Claims (10)
1. Биологически активный полипептид MF3, характеризующийся первичной структурой (аминокислотной последовательностью) SEQ ID NO.1, активный фрагмент полипептида MF3 или любое функциональное производное MF3; выше заявленный полипептид, активный фрагмент или функциональная производная способны обеспечивать эффективную устойчивость растения к инфекционным болезням и/или растительным паразитам.
2. Выделенная последовательность ДНК, изображенная в SEQ ID:2, или ее фрагмент, кодирующий функционально активный полипептид MF3 или его активный фрагмент согласно п.1; выше заявленный фрагмент ДНК может содержать вырожденные кодоны.
3. Метод приобретения устойчивости растения к фитопатогенам и/или к растительным паразитам путем введения в растение биологически активного полипептида MF3 или его активного фрагмента, или его функционального производного механически или посредством молекул-носителей.
4. Метод согласно п.3, где носителем выступает хитозан.
5. Вектор, включающий ДНК согласно п.2.
6. Метод получения трансгенного растения или культуры растительных клеток, где экспрессируется полипептид, кодируемый ДНК, использующий вектор согласно п.5.
7. Клетка организма-хозяина стабильно трансформированная или трансфецированная вектором согласно п.5.
8. Состав защитного средства растений, включающего любой из компонентов согласно п.1.
9. Активный фрагмент полипептида MF3 согласно п.1, аминокислотная последовательность которого состоит из последовательностей SEQ ID:3 или SEQ ID:4.
10. Метод выделения и очистки полипептида согласно п.1 из бактериальных клеток, экспрессирующих вышеуказанный полипептид; метод включает следующие стадии:
а) культивация бактериального штамма-продуцента и экстракция из клеток в буферном растворе при повышенной температуре;
б) осаждение неочищенного полипептида MF3 при низкой температуре с помощью осаждающего агента;
в) фракционирование растворенного осадка неочищенного полипептида MF3 с помощью анион обменной колоночной хроматографии и сбор фракций с противомикробной или инсектицидной активностью;
г) проведение электрофореза в полиакриламидном геле полипептидных фракций с противомикробной, противонематодной или инсектицидной активностями;
д) рефолдинг белка, элюированного из геля предыдущей стадии.
