Description
Oppfinnelsen vedrører en analogifremgangsmåte for fremstilling av terapeutisk aktive, nye kromanderivater med formel I
hvor
R<1> og R<2> hver betyr A,
R<4>, R<7>, R<8>, R<9> og R<10> hver betyr H,
R3 betyr OH,
R<3> og R<4> betyr tilsammen også en binding,
R<5> betyr A eller en usubstituert eller en gang med A
eller OA substituert benzylrest,
R<6> betyr H, CN, F, Cl, Br, J, H2NCO, og
A betyr alkyl med 1-6 C-atomer,
samt farmasøytisk akseptable syreaddisjonssalter derav.
Formålet med oppfinnelsen var å finne frem til nye forbindelser med verdifulle egenskaper som legemidler.
Det ble funnet at forbindelsene med formel I og deres farmasøytisk akseptable salter, har verdifulle farmakologiske egenskaper med god forenlighet. Således har de virkninger på det kardiovaskulære system, idet det som regel kan iakttas et selektivt angrep på koronarsystemet ved lavere doser, og en blodtrykkssenkende effekt ved høyere doser. På koronarsystemet inntrer det f.eks. motstandsreduksjon og strømningsøkning, idet innflytelsen på hjertefrekvensen er liten. Videre opp-viser forbindelsene en relakserende virkning på forskjellige glattmuskulære organer (mage- og tarmkanalen, respirasjons-systemet og livmoren). Virkningen av forbindelsene kan fast-slås ved hjelp av i og for seg kjente metoder, som f.eks. angitt i EP-A1-76075, EP-A1-173848 eller AU-A-45547/85 (Derwent Farmdoc Nr. 86 081 769) samt av K.S. Meesmann et al., Arznei-mittelforschung 25 (11), 1975, 1770-1776. Som forsøksdyr egner seg f.eks. mus, rotter, marsvin, hunder, katter, aper eller griser.
Forbindelsene kan således anvendes som aktive forbindelser i legemidler innen human- og veterinærmedisinen.
I de angitte formler betyr A en fortrinnsvis ufor-grenet alkylgruppe med 1-6, foretrukket 1-4, særlig 1, 2 eller 3 C-atomer, nærmere bestemt fortrinnsvis metyl, videre foretrukket etyl, propyl, isopropyl, butyl, isobutyl, ytterligere foretrukket sek.-butyl, tert.-butyl, pentyl, isopentyl(3-metylbutyl), heksyl eller isoheksyl(4-metylpentyl).
R<1> og R2 er fortrinnsvis metyl eller etyl, foretrukket metyl.
Det er foretrukket at R<5> er A (særlig metyl, videre etyl, propyl, isopropyl, butyl eller isobutyl), benzyl, o-, m-eller p-metoksybenzyl, eller o-, m- eller p-metylbenzyl.
Resten R<6> står fortrinnsvis i 6-stilling i kroman-systemet. Den kan imidlertid også stå i 5- eller 7-stilling.
Restene R<6> er fortrinnsvis forskjellig fra H og er fortrinnsvis CN.
Gjenstand for oppfinnelsen er følgelig særlig fremstilling av de forbindelser med formel I, hvor minst én av de nevnte restene har en av de ovenfor angitte foretrukne betydninger. Noen foretrukne grupper av forbindelser kan uttrykkes gjennom de etterfølgende formler Ia - le, som omfattes av formel I og hvor de ikke nærmere angitte restene har de for formel I angitte betydninger, hvor imidlertid
Gjenstand for oppfinnelsen er en analogifremgangsmåte for fremstilling av et terapeutisk aktivt kromanderivat med formel I ifølge krav 1, kjennetegnet ved at et kroman med formel II

Forbindelsene med formel I fremstilles således etter i og for seg kjente fremgangsmåter som er beskrevet i littera-turen (f.eks. i standardverkene som Houben-Weyl, Methoden der organischen Chemie, Georg-Thieme-Verlag, Stuttgart; Organic Reactions, John Wiley & Sons, Inc., New York; samt i de ovenfor angitte patentsøknader), og da under reaksjonsbetingelser som er kjent og egnet for de nevnte omsetninger. Dessuten kan man også gjøre bruk av i og for seg kjente, her ikke nærmere nevnte, varianter.
Utgangsforbindelsene kan om ønsket også dannes in situ slik at man ikke isolerer dem fra reaksjonsblandingen, men øyeblikkelig omsetter dem videre til forbindelsene med formel I.
