Oppfinnelsens område
Foreliggende oppfinnelse angår nye forbindelser, farmasøytiske preparater som omfatter forbindelsene, fremgangsmåter for dets fremstilling så vel som anvendelsen av forbindelsene for fremstilling av et medikament som spesielt virker på sentralnervesystemet.
Oppfinnelsens bakgrunn
Mange sykdommer i sentralnervesystemet påvirkes av det adrenerge, dopaminerge og serotonerge neurotransmittersystem. Serotonin er for eksempel involvert i en rekke sykdommer og tilstander som har sin opprinnelse i sentralnervesystemet. Flere farmakologiske og genetiske undersøkelser som omfatter serotoninreseptorer viser at 5-HT2C-reseptor subtypen i stor grad er involvert i reguleringen av matinntak (Obes. Res. 1995, 3, Suppl. 4,449A-462S). 5-HT2C-reseptorsubtypen transkriberes og uttrykkes i strukturer i hypotalamus som er forbundet med appetittregulering. Det er påvist at 5-HT2C-reseptoragonisten m-klorfenylpiperazin (mCPP) som til en viss grad prefererer 5-HT2C-reseptoren reduserer matinntaket hos mus som uttrykket den normale 5-HT2C-reseptor, mens forbindelsene ikke har virkning hos mus som uttrykker den muterte, inaktive form av 5-HT2c-reseptoren (Nature 1995, 374, 542-546). I et nylig utført klinisk forsøk ble det oppnådd en liten, med medvarende reduksjon av kroppsvekt hos overvektige pasienter etter 2 ukers behandling med mCPP (Psychopharmacology 1997, 133, 309-312). Nylig ble en rekke porrolo[3,2,l-ij]kinolinderivater identifisert som 5-HT2c-reseptoragonister med selektivitet for 5-HT2a-reseptoren (Isaac M., et al., Bioorg. Med. Chem. Lett. 2000,10,919-921). Det hevdes at forbindelsene innebærer en ny tilnærmingsmåte til behandling av overvekt og epilepsi.
Andre kliniske forsøk rapporterer også om vektreduksjon ved bruk av andre "serotonerge" midler (se for eksempel IDrugs 1998,1,456-470). For eksempel har 5-HT-reopptaksinhibitoren fluoksitin og 5-HT-frigjøringsmiddelet/reopptaksinhibitoren deksfenfluramin medført vektreduksjon i kontrollerte forsøk. Dagens tilgjengelige legemidler som øker den serotonerge transmisjon synes imidlertid å ha bare moderat og i noen tilfeller kortvarig virkning på kroppsvekten.
Det er også blitt foreslått at 5-HT2c-reseptorsubtypen er involvert i CNS-forstyrrelser som depresjon og angst (Exp. Opin. Invest. Drugs 1998,7,1587-1599; IDrugs, 1999, 2, 109-120).
Det er videre blitt foreslått at 5-HT2C-reseptorsubtypen er involvert i urinveislidelser som urininkontinens (IDrugs, 1999,2,109-120).
Forbindelser som har selektiv virkning på 5-HT2C-reseptoren kan derfor ha et terapeutisk potensiale for behandling av forstyrrelser som de som er nevnt ovenfor. Selvfølgelig reduserer også selektivitet risikoen for bivirkninger som er mediert av andre serotoninreseptorer.

I WO 99/03833 beskrives arylpiperazinderivater som er 5-HT2-antagonister og 5-HTia-reseptoragonister og derfor er anvendelige som hjelpemidler eller forebyggende midler ved psykonevrose.
I WO 96/02525 beskrives arylpiperazin-avledede piperazid-derivater som har antagonistisk virkning på 5-HT-reseptoren.
I WO 99/58490 beskrives aryl-hydronaftalen-alkanaminer som kan medføre delvis eller fullstendig blokkering av serotonerge 5-HT2c-reseptorer i en organisme.
Det er et formål med foreliggende oppfinnelse å tilveiebringe nye forbindelser.
Et annet formål med oppfinnelsen er et farmasøytisk preparat som omfatter som aktiv forbindelse, forbindelser for bruk i terapi.
Dessuten er et formål med oppfinnelsen en metode for å behandle eller forebygge en serotonin-relatert sykdom, særlig en sykdom som er relatert til 5-HT2C-reseptoren.
