Abstract

Oppfinnelsen angår rizatriptan eller et farmasøytisk akseptabelt derivat derav som inkluderer a) fremstilling av diazoniumsaltet av anilinhydrokloridet (II); etterfulgt av reduksjon og surgjøring, noe som gir hydrazinet (III); b) in situ omsetting av hydrazinhydrokloridet (III) med a-keto-d-valerolakton, noe som gir hydrazonet (IV); c) Fisher-indolomsetting av hydrazonet (IV), noe som gir pyranoindolonet. (V), eventuelt etterfulgt av en hydrolysereaksjon, noe som gir (VI); d) transesterifisering av (V) eller esterifisering av dens hydrolyseprodukt (VI), noe som gir (VII), hvor R betyr rett eller forgrenet C1 -C4-alkylkjede; e) omdanning av hydroksylgruppen i (VII) til dimetylamin, noe som gir indolkarboksylatet (VIII), hvor R har betydningen som definert ovenfor; f) saponifisering av 2-karboalkoksygruppen (VIII), noe som gir indolkarboksylsyren (IX); og g) dekarboksylering av indolkarboksylsyren (IX), noe som gir rizatriptan og eventuelt oppnåelsen av et farmasøytisk akseptabelt salt derav.
