Description
Bakgrunn
Foreliggende oppfinnelse angår en analogifremgangsmåte for fremstilling av en terapeutisk aktiv forbindelse med formel I,
hvor A er COOR, hvor R er H, laverealkyl eller NH2. I de fremstilte forbindelser er tre olifiniske enheter fra den syreholdige gruppen i retinsyre blitt erstattet med en etynyl-fenylfunksjon. Slike modifikasjoner i retinsyreststrukturen gjør at slike forbindelser beholder sin retinsyrelignende aktivitet.
Tidligere kjente fremgangsmåter
Nematocide forbindelser som er beskrevet i japansk patent 56-123903, har strukturen 2-(2-(((1,1-dimetyl)-dimetylsilyl)oksy-)etyl-alfa-(4-(2,6,6-trimetyl-l-cykloheksen-l-yl)-3-buten-l-ynyl)-1-cyklopenten-l-metanol og bruker l-(2',6',6'-trimetylcykloheks-l'-enyl)-but-l-en-3-yngruppen i de forbindelser som er beskrevet her. Dette fragmentet av molekylet er imidlertid den eneste likheten mellom de japanske forbindelser og de som er beskrevet her, og det foreligger derfor intet nært slektskap med de heri fremstilte forbindelser.
De fremstilte forbindelser med formel I kan anvendes for behandling av dermatoser, f.eks. filipenser, Dariers sykdom, psoriasis, ictyosyse, eksem, atopisk dermatittisk og epitelisk kreft. Forbindelsene kan også brukes for behandling av artritiske sykdommer og andre immunologiske lidelser (f.eks. lupus erytematosus), for å fremme groing av sår og ved behandling av det såkalte tørrøyesyndromet.
I farmasøytiske preparater kan en forbindelse med formel I foreligge i blanding med et farmasøytisk akseptabelt fortynnings-middel.
I fremgangsmåten ifølge oppfinnelsen omsetter man en forbindelse med formel II med en forbindelse med formel III hvor A er som angitt ovenfor og X er halogen, og eventuelt hydrolyserer en forbindelse med følgende formel
hvor A er COOR og R er laverealkyl, eller om ønsket
når A er COOH, omdanner syren til et salt; eller når A er COOH, omdanner syren til en ester; eller når A er COOH, omdanner syren til et amid.
Generelle utførelser
Definisjoner
Begrepet "ester" slik det brukes her, refererer seg til og dekker enhver forbindelse som faller innenfor den definisjon som klassisk brukes i den organiske kjemi. Begrepet innbefatter både organiske og uorganiske estere. Når A er -COOH, dekker dette begrepet de produkter som avledes fra behandling av denne funksjonen med alkoholer eller tioalkoholer. Når esteren er fremstilt fra forbindelser hvor A er -CH2OH, vil begrepet dekke forbindelser som er fremstilt fra organiske syrer som er istand til å danne estere, slik som fosforbaserte og svovelbaserte syrer eller forbindelser med formelen -CH2OCOR, hvor R er enhver substituert eller usubstituert, alifatisk, aromatisk, heteroaromatisk eller alifatisk-aromatisk gruppe.
Foretrukne estere fremstilles fra mettede alifatiske alkoholer eller syrer med ti eller færre karbonatomer, eller cykliske eller mettede alifatiske cykliske alkoholer og syrer med fra 5-10 karbonatomer. Spesielt foretrukne alifatiske estere er de som er fremstilt fra lavere alkylsyrer og alkoholer. Med begrepet lavere alkyl forstås i denne forbindelse og i den etterfølgende tekst alkylgrupper med fra 1-6 karbonatomer. Foretrukket er også fenyl eller lavere alkylfenylestere.
Amider har samme klassiske betydning som ellers i den organiske kjemi. I dette tilfelle innbefatter begrepet usubstituerte amider og alle alifatiske og aromatiske mono- og di-substituerte amider. Foretrukne amider er mono- og di-substituerte amider fremstilt fra mettede alifatiske rester med 10 eller færre karbonatomer, eller cykliske eller mettede alifatisk-cykliske rester med fra 5-10 karbonatomer. Spesielt foretrukne amider er de som er fremstilt av usubstituerte eller substituerte lavere alkylaminer. Foretrukket er også mono- og di-substituerte amider fremstilt fra substituerte og usubstituerte fenyl- eller lavere alkylfenylaminer. Usubstituerte amider er også foretrukket.
