Description
Det er tidligere bare kjent to nukleoside antibiotiske midler med
5-leddede heterocykliske ringer. Disse er showdomycin og pyrazomycin,
som henholdsvis har strukturene 1 og 2:
Blant de hittil kjente syntetiske nukleoside antivirale midler er
de tre viktigste 5-iodo-2'-deoxyuridin (5-IDU), 9-/>-D-arabinof uran-osyladenin (Ara-A) og l-^-D-ai?=ebino-furanosyl cytosin (Ara-C). Av disse midler er bare 5-IDU (i o,l vektprosent løsning) tilgjengelig som handelsvare som et spesifikt antiviralt middel, og denne for-
bindelse har den ulempe at den har lav løselighet og høy toksisitet,
som fører til den uønskede store fortynning som den nå benyttes i„
Alonso et al i Jc Heterocyclic Chem. 7 1259-72 197o beskriver frem-stillingen av l-(£-D-ribofuranosyl)-4-karboksamido-l,2,3-triazol,
men nevner ikke den biologiske aktivitet. Witkowski og Robins i "The Chemical Synthesis of .the 1,2,4-Triazole Wucleosides related to Uridine, 2'-Deoxy-uridine, Tnymidine and Cytidine", J. Org. Chem.
33, 2635-41 (197o) foreslår forskjellige 1,2,4-triazol-nukleosid-
analoge forbindelser som mulige substanser for biologisk undersøkelse,
men ingen spesiell virkning er anmerket og senere under søkelser-har vist at av derivatene som her er beskrevet utøver bare 5-brom-3-nitro-l-(2,3,5-tr i—0—acetyl-Ø-O-r ibof ur anosyl )-l,2,4-triazol en svak antiviral virkning og denne virkning er utelukkende antatt å skrive seg fra cytotoksisiteten av forbindelsen»
Den foreliggende oppfinnelse vedrører således en analogifremgangsmåte for fremstilling av farmakologisk aktive ribofuranosyl—1,2,4-triazol—3-karboksamid—forbindelser med den aenerelle formel
hvori Rp er H eller acetyl og har samme betydning som R£ eller står for fosfats og det særegne ved fremgangsmåten i henhold til oppfinnelsen er at man behandler en l-(-Q-acyl-blokkert-/^D-ribofuranosyl)-! , 2, 4-triazol-3-karboksylsyreester med den generelle formel hvori R' er lavere alkyl og Ac står for acetyl eller benzoyl ved at enten a) for fremstilling av en forbindelse med formel I hvori og R2 er H, forsepes forbindelser med formel II for å fjerne acylgruppene etterfulgt av behandling med ammoniakk, eller b) for fremstilling av forbindelsen med formel I hvori er fosfat og R2 er H, forsepes forbindelser med formel II for å fjerne acylgruppene etterfulgt av fosforylering og behandling med ammoniakk, eller c) for å danne en forbindelse med formel I hvori R^ og er acetyl omsettes en forbindelse med formel II ved sekvensen for reaksjonen under a) etterfulgt av re-acylering. Foretrukkede forbindelser omfatter slike med strukturen
såvel som 5'-fosfatet derav. Forbindelsene ifølge oppfinnelsen har en antiviral aktivitet og i ikke-acylert eller "avblokkert" tilstand har de en vesentlig større vannoppløselighet enn tidligere kjente nukleoside antivirale midler, og 1-(/3-D-ribofuranosyl)-1,2,4-triazol-3-karboksamid nukleosid har vist et bredt spektrum av antiviral aktivitet såvel som antitumoraktivi-tet.
Metyl-1,2,4-triazol-3-karboksylat er et utgangsstoff for fremstilling av 1,2,4-triazol-nukleosider ved fremgangsmåten ifølge oppfinnelsen. Denne forbindelse kan fremstilles konvensjonelt ved oksydasjon av 3-metyl-l,2,4-triazol etterfulgt av forestring av den dannede syre ifølge metoden til Cipens og Grinsteins Latvijas PSR Zinatnu Akad. Vestis., Kin. Ser.