Sammendrag av oppfinnelsen
I henhold til oppfinnelsen tilveiebringes nye forbindelser med den generelle formel (I):

n = 1-3, hvor n er antall metylengrupper. n er fortrinnsvis 1, og har den betydning at 2 oksygenatomer i formel (I) er adskilt av en CH2CH2-gruppe.
og farmasøytisk akseptable salter, solvater, hydrater, geometriske isomerer, tautomerer, optiske isomerer eller N-oksider derav.
Gruppen -(CH2N(Rs)(R6) kan være bundet til orto-, meta- eller para-stillingen i fenylringen, sett i forhold til sidekjeden alkylendioksy, fortrinnsvis i meta-stilling.
n i formel (I) er 1-3, hvor n er antall metylengrupper. n er fortrinnsvis 1, og har den betydning at 2 oksygenatomer i formel (I) er adskilt av en CH2CH2-gruppe.
Det er videre foretrukket at R5 og R^ sammen med nitrogenatomet de er bundet til danner en ring valgt fra pyrrolidin, piperazin og morfolin.
I en utførelsesform er forbindelsene ifølge foreliggende oppfinnelse valgt fra gruppen bestående av: 2-(l-piperazinyl)-3-{2-[3-(4-morfolinylmetyl)fenoksy]etoksy}pyrazin; 2-(l-piperazinyl)-3-{2-[3-(l-pyrrolidinylmetyl)fenoksy]etoksy}pyrazin; 2-(l-piperazinyl)-3-{2-[3-(4-metyl-l-piperazinylmetyl)fenoksy]etoksy}pyrazin; 2-( 1 -piperazinyl)-3- {2-[3- {[2-metoksyetyl)amino} metyl)fenoksy]etoksy} pyrazin; og 2-(l-piperazinyl)-3-{2-[3-{[isopropylamino)metyl}fenoksy]etoksy}pyrazin;
og deres farmakologisk akseptable salter og solvater.
I de tilfeller forbindelser med formel (I) kan foreligge i form av optiske isomerer, omfatter oppfinnelsen den racemiske blandingen så vel som de individuelle enantiomerer som sådan.
I de tilfeller forbindelser med formel (I) inneholder grupper som kan foreligge i tautomere former, omfatter oppfinnelsen de tautomere former av forbindelsene, så vel som blandinger derav.
I de tilfeller forbindelsene med formel (I) kan foreligge i form av geometriske isomerer, omfatter oppfinnelsen de geometriske isomerer, så vel som blandinger derav.
I et annet aspekt tilveiebringer denne oppfinnelsen en fremgangsmåte for å fremstille en forbindelse ifølge denne oppfinnelsen. Fremgangsmåten omfatter å omdanne en forbindelse med formel (II):

Forbindelsene i henhold til formel (I) ovenfor er egnet for bruk i terapi.
Et annet aspekt ved oppfinnelsen tilveiebringer et farmasøytisk preparat som omfatter en forbindelse i henhold til formel (I) ovenfor som aktiv forbindelse, fortrinnsvis sammen med en farmasøytisk akseptabel bærer og, om ønsket, andre farmasøytisk aktive midler.
Et annet aspekt ved oppfinnelsen tilveiebringer anvendelsen av forbindelsene i henhold til formel (I) ovenfor for fremstilling av et medikament for behandling av en serotoninrelatert sykdom, spesielt 5-HT2c-reseptor-relatert, særlig spiseforstyrrelser, spesielt overvekt; hukommelsesforstyrrelser, schizofreni, forstyrrelser i stemningsleie, angstlidelser, smerte, stoffmisbruk, seksuelle dysfunksjoner, epilepsi og urinveislidelser.
Detaljert beskrivelse av oppfinnelsen
I henhold til foreliggende oppfinnelse er det utviklet en klasse av nye forbindelser som bindes til 5-HT2c-reseptoren (agonister og antagonister) og som derfor er anvendelige ved behandling av serotonin-relaterte forstyrrelser.
Først vil vi forklare de forskjellige begreper som er benyttet, separat og i kombinasjoner, i den ovennevnte definisjon av forbindelsene med den generelle formel
(I).
Med "heteroatom" menes nitrogen, oksygen, svovel og i heteroaromatiske ringer også selenium.
Begrepet "aryl" omfatter fenyl, 1-naftyl og 2-naftyI.
Begrepet "heteroaryl" omfatter 5- og 6-leddet heteroaromatiske ringer som pyrrol, imidazol, tiofen, furan, selenofen, tiazol, isotiazol, tiadiazol, oksazol, isoksazol, oksadiazol, pyridin, pyrazin, pyrimidin, pyridazin, pyrazol, triazol og tetrazol.
