Description
Denne oppfinnelse angår en analogifremgangsmåte ved fremstilling av nye, farmakologisk aktive nitroostratriener, spesielt en ny klasse av 9a-hydroxy-lip-nitro-l,3,5(10)-ostratriener av den generelle formel: Q^ ^^^ J^ ,R„

hvor R er et hydrogenatom, et eventuelt halogen substituert alkylradikal med 1-6 carbonatomer, et cycloalkylraoikal med 5-6 carbonatomer eller et alifatisk eller cycloalifatisk acylradikal med 1 - 10 carbonatomer, R' er et hydrogenatom, et mono- eller di carboxy1syre-eller cycloalifatisk acylradikal med i - 10 carbonatomer, og R" er
et hydrogenatom eller et alkynylradikal med 2 - h carbonatomer og OR' og R" til sammen kan være oxygen.
De forbindelser som fremstilles ifolge fremgangsmåten viser interessant ostrogene, antigonadotrope og antiovulatorisk aktivitet og er spesielt virksomme når de administreres oralt.
De nye forbindelser fremstilles ved en analogifremgangsmåte som kan anvendes på steroider i hvilke A-ringen er aromatisk og substituert, med en aktivert gruppe, og som dessuten har en dobbelt-binding i 9(ll)-stillingen.
Fremgangsmåten for fremstilling av de nye forbindelser består deri at en forbindelse av den generelle formel:
hvor R, R' og R" har de ovenfor angitte betydninger, og som er opplost i et egnet opplosningsmiddel, såsom diisopropylether, dioxan, diethylether, ethylaceta.t eller ni'tromethan, omsettes med et nitreringsmiddel, såsom rykende salpetersyre og nitrogendioxyd, ved en temperatur fra -^O<0> C til romtemperatur i vannfri atmosfære. Det ekstraheres med et egnet organisk opplosningsmiddel, såsom ethylacetat, diethylether eller benzen, hvoretter ekstraktene nbytrali-seres ved tilsetning av natriumbicarbonat og nitrohydrosteroidet deretter isoleres ved krystallisasjon eller kromatografering.
Den ostrogene aktivitet ble provet på hunrotter, som minst
10 dager for behandlingen var blitt .coforektomisert og viste diestrus-utsondring. Forbindelsen ble administrert i en enkelt dose, enten subcutant eller oralt. De vaginale utsondringer ble de to folgende dager kontrollert og klassifisert etter Allen og Doisy.
Den minste virksomme dose (MED) er den mengde av forbindelsen som er nodvendig for å forårsake en Proestrus hos to av tre dyr eller en estrus hos et av to dyr.
Den antiovulatoriske aktivitet ble provet på hunrotter som hadde en regelmessig estrus-syklus, som ble"kontrollert minst i 15
på hverandre fblgende dager. Forbindelsen ble administrert subcutant eller oralt en gang om dagen i 5 på hverandre folgende dager, begynnende med den siste metestrus. Den venstre og hbyre eggstokk ble fjernet på den lM-nde henholdsvis den 15nde dag. Egglederne, ble fraskilt og undersokt på innhold av egg.-
Den effektive dose ^Q angir den mengde av forbindelsen som
er nodvendig for å forhindre ovulasjonen hos 50% av de behandlede dyr.
Den antigonadotrope aktivitet ble provet på parabiotiske rotter (kastrerte hanrotter - friske hunrotter). Forbindelsen ble administrert subcutant eller oralt på hanrottene i 10 på hverandre folgende dager.
Den effektive dose ^ angir den mengde av forbindelsen som
er nodvendig for å avstedkomme en 50%-ig hemning av eggstokkvekten hos hunrottene blant de ubehandlede kontrolldyr.
De forbindelser som fremstilles ifolge fremgangsmåten inndeles i tre grupper etter deres kjemiske struktur og deres farmakologiske virkning, i sammenligning med virkningen av ostradiol, ostron og 17cc-ethinyl-ostradiol. Disse resultater er angitt i tabellen.
Det fremgår av tabellen at 3-methoxy-llp-nitro-l,3,5(10)-ostratrien-9a , 17j3-diol og 3-methoxy-lip-nitro-l, 3 ,5(10) -ostratrien 9a,17p-diol-17-acetat oppviser en strogen aktivitet som er nesten 30 ganger hbyere enn for standardforbindelsen 1,3,5(10)-ostratrien-3,17P-diol. Den antiovulatori ske aktivitet av disse to forbindelser er, ved subcutan administrering, omtrent som for standardforbindelsen, mens den er minst 6-10 ganger, hoyere ved oral administrering.
Blant disse ostronderivater oppviser ll(3-nitro-l ,3 , 5- (10) - ostratrien-3 , 9a-d i ol-17-on, 3_me~thoxy-lip-nitro-l, 3, 5(10)-ostratrien-9a-ol-17-on og (+)-3-methoxy-llp-nitro-l,3,5(10)-ostratrien-9a-ol-17-on meget interessant aktivitet såvel ved oral som ved'subcutan administrering. Det er å merke at naturlige forbindelser (ostron og ostradiol), som ikke har noe alkylradikal i'17-stillingen, ikke'oppviser noen bemerkelsesverdig aktivitet ved oral administrering.
Den antiovulatoriske aktivitet av disse forbindelser er
ved oral administrering fra 2 til 20 ganger aktiviteten av ostron. Det er bemerkelsesverdig at 3-acetoxy-lip-nitro-l,3,5(10)-ostratrien-9a-ol-17-on ved oral administrering oppviser en antiovulatorisk aktivitet som er minst 15 ganger hoyere enn for ostron, mens dets ostrogene aktivitet ved oral administrering er omtrent den samme som for standardforbindelsen.
Blant ethynyl-ostradiol-3-methyl-etherderivatene er 3-methoxy-lip-nitro-17a-ethynyl-1,3, 5(10)-ostratrien-9a, 17p-ciiol aktiv ved såvel oral administrering som ved subcutan administrering, hva angår de her omtalte virkninger.
Dette produkt har 10 ganger hoyere ostrogen aktivitet og
30 ganger hoyere antiovulatorisk aktivitet enn det.analoge 3-methoxy-17a-ethynyl-ostradiol. 3-methoxy-ll|3-nitro-17a-ethynyl-l,3,5(10)-ostratrien-9a,17|3-diol har ved oral administrering 3 ganger hbyere bstr.ogen aktivitet og 8 ganger hbyere antiovulatori sk aktivitet enn ethynylbstradiol, som hittil har vært ansett som den mest aktive stand ardforbindelse.
De nedenstående eksempler vil ytterligere illustrere oppfinnelsen.
Eksempel 1
3- methoxy- lip- nitro- l. 3. 5( 10)- bstratrien- 9a. 17p- diol- 17- acetat
Til 1,8 g 3-methoxy-l,3, 5(10) ,9(lD-bstratrien-17p-ol-acetat opplost i 80 ml vannfri diethylether tilsettes ved 0°C og under omroring 12 ml rykende salpetersyre (spesifikk vekt 1,52). Blandingen omrores i 2 timer, idet temperaturen holdes ved 0°C. Deretter tilsettes is, hvoretter det ekstraheres tre ganger med diethylether. Ekstraktene vaskes nbytrale med vann, deretter med en 5%-ig natrium-bicarbonatopplosning og tilslutt med vann. Opplbsningen tbrres så over vannfritt natriumsulfat. Etter filtrering avdestilleres opp-lbsningsmidlet. Det oppsamles et residuum på 2,2 g, som omkrystalli-seres fra diethylether, hvorved det erholdes 1 g 3-methoxy-llp-nitro-1,3,5(10)-bstratrien-9a,17p-diol-17-acetat. Sm.p.: 180 - 182°C,
|-a-j <20>°= -^-9° (fra kloroform).