(1965) (2) 2o4-o8 (Se CA. 63, 13243, 1965). Ved en reaksjons-måte dannes trimetyl-silyl-derivater av metyl-1,2,4-triazol-3-karboksylatet i kvantitativt utbytte ved omsetning med heksametyldisilazan under tilbakeløp og videre omsetning med et passende O-acylert halo-sukker GX for dannelse av for eksempel en blanding av O-benzoylert 1-((S-D-ribofuranosyl)-1, 2, 4-triazol-3-karboksylsyre-metylester og 5-karboksylsyre-metylester,
d. v. s. :
hvori B er benzoyl. 3-karboksylsyre-metylesteren separeres ved fraksjonert krystallisasjon eller, fortrinnsvis ved kolonne kromatografering over silika-gel. Debenzoylering og aminolyse gir 1- (/3-D-ribof uranosyl) -1,2, 4-triazol-3-karboksamidet.
Den foretrukkede syntesemetode er ved syrekatalysert sammen-slutning av et passende substituert triazol med det O-acyl blokkerte sukker (i dette tilfelle l-0-acetyl-2,3,5-tri-0-benzoyl-/5-D-ribofuranose) etterfulgt av avblokkering og aminolyse, d.v.s. :
3—karboksamido-nukleosidene kan omdannes til det tilsvarende 5'-fosfat eller.ammonium eller alkalimetallsaltet av- fosfatet og benyttes som sådant. En fagmann vil være kjent med metoden og måten for fosforylering av nukleosider. I en foretrukket utførelsesform av. oppfinnelsen dannes 5'-fosfatet av l-O^-D-ribofuranosyl)-l,2,4-triazol-3-karboks-amidet ved omsetning av l-(/5-D-ribof uranosyl )-l, 2,4-tri'azol-3-karboksyl-syremetylester med POClg i trirnetylfosfats etterfulgt av hydrolyse i isvann for å gi det fri 5<*->fosfato Dannelse av ammoniumsaltet og aminolyse av metylesteren foregår samtidig.ved behandling.med vandig ammoniakk. Alkalimetallsalter av det fri 5'-fosfat av 1-C.Æ-D—ribof uranosyl )-l, 2, 4-triazol-3-karboksamid-nukleo3id kan således oppnås fra ammoniumsaltet av fosfatet ved ioneveksling.
Oppfinnelsen er beskrevet med henvisning til acetyl-' eller benzoyl-blokkering av 2', 3' og 51 og hydroksyl i glykosyldelen. Sutherland et al i Biochim. et Biophys Acta'l48, lo6 (1967) har beskrevet at acylering av cykliske nukleotider fremmer den cellulære txransporto ?å lignende måte kan de fri glykosylhydroksyler av forbindelsene fremstilt ifølge
I
oppfinnelsen bli acylert for forøket lipid-oppløselighet eller for eksempel forsynt med alkyl- eller alkarylsulfonylgrupper, som for eksempel tosyl, mesyl, brosyl, nisyl, etc. for det samme formål.
Mens den 1'-gruppe som blir erstattet i sukkerkomponentene diskutert ovenfor er angitt som acetyl, eller i silyleringsmetoden som halo,
vil det være klart at enhver erstattbar gruppe kan benyttes under forut-setning av at biprodukter lar seg skille ut.
Oppfinnelsen beskrives nedenfor ved hjelp av eksempler, hvor alle
deler og prosentangivelser er på vektbasis. Alle fordampningsprosedyr-ene er utført i en roterende fordamper under forminsket trykk ved 35° C.
Fremstilling av utgangsforbindelser
l-(2, 3,5-tri-O-benzoyl-^i-D-ribof uranosyl-1,2, 4-triazol-3~karboksylsyre-metylester og l-(2,3,5-tri-0-benzoyl-/5-D-ribof uranosyl )-l,2,4-triazol-5-karboksylsyre-metylester
A. Silyleringsmetoden
En suspensjon av metyl 1,2,4-triazol-3-karboksylat Cl4.o g,
llo mmol) og heksametyldisilazan (loo ml) kokes under tilbakeløp under omrøring inntil utvikling av ammoniakk opphører (ca. 2 timer)o Overskudd av heksametyl-disilazan fjernes under redusert trykk
og gir 2o,6 g { loo%) av N-trimetylsilylderivatet av metyl— 1,2,4-triazol-3-karboksylat.
