Foreliggende oppfinnelse angår en analogifremgangsmåte til fremstilling av nye terapeutisk aktive anilider av kinuklidin-2- og kinuklidin-3-karboksylsyre og deres salter med den generelle formel:

hvor R og R kan være like eller forskjellige og hver representerer et hydrogen- eller halogenatom eller en alkylgruppe med høyst 3 karbonatomer, samt terapeutisk akseptable salter av disse, forbindelser..
Disse nye derivater er stabile og har verdifulle farmakolog-
iske egenskaper. Spesielt har de vist seg å ha antiarrytmiske og/eller
lokale bedøvende effekter. Som foretrukne forbindelser fremstilt ifølge oppfinnelsen skal nevnes kinuklidin-2-karboksylsyre o-toluidid, kinuklidin-2-karboksylsyre 2,6-xylidid, kinuklidin-2-karboksylsyre 2-metyl-6-etylanilid og kinuklidin-2-karboksylsyre 2,6-dietylanilid samt terapeutisk akseptable salter derav.
Ifølge foreliggende oppfinnelse blir forbindelser med formel
I fremstilt ved at. en forbindelse med formelen:
tilsvarende syreklorid eller en funksjonell ekvivalent derav, slik som et syrebromid, en ester, spesielt en lavere alkylester, anhydridet, et blandet anhydrid, spesielt et anhydrid dannet med en alkoksymaursyre, eller et derivat oppnådd ved reaksjon mellom nevnte syre og et karbo-diimid eller N,N'-karbonyldiimidazol eller N-etyl-5-fenylisoksazolium-3?-sulfonat, eller et salt .derav, omsettes med en forbindelse med formelen:
hvor R og R har den ovenfor angitte betydning, og Y er hydrogen, Na, Mgl eller -CO-NH-^^T^, hvoretter, om ønsket, den erholdte base med formel I omdannes til terapeutisk akseptable salter derav ved omsetning med tilsvarende syrer.
Det er foretrukket å benytte kinuklidin-2-karboksylsyre eller ett av de angitte derivater som forbindelse II, mens det som forbindelse III er foretrukket å anvende en av de angitte anilinforbindelser

som har substituentene R og R i 2- og 6-stilling.
Med uttrykket "terapeutisk akseptabelt salt" forståes et syre-addisjonssalt som er fysiologisk uskadelig når det administreres i en dose og med slike tidsintervaller (dvs. administrasjonsfrekvens), som er effektive for det angitte terapeutiske bruk med den angjeldende forbindelse. Typisk terapeutisk akseptable syreaddisjonssalter av forbindelser med formel I omfatter, men er ikke begrenset til, syre av mineralsyre eller saltsyre, hydrobromsyre, fosforsyre eller svovel-syre, organiske syrer slik som melkesyre, levulinsyre, sitronsyre, fumarsyre, maleinsyre, ravsyre, vinsyre, benzosyre og sulfonsyre, som f.eks. metansulfo.nsyre og sulfaminsyre.
Utgangsforbindelser med formel II kan fremstilles på fbl-gende måte: kinuklidin-2-karboksylsyre fremstilles ifolge E. Renk et al. (Heiv. Chim. Acta 37 (1954), 2119), Qg kinuklidin-3-karboksylsyre ifolge CA. Grob og E. Renk (Heiv. Chim. Acta 37_ (1954), 1689) og reaktive derivater fremstilt av disse forbindelser ved fremgangsmåter som i seg selv er kjente.
Forbindelsene med formel I fremstilles fordelaktig ved åt en lavere alkylester av kinuklidin-karboksylsyrer omsettes med Grignard-reagensen av aniiinforbindelsen eller ved at anhydridet av kinuklidin-karboksylsyre omsettes med aniiinforbindelsen.
Ved klinisk anvendelse vil derivater fremstilt ifolge foreliggende oppfinnelse normalt bli administrert oralt eller ved injeksjon i form av farmasSytiske preparater omfattende den aktive bestanddel enten i form av den frie base eller som et vanlig terapeutisk akseptabelt salt, som f.eks. hydroklorid, sammen med et farmasøytisk akseptabelt bærestoff som kan være fast, halvfast ; eller flytende eller sammen med en fordbybar kapsel. Vanligvis vil den aktive bestanddel utgjore fra 0,1 til 85 vektprosent av preparatet, f.eks. mellom 0,5 og 5 vektprosent i preparater for injeksjon bg mellom 2 og 50 % i preparater for oral administrasjon.
Forbindelsene fremstilt ifolge foreliggende oppfinnelse er virksomme antiarrytmiske midler ettersom de foruten sterke lokale bedbvende effekter (tabell 1) også nedsetter eksitabiliteten og forlenger refraktærperioden bg overledningstidén i hulrommene i hjertet hos marsvin, (tabell 2).
Følgende eksempler illustrerer oppfinnelsen.
Eksempel 1. Kinuklidin- 2- karboksylsyre o- toluidid
En blanding av 4.5 g metylkinuklidin-2-karboksylat, 2.9-g o-toluidin og 0,1 g natrium ble oppvarmet ved l40°C i 5 timer. Reaksjonsblandingen ble så behandlet med vann og eter, det utskilte vannlag.ble ekstrahert to ganger med eter og.de samlede eterløsninger"ble så ekstrahert med fortynnet saltsyre. Det sure ekstrakt ble gjort sterkt alkalisk og det utfelte krystallinske stoff omkrystallisert fra vandig alkohol. Utbytte.2.1 g, smeltepunkt 115.5° -117°C.

