Description
Foreliggende oppfinnelse angår en analogifremgangsmåte ved fremstilling av terapeutisk aktiv (R,S)-5-amino-hepta-5,6-diensyre, (S)-4-amino-hepta-5,6-diensyre og farmasøytisk akseptable salter derav.
Biooverføringen av Y-aminosmørsyre (GA3A) til ravsyre-seraialdehyd, som katalyseres av enzymet GABA-transaminase (GABA-T), er den primære reaksjon som er ansvarlig for kata-bolismen av GABA, en inhiberende neurotransmitter for sentralnervesystemet. Det er kjent at lave nivåer av endogent GABA
er forbundet med anfallssykdommer (slik som de som er involvert i epilepsi, alkoholabstinens eller barbituratabstinens), med sykdommer innbefattende bevegelser som ikke er underkastet viljens herredømme (slik som de fremkalt av de ekstrapyri-midale virkninger av legemidler, f.eks. tardivdyskinesia) med visse psykiatriske sykdommer (slik som schizofreni og depresjon) og med muskelkramper. Blokkering av overføringen av GABA til ravsyresemialdehyd, slik som ved irreversibel inhibering av GABA-T, kan forhøye GABA-nivåer i sentralnervesystemet (CNS) og utgjør således et middel for behandling av sykdommer på sentralnervesystemet forbundet med lave GABA-nivåer.
Visse forbindelser er kjent for å være irreversible inhibitorer av GABA-T og forhøyer derved hjernenivåer av GABA. Eksempler er 4-aminohex-5-ensyre ("vinyl GABA") og 4-aminohex-5-ynsyre ("acetylenisk GABA") [se U.S. patent nr. 3960927 og 3959356; Lippert et al., Eur. J. Et ytterligere eksempel er 1-acetylen-l,4-butandiamin ("acetylenisk putrescin") (se U.S. patentskrift 4139563).
Foreliggende oppfinnelse angår således en analogifremgangsmåte for fremstilling av terapeutisk aktiv (R,S)-5-amino-hepta-5,6-diensyre, (S)-4-amino-hepta-5,6-diensyre eller et farmasøytisk akseptabelt salt derav.
Navnene "(S)-allenyl GABA" og "(R,S)-allenyl GABA" anvendes her for å angi (S)-4-amino-hepta-5,6-diensyre og (R,S)-4-amino-hepta-5,6-diensyre.
Illustrative eksempler på farmasøytisk akseptable salter av forbindelsene fremstilt ifølge oppfinnelsen, innbefatter ikke-toksiske syreaddisjonssalter dannet med uorganiske syrer, slik som saltsyre, hydrobromsyre, svovelsyre og fosforsyre, eller med organiske syrer slik som organiske carboxylsyrer, f.eks. salicylsyre, maleinsyre, malonsyre, vinsyre, citronsyre og ascorbinsyrer, og organiske sulfonsyrer, f.eks. methansulfon-syre; og ikke-toksiske salter dannet med uorganiske eller organiske baser slik som hydroxyder av alkalimetaller, f.eks. natrium, kalium og lithium, jordalkalimetaller, f.eks. calcium og magnesium, lette metaller av gruppe III A, f.eks. aluminium, organiske aminer slik som primære, sekundære eller tertiære aminer, f.eks. cyclohexylamin, ethylamin, methylamino-ethanol-amin og piperidin. Saltene fremstilles på vanlig måte.