Et farmasøytisk akseptabelt salt kan fremstilles av enhver forbindelse fremstilt ifølge foreliggende oppfinnelse med en syregruppe. Et farmasøytisk akseptabelt salt er ethvert salt som bibeholder aktiviteten fra utgangsforbindelsen og som ikke gir noen skadelige eller uønskede effekter på den pasient som tilføres eller kommer i kontakt med preparatet.
Farmasøytisk akseptable salter kan fremstilles fra organiske eller uorganiske syrer eller baser. Saltet kan være et mono- eller polyvalent ion. Av spesiell interesse er de uorganiske ioner som natrium, kalium, kalsium og magnesium. Organiske salter kan fremstilles med aminer, da spesielt ammoniumsalter såsom mono-, di- og trialkylaminer eller etanolaminer. Salter kan også fremstilles med kaffein, trometamin og lignende molekyler.
De sterkest foretrukne forbindelser er:
etyl 4-[4</->(2",6",6"-trimetylcykloheks-l"-enyl)-but-3'-en-1'-ynyl]benzoat;
4-[4</->(2",6",6"-trimetylcykloheks-l"-enyl)-but-3'-en-1'-ynyl]benzosyre; og
4-[4'-(2",6",6"-trimetylcykloheks-l"-enyl)but-3'-en-1'-ynyl]-benzamid.
Forbindelsene kan gis systemisk eller topisk avhengig av de tilstander som skal behandles, behovet for posisjons-spesifikk behandling, den mengde som skal tilføres og tallrike andre hensyn.
Ved behandling av dermatoser vil man normalt tilføre den aktive ingrediensen topisk, skjønt man i visse tilfeller,
f.eks. ved behandling av alvorlige cystiske filipenser eller psoriasis, så kan man også bruke oral tilførsel. Man kan bruke enhver kjent topikal preparatform, f.eks. en oppløsning, suspensjon, en gel, en salve eller lignende. Fremstillingen av slike topikale preparater er velkjente i den farmasøytiske industrien og er f.eks. beskrevet i Remington/ s Pharmaceutical Science, utgave 17, Mack Publishing Company, Easton, Pennsylvania. For topikal tilførsel kan disse forbindelser også tilføres i form av et pulver eller et forstøvingspreparat, spesielt i aerosol form.
Hvis den aktive ingrediens skal tilføres systemisk, kan den opparbeides i form av et pulver, en pille, tablett eller lignende, eller som en sirup eller en eliksir som er regnet for oral tilførsel. For intravenøs eller intreperitonal tilførsel kan forbindelsen opparbeidet i form av en oppløsning eller suspensjon som kan tilføres ved injeksjon. I visse tilfeller er det fordel-aktig å opparbeide forbindelsene i form av et suppositorium eller som et preparat med forsinket frigjøring for avsetning under huden eller som en intramuskulær injeksjon.
Andre aktive ingredienser kan tilsettes slike topikale preparater for sekundære formål for behandling av hudtørrhet, for beskyttelse mot lys eller for behandling av dermatoser, for å hindre infeksjon, redusere irritasjon, inflammasjon og lignende.
Behandling av dermatoser eller andre tilstander hvor man vet eller har oppdaget at de kan behandles med retinsyrelignende forbindelser, vil bli påvirket ved tilførsel av en terapeutisk effektiv dose av én eller flere forbindelser ifølge foreliggende oppfinnelse. En terapeutisk konsentrasjon vil være den konsentrasjon, som reduserer den\ spesielle tilstand man ønsker å behandle eller forsinker dens utvikling. I visse tilfeller kan forbindelsen brukes profylaktisk for å hindre utviklingen av en spesiell tilstand.
En brukbar terapeutisk eller profylaktisk konsentrasjon vil variere fra tilstand til tilstand, og kan i visse tilfeller også variere med graden av den tilstand som skal behandles og pasien-tens reaksjon på behandlingen. Man kan således ikke bruke en ensartet konsentrasjon i hvert enkelt tilfelle, men dosen vil måtte modifiseres avhengig av den lidelse og den pasient som skal behandles. Slike konsentrasjoner kan fastlegges ved hjelp av rutineeksperimenter. Det er imidlertid antatt at man ved behandling av f.eks. filipenser eller lignende dermatoser kan bruke et preparat som inneholder mellom 0,01 og 1,0 mg/ml preparat og at dette vil være en terapeutisk effektiv konsentrasjon for dette formål. Hvis den aktive ingrediens tilføres systemisk, vil man få et effektivt terapeutisk resultat hvis man tilfører den aktive ingrediensen i mengder på mellom 0,01 og 5 mg/kg kroppsvekt pr. døgn ved behandling av de mange lidelser som disse forbindelser kan brukes for.
Den retinsyrelignende aktiviteten for disse forbindelser ble fastslått ved hjelp av en klassisk måling av retinsyreaktivi-tet som innbefatter effektene av forbindelsen på ornitindekarboksylase. Det opprinnelige arbeid som klargjorde korrelasjonen mellom retinsyre og nedsatt celleformering ble utført av Verma & Boutwell, Cancer Research. 1977. 37. 2196-2201. Denne referanse beskriver at ornitindekarboksylase (ODC)-øktiviteten økte forut for en polyaminbiosyntese. Det har blitt fastslått på annet hold at en økning i polyaminsyntesen kan korroleres eller forbindes med cellulær formering. Hvis således ODC-aktiviteten kunne hemmes, ville man kunne dempe cellenes hyperformering. Skjønt man ikke kjenner alle årsaker til ODC-aktivitetsøkningen, er det kjent at 12-0-tetradekanoylforbol-13-acetat (TPA) induserer ODC-aktivitet. Retinsyre hemmer denne induksjonen av ODC-aktiviteten på grunn av TPA. Forbindelser ifølge foreliggende oppfinnelse hemmer også TPA-induksjonen av ODC slik dette kan vises ved en prøve som i alt vesentlig følger den fremgangsmåte som er angitt i Cancer Res.: 1662-1670, 1975.
Spesifikke utførelser
For å illustrere oppfinnelsen har man her beskrevet en serie trinn som har vist seg å tilveiebringe forbindelser med formel I, når de settes sammen til en syntese av den type som er beskrevet i det etterfølgende. En syntetiker vil lett kunne fastslå at de reaksjonsbetingelser som er angitt her er spesifikke utførelser som lett kan generaliseres til enhver av de forbindelser som er angitt med formel I.
Forbindelser med formel I ble fremstilt som følger:
I det ovennevnte reaksjonsskjemaet er X halogen. Resten
R' er en lavere alkylrest i dette reaksjonsskjéma.
Forbindelser med formel 2 fremstilles fra deres tilsvarende syrer. Disse syrer (formel I) er alle tilgjengelige fra kjemiske laboranter eller fabrikker, eller kan fremstilles ved kjente fremgangsmåter. Forestering utføres ved å koke syren under tilbakeløp i en oppløsning av den passende alkoholen i nærvær av tionylklorid. Koking under tilbakeløp i fra 2-5 timer gir den ønskede esteren. Esteren gjenvinnes og renses på vanlig kjent måte.
Forbindelsen med<1> formel 3 selges av Aldrich Chemical Company under navnet Beta-Ionone. Den acetyliniske funksjon introduseres ved hjelp av litiumdiisopropylamid eller en lignende base ved redusert temperatur i en inert atmosfære. Reaksjonen utføres i et oppløsningsmiddel av etertypen, f.eks. en dialkyleter eller en cyklisk eter, f.eks. tetrahydrofuran, pyran eller lignende.
Mer spesielt kan det angis at litiumdiisopropylamid kan utvikles in situ ved å blande diisopropylamin i et tørt oppløs-ningsmiddel såsom tetrahydrofuran, som deretter avkjøles til -70 og -50'C i en inert atmosfære. En ekvimolar mengde av en alky-litiumforbindelse, f.eks. N-butyllitium i et passende oppløs-ningsmiddel, blir så tilsatt ved nevnte reduserte temperatur, og blandes i tilstrekkelig langt tidsrom til at man får dannet litiumdiisopropylamid (LDA). Ketonet med formel 3 (i minst et molart overskudd på 10 prosent) oppløses i samme oppløsningsmid-del som ble brukt for selve hovedreaksjonen, hvoretter oppløsnin-gen avkjøles til samme temperatur som LDA-blandingen, og det hele blandes. Oppløsningen blir så behandlet med dialkylklorfosfat, fortrinnsvis dietylklorfosfat i et molart overskudd på ca. 2 0 prosent. Reaksjonsblandingen oppvarmes deretter gradvis til romtemperatur. Denne oppløsning blir så tilsatt en ny litiumdi-isopropylamidoppløsning (ca. 2 ekvivalenter) som fremstilles in situ ved å bruke tørt oppløsningsmiddel og en inert atmosfære, fortrinnsvis argon, ved en redusert temperatur (f.eks. -78°C). Deretter blir reaksjonsblandingen igjen oppvarmet til romtemperatur hvor den blir rørt i et lengre tidsrom, fortrinnsvis mellom 10 og 20 timer, mest foretrukket ca. 15 timer. Oppløsningen blir så surgjort, og produktet med formel 4 kan så gjenvinnes på vanlig kjent måte.
Forbindelser med formel- 5 fremstilles under betingelser hvor man har utelukket vann og oksygen. Et tørt oppløsningsmiddel av etertypen, f.eks.a en dialkyleter eller en cyklisk eter såsom furan eller pyran, da spesielt tetrahydrofuran, kan brukes som oppløsningsmiddelet. En oppløsning av forbindelsen med formel 4 fremstilles først under en inert atmosfære, f.eks. av argon eller nitrogen, hvoretter man tilsetter en sterk base såsom N-butyllitium (i et molart overskudd på ca. 10 prosent). Reaksjonen begynner ved redusert temperatur mellom -10 og +10"C, fortrinnsvis ca. 0°C. Blandingen blir rørt i et kort tidsrom, dvs. mellom en halv og to timer, og blir så behandlet med et molart overskudd på 10 prosent av smeltet zinkklorid oppløst i reaksjonsoppløs-ningsmiddelet. Blandingen blir så rørt i ytterligere 1-3 timer ved ovennevnte starttemperatur, hvoretter man lar temperaturen øke til ca. romtemperatur i løpet av 10-40 minutter.
For å frembringe dannelsen av forbindelsen med formel 6, blir alkylhalobenzoatet oppløst i det tørre reaksjonsoppløsnings-middelet. Esteren brukes i en mengde som omtrent tilsvarer den molare mengden av utgangsmengden av forbindelse 4. Oppløsningen føres inn i en suspensjon av tetrakis-trifenylfosfinpalladium (et molart overskudd på fra 5-10 prosent i forhold til reaktantene)
i reaksjonens oppløsningsmiddel ved temperaturer på mellom -10 og +10"C. Blandingen blir så rørt i ca. 15 minutter. Denne nylig fremstilte blanding blir så tilsatt en forutfremstilt oppløsning av forbindelsen med formel 5, og tilsetningen utføres ved ca. romtemperatur. Oppløsningen røres så i et lengre tidsromd, dvs. fra 15-25 timer ved romtemperatur. Reaksjonen blir så stoppet med syre og produktet utskilt og renset på vanlig kjent måte, hvorved man får forbindelsen med formel 6.
Syrene, farmasøytisk akseptable salter og amider angitt med formel 7, lar seg lett fremstille fra estere med formel 6. Basisk forsåpning med en alkalimetallbase vil gi syren. F.eks. kan en ester med formel 6 oppløses i et polart oppløsningsmiddel såsom en alkanol, fortrinnsvis under en inert atmosfære ved romtemperatur med et tremolart overskudd av en base, f.eks. kaliumhydroksyd. Oppløsningen kan røres i fra 15-20 timer, blir så avkjølt og tilsatt syre, hvoretter hydrolysatet kan gjenvinnes på vanlig kjent måte.