Fortrinnsvis fremstilles forbindelsene med formel I, hvor R<5> er forskjellig fra H ved omsetning av forbindelser med formel II med forbindelser med formel III, hensiktsmessig i nærvær av et inert oppløsningsmiddel ved temperaturer mellom 0
og 150°C.
Utgangsforbindelser med formel II hvor
(3,4-epoksy-kroman) er foretrukket.
Utgangsforbindelsene II og III er som regel kjent (jf. f.eks. DE-OS 37 26 261). Såfremt de ikke er kjent, kan de fremstilles etter i og for seg kjente fremgangsmåter. Så-
ledes lar utgangsforbindelsene med formel II
seg erholde ved omsetning av 2-hydroksyacetofenoner med formel 2-HO-R<6>R<7>C6H2-COCH3 med ketoner med formel R^CO-R<2> til tilsvarende 4-kromanoner med formel IVa

eventuelt kondensasjon med aldehyder med formel R<12->CHO (R<12> = alkyl med 1-5 C-atomer) til 3-alkyliden-4-kromanoner med formel IVb, reduksjon f.eks. med NaBH4 til kromanoler med formel IVc, dehydratisering f.eks. med p-toluensulfonsyre til kromener med formel IVd og oksidasjon f.eks. med 3-klorper-benzosyre. Den sistnevnte oksidasjon kan også skje i flere trinn. Således kan man f.eks. med N-bromsuccinimid i vandig oppløsning først fremstille bromhydrinet med formel IVe, og fra dette deretter avspalte HBr med en base, f.eks. natronlut.
Kromenene med formel IVd kan også erholdes ved kondensasjon av salisylaldehyder med formel 2-H0-R<6>R<7->C6H2-CH0 med ketoner med formel R<1->CO-CH2-R<8> til hydroksyketoner med formel 2-HO-R<6>R<7>C6H2-CH=CR8-CO-R<1>, omsetning med Organo-Li-forbindelser med formel R<2->Li og etterfølgende hydrolyse til dioler med formel 2-H0-R<6>R7C6H2-CH=CR<8->CR<1>R2-0H og ringslutning under vannavspalting.
I forbindelser med formel II (-X-Y- = -CHE-CR<3>R<8->) kommer det som "reaksjonsdyktig forestrede OH-grupper" særlig på tale med esterne med alkylsulfonsyrer (hvor alkylgruppen inneholder 1-6 C-atomer) eller med arylsulfonsyrer (hvor aryl-gruppen inneholder 6-10 C-atomer). Disse forbindelsene lar seg erholde fra 4-kromanolene med formel IVc ved omsetning med et uorganisk syrehalogenid som PC13, PBr3, S0C12 eller S0Br2 eller med et sulfonsyreklorid som metan- eller p-toluensulfonsyre-klorid.
Ved omsetningen av II med III er det hensiktsmessig å arbeide i nærvær av en base. Som baser egner seg fortrinnsvis alkalimetallalkyler, særlig metyllitium eller butyllitium, alkalimetalldialkylamider, særlig litiumdiisopropylamid, dessuten f.eks. alkalimetall- eller jordalkalimetallhydroksyder, -karbonater, -alkoholater, -hydrider eller også -amider som NaOH, KOH, Ca(0H)2, Na2C03, K2C03, Na- eller K-metylat, -etylat eller -tert.-butylat, NaH, KH, CaH2, NaNH2, KNH2, videre organiske baser som trietylamin eller pyridin, som også kan anvendes i overskudd og da samtidig tjener som oppløsnings-middel.
Som inerte oppløsningsmidler egner seg særlig alkoholer som.metanol, etanol, isopropanol, n-butanol eller tert.-butanol; etere som dietyleter, diisopropyleter, tetrahydro-furan (THF) eller dioksan; glykoletere som etylenglykolmono-metyl- eller -monoetyleter (metylglykol eller etylglykol), etylenglykoldimetyleter (diglym); ketoner som aceton eller butanon; nitriler som acetonitril; nitroforbindelser som nitrometan eller nitrobenzen; estere som etylacetat; amider som dimetylformamid (DMF), dimetylacetamid eller fosforsyre-heksametyltriamid; sulfoksider som dimetylsulfoksid (DMSO); klorerte hydrokarboner som diklormetan, kloroform, triklor-etylen, 1,2-dikloretan eller karbontetraklorid; hydrokarboner som benzen, toluen eller xylen. Blandinger av disse oppløs-ningsmidlene med hverandre er dessuten egnet.
Epoksydet II

situ, f.eks. ved innvirkning av en base på det tilsvarende bromhydrin IVe.