(S)-allenyl GABA og (R,S)-allenyl GABA bevirker in vivo irreversibel inhibering av GABA-T og kan forhøye GABA-nivåer signifikant i CNS når de administreres oralt eller parenteralt til varmblodige dyr. Således er (S)-allenyl GABA og (R,S)-allenyl GABA anvendbare for behandling av sykdommer i varmblodige dyr forbundet med lave nivåer av GABA i CNS. I særdeleshet er forbindelsene anvendbare som anti-konvulsive midler for regulering av anfall innen epilepsi. Anti-konvul-siv aktivitet kan demonstreres ved hjelp av standard testprosedyrer i laboratoriedyr overfor eksperimentelt fremkalte anfall. Eksempelvis kan (S)-allenyl GABA og (R,S)-allenyl GABA beskytte mus mot klonisk anfall fremkalt av bicucullin når de behandles etter prosedyren beskrevet av W. Buckett (Br. J. Pharm., (58, 177 (1980)) og Journal of Pharmacological Methods, 5, 35 (1981)). Forbindelsene kan også beskytte mus og rotter mot anfall fremkalt av metrazol (klonisk og tonisk), maksimalt elektrosjokk (tonisk), og/eller 3-mercaptopropionsyre (klonisk og tonisk).
I tillegg til anvendelser som antikonvulsive midler
er (S)-allenyl GABA og (R,S)-allenyl GABA anvendbare for behandling av CNS-sykdommer innbefattende bevegelser som ikke er underkastet viljens herredømme, i særdeleshet tardiv dyskinesia, for behandling av psykiatriske sykdommer slik som f.eks. schizofreni og depresjon, og/eller for behandling av muskelkramper. Ennvidere kan forbindelsene fremkalle hypo-termia, myorelaksasjon, anorexia, sedasjon og/eller anti-nocicepsjon når de administreres systemisk.
Dosen av (S)-allenyl GABA og (R,S)-allenyl GABA i varmblodige dyr vil avhenge av de arter som behandles, den bestemte forbindelse som anvendes, strengheten av den tilstand som behandles, og administreringsmåte. Generelt kan en effektiv dose som er i stand til å tilveiebringe fysiologisk nyttig økning av GABA-nivåer i CNS oppnåes i varmblodige dyr ved en dose på fra 1 til 500 mg/kg (kroppsvekt) pr. dag administrert oralt eller parenteralt. For større dyr (ca. 70 kg) kan det anvendes en dose på 5 til 100 mg/kg pr. dag. Behandlingen bør startes ved lavere doser idet dosen deretter økes i små porsjoner inntil den ønskede effekt er oppnådd.
Den GABA-T inhiberende aktivitet av forbindelsene kan demonstreres i laboratoriedyr in vivo ved metoder ifølge M. Jung et al., J. Neurochem., 2J3, 717 (1977). På mennesker kan GABA-T-inhibering måles etter systemisk legemiddel-administrering ved bestemmelse av forhøyede GABA, homocarnosin og |3-alaninnivåer i cerebrospinalvæske (CSF) , da det fore-ligger en kjent korrelasjon mellom GABA, homocarnosin og P-alaninnivåer i hjernen og i CSF.
Biologisk testing av (R)-allenyl GABA, enantiomeren av (S)-allenyl GABA, har fastslått at (R)-allenyl GABA ikke er en irreversibel inhibitor av GABA-T. Det skal forståes at når det gjelder inhibering av GABA-T og økning av hjerne-GABA-nivåer som tidligere beskrevet, kan (S)-allenyl GABA anvendes enten hovedsakelig fri for (R)-allenyl GABA eller i fysiologisk blanding med (R)-allenyl GABA, slik som race-matet (R,S)-allenyl GABA.
Det er antatt at (S)-allenisk GABA og (R,S)-allenisk GABA er "substrat-induserte irreversible inhibitorer" av GABA-T. Slike inhibitorer er også kjent innen faget som "enzym-aktiverte irreversible inhbitorer", "selvdestruerende enzyminhibitorer", "K ca t<i>nhibitorer", eller "mekanisme-
C3 "C
baserte inhibitorer". For at en forbindelse skal være en substrat-indusert irreversibel enzyminhibitor må forbindelsen ha et substrat for målenzymet, og forbindelsen må inneholde en latent reaktiv gruppe som er ømfintlig overfor å avmaskeres som resultat av den normale katalytiske virkning av enzymet. Avmaskering av den latente reaktive gruppe ved virkningen av enzymet skaper en reaktiv funksjon som alkylerer et nucleo-filt residuum tilstedeværende ved enzymets aktive sete. Således dannes det en kovalent binding mellom inhibitoren og enzymet ved det aktive sete som resulterer i irreversibel inaktivering av enzymet. Slike inhibitorer er ekstremt spesi-fikke da inhibitoren må være et substrat for målenzymet og da biooverføringen av inhibitoren med målenzymet er nødvendig før enzymet inaktiveres. Selv om det er antatt at (S)-allenyl GABA og (R,S)-allenyl GABA generelt utviser deres virkning ved hjelp av en substrat-indusert mekanisme, kan inhiber-ingen finne sted ved andre mekanismer slik som ved konkurrer-ende inhibering.