Eksempel 2. KinukTidin- 2- karboksylsyre b- kloranilid
Ved hjelp av samme fremgangsmåte som er beskrevet i eksempel 1, men ved å erstatte o-toluidin med 3-5 g o-kl<p>ranilin og utføre opp-varmningen i 15 timer, ble kinuklidin-2rkarboksylsyre o-kloranilid fremstilt. Utbytte 2.6 g, smeltepunkt 117° - 119.5°C (fra 60* vandig alkohol).

2.42 g 2.6-xylidin i 15 ml eter ble dråpevis tilsatt til en løsning av metylmagnesiumjodid fremstilt fra.0.49 g magnesium og 2.85 g metyljodid i .20 ml eter. Deretter ble 1,69 g metyl-kinuklidin-2-kar-boksylat i 10 ml eter tilsatt, og blandingen ble tilbakeløpskokt i 3 timer. Reaksjonsblandingen ble deretter behandlet med fortynnet saltsyre, den vandige fase utskilt og pH justert til 5.7.' Etter ekstrak-sjon med eter (ekstraktet, inneholdende ureagert xylidin ble kastet), ble løsningen gjort sterkt alkalisk, og den. utfelte base ekstrahert
med eter. Etter tørking over kaliumkarbonat ble basen omdannet til hydroklorid,-som så ble omkrystallisert■fra etanol-diisopropyleter. Utbytte 0.85 g, smeltepunkt 22 3° - 225°C. Cl6H22N2° x HC1 Beregnet: C 65.18*, H 7.86*, N 9.50*
Funnet: C 65.4 *, H 7.89*, N 9.68*. Eksempel 4. Kinuklidin- 2- karboksylsyre 2- metyl- 6- etylanilid.
Anhydridet av kinuklidin-2-karboksylsyre hydroklorid ble fremstilt ved hjelp av fremgangsmåten til H. Rinderknecht (Heiv. Chim. Acta 47 (1964), 162). Suspensjonen fremstilt ved å blande 3.85 g.- kinuklidin-2-karboksylsyre hydroklorid og 2.0 g trietylamin i 40 ml kloroform ble behandlet dråpevis med en løsning av 1.0 g fosgen i 10 ml toluen. Blandingen ble hensatt ved romtemperatur over natten, hvoretter en løsning av 2.7 g 2-metyl-6-etylanilin i 25 ml benzen ble tilsatt. Etter tilbakeløpskoking i 2 timer ble blandingen ekstrahert tre ganger med vann, og det sure vandige ekstrakt underkastes samme fremgangsmåte som beskrevet i eksempel 1. Man fikk fremstilt 1.2 g av hydrokloridet med smeltepunkt 203° - 208°C. Omkrystalli-sering fra metylpropylketon hevet smeltepunktet til 209° - 211.5°C.

