Abstract
Description
本発明は、ターゲット細胞へのウイルスのトランスダクション効率の改善において使用するためのペプチド及び組成物に関する。
遺伝子治療アプローチは、多くの場合、リコンビナントウイルスベクターによるターゲット細胞の低いトランスダクション効率によって妨げられる。レトロウイルスベクター、特にヒト免疫不全ウイルス1(HIV−1)ベースのレンチウイルスベクター(LV)は、遺伝子治療のための有望なビヒクルである(D'Costa et al., 2009)。これらベクターは、現在、様々な疾患、例えば免疫不全、神経変性性又は神経学的疾患、貧血症、HIV感染症を処置するための臨床用途において使用されている。レトロウイルスベクターの用途のいくつかは、ex vivoにおける特定のターゲット細胞、例えばCD34マーカーを発現する造血幹/前駆細胞のトランスダクションに依拠する。リコンビナントレンチウイルス粒子の使用に伴う制限要因は、リコンビナントレンチウイルスベクター粒子の生産中に高感染力価を得る能力である。この制限を回避する1つの方法は、精製工程中にウイルス上清を濃縮することである(Rodrigues et al., 2007)。しかしながら、GALVTR−LV(両種性マウス白血病ウイルス(MLV−A)エンベロープ糖タンパク質の細胞質尾部に融合されたテナガザル白血病ウイルスエンベロープ糖タンパク質でシュードタイピングされたLV(Sandrin et al., 2002))の場合のように、ウイルス粒子をシュードタイピングするために使用されるエンベロープ糖タンパク質に応じて、いくつかのLVについては、精製プロトコールは確立困難である。したがって、多くのレンチウイルスベクター調製物は低い力価を有し、トランスダクション効率は限られている。別の制限要因は、ターゲット細胞に感染するレンチウイルスベクターそれ自体の能力である。いくつかのエンベロープ糖タンパク質、例えばVSV−G、RD114TR、GALVTRは、レンチウイルスベクターをシュードタイピングするために使用され得、ターゲット細胞、例えばCD34+細胞に対する不定の感染性を有する(Sandrin et al., 2002)。これらの制限を回避する1つの戦略は、トランスダクションプロトコールを最適化する補因子、例えばカチオン性ポリマー(例えば、ポリブレン)又はフィブロネクチンフラグメント(例えば、レトロネクチン)を追加することである(Davis et al., 2004; Pollok et al., 1999)。米国特許第7,759,467号には、レトロウイルスによる造血細胞のトランスダクション効率を増加させるための方法であって、フィブロネクチン又はフィブロネクチンフラグメントの存在下における該細胞の感染を含む方法が記載されている。しかしながら、少なくとも2つの理由により、提案されている方法は全体的に満足なものではない。第1に、レトロウイルスの効率を改善するために使用されるフィブロネクチンフラグメントは、通常、約270個以上のアミノ酸を含むので、大きな経済的欠点を示す。さらに、フィブロネクチン又はフィブロネクチンフラグメントの使用は、培養プレートをコーティングし、ウイルス上清を固定化フィブロネクチンフラグメント上にプレロードすることを必要とする。これら2つの工程は標準化困難であり、使用されるフィブロネクチンフラグメント及びウイルス上清の濃度に応じて、ターゲット細胞のトランスダクションのある程度の飽和をもたらし得る(Novelli et al., 1999)。

本発明のペプチドは、細胞のウイルス感染を促進する。本明細書で使用される「ウイルス」は、天然に存在するウイルス及び人工ウイルスに関する。例えば、パラミクソウイルス(例えば、呼吸器合胞体ウイルス、麻疹ウイルス)、オルトミクソウイルス(例えば、インフルエンザウイルス)、フラビウイルス(例えば、C型肝炎ウイルス)、ヘパドナウイルス(例えば、B型肝炎ウイルス)、ラブドウイルス(例えば、狂犬病、VSV)、コロナウイルス(例えば、SARS)、トガウイルス(例えば、シンドビスウイルス、チクングニヤウイルス)、フィロウイルス(例えば、エボラウイルス)、アレナウイルス、ポックスウイルス、ヘルペスウイルス、ブンヤウイルス、ボルナウイルス、アルテリウイルス、バキュロウイルス、パルボウイルス、例えばアデノ随伴ウイルス。