(S)-allenyl GABA og (R,S)-allenyl GABA kan administreres på forskjellig måte for å oppnå den ønskede effekt. Forbindelsene kan administreres alene eller i kombinasjon med farma-søytisk akseptable bærere eller fortynningsmidler, hvor mengden og arten av disse bestemmes av løseligheten og de kjemiske egenskaper av den valgte forbindelse, den valgte administreringsmåte og standard farmasøytiske praksis. Forbindelsene kan administreres oralt i faste doseringsformer, f.eks. kapsler,
tabletter, pulvere, eller i væskeformer, f.eks. løsninger eller suspensjoner.. Forbindelsene kan også injiseres parenteralt i form av sterile løsninger eller suspensjoner. Faste, orale Jformer kan inneholde konvensjonelle eksipienter, f.eks. lactose, sucrose, magnesiumstearat, harpikser og lignende materialer. Vasskeformige orale former kan inneholde forskjellige smaksgivende midler, farvestoffer, konserveringsmidler, stabiliseringsmidler, oppløsningsmidler eller suspenderingsmidler. Parenterale preparater er sterile vandige eller ikke-vandige løsninger eller suspensjoner som kan inneholde forskjellige konserveringsmidler, stabiliseringsmidler, buffermidler, oppløsningsmidler eller suspenderingsmidler.
Om ønsket, kan additiver slik som fysiologisk saltvann eller glucose tilsettes for å gjøre løsningene isotoniske.
Mengden av den administrerte nye forbindelse vil variere og kan være en hvilken som helst effektiv mengde. Enhetsdoser av disse forbindelser kan f.eks. inneholde fra 100 mg til 500 mg av forbindelsene, og kan administreres én eller flere ganger daglig, f.eks. fra 1 til 4 ganger daglig.
Som anvendt her, menes med uttrykket pasient et varm-blodig dyr slik som mennesker og andre pattedyr, f.eks. katter, hunder, rotter, mus marsvin, sauer, hester og kveg.
Uttrykket "enhetsdoseringsform" anvendes her for å angi en enkel eller multippel doseform inneholdende en mengde av den aktive bestanddel i blanding med eller på annen måte i forbindelse med fortynningsmidlet eller bæreren, hvilken mengde er slik at én eller flere på forhånd bestemte enheter normalt er nødvendig for en enkelt terapeutisk administrering. Når det gjelder multiple doseformer slik som væsker eller graderte tabletter, vil angitte på forhånd bestemte enhet være en frak-sjon slik som en 5 ml mengde (teskje) av en væske eller halv-parten eller en fjerdedel av en gradert tablett av den multiple doseform.
Analogifremgangsmåten ifølge oppfinnelsen er kjenne-tegnet ved at ethynylgruppen i et amino-beskyttet derivat av en (S)- eller (R,S)-aminoalkynforbindelse av generell formel
hvori R er C^-C^-alkyl, omdannes på kjent måte til en allenyl-gruppe.
Forbindelsene av formel I er kjente forbindelser (se f.eks. U.S. patentskrift 3959356 og 4139563) eller kan fremstilles ved analoge metoder.
