Claims (55)

1. Антагонист c-Met, нарушающий димеризацию c-Met.
2. Антагонист по п.1, нарушающий функцию димеризации домена Sema.
3. Антагонист по п.1, связывающий c-Met, так что нарушена димеризация c-Met.
4. Антагонист по п.1, связывающий c-Met, так что нарушена способность домена c-Met Sema осуществлять димеризацию c-Met.
5. Антагонист c-Met, специфически связывающий последовательность в домене c-Met.
6. Антагонист по п.1 или 5, при связывании с молекулой c-Met ингибирующий димеризацию указанной молекулы.
7. Антагонист по п.6, где указанная димеризация предусматривает гомодимеризацию.
8. Антагонист по п. 1 или 5, не связывающий участок связывания HGF на c-Met.
9. Антагонист по п. 1 или 5, в основном не конкурирующий с фактором роста гепатоцитов (HGF) за связывание с c-Met.
10. Антагонист по п. 1 или 5, в основном не ингибирующий связывание фактора роста гепатоцитов с c-Met.
11. Антагонист по п. 1 или 5, конкурирующий с HGF за связывание с c-Met.
12. Антагонист по п. 1 или 5, связывающий эпитоп c-Met, отличный от эпитопа, связывающего моноклональное антитело, продуцируемое линией клеток гибридомы, хранящейся в Американской коллекции типовых культур под инвентарным номером ATCC HB-11894 (гибридома 1A3.3.13) или HB-11895 (гибридома 5D5.11.6).
13. Антагонист по п. 1 или 5, не являющийся моноклональным антителом, продуцируемым линией клеток гибридомы, хранящейся в Американской коллекции типовых культур под инвентарным номером ATCC HB-11894 (гибридома 1A3.3.13) или HB-11895 (гибридома 5D5.11.6).
14. Антагонист по п. 1 или 5, представляющий собой антитело или его фрагмент, пептид или их сочетание.
15. Антагонист по п. 1 или 5, где связывание антагониста с c-Met ингибирует зависимую или не зависимую от HGF активацию c-Met.
16. Антагонист по п. 1 или 5, где связывание антагониста с c-Met в клетке ингибирует пролиферацию, распределение, морфогенез и/или подвижность клетки.
17. Антагонист по п. 1 или 5, где указанный антагонист содержит пептид, содержащий по меньшей мере часть домена Sema c-Met или его варианта.
18. Антагонист по п.17, где указанный пептид содержит по меньшей мере одну из последовательностей, выбранной из группы, включающей в себя LDAQT (SEQ ID NO:1), LTEKRKKRS (SEQ ID NO:2), KPDSAEPM (SEQ ID NO:3) и NVRCLQHF (SEQ ID NO:4).

34. Способ по п.33, где отобранное вещество приводят в контакт с клеткой, экспрессирующей c-Met, и детектируют или оценивают количественно ингибирование димеризации c-Met в клетке.
35. Способ по п.34, где ингибирование димеризации c-Met определяют по уменьшению степени активации c-Met.
36. Способ по п.35, где степень активации c-Met определяют по степени связанной с c-Met передачи сигнала в клетке.
37. Способ по п.36, где указанную передачу сигнала в клетке определяют по фосфорилированию белка.
38. Способ модулирования активации c-Met у субъекта, где указанный способ включает введение субъекту модулятора домена Sema c-Met, посредством чего модулируют активность c-Met.
39. Способ ингибирования активированной c-Met пролиферации клетки, где указанный способ включает приведение в контакт клетки или ткани с антагонистом по п. 1 или 5, посредством чего ингибируют связанную с активацией c-Met пролиферацию клетки.
40. Способ лечения патологического состояния, связанного с дисрегуляцией активации c-Met у субъекта, где указанный способ включает введение субъекту антагониста по п. 1 или 5, посредством чего ингибируют активацию c-Met.
41. Способ ингибирования роста клетки, экспрессирующей c-Met или фактор роста гепатоцитов, или оба, где указанный способ включает приведение в контакт указанной клетки с антагонистом по п.1 или 5, посредством чего вызывают ингибирование роста указанной клетки.
42. Способ терапевтического лечения млекопитающего, страдающего злокачественной опухолью, содержащей клетки, экспрессирующие c-Met или фактор роста гепатоцитов, или оба, где указанный способ включает введение указанному млекопитающему эффективного количества антагониста по п. 1 или 5, посредством чего эффективно лечат указанное млекопитающее.
43. Способ лечения или предупреждения нарушения пролиферации клеток, связанного с увеличенной экспрессией или активностью c-Met или фактора роста гепатоцитов, или обоих, где указанный способ включает введение субъекту, нуждающемуся в таком лечении, эффективного количества антагониста любому из пп.1-21, посредством чего эффективно лечат или предотвращают указанное нарушение пролиферации клеток.
44. Способ по п.43, где указанное нарушение пролиферации клеток представляет собой рак.
45. Способ ингибирования роста клетки, где рост указанной клетки по меньшей мере частично зависит от потенцирующего рост эффекта c-Met или фактора роста гепатоцитов, или обоих, где указанный способ включает приведение в контакт указанной клетки с антагонистом по п. 1 или 5, посредством чего ингибируют рост указанной клетки.
46. Способ терапевтического лечения опухоли у млекопитающего, где рост указанной опухоли по меньшей мере частично зависит от потенцирующего рост эффекта c-Met или фактора роста гепатоцитов, или обоих, где указанный способ включает приведение в контакт указанной клетки с антагонистом по п. 1 или 5, посредством чего эффективно лечат указанную опухоль.
47. Способ по п.46, где указанная клетка представляет собой раковую клетку.
48. Способ по п.47, где указанную раковую клетку в дальнейшем подвергают облучению и/или действию химиотерапевтического средства.
49. Способ по п.47, где указанную раковую клетку выбирают из группы, включающей в себя клетку рака молочной железы, клетку рака кишечника, клетку рака легкого, клетку папиллярной карциномы, клетку рака предстательной железы, клетку лимфомы, клетку рака толстой кишки, клетку рака поджелудочной железы, клетку рака яичника, клетку рака шейки матки, клетку опухоли центральной нервной системы, клетку остеобластосаркомы, клетку почечно-клеточной карциномы, клетку печеночно-клеточной карциномы, клетку рака мочевого пузыря, клетку карциномы желудка, клетку плоскоклеточной карциномы головы и шеи, клетку меланомы и лейкемическую клетку.
50. Способ по п.47, где c-Met или фактор роста гепатоцитов, или оба более сильно экспрессированы в указанной раковой клетке по сравнению с нормальной клеткой того же тканевого происхождения.
51. Способ по п.46, где указанный фактор роста гепатоцитов экспрессирован другой клеткой.
52. Способ по п.47, где активация c-Met в указанной раковой клетке увеличена по сравнению с нормальной клеткой того же тканевого происхождения.
53. Способ по п.47, приводящей к гибели указанной клетки.
54. Способ по п.46, где в отсутствие указанного антагониста активация c-Met происходит независимо от лиганда.
55. Способ по п.46, где в отсутствие указанного антагониста активация c-Met является зависимой от лиганда.