特定の実施態様によれば、ウイルスは、例えば、カーゴ、例えば治療、診断又は任意の他の目的に有用な(例えば、ターゲット細胞内において機能的研究を行うために有用な)カーゴを含み得る人工ウイルスである。例示的なカーゴとしては、核酸カーゴ、例えば産物、特に遺伝子治療産物(例えば、タンパク質又はRNA、例えばアンチセンスRNA又はshRNA)又は診断産物をコードするDNA又はRNA配列が挙げられる。さらに、本発明によれば、ウイルスは、カーゴ、例えばタンパク質カーゴ、核酸カーゴ(例えば、DNA又はRNAカーゴ)、診断カーゴ又は薬物カーゴをさらに含有し得るウイルス様粒子(又はVLP)であり得る。本発明との関連では、ウイルスは、エンベロープウイルス又は非エンベロープウイルスのいずれかである。アデノ随伴ウイルス(又はAAV)などのパルボウイルスは、例示的な非エンベロープウイルスである。好ましい実施態様では、ウイルスは、エンベロープウイルスである。好ましい実施態様では、ウイルスは、レトロウイルス、特にレンチウイルスである。本発明者らは、本発明のペプチドが、水疱性口内炎ウイルス(VSV−G)、改変ネコ内因性レトロウイルス(RD114TR)及び改変テナガザル白血病ウイルス(GALVTR)由来のエンベロープ糖タンパク質(GP)でシュードタイピングされたHIV−1由来レンチウイルスベクター(LV)による真核細胞の感染を促進し得ることを示した。本発明者らは、本発明のペプチドが、アデノ随伴ウイルスを含むパルボウイルスなどの他のウイルスの侵入を効率的に促進することをさらに示し、それにより、それらの広範な効率を実証した。本発明のペプチドを用いて得られたトランスダクションの効率と、開示されている実験において使用したウイルスベクター及びGPの多様性とを考慮すると、本ペプチドは、真核細胞におけるエンベロープウイルス及び非エンベロープウイルスのトランスダクション効率を増加させるための一般的手段として使用され得ることが明らかである。したがって、本発明のペプチドは、多くの他のウイルス若しくは他の偽型ウイルス、例えばウイルスエンベロープ糖タンパク質(Levy, 2015)、例えば両種性マウス白血病ウイルスGP(A−MLV)若しくは改変ヒヒ内因性ウイルスGP(BaEVTR)でシュードタイピングされたレンチウイルスによる、又は非エンベロープウイルス、例えばパルボウイルス、特にAAV、例えばリコンビナントAAVベクターによる真核細胞の感染を促進し得ることも予想される。

ペプチド及び試薬
標準的なフルオレニルメチルオキシ−カルボニルクロリド固相ペプチド合成によって、Vectofusin−1、Tat−Scrambled、Tat−Beclin1及びその誘導体を生産し、分取逆相HPLCによって精製し、HPLC及び質量分析法によって分析した(Genecust, Dudelange, Luxembourg)。7−アミノ−アクチノマイシンD(7−AAD)、ポリブレン、硫酸プロタミン及びTritonX−100は、Sigma-Aldrich (St-Quentin-Fallavier, France)から入手した。
10%熱不活性化ウシ胎児血清(FCS)(Life Technologies, St-Aubin, France)を補充したダルベッコ改変イーグル培地中で、ヒト結腸直腸ガン由来HCT116細胞(CCL-247; ATCC, Manassas, VA)及びヒト胚腎臓HEK293T細胞(Merten, 2011)を37℃、5%CO2で培養した。
