Description
Foreliggende oppfinnelse vedrorer nye vannfrie oksydasereagens-opplosninger som er spesielt nyttige ved påforing av Neisseria-kolonier, som produserer enzymet cytokromoksydase.
Det er velkjent å anvende en oksydasereagensopplosning for å bestemme tilstedeværelsen av Neisseria-kolonier, såsom Neisseria gonorrhoeae eller Neisseria meningitidis. U.S. Public Health Service har etablert kriteria for presumptiv og bekreftende identifikasjon av N. gonnorhoeae. For en presumptiv diagnose må en organisme dyrket på et selektivt medium for N. gonorrhoeae identifiseres som oksydase-positiv, gram-negativ diplococci og utvise typisk kolonimorfologi. For ytterligere å bekrefte diagnosen må sukkerfermenteringer og/eller fluorescens-antile-gemebestemmelser også utfores.
I henhold til standard-laboratorieprosedyrer for bestemmelse
av N. gonorrhoeae erholdes prover med en steril dott. Sidene og tuppen av dotten rulles deretter på det selektive medium.
Et typisk fast kulturmedium for anvendelse ved dyrking av
N. gonorrhoeae er beskrevet av Martin, J. E., Jr. og Lester A.
i HSMHS Health Reports 86: 30-33, 1971. Mediet blir deretter inkubert i 24 - 48 timer og inspiseret for kolonivekst. Mis-tenkte N. gonorrhoeae-kolonier underkastes en oksydaseprove for å bestemme om enzymet cytokromoksydase er dannet. En positiv oksydaseprbvereaksjon gjenkjennes ved utvikling av en mork purpurfarge i lopet av 5 - 30 s. Hvis en omhyggelig underso-kelse av mediet ikke viser noen vekst, blir oksydasereagenset påfbrt hele mediet. Dette vil gjore det lettere å se orsmå oksydasepositive kolonier som ellers ikke ville oppdages.
Vekst av oksydasepositive kolonier gir en sikker forbestemmelse på N. gonorrhoeae. For positiv identifisering blir ubehandlede kolonier på mediet eller en annen ubehandlet kultur underkastet ytterligere bekreftende laboratorieundersøkelser, eksempelvis sukkerfermentering og fluorescens-antilegemebestemmelse.
Denkjente og kommersielt tilgjengelige oksydasereagens anvendt for å bestemme tilstedeværelsen av Neisseria-kolonier består av en 1%'ig vandig opplesning av N,N,N<1>,N'-tetrametyl-p-fenylen-diamin-dihydroklorid. Denne opplesning er imidlertid meget u-stabil. Den har en lyseblå farge når den er nyfremstilt, men går over til eh sort farge i lopet av 2 - 3 dager og er da ikke lenger effektiv med hensyn til bestemmelse av oksydase p.g.a. den raske nedbrytning av diaminsaltet. Av denne grunn er det anbefalt at den kommersielle reagensopplbsning fremstilles daglig.
P.g.a. denkjente oksydasereagensens ustabile natur er det også anbefalt at den holdes under en nitrogenatmosfære og i en her-metisk forseglet glassampulle, hvis den ikke skal anvendes umid-delbart. Ytterligere vil reagensens naturlige blåfarge repre-sentere en stadig konflikt med hensyn til den dype purpurfarge som erholdes når reagensen påfbres Neisseria-organismene og vil derfor kunne virke forstyrrende på bestemmelsen.
Det er derfor en hensikt med foreliggende oppfinnelse å fremstille en fargelbs, stabil oksydasreagensopplbsning for påvisning av mikroorganisme-kolonier, hvilken opplbsning kan lagres relativt lenge.
Med den nye oksydasereagenspreparat ifølge foreliggende oppfinnelse unngås overraskende stabilitetsproblemer, som det kjente oksydasereagens er beheftet med.
Oksydasereagensopplbsningen ifblge oppfinnelsen er fargelbs
og stabil. F.eks. forblir reagensopplbsningen ifblge oppfinnelsen fargelbs opptil 8 uker og bibeholder en utmerket aktivitet. Opplesningen kan inneholdes i dryppeflasker og be-hovet for pakking under nitrogenatmosfære er eliminert.
Ytterligere, den stabile vannfrie oksydasereagensopplosning ifblge oppfinnelsen gir en meget bedre kontrast, mellom koloniene og mediet. Anvendelsen av en fargelbs opplbsning som utgangsmate-riale forenkler vesentlig observasjonen og tolkingen av koloniene , som får en dyp purpurfarge i nærvær av reagensopplbsningen.
Den stabile vannfrie oksydasereagensopplosning ifblge foreliggende oppfinnelse omfatter tetrametyl- eller dimetyl-p-fenylen-diamin i dimetylsulfoksyd. Fenylendiaminet vil utgjore 0,1 -
1,0 vekt-% av blandingen. Fortrinnsvis er fenylendiaminet tilstede i en mengde på 0,2 - 0,5 vekt-%. Mest fordelaktig vil den stabile reagensopplbsning ifblge oppfinnelsen inneholde N,N,N' , N'-tetrametyl-p-fenylendiamin.
Oppfinnelsen innbefatter også ikke-toksiske, farmasbytisk akseptable addisjonssalter av de ovenfor definerte baser erholdt med organiske eller uorganiske syrer. Slike salter lar seg lett fremstille ved kjent teknikk. Basen omsettes enten med den stbkiometriske mengde av organisk eller uorganisk syre i et med vann blandbart opplbsningsmiddel, såsom aceton eller etanol, med isolasjon av saltet ved inndampning og avkjbling eller med et overskudd av syren i et med vann ublandbart opplbsningsmiddel , såsom etyleter eller kloroform, hvorved det bns-
kede salt faller ut direkte. Eksempler på slike organiske salter er de som erholdes med maleinsyre, fumarsyre, benzosyre, ravsyre, eddiksyre, propionsyre, vinsyre, salicylsyre, sitron-syre, stearinsyre og palmitinsyr e. Eksempler på slike uorganiske salter er de som erholdes med saltsyre, bromhydrogen-
syre, svovelsyre, fosforsyre og salpetersyre. Disse salter kan også fremstilles ved den klassiske metode ved dobbeltspalt-ning av passende salter, slik som er velkjent innen teknikkens stand.

- Stabil oksydasereagensoppløsning for påvisning av Neisseria-kolonier omfattende 0,1 - 1,0 vekt%. N,N,N',N'-tetrametyl-p-fenylendiamin eller N,N-dimetyl-p-fenylendi-amin eller syreaddisjonssalter derav, karakterisert ved at som oppløsningsmiddel er anvendt dimetylsulfoksyd.