Ci-6-alkyl som kan være rettkjedet eller forgrenet, er fortrinnsvis Ci_4-alkyl. Eksempler på alkylgrupper omfatter metyl, etyl, propyl, isopropyl, butyl, sec-butyl, tert-butyl, pentyl, isopentyl, heksyl og isoheksyl.
Ci.4-alkoksy kan være rettkjedet eller forgrenet. Eksempler på alkoksygrupper omfatter metoksy, etoksy, propoksy, isopropoksy, butoksy, sec-butoksy og tert-butoksy.
C2-4-alkenyl kan være rettkjedet eller forgrenet. Eksempler på alkenylgrupper omfatter vinyl, 2-propenyl og l-metyl-2-propenyl.
Ci-C4-alkoksy-C2-C4-alkyl kan være rettkjedet eller forgrenet. Eksempler på grupper omfatter 2-(metoksy)etyl, 3-metoksy-l-propyl, 4-etoksy-l -butyl og lignende.
Eksempler på aryl-Ci-C2-acyl omfatter benzoyl og fenylacetyl. Eksempler på heteroaryl-Ci-C2-acyl omfatter nikotinoyl og 3-pyridinylacetyl og lignende.
C2.4-acyl kan være mettet eller umettet. Eksempler på acylgrupper omfatter acetyl, propionyl, butyryl, isobutyryl og butenoyl (for eksempel 3-butenoyl).
Halogen omfatter fluor, klor og brom.
Når det er oppgitt ovenfor at aryl- og heteroarylrester kan være substituert, vises det til aryl og heteroaryl per se, så vel som hvilke som helst kombinert i grupper som inneholder aryl- eller heteroarylrester, slik som heteroaryl-Ci-C2-alkyl og aryl-Ci-C2-acyl.
Begrepet "N-oksider" betyr at et eller flere nitrogenatomer, når de foreligger i en forbindelse, er i N-oksidform (N-»0).
Begrepet "prodrug-former" betyr et farmakologisk akseptabelt derivat, som en ester eller et amid, hvor derivatet biotransformeres i kroppen og danner det aktive legemiddelet. Det vises til Goodman og Gilman's, The Pharmacological basis of Therapeutics, 8. utg., McGraw-Hill, Int. Ed. 1992, Biotransformations of Drugs, s. 13-15.
"Farmasøytisk akseptabel" betyr å være anvendelig ved fremstilling av et farmasøytisk preparat som vanligvis er trygt, ikke-toksisk og hverken biologisk eller på annen måte uønsket, og som omfatter å være anvendelig ved veterinær bruk så vel som human farmasøytisk bruk.
"Farmasøytisk akseptable salter" betyr salter som er farmasøytisk akseptable, som definert ovenfor, og som har den ønskede farmakologiske virkning. Slike salter omfatter syreaddisjonssalter som er dannet med organiske og uorganiske syrer som hydrogenklorid, hydrogenbromid, hydrogenjodid, svovelsyre, fosforsyre, eddiksyre, glykolsyre, maleinsyre, malonsyre, oksalsyre, toluensulfonsyre, metansulfonsyre, fumarsyre, ravsyre, vinsyre, sitronsyre, benzosyre, askorbinsyre og lignende.
Som nevnt ovenfor er forbindelsene ifølge foreliggende oppfinnelse anvendelige for behandling (inkludert forebyggende behandling) av serotonin-relaterte forstyrrelser, særlig 5-HT2c-reseptor-relaterte, hos et menneske eller et dyr (inkludert for eksempel kjæledyr), slik som spiseforstyrrelser, særlig overvekt; hukommelsesforstyrrelser slik som Alzheimers sykdom; schizofreni; forstyrrelser i stemningsleie inkludert, men ikke begrenset til alvorlig depresjon og bipolar depresjon, inkludert både mild og manisk bipolar forstyrrelse, vinterdepresjon (seasonal affective disorder = SAD); angstlidelser inkludert situasjonsbetinget angst, generelle angstlidelser, primære angstlidelser (panikklidelser, fobier, obsessiv-kompulsive forstyrrelser og post-traumatiske stressforstyrrelser) og sekundære angstlidelser (for eksempel angst forbundet med stoffmisbruk); smerte; stoffmisbruk; seksuelle dysfunksjoner; epilepsi og urinveislidelser, slik som urinveisinkontinens.
