Description
Fremgangsmåte for behandling av matvarer.
Foreliggende oppfinnelse angår en fremgangsmåte for behandling av matvarer. Mer spesielt angår den en fremgangsmåte for forbedring av smak eller lukt av matvarer ved tilsetning av minst en de-oksy-5'-inosinsyre, deoksy-5'-guanylsyre og deres salter.
Det har vært kjent at mononatriumglutamat eller natriumsuccinat har evnen til å fremheve matvarers naturlige lukt og smak, og at begge disse er anvendt for forbedring av lukt og smak av matvarer i hjem og restauranter, og ved behandling av slike ting som vin, kjøtt og hermetiske matvarer. Natriuminosinat har også vært kjent som et stoff som har liknende virk-ning.
Den fyldige lukt og smak av suppe-bestanddeler av enhvert art antas å være et resultat av oppløselige bestanddeler i slike stoffer, særlig av de ovennevnte kjemiske forbindelser. Hydrolysater av animalske eller vegetabilske proteiner, som fås ved virk-ning av syrer eller protease av mikroorganismer, eller avledede stoffer, er også kjent og anvendt som kjemiske tilsetninger for matvarer. Kjemiske tilsetninger av denne art inneholder generelt forskjellige amino-syrer som hovedbestanddeler.
Alle de hittil kjente kjemiske tilsetninger anvendes i matvareindustrien eller ved tilberedning, eller de kan noen ganger anvendes sammen med to eller flere av liknende kjemiske tilsetninger. F. eks. kan 5'-inosinsyre noen ganger anvendes i for-bindelse med mononatriumglutamat. Eller kjente kjemiske tilsetninger anvendes noen ganger sammen med et naturlig krydder, slik som tørket bonito (såkalt katsuobushi) for å forbedre matvarers smak eller aroma. I dette tilfelle må smaken eller aromaen som er forbedret på denne måte, skrive seg fra synergi mellom de kjemiske kryddere og 5'-inosinsyre inneholdt i den tørkede bonito.
Alle disse kjemiske kryddere er nyttige for forbedring av smak eller aroma i matvarer, men den respektive smak eller aroma som er fremhevet på denne måte ved disse tilsetninger er ikke de samme. Med andre ord, hvert av disse stoffer har sin egen spesialitet ved behandling av matvarer.
Oppfinneren har også undersøkt mu-ligheten for å finne andre kjemiske forbindelser som kan forbedre eller fremheve smak eller aroma av matvarer i et videre område, særlig i det felt som angår deoksyribonukleinsyrer (DNA) og deres hydrolysater. Som et resultat er det blitt klarlagt at smak og aroma av matvarer kan forbedres eller fremheves ved tilsetning av deoksy-5'-inosinsyre, deoksy-5'-guanylsyre, eller deres salter eller derivater, hvilke tidligere aldri er blitt anvendt som kjemisk krydder og deres evne til å forbedre eller fremheve smak eller aroma i matvarer har ikke vært kjent. Det er således nå blitt funnet at de-oksy-5'-xantylsyre, dens salter eller derivater gir en utmerket smak og aroma til matvarer, således at de kan anvendes som kjemisk krydder for behandling av matvarer, og 5'-deoksyribonukleotider, slik som deoksy-5'-uridylsyre, deoksy-5'-cytidylsyre eller deres derivater har selv neppe noen god smak, men de kan anvendes som kjemisk krydder i form av en blanding med deoksy-5'-inosinsyre etc.
Hensikten med foreliggende oppfinnelse er å forbedre eller fremheve smaken eller aromaen i matvarer i et større område ved tilsetning av deoksy-5'-inosinsyre eller deoksy-5'-guanylsyre eller deres derivater. Foreliggende oppfinnelse kan således særlig utnyttes i matvareindustrien eller ved husholdningskokning.
