Området for oppfinnelsen
Foreliggende oppfinnelse angår avliving uten lysis av celler i vekstkultur eller fermenteringsblanding av gjær, bakterier eller sopp, for forbedret gjenvinning av utskilt/ sekretert produkt fra cellene.
Bakgrunnen for oppfinnelsen
I de forskjellige fremgangsmåter ved dyrkning eller fermentering av mikro-organismer er det enkelte ganger nødvendig ved slutten av kulturveksten eller slutten av fermenteringsprosessen å være istand til å avlive de aktive celler i blandingen, slik at vekstaktiviteten stoppes og det ønskede produkt kan gjenvinnes fra kulturen eller fermenteringsblandingen. Dette gjelder spesielt når organismer som inneholder rekombinant DNA dyrkes som produksjonsvert i organismer og det er nødvendig å forhindre enhver leve-dyktig rekombinant organisme fra å bli frigjort i det omliggende miljø.
Det er enkelte ganger ønskelig også å lysere cellene på den tiden cellene blir avlivet for å gjenvinne ethvert ønsket produkt som blir dannet intracellulært. En vanlig måte celler blir avlivet og lysert på er ved bruk av varme. US patent 4.601.986 til Wegner et al, er et eksempel på bruken av varme for å avlive cellene og stoppe veksten av mikro-organismekulturer. En annen metode som er anvendelig på visse mikro-organismer er å endre det osmotiske trykk som gjør at cellene lyserer. Et eksempel på denne metode er illustrert i US patent 4.299.858 til Aubert et al. En annen vanlig metode brukt for å lysere celler er ved innføring av enzymer som bryter ned celleveggene eller membranene. Eksempler på denne metode er beskrevet i US 3.816.260 til Sugiyama, US 3.890.198 til Kobayashi et al og US 3.917,510 til Kitamura et al. Beskrivelsen av de overnevnte patenter er innbefattet heri pr. referat.
Imidlertid, i andre tilfeller er det ønskelig simpelthen å avlive cellene for å stoppe mikro-organismeaktiviteten uten å lysere cellene. Dette gjelder spesielt de systemer hvor cellene danner og skiller ut det ønskede produkt. I slike systemer er det meget ønskelig å avlive cellen uten å lysere cellene fordi lysering av cellene frigjør ytterligere celleavfall og materialer og gjør således gjenvinning og opprensning av det ønskede utskilte produkt mer vanskelig og kostbart. Følgelig når cellene i slike systemer kan avlives uten å lysere dem, blir økede prosesseffektiviteter i gjenvinning og opprensning av de utskilte produkter oppnådd.
I mange systemer er vertsmikro-organismer vanskelig å avlive, f.eks. sopp. Vanlige metoder så som varme er for kraftige og vil ødelegge eller endre det ønskede utskilte produkt før cellene blir avlivet. I slike systemer må produktet bli gjenvunnet uten å avlive cellene noe som krever bruk av tidkrevende og kostbare innesluttelsespro-sedyrer og utstyr.
I kommersielle fermeteringsprosesser i stor skala er det ønskelig å ha mer effektive og raskere metoder for å avlive cellene og stoppe celleveksten slik at den resulterende fermenteringsblanding kan behandles for å ekstrahere og gjenvinne det ønskede produkt som blir dannet uten å lysere cellene og således eliminere behovet for inneslutting. Varmemetoden og andre kjente metoder for å avlive celler er for langsomme og energiineffektive for kommersiel bruk og resulterer ofte i uønsket lysering av cellene. I tillegg er mange av de vanlige metoder for å avlive celler ikke kompa-tible med kultur og fermenteringsprosesser for mikrobiell fremstilling av enzymer. Konvensjonelle metoder denaturerer eller endrer ofte det ønskede enzym før det kan isoleres og gjenvinnes, eller disse metoder innfører materialer f.eks. andre enzymer som gjør isolering, gjenvinning og opprensning av det ønskede enzymprodukt mer vanskelig, mindre effektiv og følgelig mer kostbart.