以前に記載されているように(Fenard, 2013)、LVを作製した。簡潔に言えば、4つのプラスミド:gagpol(pKLgagpol)及びrev(pKrev)発現プラスミド、トランスファープラスミド(pCCLsin.cPPT.hPGK.eGFP.WPRE)並びにVSV−G(pMDG)エンベロープ糖タンパク質(GP)、GALVTR GP(pBA.GALV/Ampho−Kana)又はRD114TR GP(pHCMV−RD114TR)のいずれかをコードするプラスミド.によるリン酸カルシウムトランスフェクションを使用して、HEK293T細胞を一過性トランスフェクションする。24時間の産生後、生ウイルス上清を採取し、ろ過し(0.45μm)、−80℃で凍結した。いくつかの膜ベースの工程及びクロマトグラフィー工程によるGFP発現VSV−G−LVの精製が以前に記載されている(Merten, 2011)。市販のELISAキット(Perkin Elmer, Courtaboeuf, France)を使用してHIV−1 p24カプシド含有量を測定することによって、物理的粒子力価を決定した。フローサイトメトリー(FACSCalibur, BD Biosciences, Le Pont de Claix, France)によるGFPの検出(力価は、1ミリリットル当たりのトランスダクション単位(TU/ml)として表される)を使用して、又はQ−PCR(力価は、1ミリリットル当たりの感染ゲノム(ig/ml)として表される)を使用して、HCT116細胞において、感染力価を決定した(Merten, 2011)。
レンチウイルストランスダクションでは、48プレート中、培養添加物の非存在下又は存在下、所望の量のレンチウイルスベクター上清でHCT116細胞を6時間トランスダクションした。次に、新鮮培地中で細胞をインキュベーションし、さらに3〜4日間培養した。
French Ministry of Research and Higher Studiesへの申告N° DC-201-1655の下、国際倫理原則及びフランスの国内法(生命倫理法n°2011-814)にしたがって、Centre Hospitalier Sud Francilien, Evry, Franceにおいて、合併症を伴わない出産後にインフォームドコンセントを得て、臍帯血(UCB)サンプルを収集した。免疫磁気選択(Miltenyi Biotec, Paris, France)によって、ヒトCD34+細胞を単離した。以前に記載されているように(Ingrao et al., 2014)、hCD34+細胞の予備活性化を一晩実施した。予備活性化細胞を96ウェルプレートにプレーティングし、目的のペプチドと混合した又は混合していない所望の量のLV粒子を追加することによって、トランスダクションを開始した。トランスダクションの6時間後、分化培地を各ウェルに追加することによって、反応物を希釈した。4〜6日後、7−AAD標識及びGFP発現の測定によって、細胞死亡率及びトランスダクション効率をそれぞれ評価した。
以前に記載されているプロトコール(Yolamanova, 2013)から、培養添加物の存在下におけるLV粒子のプルダウンを適合させた。簡潔に言えば、室温で平衡化したX-Vivo20培地で、VSV−G−LV上清を100ng/ml p24に希釈した。次に、10μMの示されている培養添加物の非存在下又は存在下で、LV懸濁液 500μlを1.5mlチューブにロードした。ホモジナイズした後、サンプルを低速(15,000g)、室温で5分間遠心分離した。次いで、上清を廃棄し、ペレットを新鮮培地 100μlに懸濁し、−20℃で凍結した。各条件について、上記のように市販のHIV−1 p24 ELISAキットを使用して、ペレット化p24の量を評価した。
以前の研究(Fenard, 2013)から、ターゲット細胞に対するLV接着のプロトコールを適合させた。簡潔に言えば、Tat−Scrambled又はTat−Beclin1(5μM)の非存在下又は存在下で、HCT116細胞を37℃で30分間プレインキュベーションした。次いで、培養添加物の非存在下又は存在下で、細胞をウイルス上清と共に4℃でさらに2.5時間インキュベーションした。次に、冷PBS1×で細胞を3回洗浄し、1%TritonX−100及びプロテアーゼカクテル阻害剤Complete (Roche diagnostics, Meylan, France)を含有するPBS1×で溶解した。市販のHIV−1 p24 ELISAキットを使用して溶解物中のp24含有量を評価し、DCタンパク質アッセイ(Biorad, Ivry-sur-Seine, France)を使用して全タンパク質含有量に対してデータを正規化した。ウイルス融合アッセイ(BLAM−LV)アッセイについて、プロトコールは以前に詳細に記載されていた(Ingrao et al., 2014)。
