Denne oppfinnelse vedrører membran-omspennende proteiner og nukleinsyremolekyler som har biotin-bindende aktivitet.
Bakgrunn for oppfinnelsen
Biotin (vitamin H) er en lett vannløselig substans som finnes i lave konsentrasjoner i blod og vev. Biotins biologiske rolle er som bærer av aktivert C02 og det muliggjør overføring av C02 til akseptorer uten behov for ytterligere fri energi. Det aktiverte karboksybiotin er vanligvis forbundet med et enzym som fordres for dannelsen av karboksybiotin. For eksempel kan biotin være forbundet med pyruvatkarboksylase som i nærvær av acetyl CoA, katalyserer dannelsen av karboksybiotin og den etterfølgende overføring av den aktiverte karboksylgruppe til pyruvat, for å danne oksaloacetat.
Biotin binder seg også med én av de høyest kjente naturlige affiniteter, til avidin, et 63 kDa glykoprotein fra hønseggehvite, og til streptavidin, et ikke-glykosylert protein fra bakterien Streptomyces avidinii. WO 8705026 beskriver isolering av DNA sekvensen til streptavidin fra Streptomyces avidinii og fusjonering av denne sekvensen til genet som koder for den humane low density lipoprotein (LDL) reseptoren.
WO 9719957 beskriver en metode for målrettet avlevering av toksiner eller nukleinsyrer i celler ved bruk av et fusjonsprotein som består av streptavidin og protein A.
Sammenfatning av oppfinnelsen
Det er nå fastslått at den biotin-bindende aktivitet av avidin og streptavidin kan utnyttes ved produksjon av transmembranproteiner som kan binde biotinylerte molekyler.
Proteiner ifølge foreliggende oppfinnelse kan omfatte et cytoplasmatisk domene, et membran-omspennende domene og et ekstracellulært domene, hvor det ekstracellulære domenet omfatter biotin-bindende aktivitet. Det ekstracellulære domenet kan omfatte avidinets eller streptavidinets funksjonelle aktivitet.

Beskrivelse av oppfinnelsen
Foreliggende oppfinnelse vedrører følgelig protein for å målrette bitinylerte molekyler mot spesifikke seter i vev, kjennetegnet ved at det omfatter et membran-omspennende domene av en endocytotisk reseptor og et ekstracellulært domene, som omfatter biotin-bindende aktivitet.
Det er også beskrevet nukleinsyre-molekyl for å målrette biotinylerte molekyler mot spesifikke seter i vev, kjennetegnet ved at det koder for et protein i henhold til et hvilket som helst av kravene 1 til 5.
Proteiner ifølge foreliggende oppfinnelse kan fremstilles ved bruk av konvensjonell rekombinant DNA-teknologi. Typisk manipuleres en DNA-sekvens som koder for det funksjonelle domenet av et biotin-bindende protein, så som avidin, streptavidin eller et beslektet protein, inn i en genetisk konstruksjon som omfatter en DNA-sekvens som koder for et protein som har membran-omspennende egenskaper. Eksempler på avidin- og streptavidin-relaterte proteiner er AVR-1-AVR-5, AVR-X-AVR-V, Stv1 og Stv2.
De enkelte domenene av fusjonsproteinet kan amplifiseres ved PCR (polymerase chain reaction) eller isoleres fra den opphavelige cDNA ved å benytte restriksjonsenzymoppslutning, isolering og rensing, f.eks. ved å benytte gelelektroforese og påfølgende ligering, f.eks. ved å benytte DNA-ligase. Konstruksjonen kan også benyttes som nakent DNA eller som et plasmid/liposom-, plasmid/polyetylenimin-, plasmid/dendrimer- eller plasmid/peptid-kompleks.