Det er derfor et mål for foreliggende oppfinnelse å fremskaffe en raskere, mer effektiv metode for å avlive celler og preparere fermentering eller kultur av mikro-organismer for ekstraksjon og produktgjenvinningsbehandling. Det er et ytterligere mål for foreliggende oppfinnelse å fremskaffe en metode for å utføre ønsket celleavliving som er kompatibel med mikrobiell, produksjon av enzymer og gjenvinning og opprensning av slike mikrobielt fremstilte enzymer.
Oppsummering av oppfinnelsen
I sitt videste aspekt er foreliggende oppfinnelse en fremgangsmåte for å avlive celler i vekst, kultur eller fermentering av gjær, bakterier eller sopp hvor fremgangsmåten er særpreget ved å (a) justere pH til vekst, kultur eller fermenteringsblanding inneholdende gjæren, bakterien eller soppen til en verdi lik eller mindre enn pKafor en på forhånd valgt organiske syre og (b) tilsette en tilstrekkelig mengde av nevnte på forhånd valgte organiske syre eller salt derav til blandingen for å fremskaffe en i hovedsak fullstendig celleavliving i blandingen, uten lysis av cellene, og (c) "høste" det sekreterte/utskilte produkt fra cellene.
I et foretrukket aspekt er foreliggende oppfinnelse en fremgangsmåte for å avlive celler i vekst, kultur eller fermentering av gjær, bakterier eller sopp, omfattende trinnene punkt (a) å justere pH til veksten, kulturen eller fermenteringsblandingen inneholdende gjæren, bakterien eller soppen til omkring 4.75 eller mindre, og derpå (b) tilsette tilstrekkelig eddiksyre til blandingen for å fremskaffe en i hovedsak fullstendig celleavliving i blandingen .
Beskrivelsen av oppfinnelsen
I utviklingen av foreliggende oppfinnelse er det blitt funnet at endringen i pH alene hos en fermenteringsblanding ikke fremskaffer en fullstendig eller tilstrekkelig celleavliving. F.eks. ved en fermentering av Asperqillus Niger for fremstilling av chymosin, oppnås reduksjon av pH til omkring 2 ved å bruke svovelsyre, men ikke den ønskede grad av celleavliving. Følgelig har det tidligere vært nødvendig å oppvarme fermenteringsblandingen for tilstrekkelig å avlive cellene for å stoppe fermenteringsprosessen for å fremstille blandingen for gjenvinning av chymosin produktet .
I en foretrukket utførelsesform av foreliggende oppfinnelse er det blitt funnet at eddiksyre er spesielt anvendelig ved avliving av celler i fermenteringsprosesser forutsatt at pH til fermenteringsblandingen først justeres til omkring 4.75 eller under ved tilsetning av en mineralsyre såsom svovelsyre hvorpå eddiksyren tilsettes. Det er overraskende blitt funnet at når denne metode blir brukt, er mengden av eddiksyre som er nødvendig for å oppnå i hovedsak fullstendig avliving av cellene i fermenteringsblandingen ganske liten. Generelt vil en fullstendig celleavliving bli erholdt ved denne metode med kun omkring 1 til 2 vekt-% eddiksyre. I enkelte kultur- eller fermenteringsblandinger kan det være nødvendig å bruke større mengder eddiksyre såsom omkring 10 vekt-% eller mer basert på den totale vekt av blandingen, mens i andre prosesser kan et tilfredsstillende nivå av celleavliving bli erholdt ved å bruke så lite som 0.25 vekt-%. Generelt er det imidlertid blitt funnet at mengden av eddiksyre tilsatt etter justeringen av pH til blandingen vil være mellom omkring 0.5 vekt-% til omkring 10 vekt-%, fortrinnsvis mellom omkring 0.75 vekt-% og 5 vekt-% mer foretrukket mellom 1 og 3 vekt-%.