Funnet: C 65.9 *, H 8.14*, N 9.07*, Cl 11.5 *. Eksempel 5. Kinuklidin- 2- karboksylsyre 2, 6- dietylanilid.
Dette anilid ble fremstilt på samme måte som beskrevet i eksempel 4 fra anhydridet og 2,6-dietylanilin. Hydrokloridet hadde et smeltepunkt på 209.5° - 211.5°C (fra acetonitril).
Cl8H26N2° x HC1 Beregnet: C 66.96*, H 8.43*, N 8.68*, Cl.10.97*
Funnet: C 66.7 *, H 8.20*, N 8.66*, Cl.11.1 *. Eksempel 6. Kinuklidin- 3- karboksylsyre o- kloranilid
En blanding av 1.9 g kinuklidin-3-karboksylsyrehydroklorid og 20 ml tionylklorid ble refluksert i 2.5 timer. Overskuddet av tionylklorid ble så avdestillert, til slutt ved hjelp av to porsjoner benzen. Residuet ble oppløst i 25 ml kloroform hvoretter 6.5 g o-kloranilin ble tilsatt. Etter at den svakt eksoterme reaksjon var stoppet opp, ble blandingen tilbakeløpskokt i 1 time. Det utfelte anilinhydroklorid ble frafiltrert, og filtratet ble ekstrahert med fortynnet saltsyre. pH i ekstraktet ble justert til 5.5 og overskuddet av kloranilinet ekstrahert med eter. Løsningen ble så gjort alkalisk (pH 10). Den utfelte base veide 1,8 g med et smeltepunkt på 157° 165°C. To om-krystalliseringer fra métyl-isobutylketon hevet smeltepunktet til 166.5° - 168.5°C.

Funnet: C 62.5 *, H 6.33*, N 10.9 *, Cl-13.48*.
Eksempel 7» Kinukli din- 3- karboksylsyreariilid
1.55 g kinuklidin-3-karboksylsyre og 2.12 g N,N'-difenylurea ble blandet og oppvarmet til 210°C med omrøring. Den oppnådde smelte ble holdt på denne temperatur i 4 timer med kontinuerlig omrøring.
Etter avkjøling ble den svartbrune reaksjonsblanding oppløst
i fortynnet saltsyre. Denne løsning<?>s pH ble justert til 5.7, og løs-
ningen ble så vasket med eter.. Den ble derpå gjort sterkt alkalisk hvoretter den utfelte base ble ekstrahert med eter. Ekstraktene ble tørket over magnesiumsulfat. Den filtrerte eterløsning ble noe kon-
sentrert og deretter avkjølt, hvorpå, man fikk et utbytte på 0.50- g av produktet med smeltepunkt 177° - 1.79.5°C. Omkrystallisering fra. metyl-
isobutylketon hevet smeltepunktet til 178° - 180°C.

1. Analogif remgangsmåte t-il fremstilling av terapeutisk aktive
anilider av kihuklidin-2- og kinuklidin-3-karboksylsyre med formelen: 12.
hvor R og R er like eller■forskjellige og hver er et hydrogen- eller
halogenatom eller en 'alkylgruppe med høyst 3 karbonatomer, og terapeu
tisk akseptable salter derav, karakterisert ved at
en forbindelse med formelen: •,,'.• ..-
eller et
tilsvarende syreklorid eller en funksjonell ekvivalent derav, slik som
et syrebromid, en ester, spesielt en lavere alkylester, anhydridet, et
blandet anhydrid, spesielt et anhydrid dannet med en alkoksymaursyre,
eller et derivat oppnådd ved reaksjon mellom nevnte syre og et karbodi-
imid eller N,N'-karbonyldiimidazol eller N-etyl-5-fenylisoksazolium-3'-
sulfonat, eller et sålt derav, omsettes med en forbindelse med formelen: 1 2

3. Analogifremgangsmåte ifolge krav 1, karakterisert ved at metyl-kinukli.din-2-karboksylat omsettes med 2,6-xylidin-magnesiumjodid.
4. Analogifremgangsmåte ifolge krav 1, karakterisert ved at kinuklidin-2-karboksylsyre hydrokloridanhydrid omsettes med 2-metyl-6-etylanilin.
5. Analogifremgangsmåte ifolge krav 1, karakterisert ved at kinuklidin-2-karboksylsyre hydrokloridanhydrid omsettes med 2,6-dietylanilin.