En særlig foretrukket arbeidsmåte består i at man først omsetter diisopropylamin med butyllitium i THF/heksan til litiumdiisopropylamid, bringer dette til reaksjon med pyridon III in situ, tilfører epoksydet II og koker til slutt i ca. 0,5-20 timer.
En forbindelse med formel I hvor R3 = OH og R<4>= H, kan omdannes til en forbindelse med formel I hvor R<3> og R<4 >tilsammen betyr en binding, ved behandling med et dehydrati-seringsmiddel. Det lykkes f.eks. ved innvirkning av en av de angitte baser, f.eks. NaOH eller NaH, i et av de angitte opp-løsningsmidler, f.eks. THF, dioksan eller DMSO, ved temperaturer mellom 0 og 150°C.
Dessuten kan man i en forbindelse med formel I om-danne en cyangruppe til en karbamoylgruppe eller omvendt.
Eksempelvis er det mulig at man omdanner en cyangruppe (f.eks. med KOH i tert.-butanol) til en karbamoylgruppe, og/eller dehydratiserer en karbamoylgruppe (f.eks. med P0C13) til en cyangruppe.
En base med formel I kan overføres med en syre til det tilsvarende syreaddisjonssalt. For denne omsetningen kommer det bare på tale med syrer som gir farmasøytisk akseptable salter. Således kan uorganiske syrer anvendes, f.eks. svovelsyre, salpetersyre, hydrogenhalogensyrer som saltsyre eller hydrogenbromsyre, fosforsyrer som ortofosforsyre, sulfaminsyre, videre organiske syrer, særlig alifatiske, ali-sykliske, aralifatiske, aromatiske eller heterosykliske en-eller flerbasiske karboksyl-, sulfon- eller svovelsyrer, f.eks. maursyre, eddiksyre, propionsyre, pivalinsyre, dietyl-eddiksyre, malonsyre, ravsyre, pimelinsyre, fumarsyre, malein-syre, melkesyre, vinsyre, eplesyre, benzosyre, salisylsyre, 2-eller 3-fenylpropionsyre, sitronsyre, glukonsyre, askorbin-syre, nikotinsyre, isonikotinsyre, metan- eller etansulfon-syre, etandisulfonsyre, 2-hydroksyetansulfonsyre, benzen-sulfonsyre, p-toluensulfonsyre, naftalin-mono- og disulfon-syrer og laurylsvovelsyre. Salter med fysiologisk ikke akseptable syrer, f.eks. pikrater, kan anvendes til rensing av forbindelsene med formel I.
Forbindelsene med formel I kan ha ett eller flere kirale sentra. De kan således ved fremstillingen erholdes som racemater eller, dersom det anvendes optisk aktive utgangsforbindelser, også i optisk aktiv form. Dersom forbindelsen har to eller flere kirale sentra, kan de ved syntesen falle ut som blandinger av racemater, hvorfra man kan isolere de enkelte racemater i ren form, f.eks. ved omkrystallisering fra inerte oppløsningsmidler. Således har f.eks. forbindelser med formel I hvor R<1>=R<2>, R3 = OH og R4 = H, to kirale sentra; ved fremstillingen ved hjelp av omsetning av II med III fås imidlertid i alt overveiende grad bare et racemat med transstil-ling av l-R<5->l,2-dihydro-2-okso-3-pyridylgruppen og substi-tuentene R<3> = OH. Om ønsket kan erholdte racemater oppløses mekanisk, kjemisk eller biokjemisk i sine enantiomerer etter i og for seg kjente metoder. Således kan det dannes diastereo-merer fra racematet ved omsetning med et optisk aktivt oppløs-ningsmiddel. Som oppløsningsmiddel for basiske forbindelser med formel I, egner seg f.eks. optisk aktive syrer, som D- og L-formene av vinsyre, dibenzoylvinsyre, diacetylvinsyre, kamfansyre, kamfersulfonsyre, mandelsyre, eplesyre eller melkesyre. Karbinoler (I, R<3> = OH) kan videre forestres ved hjelp av kirale acyleringsreagenser, f.eks. D- eller L-a-metylbenzylisocyanat, og så oppløses (jf. EP-A1-120 428). De forskjellige formene av diastereomerene kan separeres på i og for seg kjent måte, f.eks. ved fraksjonert krystallisasjon, og enantiomerene med formel I kan på i og for seg kjent måte fri-settes fra diastereomerene. Enantiomeroppløsning lykkes videre ved kromatografering på optisk aktive bærermaterialer.