Mer generelt kan prosessen ifølge foreliggende oppfinnelse bli anvendt ved å bruke enhver ønsket organisk syre fulgt av overnevnte trinn, forutsatt at pH til kultur- eller fermenteringsblandingen først justeres ved å bruke en mineralsyre til en pH omtrentlig lik eller mindre enn pKatil'en organiske syre som er valgt for bruk for celleavli vingen etter pH er justert til det passende nivå idet den organiske syre blir tilsatt i en mengde som er tilstrekkelig til å fremskaffe den ønskede celleavliving. F.eks. dersom maursyre blir brukt for å utføre celleavlingen, blir pH til blandingen justert med en mineralsyre til omkring 3.75 eller mindre, derpå tilsettes maursyren for å utføre celleavlivingen. Dersom propionsyre blir valgt for bruk, vil pH bli justert til omkring 4.87 eller mindre, derpå tilsettes propionsyren i blandingen. Den organiske syre kan være enhver egnet og kompatibel syre med 1 til omkring 5 karbonatomer. Den valgte organiske syre bør være en som er kompatibel med, og ikke destruktiv overfor det ønskede produkt som blir dannet i kulturen eller fermeteringsblan-dingen og bør være slik at den ikke innvirker på separa-sjon-, gjenvinnings- og opprensnigsmetoder brukt for å gjenvinne det ønskede produkt fra blandingen.
Det er også blitt funnet at det ikke er nødvendig å justere pH til blandingen før tilsetning av den organiske syre. Den organiske syre kan tilsettes til blandingen, derpå tilsettes mineralsyren for å justere pH til det ønskede nivå for utføring av foreliggende oppfinnelse. Som nevnt nedenfor er det samme tilfelle for bruk av saltet av den organiske syre. Saltet kan tilsettes, derpå kan pH justeres. I sitt bredeste aspekt er det bare viktig ved utføring av foreliggende oppfinnelse å ha den organiske syre eller saltet derav i blandingen som har en pH ved eller under pKaverdien til den organiske syre utvalgt for bruk.
Selvom oppfinnelsen ikke er begrenset til eller nødvendig
basert på den følgende teori, er det antatt at foreliggende oppfinnelse oppnår den uventede og effektive og fullstendi-ge celleavliving med den følgende mekanisme. Ved å redusere pH av blandingen eller mediet til en verdi lik eller mindre enn pKatil den organiske syre som skal bli brukt, blir
syren protonert eller uladet og blir "usynlig" for cellen som en syre. Cellen kan så ta opp eller innføre den nøytra-le syreforbindelse på vanlig måte som et næringsmiddel
fordi cellen ikke ser forbindelsen som en syre. Når den er inne i cellen blir syren reionisert og endrer så pH inne i cellen noe som dreper cellen. Ifølge denne teori av meka-nismen er det klart ønskelig å velge ut en organisk syre som cellen sannsynligvis vil ta inn som næringsmiddel i syrens protonerte form. En foretrukket syre er eddiksyre fordi den er effektiv med et bredt område av celler og fordi den er en av syrene med lavest tilgjengelig pris. Andre effektive syrer kan bli brukt avhengig av cellekul-turen det er tale om og kostnaden ved prosessen.
Konsentrasjonen av den organiske syre er ikke kritisk, men bør være av en høy nok konsentrasjon slik at celleblandingen ikke blir for mye fortynnet når den organiske syre tilsettes. Når eddiksyre blir brukt er iseddiksyre en passende form.
Det vil også bli funnet av fagmannen at saltene av de organiske syrer også kan bli brukt. F.eks. i stedet for eddiksyre kan natriumacetat bli brukt sammen med eddiksyre. Syresaltet, eller minst en del derav vil bli protonert i oppløsningen hvor pH har blitt senket av mineralsyren til en verdi under pKatil den organiske syre. Som det også vil være tydelig for fagmannen, vil pH-justeringen av oppløs-ningen være forskjellig når et syresalt blir brukt enn når syren i seg selv blir brukt fordi saltet ikke vil påvirke pH slik som den organiske syre. Ethvert organisk syresalt kan bli brukt som er kompatibelt med oppløsningen og kompo-nentene av oppløsningen som skal gjenvinnes fra oppløsnin-gen etter celleavliving er blitt utført. Som nevnt ovenfor kan pH til blandingen bli justert til syrens pKa-verdi eller lavere før eller etter den organiske syre derav tilsettes blandingen. Imidlertid er den foretrukne utfø-relse av oppfinnelsen at pH først justeres til den ønskede verdi med en mineralsyre og derpå tilsettes den organiske syre.