I radiomerket form kan forbindelsene ifølge foreliggende oppfinnelse anvendes som et diagnostisk middel.
Forbindelsene ifølge den generelle formel (I) ovenfor kan fremstilles ved hjelp av en fremgangsmåte ifølge denne oppfinnelsen eller analogt med en konvensjonell fremgangsmåte. Denne oppfinnelsen angår fremgangsmåte for å fremstille forbindelser med enhver formel heri, som omfatter å omsette hvilken som helst, en eller flere, av forbindelsene eller formlene som er skissert heri, inkludert hvilke som helst prosesser som er skissert heri.
Som vist i Skjema 1 kan for eksempel en forbindelse med formel (I) fremstilles ved
først å behandle en forbindelse med formel (II) hvor Hal er halogen og R3 og R4 er som definert ovenfor, med et passende piperazin med formel (III), hvor Ri er hydrogen eller en nitrogenbeskyttende gruppe, slik som trityl, benzyl eller tert-butoksykarbonyl, og R2 har samme betydning som i formel (I) for å tilveiebringe en forbindelse med formel (IV). Reaksjonen utføres i et løsningsmiddel slik som acetonitril, dioksan,
tetrahydrofuran (THF), n-butanol, N,N-dimetylformamid (DMF) eller i en blanding av løsningsmidler slik som DMF/dioksan, eventuelt i nærvær av en base slik som K2CO3, Na2C03, Cs2C03, NaOH, trietylamin, pyridin eller lignende, ved 0-200°C i 1-24 timer.
Forbindelsen med formel (IV) omsettes med en diol med formel (V), hvor n har samme betydning som formel (I), for å tilveiebringe intermediatet (VI). Reaksjonen utføres i et løsningsmiddel slik som dioksan, THF, DMF eller pyridin og lignende, i nærvær av en base slik som K-t-BuO, Na-t-BuO, NaH eller lignende ved 0-150°C i 1-24 timer.
Intermediat (VI) omsettes med en hydroksybenzaldehydforbindelse med formel (VII), hvor R7 har samme betydning som i formel (I), for å tilveiebringe aldehydintermediatet (VIII) . Reaksjonen kan utføres i nærvær av dietylazodikarboksylat (DEAD) eller 1,1'-azobis(N,N-dimetylformamid) (Tetrahedron Lett. 1995, 365, 3789-3792), fortrinnsvis DEAD og trifenylfosfin (PPh3) i et løsningsmiddel som THF eller diklormetan (Mitsunobu-reaksjon; se Org. React. 1992, 42, 335-656).
Når intermediat (VIII) gjennomgår en vanlig reduktiv alkyleringsprosedyre (slik som beskrevet i J. Org. Chem. 1996, 61, 3849-3862), med et passende amin med formel (IX) , hvor R5 og R<5, uavhengig av hverandre, er hydrogen, Ci-C4-alkoksy-C2-C4-alkyl, hydroksy-C2-C4-alkyl, Ci-C6-alkyl eller fenyl som kan være uavhengig substituert i en eller flere stillinger med Ci-4-alkoksy; eller R5 og Re danner sammen med nitrogenatomet de er bundet til en morfolin-, pyrrolidin- eller piperazinring, som kan være substituert med metyl; resulterer det i en forbindelse ifølge denne oppfinnelsen (I). innen teknikkens stand og kan for eksempel utføres ved fraksjonell krystallisering av salter med optisk aktive (kirale) syrer eller ved kromatografiseparering på kirale kolonner.
I henhold til foreliggende oppfinnelse kan forbindelsene med formel (I) i form av frie baser eller salter med fysiolgisk akseptable syrer innarbeides i egnede galeniske former slik som preparater for oral bruk, for injisering, for administrering i form av nesespray eller lignende, i samsvar med aksepterte farmasøytiske prosedyrer. Slike farmasøytiske preparater i henhold til oppfinnelsen omfatter en effektiv mengde av forbindelsene med formel (I) sammen med forenelige farmasøytisk akseptable bærermaterialer eller fortynningsmidler, som er velkjent innen teknikkens stand. Bærerne kan være ethvert inert materiale, organisk eller uorganisk, som er egnet for enteral, perkutan, subkutan eller parenteral administrering slik som vann, gelatin, gummi arabikum, laktose, mikrokrystallinsk cellulose, stivelse, natrium stivelsesglykolat, kalsiumhydrogenfosfat, magnesiumstearat, talkum, kolloid silikondioksid og lignende. Slike preparater kan også inneholde andre farmakologisk aktive midler og vanlige tilsetningsstoffer som stabilisatorer, fuktemidler, emulgatorer, smaksstoffer, buffere og lignende.
