Claims (30)
1. Трехмерная сетка, обладающая поверхностью и внутренней частью, указанная трехмерная сетка включает:
(a) сшитый полимер, ковалентно связанный с поверхностью субстрата;
(b) один или большее количество каналов; и
(c) молекулы-зонды, ковалентно связанные с полимером.
2. Трехмерная сетка по п. 1, в которой по меньшей мере один из каналов обладает одной, двумя или тремя из следующих характеристик:
(a) канал простирается во внутреннюю часть от точки, которая находится на расстоянии менее 5 мкм от поверхности сетки или простирается во внутреннюю часть от точки на поверхности сетки;
(b) канал обладает длиной, которая составляет не менее 10% от наибольшего размера сетки; и
(c) канал обладает минимальным сечением, которое по меньшей мере в 5 раз больше размера отверстий сетки.
3. Трехмерная сетка по п. 1 или 2, включающая не менее 5 каналов, в которой множество каналов сходится в точке во внутренней части сетки, так что боковое расстояние между каналами уменьшается при переходе от поверхности к точке во внутренней части.
4. Трехмерная сетка по любому из пп. 1-3, в которой сшитый полимер набухает в воде.
5. Трехмерная сетка по п. 4, где сетка в своем гидратированном состоянии обладает отверстиями размером от 5 до 75 нм.
6. Трехмерная сетка по любому из пп. 1-5, в которой большинство молекул-зондов находится во внутренней части сетки и/или соприкасается с каналом.
7. Трехмерная сетка по любому из пп. 1-5, в которой полимер включает сополимер, полученный сополимеризацией диметилакриламида (ДМАА), метакрилоилоксибензофенона (МАБФ) и 4-винилбензолсульфоната натрия (SSNa).
8. Массив, включающий множество трехмерных сеток по любому из пп. 1-7 на субстрате, в котором (а) трехмерные сетки иммобилизованы на субстрате и (b) каждая из трехмерных сеток находится в отдельном пятне на субстрате.
9. Массив по п. 8, который можно использовать повторно не менее 10 раз.
10. Способ получения трехмерной сетки по любому из пп. 1-7, включающий стадии:
(a) обработку смеси, находящейся на поверхности субстрата смеси при условиях, обеспечивающих образование игольчатых кристаллов, указанная смесь включает (i) водный раствор соли, который необязательно является раствором соли одновалентного катиона, (ii) полимер, (iii) сшивающий реагент и (iv) молекулы-зонды, тем самым образование смеси, содержащей один или большее количество игольчатых кристаллов соли;
(b) обработку смеси, содержащей один или большее количество кристаллов соли, при условиях, обеспечивающих сшивку, тем самым образование сшитой полимерной сетки, содержащей один или большее количество игольчатых кристаллов соли; и
(c) взаимодействие сшитой полимерной сетки, содержащей один или большее количество кристаллов соли, с растворителем, в котором растворим один или большее количество кристаллов соли, тем самым растворение игольчатых кристаллов соли и образование одного или большего количества каналов вместо кристаллов соли.
11. Способ по п. 10, в котором:
(а) условия, обеспечивающие образование игольчатых кристаллов соли, включают:
(i) дегидратацию смеси; или
(ii) охлаждение смеси; и/или
(b) растворителем является буфер на водной основе.
12. Способ получения массива, включающий (а) образование множества трехмерных сеток способом по п. 10 или 11 на отдельных пятнах на поверхности одного того же субстрата и (b) сшивку множества трехмерных сеток с поверхностью.
13. Способ определения того, содержится ли анализируемое вещество в образце, включающий:
(a) взаимодействие трехмерной сетки по любому из пп. 1-7 или массива по п. 8 или 9, включающего молекулы-зонды, которые способны связываться с анализируемым веществом, с образцом; и
(b) обнаружение связывания анализируемого вещества с молекулами-зондами в трехмерной сетке, тем самым определение того, содержится ли анализируемое вещество в образце, и необязательно количества анализируемого вещества.
14. Способ по п. 13, в котором сетку или массив использовали и промывали не менее 10 раз до стадии (а) или который дополнительно включает повторное использование сетки или массива не менее 10 раз после стадии (b).
15. Способ по п. 14, который дополнительно включает количественное определение связывания анализируемого вещества с молекулами-зондами в трехмерной сетке.