Alternativt kan konstruksjonen innføres i et replikasjonsdefekt virus som kan benyttes til å målrette konstruksjonen mot spesifikke seter in vivo. For eksempel kan konstruksjonen være en retroviral vektor, som omfatter den riktige cDNA for fusjonsproteinet. Et replikasjonsdefekt retrovirus, f.eks. Moloney murint retrovirus, kan deretter benyttes for den stabile transfeksjon av målceller og vev. Egnede reseptorer som kan benyttes i henhold til denne oppfinnelse omfatter scavenger-reseptor klasse A, low density lipoproteinreseptor, very low density lipoproteinreseptor, transferrin-reseptor og LOX-1-reseptoren. Fusjonsproteinet kan også omfatte en linker mellom reseptroproteinet og avidin-peptidsekvensene. Linkeren kan ha en hvilken som helst lengde, forutsatt at den funksjonelle aktivitet av de forskjellige komponentene av fusjonsproteinet bibeholdes.
I alminnelighet er fusjonen mellom avidin- eller streptavidin-peptidsekvensene og reseptorpeptidsekvensene mellom reseptorproteinets ekstracellulære domene og et hvilket som helst sete utenfor det biotin-bindende sete til avidin eller streptavidin.
Det følgende eksempel illustrerer oppfinnelsen.
Eksempel
En DNA-konstruksjon ble opprettet mellom den bovine scavenger-reseptor klasse A (ScR) (Kodama et al., (1990) Nature, 343:531-535) og avidin (Green (1975) Adv. Prot. Chem. 29:85-133) som koder for et protein som har et ScR-cytoplasmatisk domene, membran-omspennende domene og cc-heliks-domene, ligert til et biotin-bindende domene. Den fullstendige aminosyresekvens av fusjonsproteinet er vist i SEKV. ID. NR:2, hvor aminosyrene 1-53 representerer det cytoplasmatiske domenet; aminosyrene 55-79 representerer transmembrandomenet; aminosyrene 81-111 representerer et spacer-domene og aminosyrene 113-272 representerer a-heliks-domenet. Aminosyrene 273-400 representerer den modne avidin-peptidsekvens avledet fra avidin-cDNA (Gope et al., (1987) Nucleic Acids Res. 15:3595-3606) som mangler et sekresjonssignal.
I korthet ble cDNA for ScR oppnådd fra dyrkede celler som tidligere var transfektert med et plasmid (PLScRNL) inneholdende ScR cDNA med en intern Rous sarkomavirus promoter og /-//ndlll-retriksjonsseter. Den isolerte cDNA ble deretter innskutt i et /-//ndlll-sete i retrovirusvektoren pLSIARNL. Avidin-cDNA ble produsert ved PCR og deretter innskutt i retrovirusvektoren ved et Sty 1 restriksjonssete på ScR cDNA. Den cDNA som utgjør oppfinnelsen er vist som SEKV. ID. NR:1, hvor nukleotidene 1-989 representerer en lang terminal repetisjon fra Mo-MuSV; nukleotidene 1071-2270 representerer den kodende region for fusjonsproteinet; nukleotidene 2376-3101 representerer den utranslaterte region fra bovin scavenger-reseptor I cDNA; nukleotidene 3107-3376 representerer en RSV promoterregion; nukleotidene 3727-4522 representerer et neo R-gen og nukleotidene 4540-5177 representerer en lang terminal repetisjon fra Mo-MuLV.
Fig. 2 og 3 refererer til prosesser benyttet i dette eksempel. Nærmere bestemt viser Fig. 2 hvorledes ScR cDNA med en intern RSV-promoter, ble kuttet fra plasmid pLScRNL med Hind\\\ og klonet inn i et H/ncflll-sete i en skyttelvektor. Fig. 3 viser hvorledes ScR-avidin-RSV cDNA ble klonet inn i et retrovirusvektor pLRNL HindlU-sete.
Ekspresjonen av fusjonsproteinet i celler transfektert med vektoren, kan bekreftes ved northernblotting og immuncytokjemisk farving med et antistoff utviklet mot avidin.