Preparatene i henhold til oppfinnelsen kan for eksempel fremstilles i fast eller flytende form beregnet for oral administrering, slik som tabletter, piller, kapsler, pulvere, siruper, miksturer ("elixirs"), dispergerbare granuler, "cachets", suppositorier og lignende i form av sterile løsninger, suspensjoner eller emulsjoner beregnet for parenteral administrering, sprayer, for eksempel nesespray, transdermale preparater, for eksempel plastre og lignende.
Som nevnt ovenfor kan forbindelsene ifølge oppfinnelsen anvendes ved behandling av serotonin-relaterte forstyrrelser hos et menneske eller et dyr, slik som spiseforstyrrelser, spesielt overvekt, hukommelsesforstyrrelser, schizofreni, forstyrrelser i stemningsleie, angstlidelser, smerte, stoffmisbruk, seksuelle dysfunksjoner, epilepsi og urinveislidelser. Dosestørrelse og frekvens av dosering av den aktuelle forbindelse vil variere avhengig av mange faktorer som omfatter virkeevnen til den aktuelle forbindelse, metabolsk stabilitet og virkningstid for forbindelsen, pasientens alder, kroppsvekt, generelle helsetilstand, kjønn, kosthold, administreringsmåte og tidspunkt for denne, ekskresjonshastighet, legemiddel kombinering, alvorlighetsgraden av tilstanden som skal behandles og pasienten som gjennomgår terapi. Den daglige dose kan for eksempel være i området fra ca. 0,01 mg til ca. 100 mg/kg kroppsvekt,
En fremstilt forbindelse med formel (I) kan omdannes til en annen forbindelse med formel (I) ved hjelp av fremgangsmåter som er velkjente innen teknikkens stand. Fremgangsmåtene som er beskrevet ovenfor kan utføres slik at det gir en forbindelse ifølge oppfinnelsen i form av en fri base eller som et syreaddisjonssalt. Et farmasøytisk akseptabelt syreaddisjonssalt kan fremstilles ved å oppløse den frie basen i et egnet organisk løsningsmiddel slik som eter eller en blanding av eter og metanol, å behandle løsningen med en syre i samsvar med vanlige prosedyrer for fremstilling av syreaddisjonssalter fra base-forbindelser. Eksempler på addisjonssaltdannende syrer er maleinsyre, fumarsyre, ravsyre, metansulfonsyre, eddiksyre, oksalsyre, benzosyre, saltsyre, svovelsyre, fosforsyre og lignende.
Forbindelsene med formel (I) kan ha ett eller flere kirale karbonatomer, og de kan
derfor oppnås i form av optiske isomerer, for eksempel som en ren enantiomer eller som en blanding av enantiomerer (racemat) eller en blanding som inneholder diastereomerer. Separering av blandinger av optiske isomerer for å oppnå rene enantiomerer er velkjente
administrert som en enkelt eller flere doser, for eksempel fra ca. 0,01 mg til ca. 25 mg hver. Vanligvis gis en slik dose oralt, men parenteral administrering kan også velges.
De spesifikke eksempler nedenfor skal anses kun som forklarende og ikke som begrensende for resten av beskrivelsen på noen som helst måte. Det antas at en fagkyndig på basis av beskrivelsen heri, uten ytterligere utdyping, kan utnytte foreliggende oppfinnelse i sin fulle utstrekning.
EKSEMPLER
Generelt
NMR-spektra ble registrert på et spektrometer av typen Bruker Advance DPX 400 MHz ved 25°C. Kjemiske skift er oppgitt i ppm i forhold til tetrametylsilan. LC/MS-data ble oppnådd ved hjelp av et HP 1100 hplc-system som var koblet til et Micromass platform-LC-massespektrometer som kjørte MassLynx. HPLC-detaljer er: Kolonne, Phenomenex Cl8 Luna, 30 x 46 mm ved 40 + 1°C. Elueringsgradient T = 0,95% (0,1% maursyre i vann) og 5% (0,1% maursyre i acetonitril), deretter en lineær gradient til T = 2,5 minutter, 5% (0,1% maursyre i vann) og 95% (0,1% maursyre i acetonitril), deretter ytterligere 1 minutt under disse betingelsene. Flow-hastighet for elueringsmiddelet var 2 ml/min. Deteksjon ble utført ved hjelp av UV-diodematrise ved vindu 210-400 nm. Alternerende APCI-massespektra for +ve- og -ve-ion ble registrert i løpet av de 3,5 minutter med scanning mellom 100 og 650 masseenheter. Høyoppløsnings MS ble oppnådd på et Micromass LCT-spektrometer. Løsningsmidler for fremkalling av tynnsjiktskromatografi på silika var di-isopropyleter eller etylacetat/lett petroleum-blandinger.