Den aminobeskyttende gruppe velges under hensyntagen til arteri av den relevante reaksjon og den letthet med hvilken gruppen kan fjernes for å frigjøre aminogruppen. Den beskyttende gruppe kan velges fra f.eks. acyl, f.eks. lavere alkanoyl, f.eks. acetyl, propionyl, trifluoracetyl og lignende; aroyl, f.eks. benzoyl, toluoyl og lignende; lavere alkoxycarbonyl, f.eks. methoxycarbonyl, ethoxycarbonyl, tert-butoxycarbonyl
(BOC)og lignende; carbobenzoxy, benzensulfonyl og tosyl. -Begge aminohydrogenatomer kan være substituert med en enkelt beskyttende gruppe slik som f.eks. fthalyl. Den for tiden foretrukne
beskyttende gruppe er tert-butoxycarbonyl (BOC). De beskyttende grupper innføres på kjent måte, f.eks. ved omsetning av aminet med et lavere alkanoyl- eller aroylklorid, anhydrid eller sulfonylklorid. For å innføre BOC-gruppen kan forbindelsen av formel I behandles med tertiobutoxycarbonyloxyimino-2-fenyl-acetonitril (BOC-ON), di-tertiobutyldicarbonat ((BOC)20) eller tertiobutoxycarbonylklorid.
Fjerning av den beskyttende gruppe etter at den nødvendige reaksjon er fullført, kan utføres på kjent måte for den relevante beskyttende gruppe. Vanligvis vil fjerningen bli fore-tatt ved hydrolytisk splitting under anvendelse av en sterk organisk eller uorganisk syre slik som f.eks. trifluoreddik-syre, saltsyre og lignende syrer; eller med hydrogenkloridgass under vannfrie betingelser. Anvendte løsningsmidler velges avhengig av betingelsene for fjerning av den beskyttende gruppe. Eksempelvis kan ethere slik som f.eks. diethylether anvendes for splitting under anvendelse av hydrogenkloridgass.
Omdannelsen av ethynylgruppen til den krevede allenyl-gruppe kan utføres etter den generelle metode som er beskrevet av P. Crabbé et al. (J.C.S. Chem. Comm. 1979, 859-860) og H. Fillion et al. (Tet. Letters, 1980, 929-930) for allenisk alkohol. I denne metode oppvarmes det aminobeskyttende derivat av en forbindelse av formel I med formaldehyd og et sekundært amin med et hydrogenatom på a-carbonatornet, i et organisk løs-ningsmiddel og i nærvær av et uorganisk salt. Fortrinnsvis utføres oppvarmingen til under tilbakeløpsbetingelser. Det foretrukne amin er diisopropylamin, og det foretrukne uorganiske salt er et kobbersalt, spesielt kobberbromid eller kobber-klorid. Egnede løsningsmidler innbefatter dioxan, tetrahydro-furan, 1,2-dimethoxyethan, benzen, acetonitril og/eller toluen.
Omdannelsen er antatt å forløpe via det tilsvarende aminobeskyttede derivat av den sekundære aminopropynylforbind-else, spesielt diisopropylaminopropynylforbindelsen av følgende formel II:
hvori R er som definert i forbindelse med formel I.
(S)-allenyl GABA kan også erholdes på kjent måte, f.eks. ved oppløsning av (R,S)-alienisk GABA under anvendelse av en chiralsyre slik som (+)- eller (-)-binafthylfosforsyresalt ved metoden beskrevet av R. Viterbo et al., i Tetrahedron Letters 48, 4617-4620 (1971) og i U.S. patentskrift 3848030 eller (+)-kamfer-lO-sulfonsyre.
(S)-4-amino-hex-5-ynsyre kan erholdes på kjent måte ved oppløsning av (R,S)-4-amino-hex-5-ynsyre, f.eks. under anvendelse av en chiral syre, fortrinnsvis (+)- eller (-)-binafthyl-fosforsyre ved metoden ifølge Viterbo. Anvendelse av (+)-binafthylfosforsyre er foretrukket.
Forbindelsene fremstilt ved de foregående prosesser, kan isoleres enten som sådanne eller som salter, vanligvis syreaddisjonssalter.
Syreaddisjonssaltene er fortrinnsvis de farmasøytisk akseptable, ikke-toksiske addisjonssalter med egnede syrer slik som de her tidligere beskrevne. Bortsett fra farmasøytisk akseptable syreaddisjonssalter kan andre syreaddisjonssalter slik som f.eks. de med picrinsyre eller oxalsyre anvendes,
idet disse kan tjene som mellomprodukter ved rensing av forbindelsene ifølge oppfinnelsen eller ved fremstilling av andre, f.eks. farmasøytisk akseptable syreaddisjonssalter, eller kan være anvendbare for identifisering eller karakterisering av basene.
Et resulterende syreaddisjonssalt kan omdannes til den frie forbindelse etter kjente metoder, f.eks. ved behandling med et alkali- eller jordalkalimetallhydroxyd eller -alkoxyd, med et alkali- eller jordalkalimetallcarbonat eller -hydrogen-carbonat, med trialkylamin, eller med en anionbytterharpiks.
Et resulterende syreaddisjonssalt kan også omdannes til et annet syreaddisjonssalt etter kjente metoder, eksempelvis kan et salt med en uorganisk syre behandles med et metallsalt, f.eks. et natrium-, barium- eller sølvsalt av en syre i et egnet fortynningsmiddel hvori et resulterende uorganisk salt er uløselig og som således kan fjernes fra reaksjonsmediet.

A) F remsti lling av: ( R, S)- methyl- 4-( N- tertiobutoxycarbonylamino)- hex- 5- ynoat En løsning av 12,7 g (0,1 mol) (R,S)-4-amino-hex-5-yn-syre (fremstilt som beskrevet i U.S. patentskrift 3959356) 1 tørr methanol ble mettet med tørr hydrogenkloridgass og fikk stå over natten ved romtemperatur. Løsningen ble for-dampet under redusert trykk under dannelse av det urene (R,S)-methyl-4-amino-hex-5-ynoat-hydroklorid som residuum. Dette residuum ble suspendert i en løsning av 60 g ditertiær-butyldicarbonat (0,1 mol) i 100 ml kloroform. Suspensjonen ble avkjølt til 0°C, og 14 ml (0,1 mol) triethylamin ble dråpe-vis tilsatt. Den resulterende klare løsning ble oppvarmet i 2 timer under tilbakeløpskjøling, ble konsentrert under redusert trykk og ble fortynnet med 200 ml diethylether og vasket med 5 x 50 ml vann. Det organiske lag ble tørket over magnesiumsulfat og konsentrert under dannelse av 20 g (80%) av den urene tittelforbindelse. Det urene produkt ble renset ved krystallisering i diethylether/pentan. B) Fremstilling av: ( R, S)- methyl- 4-( N- tertiobutoxycarbonylamino)- hepta- 5, 6-dienoat
En løsning av 4,8 g (0,02 mol) urent (R,S)-methyl-4-(N-tertiobutoxycarbonylamino)-hex-5-ynoat fremstilt som beskrevet i trinn A, 2,7 ml av en 37%-ig vandig formaldehyd-løsning (0,036 mol), 3,2 ml (0,025 mol) diisopropylamin og 1 g (0,006 mol) kobberbromid i 50 ml dioxan ble oppvarmet under tilbakeløpskjøling i 2 timer. Løsningen ble slukket med 50 ml IN vandig eddiksyre og ekstrahert med diethylether. Det organiske lag ble tørket over magnesiumsulfat og konsentrert under redusert trykk. Residuet ble renset ved kolonne-kromatografi på silicagel under anvendelse av diethylether :petroleumether (50:50) som elueringsmiddel under dannelse av 2,2 g av tittelforbindelsen. C) Fremstilling av:
( R, S)- 4- amino- hepta- 5, 6- diensyre- hydroklorid
En løsning av 1,275 g (0,005 mol) (R,S)-methyl-4-(N-tertiobutoxycarbonylamino)-hepta-5,6-dienoat fremstilt som beskrevet i trinn B og 0,12 g (0,005 mol) lithiumhydroxyd i 10 ml dimethoxyethan og 3 ml vann fikk stå ved romtemperatur i 3 timer. Løsningen ble fortynnet med diethylether og vann, og det organiske lag ble surgjort med 0,01N vandig saltsyre, ble mettet med natriumklorid og ekstrahert med diethylether. Det resulterende organiske lag ble tørket over magnesiumsulfat og konsentrert under redusert trykk, og residuet ble omkrystallisert fra ether/pentan under dannelse av 1,1 g (R,S)-4-(N-tertiobutoxycarbonylamino)-hepta-5,6-diensyre. Denne syre ble tilsatt til en mettet løsning av tørt hydrogenklorid i 20 ml tørr diethylether og fikk stå over natten ved romtemperatur. Hydrokloridet ble dannet direkte som krystaller som ble filtrert, vasket med diethylether og tørket under dannelse av den rene tittelforbindelse i praktisk talt kvantitativt utbytte (0,7 g, smp. 112°C).
