Description
Foreliggende oppfinnelse angår fremstilling av nye anti-atherosclerose-midler som er egnet til senkning av for høye kolesterol-nivåer eller lipider i blodet.
Atherosclerose er en voksen-sykdom som man ikke kjenner noen tilfredsstillende helbredelsesmetode for. Selv om årsakene til atherosclerose ennå ikke er kjent til tross for disku-sjoner i akademiske kretser, har man stort sett erkjent at en av de tydligste histo-patalogiske manifestasjoner for atherosclerose er avsetning av lipider i blodet. Følgelig har man rettet forskning mot forstyrret fettstoff-metabolisme og har viet oppmerksomheten til det ekstraordinært høye innholdet av kolesterol i blodet.
Mange eksprimentelle og kliniske data er rapportert, som tyder på en sammenheng mellom atherosclerose og høye kolesterol-eller fettstoff-innhold i blodet. Følgelig anser man at ut-vikling av midler som kan nedsette for høye kolesterol- og fettstoff-innhold i blodet, er meget viktige for å hindre atherosclerose.
Man har tidligere forsøkt å finne slike midler som senker kolestérolinnholdet og fettstoffinnholdet i blodet og en rekke forbindelser er testet klinisk, men ingen av dem har vist seg å være helt tilfredsstillende. Enkelte er temmelig effektive, men fremkaller tydelige og uheldige bivirkninger og andre har utilstrekkelig virkning,slik at de må gis i store doser.
En gruppe forbindelser som for tiden brukes for formålet, er etyl a- (p-klorf enoks_y) isobutyrat. Imidlertid er virk-
ningen ikke særlig god og forbindelsen har tendens til å gi skadelige bivirkninger, som lever-hypertrofi. Man har nå funnet en ny klasse forbindelser som er effektive som kolesterol-
senkende midler og som er i det vesentlige ugiftige.
Fra norsk patent nr. 127298 er det kjent forbindelser som er nær beslektet med forbindelsene som fremstilles ifølge foreliggende oppfinnelse, men dé sistnevnte har en bedre kolesterolsenkende virkning enn de fra patentet kjente forbindelser. En sammenligning som viser dette er angitt senere i dette skrift.
De nye terapeutisk virksomme cykloalkylfenoksykarboksylsyre-derivatene som fremstilles ifølge oppfinnelsen, har den generelle formel:
hvor R1? R2 og R3 uavhengig av hverandre betegner hydrogen eller C1~C4 alkyl, og n betegner et helt tall,på 4 eller 5.
Ifølge oppfinnelsen fremstilles forbindelsene med formel (I) ved at man
(a) omsetter ét bisfenolderivat med formelen:
hvor n har den overfor angitte betydning, med en•ketonforbindelse med formelen: -'■ . hvor og R_ ^ar betydning som angitt ovenfor, samt kloroform, i nærvær av alkali, for dannelse av et cykloalkylfenoksykarboksylsyre-derivat med formelen: hvor R^, R2 og n har betydning som angitt ovenfor, eller (b) omsetter et bisfenol-derivat med formelen:
hvor n har betydning som angitt ovenfor, med et karboksylsyrederivat med formelen:
hvor X betegner halogen eller hydroksyl, og R^, R2 og R^ har betydning som angitt ovenfor, for fremstilling av et cyklo-alkylf enoksykarboksylsyre-derivat med formel (I),
og, om ønsket, omdannelse av et erholdt produkt til et farmasøytisk akseptabelt salt derav.
Ovennevnte fremgangsmåte illustreres detaljert på følgende måte:
Metode a)
Omsetning av et bisfenolderivat ( II) med kloroform og en keto- forbindelse ( III) i nærvær av alkali.
For gjennomføring av denne reaksjon settes minst 1 mol kloroform dråpevis til en blanding som inneholder 1 mol bisfenol-derivat (II) og minst 1 mol keto-forbindelse (III) i nærvær av minst 3 mol av en alkaliforbindelse. Eksempler på brukbar alkali er natriumhydroksyd og kaliumhydroksyd. Reaksjonen krever en temperatur på 0-150°C, og vanligvis en temperatur på 5-70°C,
og en reaksjonstid på 3-40 timer. For å få et fenoksydikarboksyl-
syre-derivat (Ia), bør reaksjonsforholdene som f.eks. mengdeforhold mellom de anvendte reaktanter, reaksjonstemperaturen og reaksjonstiden, passes nøye. Ved anvendelse av kéto-forbindelse, kloroform og alkali i overskuddsmengder, får man et bisfenoksydikarboksylsyre-derivat (Ia).
Reaksjonen kan foretas i nærvær av overskudd av kloroform og keto-forbindelse (III), i nærvær av eller uten et inert reaksjonsmedium. Eksempler på slike reaksjonsmedia er dioksan, benzen, toluen etc.
Metode b)
Kondensasjonsreaksjon mellom et bisfenolderivat ( II) med et a- halogen- eller hydroksy- karboksylsyre- derivat ( IV).