En løsning som inneholder metyl-N-(trimetylsilyl)-l,2,4-triazol-3-karboksylat (2b.6 g, llo mmol) og 2,3,5-tri-O-benzoyl-D-ribofuranosylbromid (52.5 g, loo mmol) i vannfritt acetonitril (3oo ml) holdes ved 25°C i tre dager. Løsningsmidlet fjernes og resten krystalliseres fra etanol. Omkrystallisasjon av materialet fra etylacetat-etanol og kolonne-kromatografering av filtratet over silika-gel med kloroform gir ren l-(2,3,5- tri-o-benzoyl-/J-D-ribof uranosyl )-l ,2,4-triazol-3-karboksylsyre-metylester (25.1 g, 44„o%) med smeltepunkt 137 - 139° C. I

Den hurtigst vandrende komponent fra silika-gel-kolonnen krystalliseres fra etanol og gir l-(2,3,5-tri-O-benzoyl-zS-D-ribofuranosyl )-l,2,4-triazol-5-karboksylsyre-metylester (13.2 g, 23.1%) med smeltepunkt 122 - 124° C.
Analyse: Funnet: C 63,26, - H <=> "4,35, N 7,12
B. Smeltemetoden
En blanding av metyl-1,2,4~triazol-3—karboksylat (12„7 g, loo
mmol) og l-0-acetyl-2,3,5-tri-0-benzoyl-$-|Jr-ribofuranbse
(55.4 g, llo mmol) oppvarmes i et bljebad som holdes ved 16o - 165° C. Etter at sukkeret er smeltet tilsettes bis(p.-nitrofenyl)fosfat (4oo mg) under omrtøring og blandingen oppvarmes under redusert trykk ved 16o - 165° C i 15 - 2o minutter. Krystallisasjonen av resten fra etylacetat-etanol ga 42,5 g (74,5%) av det samme produkt med smeltepunkt 137 139° G.
2. 1-C2, 3,5-tri-0-acetyl-/8-D-ribof ur anosyl )-1,2,4-triazol-3-karboksylsyre-metylester
En blanding av metyl-1,2,4-triazol-3-karboksylat (12.7 g,
o.lo mol) og 1,2,3,5-tetra-O-acetyl-P-D-ribofuranose (31, 8 g, o,lo mol) oppvarmes i et oljebad som holdes ved 166 165° C inntil sukkeret er smeltet. Bis(]D-nitrofenyl)fosfat (25o mg) tilsettes og oppvarmes ved 16o - 165° C under omrøring og redusert trykk i ,.15 — 2o. minutter..
Resten løses i varm benzen, løsningen filtreres og cykloheksan tilsettes til filteret for å gi et krystallinsk produkt (3o,o g, 77,8%) med smeltepunkt lo7 - lo9° C.

Eksempel I
1- Qfl- Q- ribofuranosyl)- l, 2, 4- triazol- 3- karboksamid Metode 1
En løsning av 1-(2,3,5-tri-Q-benzoyl-^-D-ribofuranosyl)-1,2, 4-triazol-3-karboksylsyre-metylester (16,o g, 28,o ,,ol) i metanol (3oo, ml, på forhånd mettet med vannfri ammoniakk ved 0° C) holdes i en lukket trykkflaske ved 25° C i 3 dager. Løsningsmidlet fjernes og produktet krystalliseres fra etanol og gir 6,7 g (98%) produkt med smeltepunkt 174 -176° C. Omkrystallisasjon av produktet fra vandig etanol gir en annen krystallinsk form av nukleosidet med smeltepunkt 166 - 168° C.
Analyse: Teoretisk for <c>8Hi2N4°5 : C = 39'34' H = 4'95'

Funnet: C = 39.08, H = 5,lo, N = 22,67.
Metode 2
En løsning av 1-(2,3,5-tri-0-acetyl-/6-D-ribof uranosyl)-1,2, 4-triazol-3-karboksylsyre-metylester (lo,o g, 26,o mmol) i metanol (7o ml) mettet ved 0° C med vannfri ammoniakk holdes i en lukket trykkolbe ved 25° C i 18 timer. Produktet krystalliseres fra etanol og gir 1- (/5-D-ribofuranosyl) -1, 2, 4, -triazol-3-karboksamid (5,7o g, 9o. o%).
Metode 3
En løsning av 1- (2, 3, 5-tri-0-benzoyl-£-jD-ribofuranosyl)-1, 2, 4-triazol-3-karboksylsyre-metylester (25,o g, 43,8 mmol) og natrium-metoksyd (4oo mg) i metanol (2oo ml) holdes under tilbakeløp i 45 minutter. Løsningen nøytraliseres med "Bio-Rad AG5o-X2(H)", filtreres og filtratet konsentreres til en sirup. Sirupen krystalliseres fra metanol-etylacetat og gir 8,og (7o,5%)
produkt med smeltepunkt 117-119° C.