Forbindelsene med formel I og deres farmasøytisk akseptable salter kan anvendes til fremstilling av farma-søytiske preparater, særlig på ikke-kjemisk måte. Således kan de bringes på en egnet doseringsform sammen med minst et fast, flytende og/eller halvflytende bærer- eller hjelpestoff, og eventuelt i kombinasjon med en eller flere ytterligere aktive forbindelser.
Forbindelsene med formel I kan således anvendes i farmasøytiske preparater som inneholder minst én av forbindelsene med formel I og/eller et av deres farmasøytisk akseptable salter.
Disse preparatene kan anvendes som legemidler i human- eller veterinærmedisinen. Som bærerstoffer kommer det på tale med organiske eller uorganiske stoffer, som egner seg for enteral (f.eks. oral), parenteral eller topisk applikasjon, og som ikke reagerer med de nye forbindelsene, f.eks. vann, planteolje, benzylalkoholer, polyetylenglykoler, glyse-roltriacetat, gelatin, karbohydrater som laktose eller stivel-se, magnesiumstearat, talkum, lanolin og vaselin. For oral anvendelse tjener særlig tabletter, dragéer, kapsler, siruper, safter eller dråper, for rektal anvendelse suppositorier, for parenteral anvendelse oppløsninger, fortrinnsvis olje- eller vannoppløsninger, videre suspensjoner, emulsjoner eller implantater, for den topiske anvendelse salver, kremer, pastaer, lotions, geléer, sprayer, skum, aerosoler, oppløs-ninger (f.eks. oppløsninger i alkoholer som etanol eller isopropanol, acetonitril, DMF, dimetylacetamid, 1,2-propandiol eller blandinger derav med hverandre og/eller med vann) eller pudder. De nye forbindelsene kan også lyofiliseres og de erholdte lyofilisater anvendes f.eks. til fremstilling av injek-sjonspreparater. Særlig for den topiske anvendelse kommer det i betraktning liposomale preparater. De angitte preparater kan være steriliserte og/eller inneholde hjelpestoffer som glide-, konserverings-, stabiliserings- og/eller fuktemiddel, emul-gatorer, salter for påvirkning av det osmotiske trykk, buffer-stoffer, farge-, smaks- og/eller aromastoffer. Om ønsket kan de også inneholde ett eller flere ytterligere aktive forbindelser, f.eks. ett eller flere vitaminer.
Forbindelsene med formel I og deres fysiologisk akseptable salter kan administreres til mennesker eller dyr, særlig pattedyr som aper, hunder, katter, rotter eller mus, og anvendes ved den terapeutiske behandling av menneske- eller dyrekroppen både ved bekjempelsen av sykdommer, særlig ved terapi og/eller profylakse av forstyrrelser i det kardiovaskulære system, særlig dekompensert hjerteinsuffisiens, angina pectoris, arytmi, perifere eller cerebrale karsykdommer, så vel som sykdomstilstander som er forbundet med høyt blodtrykk, videre av sykdommer som er forbundet med forandringer i den ikke-vaskulære muskulatur, f.eks. astma og urinblæreinkonti-nens.
Således administreres forbindelsene fremstilt ifølge oppfinnelsen som regel analogt med de kjente antianginosa eller blodtrykkssenkende midler, f.eks. "Nicorandil" eller "Cromakalim", fortrinnsvis i doseringer mellom ca. 0,01 og 5 mg, særlig mellom 0,02 og 0,5 mg, pr. doseringsenhet. Den daglige dosering ligger fortrinnsvis mellom ca. 0,0001 og 0,1, særlig mellom 0,0003 og 0,01 mg/kg kroppsvekt. Den spesielle dose for hver bestemte pasient avhenger imidlertid av de for-skjelligste faktorer, f.eks. aktiviteten av den spesielle forbindelse som anvendes, alderen, kroppsvekten, den generelle helsetilstand, arv, kosten, administreringstidspunkt og -vei, utskillingshastighet, legemiddelkombinasjon og den aktuelle sykdoms alvorlighet, som behandlingen gjelder. Den orale applikasjon er foretrukket.
Forbindelsene med formel I og deres salter egner seg særlig ved topisk anvendelse, videre for behandling av Alo-pecia areata. For dette anvendes særlig farmasøytiske preparater som er egnet for topisk behandling av hodehuden, og som er nevnte ovenfor. De inneholder ca. 0,005-10, fortrinnsvis 0,5-3 vekt% av minst en forbindelse med formel I, og/eller minst et salt derav. For øvrig kan disse forbindelsene anvendes mot alopesi analogt med angivelsene i WO 88/00822.