Når X betegner halogen, kontaktes 1 mol bisfenolderivat (II) i oppløst eller suspendert form i inert reaksjonsmedium med minst 1 mol alkali under dannelse av et alkalisalt, og derpå settes minst 1 mol a-halogenert karboksylsyrederivat (IV) (dvs. X = halogen) til den dannede reaksjonsblanding for gjennom-føring av kondensasjonsreaksjonen. Når reaksjonen er avsluttet gjennomgår reaksjonsblandingen videre vanlig renseprosesser for opparbeidelse av de ønskede fenoksykarboksylsyre-derivater (I). Eksempler på inerte reaksjonsmedia som brukes til denne frem-gangsmåten er benzen, toluen, metanol, etanol, eter, dioksan, dimetylsulfoksyd, N,N-metylformamid etc. Eksempler på alkalier er kaliumhydroksyd, natriumhydroksyd, alkalimetallalkoholater, alkalimetallkarbonater, metallisk natrium, natriumhydrid og organiske tertiære'aminer, som trimetylamin, trietylamin og pyridin. Reaksjonen krever eri temperatur på 0-120°C.
For å få et fenoskydikarboksylsyre-derivat (I), må reaksjonsbetingelsene, som mengdeforhold mellom reaktanter, reaksjonstemperatur og reaksjonstid, velges omhyggelig. Hvis alkalisk reagens og/eller ct-halogenkarboksylsyre-derivat (IV) brukes i ekvimolar mengde til bisfenolderivat (II), får man hovedsakelig et fenoksymonokarboksylsyre-derivat (I). Hvis man på den annen side bruker 2 eller flere mol av både alkali og a-halogenkarboksylsyre-derivat (IV) pr. mol bisfenolderivat (II), får man et fenoksydikarboksysyre-derivat (I) i hovedsak.
Når X er hydroksyl, føres 1 mol bisfenolderivat (II) i kontakt med minst 1 mol a-hydroksykarboksylsyre-derivat (IV)
(X = OH) i nærvær av en syrekatalysator, som svovelsyre,
p-toluensulfonylklorid, arsensyre, borsyre, natriumhydrogensul-fat, kaliumhydrogensulfat, etc. i nærvær eller fravær av inert reaksjonsmedium. Eksempler på reaksjonsmedier er benzen, toluen, dioksan, etc. Den anvendte syrekatalysator brukes i en mengde på 0,01-0,5 mol pr. 1 mol bisfenolderivat. Reaksjonen krever en temperatur på 10-90°C. Det er viktig å velge reaksjonsbetingelsene, som mengdeforhold, reaksjonstemperatur og reaksjonstid, nøye.
Et fenoksykarboksylsyre-derivat (Ia) eller dets reaktive ester omdannes til en ester ved vanlige forestringsmetoder,
f.eks. ved behandling med et forestringsmiddel. Uttrykket "reaktiv ester" betegner her et fenoksykarboksylsyre-derivat (Ia), et acylhalogenid, syreanhydrid, ester av en syre, syresalt, etc. og uttrykket "forestringsmiddel" betegner en alkohol, diazometan, et dialkylsulfat, alkylhalogenid, alkylhalogensulfitt etc.
Det fremstilte fenoksykarboksylsyre-derivat (I) hvor
R^ betegner hydrogen, omdannes til et salt ved behandling med
et alkali. Saltet dannes ved karboksyl- og/eller den fenoliske hydroksylgruppe. Et alkalimetallsalt kan fremstilles ved å bringe fenoksykarboksylsyre-derivat (I), hvor R^ betegner hydrogen, i kontakt med natriumhydroksyd, kaliumhydroksyd, natrium-karbonat, kaliumkarbonat, natriumbikarbonat eller ammoniakk, etc. eller med et alkoholat av et alkalimetall, som natriummetylat, i et organisk oppløsningsmiddel, fortrinnsvis i en lav alkanol,
som metanol eller etanol, eller med et hydroksyd, karbonat eller bikarbonat av et alkalimetall i et organisk oppløsnings-middel, fortrinnsvis aceton eller metanol, om nødvendig i nær-
vær av en liten mengde vann. Det dannede alkalimetallsalt kan overføres til et jordalkalimetall ved behandling med et jord-alkalimetallsalt, som kalsiumklorid.
Bisfenol-derivater med formel (II) som brukes som ut-gangsstoffer, kan f.eks. være 1,2-bis(4'-hydroksyfenyl)-cykloheksan, 1,3-bis(4'-hydroksyfenyl)cyklopentan, l,4-bis(4'-hydroksyfenyl)cyklopentan, 1,2-bis(4<1->hydroksyfenyl)cykloheptan, 1,3-bis-(4<1->hydroksyfenyl)cykloheptan og 1,4-bis(4'-hydroksyfenyl)-cykloheptan. F.eks. kan 1,2-bis(4',hydroksyfenyl)cykloheksan fremstilles i henhold til en metode som er beskrevet i J. Amer. Chem. Soc, 11, 3313 (1949), 1, 3-bis (4 1-hydroksyf enyl) cyklo-heksan kan fremstilles ifølge J. Amer. Chem. Soc, 7_3/ 2377 (1951), og 1,4-bis(4'-hydroksyfenyl)cykloheksan ifølge J.Amer. Chem. Soc., 74, 5631~(1952).