Det er en hensikt med foreliggende oppfinnelse å skaffe en ny anvendelse av deoksy-5'-inosinsyre, deoksy-5'-guanylsyre og deres derivater i matvareindustrien eller ved kokning. Deoksy-5'-inosinsyre og deoksy-5'-guanylsyre kan fåes fra et hy-drolysat av deoksyribonukleinsyre eller oli-godeoksyribonukleotider, som er ekstrahert fra slike organismer eller organer eller vev som bakterier, féthumus, fésemen, sildese-men etc. Hydrolysen kan utføres ved hjelp av et enzymsystem av slangevenom eller av en mikroorganisme. Hydrolysen kan ikke utføres ved hjelp av en rent kjemisk fremgangsmåte, fordi kjemisk hydrolyse ikke gir 5'-deoksyribonukleotider. Da enzymsystemet av slangevenom er vanskelig tilgjen-gelig, foretrekkes det heller å anvende et system av mikroorganismer. Enzymsystemet som kan hydrolysere deoksyribonukleinsyrer eller oligodeoksyribonukleiotider til 5'-deoksyribonukleotider etc, er fordelt blant slike mikroorganismer som hører til klassen Schizomycetes, Ascomycetes etc, og mer spesielt blant mikroorganismer som hører til Fungi Imperfecti (f. eks. genera Fusarium, Verticillium, Gliomastix eller Helminthosporium), Eubacterialer (f. eks. genus Bacillus), aktinomycetaler (f. eks. genus Streptomyces), sferialer (f. eks. genera Anixiella, Botryosphaeria, Chaetomi-dium, Glomerella, Neurospora, Ophiobo-lus, Sordaria, Tilaclidium, etc.) og Plectas-cales (f. eks. genus Aspergillus).
Deoksyribonukleinsyrer eller olegode-oksynukleotider kan hydrolyseres med gun-stig resultat ved hjelp av enzymsystemet fra slike mikroorganismer, som nevnt ovenfor. Hydrolysen kan utføres ved 1) inkube-ring av mikroorganismen på et deoksyribonukleinsyre- og/eller olegodeoksynukle-otidholdig medium, 2) hvoretter kulturmediet, filtrert medium eller celle- eller my-celium-suspensjon av mikroorganismene tillates å reagere med deoksy-ribonuklein-syrer og/eller olegodeoksyribonukleotider, eller 3) å tillate enzymsystemet, etter rensning, å reagere med deoksy-ribonukleinsy-rer og/eller olegodeoksy-ribonukleotider Blant disse er den annen fremgangsmåte den mest foretrukne for industrielle formål. Mer spesielt, inkuberes mikroorganismen ved hjelp av areobisk neddyppet kultur i et vandig medium ved en egnet temperatur i et tidsrom for akkumulering av det ønskede enzymsystem. Kulturmediet kan anvendes som enzymkilde eller kan filtreres for å oppnå en rå enzymatisk oppløsning og celle- eller myceliumlegeme, og hver av dem kan anvendes som enzymkilde for hydrolysen. Den således erholdte enzymkilde settes til en oppløsning som inneholder deoksyribonukleinsyre og/eller olegodeoksy-nukleotider, og deretter hensettes blandingen ved en egnet temperatur i et passende tidsrom for å fullstendiggjøre den ønskede hydrolyse. Når enzymkilden er forurenset med fosformonoesterase, hydrolyseres de ønskede deoksy-5'-ribonukleotider til de til-svarende deoksyribonukleotider, således at det er nødvendig å utføre en behandling
for å hindre fosformonoesteraseaktivitet.
F. eks. oppnås dette ved tilsetning av en
fosformonoesterase-inhibitor, slik som ar-senater, fosfater, cyanater, fluorider eller en slik fenolforbindelse som fenol, kresol etc, til reaksjonsblandingen, eller det kan utføres ved varmebehandling ved en temperatur på 40—60° C ved en pH på 3,5—5,0.