I eksemplene nedenunder betyr "vanlig opparbeidelse": Om nødvendig tilsettes vann, det ekstraheres med et organisk oppløsningsmiddel som etylacetat, det separeres, den organiske fase tørkes over natriumsulfat, filtreres, inndampes og renses ved kromatografi og/eller krystallisasjon.
Ovenfor og nedenunder er alle temperaturer angitt i
°C.
Eksempel 1
En oppløsning av 1,4 ml diisopropylamin i 10 ml THF ble under omrøring og innføring av N2 ved 0°C, tilsatt 6,7 ml av en 15 %-ig oppløsning av butyllitium i heksan. Deretter ble det tildryppet en oppløsning av 1,85 g N-benzyl-2-pyridon i 10 ml THF ved 0°C. Etter 0,5 timer med omrøring ble det tildryppet en oppløsning av 2 g 2,2-dimetyl-3,4-epoksy-6-cyan-kroman i 10 ml THF ved 0°C, det ble kokt i 2 timer og over-skytende butyllitium dekomponert ved tildrypping av metanol. Etter vanlig opparbeidelse fikk man 2,2-dimetyl-4-(1-benzyl-1,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol, sm.p. 120-122°C.
Analogt fikk man de tilsvarende pyridoner: 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyri-dyl)-6-brom-3-kromanol, sm.p. 192-193°,
2,2-dimetyl-4-(1-metyl-l,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol ("A"), sm.p. 247-248,5°C, og
2,2-dimetyl-4-(1-p-metoksybenzyl-l, 2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol, sm.p. 120°.
Eksempel 2
En oppløsning av 2,82 g 2,2-dimetyl-4-brom-6-cyan-3-kromanol og 1,09 g 1-metyl-l,2-dihydro-2-okso-pyridin i 70 ml DMSO ble tilsatt 1,2 g 80 %-ig NaH og omrørt i 3 dager ved 20°C. Etter vanlig opparbeidelse fikk man "A", sm.p. 247-248,5°C.
Eksempel 3
En blanding av 384 mg 2,2-dimetyl-4-[l-(2-metoksy-etoksymetyl)-l,2-dihydro-2-okso-3-pyridyl]-6-cyan-3-kromanol [som fås av l-(2-metoksyetoksymetyl)-l,2-dihydro-2-okso-pyridin og 2,2-dimetyl-3,4-epoksy-6-cyan-kroman], 600 mg finpul-verisert ZnBr2 og 4 ml diklormetan ble omrørt i 16 timer ved 25°C. Det ble vasket med mettet NaHC03-oppløsning, så med mettet NaCl-oppløsning, vaskeoppløsningene ble ekstrahert med eter, de organiske fasene slått sammen og det ble opparbeidet på vanlig måte. Det ble erholdt 2,2-dimetyl-4-(1,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol, sm.p. 246-247°.
Eksempel 4
Til en oppløsning av 1 g 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol i 40 ml dioksan ble det under omrøring og innføring av N2 tilsatt 0,4 g NaH, deretter kokt i 10 timer, inndampet og opparbeidet på vanlig måte. Det ble erholdt 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromen, sm.p. 130-132°.
Analogt fikk man fra den tilsvarende 3-kromanol: 2,2-dimetyl-4-(1-metyl-l,2-dihydro-2-okso-3-pyridyl)-
6-cyan-3-kromen, sm.p. 187-188°.
Eksempel 5
En blanding av 5,8 g 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl )-6-cyan-3-kromanol ble kokt i 50 minutter, avkjølt og helt over på is. Etter vanlig opparbeidelse fikk man 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-3-kromen, sm.p. 199-202°.
Eksempel 6
En oppløsning av 4 g 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-3-kromen i 200 ml metanol ble hydrogenert med 4 g 20 %-ig Pd-C ved 20°C og 1 bar, det ble filtrert, inndampet og erholdt 2,2-dimetyl-4-(1, 2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-kroman, sm.p. 177-179°C.
Eksempel 7
En blanding av 2,98 g 2,2-dimetyl-4-(1,2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-kroman, 2 ml P0C13 og 200 ml 1,2-dikloretan ble kokt i 45 minutter. Etter avkjøling og vanlig opparbeidelse fikk man 2,2-dimetyl-4-(1, 2-dihydro-2-okso-3-pyridyl)-6-cyan-kroman, sm.p. 174-175,5°C.