Forsøkene viste at det fulle mRNA-transkript ble translatert inn i 55 kDa monomerer som var i stand til å danne sekundærstrukturer på 110 kDa dimerer tilkoblet ved S-S-bindinger under ikke-reduserende betingelser.Omtrent 110 kDa dimere og 55 kDa monomere peptider ble påvist under bruk av denaturerende betingelser. Resultatene er sammenlignbare med datamaskinberegninger for det monomere fusjonsprotein, 45 kDa. Under ikke-denaturerende betingelser (dvs. bruk av acetylering før westernblotting) var det sterkeste signalet ca. 220 kDa, som ble denaturert til en ca. 110 kDa dimer og en 55 kDa monomer, hvilket tyder på dannelse av tetramerer. Nærværet av proteiner på 220 kDa ble også verifisert ved bruk av kjemiske kryss-linkere, f.eks. NHS-estere. Resultatene viser at avidin forblir løselig og er i stand til å danne tetramerer selv når det er koblet til membran-omspennende domener av endocytotiske reseptorer.
Fusjonsproteinet ble vist å være et funksjonelt protein som var i stand til å binde FITC-biotin når det ble analysert ved konfokal mikroskopi og «atomic force» mikroskopi. Utransduserte celler og celler transfektert med en retrovirusvektor inneholdende LacZ-genet, ble benyttet som kontroller. Det ble ikke påvist uspesifikk binding av biotinprober til LacZ-transduserte kontrollceller ved «atomic force» mikroskopi. Som forventet oppviste de transfekterte cellene spesifikk binding som var gjentatt målbar i ikke-fikserte prøver. De målte bindingskreftene utgjorde multipler av gjennomsnittet 149 ± 19 pN (gjennomsnitt ± sd) som, som også forventet, ligger innenfor området for den tidligere rapporterte biotin/streptavidin-bindingskraft på 160 pN (Florin et al., (1994), Science 264:415-417).
Konstruksjonens funksjonalitet kan også bekreftes in vivo ved å vise bindingen av fluorescens-merkede biotinmolekyler til celler som har fusjonsproteinkonstruksjonen, ved å benytte FACS-analyse.
Den funksjonelle aktivitet av fusjonsproteinet ble analysert in vivo i en malign rottegliom-modell. BT4C villtype gliomceller ble implantert intrakranialt i høyre corpus callosum i en dybde på 2,5 mm i hjernen til innavlede BDIX hunnrotter. Tumorveksten ble hyppig monitert med høyoppløsende MRI (magnetisk resonans-avbildning). 3 uker etter tumorcelle-inokuleringene ble pseudotypede retrovirus som var bærere av cDNA for fusjonsproteinet eller LacZ-genet i titere på henholdsvis 2 x 10<6> cfu/mL og 1,3 x 106 cfu/mL ført inn i tumoren, først i en dybde på 2,5 mm og deretter i en dybde på 1,5 mm, med et intervall på 10 minutter. Genoverføringen ble gjentatt etter to dagers vekst. Dyrene ble avlivet og perfusjons-fiksert med 4% PFA 3 dager etter den siste injeksjonen. Hjernene ble tatt ut og delt ved injeksjonsstedet i to koronale deler, snittet på is og analysert ved immunoreaktivitet mot anti-avidin-antistoff. Resultatene viste at fusjonsproteinet ble uttrykt in vivo i malignt rottegliom. Protein ble påvist i gliomceller og i ring-lignende strukturer, som lignet vaskulære endotelceller i tumor-blodkar.
Claims (6)
1. Protein for å målrette bitinylerte molekyler mot spesifikke seter i vev, karakterisert ved at det omfatter et membran-omspennende domene av en endocytotisk reseptor og et ekstracellulært domene, som omfatter biotin-bindende aktivitet.
2. Protein ifølge krav 1, karakterisert ved at det videre omfatter et cytoplasmatisk domene.
3. Protein ifølge krav 1 eller krav 2, karakterisert ved at det ekstracellulære domenet omfatter en biotin-bindende domene av avidin eller streptavidin.
4. Protein ifølge et hvilket som helst av de foregående krav, karakterisert ved at reseptoren er scavenger-reseptor klasse A.
5. Protein ifølge krav 1, karakterisert ved at det omfattter SEKV. ID. NR:2.
6. Nukleinsyremolekyl for å målrette biotinylerte molekyler mot spesifikke seter i vev, karakterisert ved at det koder for et protein i henhold til et hvilket som helst av kravene 1 til 5.