Da det videre inneholdes diaminase i disse enzymkilder, deamineres deoksy-5'-adenylsyren, en komponent av deoksyribonukleinsyre, slik at det dannes deoksy-5'-inosinsyre. Denne fremgangsmåte, hvor det anvendes en av disse enzymkilder, er således meget fordelaktig for fremstilling av deoksy-5'-inosinsyre med meget godt ut-bytte.
Således fremstilles i reaksjonsblandingen den ønskede deoksy-5'-inosinsyre og deoksy-5'-guanylsyre sammen med andre 5'-deoksyribonukleotider, slik som deoksy-5'-adenylsyre, deoksy-5'-uridylsyre, deok-sy-5'-tymidylsyre etc, som har liten ønsket aktivitet. Reaksjonsblandingen inneholder forskjellige forurensninger, slik som proteiner, sukkere, celler eller mycelia, uorganiske salter, en del andre kulturprodukter etc. Naturligvis kan deoksy-5'-inosinsyre eller deoksy-5'-guanylsyre anvendes for formålet med foreliggende oppfinnelse så lenge de ikke er forurenset med en bestanddel eller bestanddeler som har ubehagelig eller uønsket lukt eller smak eller er giftige eller skadelige. Når slike stoffer ikke er forurenset, kan filtratet av reaksjonsblandingen konsentreres eller tørkes på vanlig måte, slik at det fås en væske eller et pulver av deoksy-5'-inosinsyre- og/ eller deoksy-5'-guanylsyre, som inneholder stoff som kan anvendes som krydder ifølge foreliggende oppfinnelse. Når det inneholdes slike bestanddeler som har ubehagelig lukt eller smak i reaksjonsblandingen, er det ønskelig å underkaste reaksjonsblandingen eller dens filtrat en renseprosess.
Rensningen av deoksy-5'-ribonukleotider som fås på denne måte kan utføres ved hjelp av en sterkt sur kationutvekslerhar-piks slik som Amberlite IR-120 (Rohm & Haas Co.). En slik harpiks kan adsorbere deoksy-5'-inosinsyre, deoksy-5'-guanylsyre, deoksy-5'-uridylsyre, og deoksy-5'-cytidyl-syre, og oppløsningen som fåes ved eluering av harpiksen med et vandig oppløsnings-middel, inneholder deoksy-5'-inosinsyre og/eller deoksy-5'-guanylsyre. Eller reaksjonsblandingen behandles med en svakt sur ioneutvekslingsharpiks for å fjerne ba-siske forurensninger, og stoffet som etter-lates uadsorbert inneholder deoksy-5'-ribonukleotider. Fra den nevnte oppløsning, kan rå deoksy-5'-inosinsyre, deoksy-5'-gu-anylsyre, etc, oppsamles separat eller sam-tidig ved hjelp av konsentrering. Oppløs-ningen kan også nøytraliseres for å oppnå alkalisalter av deoksy-5'-inosinsyre og de-oksy-5'-guanylsyre. Foruten som nevnt ovenfor, kan reaksjonsblandingen behandles ved hjelp av elektroforese, idet det anvendes en ioneutvekslingsharpiks-membran for å rense deoksy-5'-inosinsyre og/eller deoksy-5'-guanylsyre.
Videre rensning kan utføres ved ad-sorbsjon eller oppdelingskromatografi på et egnet adsorbsjonsmiddel slik som aktivkull, silikagel og cellulosepulver.
Deoksy-5'-inosinsyre eller deoksy-5'-guanylsyre er egnet for formålet ifølge foreliggende oppfinnelse som sine fri syrer, alkalimetallsalt, jordalkalimetallsalt, ammoniumsalt, histidinsalt, syreamid etc. I tillegg til deres rene former, kan deoksy-5'-inosinsyre og deoksy-5'-guanylsyre, som inneholder forurensninger som ikke har noen ubehagelig lukt eller smak eller er giftige, anvendes. Ubetydelige forurensninger er uorganiske salter, nukleotider eller nukleosider. Således kan et rått hydroly-sat av deoksyribonukleinsyre eller av ole-gonukleotider hvorfra uønskede bestanddeler er blitt fjernet, også anvendes ifølge foreliggende oppfinnelse.