Mineralsyrene som kan bli brukt for å justere pKaav blan-dingene i henhold til fremgangsmåten ifølge oppfinnelsen innbefatter svovelsyre, saltsyre og andre mineralsyrer som er istand til å redusere pH av celleblandingen og til en verdi lik eller mindre den til pKaav den organiske syre som skal bli brukt for å avlive cellene i blandingen. Det er ønskelig å velge ut en mineralsyre for justering av pH som er kompatibel med fremgangsmåtene og utstyret som skal bli brukt for å adskille eller ekstrahere det ønskede produkt fra fermenteringen eller kulturblandingen eller mediet. Konsentrasjonen av mineralsyren som skal brukes bør være høy nok slik at pH til celleblandingen kan justeres til det ønskede nivå uten unødvendig fortynning av blandingen.
Etter å ha beskrevet generelle aspekter av foreliggende oppfinnelse vil nå oppfinnelsen bli illustrert ved de spesielle utførelsesformer beskrevet i de følgende eksempler.
Eksempler
I de følgende eksempler ble prøvene erholdt fra fermenteringen av en Aspergillus niger var. awamori typisk foretatt med 6 % eller 10 % soyamel/glukose og innhøstet etter 4 til 5 dager. For formålet med å undersøke celleavlivingen fremskaffet ved foreliggende oppfinnelse er spesielle celleproduksjonsteknikker ikke viktige. De følgende kompa-rative resultater illustrerer forskjellige utførelsesformer av foreliggende oppfinnelse ved å bruke en standard seriell fortynningstest for å bestemme mengden av levende celler som er tilbake etter celleavlivingen er utført. Etter celleavlivingsbehandlingen blir prøvene brakt til pH 5.5, serielt fortynnet i 0.85 % NaCl, sådd på CMA-plater inkubert ved 37°C i 72 timer og angitt i CFU/ml.
Eksempel I
Dette eksempel illustrerer effektiviteten av metodene ifølge foreliggende oppfinnelse til å fremskaffe i hovedsak fullstendig celleavliving av A. niger var, awarmori.
To prøver av en A. niger var, awarmori. fermateringsblan-ding ble justert til pH 2.0 med svovelsyre. Etter at syren var godt blandet ble 4 vol-% iseddiksyre tilsatt til en prøve. Begge prøver ble lagret over natten i et kjølerom og derpå undersøkt for celleavliving. Resultatet fra den serielle fortynningstest var som følger:

Dette eksempel viser at svovelsyren alene ikke fremskaffet en fullstendig celleavliving, men kombinasjonen av svovelsyren og eddiksyren fremskaffet en fullstendig celleavliving.
Eksempel II
I dette eksempel ble en del av en fermenteringsblanding lik den i eksempel I avkjølt til 12°C og oppbevart i 60 timer. Tre prøver ble tatt fra blandingen: én var ubehandlet og en ble behandlet med H2S04alene til pH 2.0. Den tredje prøven ble behandlet med H2S04til en pH 2.0, iseddiksyre ble tilsatt i en mengde på 1 vekt-% av prøveblandingen og derpå ble blandingen aerert og omrørt i omtrent 1 time. Alle tre prøver ble justert til pH 5.5, med NaOH, serielt fortynnet, utsådd og inkubert i 5 dager ved 37°C. Forsøksresultatene var som følger:
Dette eksempel indikerer at eddiksyre/svovelsyrebehandling gir minst 6 log cellereduksjon.
Eksempel III
1 dette eksempel ble et fermenteringsmedium lik det i eksempel I brukt for å vise effekten av pH-justeringen på celleavlivingen. I tillegg blir i dette eksemplet et organisk syresalt benyttet i stedet for den organiske syre. I det følgende ble prøver 1-5 brukt som sådanne og 6-10 hadde 2 % acetat (som 4.53 g natriumacetat pr. 100 ml) tilsatt før pH-justering. pH av prøvene 1-5 før justering var omkring 5.8 og av prøvene 6-10 omkring 6.0. pH hos alle prøver ble så justert til verdiene vist nedenfor bortsett fra f.eks. 4 som ikke ble justert og som virket som kon-trollprøve for mediet. pH ble justert i hver prøve med H2S04eller NH4OH for å erholde den indikerte pH.