Analyse: Teoretisk for <C>9Hi3N3°6 : c = 41,7o, H = 5,o6,
N = 16,21
Funnet: C = 41,57, H = 5,13, N = 16,16.
1-(/5-D-ribofuranosyl)-1,2,4-triazol-3-karboksylsyre-metylesteren i metanol (på forhånd mettet med vannfri ammoniakk ved 0° C) ble holdt i en lukket trykk-kolbe ved 25° C i 3 døgn. Løsningsmidlet ble.fjernet og produktet krystallisert fra etanol til å gi det ønskede 1- (/&-D-ribofuranosyl) -1,2, 4-triazol-3-karboksamid.
Eksempel II
1- (/ 3- D- ribof uranosyl) - 1, 2, 4- triazol- 3- karboksamid 5' - fosf at-ammoniumsalt
En løsning av 1- (Æ-D-ribofuranosyl)-1,2,4-triazol-3-karboksylsyre-metylester (259 mg, l,oo mmol), trimetylfosfat (3.o ml) og fosforyl-klorid (o.2o ml) omrøres ved 0° C i 1,5 time. Det tilsettes isvann og løsningen nøytraliseres med vandig natriumhydrogenkarbonat. Løsningen ekstraheres med kloroform og den vandige fase avkjøles til 0° C og mettes med ammoniakk. Løsningen holdes ved 25° C i 16 timer og da filtreres den og filtratet konsentreres til et lite volum.
Tilsetning av etanol ga en utfelling som løses i vann og sendes gjennom en Bio-Rad AG5oW-X2 (NH4) kolonne (2o ml). Konsentrasjonen av fraksjonene som inneholdt nukleosidet ga ammoniumsaltet av 1- (/3-D-ribofuranosyl) -1, 2, 4-triazol-3-karboksamid 5' -fosfat
(19o mg, 53, o%).
Analyse: Teoretisk for CgH^NgOgP.f^O : C = 26,74, H = 5, o5,
N = 19,5o.
Funnet: C = 26,78, H = 5,23, N = 19,68.
Eksempel III
1- ( 2, 3, 5- tri- D=racetvl- - D- ribofuranosyl) - 1, 2, 4- triazol- 3-karboksamid
En løsning av 1-(tø-JD-ribofuranosyl)-1,2,4-triazol-3-karboksamid (488 mg, 2,oo mmol) i pyridin (lo-,o ml) som inneholder eddiksyre-anhydrid (l,o ml) holdes ved o - 5° C i 22 timer. Etanol (l,o ml) tilsettes og løsningen omrøres ved romtemperatur i 2 timer. Løsningsmidlet fjernes og vann tilsettes til resten. Blandingen ekstraheres med metylenklorid (3 porsjoner hver på 2o ml) og det organiske lag ekstraheres méd vannholdig natriumhydrogenkarbonat og vann. Den organiske fase tørkes så over magnesiumsulfat, filtreres og inndampes til en sirup, som tilføres til en kolonne av silika-gel. Det elueres med kloroform-metanol i forhold 19:1 som gir et rent produkt som et amorft fast stoff (56o mg, 75.6%).
Analyse: Teoretisk for ci4Hi8N4°8 : c = 45,4o, H = 4,9o,
N = 15,13.
Funnet: C = 45,36, H = 4,66, N = 14,9o.
Farmakologisk prøving
1- (2, 3, 5-tri-0-acetyl-/3-D-ribof uranosyl) -1, 2, 4-triazol-3-karboksamid gir en vesentlig økning av antall overlevende dyr såvel som midlereoverlevelsestid for de behandlende dyr relativt til kontrolldyr når forbindelsen tilføres oralt to ganger over et tidsrom av 9 dager i doser av 125 og 25o mg/kg pr. dag til mus som er smittet med influensa virus (Jap 3o5).
Således ble antivirale midler, fremstilt ifølge oppfinnelsen testet overfor både små og store virus av både DNA og RNA typer ved en virusvurderings (VR)-metode ifølge Sidwell :et al> Appl. Microbiol. 22, 797 (1971). V.R.> l.o indikerer en entydig antiviral aktivitet. V.R. i området o,5 - o,9 indikerer en moderat antiviral aktivitet, V.R. <o.5 antyder en liten eller ingen synlig antiviral aktivitet. Resultatene er angitt i tabellen som for sammenlignings skyld omfatter data fra forskjellige kjente antivirale midler, og resultatene er oppnådd ved forsøk på Microtest II (Falcon Plastics) plastplater med et enkelt lag av
KB eller RK13 celler.