製造業者の説明書にしたがって、Methocult (H4434, Stemcell Technologies)1ミリリットル当たりトランスダクション細胞又は非トランスダクション細胞 500個をプレーティングすることによって、CFCアッセイを2回反復で実施した。2週間の培養後、標準的な視覚的基準を用いて倒立顕微鏡を使用して、赤芽球バースト形成単位(BFU−E);顆粒球−単球コロニー形成単位(CFU−GM)及び顆粒球−赤血球−マクロファージ−巨核球コロニー形成単位(CFU−GEMM)をカウントした。
一過性リン酸カルシウムトランスフェクション法を使用して、pBABE−puro−mCherry−eGFP−LC3発現プラスミドでHEK293T細胞をトランスフェクションした。Tat−Beclin1(5μM)の非存在下若しくは存在下で、又はアール平衡塩類溶液(EBSS)中で、細胞(細胞 5×10E5個/ウェル)を37℃、5%CO2で6時間インキュベーションした。次に、PBS1×で細胞をで洗浄し、固定し(1.2%PFA)、ImageStream (Amnis corporation, Seattle, WA, USA)を使用して分析した。感度を最適化するために40倍対物レンズ及び最低流速を使用して、明視野チャネル、mCherry蛍光チャネル及び最後にSSCチャネル(742nm)で、細胞の画像を取得した。取得したら、Ideas(登録商標)ソフトウェアを用いて、画像を処理した。ヒストグラムで焦点画像をゲートし、40〜90の明視野チャンネルでGradient_RMS特徴値を示した。次いで、(明視野チャンネルの)面積対アスペクト比の散布図を使用して、単一細胞をゲートし、細胞のダブレットを除去した。単一細胞をゲートしたら、Ideas(登録商標)ソフトウェアに含まれる自動スポットカウントウィザードを使用して、各細胞上のスポットの数をmCherryチャネルでカウントした。テキストファイルとしてデータをエクスポートし、GraphPad Prismを用いて処理した。
低用量のTat−Beclin1は、様々な偽型による細胞株のレンチウイルストランスダクションを強く改善した。
レンチウイルストランスダクションに対するTB1(配列番号:75)ペプチドの効果を評価するために、低用量又は高用量のTB1又はコントロールペプチドTat−Scrambled(TS)存在下、広く使用されているVSV−G糖タンパク質エンベロープ(VSV−G−LV)でシュードタイピングしたレンチウイルスベクター(LV)でHCT116細胞をトランスダクションした(図1A)。興味深いことに、低用量のTB1の使用は、レンチウイルストランスダクションを強く改善した(10倍)。さらに、1対数濃度のVSV−G−LV、10E5〜10E6 TU/ml(MOI 0.5〜5に相当)では、TB1効果は飽和せず、最大84%のトランスダクション効率に達した(図1B)。
基礎研究のために細胞株のレンチウイルストランスダクションを促進することは興味深いが、別の関心対象は、ex vivo遺伝子治療アプローチのために、造血幹/前駆細胞(HSPC)をターゲティングするレンチウイルストランスダクションの現在の臨床プロトコールを最適化することである。図2Aに示されているように、高精製VSV−G−LV粒子を使用して、ヒトCD34+HSPCをトランスダクションした。TB1の存在下では、レンチウイルストランスダクションの2倍の増加が観察された。HSPCのレンチウイルストランスダクションを促進するためのTB1の最適用量は、約10μMと定義された(図2A〜B)。