Eksempel 8
En oppløsning av 280 mg 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-brom-3-kromanol i 80 ml metanol ble hydrogenert med 140 mg 20 %-ig Pd-C ved 20°C og 1 bar, det ble filtrert, inndampet og erholdt etter kromatografisk rensing på silikagel 2,2-dimetyl-4-(1,2-dihydro-2-okso-3-pyri-dyl)-3-kromanol, sm.p. 244-246°C.
Eksempel 9
En blanding av 1 g 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol og 10 ml 85 %-ig fosforsyre ble oppvarmet til 100°C i 2 timer. Etter avkjøling og vanlig opparbeidelse fikk man 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-3-kromanol, sm.p. 230-235°C.
Eksempel 10
Analogt med eksempel 9 fikk man fra 2,2-dimetyl-4-(1-benzyl-1,2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-3-kromanol med H20 på 20 %-ig Pd-C i metanol 2,2-dimetyl-4-(1,2-dihydro-2-okso-3-pyridyl)-6-karbamoyl-3-kromanolen, intet smeltepunkt opp til 280°C.
Eksempel 11
Analogt med eksempel 4 fikk man fra 2,2-dimetyl-4-(1-benzyl-1,2-dihydro-2-okso-3-pyridyl)-6-brom-3-kromanol og NaH i dioksan 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyri-dyl)-6-brom-3-kromen (sm.p. 118-119°C), som analogt med eksempel 9 ble hydrogenert til 2,2-dimetyl-4-(1,2-dihydro-2-okso-3-pyridyl)-kroman, sm.p. 185-186°C.
De etterfølgende eksempler vedrører farmasøytiske preparater som inneholder forbindelser med formel I og/eller fysiologisk akseptable salter derav.
Eksempel A: Tabletter
En blanding av 0,2 kg "A", 136,3 kg kalsiumhydrogen-fosfat, 15 kg maisstivelse, 10 kg mikrokrystallinsk cellulose, 5,5 kg uoppløselig polyvinylpyrrolidon (PVP), 1,5 kg høydis-perst silisiumdioksid og 1,5 kg magnesiumstearat, ble presset på vanlig måte til tabletter. Hver tablett på 170 mg inneholdt 0,2 mg aktiv forbindelse.
Eksempel B: Dragéer
Analogt med eksempel A, imidlertid uten tilsetning av PVP, ble det presset tabletter som deretter ble overtrukket på vanlig måte med et belegg av sakkarose, maisstivelse, talkum, tragant og fargestoff.
Eksempel C: Lakktabletter
Fra 0,2 kg 2,2-dimetyl-4-(1-benzyl-l,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromanol, 151,3 kg laktose, 10 kg mikrokrystallinsk cellulose, 5,5 kg uoppløselig PVP, 1,5 kg høydisperst silisiumdioksid og 1,5 kg kalsiumstearat ble det presset tablettkjerner (170 mg), som deretter ble lakkert på vanlig måte slik at hver lakktablett var overtrukket med 3,922 mg av en lakk som bestod av 2,2 mg hydroksypropylmetylcellulose, 0,53 mg polyetylenglykol 400, 0,85 mg titandioksid, 0,12 mg jern(III)-oksyd (gult), 0,002 mg jern(III)-oksyd (rødt) og 0,22 mg silikonolje.
Eksempel D: Kapsler
Det ble tilberedt et granulat av 10 g 2,2-dimetyl-4-(1-metyl-l,2-dihydro-2-okso-3-pyridyl)-6-cyan-3-kromen,
27,5 kg laktose, 0,35 kg hydroksypropylmetylcellulose og 0,7 kg maisstivelse, dette ble blandet med 0,15 kg høydisperst silisiumdioksid og 0,3 kg magnesiumstearat, og blandingen ble fylt på vanlig måte i hardgelatinkapsler, slik at hver kapsel inneholdt 0,1 mg aktiv forbindelse.
Eksempel E: Ampuller
En oppløsning av 10 g "A" i 70 1 1,2-propandiol, ble oppfylt til 100 1 med to ganger destillert vann, sterilfilt-rert og fylt i 1 ml ampuller, som så ble lukket sterilt. Hver ampulle inneholdt 0,1 mg aktiv forbindelse.

1. Analogifremgangsmåte for fremstilling av et terapeutisk aktivt kromanderivat med formel I

A betyr alkyl med 1-6 C-atomer,
samt farmasøytisk akseptable syreaddisjonssalter derav, karakterisert ved at et kroman med formel II