EKSEMPEL 1
2-(l-piperazinyl)-3-{2-[3-(4-morfolinylmetyl)fenoksy]etoksy}pyrazin Trinn 1: 2- klor- 3-( 4- tert- butoksvkarbonyl- l- piperazinvl) pvrazin Tittelforbindelsen ble fremstilt i henhold til fremgangsmåten som er beskrevet i WO 00/76984. En blanding av N-Boc-piperazin (11,47 g, 61,5 mmol), K2C03 (8,5 g, 61 mmol) og 2,3-diklorpyrazin (9,20 g, 61,7 mmol) i acetonitril (100 ml) ble omrørt ved 100°C i 40 timer. Reaksjonsblandingen ble konsentrert, oppløst i toluen, vasket med vann, tørket (MgS04) og konsentrert. Residuet ble renset med kromatografi på silikagel med toluen/EtOAc (7:3) som elueringsmiddel, hvilket ga 18,3 g (100%) av tittelforbindelsen.
HRMS m/z beregnet for Ci3H19N402 (M)<+> 298,1197, målt 298,1206.
Trinn 2: 2- r3-( 4- tert- butoksvkarbonvl- l- piperazinyn- 2- pvrazinvloksv1etanol Tittelforbindelsen ble fremstilt i henhold til fremgangsmåten som er beskrevet i WO 00/76984. KO-t-Bu (9,92 g, 103 mmol) ble tilsatt til en blanding av produktet oppnådd i Trinn 1 (18,14 g, 60,7 mmol) og etylenglykol (25 ml, 448 mmol) i pyridin (125 ml) ved 85°C. Reaksjonsblandingen ble omrørt i 15 timer og deretter helt i isvann og ekstrahert med toluen. Den organiske fase ble tørket (MgS04) og konsentrert. Residuet ble renset med kromatografi på silikagel med toluen/EtOAc (1:1) som elueringsmiddel, hvilket ga 16,9 g (85%) av tittelforbindelsen.
HRMS m/z beregnet for Ci5H24N404 (M)<+> 324,1798, målt 324,1784.
Trinn 3: tert- butvl- 4-| 3-[ 2-( 3- formylfenoksy) etoksv]- 2- pvrazinvll- 1 - piperazinkarboksylat
En løsning av forbindelsen som ble oppnådd i Trinn 2 ovenfor (1,5 g, 4,7 mmol) i tørr tetrahydrofuran (THF; 10 ml) ble behandlet med 3-hydroksybenzaldehyd (0,74 g, 6,06 mmol) og trifenylfosfin (1,59 g, 6,06 mmol). Denne løsningen ble omrørt ved romtemperatur og deretter behandlet med dietylazodikarboksylat (0,96 ml, 6,06 mmol) i tørr THF (5 ml). Etter 1 time viste tynnsjiktskromatografi noe gjenværende 2-[3-(4-tert-butoksykarbonyl-l-piperazinyl)-2-pyrazinyloksy]etanol. Reaksjonsblandingen ble oppvarmet ved tilbakeløp under nitrogen i 5 timer, og deretter hensatt for avkjøling til romtemperatur over natten. Ny tynnsjiktskromatografi (TLC) viste ureagert utgangsmateriale. Blandingen ble behandlet med ytterligere trifenylfosfin (0,80 g, 3,03 mmol), dietylazodikarboksylat (0,5 ml, 3,03 mmol) og 3-hydroksybenzaldehyd (0,40 g, 3,03 mmol), og ble deretter omrørt ved romtemperatur i ytterligere 3 timer (fullstendig reaksjon ifølge TLC). Flyktige stoffer ble fjernet under vakuum og residuet ble renset med flashkolonne på silikagel med petroleumseter/etylacetat (2:1) som elueringsmiddel. Dette ga 0,33 g (16%) av tittelforbindelsen i form av en fargeløs olje.