Amidet kan fremstilles ved en passende amidering av den type som er velkjent av den kjemiske industri. En fremgangsmåte for fremstilling av slike forbindelser er først og fremst til et syreklorid og så behandle dette med ammoniumhydroksid eller et passende amin. F.eks. kan syren behandles med en alkoholisk oppløsning av en base såsom etanolisk KOH (i et molart overskudd på ca. 10 prosent) og så omsettes ved romtemperatur i ca. en halv time. Oppløsningsmiddelet kan fjernes og resten løses opp i et organisk oppløsningsmiddel såsom eter, hvoretter det behandles med et dialkylformamid og så med et 10 gangers overskudd av oksalylklorid. Dette utføres ved reduserte temperaturer mellom -10° og +10°C. Den sistnevnte oppløsning blir så rørt ved redusert temperatur i fra 1-4 timer, fortrinnsvis 2 timer. En fjerning av oppløsningsmiddelet gir en rest som oppløses i et inert og uorganisk oppløsningsmiddel såsom benzen, avkjøles til ca. 0"C og behandles med konsentrert ammoniumhydroksid. Den resulterende blanding røres ved redusert temperatur i fra 1-4 timer. Produktet kan så innvinnes på vanlig kjent måte.
Forbindelser hvor A er hydrogen eller lavere alkyl kan fremstilles fra den tilsvarende jodfenylforbindelsen. Denne kan reageres med etynylzinkkloridforbindelsen, slik det er beskrevet i reaksjonsskjema I, mere spesielt i eksempel 3. Jodfenylforbin-delser hvor A er hydrogen eller lavere alkyl, er kommersielt tilgjengelig eller kan fremstilles ved fremgangsmåter som i seg selv er kjente.
De følgende eksempler illustrerer oppfinnelsen uten at denne er begrenset til disse.
Eksempel 1
Etyl 4- iodbenzoat
I en suspensjon av 10 g (40,32 mmol) 4-jodbenzosyre (Alfa Products Thiokol/Ventron Division) i 100 ml absolutt etanol ble tilsatt 2 ml tionylklorid. Blandingen ble så kokt under tilbake-løp i 3 timer. Oppløsningsmiddelet ble fjernet i vacuum og resten oppløst i 100 ml eter. Eteroppløsningen ble vasket med mettet NaHC03 og mettede NaCl-oppløsninger og tørket (MgS04). Oppløs-ningsmiddelet ble så fjernet i vakuum og resten destillert på et kugelrohr-apparat (100°C; 0,55 mm), noe som ga 9 g av tittelforbindelsen som er farveløs olje. PMR (CDC13)5 1,42 (3H, t, J~7Hz) , 4,4 (2H, q, Js:7Hz) , 7,8 (4H, s) .
Ved å gå frem på samme måte, men ved å bruke 2-jodbenzosyre og 3-jodbenzosyre, fikk man fremstilt de følgende estere: etyl 3-jodbenzoat - PMR(CDC13)S 1,4 (3H,t, Js:7Hz) , 4,37 (2H, q, J~7Hz), 7,12 (1H, t, JX8HZ), 7,95 (2H, m), 8,37 (1H, s); og
etyl 2-jodbenzoat - PMR(CDC13)S 1,4 (3H, t, JsSHz), 4,35 (2H, q, Js:8Hz) , 7,25 (2H, m) , 7,65 (2H, m) .
Denne fremgangsmåten kan brukes for fremstilling av enhver ester som er nødvendig for fremstilling av de foreliggende forbindelser.