D) Fremstilling av:
( R, S)- 4- amino- hepta- 5, 6- diensyre
(R,S)-4-amino-hepta-5,6-diensyre-hydroklorid erholdt i trinn C ble nøytralisert med 1 ekvivalent triethylamin i ethanol. Etter konsentrering under redusert trykk ble residuet tatt opp med kloroform. Det uløselige materiale ble bmkrystallisert fra ethanol:vann.
Eksempel ' 2
( S)- 4- amino- hepta- 5, 6- diensyre
Prosedyren beskrevet i eksempel 1 ble gjentatt idet man startet med (S)-4-amino-hex-5-ynsyre under dannelse av (S)-4-amino-hepta-5,6-diensyre (smp. 170°C fa]D = +44°,
C = 0,25/H20) via mellomproduktet (S)-4-(N-tertiobutoxy-carbonylamino) -hepta-5 , 6-diensyre (smp. 79°C [ct]D = -61,2°,
C = 0,25/CHCl3>.
Tilsvarende ble R-isomeren (smp. 169°C, !a]D = -38°,
C = 0,25/H20) fremstilt fra (R)-4-amino-hex-5-ynsyre via mellomproduktet (R)-4-(N-tertiobutoxycarbonylamino)-hepta-5,6-diensyre (smp. 69°C [a] = +68,80°, C = 0,25/CHCl3).
I biokjemiske tester ble (S)(+)-isomeren funnet å være en irreversibel inhibitor av GABA-T. (R)(-)-isomeren var ikke aktiv.
Eksempel 3
(S)-allenyl GABA og (R,S)-allenyl GABA's evne til å inhibere GABA-T enzym og øke GABA-nivåer i hjernen kan demonstreres i følgende testprosedyrer pa mus.
Hann albino CDl-mus som hver veier ca. 34 g ved starten av testen, ble gitt en intraperitoneal injeksjon av testforbindelsen i vandig løsning daglig i 7 fortløpende dager. Halv-parten av dyrene ble avlivet ved halshugging 24 timer etter den siste dose av testf orbindelsen. Den andre, halvpart av dyrene ble observert i opptil 12. dager med hensyn til toksin sitet (som indikert ved vekttap og død). Kontrolldyr mottok en injeksjon av bare bæreren.
Hjernene ble fjernet fra de døde mus og ble oppdelt i
to porsjoner ved sagittalsnitt. En halvpart ble anvendt for måling av GABA-T-aktiviteten mens den andre ble anvendt for måling av GABA-innholdet. GABA-T-aktiviteten måles under anvendelse av kjente metoder slik som beskrevet av M. Jung et al., J. Neurochem., 28, 717 (1977) og 29, 797 (1977). GABA-innholdet måles med perklorsyre eller trikloreddiksyreeks-trakter under anvendelse av en aminosyreanalysator utstyrt med en fluorescensdetektor.
Når (R,S)-allenyl GABA ble testet som ovenfor beskrevet, ga denne de resultater som er angitt i etterfølgende tabell 1.
Ingen tydelige tegn på toksisitet, intet vekttap og ingen store adferdsforandringer ved 50 eller 100 mg/kg/dag av
allenyl GABA.

