Description
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av en forbindelse med formelen: hvor M er hydrogen eller en alkylgruppe med opptil 3 karbonatomer, A er en nukleofil substituent valgt fra halogen eller en gruppe med følgende formel: eller CN, hvor R er hydrogen, en grenet eller rettkjedet alkylgruppe med fra 1 til 4 karbonatomer, eller fenyl, og mellomprodukter for anvendelse deri. Fremgangsmåteproduktet kan brukes som mellomprodukter for en rekke forskjellige modifiserte nukleosider med biologisk aktivitet.
Man har nylig oppdaget at revers transkriptase har en hemmende aktivitet på forskjellige modifiserte nukleosider,
noe som har gjort at de allerede brukes og kan videreutvikles som terapeutiske midler ved behandling av AIDS og HIV infeksjoner. Dette har stimulert interessen for forbedrete fremgangsmåter for fremstilling av slike modifiserte nukleosider. Av spesiell interesse er nye fremgangsmåter for fremstilling av 3'-substituerte 2',3'-dideoksyribonukleosider så som 3'-azido-3'-deoksytymidin (AZT) og 3'-deoksy-3'-fluortymidine (FLT) som er angitt å være sterkt hemmende for HIV indusert cytopatogenitet. Det har vært publisert omfattende studier og arbeider over syntesen og den biologiske aktiviteten til 3'-azido-, 3'-amino- og 3'-fluorpyrimidin og purin 2', 3'-dideoksyribonukleosid-analoger.
Fremgangsmåtene for å fremstille slike 3'-substituerte nukleosider har vanligvis utviklet seg langs to separate veier: (1) substitusjon av 3'-0H-funksjonen i et 2'-deoksynukleosid, noe som er tilfellet for syntesen av AZT eller FLT fra
tymidin, eller
(2) fremstillingen av en 3-substituert furanosid-forbindelse fulgt av koplingen av en egnet purin- eller pyrimidin-base, så som tymin. Den sistnevnte fremgangsmåten har visse fordeler, ettersom den bruker enklere utgangsforbindelser og gir lettere substitusjon av en rekke nukleofiler i 3'-stillingen, noe som gir mellomprodukter som er meget godt egnet for effektiv kopling med purin- eller pyrimidin-baser. Sistnevnte fremgangsmåte har derfor de største muligheter for en syntese av 3'-substituerte nukleosider i større skala.
Det er beskrevet flere fremgangsmåter for syntese av 3-substituerte furanosid-sukre, men de er alle kompliserte og krever mange trinn og kostbare reagenser. Fleet, G.W. et al: Tetrahedron 1988, 44(2) 625-636 beskriver syntesen av metyl-5-o- tert-butyldifenylsilvl-2-deoksy-0! ( 8) -D- threo-pento-furanosid fra D-xylose og dets omdannelse til azido, fluor- og cyano-sukre, etterfulgt av en kopling av disse derivater med beskyttet tymidin, noe som gir tymidin-forbindelsene. Bravo, P. et al.. J. Org. Chem. 1989, 54, 5171-5176 beskriver den assymetriske syntesen av 3-fluorfuranoser idet man starter fra en forbindelse med følgende formel:
som er monoalkylert på det fluorinerte karbonatomet med allylbromid. Fjerning av sulfinylgruppenn fulgt av en reduktiv opparbeiding og oksydativ avspaltning av dobbeltbindingen, gir 5-0-benzoyl-2,3-di-deoksy-3-fluorfuranose.
Den foreliggende oppfinnelse beskriver en forbedret alternativ fremgangsmåte for syntesen av 3-substituerte furanoser og furanosider. Fremgangsmåten er ukomplisert, bruker et lite antall trinn, og enkle og billige utgangsforbindelser.
Sammendra<g> av oppfinnelsen
Foreliggende oppfinnelse vedrører en forbedret fremgangsmåte for den assymetriske syntesen av 3-substituerte furanosidforbindelser som innledningsvis definert. Den omfatter ifølge oppfinnelsen følgende trinn:
a) omsetning av en forbindelse med formelen:
hvor X er klorid eller bromid, med propargylalkohol for fremstilling av heks-5-en-2-yn-l-ol; b) omsetning av produktet fra trinn a) med litiumaluminiumhydrid for fremstilling av 2,5-heksadien-l-ol; c) omsetning av produktet fra trinn b) i nærvær av di-isopropyl-D-tartrat for fremstilling av 2R,3R-epoksyheks-5-en-l-ol ;
d) omsetning av produktet fra trinn c) med
Ti(OR')nA4-n
hvor n er et heltall fra 1 til 3; R' er en grenet eller rettkjedet alkylgruppe med opptil 4 karbonatomer, og A er enten F, Cl, Br, I, eller kan velges fra en gruppe med følgende formel:
eller CN, hvor R er hydrogen, en grenet eller ugrenet alkyl-gruppe med opptil 4 karbonatomer eller fenyl, som har den nukleofile substituenten A, hvorved man får fremstilt en forbindelse med formelen: e) omsetning av produktet i trinn d) med ozon fulgt av en katalytisk reduksjon med hydrogen over palladium-på-karbon, hvorved man får fremstilt en forbindelse med formelen:

Detaljert beskrivelse
I overensstemmelse med Reaksjonsskjerna I, blir propargylalkohol 1 omsatt med allylklorid eller allylbromid 2, noe som gir alkoholen 3_. Reduksjon av denne med litiumaluminiumhydrid gir olefin 4.. Epoksydering av 4 gir oksyran 5. Områdeselektiv oksyran-ringåpning av 5 ved behandling med en passende nukleofilisk reagens med substituent A, gir diol 6, som så hydrogeneres i nærvær av 10% palladium og karbon, noe som gir den substituerte pentose-forbindelsen med formel 7.
I den foretrukne utførelsen av foreliggende oppfinnelse, blir de 3-substituerte furanosid-sukrene med formel 1 fremstilt ved hjelp av følgende trinn: (a) propargylalkohol 1 kondenseres med allylklorid eller allylbromid 2 i vann ved pH 8-9 ved en temperatur på 65-70°C under røring med CuCl eller CuBr, noe som gir heks-5-en-2-yn-l-ol 3; (b) forbindelsen 3 blir så redusert med LiAlH4 i tetrahydrofuran, hvilket gir allylalkohol, 4, trans-2,5-heksadien-l-ol; (c) allylalkoholen 4 blir så asymmetrisk epoksydert i nærvær av di-isopropyl-D-(-)-tartrat, noe som gir oksiranforbindelsen 5, dvs. 2R,3R-epoksyheks-5-en-l-ol; (d) oksiranforbindelsen 5 blir så underkastet en område-selektiv eller regio-selektiv nukleofil ringåpning ved behandling med et egnet nukleofilt reagens med følgende formel:

hvor n er et heltall fra 1 til 3, R' er en grenet eller ugrenet alkylgruppe med opp til 4 karbonatomer, og A er som definert ovenfor, hvilket gir diolforbindelsen med formel 6;
(e) diolforbindelsen 6 blir så behandlet med
(i) ozon; (ii) reduksjon ved hydrogenering med H2 på palladium-på-karbon, og; (iii) alkoholyse, noe som gir 3-substituerte 2,3-dideoksy-D-erytropentosider med formel 7, som en blanding av a og isomerer. Som angitt ovenfor i trinn (c), blir allylalkoholfor-bindelsen A asymmetrisk epoksydert ved den fremgangsmåte som er beskrevet av Y. Gao et al., J. Amer. Chem. Soc.. 109, 5765-5780 (1987), som herved inngår som en referanse. Denne fremgangsmåten gir epoksider fra olefiner i et minst 94% enantiomerisk overskudd. Olefiner kan omdannes til det tilsvarende epoksydet ved behandling med en katalytisk mengde av en katalysator, fremstilt fra tartrat, såsom dietyl eller di-isopropyltartrat og titan (IV) isopropoksid. Det beste forholdet titan/tartrat er 1:1,2. Det er viktig at reaksjonsblandingen holdes fri for fuktighet. Pulveriserte, aktiverte molekylære siler kan også brukes. Videre kan man anvende opp-løsningsmidler som diklormetan, toluen eller iso-oktan. Reak-sjonene blir vanligvis utført ved temperaturer omkring -20°C. Alle reaksjoner utføres i nærvær av tert-butyl-hydroperoksid (TBHP), skjønt man også kan bruke andre peroksider. Vanligvis blir tartrat-katalysatoren fremstilt ved å blande det valgte tartratet og titan (IV) isopropoksidet ved -20°C i et oppløsningsmiddel som metylenklorid, hvoretter man enten tilsetter den olefiniske alkoholen eller det tertiære butyl-hydroperoksidet. I ethvert tilfelle vil de tre ingre-diensene bli rørt i ca. 30 minutter før man tilsetter den siste reagensen, enten dette er alkoholen eller det tertiære butylhydroperoksidet. Alle reaksjoner utføres i nærvær av pulveriserte, aktiverte, molekylære siler. De 30 minutters røring betegnes ofte som "eldingsperioden", og er en meget viktig faktor for å oppnå høy enantioselektivitet.
I trinn (d) ovenfor vil oksiranforbindelsen 5 blir underkastet en regioselektiv nukleofil ringåpning ved mild behandling med en reagens med følgende formel:

hvor n er et heltall fra 1 til 3, R' er en grenet eller rettkjedet alkylgruppe med fra 1 til 4 karbonatomer, og A er som definert ovenfor, noe som gir diolforbindelsen med formel 6.
De nukleofile reagenser med formelen Ti(0R')nA4_n kan hensiktsmessig fremstilles fra titan (IV)-alkoksider med formelen Ti(OR')4 ved en reaksjon med en hensiktsmessig molar konsentrasjon av de tilsvarende syre- anhydrid- eller trimetyl-silyleter-forbindelsene som følger:

hvor R'' er hydrogen, CH3CO, benzolyl eller Si(CH3)3- For eksempel kan titantri-isopropylklorid fremstilles ved å omsette titan (IV) isopropoksid med trimetylsilylklorid. Titan-tri-isopropolyazid kan fremstilles fra titan (IV) isopropoksid ved en reaksjon med en HN3 i pentan. Titan-tri-isopropyltiobenzoat kan fremstilles ved å omsette titan (IV) isopropoksid med tiobenzosyre.
Det foretrukne nukleofile reagenset, i det tilfellet hvor A er fluor, er titan (IV) difluor-di-isopropoksid, [TiF2(OiPr)2]/ som hensiktsmessig kan fremstilles ved å tilsette titan (IV) isopropoksid ved 20 til 25°C til benzoylfluorid eller CH3COF i heksaner. Produkter, dvs. titan (IV) di-isopropoksid, kan oppsamles ved filtrering i en inert atmosfære.
Når substituenten A er forskjellig fra fluor, så vil det foretrukne nukleofile reagenset for ringåpningsreaksjonen være titan (IV) tri-isopropoksid. Dette reagenset ble brukt for den regioselektive ringåpningen av epoksydforbindelsen med følgende formel: hvor reaksjonsbetingelsene er som angitt i Tabell I:
De foran beskrevne reaksjonsbetingelser kan på liknende måte anvendes for en regioselektiv ringåpning av forbindelsen 5, dvs. 2R,3R-epoksyheks-5-en-l-ol. Således vil titan (IV) tri-isopropoksid-forbindelsene gjøre det mulig å gi en mild område-selektiv oksiran-ringåpning ved hjelp av den nukleofiliske gruppen A med høy effektivitet. Denne fremgangsmåten kan brukes for en rekke oksiran-åpningsreaksjoner hvor A kan være enhver nukleofil gruppe.
Den nukleofile ringåpningsreaksjonen (trinn d) kan utføres i en rekke forskjellige oppløsningsmidler såsom benzen, toluen, kloroform, metanol eller blandinger av disse. Vanligvis er det ønskelig eller foretrukket med et 1,5 molart overskudd av reagenset. Temperaturbetingelsene kan variere fra 0 til 130°C, fortrinnsvis mellom 80 og 120°C, hvor reaksjonen løper i minst 1 time, noe som gir mer enn 9 0% omdannelse. Sluttproduktene kan isoleres fra reaksjonsblandingen ved kromatografi.
I trinn (e) ovenfor, blir diolforbindelsen med formel 6 oppløst i metylalkohol, og under avkjøling til -60 til -70°C tilsetter man tørr ozon gjennom et bobleapparat inntil reaksjonen er ferdig. Etter oppvarming til OOC, tilsettes palladium på karbon, og reaksjonsblandingen hydrogeneres inntil hydrogenopptaket er komplett. En tilsetning av saltsyre fullfører glykosideringen, noe som gir den 3-substituerte-2,3-di-deoksy-erytropentose-forbindelsen med formel 7, som en blanding av a og 6 isomerer.
Andre hensikter og fordeler ved oppfinnelsen vil fremgå av den foreliggende beskrivelse og de etterfølgende eksempler. Disse beskriver oppfinnelsen mer detaljert uten at oppfinnelsen som sådan er begrenset til eksemplene.
Eksempel 1
Heks- 5- en- 2- vn- l- ol
En rørt blanding av 250 ml mettet natriumkloridoppløsning ble tilsatt 1 ml saltsyre, 8g kopper (I) klorid og 28g propargylalkohol ved romtemperatur i en 40% natriumhydroksyd-oppløsning inntil pH ble justert til 9. Reaksjonsblandingen ble oppvarmet i et bad ved 70OC, og man tilsatte dråpevis en oppløsning av 120 ml allylalkohol i 80 ml metylalkohol. pH på reaksjonsblandingen ble holdt mellom 8 og 9 ved tilsetning av 40% natriumhydroksyd etter behov. Etter tilsetningen av allylkloridoppløsningen, og med en pH mellom 8 og 9, ble blandingen rørt ved 70<Q>C i 3,5 timer. Kolben ble avkjølt til romtemperatur, og saltsyre ble tilsatt inntil pH var falt til 2. Den organiske fasen ble utskilt, og det vandige laget ekstrahert med 3 x 50 ml eter. De organiske lag ble slått sammen, og flyktige forbindelser fjernet i vakuum, noe som ga en olje, som ble vakuumdestillert. Dette ga 45g av det ønskede produktet som en vannklar væske, kokepunkt 67-68°C/10 mm no20 1,4670.
13C-NMR: 137,69(C-5), 115,98(C-6), 82,65 og 81,54(C-2 og C-3), 50,59(C-1), 23,21(C-4).
Analyse. Beregnet for C6HsO: C, 74,97; H, 8,39.
Funnet: C, 75,11; H, 8,18.
Eksempel 2
Trans- 2. 5- heksadien- l- ol
En rørt blanding av 3,8g litium aluminiumhydrid i 150 ml tetrahydrofuran avkjølt til 0°C, ble dråpevis tilsatt en opp-løsning av 9,6g av produktet fra eksempel 1 i 50 ml tetrahydrofuran. Etter at tilsetningen var ferdig, ble kjølingen fjernet, og man lot temperaturen stige til romtemperatur fulgt av 3 0 minutters røring. Temperaturen ble så hevet til 45°C i 3 timer, og deretter ble blandingen avkjølt til 0°C. lOOml mettet ammoniumklorid ble forsiktig, dråpevis tilsatt, og den resulterende blanding ble filtrert. Filterkaken ble vasket med 3 x 20 ml dietyleter, og de samlede filtrater tørket over magnesiumsulfat. Flyktige forbindelser ble fjernet i vakuum, og resten ble vakuumdestillert til 7,9g av det ønskede produktet som en vannklar væske, kokepunkt 70 - 72°C/15 mm, HD20 1,4530. 13C-NMR: 137,57(C-5), 132,01 og 128,89(C-2, C-3), 115,40(C-6), 63,00(C-1), 36,80(C-4).
Analyse. Kalkulert for C6H10O: C, 73,43; H, 10,27.
Funnet: C, 72,98; H, 10,01.
Eksempel 3
2R. 3R- epoksyheks- 5- en- l- ol
En blanding av 3,Og av en 4A pulverisert, aktivert molekylærsil og 300 ml tørr metylenklorid ble avkjølt til -20°C, og man tilsatte først 2,81g (2,36 ml) di-isopropyl D-(-)-tartrat og deretter 2,84g (2,99 ml) titan (IV) isopropoksid under kontinuerlig røring. Reaksjonsblandingen ble rørt ved -20°C samtidig som man over 5 minutter tilsatte 40 ml 5 M TBHP i metylenklorid. Røring ble fortsatt ved -20°C i en halv time, hvoretter man dråpevis tilsatte en oppløsning av 9,8g av produktet fra eksempel 2 i 50 ml metylenklorid, mens temperaturen ble holdt mellom -25 og -20°C. Blandingen ble så rørt ved denne temperaturen i fra 8 til 10 timer. Reaksjonen ble stoppet ved å tilsette 8 ml av en 10% vandig oppløsning av natriumhydroksyd mettet med natriumklorid (10g NaCl + 10g NaOH + 95 ml vann), som på forhånd var avkjølt til -20°C. Eter ble tilsatt til man fikk en 10% volum/volum-reaksjonsblanding. Kjølebadet ble fjernet, og røringen fortsatte med oppvarming til 10°C fulgt av ytterligere 10 minutters røring. Under røring ble så 80 g magnesiumsulfat og lg diatomerjord tilsatt. Røring ble fortsatt i ytterligere et kvarter, fulgt av en filtrering gjennom et lag av diatomerjord. Filterkaken ble vasket med 3 x 50 ml eter. De samlede eterfiltrater ble tørket over magnesiumsulfat, og flyktige forbindelser fordampet i vakuum, noe som ga en rest som ble vakuumdestillert til 8,9g av det forønskede produktet, kokepunkt 85-87OC/10 mm.
