Abstract
1. Обогащенная гетерогенная популяция клеток почек млекопитающих, которая экспрессирует биомаркеры GGT-1 и цитокератин.2. Популяция клеток по п. 1, отличающаяся тем, что биомаркеры выявляют с помощью моноклонального или поликлонального антитела.3. Популяция клеток по п. 1, отличающаяся тем, что более 18% клеток экспрессируют GGT-1.4. Популяция клеток по п. 1, отличающаяся тем, что цитокератин выбирают из СК8, СК18, СК19 и их комбинаций.5. Популяция клеток по п. 4, отличающаяся тем, что уровень экспрессии цитокератина составляет более 80%.6. Способ по п. 11, отличающийся тем, что биомаркер выбран из: AQP1, AQP2, AQP4, кальбиндина, кальпонина, CD117, CD133, CD146, CD24, CD31 (РЕСАМ-1), CD54 (ICAM-1), CD73, СК18, СК19, СК40 до 67, СК7, СК8, СК8, СК18, СК19, комбинаций СК8, СК18 и СК19, коннексина 43, кубилина, CXCR4 (фузин), DBA,
Claims (19)
1. Обогащенная гетерогенная популяция клеток почек млекопитающих, которая экспрессирует биомаркеры GGT-1 и цитокератин.
2. Популяция клеток по п. 1, отличающаяся тем, что биомаркеры выявляют с помощью моноклонального или поликлонального антитела.
3. Популяция клеток по п. 1, отличающаяся тем, что более 18% клеток экспрессируют GGT-1.
4. Популяция клеток по п. 1, отличающаяся тем, что цитокератин выбирают из СК8, СК18, СК19 и их комбинаций.
5. Популяция клеток по п. 4, отличающаяся тем, что уровень экспрессии цитокератина составляет более 80%.
6. Популяция клеток по п. 1, отличающаяся тем, что цитокератин выявляют с помощью поликлонального антитела.
7. Популяция клеток по п. 1, отличающаяся тем, что данная популяция клеток экспрессирует AQP.
8. Популяция клеток по п. 1, отличающаяся тем, что менее 40% клеток экспрессируют AQP2.
9. Популяция клеток по п. 7, отличающаяся тем, что AQP2 выявляют с помощью моноклонального антитела.
10. Популяция клеток по п. 1, отличающаяся тем, что млекопитающее является человеком.
11. Способ идентификации гетерогенной популяции клеток почек, подходящей для имплантации и/или получения регенеративного ответа, указанный способ включает этапы:
выделения клеток из образца почки млекопитающего;
воздействия на указанные выделенные клетки одним или более мечеными компонентами для детекции, причем каждый меченый компонент для детекции нацелен на уникальный биомаркер и помечен уникальной меткой;
определения процентного содержания клеток, которые экспрессируют каждый из указанных уникальных биомаркеров.
12. Способ по п. 11, отличающийся тем, что биомаркер выбран из: AQP1, AQP2, AQP4, кальбиндина, кальпонина, CD117, CD133, CD146, CD24, CD31 (РЕСАМ-1), CD54 (ICAM-1), CD73, СК18, СК19, СК40 до 67, СК7, СК8, СК8, СК18, СК19, комбинаций СК8, СК18 и СК19, коннексина 43, кубилина, CXCR4 (фузин), DBA, Е-кадгерина (CD324), ЭПО (эритропоэтин), GGT1, GLEPP1 (гломерулярный эпителиальный белок 1), гаптоглобулина, Itgb1 (интегрин β1), ΚΙΜ-1/ΤΙΜ-1 (молекула повреждения почек-1 /Т-клеточный
иммуноглобулин и муцин-содержащая молекула), МАР-2 (ассоциированный с микротрубочками белок 2), мегалина, N-кадгерина, нефрина, NKCC (Na-K-Cl-котранспортеры), OAT-1 (транспортер 1 органических анионов), остеопонтина, пан-кадгерина, PCLP1 (подобная подокаликсину 1 молекула), подоцина, SMA (альфа-актин гладких мышц), синаптоподина, ТНР (белок Тамма-Хорсфалла), виментина, αGST-1 (альфа глутатион-S-трансфераза) и их комбинаций.
13. Способ по п. 11, отличающийся тем, что меченый компонент для детекции представляет собой антитело.
14. Способ лечения болезни почек у млекопитающего, включающий введение гетерогенной популяции клеток почек пациенту, причем данная популяция клеток экспрессирует два или более биомаркеров, выбранных из группы, состоящей из: GGT-1, AQP2 и цитокератина.
15. Способ лечения болезни почек у млекопитающего, включающий введение гетерогенной популяции клеток почек пациенту, причем клетки данной популяции клеток имеют уровни экспрессии GGT-1 и СК18, которые превышают, соответственно, 18% и 80%.
16. Способ лечения болезни почек у млекопитающего, включающий введение гетерогенной популяции клеток почек пациенту, причем клетки данной популяции клеток экспрессируют GGT-1, AQP2 и СК18 на уровнях экспрессии, указанных в Таблице 12.4.
17. Способ по п. 15 или 16, отличающийся тем, что популяцию клеток культивируют на матрице.
18. Способ по п. 15 или 16, отличающийся тем, что популяцию клеток суспендируют в биоматериале на основе желатина.
19. Способ по п. 15 или 16, отличающийся тем, что млекопитающее является человеком.
