-конструкцию по п. 13.
21. Композиция, содержащая вариантный полипептид Cry1B по любому из пп. 1-8.
22. Композиция по п. 21, где композиция содержит от 1% до 99% по весу вариантного полипептида Cry1B.
23. Способ контроля популяции чешуекрылого вредителя, предусматривающий приведение указанной популяции в контакт с пестицидно эффективным количеством вариантного полипептида Cry1B по любому из пп. 1-8.
24. Способ уничтожения чешуекрылого вредителя, предусматривающий приведение указанного вредителя в контакт с пестицидно эффективным количеством вариантного полипептида Cry1B по любому из пп. 1-8 или его скармливание указанному вредителю.
25. Способ получения полипептида с пестицидной активностью, предусматривающий культивирование клетки-хозяина по п. 13 в условиях, при которых экспрессируется полинуклеотид, кодирующий полипептид.
26. Растение или клетка растения, в геном которых стабильно встроена ДНК-конструкция по п. 14.
27. Способ защиты растения от насекомого-вредителя, предусматривающий введение в указанное растение ДНК-конструкции по п. 14.
28. Способ по п. 27, где вариантный полипептид Cry1B обладает повышенной инсектицидной активностью против кукурузной совки и/или травяной совки по сравнению с полипептидом под SEQ ID NO: 47.
29. Способ по п. 27 или п. 28, где насекомое-вредитель является устойчивым к инсектицидному полипептиду, отличному от Cry1B.
30. Способ по любому из пп. 27, 28 или п. 29, где насекомое-вредитель представляет собой кукурузную совку, травяную совку или кукурузного мотылька.
31. Химерный полипептид Cry1B, содержащий домен I первого полипептида Cry1B и домен II и домен III второго полипептида Cry1B.
32. Химерный полипептид Cry1B, содержащий домен II первого полипептида Cry1B и домен I и домен III второго полипептида Cry1B.
33. Химерный полипептид Cry1B, содержащий домен I, домен II и домен III первого полипептида Cry1B, где одна, две или три альфа-спирали домена I замещены соответствующими альфа-спиралями второго полипептида Cry1B.
34. Химерный полипептид Cry1B по любому из пп. 31, 32 или п. 33, где первый полипептид Cry1B и второй полипептид Cry1B выбраны из Cry1Bd под SEQ ID NO: 1 и MP258 под SEQ ID NO: 47.
35. Рекомбинантный полинуклеотид, кодирующий химерный полипептид Cry1B по любому из пп. 31, 32, 33 или п. 34.
36. ДНК-конструкция, содержащая полинуклеотид по п. 35, функционально связанный с гетерологичным регуляторным элементом.
37. Трансгенное растение, содержащее ДНК-конструкцию по п. 36.
38. Семя, содержащее ДНК-конструкцию по п. 36.
39. Композиция, содержащая химерный полипептид Cry1B по любому из пп. 31, 32, 33 или п. 34.
40. Способ контроля популяции чешуекрылого вредителя, предусматривающий приведение указанной популяции в контакт с пестицидно эффективным количеством химерного полипептида Cry1B по любому из пп. 31, 32, 33 или п. 34.
41. Способ защиты растения от насекомого-вредителя, предусматривающий введение в указанное растение ДНК-конструкции по п. 36.
42. Способ по п. 41, где способ дополнительно предусматривает обеспечение экспрессии химерного полипептида Cry1B в растении.