Eksempel 2
l -( 2 '. 6'. 6/- Trimetylcykloheks- l/- envl) but- l- ene- 3- yn En oppløsning av 12,17 g (120,27 mmol) diisopropylamin i 200 ml tørr tetrahydrofuran ble avkjølt til -78 °C under argon og dråpevis ved hjelp av en sprøyte behandlet med 75 ml 1,6M (120 mmol) n-butyllitium i heksan. Blandingen ble rørt ved -78°C i 1 time og så behandlet via en kannyle med en avkjølt (-78°C) oppløsning av 21,99 g (114,35 mmol) p<->ionon (Aldrich Chemical Company) i 20 ml tørr tetrahydrofuran. Blandingen ble rørt ved - 78°C i 1 time, dråpevis behandlet med 21,73 g (125,93 mmol) dietylklorfosfat og hensatt for oppvarming til romtemperatur over 2 timer. Blandingen ble så via en kannyle overført til en oppløsning av LDA fremstilt ved at man under argon rørte en oppløsning av 26,57 g (262,57 mmol) av diisopropylamin i 150 ml tørr tetrahydrofuran og 164 ml 1.6M (262,4 mmol) n-butyllitium i heksan i en halv time ved -786C. Blandingen ble oppvarmet til romtemperatur, rørt i 15 timer og surgjort med 250 ml 3N HC1 og så ekstrahert med pentan. Det organiske ekstraktet ble vasket med 1 N HCl, vann, mettet NaHC03 og metted NaCl og så tørket over magnesiumsulfat. Produktet ble konsentrert og destillert i et kugelrohr-apparat (50°C; 0,1 mm Hg), noe som ga tittelforbindelsen som en farveløs olje.

Eksempel 3
Etyl 4- r 4'-( 2". 6". 6"- trimetvlcvkloheks- 1"- envl)-but- 3'- en- 1'- ynyl1benzoat
Reaksjonskar som ble brukt til denne fremgangsmåten ble flammetørket under vakuum, og alle operasjoner ble utført i en oksygenfri argon- eller nitrogenatmosfære. I en oppløsning av 970 mg (5,5656 mmol) av den acetyleniske forbindelsen (formel 4) fremstilt som beskrevet i eksempel 2 i 5 ml tørr tetrahydrofuran, ble ved 0<*>C tilsatt 6,3 ml av en 1,6 M (5,76 mmol) n-butyllitium-oppløsning i heksan. Blandingen ble rørt ved 0"C i 1,25 timer. Blandingen ble så via en kannyle behandlet med en oppløsning av 790 mg (5,7968 mmol) smeltet zinkklorid i 4 ml tetrahydrofuran og så rørt ved 0<*>C i 2 timer og så ved romtemperatur i 20 minutter. En oppløsning av 1,53 g (5,542 mmol) etyl 4-jodbenzoat i 4 ml tørr tetrahydrofuran ble overført ved hjelp av kannylen over i en suspensjon av 450 mg (0,3894 mmol) tetrakis-trifenylfosfinpalladium i 4 ml tørr tetrahydrofuran ved 0<*>C. Blandingen ble rørt ved 0<*>C i et kvarter. Den ble så ved hjelp av en kannyle behandlet med den ovenfor beskrevne oppløsning av alkynylzinkkloridforbindelsen, og blandingen ble så rørt ved romtemperatur i 20 timer. Reaksjonen ble stoppet ved å tilsette 30 ml 2N HC1, og den ble så ekstrahert med 100 ml blandede heksaner og 100 ml eter. De samlede organiske ekstrakter ble vasket med mettet NaHC03 og NACl-oppløsninger, tørket over magnesiumsulfat og konsentrert til en brun olje. Denne ble renset ved middels trykkvæskekromatografi (Waters 500; 2x silikaprepak; 20% CH2C12 i heksan), noe som ga tittelforbindelsen som en blek, gul olje. PMR (CDC13):S 1,05 (2CH3, s), 1,35 (3H,
t, Js:7Hz) , 1,5 (2H, m) , 1,75 (CH3 , s) , 2,0 (2H, M9 , 4,55 (2H,
q, J~7Hz) , 5,65 (1H, d, Jsl6,5 Hz), 6,70 (1H, d, Js:16,5Hz), 7,4 (2H, d, Js:8Hz) , 7,9 (2H, d, Js:8Hz) .