ターゲット細胞へのLVの侵入は律速段階である。したがって、本発明者らは、低用量のTB1が、LVとターゲット細胞膜との接着及び融合を増強することができるかを試験することを決定した。そのために、BLAM−LVアッセイを使用した(Ingrao, 2014)。図3Aに示されているように、細胞株(HCT116)又はhCD34+HSPCのような関連一次細胞のいずれかを用いた場合、TB1は、ウイルス融合段階を強く増加させた。次に、4℃でターゲット細胞と相互作用するウイルス粒子の数を定量することによって、本発明者らは、TB1の存在下では、ウイルス接着のレベルが、Vectofusin−1を使用したコントロール条件と同程度のレベルまで強く増加することを示した(図3B)。しかしながら、ウイルスプルダウンアッセイを使用して、本発明者らは、ウイルス接着のこの増加が、Vectofusin−1(ナノフィブリルペプチド)の場合のようにウイルス粒子のTB1誘導性凝集の結果ではないことを示した(図3C)。したがって、TB1ペプチドは、ウイルス粒子の凝集を伴わない分子機構を介して、ウイルス接着及び融合段階を増加させることができる。
レンチウイルストランスダクションに対するTB1の作用の特異性を評価するために、Tatトランスダクションドメイン(Tat)及び267−284改変beclin1ドメイン(Bec)を合成し、VSV−G−LVによるHCT116細胞のトランスダクション中に試験した。図4に示されているように、Tat及びBecペプチドはいずれも、レンチウイルストランスダクションを促進することができない。したがって、レンチウイルストランスダクションに対するTB1のポジティブな効果は、Tatの細胞透過性ペプチド(CPP)活性によるものではない。また、Beclin1ドメインは、トランスダクションドメインに連結されてその効果を発揮するが、これは、Shoji-Kawataの研究に記載されるGAPR−1タンパク質との特異的相互作用を確かに介して、又はまだ同定されていない別の細胞パートナーとの相互作用を介して、それが細胞内で作用していることを示唆している。
アデノ関連ベクター(AAV)は、遺伝子治療アプローチのための最も頻繁に使用されるウイルスベクターである。異なる血清型の中でも、AAV−8は臨床使用が成功しており、肝臓特異的遺伝子治療にかなり有望である(Nathwani et al, 2014)。したがって、AAV8トランスダクション効率に対するTB1の効果をin vitroで評価した。図6に示されているように、TB1は、リコンビナントAAV8の感染性を用量依存的に増強することができるが、Tat−Scrambledコントロールペプチドは効果を有しない。この結果は、TB1の作用が広範囲であり、エンベロープウイルスベクター及び非エンベロープウイルスベクターの両方に作用することを強調している。
TB1の作用機構をより良く理解するために、本発明者らは、レンチウイルストランスダクションの増加がオートファジープロセスの活性化の結果であるかを調査しようとした。単一細胞レベルでオートファジーをモニタリングするために、mCherry−eGFP−LC3融合タンパク質を発現するプラスミドでHEK293T細胞をトランスフェクションした。次に、細胞をTB1(5μM)又はEBSS飢餓溶液と共にインキュベーションし、イメージングフローサイトメーターを用いて分析した。予想通り、EBSSは、1細胞当たりのオートファゴリソソーム(mCherryスポット)の数を増加させている(図7)。反対に、TB1処置HEK293T細胞におけるオートファゴリソソームの数は、ペプチドの非存在下(なし)におけるコントロール条件と同程度である。これらのデータは、TB1の存在下で観察されるレンチウイルストランスダクションの改善が確かに、オートファジーフラックスの誘導の結果ではないことを示唆している。
ヒトBeclin1及びBeclin2タンパク質の配列アラインメントは、ECDドメインにおける相同性を示している。したがって、本発明者らは、Tat−Bec2WTペプチド(Tat(47−57)トランスダクションペプチドとヒトBeclin2 ECD249−266ドメインとの融合物)を設計することを決定した(図8A)。TB1は3つの突然変異(H275E、S279D及びQ281E)を含有するので、Tat(47−57)ペプチドと野生型ヒトBeclin1 ECD267−284ドメインとの融合物に相当するTat−BecWTペプチドも設計し、配列アラインメントに示されている(図8A)。広範囲の濃度にわたって、レンチウイルス産生を促進する能力について、これら3つのペプチドを試験した。