'H NMR (CDC13) 8 1,5 (s, 9H), 3,5 (bs, 8H), 4,45 (m, 2H), 4,75 (m, 2H), 7,2 (d, 1H), 7,45 (s, 1H), 7,5 (m, 2H), 7,6 (s, 1H), 7,75 (s, 1H).
Trinn 4: tert- butvl- 4-( 3-( 2-[ 3-( 4- morfolinylmetvl) fenoksvletoksvl- 2- pvrazinvl)-1 - piperazinkarboksylat
En omrørt løsning av aldehydet fra Trinn 3 ovenfor (71,2 mg, 0,166 mmol) i 1,2-dikloretan (5 ml) ble behandlet med morfolin (19 mg, 0,22 mmol), 3Å molekylsikter og natriumtriacetoksyborhydrid (52 mg, 0,25 mmol). Blandingen ble omrørt ved romtemperatur i 5 timer (TLC-overvåking). Løsningen ble filtrert og filtratet ble behandlet med overskudd av mettet, vandig natriumbikarbonat. Eterekstraktene ble fraseparert og tørket over magnesiumsulfat. Blandingen ble filtrert og løsningsmiddelet ble fjernet under vakuum, hvilket ga 54 mg (65%) av tittelproduktet i form av en gul olje. Trinn 5: 2-( 1 - piperazinvl)- 3 - ( 2- f 3 -( 4- morfolinvlmetvl) fenoksy] etoks vi pyrazin Produktet fra Trinn 4 ovenfor (54 mg, 0,11 mmol) ble oppløst i tørr eter (20 ml), omrørt ved romtemperatur og behandlet med hydrogenklorid i eter (~6M; 5 ml). Den resulterende hvite suspensjon ble omrørt i 2 timer, og deretter raskt frafiltrert. Hydrokloridsaltet (hydroskopisk) ble oppløst i vann og nøytralisert med natriumkarbonat. Den frie basen ble ekstrahert inn i diklormetan. De organiske sjiktene ble tørket (magnesiumsulfat), filtrert og konsentrert under vakuum, hvilket ga 13 mg (29%) av tittelforbindelsen i form av en blekgul olje. LS/MS-renhet 100%.

De følgende forbindelser ble fremstilt analogt fra tert-butyl-4-{3-[2-(3-formylfenoksy)etoksy]-2-pyrazinyl}-l-piperazinkarboksylat (oppnådd i Eksempel 1, Trinn 3) og det nødvendige amin.
EKSEMPEL 2
2-(l-piperazinyl)-3-{2-[3-(l-pyrrolidinylmetyl)fenoksy]etoksy}pyrazin
Utbytte 31%. LS/MS-renhet 100%. <!>H NMR (CDC13) 8 1,7 (m, 4H), 2,45 (m, 4H), 2,9 (m, 4H), 3,4 (m, 4H), 3,5 (s, 2H), 4,3 (m, 2H), 4,6 (m, 2H), 6,7 (d, 1H), 6,85 (m, 2H), 7,15 (t, 1H), 7,45 (s, 1H), 7,7 (s, 1H).
EKSEMPEL 3
2-(l-piperazinyl)-3-{2-[3-(4-metyl-l-piperazinylmetyl)fenoksy]etoksy}pyrazin Utbytte 56%. LS/MS-renhet 100%. 'H NMR (CDCI3) 8 2,15 (s, 3H), 2,35 (b, 9H), 2,85 (m, 4H), 3,35 (m, 6H), 4,2 (m, 2H), 4,55 (m, 2H), 6,7 (d, 1H), 6,8 (m, 2H), 7,1 (t, 1H), 7,4 (s,lH), 7,6 (s, 1H).

EKSEMPEL 5

FREMSTILLING AV FARMASØYTISKE PREPARATER
Eksempel: Fremstilling av tabletter
Den aktive forbindelse 1 blandes med ingrediensene 2, 3,4 og 5 i ca. 10 minutter. Deretter tilsettes magnesiumstearat, og den resulterende blanding blandes i ca. 5 minutter og komprimeres til tabletter med eller uten filmovertrekk.
Farmakologiske tester
Den evne en forbindelse ifølge oppfinnelsen har til å binde seg til eller virke ved spesifikke 5-HT-reseptorsubtyper kan bestemmes ved hjelp av in vitro- og in vivo-undersøkelser som er kjent innen teknikkens stand. Den biologiske virkning av forbindelser fremstilt i henhold til eksemplene ble undersøkt ved hjelp av forskjellige tester.