n<D>20 1,4458 [a]D<20> + 23,2 (c 10, CH3OH). <13>C-NMR: 134,49 (C-5), 117,28(C-6), 62,69(C-1), 58,65(C-2), 55,12(C-3), 36,45(C-4).

Funnet: C, 62,88; H, 8,19.
Eksempel 4
Titan ( IV) difluor- di- isopropoksid
En rørt blanding av 37,4g benzoylfluorid i 100 ml heksan under avkjøling i et 20-25°C bad ble dråpevis tilsatt 28,4g titan (IV) isopropoksid. Reaksjonen skjer under en temperaturøkning. Det dannet seg et hvitt, fast stoff som ble fra-filtrert i en inert atmosfære, deretter tørket under vakuum til 13,6g av det forønskede produkt som et hvitt, fint pulver.
Eksempel 5
2R, 3S- 3- fluorheks- 5- en- l. 2- diol
En blanding av 13,6g titan (IV) di-fluor-di-isopropoksid i 180 ml tørr toluen ble kokt under tilbakeløp med røring på et oljebad ved 120°C. Man tilsatte så dråpevis en oppløsning av 5,8g 2R,3R-epoksyheks-5-en-l-ol i 10 ml tørr toluen til den kokende reaksjonsblandingen. Etter tilsetningen ble badet fjernet og røring fortsatt mens temperaturen ble senket til 25-30°C. Ca. 15 ml mettet natriumbikarbonat ble så tilsatt under kraftig røring. Røringen ble fortsatt i 2 timer, og pH ble gjort nøytralt til svak alkalisk. Ytterligere mettet natriumbikarbonatoppløsning ble tilsatt etter behov. Reaksjonsblandingen ble filtrert gjennom diatomé-jord, og filterkaken vasket med aceton. De samlede filtrater ble tørket med magnesiumsulfat, hvorettet de flyktige forbindelser ble fjernet i vakuum. Resten ble renset ved kromatografi på silisium-dioksydgel ved hjelp av eluering med 50:1 kloroform-isopropyl-alkohol, noe som ga 3,4g av det forønskede produktet som et hvitt, krystallinsk, fast stoff, smeltepunkt 37-38°C. TLC (9:1 kloroform-isopropyl-alkohol) Rp = 0,41.
13C-NMR: 134,85(d, JC-F 3,5Hz, C-5), 117,62(C-6), 93,52(d, Jc_ F 171,6Hz, C-3), 73,30(d, JC-F 23,1Hz, C-2), 63,33(d, Jc-F 5,5Hz, C-l), 35,98(d, JC-F 21,1Hz, C-4).
Analyse. Beregnet for C6H11O2F: C, 53,72; H, 8,27; F, 14,17 Funnet: C, 53,92; H, 8,01; F, 13,28.
Eksempel 6
Metyl 3- fluor- 2. 3- dideoksy- a, lf- D- erytropento- furanosid
En oppløsning av l,5g 2R,3S-3-fluorheks-5-en-l,2-diol ble oppløst i 150 ml metylalkohol og avkjølt til -60 til -70°C. Ovnstørket ozon ble boblet gjennom reaksjonsblandingen i 3
timer. Man lot reaksjonsblandingen oppvarmes til 0°C og tilsatte 0,05g palladium-på-karbon. Under røring ble 250 ml hydrogen absorbert. Reaksjonsblandingen ble så filtrert, og man tilsatte 1 ml 10% HC1 i metylalkohol. Etter at reaksjonen var ferdig, noe som ble vist ved TLC (10:1 kloroform-metanol),
ble reaksjonsblandingen nøytralisert med tørr kaliumkarbonat, deretter filtrert og inndampet, hvilket ga l,0g av det for-
ønskede produkt som en blanding.