Eksempel 4
4- r4'-( 2". 6". 6".- triametylcvkloheks- l"- envl)- but- 3'-en- 1' - ynyl ] benzosyre
I en oppløsning av 292 mg (0,9056 mmol) etyl 4-[4'-(2",6",-6"-trimetylcykloheks-l"-enyl)-but-3'-en-1'-ynyljbenzoat i 1 ml etanol under argon ble dråpvis tilsatt en oppløsning av 180 mg (3,2 mmol) KOH i 2 ml etanol og 0,5 ml vann. Blandingen ble rørt ved romtemperatur i 18 timer, avkjølt og surgjort med 3N HC1. Det resulterende bunnfall ble oppløst i eter, eteroppløsningen ble så vasket med mettet NaCl og konsentrert ved et faststoff som ble omkrystallisert fra metanol/vann, noe som ga tittelforbindelsen som et gulblekt faststoff. PMR (CDC13):S 1,05 (2CH3, S), 1,47 (1H, m), 1,62 (1H, m), 1,78 (CH3, S), 2,05 (2H, m), 5,78 (1H, d, J~16Hz) , 6,78 (1H, d, Js:16Hz) , 7,58 (2H, d, JS:8Hz) , 8,02 (2H, , JS8HZ).
Eksempel 5
4- r 4 ' - ( 2 " . 6" . 6"- trimetvlcvklol- c' :s- 1"- envl) -
but- 3'- en- 1'- vnyltbenzamid
En blanding av 48,8 mg (0,1658 mmol) av syren fra eksempel 4 og 1,3 ml av 0.14M (0,182 mmol) etanolisk KOH-oppløsning ble rørt ved romtemperatur i en halv time, hvoretter oppløsningsmid-delet ble fjernet i vakuum. Resten ble oppløst i 2 ml eter, behandlet med 1 dråpe dimetylformamid, avkjølt til 0<*>C og så behandlet med 143 mg (1,1 mmol) oksalylklorid. Etter røring ved 0°C i 2 timer ble blandingen filtrert, resten vasket med eter og de samlede organiske oppløsninger ble så konsentrert. Resten ble oppløst i benzen, avkjølt under 0<*>C og behandlet dråpevis med lml konsentrert NH4OH. Blandingen ble så rørt ved 0°C i 2 timer, fortynnet med 25 ml vann og ekstrahert med eter. Eterekstraktet ble vasket med mettet NaCl, tørket ov*»*- magnesiumsulfat og konsentrert i vakuum, noe som ga tittelforbindelsen som et hvitt, faststoff. PMR(CDC13):S 1,05 (2CH3, 2), 1,46(1H, m), 1,6(1H, m), 1,76(CH3, s), 2,05 (2H, m), 5,7 (1H, d, Jxi6Hz), 6,27 (2H, broad s) , 6,72 (1H, d, Js:16Hz) , 7,5(2H, d, Js:8Hz) , 7,76 (2H, d, J%8Hz) .
Ved å bruke tilsvarende fremgangsmåte kan forbindelser fremstilt som beskrevet i eksempel 4, omdannes til sine tilsvarende amider.
Claims (4)
1. Analogifremgangsmåte for fremstilling av en terapeutisk aktiv forbindelse med formel I,
hvor A er COOR, hvor R er H, laveralkyl eller NH2, karakterisert ved at man a) omsetter en forbindelse med formel II med en forbindelse
med formel III
hvor A er som angitt ovenfor og X er halogen, og eventuelt hydrolyserer en forbindelse med følgende formel
hvor A er COOR og R er laverealkyl, eller om ønsket
når A er COOH, omdanner syren til et salt; eller når A er COOH, omdanner syren til en ester; eller når A er COOH, omdanner syren til et amid.
2. Fremgangsmåte ifølge krav 1 ved fremstilling av etyl-4-[ 4' - (2" , 6" , 6"-trimetylcykloheks-lll-enyl) but-3' -en-1'-ynyl ] - benzoat,
karakterisert ved at man anvender tilsvarende substituerte utgangsmaterialer.