Av de virustypene som benyttes i undersøkelsen er herpes type 1 anvendt i labialis (forkjølelses-sår), herpes keratitis og herpes encephalitis. Herpes type 2 forårsaker herpes genitalis, en vanlig og smittsom form for venerisk sykdom. Myxoma forårsaker død hos tamme og ville kaniner med forutgående åndedrettsvansker og kraftig oppsvelling. Pseudorabies forårsaker smittsom bulbarlammelse kjent som "mad itch" sykdom hos kveg, sauer, griser, hunder og mink. Parainfluensavirus forårsaker en mild øvre luftveisykdom hos mennesker, spesielt hos små barn, og opptrer ved kvegkonjunktivitis. Vaccinia er en avirulent form av koppevirus benyttet for koppe-vaksinasjon og bruk av denne resulterer av og til i uønskede virkninger. Rhino type 13 er en av de mange virusene som opptrer ved en vanlig forkjølelse. Den første forbindelse i tabellen ble testet med kaniner og hamstere med ikke-giftige doser og den viser vesentlig anti-HSV-keratitis aktivitet. Forbindelsen hindrer vesentlig ut-viklingen av HSV-induserte lesjoner i musehaler når det tilføres lokalt til infeksjon. Den har også blitt testet i mus mot influensa A2, influensa B og parainfluensa 1 og disse resultatene har vist en meget betydelig antiviral aktivitet. Denne forbindelse slik som det fremgår av tabellen, viser et spektrum av antiviral aktivitet som vesentlig er bredere enn det som vises av IDU, Ara-A eller Ara-C. I tillegg til den antivirale aktivitet angitt ovenfor er det også funnet at 1- (/?,^D-ribofuranosyl)-1, 2, 4-triazol-3-karboksamid hindrer vekst av bakterien Pseudomonas aeruqinosa og soppen Candida albicans og Cryptococcus diffluents.
Forbindelsene fremstilt ifølge oppfinnelsen viser lav cyto-toksisitet og de er løselige i vann.
1- (/3-D-ribof uranosyl)-1, 2 , 4-triazol-3-karboksamid viser også en interessant antitumor-aktivitet. Tre grupper av C 57 Blk/6 mus
(6 dyr i hver gruppe) ble gitt subkutan tilførsel av adeno-karcinoma-755. To grupper mottok forbindelsen (loo mg/kg x 7 dager og 2oo mg/kg x 7 dager) intraperitonealt, mens den tredje kontroll-gruppe bare mottok en saltløsning. Etter 16. døgn viste de behandlede dyr henholdsvis 18% og 63% hemning av tumor sammenlignet med kontrolldyrene. I et lignende eksperiment ble DBA/2 mus inokulert med L-121o leukemia (1 x lo celler pr. dyr), og det viste 31% økning i midlere overlevelsestid sammenlignet med kontrolldyrene når dyrene ble behandlet med forbindelsen (25o mg(kg x 7 dager).
Ved tilførsel av en dose lik 25o mg/kg i 14 dager ga forbindelsen 8o% overlevende mus (Swiss) som har intraperitonal tilførsel av Ehrlich ascites carcinoma. Marginal in vivo aktivitet overfor Novikoff hepatoma var også observert.
Claims (1)
- Analogifremgangsmåte for fremstilling av farmakologisk aktive ribofuranosyl-1,2,4-triazol-3-karboksamid—forbindelser med den generelle formel hvori Rp er H eller acetyl og R, har samme betydning som R2 eller står for fosfat,karakterisert ved 'at man behandler en l-C-O-acyl-blokkert-A-D-ribofuranosyl)-l,2,4-triazol-3-karboksyl- svreester'med den generelle formel hvori R' er lavere alkyl og Ac står for acetyl eller benzoyl ved at enten a) for fremstilling av en forbindelse med formel I hvori R, og R er H, forsepes forbindelser med formel II for å fjerne acylgruppene etterfulgt av behandling med ammoniakk, eller b) for fremstilling av forbindelsen med formel I hvori R, er fosfat og R2 er H, forsepes forbindelser med formel II for å fjerne acylgruppene etterfulgt av fosforylering og behandling med ammoniakk, eller c) for å danne en forbindelse med formel I hvori R^ og R2 er acetyl omsettes en for* indelse med formel II ved sekvensen for reaksjonen under a) etterfulgt av re-acylering.