Affinitetsforsøk
5-HT2C-reseptor-affiniteten til forbindelser i henhold til eksemplene ble bestemt i kompetitive eksperimenter hvor evnen til hver enkelt forbindelse i seriefortynning, til å fortrenge <3>H-merket 5-HT som er bundet til membraner fremstilt fra en transfisert (transfectet) HEK293-cellelinje som ga stabil (stably) ekspresjon av det humane 5-HT2C-reseptorprotein, ble bestemt ved hjelp av forsøk som omfatter proksimal scintillasjonsteknologi (Scintillation Proximity Assay technology). Ikke-spesifikk binding ble bestemt ved hjelp av 5 uM mianserin. Resultater som ble oppnådd for eksempler på forbindelser ifølge oppfinnelsen er oppgitt i Tabell 1 nedenfor. Verdier for 5-HT2c-reseptoraffinitet (Kj, nM) var vanligvis i området fra 1 nM til 1500 nM, fortrinnsvis 1 nM til 100 nM.
Virkningsforsøk
Den agonistvirkning som forbindelsen i henhold til eksemplene har på 5-HT2C-reseptoren ble bestemt ved å måle den evne hver enkelt forbindelse har til å mobilisere intracellulært kalsium i transfiserte (transfected) HEK293-celler som gir stabil (stably) ekspresjon av den humane 5-HT2C-reseptorprotein, ved hjelp av det kalsium-kelerende fluoriserende fargestoff FLUO-3 (Sigma, St. Louis, MO, USA).
Maksimum respons for 5-HT2C-agonister var vanligvis i området fra 15-100%, i forhold til maksimum respons av 5-HT (serotonin) i en konsentrasjon på 1 uM.
Claims (14)
1.

Ri, R2, R3 og R4 er hydrogen;
R5 og Rg er uavhengig av hverandre hydrogen, Ci-C4-alkoksy-C2-C4-alkyl, hydroksy-C2-C4-alkyl, Ci-C6-alkyl eller fenyl som kan være uavhengig substituert i en eller flere stillinger med Ci-C4-alkoksy; eller
R5 og Ré danner sammen med nitrogenatomet de er bundet til en morfolin-, pyrrolidin- eller piperazinring;
R7 er hydrogen; og
n=l-3;
og farmasøytisk akseptable salter, solvater, hydrater, geometriske isomerer, tautomerer, optiske isomerer eller N-oksider derav.

Forbindelse i henhold til krav 1, hvor gruppen -CH2N(R5)(R6) er bundet i meta-stilling, i forhold til sidekjeden alkylendioksy i fenylringen.

2-(l-piperazinyl)-3-{2-[3-(4-morfolinylmetyl)fenoksy]etoksy}pyrazin; 2-(l-piperazinyl)-3-{2-[3-(l-pyrrolidinylmetyl)fenoksy]etoksy}pyrazin; 2-(l-piperazinyl)-3-{2-[3-(4-metyl-l-piperazinylmetyl)fenoksy]etoksy}pyrazin; 2-(l-piperazinyl)-3- {2-[3- {[2-metoksyetyl)amino}metyl)fenoksy]etoksy}pyrazin; og 2-( 1 -piperazinyl)-3 - {2-[3 - {[isopropylamino)metyl} fenoksy] etoksy} pyrazin
og deres farmakologisk akseptable salter og solvater.

Farmasøytisk preparat som omfatter en forbindelse i henhold til hvilket som helst av kravene 1-5 som aktiv forbindelse, sammen med en farmakologisk og farmasøytisk akseptabel bærer.
7.
Anvendelse av en forbindelse i henhold til hvilket som helst av kravene 1-5 ved fremstilling av et medikament for å forebygge eller behandle en serotonin-relatert sykdom.
8.
Anvendelse i henhold til krav 7, hvor nevnte sykdom er en 5-HT2C-reseptor-relatert sykdom.
9.
Anvendelse i henhold til krav 7 eller 8, hvor nevnte sykdom er valgt fra spiseforstyrrelser, hukommelsesforstyrrelser, schizofreni, forstyrrelser i stemningsleie, angstlidelser, smerte, stoffmisbruk, seksuelle dysfunksjoner, epilepsi og urinveislidelser.

Anvendelse i henhold til krav 9, hvor spiseforstyrrelsen er overvekt.

Fremgangsmåte for fremstilling av en forbindelse i henhold til krav 1, som omfatter å omdanne en forbindelse med formel (II):
hvor R-3 og R» er hydrogen; og
hver av Hali og Hah er uavhengig halogen;