Skjønt deoksy-5'-xantylsyre aldri er blitt oppdaget som en bestanddel av naturlig deoksyribonukleinsyre, kan den avledes fra deoksy-5'-guanylsyre. Semisyntesen av deoksy-5'-xantylsyre fra deoksy-5'-guanyl-syre kan f. eks. utføres som følger: Deoksy-5'-guanylsyre omdannes først til dens 2-diazoniumsalt ved hjelp av et nitritt og en syre, og diazoniumsaltet hydrolyseres til hydroksylgruppen, med en syre. Deoksy-5'-xantylsyre synes å være litt svakere enn de-oksy-5'-guanylsyre ved forbedring av smak eller aroma i matvarer. Men smaken eller aromaen som er forbedret eller fremhevet på denne måte med deoksy-5'-xantylsyre er betydelig.
Disse nye kjemiske kryddere er stabile mot varme og de kan derfor hensiktsmes-sig isoleres eller oppsamles fra hydrolysatet av deoksyribonukleinsyre etc, og anvendes for kokning eller for lagring av matvarer.
Smaken eller aromaen av alle de matvarer for hvilke de hittil kjente kjemiske kryddere er blitt anvendt er forbedret eller fremhevet ved tilsetning av kjemiske kryddere ifølge foreliggende oppfinnelse, uan-sett type eller formen for matvarer, f. eks. supper, sauser, pastaer, pølser, hermeti-serte matvarer, salat, kokte matvarer, pick-les, alkoholiske drikker, eddik, bordsalt, brød, fruktsafter, fermenterte matvarer etc. De kjemiske kryddere ifølge foreliggende oppfinnelse kan settes til matvarer under eller etter kokning. Med andre ord, kan de settes til råmaterialer av matvarer eller kokte matvarer.
Mengden av de nye kjemiske kryddere som skal settes til matvarer kan lett be-stemmes ifølge form, type eller annen til-stand for matvarene. Imidlertid gir mer eller mindre overskudd av krydderne neppe noen ubehagelig smak. Smaken eller aromaen av deoksy-5'-inosinsyre eller de-oksy-5'-guanylsyre er følbar i en konsentrasjon på ca. 0,015 pst., men egnet konsentrasjon er omkring 0,2 til 0,002 prosent i flytende matvarer.
De nye kjemiske kryddere forbedrer i smak og aroma på liknende måte som for 5'-inosinsyre og de anvendes fortrinsvis sammen med de hittil kjente kjemiske kryddere, slik som mononatriumglutamat, natriumsuccinat, natriuminosinat etc, for forbedring eller fremheving av smak og aroma. Det er derfor også effektivt å sette de nye kjemiske kryddere til de andre kjente kryddere, slik som aminosyrekryd-der, og særlig deres blanding med natriumglutamat gir meget godt resultat.
Når det nye kjemiske krydder settes til fermenterte matvarer, slik som bønnepasta i (miso), sake, whisky, soya etc, er det øn-- skelig at fosformonoesterasen i matvarene inaktiveres eller fjernes før tilsetning av kjemiske kryddere.
De riktige konsentrasjoner av natrium-
deoksy-5'-inosinat som skal settes til flere typer matvarer er angitt i det følgende sammenlignet med for natriumglutamat.
Eksempel 1.