Eksempel 4
A. Prosedyren beskrevet i eksempel 3 ble gjentatt under anvendelse av en enkel intraperitoneal dose av testforbindelsen, og musene ble avlivet 6 timer etter den intraperitoneale injeksjon. Resultatene for (S)-allenyl GABA ("S") og (R,S)-allenyl GABA ("R,S") er angitt i etterfølgende tabell 2.
Resultatene i tabell 2 viser at (S)-allenyl GABA er dobbelt så kraftig som (R,S)-allenyl GABA når det gjelder økning av GABA-nivåer.
B. Prosedyren i del A ble gjentatt under anvendelse av både orale og intraperitoneale doser av (R,S)-allenyl GABA. Resultatene er angitt i etterfølgende tabell 3:
Resultatene i tabell 3 indikerer at innen mindre avvik på grunn av biologiske variasjoner er (R,S)-allenyl GABA og (S)-allenyl GABA like effektive når de gies ved orale og intraperitoneale administreringsmåter.
Eksempel 5
A. Toksisiteten av (S)-allenyl GABA og (R,S)-allenyl GABA ble bestemt etter intraperitoneal administrering til hannmus. Musene ble observert opptil 8 dager etter injeksjon. Dødelig-heten av forbindelsene (angitt som antall døde dyr pr. gruppe) er angitt i det etterfølgende:
De ovenfor angitte resultater indikerer at (S)-allenyl GABA er ikke-toksisk etter intraperitoneal administrering av meget store doser svarende til maksimal biologisk effekt.
(R,S)-allenyl GABA er imidlertid toksisk ved doser som er 4-5 ganger høyere enn den laveste biokjemisk effektive dose.
B. Toksisiteten av (S)-allenyl GABA ("S") og (R,S)-allenyl GABA ("R,S") ble bestemt som i del A både etter oral og intraperitoneal administrering. Dødeligheten av forbindelsene på dag 4 etter injeksjon (angitt som antall døde dyr pr. gruppe) er angitt i det etterfølgende:
De ovenfor angitte data indikerer at (R,S)-allenyl GABA ikke utviser toksisitet ved oral administrering opptil en dose på 1500 mg/kg. Dataene indikerer også at (S)-allenyl GABA er ikke-toksisk ved oral administrering.
Eksempel 6
(R,S)-allenyl GABA's evne til å beskytte mus mot anfall og død fremkalt av mercaptopropionsyre ble demonstrert som følger:
Mus (3,5) ble gitt en enkel intraperitoneal injeksjon av testforbindelsen. 6 timer senere mottok dyrene mercaptopropionsyre i en dose på 53 mg/kg i.p. Dyrene ble observert for tilsynekomst av klonisk toksiske anfall og dødsfall. Resultatene av testen er angitt i det etterfølgende:
Mens (R,S)-allenyl GABA ikke fullstendig beskyttet mot anfall i denne modell, reduserte (R,S)-allenyl GABA betydelig antallet av døde dyr.
Claims (3)
1. Analogifremgangsmåte for fremstilling av terapeutisk aktiv (R,S)-5-amino-hepta-5,6-diensyre, (S)-4-amino-hepta-5,6-diensyre eller et farmasøytisk akseptabelt salt derav, karakterisert ved at ethynylgruppen i et amino-beskyttet derivat av en (S)- eller (R,S)-aminoalkyn-forbindelse av generell formel
hvori R er C-^-C^-alkyl, omdannes på kjent måte til en allenyl-gruppe.
2. Fremgangsmåte ifølge krav 1 for fremstilling av (S)-4-amino-hepta-5,6-diensyre eller et salt derav, karakterisert ved at tilsvarende utgangs-materialer anvendes.
3. Fremgangsmåte ifølge krav 1 for fremstilling av (R,S)-5-amino-hepta-5,6-diensyre og (S)-isomeren derav eller salt derav,karakterisert ved at tilsvarende utgangs-materialer anvendes.