Til 1 g 3-raethoxy-l ,3, 5(1°) , 9(H)-bstratetraen-17-on opplost i 50 ml vannfri diethylether tilsettes ved 0° C 6 ml rykende salpetersyre (spesifikk vekt 1,52). Blandingen omrores i 30 minutter ved 0° C, hvoretter den fortynnes med isvann og steroidet ekstraheres med diethylether. Etherekstraktene vaskes med en 5$-ig natriumbicarbonatopplbsning og deretter med vann, tbrres over vannfritt natriumsulfat og inndampes til tbrrhet i vakuum. Residuet krystalliseres fra aceton/petrolether, hvorved man får 3-methoxy-llfS-nitro-1,3,5(10)-bstratrien-9a-ol-17-on. Sm.p.: 218 - 221°C; [Q]20° =

En opplbsning av ^80 mg 3-methoxy-1,3,5(10),9(11)-ostratetraen-17(3-ol i 25 ml vannfri diethylether, holdt ved -5°C, tilsettes under omrbring 3 ml rykende salpetersyre' (spesifikk vekt 1,52). Temperaturen holdes ved 0°C og oppløsningen omrores i 20 minutter, hvoretter den fortynnes med vann og is og steroidet ekstraheres med ethylacetat. Ekstraktene vaskes nbytrale, forst med en 5$-ig natriumbicarbonatopplbsning og deretter med vann, tbrres over vannfritt natriumsulfat og inndampes til tbrrhet. 3-methoxy-ll|3-nitro-l,3, 5(10)-bstratrien-9a,17P~diol skilles ut som en krystallinsk forbindelse, men utfelles imidlertid som et skum fra petrolether, hvilket skum behandles med eddiksyreanhydrid for dannelse av det tilsvarende 17-acetat, som har samme egenskaper som forbindelsen ifolge eksempel 1.