100 g deoksyribonukleinsyre fremstilt fra sildemelke, tilsettes 10 1 kulturfiltrat av Streptomyces aureus (Waksman et Curtis) Waksman et Henrici (IFO-3303, ATCC-13404). Videre tilsettes blandingen natriumarsenat slik at den endelige konsentrasjon blir 10 mM. Deretter reguleres pH-verdien i blandingen til 8,5 og blandingen holdes ved 37° C i 8 timer. Hydrolysatet sendes gjennom et lag av aktivt kull for å adsorbere deoksyribonukleotidene. Aktiv - kullet elueres med en blanding av ammoniakk og etanol, og deretter konsentreres eluatet til tørrhet for å oppnå et pulveraktig krydder inneholdende deoksy-5'-guanyl-syre og deoksy-5'-inosinsyre, idet den totale mengde av bestanddelene er ca. 50 pst. av det pulveraktige krydder. Rå soya som er blitt brygget ved vanlig fermente-ring og filtrert gjennom en filterpresse oppvarmes for å inaktivere fosformonoesterasen i den anvendte maltede ris. I 1 liter av den således behandlede rå soya oppløses 4 g krydder fremstilt som ovenfor for å fremheve den bedre smak eller aroma av soyaen.
Eksempel 2.
100 g desoksyribonukleinsyre fra sildemelke tilsettes 10 liter kulturfiltrat av Aspergillus quercinus (Bainier) Thorn et Church (IFO-4363). Deretter reguleres blandingens pH-verdi til 8,5 og blandingen hydrolyseres ved 37° C i 8 timer, etterfulgt av tilsetning av 50 g kulturmycelia.
Den således tilberedte blanding under-kastes, ved regulering av pH-verdien til 5,0, hydrolyse ved 37° C i 1 time for å omdanne deoksy-5'-adenylsyre til deoksy-5'-inosinsyre, og sendes deretter gjennom et lag av aktivkull for å adsorbere deoksy-5'-inosinsyre. Aktivkullet elueres med en blanding av ammoniakk og etanol, og deretter konsentreres eluatet til tørrhet for å oppnå et pulveraktig krydder inneholdende deoksy-5'-guanylsyre og deoksy-5'-inosinsyre, idet den totale mengde av bestanddelene er ca. 40 pst. av det pulveraktige krydder. Raffi-nert sake, fremstilt ved å fjerne bermen fra moromi oppvarmes for å inaktivere fosformonoesterase. 1 liter av den således behandlede sake tilsettes 0,2 g av krydderet fremstilt som ovenfor, for å øke smak og aroma av den raffinerte sake.
Eksempel 3.
1 liter grønnsaksuppe, fremstilt ved tilsetning av en passende mengde bordsalt, tilsettes 0,2 g av krydderet erholdt ifølge eksempel 2 og 0,5 g mononatriumglutamat, hvoretter smaken eller aromaen i suppen i høy grad forbedres.
Eksempel 4.
100 g deoksyribonukleinsyre fra sildemelke tilsettes 10 liter kulturfiltrat av Botryosphaeria ribis chromogena G. et D.
(IFO-4837; ATCC-13834). Blandingen hydrolyseres ved 37° C i 10 timer og pH-verdien holdes omkring 8,5. Hydrolysatet sendes gjennom et lag av aktivkull for å adsorbere deoksyribonukleotider. Aktivkullet elueres med en blanding av ammoniakk og etanol, og deretter konsentreres eluatet.
Ved hjelp av ioneutvekslende harpiks, fraksjoneres konsentratet til fire bestanddeler av deoksyribonukleotider. Deoksy-5'-adenylsyre avledet fra den opprinnelige deoksyribonukleinsyre er blitt omdannet til deoksy-5'-inosinsyre ved deaminering av den sistnevnte med salpetersyrling. I 1 liter rå soya oppløses 2 g av hver av de så-kalte erholdte deoksy-5'-inosinsyre og de-oksy-5'-guanylsyre for å frembringe bedre smak eller aroma av soyaen.