図8Bに示されているように、前記ペプチドは全て、レンチウイルストランスダクションを効率的に促進することができるが、それらの最適用量(TB1の場合には3μM、Tat−BecWTの場合には1μM及びTat−Bec2WTの場合には500nM)にはかなりの変動がある。100nMのTat−Bec2WTのみがレンチウイルストランスダクションを3倍(10から30%に)増加させることができ(8B)、この濃度では、TB1及びTat−BecWTは効果を有しない。Tat−Scr2WT(Tat−Bec2WTのスクランブルバージョン)は、レンチウイルストランスダクションを促進しているが、Tat−Bec2WTに対する効果の程度の減少に相当する用量では、確かにTat依存的活性である。結論として、Tat−Bec2WTは、非常に低用量で、レンチウイルストランスダクションの強力なエンハンサーである。
Claims (37)
- (i)CPP部分;及び
(ii)14〜22個のアミノ酸残基長を有するBeclin由来ペプチド部分
を含むペプチドであって、ただし、配列番号:75、配列番号:77及び配列番号:98〜101に示されているペプチドではない、ペプチド。 - CPP部分が、
−ヒト免疫不全ウイルス(HIV)1型の転写トランス活性化因子(tat)ペプチド由来のペプチド;又は
−MAPペプチド
である、請求項1に記載のペプチド。 - CPP部分が、配列番号:1〜配列番号:62、特に配列番号:1〜配列番号:5からなる群より選択され、より具体的にはCPP部分が、配列番号:1及び配列番号:4から選択される、請求項1又は2に記載のペプチド。
- Beclin由来ペプチド部分が、Beclin−1由来ペプチド部分、例えばBeclin−1の配列若しくはBeclin−1のβ1シート領域を含む前記配列の機能的誘導体、又はBeclin−2由来部分、例えば配列番号:89のアミノ酸配列若しくはその機能的変異体を含むBeclin−2由来部分である、請求項1〜3のいずれか一項に記載のペプチド。
- Beclin−1由来ペプチド部分が、配列番号:65に示されているアミノ酸配列又はその機能的変異体を含む、請求項4に記載のペプチド。
- Beclin−1由来ペプチド部分が、配列番号:65〜73からなる群において選択される配列を含む、請求項4又は5に記載のペプチド。
- CPP部分及びBeclin由来ペプチド部分が、リンカー、例えばペプチドリンカー、より具体的にはジペプチドリンカー、例えばGGジペプチドリンカーを介して互いに連結されている、請求項1〜6のいずれか一項に記載のペプチド。
- ペプチドが、配列番号:74、配列番号:76、配列番号:78〜83及び配列番号:90〜91のいずれか1つに示されている配列を含むか、又はそれからなる、請求項1〜7のいずれか一項に記載のペプチド。
- ウイルス又はウイルスベクターによる細胞のトランスダクションを促進するためのin vitro方法であって、該細胞と、該ウイルスと、
−細胞透過性ペプチド(CPP)部分;及び
−Beclin由来ペプチド部分
を含むペプチドとを接触させることを含む、方法。 - ウイルスがレトロウイルス、特にレンチウイルスである、請求項9に記載の方法。
- レンチウイルスが偽型のレンチウイルスである、請求項10に記載の方法。
- ウイルスがパルボウイルス、特にアデノ随伴ウイルス(AAV)である、請求項9に記載の方法。
- 細胞が造血幹/前駆細胞、好ましくはhCD34+細胞である、請求項9〜12のいずれか一項に記載の方法。
- 別のウイルストランスダクションエンハンサー、例えばベクトフシン(vectofusin)、ヒトフィブロネクチンフラグメント、ウイルス感染の様々な精液由来エンハンサー又はHIV−1エンベロープ糖タンパク質由来のペプチドを接触させることをさらに含む、請求項9〜13のいずれか一項に記載の方法。

- 請求項1〜8に記載のペプチドをコードするポリヌクレオチドを含む、核酸構築物。
- 1つ以上のコントロール配列をさらに含む、請求項26に記載の核酸構築物。
- 請求項1〜8に記載のペプチド又は配列番号:75及び配列番号:77に示されているアミノ酸配列を含むか若しくはそれからなるペプチドと、ウイルスベクターとを含む、キット。