Alle prøver ble lagret på is i 4 timer og så ble pH justert til 5,5 for utsåing på CMA-plater med antibiotika. Platene ble inkubert ved SPC i 7 dager. Forsøksresultatene var som følger:

Dette eksempel illustrerer viktigheten av å justere pH av blandingen til en verdi ved eller fortrinnsvis under pKatil den anvendte organiske syre. En mer fullstendig celle-avling ville bli erholdt ved lavere pH i områder dersom høyre mengder acetat ble brukt så som 4 %. Imidlertid ble det nedre nivå for acetat brukt i dette eksempel slik at effekten av pH kunne bli oberservert.
Eksempel IV
Dette eksempel illustrerer bruken av foreliggende oppfinnelse til å avlive gjærceller. For dette eksempel ble en gjaertype kjent som Saccharom<y>ces cerevisiae dyrket på standard "YM" medium tilgjengelig fra Difco, ved 250 rpm i 24 timer ved 37°C. Som i eksempel 3 ovenfor ble 10 prøver tatt, prøvene 6-10 ble behandlet med 2 % acetat (som natriumacetat) pH ble justert til verdien vist nedenfor og derpå utsådd og inkubert i 4 timer.

Som det kan konkluderes med av det ovenstående ble omkring 20 % avliving erholdt ved pH 2,8, omkring 30 % ved pH 3,6 og ingen signifikant avliving ved pH 4.7, 5.7 eller 6.75. Selvom dette eksempel ble foretatt for å bestemme effekten av pH på effektiviteten av avlivingen, er det tydelig at en mer effektiv avliving ville bli oppnådd ved høyere acetat-nivå f.eks. 4 %. I tillegg, som det vil bli forstått er det vanskeligere å mengdebestemme nøyaktig celleavlivingen og kulturveksten til gjær enn sopp, men dette eksempel viser anvendeligheten av foreliggende oppfinnelse for gjær.
Etter å ha beskrevet foreliggende oppfinnelse og illustrert spesielle utførelsesformer av oppfinnelsen blir nå omfanget av foreliggende oppfinnelse definert ved kravene som føl-ger.
Claims (7)
1. Fremgangsmåte ved avliving uten lysis av celler i vekstkultur eller fermenteringsblanding av gjær, bakterier eller sopp, for forbedret gjenvinning av utskilt/sekretert produkt fra cellene,
karakterisert vedat den omfatter: (a) å justere pH for vekst-, kultur- eller fermenteringsblandingen inneholdende gjæren, bakterien eller soppen til en verdi lik eller mindre enn pKafor en på forhånd valgt organisk syre, og (b) tilsette en tilstrekkelig mengde av den på forhånd valgte organiske syre eller et salt derav til blandingen for å fremskaffe en i hovedsak fullstendig celleavliving i blandingen, uten lysis av cellene, (c) "høste" det sekreterte/utskilte produkt fra cellene .
2. Fremgangsmåte ifølge krav 1,karakterisert vedat den organiske syre er maursyre, eddiksyre, propionsyre eller et salt derav.
3. Fremgangsmåte ifølge krav 1,karakterisert vedatpH justeres ved å tilsette en mineralsyre til blandingen.
4. Fremgangsmåte ifølge krav 2,karakterisert vedatpH justeres ved å tilsette en mineralsyre til blandingen.
5. Fremgangsmåte ifølge krav 2,karakterisert vedat den organiske syre er eddiksyre og pH justeres til omkring 4,75 eller mindre.
6. Fremgangsmåte ifølge krav 1,karakterisert vedat pH justeres før den organiske syre eller saltet tilsettes.
7. Fremgangsmåte ifølge krav 1,karakterisert vedat den organiske syre eller saltet tilsettes før pH justeres.