En opplbsning av 2 g 3-acetoxy-l,3,5(10),9(11)-bstratetraen-17-on i 100 ml vannfri diethylether tilsettes ved 0°C og under omrbring 15 ml rykende salpetersyre (spesifikk vekt 1,52). Temperaturen holdes ved 0°c. Etter tilsetning av syren omrores reaksjons-blandingen ytterligere i 30 minutter ved 0°C, hvoretter den fortyn-
nes med vann og is og steroidet ekstraheres med diethylether. De organiske skikt slåes sammen, vaskes til nbytral reaksjon med en 5%-ig natriumbicarbonatopplbsning og deretter med vann, tbrres over vannfritt natriumsulfat og inndampes til tbrrhet. Residuet gir ved omkrystallisering fra aceton/diethylether 3_acetoxy-llp-nitro-l,3,5_ (10)-bstratrien-9a-ol-17-on med smeltepunkt 228 - 235°C. Etter omkrystallisering av forbindelser fra aceton/diethylether oker smelte-punktet til 235 - 238°C; [a]<2>°° = +3^° (CHC13);X C2H^0H - 268 m^;

En opplbsning av 700 mg 3-methoxy-17a-ethynyl-1,3, 5( 1°)-9(H)-bstrat.e tråen -17B-ol i 35 ml vannfri diethylether, som er kjblt
til -20° C, tilsettes '+,2 ml rykende salpetersyre. Temperaturen holdes ved -20°C, og blandingen omrores i 20 minutter, hvoretter don fortynnes med 50 mi av en 20%-ig natriumacetatopplosning og to ro kl ot ekstraheres med diethylether. 3-( 2'- klorethoxy)- llB- nitro- 1, 3. 5( 10)- bstratrien- 9a- ol- 17- on
2,^5 g 1,3,5(10),9(ll)-bstratetraen-3,ol-17-on ble behandlet ned ethylencarbonat i en natriumamylatlbsning, hvorved det ble erholdt 1,76 g 3-(2'-hydroxy-ethoxy)-l,3, 5(10) ,9(H)-bstratetraen-17-on som deretter ble omsatt med mesylklorid i pyridin, og deretter med lithiumklorid i isopropylalkohol, hvorved det ble erholdt 1,62 g 3-(2'-klorethoxy)-l,3, 5(10) ,9(H)-ostratetraen-17-on. 1,62 g urent 3-(2'-klorethoxy)-l,3,5(10),9(11)-bstratetraen-17-on ble opplost i L00 ml vannfri ethylether. Lbsningen ble avkjblt til -15 C, og 2 ml litrogendioxyd som var lost i 20 ml vannfri ether ble tilsatt under Dmrbrinf* i lbpet av 10 minutter. 3- methoxy- llB- nitro- l. 3, 5( 10)- ostratrien- 9a, 17B- diol 17- cyclopentylpropionat 1 g uren 3-methoxy-l,3,5(10), 9(11)-ostratetraen-17B-ol cyclopentylpropionat erholdt ved omsetning av 1,2 g 3-methoxy-l,3,5
(10),9(ll)-ostratetraen-17B-ol og 1 ml cyclopentylpropionylklorid i
10 ml pyridin, ble opplost i 50 ml nitromethan, hvoretter den ethylen-avkjolte losning til - 10°C ble tilsatt under omroring og i lopet av 10 minutter, 0,7 ml hitrogendioxyd lost i 10 ml nitromethan. Losningen ble omrort ved -10°C i 10 minutter, og deretter 5 minutter ved romtemperatur, og ble helt over i en 5%-ig natriumbicarbonatlosning. Steroidet ble ekstrahert flere ganger med ethylether, de organiske
lag ble samlet, vasket med vann til noytral reaksjon, torket over vannfritt natriumsulfat og fordampet til torrhet.
Residuet ble renset ved kromatografi. 3-methoxy-llB-nitro-1,3,5(10)-ostratrien-9a,17B-diol 17-cyclopentylpropionat ble erholdt og krystallisert fra ethylether-petroleumether. Sm.p.: 1^5 - 1<1>+6°C:

Claims (1)
- Analogifremgangsmåte ved fremstilling av farmakologisk aktive nitroostratriener av den generelle formel:hvor R er et hydrogenatom, et eventuelt halogensubstituert alkylradikal med 1-6 carbonatomer, et cycloalkylradikal med 5-6 carbonatomer eller et alifatisk eller cycloalifatisk acylradikal med 1-10 carbonatomer, R' er et hydrogenatom, en mono- eller dicarboxylsyre- eller cycloalifatisk acylrest med 1-10 carbonatomer, og R" er et hydrogenatom eller et alkynylradikal med 2 - h carbonatomer og OR' og R" tilsammen kan være oxygen, karakterisert ved- at en forbindelse av den generelle formel:hvor R, R' og R" har de ovenfor angitte betydninger, omsettes med et egnet nitreringsmiddel som består av rykende salpetersyre eller nitrogendioxyd ved en temperatur mellom -<>>+0°C og romtemperatur, og at nitrosteroidet deretter isoleres.