Kolesterol-sehkende midler fremstilt i henhold til oppfinnelsen kan f.eks. gis oralt, vanligvis er den orale dosen 0,01-10 g pr. dag/voksen pasient, fortrinnsvis 0,05-3 g pr. dag/voksen pasient. De kolesterol-senkende midler kan gis en hvilken som helst vanlig form for oral administrasjon. Så-ledes kan de gis som kapsler eller i flytende form, tablettform eller som pulver. For fremstilling av preparatene i disse for-skjellige former, kan den aktive forbindelse blandes med eller impregneres i farmasøytiske bærestoffer og blandemedia, som laktose, potetstivelse, maisstivelse, cellulosederivater, gelatin, maisolje, bomullsfrøolje, etc.
Den kolesterolsenkende virkning av forbindelsene med formel (I) på mus, ble undersøkt ved å injisere forbindelsene intravenøst med 500 mg pr. kg "Triton WR 1399" (oksyetylert tert-oktylfenolformaldehyd-polymer).
Forsøksforbindelsen ble gitt oralt i en dose på 12,5 mg pr. kg umiddelbart etter injeksjon. Musene ble drept og man gjennomførte en analyse av serum-kolesterolet. Den'kolesterol-senkende virkning ble beregnet ut fra følgende ligning:
hvor C = serumkloesterolinnhold (mg/100 ml) i en gruppe på 24 mus målt etter injeksjon med triton og før behandling med for-søksf orbindelsen,
T = serumkolesterolinnhold (mg/100 ml) i en gruppe på 12 av musene injisert med triton og behandlet med forsøksfor-bindelse målt etter injeksjon og behandling, og
N = serumkolesterolinnhold (mg/100 ml) hos en gruppe på 12 ubehandlede mus (ingen triton eller forsøksforbindelse gitt).
Et eksempel på resultater oppnådd ved disse forsøk fremgår av tabell 1. I tabell 1 refererer tallene seg til forbindelsene fremstilt i de tilsvarende eksempler.
Oppfinnelsen skal nå beskrives mere detaljert i lys av følgende eksempler.
Eksempel 1
Til en blanding av 15 g 1,3-bis(4'-hydroksyfenyl)cyklo-heksan og 250 g aceton satte man 75 g kaliumhydroksyd. Derpå tilsatte man 40 g kloroform til blandingen under røring ved 10-20°C, og blandingen ble oppvarmet ved 30-40^C i 3 timer for å avslutte reaksjonen. Derpå ble den konsentrert til et residum. Etter oppløsning av konsentrasjonsresten i vann, ble oppløs-ningen behandlet med aktiv kull og syregjort med fortynnet saltsyre, som gav en oljeaktig forbindelse. Denne oljen ble ekstrahert med eter og eteroppløsningen ekstrahert på nytt med vandig fortynnet Na2CC>2-oppløsning. Det fraskilte alkaliske vannsjikt ble surgjort-og ekstrahert med eter. Etersjiktet ble tørket over vannfri natriumsulfat og konsentrert til et rå-produkt som ble renset ved omkrystallisering fra toluen. Man fikk den ønskede fenoksydikarboksyl-syre, 18 g, smeltepunkt 160,5°-162°C.

Eksempel 2- 6
På analog måte som beskrevet i eksempel 1 fremstilte man de forbindelser som fremgår av tabell 2.
Eksempel 8 Til en blanding av 3 g 1,3-bis(4<1->hydroksyfenyl)cyklo-pentan og 25 ml tørr toluen, satte man 0,5 g natriummetoksyd. Derpå tilsatte man dråpevis 3 g 2-brom-2-metylsmørsyremetylester oppløst i toluen til blandingen, under røring. Blandingen ble oppvarmet ved 60-80°C i 10 timer. Reaksjonsblandingen ble vasket med vann. Derpå ble toluen avdestillert, destillasjons-resten renset ved kolonnekromatografi gjennom kolonne fylt med aktivert aluminiumoksyd. Man fikk den ønskede ester i en mengde 27 på 2,5 g, n 1,5215. Elementær analyse:

På lignende måte som angitt i eksempel 8 fremstilte man de stoffer som fremgår av tabell 3.
Sammenligningsforsøk
Man har foretatt sammenligningsforsøk mellom forbindelser fremstilt ifølge foreliggende oppfinnelse og forbindelser som er kjent fra det ovennevnte norske patent nr. 127.298. Den forsøksmetode som ble benyttet er den samme som beskrevet tidligere i beskrivelsen. De oppnådde resultater er som følger:
