Description
Fremgangsmåte til fremstilling av et godt vannoppløselig rescinnaminsalt.
I hovedpatent nr. 87.348 er det bl. a.
beskrevet fremstilling av rescinnamin,
3,4,5-trimetoksy-kanelsyreesteren av me-tylreserpat, samt fremstilling av salter av
reserpinsyre-diestere. Rescinnamin, som er
et verdifullt terapeutisk virksomt alkaloid,
som utmerker seg ved sedativ og hypotensiv
virkning, er vannuoppløselig og dermed
bare lite egnet for parenteral anvendelse.
Heller ikke det saltsure og svovelsure salt
av denne forbindelse har den vannoppløse-lighet som er ønsket for en slik anvendelse.
Det ble nå funnet at man kan fremstille et godt vannoppløselig og ikke giftig
salt av rescinnamin når man overfører rescinnamin i dets fosforsure salt. Man kan
derved arbeide på i og for seg kjent måte.
Således lar rescinnamin eller et av dets
salter seg omsette med fosforsyre eller et
av dens salter. Fortrinnsvis oppløser man
rescinnaminet i metanolisk fosforsyre og
feller det dannede salt med eter.
Det nye salt er overraskende godt opp-løselig i vann og dermed på fremragende
måte egnet for vandige parenteral-anven-delige oppløsninger, som har den samme
virkning som selve det vannuoppløselige
rescinnamin. Slike oppløsninger kan inne-holde 0,1 til 250 mg, fortrinnsvis 0,2—50 mg,
rescinnamin-fosfat pr. enkeltdose.
Oppfinnelsen beskrives i det følgende
eksempel. Temperaturen er angitt i celsius-grader.
Eksempel:
10 g rescinnamin oppløses i en oppløs-
ning av 2,01 g 85 pst.'s fosforsyre i 30 cm3
metanol, og oppløsningen befris ved filtre-ring fra spor av uoppløselige stoffer. Filt-ratet tilsettes under god omrøring 300 cm» eter, deretter røres det enda noen minutter, og det utfelte salt samles på en Buchner-trakt. Man vasker etter med 200 cm3 eter og suger fra i vakuum. Resten vasker man deretter for å fjerne resterende fosforsyre to ganger med hver gang 50 cm« av en 8 : 2-blanding eter-metanol, og endelig
flere ganger med litt eter. Deretter tørker man resten ved romtemperatur over kalsi-umklorid og fosforpentoksyd i to dager i vakuum (ved omtrent 20 mm kvikksølv - trykk) og over fosforpentoksyd videre i 18 timer ved omtrent 1 mm kvikksølvtrykk. Man får således 10,6 g svakt gulaktig res-cinamin-fosfat, som inneholder omtrent 2 mol vann. Det sintrer ved 181° og smelter ved 183—188°. Ved forsterket tørkning får man rescinnaminfosfat i praktisk taltvann-fri form.
En parenteralt anvendelig oppløsning av denne forbindelse lar seg fremstille på følgende måte: Man oppløser 100 mg rescinnamin-fosfat i 10 cm<;>i vann, filtrerer gjennom et mikrobiologisk porselensfilter, hvoretter den sterile oppløsning kan anven-des direkte for injeksjoner. Den lar seg også, i tilfelle det er ønsket, lyofilisere i am-puller.
Claims (2)
1. Fremgangsmåte til fremstilling av et godt vannoppløselig rescinnaminsalt ifølge hovedpatent nr. 87.348, karakterisert ved at man overfører rescinnamin i dets
fosforsure salt ved omsetning av rescinnamin eller et av dets salter med fosforsyre eller et av dens salter.
2. Fremgangsmåte ifølge påstand 1,karakterisert ved at man oppløser rescinn
amin i metanolisk fosforsyre og feller ut det dannede salt med eter.