Eksempel 5. 50 g kalvetymus-deoksyribonukleinsyre tilsettes 20 liter kulturfiltrat av Helminthosporium sigmoideum var. irregulare Cralley et Tullis (IFO-5273, ATCC-13406). Etter tilsetning av natriumsulfat for å gjøre blandingens konsentrasjon til 10 mM, hydrolyseres blandingen ved 37° C i 15 timer, og pH-verdien holdes omkring 8,0. De-oksy-5'-inosinsyre og deoksy-5'-guanylsyre fåes ved hjelp av samme fremgangsmåte som i eksempel 4. 1 liter grønnsaksuppe fremstilt ved tilsetning av en passende mengde bordsalt tilsettes 0,2 g deoksy-5'-guanylsyre, 0,2 g deoksy-5'-inosinsyre og 0,5 g mononatriumglutamat, hvoretter smak og aroma av suppen er langt bedre.
Eksempel 6.
100 g deoksyribonukleinsyre fremstilt fra sildemelke tilsettes 10 liter kulturfiltrat av Streptomyces aureus (Waksman et' Curtis) Waksman et Henrici (IFO-3303; ATCC-13404). Videre settes natriumarsenat til blandingen for å gjøre dens endelige konsentrasjon 10 mM. Deretter reguleres pH-verdien i blandingen til 8,5 og blandingen holdes ved 37° C i 8 timer. Hydrolysatet sendes gjennom et lag av aktivkull for å adsorbere deoksyribonukleotider. Aktivkullet elueres med en blanding av ammoniakk og etanol. Eluatet tillates å strømme gjennom «Dowex-1 (format-type)» (vare-merke for en anionutvekslerharpiks som selges av Dow Chemical Co., U.S.A.) for å adsorbere deoksy-5'-inosinsyre og deoksy-5'-guanylsyre, og harpiksen elueres deretter med en vandig oppløsning som inneholder 0,01 N.-maursyre og 0,05 N. natriumformat for å frigjøre deoksy-5'-inosinsyre. Eluatet bringes igjen i kontakt med aktivkull for å adsorbere deoksy-5'-inosinsyre og kullet elueres med ammoniakalsk etanol. Eluatet konsentreres til tørrhet for å oppnå deoksy-5'-inosinsyre. 1 liter kyllingsky tilsettes 100 mg de-oksy-5'-inosinsyre, som er fremstilt på denne måte, 1 g natriumglutamat og en passende mengde bordsalt og krydder for å fremstille suppe.
Eksempel 7.
Harpiksen som er behandlet på samme måte som i eksempel 6 elueres videre med en vandig oppløsning som inneholder 0,1 N. maursyre og 0,1 N. natriumformat for å fri-gjøre deoksy-5'-guanylsyre. Eluatet bringes i kontakt med aktivkull for å adsorbere deoksy-5'-guanylsyre og kullet elueres med ammoniakalsk etanol. Eluatet konsentreres til tørrhet for å oppnå deoksy-5'-gua-nylsyre. 889 ml kyllingsky tilsettes 50 mg således fremstilt deoksy-5'-guanylsyre og 1 g natriumglutamat, og det tilsettes deretter 7,2 g tørrmelk, 29 g rismel, 0,8 g bordsalt, 0,82 g løkpulver, 0,25 g sort pepper og 0,2 g salvie. Blandingen kokes for å fremstille kyllingsuppe. i
Claims (3)
1. Fremgangsmåte for økning av smak og aroma i matvarer, karakterisert v e d at det tilsettes minst ett stoff valgt fra den gruppe som består av deoksy-5'-inosinsyre, deoksy-5'-guanylsyre, deoksy-5'-xantylsyre og deres ikke-giftige salter til matvarene.
2. Fremgangsmåte ifølge påstand 1, karakterisertvedatde angitte stoffer tilsettes sammen med natriumglutamat til matvarene.
3. Fremgangsmåte ifølge påstandene 1—2, karakterisert ved at matvarene tilsettes natriumsaltet av de angitte syrer.
